Abstract:Accumulations of high molecular weight glutenin subunits(HMW-GS) and glutenin macropolymer(GMP) play key roles in grain quality of wheat(Triticum aestivum L.).The aims of this study were to analysis dynamic changes in contents of each high molecular weight glutenin subunit and GMP,and evaluate effects of nitrogen rates on contents of HMW-GS and GMP in grains of two wheat cultivars,Xuzhou 26 and Ningmai 9,which contained similar type of HMW-GS while differed in protein content in grain.HMW-GS were firstly separated using the sodium dodecyl sulfate polyacrylamide gel electrophoresis(SDS-PAGE),and each of the HMW-GS in the gel was cut and extracted,then measured quantitatively by colorimetric analysis.Five nitrogen rates of 0,75,150,225 and 300 kg·ha~(-1) were established for each cultivar.The results showed that all HMW-GS in high grain protein content cultivar Xuzhou 26 were formed in grain at 14 days after anthesis(DAA),while those of low grain protein content cultivar Ningmai 9 at 21 DAA.Nitrogen application showed no impact on the early formation of HMW-GS in grain of both cultivars.Grain HMW-GS accumulation rate was increased and the rapid accumulation period prolonged with the increasing nitrogen rate in Xuzhou 26,while HMW-GS accumulation rate during late grain filling period was enhanced in Ningmai 9.Thereafter,the contents of total HMW-GS and GMP increased with the nitrogen rate in grain of Xuzhou 26.In Ningmai 9,however,the contents of total HMW-GS and GMP increased within the nitrogen rates from 0 to 225 kg·ha~(-1),while decreased when excessive nitrogen of 300 kg·ha~(-1) was applied.Under the same nitrogen rate,each content of HMW-GS in grain of Xuzhou 26 was higher than that of Ningmai 9,indicating the key role of HMW-GS in wheat quality.In conclusion,the higher HMW-GS content is related to the earlier starting time of HMW-GS synthesis,and moderate nitrogen application favors synthesis of HMW-GS in wheat grain.
Effect of Nitrogen Application Rate on Content of Glutenin Macropolymer and High Molecular Weight Glutenin Subunits in Grains of Two Winter Wheat Cultivars

Abstract:Two winter wheat (Triticum aestivum L.) cultivars differing in grain protein content were selected to study the effect of N application rate on changes in contents of glutenin macropolymer (GMP) and high molecular weight glutenin subunits (HMW-GS) during grain filling. Contents of GMP and HMW-GS were much higher in the high GPC cultivar, Xuzhou 26, than those in low GPC cultivar, Ningmai 9. N increased contents of GMP and HMW-GS in Xuzhou 26 with N rate between 0 and 300kgha−1, while at the very high N rate of 300kgha−1 the contents of GMP and HMW-GS in Ningmai 9 decreased. The high contents of GMP and HMW-GS at maturity were closely related to the rapid increase in contents of GMP and HMW-GS during the initial period of their synthesis. HMW-GS and GMP content were closely correlated. The total HMW-GS content was important in determining GMP content than the content of any HMW-GS pair or any individual HMW-GS present in the selected cultivars. The pattern of response of GMP content to N application rate was closely related to the regulatory effect of N on HMW-GS synthesis.

