Abstract:Oxygen (O2)-delivering tissue substitutes have shown tremendous potential for enhancing tissue regeneration, maturation, and healing. As O2 is both a metabolite and powerful signaling molecule, providing controlled delivery is crucial for optimizing its beneficial effects in the treatment of critical-sized injuries. Here, we report the design and fabrication of 3D-printed, biodegradable, O2-generating bone scaffold comprising calcium peroxide (CPO) that once hydrolytically activated, provides long-term generation of oxygen at a controlled, concentration-dependent manner, and polycaprolactone (PCL), a hydrophobic polymer that regulate the interaction of CPO with water, preventing burst release of O2 at early time points. When anoxic conditions were simulated in vitro, CPO-PCL scaffolds maintained the survival and proliferation of human adipose-derived stem/stromal cells (hASCs) relative to PCL-only controls. We assessed the in vivo osteogenic efficacy of hASC-seeded CPO-PCL scaffolds implanted in a non-healing critical-sized 4-mm calvarial defects in nude mice for 8 weeks. Even without exogenous osteoinductive factors, CPO-PCL scaffolds demonstrated increased new bone volume compared to PCL-only scaffolds as verified by both microcomputed tomography analysis and histological assessments. Lastly, we employed a quantitative 3D lightsheet microscopy platform to determine that O2-generating scaffolds had similar vascular volumes with slightly higher presence of CD31hiEmcnhi pro-osteogenic, type H vessels and increased number of Osterix+ skeletal progenitor cells relative to PCL-only scaffolds. In summary, 3D-printed O2 generating CPO-PCL scaffolds with tunable O2 release rates provide a facile, customizable strategy for effectively treating, craniofacial bone defects. STATEMENT OF SIGNIFICANCE: Oxygen(O2)-delivering bone substitutes show promise in defect repair applications by supplying O2 to the cells within or around the graft, improving cell survivability and enhancing bone matrix mineralization. A novel O2-generating bone scaffold has been 3D printed for the first-time which ensures patient and defect specificity. 3D printed calcium peroxide-polycaprolactone (CPO-PCL) bone scaffold provides uninterrupted O2 supply for 22 days allowing cell survival in deprived O2 and nutrient conditions. For the first time, O2-driven bone regenerative environment in mice calvaria has been captured by light-sheet imaging which illuminates abundance of Osterix+ cells, angiogenesis at a single cell resolution indicating active site of bone remodeling and growth in the presence of O2.

Abstract:Oxygen is essential for cell survival and tissue regeneration. Scaffolds releasing oxygen have been hypothesized as an ideal strategy for bone repair. However, excessive oxygen supply will disturb the redox balance, lead to oxidative stress, and affect bone regeneration. In this study, we synthesized a hydrogel from sodium alginate and loaded it calcium peroxide nanoparticles as an oxygen generating material and vitamin C as a pH regulator and antioxidant. The composite hydrogel, with a pH value close to physiological humoral fluid, could release oxygen to alleviate hypoxia in the bone defect and reduce the side effects of excessive hydrogen peroxide. In in vitro experiments, the composite hydrogel promoted the osteogenic differentiation and ALP and mineralization ability of rat bone marrow mesenchymal stem cells in a hypoxic environment (2% O 2 ). In animal experiments, the composite hydrogel was applied in rat skull defect models. It promoted the healing of bone defects. These results suggest that sodium alginate hydrogel releasing oxygen and vitamin C is suitable for cell survival and tissue regeneration in a hypoxic environment and has good application prospects in bone defect repair.

Abstract:Limited cells and factors, inadequate mechanical properties, and necrosis of defects center have hindered the wide clinical application of bone-tissue engineering scaffolds. Herein, we construct a self-oxygenated 3D printed bioactive hydrogel scaffold by integrating oxygen-generating nanoparticles and hybrid double network hydrogel structure. The hydrogel scaffold possesses the characteristics of extracellular matrix; Meanwhile, the fabricated hybrid double network structure by polyacrylamide and CaCl2-crosslinked sodium carboxymethylcellulose endows the hydrogel favorable compressive strength and 3D printability. Furthermore, the O2 generated by CaO2 nanoparticles encapsulated in ZIF-8 releases steadily and sustainably because of the well-developed microporous structure of ZIF-8, which can significantly promote cell viability and proliferation in vitro, as well as angiogenesis and osteogenic differentiation with the assistance of Zn2+. More significantly, the synergy of O2 and 3D printed pore structure can prevent necrosis of defects center and facilitate cell infiltration by providing cells the nutrients and space they need, which can further induce vascular network ingrowth and accelerate bone regeneration in all areas of the defect in vivo. Overall, this work provides a new avenue for preparing cell/factor-free bone-tissue engineered scaffolds that possess great potential for tissue regeneration and clinical alternative.

