Abstract:In this study, we investigated the mechanism of transcutaneous adjuvant activity of the CpG-oligonucleotide (K3) in mice. Transcutaneous immunization (TCI) with an ovalbumin-loaded self-dissolving microneedle patch (OVA-sdMN) and K3-loaded hydrophilic gel patch (HG) increased OVA-specific Th2- and Th1-type IgG subclass antibody titers more rapidly and strongly than those after only OVA-sdMN administration. However, the antigen-specific proliferation of OVA-specific CD4+ T cells was similar between the OVA-only and the OVA+K3 groups. Population analysis of various immune cells in draining lymph nodes (dLNs) in the primary immune response revealed that the OVA+K3 combination doubled the number of dLN cells, with the most significant increase in B cells. Phenotypic analysis by flow cytometry revealed that B-cell activation and maturation were promoted in the OVA+K3 group, suggesting that direct B-cell activation by K3 largely contributed to the rapid increase in antigen-specific antibody titer in TCI. In the secondary immune response, a significant increase in effector T cells and effector memory T cells, and an increase in memory B cells were observed in the OVA+K3 group compared with that in the OVA-only group. Thus, K3, as a transcutaneous adjuvant, can promote the memory differentiation of T and B cells.

Abstract:Background Transcutaneous vaccines have received wide attention due to their easy-to-use, needle-free, noninvasive delivery. However, the novel barrier function of stratum corneum hinders the transport of antigen and adjuvant in transcutaneous immunization. Novel nanoscale delivery systems employing, for example, liposomes and nanoparticles, have been widely investigated to overcome the penetration barrier of stratum corneum for effective transcutaneous immunization. Objective The objective of this study was to prepare two types of flexible liposomes and determine their efficacies for the transcutaneous delivery of antigen and the subsequent immune response induced in vivo. Methods Ovalbumin (OVA) liposome-based transcutaneous vaccines were prepared using reverse-phase evaporation and film-dispersion methods. Particle sizes and antigen encapsulating efficiency were then evaluated. After application to bare mouse skin, topical sites were examined for the presence of fluorescence-labeled liposome. The efficacy of the transcutaneously delivered OVA-loaded flexible liposome in activating the immune responses was investigated by detecting serum immunoglobulin G levels. The influence of an adjuvant, imiquimod, in the transcutaneous immunization was also tested. Results Two flexible liposomes with well-encapsulated OVA were successfully prepared by film-dispersion or reverse-phase evaporation methods. The sizes of the prepared flexible liposomes ranged from 200 to 400 nm. In vivo, the fluorescence-labeled liposome was detected in hair-follicle ducts, indicating that the flexible liposome can penetrate the skin barrier through the hair follicles. Upon transcutaneous administration, the OVA-encapsulated flexible liposome elicited a strong immune response similar to that of positive control (ie, OVA solution administrated by subcutaneous injection with Al(OH)3 as an adjuvant). Co-administration of imiquimod with the OVA-loaded liposome expressed a significant enhancement on the transcutaneous immune responses. Conclusion Results of this study highlight the nanoscale formulation, flexible liposome, as a promising carrier for the transcutaneous delivery of antigen proteins. Imiquimod was shown to be an effective adjuvant as a transcutaneous immunization enhancer with the potential for transcutaneous vaccine development.

Abstract:Transcutaneous immunization (TCI) is an appealing vaccination method. Compared with conventional injectable immunization, TCI is easier and less painful. We previously developed a dissolving microneedle (MN) patch and demonstrated that TCI using MN patches demonstrates high vaccination efficacy without adverse events in humans. In this study, we investigated the immune induction mechanism of TCI using our MN patch, focusing on inflammatory responses in the skin and on the dynamics, activation, and differentiation of various immunocompetent cells in draining lymph nodes (dLNs). We demonstrate that inflammatory cytokines such as IL-6 and TNF-α increased in the skin at an early stage after MN patch application, inducing the infiltration of macrophages and neutrophils and promoting the activation and migration of skin-resident antigen-presenting cells (Langerhans and Langerin- dermal dendritic cells) to dLNs. Moreover, the activated antigen-presenting cells reaching the dLNs enhanced the differentiation of T (Teff, Tem, and Tcm) and B (plasma and memory) cells. This may contribute to the efficient antigen-specific antibody production induced by TCI using MN patches. We believe that our findings reveal a part of the immune induction mechanism by TCI and provide useful information for the development and improvement of TCI formulations based on the immune induction mechanism.