Abstract:Wheat processing quality has close relationships with high molecular weight glutenin subunits(HMW-GS) and glu-tenin macropolyer(GMP) in grains.Nitrogen fertilization scheme could affect the accumulation of HMW-GS and the size distri-bution of GMP.In this study,we used a strong gluten winter wheat cultivar,Jinan 17,and a weak gluten winter wheat cultivar,Lumai 21,to investigate the effects of nitrogen topdressing stage on the accumulation of HMW-GS and the size distribution of GMP in grains.The nitrogen fertilizer rate during the whole growth period was 225 kg ha?1,of which a half was applied as base fertilizer and the other half was topdressed at pseudo stem erection(SE,GS 30),jointing(JT,GS 32),or booting(BT,GS 41) stage.The spike samples were collected from 7 d after anthesis(DAA) to maturity with a 7-day interval.The HMW-GS in grains formed before 14 DAA,and Jinan 17 had higher contents of HMW-GS and GMP than Lumai 21,which indicated that more glu-tenin is accumulated in the strong gluten cultivar than in the weak gluten cultivar.At maturity,the contents of HMW-GS and GMP in Jinan 17 showed decreasing trends with the delay of topdressing practice,and the highest values were observed in the SE treatment.In Lumai 21,compared to the SE and BT treatments,the JT treatment significantly accelerated the HMW-GS accumu-lation at middle-late stage of grain filling,and the rapid accumulation period was prolonged.At middle-late stage of grain filling,the accumulation rates of x-type subunits(1,4,5,and 7) were increased significantly in the SE treatment of Jinan 17 and the JT treatment of Lumai 21,whereas the y-type subunits were seldom affected by nitrogen application timing.For both cultivars,delay of nitrogen topdressing enhanced the percentages of volume and surface of GMP particle with diameter less than 12 μm,but re-duced those of GMP particle with diameter larger than 100 μm.The percentage of GMP particle number with diameter larger than 12 μm was increased in Jinan 17 with the delay of nitrogen topdressing but decreased in Lumai 21.The percentage of large GMP particle volume was higher in Jinan 17(containing subunit pair 4+12) than in Lumai 21(containing subunit pair 5+10).This result suggested that polymerization of subunits and GMP particles are related to not only the subunit type but also the subunit content per unit of flour.

Abstract:Wheat gluten properties can be improved by the application of nitrogen. This study investigates the effects of nitrogen application in the booting stage on glutenin polymerization during grain-filling and structural–thermal properties of gluten based on the high-molecular-weight glutenin subunits (HMW-GSs) using near-isogenic lines (Glu-1Da and Glu-1Dd). The nitrogen rate experiment included rates of 0, 60, 90, and 120 kg N ha−1 applied with three replicates. Nitrogen significantly improved the grain quality traits (wet gluten contents, Zeleny sedimentation values, and maximum resistance) and dough strength (dough development time, dough stability time, and protein weakening), especially in wheat with the Glu-1Da allele. Nitrogen increased the protein composition contents, proportions of glutenins and HMW-GSs, and disulfide bond concentration in the flours of Glu-1Da and Glu-1Dd, and accelerated the polymerization of glutenins (appearing as glutenin macropolymer) during grain-filling, where nitrogen enhanced the accumulation and polymerization of glutenins more for line containing Glu-1Da than Glu-1Dd. The β-sheets, α-helix/β-sheet ratio, microstructures, and thermal stability were also improved to a greater degree by nitrogen for gluten with Glu-1Da compared to Glu-1Dd. Nitrogen treatment was highly effective at improving the gluten structural‒thermal properties of wheat in the booting stage, especially with inferior glutenin subunits.

Abstract:The high-molecular-weight glutenin subunits (HMW-GS) at Glu-D1 have a larger contribution to the processing property of wheat flours than those at the Glu-A1 and Glu-B1 loci and are always expressed in nature. Wheat lines missing one subunit at Glu-D1, either x or y, attract great interest for their potential quality value. Major quality parameters including SDS-sedimentation value, gluten-related indices, and other dough strength parameters such as development time, stability time, degree of softening, and farinograph quality numbers were tested in wheat lines solely expressing HMW-GS 1Dy12 from different backgrounds and compared with a weak gluten wheat control CN 16 to elucidate their quality potential. Wheat line solely expressing HMW-GS 1Dy12 derived from different genetic backgrounds showed similar but low SDS-sedimentation values (∼10 mL) and low wet gluten (∼20–25 g) and dry gluten contents (∼10 g). Additionally, wheat line solely expressing HMW-GS 1Dy 12 also showed a lower development time (<1 min), stability time (<1 min), and farinograph quality number (∼5–15), and a higher degree of softening (∼170–301), than the control CN 16. Wheat line solely expressing HMW-GS 1Dy 12 had a weak dough strength and may be used as a potential material for producing weak-gluten foods.

