Abstract:Antimicrobial wound dressings can aid wound healing by preventing bacterial infection. This is particularly true of electrospun ones, which have a porous structure and can be easily loaded with antimicrobial drugs. Here, Poly lactic acid (PLA), Silk Fibroin (SF) and antimicrobial agents of Silver nanoparticles (Ag NPs) and Silver oxide (Ag2O) to prepare the PLA/SF composites antimicrobial nanofiber membrane by electrospinning. The PLA with 30 % SF nanofiber membrane show the water vapor permeability (WVP) and the liquid absorption of 36 g·mm/(m2·d·kPa) and 1721 %. With the increasing of SF contents, the degradation rate and surface hydrophilicity of the nanofiber membrane increase significantly. The nanofiber membrane exhibited excellent antimicrobial activity against Pseudomonas aeruginosa (P. aeruginosa) with the inhibition circle reach at 18.2 mm. The resultant nanofiber membrane showed high cytosolic activity, good cytocompatibility and strong antimicrobial ability, which laid a theoretical foundation for the construction of a new PLA/SF composites antimicrobial fiber membrane.

Abstract:Antibacterial wound dressing, an effective barrier between the wound and the foreign environment, has the ability to inhibit and even kill the bacteria around the wound so that it can accelerate the wound healing. However, the loaded antibacterial agents of antibiotics and metal ions on wound dressings caused the new problems of drug resistance and toxicity. To develop a novel wound dressing with the natural antibacterial property agent, here, natural Manuka honey (MH), silk fibroin (SF), and polycaprolactone (PCL) were mixed to prepare nanofibrous membranes by electrospinning. The SEM observation showed the good microscopic morphology of SF/PCL/MH nanofibrous membranes that the fibers were even and smooth. With the addition of MH, the membranes gained the accepted stress and the excellent flexibility to adapt to wound contour. In addition, their hydrophilic property was beneficial to cell adhesion and proliferation. Most importantly, SF/PCL/MH nanofibrous membranes exhibited excellent sustained release effect and antibacterial properties. The antibacterial activity against Escherichia coli, Staphylococcus aureus, and Candida albicans reached 99.88%, 99.70%, and 98.95%, respectively when 75% natural antimicrobial agent MH was loaded. Moreover, SF/PCL/MH membranes showed good cytocompatibility. These results indicated the fabricated SF/PCL/MH nanofibrous membranes are a kind of novel dressing with good antimicrobial property.

Abstract:Many complex diseases, such as bacterial infections, frequently accompany cutaneous wound healing, adding to the difficulty of clinical wound management. Consequently, in addition to displaying strong biocompatibility and actively promoting wound healing, an optimal wound dressing should also possess antimicrobial qualities to address issues with bacterial infection. This paper developed natural silk fiber (SF) membranes (also known as a flat silk cocoon (FSC)) with antimicrobial properties as a dressing for skin wounds. By changing the spinning tools and environment of silkworm larvae, a novel natural SF membrane with a cocoon structure and controllable size was prepared. The functional SF membranes were obtained via a hot press process and grafted with ε-Poly-l-lysine (EPL). The results showed that the SF membrane dressing was adjustable in size with a similar structure to the extracellular matrix (ECM), displaying inherent mechanical properties, excellent antimicrobial qualities, and biocompatibility. In vivo experiments using a full-thickness skin defect model indicated that EPL-modified SF membranes significantly promoted the rate of wound healing, exhibiting thicker granulation tissue and higher collagen disposition than commercial dressings (TegadermTM film). Therefore, the excellent mechanical qualities and cytocompatibility of the antimicrobial EPL-modified SF membranes substantially promote their potential application as a chronic wound dressing.