Abstract:Transcutaneous immunization (TCI) with dissolving microneedle arrays (DMAs) is a promising vaccine administration method. In this work, we developed a TCI device consisting of dissolving polyvinylpyrrolidone (PVP) microneedles array, where in the tips are loaded with antigen and adjuvant encapsulated in liposomes. The microneedles could effectively be inserted into the skin and completely dissolve within 3 min. As a test-case, we selected ovalbumin (OVA) as a model antigen, CpG OND as adjuvant and cationic liposome (Lip) as a microparticulate vehicle for co-deliver antigens and adjuvant. Mice were immunized transcutaneously with DMAs containing OVA, OVA-CpG OND, OVA encapsulated in Lip, OVA-CpG OND encapsulated in Lip and conventional intramuscular injection (IM) with OVA solution, respectively. The results show that the anti-OVA IgG antibody level in the group immunized with the DMA containing OVA-CpG OND encapsulated in Lip was significantly higher than that of the other groups. Furthermore, it significantly increased the level of IgG2a (P<0.05) and achieved the shift of immune type from predominate Th2 type to a balance Th1/Th2 type. In conclusion, the DMA TCI device can effectively deliver the Lip encapsulating CpG OND-OVA into skin, enhancing the immune response and change the immune type.

Abstract:An in vitro immunization (IVI) protocol enables antigen specific antibody production from L-Leucyl-L-Leucine methyl ester (LLME)-treated human peripheral blood lymphocytes (PBL) upon antigen stimulation in the presence of IL-2, IL-4, and muramyl dipeptide. In the course of our studies, we have found that IL-10 added at the antigen sensitization significantly augmented antibody production level from the LLME-treated PBL. In the present study, we tried to demonstrate the role of IL-10 in the augmentation of antibody production in an IVI protocol by clarifying the cytokine expression profiles in CD4(+) and CD8(+) T cells. The results showed that IL-10 skewed the Th1/Th2 balance to Th2-type responses by suppressing Th1-type cytokine production and augmenting Th2-type cytokine production in CD4(+) and CD8(+) T cells, as well as in CD19(+) B cells. Furthermore, IL-10 augmented the expression of CD38, an antigen marker of plasma cells, on B cells, which clearly indicates that IL-10 promoted differentiation and maturation of B cells in an IVI protocol. These results indicate that IL-10 plays an important role in setting the cellular milieu to produce antibodies in an IVI protocol.
Modulation of Th1 and Th2 Cytokine Production with Imiquimod in the PBLN Cell Cultures from Ovalbumin-Sensitized Rats

Abstract:Objective To investigate the mechanism of the immune response modifier imiquimod.Methods PBLN was isolated and cultured with different concentrations of imiquimod(0.10,1.0,or 10μg/ml)for 0,3,6,12,24 or 48 h.The cell-free supernatants were harvested and expressions of interlukin(IL)-4,IL-5,IL-12 and IFN-γ determined by ELISA.Results Concentrations of IL-12 and IFN-γ were increased significantly by imiquimod of 1.0 and 10μg/ml,while the levels of IL-4 and IL-5 were increased slowly by ovalbumin alone(P0.05).The effect began at 6h,achieved a peak level at 12h,and lasted for 24h.Conclusion Imiquimod shows its effect by marked inhibition of the expression of Th2-type cytokines IL-4,IL-5,and a significant increase of expression of Th1-type cytokines IL-12,IFN-γ.It may be an effective approach in the management of atopic disease.