Abstract:A set of high-molecular-weight glutenin subunit (HMW-GS) deletion lines were used to investigate the influences of HMW-GS on wheat gluten, and dough properties were investigated using a set of HMW-GS deletion lines. Results showed that HMW-GS deletion significantly decreased the dough stability time, as well as viscoelastic moduli (G' and G″), compared with the wild type, where the deletion of x-type HMW-GSs (Ax1d, Bx7d, and Dy12d) decreased more than y-type HMW-GSs (By8d and Dy12d). The deletion of HMW-GS significantly decreased HMW-GS contents and increased α-/γ-gliadin contents. A proteomic study showed that the HMW-GS deletion down-regulated the HMW-GS, β-amylase, serpins, and protein disulfide isomerase and up-regulated the LMW-GS, α/γ-gliadin, and α-amylase inhibitor. Meanwhile, HMW-GS deletion significantly decreased contents of β-turn and β-sheet. In addition, less energetically stable disulfide conformations (trans-gauche-gauche and trans-gauche-trans) were abundant in HMW-GS deletion lines. Furthermore, analysis of five HMW-GSs based on amino acid sequences proved that Dx2 and Bx7 had a more stable structure, followed by Ax1, then Dy12, and finally By8.
Deletion of high-molecular-weight glutenin subunits in wheat significantly reduced dough strength and bread-baking quality

Abstract:Background: High-molecular-weight glutenin subunits (HMW-GS) play important roles in the elasticity of dough made from wheat. The HMW-GS null line is useful for studying the contribution of HMW-GS to the end-use quality of wheat. Methods: In a previous work, we cloned the Glu-1Ebx gene from Thinopyrum bessarabicum and introduced it into the wheat cultivar, Bobwhite. In addition to lines expressing the Glu-1Ebx gene, we also obtained a transgenic line (LH-11) with all the HMW-GS genes silenced. The HMW-GS deletion was stably inherited as a dominant and conformed to Mendel's laws. Expression levels of HMW-GS were determined by RT-PCR and epigenetic changes in methylation patterns and small RNAs were analyzed. Glutenins and gliadins were separated and quantitated by reversed-phase ultra-performance liquid chromatography. Measurement of glutenin macropolymer, and analysis of agronomic traits and end-use quality were also performed. Results: DNA methylation and the presence of small double-stranded RNA may be the causes of post-transcriptional gene silencing in LH-11. The accumulation rate and final content of glutenin macropolymer (GMP) in LH-11 were significantly lower than in wild-type (WT) Bobwhite. The total protein content was not significantly affected as the total gliadin content increased in LH-11 compared to WT. Deletion of HMW-GS also changed the content of different gliadin fractions. The ratio of ω-gliadin increased, whereas α/β- and γ-gliadins declined in LH-11. The wet gluten content, sedimentation value, development time and stability time of LH-11 were remarkably lower than that of Bobwhite. Bread cannot be made using the flour of LH-11. Conclusions: Post-transcriptional gene silencing through epigenetic changes and RNA inhibition appear to be the causes for the gene expression deficiency in the transgenic line LH-11. The silencing of HMW-GW in LH-11 significantly reduced the dough properties, GMP content, wet gluten content, sedimentation value, development time and stability time of flour made from this wheat cultivar. The HMW-GS null line may provide a potential material for biscuit-making because of its low dough strength.
Effects of Low-Molecular-Weight Glutenin Subunit Encoded by Glu-A3 on Gluten and Chinese Fresh Noodle Quality