Abstract:Wound dressing is commonly used for skin injuries. The design of wound dressing typically stems from the principles of open-wound management such as infection prevention, moisture balance and healing response. A new wound dressing comprising polyvinylpyrrolidone (PVP)–berberine hydrochloride (BHC)/PVP–cellulose acetate (CA)–BHC/CA-aloin tri-layer Janus fiber was successfully fabricated using trifluid side-by-side electrospinning for antibacterial and wound healing functions. The fibrous membrane can hold 5.090 ± 0.276 times of its own water mass and achieve 6 h of microenvironment moisture retention. In vitro antibacterial results show that BHC exhibited selective resistance to S. aureus . The nanofibers were noncytotoxic and showed good cell adhesion properties and enhanced cell proliferation effect compared with BHC or aloin only tri-layer nanofibers. Owing to the specially designed tri-layer Janus structure, the in vitro drug-release profile of drugs-loaded fibrous membrane showed a combination of two-stage release for BHC (76.093 % ± 1.813 % release in first hour and 98.526 % ± 0.604 % release in 6 h) and sustained release for aloin (88.376 % ± 5.282 % release in 6 h). Therefore, the tri-layer Janus structure can be a potential choice for wound dressing application.

Abstract:A sustained release of anti-infection drugs is beneficial for the reduction of required dressing change times and the protection of the granulation tissue against further infection. In this work, the strategy of encapsulating mesoporous SBA-15 particles in a permeable cellulose membrane (CM) was used in the preparation of antibacterial cellulose based composite membranes to depot store and deliver drugs with prolonged anti-microbial drug release characteristics. It was found that SBA-15 with the protection of phosphate buffered saline prefilled in SBA-15 mesoporous channel could resist strong alkali environment during composite membranes preparation and its mesoporous structure remained intact, which was verified by SEM, TEM and BET results. The resultant cellulose based composite membrane containing 30 wt% SBA-15 (denoted as P-CM-SBA (30%)) had achieved 3.6 wt% drug loading and demonstrated the sustained release of chloramphenicol for 250 h, which was resulted from SBA-15/cellulose structure-inducing two-stage release behavior. Strong antibacterial activity of P-CM-SBA (30%) against S. aureus and E. coli. could last 144 h. In addition, the tensile strength, water vapor transmission rate and swelling properties of P-CM-SBA (30%) conformed to the primary requirements of wound dressing materials. Abstract:Chitosan, a naturally occurring polysaccharide with abundant resources, has been extensively exploited for various biomedical applications, typically as wound dressings owing to its unique biocompatibility, good biodegradability and excellent antibacterial properties. In this work, composite nanofibrous membranes of chitosan (CS) and silk fibroin (SF) were successfully fabricated by electrospinning. The morphology of electrospun blend nanofibers was observed by scanning electron microscopy (SEM) and the fiber diameters decreased with the increasing percentage of chitosan. Further, the mechanical test illustrated that the addition of silk fibroin enhanced the mechanical properties of CS/SF nanofibers. The antibacterial activities against Escherichia coli (Gram negative) and Staphylococcus aureus (Gram positive) were evaluated by the turbidity measurement method; and results suggest that the antibacterial effect of composite nanofibers varied on the type of bacteria. Furthermore, the biocompatibility of murine fibroblast on as-prepared nanofibrous membranes was investigated by hematoxylin and eosin (H&E) staining and MTT assays in vitro, and the membranes were found to promote the cell attachment and proliferation. These results suggest that as-prepared chitosan/silk fibroin (CS/SF) composite nanofibrous membranes could be a promising candidate for wound healing applications.

Abstract:Aims: In models of diabetes, distal nephron cells contribute to glucose uptake and oxidation. How these cells contribute to the use of glucose for the regulation of H+ extrusion remains unknown. We used Madin-Darby Canine Kidney (MDCK) cells to investigate the effect of acute or chronic high glucose concentration on the abundance and activity of the Na+/H+ exchanger (NHE-1). Methods: Using RT-PCR, we also evaluated the mRNA expression for sodium glucose co-transporters SGLT1 and SGLT2. Protein abundance was analyzed using immunoblotting, and intracellular pH (pHi) recovery was evaluated using microscopy in conjunction with the fluorescent probe BCECF/AM. The Na+-dependent pHi recovery rate was monitored with HOE-694 (50 µM) and/or S3226 (10 µM), specific NHE-1 and NHE-3 inhibitors. Results: MDCK cells did not express the mRNA for SGLT1 or SGLT2 but did express the GLUT2, NHE-1 and NHE-3 proteins. Under control conditions, we observed a greater contribution of NHE-1 to pHi recovery relative to the other H+ transporters. Acute high glucose treatment increased the HOE-694-sensitive pHi recovery rate and p-Erk1/2 and p90RSK abundance. These parameters were reduced by PD-98059, a Mek inhibitor (1 µM). Chronic high glucose treatment also increased the HOE-694-sensitive pHi recovery rate and p-p38MAPK abundance. Both parameters were reduced by SB-203580, a p38MAPK inhibitor (10 µM). Conclusion: These results suggested that extracellular high glucose stimulated NHE-1 acutely and chronically through Mek/Erk1/2/p90RSK and p38MAPK pathways, respectively.