Abstract:Globally, an annual count of more than two million bone transplants is conducted, with conventional treatments, including metallic implants and bone grafts, exhibiting certain limitations. In recent years, there have been significant advancements in the field of bone regeneration. Oxygen tension regulates cellular behavior, which in turn affects tissue regeneration through metabolic programming. Biomaterials with oxygen release capabilities enhance therapeutic effectiveness and reduce tissue damage from hypoxia. However, precise control over oxygen release is a significant technical challenge, despite its potential to support cellular viability and differentiation. The matrices often used to repair large-size bone defects do not supply enough oxygen to the stem cells being used in the regeneration process. Hypoxia-induced necrosis primarily occurs in the central regions of large matrices due to inadequate provision of oxygen and nutrients by the surrounding vasculature of the host tissues. Oxygen generating biomaterials (OGBs) are becoming increasingly significant in enhancing our capacity to facilitate the bone regeneration, thereby addressing the challenges posed by hypoxia or inadequate vascularization. Herein, we discussed the key role of oxygen in bone regeneration, various oxygen source materials and their mechanism of oxygen release, the fabrication techniques employed for oxygen-releasing matrices, and novel emerging approaches for oxygen delivery that hold promise for their potential application in the field of bone regeneration.

Abstract:Bone defects resulting from trauma, disease, or aging present significant challenges in the clinic. Although biomaterial scaffolds for bone-tissue engineering have shown promising results, challenges remain, including the need for adequate mechanical strength and suitable bioactive agents within scaffolds to promote bone formation. Oxygen is a critical factor for successful bone formation, and low oxygen tension inhibits it. In this study, we developed gelatin methacryloyl (GelMA) hydrogel-impregnated electrospun polycaprolactone (PCL) scaffolds that can release oxygen over 3 weeks. We investigated the potential of composite scaffolds for cell survival in bone-tissue engineering. Our results showed that the addition of an increased amount of CaO2 nanoparticles to the PCL scaffolds significantly increased oxygen generation, which was modulated by GelMA impregnation. Moreover, the resulting scaffolds showed improved cytocompatibility, pre-osteoblast adhesion, and proliferation under hypoxic conditions. This finding is particularly relevant since hypoxia is a prevalent feature in various bone diseases. In addition to providing oxygen, CaO2 nanoparticles also act as reinforcing agents improving the mechanical property of the scaffolds, while the incorporation of GelMA enhances cell adhesion and proliferation properties. Overall, our newly developed self-oxygenating composite biomaterials are promising scaffolds for bone-tissue engineering applications.

Abstract:Despite three-dimensional biomaterial scaffolds emerging for facilitating tissue remodeling and recovery in tissue engineering, the scaffolds with an insufficient oxygen supply are limited by various concerns, such as their retention, localization, survival, and lineage fate control of post-transplantation of stem cells. Herein, we report oxygen-enriched osteoinductive nanoerythrocytes as a facile biomimetic platform for bone formation, combining the biocompatibility of natural red blood cell membranes with the oxygen-carrying perfluorocarbons and the osteoinductive oxysterols. The resulting nanoerythrocytes exhibit a high capacity for oxygen delivery, improving cell survival and proliferation and mitigating hypoxic stress in cell-laden hydrogels as a three-dimensional biomaterial device in vitro and in vivo. Notably, the introduction of oxysterols incites the osteoinductive activity for the differentiation of progenitor cells toward osteogenic lineage through the positive regulation of Hedgehog signaling and in vivo reossification activity against a nonhealing calvarial defect. Given the beneficial properties of the assembled parts in a facile single system, this bioactive and biomimetic nanodelivery system can have the potential to address desired needs in the biomaterial scaffolds for cell delivery and bone tissue engineering.