Abstract:Low-molecular-weight glutenin subunits (LMW-GS) account for 40% of the total wheat grain gluten protein fraction, which plays a significant role in the formation of noodle processing quality. The goal of this study was to clarify the effects of the major LMW-GS encoded by Glu-A3 on gluten and Chinese fresh noodle (CFN) quality. Four near-isogenic lines (NILs) were used as materials in this study, respectively carrying alleles Glu-A3a, Glu-A3b, Glu-A3c, and Glu-A3e, against the background of wheat variety Xiaoyan 22. The grain protein and its component contents and the gluten content, gluten index, farinograph properties, cooking quality, and textural quality of CFN were investigated. The results show that the ratios of glutenin to gliadin (Glu/Gli) in the NILs ranked them as Glu-A3b > Glu-A3c/Glu-A3a > Glu-A3e, and the unextractable polymeric protein content (UPP%), gluten index (GI), and farinograph quality in the NILs ranked them as Glu-A3b > Glu-A3c > Glu-A3a/Glu-A3e. Compared to Glu-A3b and Glu-A3a, the NILs carrying alleles Glu-A3c and Glu-A3e had better cooking and texture properties in CFN. All these findings suggest that the introduction of alleles Glu-A3c or Glu-A3e is an efficient method for quality improvement in CFN, which provides an excellent subunit selection for improving CFN quality.

Abstract:High molecular weight glutenin subunit(HMW-GS) and glutenin macropolymer(GMP) play key roles in determining grain quality in wheat(Triticum aestivum L.).The aim of this study was to evaluate effects of drought and waterlogging post anthesis on HMW-GS and GMP contents in grains of two wheat cultivars,Yumai 34(strong gluten) and Yumai 9(weak gluten).According to our previously reported quantification protocol,each HMW-GS content was calorimetrically determined,following the sodium dodecyl sulfate polyacrylamide gel electrophoresis(SDS-PAGE).The treatments of waterlogging,drought,and moderate water supply(CK)were used from anthesis to maturity.The results showed that HMW-GS of both cultivars under drought and those of Yumai 34 under waterlogging appeared in grains at about 10 days post anthesis(DPA),while those of Yangmai 9 under waterlogging and of both cultivars under moderate water supply were firstly found at about 15 DPA.The results indicated that water stress post anthesis showed significantly negative impact on the formation of HMW-GS in grains of both cultivars, and drought enhanced the initial formation of HMW-GS in grains of both cultivars.Compared with the control,drought promoted the HMW-GS accumulation at early grain filling stage,while the case was just opposite at late grain filling stage.Waterlogging showed more obviously negative impact on HMW-GS and GMP accumulation than drought.Finally,the control had the highest total HMW-GS and GMP contents,followed by drought treatment,and waterlogging was the lowest for both wheat cultivars.Under the same water treatment,each HMW-GS content of grains was higher in Yumai 34 than in Yangmai 9.
Wheat GSPs and Processing Quality Are Affected by Irrigation and Nitrogen through Nitrogen Remobilisation