Abstract:The application prospects of composite sponges with antibacterial and drug-carrying functions in the field of medical tissue engineering are extensive. A solution of cassava silk fibroin (CSF) was prepared with Ca(NO3)2 as a solvent, which was then combined with chitosan (CS) to create a sponge-porous material by freeze-drying. The CSF-CS composite sponge with a mesh structure was successfully fabricated through hydrogen bonding. Scanning electron microscopy (SEM), Fourier transform infrared absorption (FTIR) and X-ray diffraction (XRD) were employed to investigate the appearance and structure of the cassava silk's fibroin materials, specifically examining the impact of different mass percentages of CS on the sponge's structure. The swelling rate and mechanical properties of the CSF-CS sponge were analyzed, along with its antibacterial properties. Furthermore, by incorporating ibuprofen as a model drug into these loaded sponges, their potential efficacy as efficient drug delivery systems was demonstrated. The results indicate that the CSF-CS sponge possesses a three-dimensional porous structure with over 70% porosity and an expansion rate exceeding 400% while also exhibiting good resistance against pressure. Moreover, it exhibits excellent drug-carrying ability and exerts significant bacteriostatic effects on Escherichia coli. Overall, these findings support considering the CSF-CS composite sponge as a viable candidate for use in drug delivery systems or wound dressings.

Multifunctional electrospun membranes with hydrophilic and hydrophobic gradients property for wound dressing

Abstract:Achieving sustained and stable release of macromolecular antibacterial agents and unidirectional transport of liquids in targeted environment is still a challenge to be addressed in the management of wounds with large amounts of tissue exudates. In this work, a multilayer electrospun membrane (ethylcellulose-ethylcellulose/gelatin-quercetin/Eudragit L-100/polyethylene glycol, EC-EC/Gel-Q/EL/PEG) was designed with hydrophobic-hydrophilic gradients and drug sustained-release properties controlled by self-pumping effect and prepared using sequential electrospinning technology. The capillary force of different layers in the multilayer membrane could be controlled by precisely tuning the polymer concentrations of the inner and middle layers to extract water directly from hydrophobic inner ethylcellulose (EC) layer to hydrophilic middle ethylcellulose/gelatin (EC/Gel) layer. The droplets could not penetrate the hydrophobic side, but the drug molecules in the outer layer quercetin-loaded Eudragit L-100 (Q/EL/PEG) membrane moved after absorbing a large amount of water. The drug release behavior of multilayer wound dressing mainly followed the Korsmeyer-Peppas model. This multifunctional electrospun membrane could rapidly drive the biofluid outflow, effectively block the invasion of external contaminants and continuously release anti-inflammatory drugs, without any obvious cytotoxicity to mouse fibroblast cells. Hence, the above results indicate the excellent therapeutic potential of the proposed biomaterial as a wound dressing for diabetic patients.
Development of a tobramycin-loaded calcium alginate microsphere/chitosan composite sponge with antibacterial effects as a wound dressing