Abstract:Imiquimod (IMQ; brand name Aldara®) is a registered topical agent that has been proven to induce local inflammation via the Toll-like receptor (TLR)7 pathway. The purpose of this study was to characterize TLR7-mediated inflammation following 7 days (168 h) of topical IMQ exposure in healthy volunteers, and to compare the effects of short exposure (48 h-72 h) with prolonged exposure (120 h-168 h). IMQ (100mg) was applied under occlusion to 5 different tape-stripped treatment sites on the back of 10 healthy participants for a maximum of 7 consecutive days. Erythema and skin perfusion were measured daily up to 168h. Biopsies for immunohistochemical staining and RNA sequencing were collected at 0h, 48h, 72h, 120h and 168h post IMQ application. IMQ triggered an inflammatory response starting at 48h after application, including erythema and perfusion of the skin. At the transcriptomic level, IMQ induced TLR7 signalling, IRF involvement and activation of TNF signalling via NF-κB. Furthermore, an enhanced inflammatory response at the cellular level was observed after prolonged IMQ exposure, with cellular infiltration of dendritic cells, macrophages and T cells which was also corroborated by transcriptomic profiles. No difference was found in the erythema and perfusion response after 168h of IMQ exposure compared to 72h. Prolonged IMQ exposure revealed enhanced cellular responses and additional pathways with modulated activity compared to short exposure and can therefore be of interest as a model for investigational compounds targeting innate and adaptive immune responses.

Abstract:Transcutaneous immunization (TCI) is easy to use, minimally invasive, and has excellent efficacy in vaccines against infections. We focused on toll-like receptor (TLR) ligands as applicable adjuvants for transcutaneous formulations and characterized immune responses. TCI was performed using poke-and-patch methods, in which puncture holes are formed with a polyglycolic acid microneedle on the back skin of mice. Various TLR ligands were applied to the puncture holes and covered with an ovalbumin-loaded hydrophilic gel patch. During the screening process, K3 (CpG-oligonucleotide) successfully produced more antigen-specific antibodies than other TLR ligands and induced T helper (Th) 1-type polarization. Transcutaneously administered K3 was detected in draining lymph nodes and was found to promote B cell activation and differentiation, suggesting a direct transcutaneous adjuvant activity on B cells. Furthermore, a human safety test of K3-loaded self-dissolving microneedles (sdMN) was performed. Although a local skin reaction was observed at the sdMN application site, there was no systemic side reaction. In summary, we report a K3-induced Th1-type immune response that is a promising adjuvant for transcutaneous vaccine formulations using MN and show that K3-loaded sdMN can be safely applied to human skin.

Abstract:Adjuvants are essential for the induction of robust immune responses against vaccine antigens. Small-molecule TLR7 agonists hold high potential for this purpose. In this communication, imiquimod (IMQ) bearing a cholesterol lipid moiety derivative, IMQ-Chol, was designed and synthesized as a vaccine adjuvant, which could release parent IMQ molecules in aqueous conditions via amide bond hydrolysis. We performed a series of immunological evaluations by cooperating with the inactivated foot-and-mouth disease virus (FMDV). All of the results confirmed that IMQ-Chol could stimulate the body for a prolonged time to produce strong humoral and cellular immunity with a balanced Th1/Th2 immune response through a TLR7-related MAPK pathway. In addition, the results of the proof-of-concept vaccine indicated IMQ-Chol had a good effect on preventing and treating FMD in pigs.