Abstract:The rheological properties and end-use qualities of many foods are mainly determined by the types and levels of grain storage proteins (GSPs) in wheat. GSP levels are influenced by various factors, including tillage management, irrigation, and fertiliser application. However, the effects of irrigation and nitrogen on GSPs remain unclear. To address this knowledge gap, a stationary split-split block design experiment was carried out in low- and high-fertility (LF and HF) soil, with the main plots subjected to irrigation treatments (W0, no irrigation; W1, irrigation only during the jointing stage; W2, irrigation twice during both jointing and flowering stages), subplots subjected to nitrogen application treatments (N0, no nitrogen application; N180, 180 kg/ha; N240, 240 kg/ha; N300, 300 kg/ha), and cultivars tested in sub-sub plots (FDC5, the strong-gluten cultivar Fengdecun 5; BN207, the medium-gluten cultivar Bainong 207). The results showed that GSP levels and processing qualities were significantly influenced by nitrogen application (p < 0.01), N240 was the optimal nitrogen rate, and the influence of irrigation was dependent on soil fertility. Optimal GSP levels were obtained under W2 treatment at LF conditions, and the content was increased by 17% and 16% for FDC5 and BN207 compared with W0 under N240 treatment, respectively. While the optimal GSP levels were obtained under W1 treatments at HF conditions, and the content was increased by 3% and 21% for FDC5 and BN207 compared with W0 under N240 treatment, respectively. Irrigation and nitrogen application increased the glutenin content by increasing Bx7 and Dy10 levels in FDC5, and by increasing the accumulation of Ax1 and Dx5 in BN207. Gliadins were mainly increased by enhancing α/β-gliadin levels. Correlation analysis indicated that a higher soil nitrate (NO3-N) content increased nitrogen remobilisation in leaves. Path analysis showed that Dy10, Dx5, and γ-gliadin largely determined wet glutenin content (WGC), dough stability time (DST), dough water absorption rate (DWR), and sedimentation value (SV). Therefore, appropriate irrigation and nitrogen application can improve nitrogen remobilisation, GSP levels, and processing qualities, thereby improving wheat quality and production.
Expression of the 1Ax1 transgene in an elite Chinese wheat variety and its effect on functional properties.

Abstract:BACKGROUND: The transfer of transgenes from 'model'wheat genotypes into elite wheat cultivars using conventional plant breeding is an alternative strategy for improving the dough quality of wheat. Thus a cross was made between a popular Chinese elite wheat cultivar of the Yangzi River down-central area that expresses high-molecular-weight glutenin subunit (HMW-GS): 1 Bx7+1 By8 plus 1 Dx2+1 Dy1 2, and a model transgenic wheat line B1 02-1-2 which over-expresses HMW-GS 1 Ax1 in an L88-31 genetic background that includes HMW-GS 1 Bx1 7+By1 8. RESULTS: F-1 to F-6 generations of crosses between B102-1-2 (paternal) and Emai (maternal) were analysed for their HMW-GS compositions, allowing the selection of pure F-6 lines over-expressing HMS-GS 1Ax1 in the presence of the endogenous HMW-GS: 1 Bx7+1 By8 with 1 Dx2+1 Dy1 2, and 1 Bx1 7+1 By1 8 with 1 Dx2+1 Dy1 2. Analysis of the F-6 lines showed changes in the extensograph parameters, with increases in peak area, resistance to extension, and extensibility and peak resistance to extension, similar to those observed in the transgenic paternal line B1 02-1-2. CONCLUSION: This work showed that the expression levels of the 1Ax1 transgene and the effects on dough properties were similar in the transgenic parental line B102-1-2 and the F6 progeny generated from a cross with an elite Chinese cultivar Emai. Abstract:Basis for the effects of nitrogen (N) on wheat grain storage proteins (GSPs) and on the establishment of processing quality are far from clear. The response of GSPs and processing quality parameters to four N levels of four common wheat cultivars were investigated at two sites over two growing seasons. Except gluten index (GI), processing quality parameters as well as GSPs quantities were remarkably improved by increasing N level. N level explained 4.2~59.2% and 10.4~80.0% variability in GSPs fractions and processing quality parameters, respectively. The amount of N remobilized from vegetative organs except spike was significantly increased when enhancing N application. GSPs fractions and processing quality parameters except GI were only highly and positively correlated with the amount of N remobilized from stem with sheath. N reassimilation in grain was remarkably strengthened by the elevated activity and expression level of glutamine synthetase. Transcriptome analysis showed the molecular mechanism of seeds in response to N levels during 10~35 days post anthesis. Collectively, we provided comprehensive understanding of N-responding mechanisms with respect to wheat processing quality from N source to GSPs biosynthesis at the agronomic, physiological and molecular levels, and screened candidate genes for quality breeding.