Abstract:Traditional dressings exhibit several disadvantages, as they frequently lead to bacterial infections, cause severe tissue adhesion and perform a relatively single function. Therefore, in this study, a composite sponge dressing with antibacterial properties and excellent physicochemical properties was developed. Six groups of tobramycin-loaded calcium alginate microspheres were prepared by changing the amount of tobramycin added, and the optimal group was selected. Then, seven groups of tobramycin-loaded calcium alginate microsphere/chitosan composite sponges were fabricated via a solvent blending process and a freeze-drying method. The surface morphology, physicochemical properties,in vitrodegradation properties,in vitrodrug release properties, antibacterial properties and cytotoxicity of the composite sponges were examined. Group 3.0 contained the best microspheres with the largest drug loading capacity, good swelling performance and cumulative drug release rate, obvious and sustained antibacterial activity, and good cytocompatibility. The tobramycin-loaded calcium alginate microsphere/chitosan composite sponges exhibited three-dimensional porous structures, and their porosity, swelling rate, water absorption and water retention rates and water vapor transmission rate met the standards needed for an ideal dressing. The comprehensive performance of the sponge was best when 20 mg of drug-loaded microspheres was added (i.e. group 20). The cumulative drug release rate of the sponge was 29.67 ± 4.14% at 7 d, the diameters of the inhibition zones against the three bacteria were greater than 15 mm, and L929 cell proliferation was promoted. These results demonstrated that the tobramycin-loaded calcium alginate microsphere/chitosan composite sponge with 20 mg of tobramycin-loaded microspheres shows promise as a dressing for infected wounds.

Abstract:Optimal wound healing requires a wet microenvironment without over-hydration. Inspired by capillarity and transpiration, we have developed a sandwich-like fibers/sponge dressing with continuous exudate drainage to maintain appropriate wound moisture. This dressing is prepared by integrating a three-layer structure using the freeze-drying method. Layer I, as the side that contacts with the skin directly, consists of a hydrophobic silk fibroin membrane; Layer II, providing the pumping action, is made of superabsorbent chitosan-konjac glucomannan sponge; Layer III, accelerating evaporation sixfold compared to natural evaporation, is constructed with a graphene oxide soaked hydrophilic cellulose acetate membrane. Animal experiments showed that the composite dressing had superior wound-healing characteristics, with wounds decreasing to 24.8% of their original size compared to 28.5% for the commercial dressing and 43.2% for the control. The enhanced wound healing can be ascribed to the hierarchical porous structure serves as the fluid-driving factor in this effort; the hydrophilicity of a membrane composed of silk fibroin nanofibers is adjustable to regulate fluid-transporting capacity; and the photothermal effect of graphene oxide guarantees exudates that have migrated to the top layer to evaporate continuously. These findings indicate the unidirectional wicking dressing has the potential to become the next generation of clinical dressings.

Abstract:Nanomaterial-based wound dressings have been extensively studied for the treatment of both minor and life-threatening tissue injuries. These wound dressings must possess several crucial characteristics, such as tissue compatibility, non-toxicity, appropriate biodegradability to facilitate wound healing, effective antibacterial activity to prevent infection, and adequate physical and mechanical strength to withstand repetitive dynamic forces that could potentially disrupt the healing process. Nevertheless, the development of nanostructured wound dressings that incorporate various functional micro- and nanomaterials in distinct architectures, each serving specific purposes, presents significant challenges. In this study, we successfully developed a novel multifunctional wound dressing based on poly(lactic acid) (PLA) fibrous membranes produced by solution-blow spinning (SBS) and electrospinning. The PLA-based membranes underwent surface modifications aimed at tailoring their properties for utilization as effective wound dressing platforms. Initially, beta-chitin whiskers were deposited onto the membrane surface through filtration, imparting hydrophilic character. Afterward, silver nanoparticles (AgNPs) were incorporated onto the beta-chitin layer using a spray deposition method, resulting in platforms with antimicrobial properties against both Staphylococcus aureus and Escherichia coli. Cytotoxicity studies demonstrated the biocompatibility of the membranes with the neonatal human dermal fibroblast (HDFn) cell line. Moreover, bilayer membranes exhibited a high surface area and porosity (> 80%), remarkable stability in aqueous media, and favorable mechanical properties, making them promising candidates for application as multifunctional wound dressings.