Abstract:Low oxygen (O2) diffusion into large tissue engineered scaffolds hinders the therapeutic efficacy of transplanted cells. To overcome this, we previously studied hollow, hyperbarically-loaded microtanks (μtanks) to serve as O2 reservoirs. To adapt these for bone regeneration, we fabricated biodegradable μtanks from polyvinyl alcohol and poly (lactic-co-glycolic acid) and embedded them to form 3D-printed, porous poly-ε-caprolactone (PCL)-μtank scaffolds. PCL-μtank scaffolds were loaded with pure O2 at 300-500 psi. When placed at atmospheric pressures, the scaffolds released O2 over a period of up to 8 h. We confirmed the inhibitory effects of hypoxia on the osteogenic differentiation of human adipose-derived stem cells (hASCs and we validated that μtank-mediated transient hyperoxia had no toxic impacts on hASCs, possibly due to upregulation of endogenous antioxidant regulator genes. We assessed bone regeneration in vivo by implanting O2-loaded, hASC-seeded, PCL-μtank scaffolds into murine calvarial defects (4 mm diameters × 0.6 mm height) and subcutaneously (4 mm diameter × 8 mm height). In both cases we observed increased deposition of extracellular matrix in the O2 delivery group along with greater osteopontin coverages and higher mineral deposition. This study provides evidence that even short-term O2 delivery from PCL-μtank scaffolds may enhance hASC-mediated bone tissue regeneration.

Abstract:The lack of oxygen supply in engineered constructs has been an ongoing challenge for tissue engineering and regenerative medicine. Upon implantation of an engineered tissue, spontaneous blood vessel formation does not happen rapidly, therefore, there is typically a limited availability of oxygen in engineered biomaterials. Providing oxygen in large tissue-engineered constructs is a major challenge that hinders the development of clinically relevant engineered tissues. Similarly, maintaining adequate oxygen levels in cell-laden tissue engineered products during transportation and storage is another hurdle. There is an unmet demand for functional scaffolds that could actively produce and deliver oxygen, attainable by incorporating oxygen-generating materials. Recent approaches include encapsulation of oxygen-generating agents such as solid peroxides, liquid peroxides, and fluorinated substances in the scaffolds. Recent approaches to mitigate the adverse effects, as well as achieving a sustained and controlled release of oxygen, are discussed. Importance of oxygen-generating materials in various tissue engineering approaches such as ex vivo tissue engineering, in situ tissue engineering, and bioprinting are highlighted in detail. In addition, the existing challenges, possible solutions, and future strategies that aim to design clinically relevant multifunctional oxygen-generating biomaterials are provided in this review paper.

Abstract:Tissue-engineered bones have therapeutic potential for critical-size bone defects; however, the production of high quantities of the tissue-engineered bones with osteo-induction ability remains a huge challenge. Hyperthermia has been shown to up-regulate the expression of osteogenesis-related proteins to efficiently to promote bone regeneration. In this study, the authors develop a novel photothermal tissueengineered bone (PTEB) with osteo-induction ability and a biomimetic cellular environment. PTEB is generated by seeding rat bone mesenchymal stem cells (rBMMSCs) in the photothermal MoS2-biotin-garose-gelatin scaffold, and then overlaying the scaffold using osteo-induction extracellular matrix (OiECM). The rBMMSCs act as seeding cells, while OiECM provide a biomimetic microenvironment for repairing critical-sized cranial defects in rats. The results show that the PTEB exhibit high biocompatibility and osteo-induction ability under near-infrared (NIR) radiation. Results of in vitro experiments show that PTEB under NIR radiation promote proliferation and osteogenic differentiation of rBMMSCs. Furthermore, the PTEB implantation under NIR radiation significantly induces regeneration of bone in critical-size bone defects in rats 12 weeks after implantation. These findings indicate that PTEB has great potential in regenerative medicine and may represent an effective replacement for autografts used commonly in bone tissue engineering.