Four wheat cultivars (Shiluan 02-1, Yannong 24, Jinan 17 and Lumai 21) with different end-use qualities were used to investigate the HMW-GS and GMP contents, and the GMP particle distributions in grain produced under irrigated and rainfed conditions. The percent volume of GMP particles and the contents of HMW-GS and GMP were affected by genotype and soil water. Genotype × soil water interaction was significant only for GMP particles < 12 μm and > 100 μm in the growing season of 2010–2011. Irrigated and rainfed conditions had different influences on the GMP particle distribution in the four cultivars. Compared to irrigated treatment, the rainfed treatment had higher accumulations of HMW-GS and GMP, especially in cultivars Yannong 24, Jinan 17 and Lumai 21. Rainfed conditions also increased the proportion of large size particles of GMP, indicating that different water regimes had an effect on grain quality. According to correlation coefficients (r), the contents of HMW-GS and GMP in grains were negatively correlated with the volume of < 12 μm GMP particles, but positively correlated with GMP particles > 100 μm.
Molecular Characterization and Marker Development of the HMW-GS Gene from Thinopyrum elongatum for Improving Wheat Quality

Abstract:The quality of wheat primarily depends on its storage protein quality, especially in regards to gluten content and high-molecular-weight glutenin subunits (HMW-GS). The number of HMW-GS alleles is limited in bread wheat (Triticum aestivum L.), whereas it is abundant in wheat relatives. Therefore, HMW-GS alleles from wheat relatives could provide a potential for improving quality in wheat breeding. Thinopyrum elongatum (EE) is one of the relatives of wheat. The E genome is closely related to the ABD genome in wheat; therefore, Th. elongatum is often used as an excellent exogenous gene donor for wheat genetic improvement. In this study, the high-molecular glutenin subunit gene was cloned and sequenced from Th. elongatum. A specific molecular marker for identifying the Glu-1Ey subunit gene was developed and applied to detected wheat-Th. elongatum alien introgression lines. Quality analysis indicated that the substitution and addition lines containing Th. elongatum alleles significantly (p < 0.05) increased grain protein content by 3.76% to 5.11%, wet-gluten content by 6.55% to 8.73%, flour 8-MW by 0.25% to 6.35%, and bread volume value by 33.77 mL to 246.50 mL, in comparing it with Chinese Spring. The GMP content and lactic acid SRC showed significant positive correlations with flour processing quality and might be used as indicators for wheat quality. The results were expected to provide a novel route for improving processing quality in wheat quality breeding.

Abstract:An increased number of high molecular weight glutenin subunits (HMW-GSs) is advantageous to the formation of glutenin macropolymer, the major determinant of end-product quality. Out of the six HMW-GS genes, Glu-1Ay is typically inactive in most hexaploid wheat cultivars. In the current study, we characterized two active Glu-1Ay genes Glu-1AyN11 and Glu-1AyC422 from a hexaploid wheat cultivar N11 and a durum wheat cultivar C422, respectively. The molecular masses of the 1Ay subunit of N11 (Glu-1AyN11) and C422 (Glu-1AyC422) were 61.1 kDa and 63.9 kDa, respectively, measured by matrix-assisted laser desorption/ionization time of flight mass spectrometry. Molecular characterization of Glu-1AyN11 and Glu-1AyC422 showed their complete coding regions were 1764 bp and 1827 bp in length, respectively. The deduced amino acid sequence of both Glu-1AyN11 and Glu-1AyC422 has a similar typical primary structure as the other y-type HMW-GSs, but in comparison to Glu-1By and Glu-1Dy subunits, the Glu-1Ay subunit has one less conserved cysteine residue in the repetitive domain. Phylogenetic analysis revealed that the inactive Glu-1Ay had a closer evolutionary relationship with Glu-1AyC422 than with Glu-1AyN11. The divergence time among Glu-1AyN11, Glu-1AyC422, and other y-type subunit genes was also estimated.