Abstract:Silk fibroin (SF) and alginate (ALG), as naturally-based polymers, are ideal for wound dressings due to their non-allergenicity, low toxicity, biocompatibility, biodegradability features, and ability to promote wound healing. Researches revealed that exosomes secreted by human bone marrow mesenchymal stem cells (HBM-MCS-Exo) demonstrate a paracrine effect on tissue repair procedure. In this research, a wound dressing based on SF nanofibers and ALG-HBM-MCS-Exo hydrogel has been designed. According to Dynamic Light Scattering (DLS) and Western blot analysis, the exosomes were 24.8 nm in diameter and expressed HMCS-Exo biomarkers. The scanning electron microscopy (SEM) results indicated that the entrapment of the ALG hydrogel in SF nanofibers with a diameter of 168 ± 19 nm resulted in an increase in the diameter of the fibers. The produced constructs were found to have reasonable water vapor transfer rates (WVTR) that prevent being infected due to seepage generation between skin and dressing. This feature, accompanied by the good swelling properties, makes them suitable for dressing wounds. Also, evaluation of cell viability revealed sufficient proliferation of human foreskin fibroblast cells and non-toxicity of designed wound dressings.

Abstract:The sustained release of antimicrobial therapeutics for wound dressing has become an attractive design strategy for prolonging the timespan of wound dressings and for reducing the risk of chronic wound infection. Recently, cellulose-based membrane has become a preferred option of wound dressings for the treatment of burn wounds and skin ulcers. In this work, novel cellulose membrane incorporated with mesoporous silica particles (SBA-15) was developed as an antimicrobial wound dressing with desirable sustained release functionality for targeting persistent bacterial pathogens. Attributed to a coated layer of calcium carbonate (CaCO3), SBA-15 particles were free from corrosion in alkaline condition during the preparation of cellulose-based composite membranes. SEM, TEM and BET results showed that the morphology, specific surface area, pore size and pore volume of pristine SBA-15 were preserved after the incorporation of CaCO3-coated SBA-15 into the cellulose matrix, while the mesoporous structure of SBA-15 was significantly disrupted without the use of CaCO3 coating. The resultant composite membranes containing 30 wt% SBA-15 (denoted as CM-Ca2-SBA(30%)) achieved 3.6 wt% of antimicrobial drug loading. Interestingly, CM-Ca2-SBA(30%) demonstrated the sustained release property of chloramphenicol for 270 h, driven by a two-stage drug release processes of SBA-15/cellulose. The water vapor permeability (WVTR) and swelling properties of composite membranes were shown to have complied with the primary requirements of wound dressing. Antibacterial assays revealed that strong antibacterial activities (144 h) of the composite membranes against Staphylococcus aureus and Eschericia coli were achieved. All results displayed that the strategy of coating silica with CaCO3 helps to obtain cellulose–silica composite membranes with desirable sustained release profiles and strong antibacterial activities. The antibacterial SBA-15/cellulose composite membranes show potential for the application of wound dressing.

Abstract:Hydrogel has emerged as one of the most promising dressings for wound healing owing to the creation of moist environment. However, it suffers from poor water retention capability and weak mechanical property. Therefore, development of dressings with durable moisture retention, robust mechanical property, and self-healing performance is urgently needed in practical applications. Herein, we developed a bilayered composite dressing consisting of breathable and hydrophobic polyurethane/polydimethylsiloxane (PU/PDMS) nanofibrous membrane and double-crosslinked chitosan-based hydrogel with dynamic Schiff base bonds and non-dynamic photo-crosslinking bonds. Combined with the nanofibrous membrane as protection and mechanical support, the bilayered hydrogel composites presented enhanced water retention capability to provide sustained moist environment, excellent stretchable and compressive performance to bear deformation, fast self-healing and good cyclic resilience to endure frequent movements. The in vivo study indicated their effective therapeutic effect in a rat model with full-thickness skin wounds. This study demonstrated that the hydrogel/nanofibrous membrane composite represented a promising candidate as dressing material for wound healing.

Abstract:The purpose of this study is to prepare polysaccharide-based hydrogel dressing integrated with microspheres which has excellent self-healing ability and sustained antibacterial function for wound healing application. The hydrogel is derived from carboxymethyl chitosan (CMCS) and carboxymethyl cellulose (CMC) via the Schiff’s base reaction, without the addition of chemical catalyst. Also, the CMCS microspheres containing an antibacterial drug, e.g., tetracycline hydrochloride (TH), were prepared by an emulsion cross-linking. Parameters were optimized during the preparation experiment to improve various properties of the microspheres/hydrogel composite dressing. This gel dressing has good elasticity, when compressive strain was 0.7 mm/mm, the tangent elastic modulus was up to 1.7 MPa. Furthermore, its drug sustained release performance was significantly better than the drug directly embedded in the hydrogel. TH could sustain release stably within 12 h, without obvious burst release phenomenon. Antibacterial experiments showed that 20 mg/mL microspheres made composite dressing a better performance than control microspheres and hydrogel. Potential of the composite scaffolds was further demonstrated by culture of human adipose-derived stem cells (ASCs) in vitro. Cell culture showed that this composite hydrogel could support survival and proliferation of ASCs. These characteristics provide a potential opportunity to use this composite gel dressing in drug delivery and wound healing applications.