Abstract:The failure or delay in healing of critical bone defects is primarily due to early local anoxic conditions and reduced osteogenic activity. In this research, we integrated calcium peroxide (CPO) embedded polycaprolactone (PCL) microspheres and osteoinductive nanoparticles (Hydroxyapatite/Laponite) into a thermosensitive hydrogel (Pluronic F127), thereby formulating an injectable oxygen-releasing osteogenic thermosensitive hydrogel. Notably, the oxygen-releasing microspheres (ORMs) within the composite hydrogel provide stable oxygen release for up to 21 days, ensuring the survival, migration, and bioactivity of both mesenchymal stem cells and endothelial cells under anoxic conditions. Additionally, the composite hydrogel significantly augments the osteogenic potential of bone marrow mesenchymal stem cells by providing a biomimetic microenvironment with the incorporation of nano-hydroxyapatite/laponite. Ultimately, the injectable composite hydrogel successfully stimulated bone regeneration within a cranial defect in a rat model after 8 weeks, with enhanced vascularization and bone quality. The engineered hydrogel provides a minimally invasive approach to stimulate bone regeneration with a sustained oxygen supply and osteogenic microenvironment provision, underlining its potential for treating critical bone defects.

Abstract:An oxygenating hydrogel microsphere (CSP‐GM) integrated with CSP nanoparticles is developed as a multifaceted microenvironment regulator. This innovative system effectively remodels the regenerative microenvironment by synergistically producing oxygen and scavenging ROS, recovering mitochondrial homeostasis and antioxidant defenses of BMSCs, promoting angiogenesis and osteogenesis under hypoxia conditions via precisely modulating the Nrf2/HO‐1 signaling pathways, thereby facilitating bone regeneration. Hypoxia and reactive oxygen species (ROS) overaccumulation cause persistent oxidative stress and impair intrinsic regenerative potential upon tissue injury. For local tissue injury with hypoxia, such as bone fracture and defects, a localized‐sufficient oxygen supply is highly desirable but remains challenging. Therefore, to explore a strategy and its intrinsic mechanism for supplying oxygen locally and remodeling the regenerative microenvironment, an innovative oxygenating hydrogel microsphere system with sustained oxygenation and antioxidant properties is introduced by loading CaO2@SiO2@PDA (CSP) nanoparticles. Specifically, the CSP nanoparticles exhibited broad‐spectrum free radicals scavenging ability, along with prolonged controlled‐release of oxygen once integrated into the gelatin methacrylate anhydride (GelMA) microspheres (CSP‐GM). The CSP‐GM with extra cellular matrix (ECM)‐mimicking structures reconstructed living niches, promoting the adhesion and proliferation of bone marrow stromal cells (BMSCs). As a multifaceted microenvironment regulator, CSP‐GM remodeled the regenerative microenvironment by synergistically producing oxygen and scavenging ROS, recovering mitochondrial homeostasis and antioxidant defenses of BMSCs, promoting angiogenesis and osteogenesis under hypoxia conditions via precisely modulating the Nrf2/HO‐1 signaling pathway. The multiple pro‐regenerative effects of the implantable functionalized micro‐oxygen reservoir on bone repair are further corroborated by the enhanced vascularized bone formation in rat femoral defects, presenting a comprehensive and promising strategy for tissue repair.

Abstract:Engineering scaffolds combinging natural biomineral and artificially synthesized material hold promising potential for bone tissue regeneration. In this study, novel bioactive calcium sulfate/oyster shell (CS/OS) composites were prepared. Comparing to CS scaffold, the CS/OS composites with a controllable degradation rate displayed enhanced mineral nodule formation, higher alkaline phosphate (ALP) activity and increased proliferation rate while treated osteocytes. In CS/OS composites group, elevated mRNA levels of key osteogenic genes including bone morphogenetic protein-2 (BMP-2), runt-related transcription factor 2 (Runx2), osterix (Osx), and osteocalcin (OCN) were observed. Furthermore, The up-regulation of BMP-2 and type I collagen (COL-I) was observed for CS/OS composites relative to a CS group. Scaffolds were implanted into critical-sized femur cavity defects in rabbits to investigate the osteogenic capacity of the composites in vivo. The CS/OS scaffolds with proper suitable times and mechanical strength strongly promoted osteogenic tissue regeneration relative to the regeneration capacity of CS scaffolds, as indicated by the results of histological staining. These results suggest that the OS-modified CS engineering scaffolds with improved mechanical properties and bioactivity would facilitate the development of a new strategy for clinic bone defect regeneration.