Abstract:In order to determine the genetic differences between high-molecular weight glutenin subunits (HMW-GS) 7OE + 8* and 17 + 18, HMW-GS 7OE + 8* from a Canadian extra-strong wheat cultivar Glenlea was introduced to Longmai 20 (1, 17 + 18, 5 + 10) by 6 consecutive backcrosses with biochemical marker-assisted selection. The near-isogenic lines (NILs) of Longmai 20 possessing HMW-GS 7OE + 8* and 17 + 18 were grown in the experimental field of the Crop Breeding Institute of Heilongjiang Academy of Agricultural Sciences from 2006 to 2009. The field experiments were conducted in a two-column contrast arrangement with four replicates. Compared with the NILs possessing subunits 17 + 18, the quality parameters of NILs with subunits 7OE + 8*, which include protein content, dry gluten content, ratio of wet gluten/dry gluten, ratio of Zeleny sedimentation/dry gluten, stability, maximum resistance and area of extensogram, increased by 1.5% (P = 0.06), − 0.7% (P = 0.47), − 1.9% (P < 0.01), 6.6% (P = 0.02), 11.2% (P = 0.6), 20.3% (P < 0.01) and 19.9% (P < 0.01), respectively. HMW-GS 7OE + 8* have a greater positive effect on the rheological properties of flour, such as the maximum resistance and area of extensogram, which are less influenced by environmental conditions compared to stability of Farinogram.

Abstract:Wheat gluten proteins, especially high-molecular-weight glutenin subunits (HMW-GS), are the main contributor to flour processing quality. Tannic acid (TA) consisting of a central glucose unit and ten gallic acid molecules is a phenolic acid that improves the processing quality. However, the underlying mechanism of TA's improvement remains largely unknown. Here, we showed that TA's improving effects on gluten aggregation, dough-mixing and bread-making properties were directly associated with the kinds of HMW-GS expressed in wheat seeds in HMW-GS near-isogenic lines (NILs). We established a biochemical framework, elucidated the additive effects of HMW-GS-TA interaction and discovered that TA cross-linked specifically with wheat glutenins but not gliadins, and reduced gluten surface hydrophobicity and SH content depending on the kinds of expressed HMW-GS in the wheat seeds. We also demonstrated that hydrogen bonds play an essential role in TA-HMW-GS interactions and improvement of wheat processing quality. Additionally, the effects of TA on the antioxidant capacity and on nutrient (protein and starch) digestibility were also investigated in the NILs of HMW-GS. TA increased antioxidant capacity but did not affect the digestion of starches and proteins. Our results revealed that TA more effectively strengthened wheat gluten in the presence of more HMW-GS kinds, highlighting TA's potential as an improver toward healthy and quality bread and demonstrating that manipulating hydrogen bonds was a previously overlooked approach to improve wheat quality.

Abstract:The difference in accumulation of high‐molecular‐weight glutenin subunits (HMW‐GS) in superior (basal) and inferior (distal) grains results in the nonuniformity of grain quality in a winter wheat (Triticum aestivum L. ‘Yangmai 158’). The HMW‐GS accumulation and glutenin macropolymer (GMP) content were studied in superior and inferior grains during the grain‐filling period. Compared with inferior grains, HMW‐GS was formed earlier and total accumulation amount was higher in superior grains. The total HMW‐GS content was higher in superior grain than inferior grain, except at maturity. For individual HMW‐GS types, the accumulation and content of subunit 7 were the highest, followed by subunit 12, and those of subunit 8 were the lowest, followed by subunit 2 in superior grain. In contrast, the accumulation and content of subunit 7 at maturity were significantly higher than subunit 8 but similar between subunit 2 and subunit 12 in inferior grain. Moreover, the accumulation of subunit 7 and 12 in superior grain was significantly higher than in inferior grain. However, compared with the inferior grain, the GMP accumulation was higher but content was lower in superior grain at maturity.