Abstract:Nucleation of microtubules by eukaryotic microtubule organizing centers (MTOCs) is required for a variety of functions, including chromosome segregation during mitosis and meiosis, cytokinesis, fertilization, cellular morphogenesis, cell motility, and intracellular trafficking. Analysis of MTOCs from different organisms shows that the structure of these organelles is widely varied even though they all share the function of microtubule nucleation. Despite their morphological diversity, many components and regulators of MTOCs, as well as principles in their assembly, seem to be conserved. This review focuses on one of the best-characterized MTOCs, the budding yeast spindle pole body (SPB). We review what is known about its structure, protein composition, duplication, regulation, and functions. In addition, we discuss how studies of the yeast SPB have aided investigation of other MTOCs, most notably the centrosome of animal cells.
Poly (lactic acid) electrospun nanofiber membranes: Advanced characterization for biomedical applications with drug loading performance studies

Abstract:Traditional dressings have shortcomings such as poor moisture absorption and easy to adhere, making the development of new dressings crucial. In this work, a PLA/PVP crosslinked drug-loaded nanofiber membrane was prepared through electrospinning and ultraviolet crosslinking, with poly (lactic acid) (PLA), polyvinylpyrrolidone (PVP), and salicylic acid (SA) as starting materials. The results demonstrated that the inclusion of PVP notably boosted the viscosity and conductivity of the blend spinning solution. The roughness of the fabricated fiber was elevated, and the diameter of the fibers was more uniform. Additionally, the incorporation of PVP not only enhanced the porosity of the fiber membrane but also effectively decreased its contact angle. Notably, when the PVP content reached 40 %, the contact angle underwent a substantial reduction, decreasing significantly from 125.4° to 82.2°. The SA drug-loaded fiber membrane exhibited a notable bacteriostatic effect against Escherichia coli and Staphylococcus aureus, with its release behavior adhering to Fick's diffusion law. In the cell viability experiment, the cell proliferation rate increased from 94 % to 129 % after 3 days. This shows that the prepared membrane has good antibacterial effect and cell compatibility, which provides a theoretical basis for the construction of a new medical dressing.
Preparation and characterization of unidirectional water-transported bilayer PLA/ZnO-PAN/SPA nanofibrous membrane for wound healing

Abstract:Wound infection caused by excessive exudate leads to overhydration of wounds and prevents wound healing. Thus, there is a need for a wound dressing to be developed that can unidirectionally export excess biological fluid while inhibiting bacterial infection. Herein, a nanofibrous bilayer membrane with a unidirectional water transport function was fabricated by electrospinning as a novel wound dressing, which can export excess biological fluid efficiently and further inhibit bacterial infection. The inner layer of the bilayer nanofiber membrane is polylactic acid (PLA)-zinc oxide (ZnO) nanofibers, which are hydrophobic and antibacterial. The outer layer is polyacrylonitrile (PAN)-sodium polyacrylate (SPA) nanofibers, which are hydrophilic and liquid-absorbent. The bilayer nanofibrous membrane's morphology, mechanics, thermal stability, wettability , water absorption, gas permeability, and unidirectional water transport ability were characterized and evaluated. The study showed that the nanofibrous bilayer membrane exhibited excellent physicochemical properties and biological performance, and its mechanical properties can reach 4.6 MPa. In addition, when the thickness of the hydrophobic layer is 3 μm and the thickness of the hydrophilic layer is 44 μm, the double-layer fiber membrane can effectively export the excess biological fluid in 5.09 s and prevent the wound from wetting again. The nanofibrous bilayer membrane with excellent water absorption, air permeability, thermal stability, and mechanical strength provides a feasible solution for chronic wound healing.