Abstract:One of the most hindrance for bone tissue engineering to reconstruct a bone defect is that the insufficient oxygen supply, which has an extremely negative influence upon the survival of seed cells in the scaffold, especially during the initial transplantation period. Herein, we devised a controlled-oxygen-generating composite scaffold that removes this limitation. The microparticles, which are composed of gelatin and CaO2, are hydrolytically activated to generate oxygen when they make contact with water. The hydrogel, self-assembling peptide(SAP), and PLGA membrane were designed to prevent direct exposure of the gelatin-CaO2 microspheres to a large quantity of water and provide a three-dimensional environment for cell growth. The oxygen-generating composite scaffold has the capability of sustainably controlling oxygen generation for about 21 days in vitro and 28 days in vivo. The capacities of this oxygen-generating composite scaffold to improve seed cell survival and the reparation of bone defects were tested using peripheral blood-derived mesenchymal stem cells and a critical-sized cranial defect. Compared with the control groups, survival of seed cells both in vitro and in vivo was significantly improved when the cells were co-cultured or co-transplanted with the oxygen-generating microspheres. Moreover, the presence of the gelatin-CaO2 microspheres significantly improved the repair of bone defects. These results indicated that the oxygen-generating composite scaffold in this study has great potential to increase seed cell survival and enhance bone defect reconstruction.

Axillary surgery: clinical judgment required.

Abstract:The Halsted radical mastectomy included an en bloc resection of axillary contents. Now, more than a century later, there is little controversy about the use of breast-conserving surgical treatments in place of mastectomy, while the routine use of axillary dissection, or at least axillary sampling, has persisted despite the fact that it is the greatest source of morbidity from breast surgery. Is it time to abandon this procedure all together, at least in some patients? Halsted and his successors removed and/or irradiated lymph nodes because they thought that breast cancer always spreads regionally before metastasizing distantly, and that all regional disease had to be removed to improve survival. The National Surgical Adjuvant Breast and Bowel Project (NSABP) demonstrated in trial B04 that this is not the case. 1 One thousand seventy-nine patients without clinical evidence of axillary adenopathy were randomly assigned to simple mastectomy, a radical mastectomy with lymph node dissection, or simple mastectomy with radiation therapy to the chest wall and axilla. Sixty-eight (19%) of the patients randomly assigned to receive no surgery or radiotherapy to the axilla eventually developed positive axillary nodes. This was about half of the rate expected from the incidence of positive nodes in the radical mastectomy arm of the study, and proved that disease left behind can be kept in check or eradicated by the body’s own defense mechanisms. It was possible to remove the axillary recurrence in all but one of the 68 patients, thus removing the argument that axillary recurrences cannot be managed surgically. Most importantly, with 25 years of follow-up, no significant survival differences have emerged. There have been no randomized trials of similar or larger size addressing the role of axillary surgery without axillary radiotherapy subsequent to NSABP B04. In this issue of the Journal of Clinical Oncology, the International Breast Cancer Study Group (IBCSG) 2 reports the result of a trial in which older patients (defined as those 60 years of age) who were to be treated with tamoxifen regardless of nodal status were randomly assigned to receive clearance of the axillary lymph nodes or no surgical intervention in the axilla after either mastectomy or lumpectomy. Among those randomly assigned to axillary clearance, 28% were node-positive. Radiotherapy was not recommended for patients treated with mastectomy; 60% of those treated with breast-conserving therapy (33% of all patients in the study) had radiotherapy. There was significantly more restriction in arm movement and arm pain during the first few months after treatment among those randomly assigned to axillary clearance, but these differences were small and not significant after longer followup, and there were no differences in overall quality of life parameters. There were no differences in disease-free or overall survival, but there were only 473 patients in the study and the median follow-up was just 6.6 years. This study was originally designed to be much larger, but because of poor accrual the study goals were amended to focus on quality-of-life issues. It seems plausible that the poor accrual reflects a lack of uncertainty by the patients and physicians involved in this trial about the value of axillary node dissection in this patient population. Abstract:As we enter the 21st century, the year 2000, or Y2K, what are some of the issues that we, as plastic surgeons, will face? We all know that change is life’s one constant but that change also presents opportunities. We should also recognize that change has positive and negative aspects. As Sir Francis Bacon said: “He who will not apply new remedies must expect new evils.” That said, I want to examine several issues of critical importance, as I see it, to plastic surgery today. First, I want to look at their positive as well as negative aspects. Then I would like to make specific recommendations—courses of action that to me seem to be essential to our future growth and success. Change is not coming; change is here now. And like all Darwin’s creatures, we will have to adapt smartly if we intend to survive. First, I think we all would agree that the loss or diminution of physician autonomy has had a tremendous impact on our lives and our profession. I know that in my case one of the strong incentives for pursuing a career in medicine was that one day I could “hang out my shingle” and direct the course of my professional career. I think that for many of us the dream was to study, learn, and go forth to treat patients. Today, young plastic surgeons, just completing residency training and saddled with educational loans, have little chance of “hanging out their shingles.” There are fewer practice opportunities; start-up costs are enormous and the system of reimbursement changes daily. I began by writing that we had to look at both sides of every issue. One could, in fact, argue eloquently that the lack of physician autonomy has a brighter side than the one I just described. Anyone who has ever run his or her own practice knows well the meaning of the phrase “you don’t own a small business; it owns you.” Giving up what we might call such “indentured autonomy” generally means getting out from under a mountain of tedious detail, allowing you to spend more time with your patients— and losing autonomy can probably be good for your health. But to me and my generation of physicians, the concept of “shift medicine” was looked down upon. Obstetricians would never think of leaving a woman in labor just because their shift was up. Normal schedules were something only for “regular” human beings. Recently we have come to realize that physicians are regular human beings—that they work more effectively if they can get off the treadmill once in a while—that creative insight is often the product of rest and time away from the practice. And, in this sense, shift medicine may actually lead to a higher quality practice of medicine. But turn the coin over again—I do not want someone in my family to be the woman in active labor when the doctor announces, “Shift over, see you tomorrow. Meet Doctor X and the next crew.” Nor would I particularly like to be the patient of a group practice that, to survive economically, must restrict time with each patient to 12 minutes. Abstract:Angiogenesis plays a crucial role in bone regeneration. Hypoxia is a driving force of angiogenesis at the initial stage of tissue repair. The hypoxic microenvironment could activate the hypoxia-inducible factor (HIF)-1α signaling pathway in cells, thereby enhancing the proliferation, migration and pro-angiogenic functions of stem cells. However, long-term chronic hypoxia could inhibit osteogenic differentiation and even lead to apoptosis. Therefore, shutdown of the HIF-1α signaling pathway and providing oxygen at later stage probably facilitate osteogenic differentiation and bone regeneration. Herein, an oxygen tension regulating hydrogel that sequentially activate and deactivate the HIF-1α signaling pathway were prepared in this study. Its effect and mechanism on stem cell differentiation were investigated both in vitro and in vivo. We proposed a gelatin-based hydrogel capable of sequentially delivering a hypoxic inducer (copper ions) and oxygen generator (calcium peroxide). The copper ions released from the hydrogels significantly enhanced cell viability and VEGF secretion of BMSCs via upregulating HIF-1α expression and facilitating its translocation into the nucleus. Additionally, calcium peroxide promoted alkaline phosphatase activity, osteopontin secretion, and calcium deposition through the activation of ERK1/2. Both Cu2+ and calcium peroxide demonstrated osteogenic promotion individually, while their synergistic effect within the hydrogels led to a superior osteogenic effect by potentially activating the HIF-1α and ERK1/2 signaling pathways.

Abstract:Mobilizing endogenous progenitor cells to repair damaged tissue in situ has the potential to revolutionize the field of regenerative medicine, while the early establishment of a vascular network will ensure survival of newly generated tissue. In this study we describe a gene‐activated scaffold containing a stromal derived factor 1α plasmid (pSDF1α), a pro‐angiogenic gene that is also thought to be involved in the recruitment of mesenchymal stromal cells (MSCs) to sites of injury. We show that over‐expression of SDF1α protein enhanced MSC recruitment and induced vessel‐like structure formation by endothelial cells in vitro. When implanted subcutaneously, transcriptomic analysis revealed that endogenous MSCs were recruited and significant angiogenesis was stimulated. Just one‐week after implantation into a calvarial critical‐sized bone defect, pSDF1α‐activated scaffolds had recruited MSCs and rapidly activated angiogenic and osteogenic programs, upregulating Runx2, Dlx5, and Sp7. At the same time‐point, pVEGF‐activated scaffolds had recruited a variety of cell types, activating endochondral ossification. The early response induced by both scaffolds led to complete bridging of the critical‐sized bone defects within 4‐weeks. The versatile cell‐free gene‐activated scaffold described in this study is capable of harnessing and enhancing the body's own regenerative capacity and has immense potential in a myriad of applications.