Abstract:Abstract Novel effective vaccines are key to solving many global health problems. Past efforts to rationally design vaccines that elicit strong cell-mediated responses have largely failed. Vaccine adjuvants, particularly toll-like receptor (TLR) agonists, are a promising tool to shape immune responses by polarizing CD4 T cells to distinct fates. Use of these adjuvants to design novel vaccines requires the full characterization of their effects on CD4 T cell differentiation, particularly in regard to the route of vaccination. In a large scale in vitro screen of vaccine adjuvants, we demonstrated that Pam3CSK4, a synthetic mimic of bacterial lipoproteins with TLR1/2 agonist activity, strongly polarized CD4 T cells to the Th1 fate (IFN-γ+, T-bet+) supporting cell-mediated responses. We adapted a vaccination system to study CD4 T cell polarization in vivo using ovalbumin-specific transgenic CD4 T cells. This in vivo system showed a route-specific effect, with strong Th1 responses from intravenous immunization with ovalbumin plus Pam3CSK4, but a lack of Th1 responses with immunization through more clinically relevant cutaneous routes. Interestingly, Pam3CSK4 in combination with a known Th1 polarizing adjuvant, CpG ODN, showed little to no Th1 response suggesting a dominant suppressive mechanism. Furthermore, suppression of Th1 differentiation occurred in response to both TLR1/2 and TLR6/2 agonists. Using a variety of cell-ablative mouse models we were able to reverse suppression of Th1 responses and identify specific candidates of innate immune cells mediating suppression during vaccination with TLR2 agonists. Current work focuses on mechanisms of suppression by TLR2 agonists in the skin and relevance to improving vaccine design.

Abstract:Objective: Transcutaneous immunization (TCI) is a novel route of vaccination through application of a topical vaccine antigen on skin. Phosphorylcholine (PC) is a structural component of a variety of pathogens, and anti-PC immune responses protect mice against invasive bacterial diseases. The purpose of the study was to examine the effect of TCI using PC in back skin or auricle skin in BALB/c mice. Methods: TCI was performed in BALB/c mice in back skin or auricle skin using PC-keyhole limpet hemocyanin (KLH) plus cholera toxin (CT). Inoculations were given once each week for six consecutive weeks. Immunogenicity was evaluated by measuring PC-specific IgG and specific IgG1, IgG2a, IgM, IgA, and secretory IgA antibodies by ELISA. IL-4, IL-5, IL-10, IL-12, IL-13 and IFN-γ levels were also measured by ELISA. Results: Serum IgG after TCI in auricle skin was significantly higher than after TCI in back skin and in controls. Secretory IgA antibodies after TCI in auricle skin were also significantly higher than after TCI in back skin and in controls in nasal, BALF, vaginal and fecal samples. PC-specific IgG1 and IgG2a were significantly higher after TCI in auricle skin compared to controls and compared to TCI in back skin. IgG1 was significantly higher than IgG2a after TCI in auricle skin. Production of IFN-γ, IL-4 and IL-10 from CD4+ cells was significantly higher after TCI in auricle skin than after TCI in back skin and in controls, whereas IL-5, IL-12 and IL-13 were not detected in any mice. Conclusion: These results suggest that TCI in auricle skin using PC plus CT in BALB/c mice is a simple approach for induction of systemic and mucosal immune responses that are shifted in the Th2 direction.

Abstract:Paclitaxel, a diterpenoid isolated from the bark of the Taxus cuspidate cv. Nana, was evaluated for its adjuvant effect on the immune responses in a mouse model. Fifty-six mice were randomly distributed into seven groups with 8 mice in each. Animals were subcutaneously immunized on days 1 and 21 with 100μg of paclitaxel, 10μg of ovalbumin (OVA), OVA with paclitaxel (50, 100 or 200μg) or with aluminum hydroxide (alum). Two weeks after the primary and boost immunizations, blood samples were collected for measurement of serum antibodies. Splenocytes were separated for detection of lymphocyte proliferation in responses to concanavalin A (Con A), lipopolysaccharide (LPS) and OVA, and mRNA expression of Th1 cytokines (IFN-γ and IL-12), Th2 cytokines (IL-10 and IL-5) and transcription factors T-bet/GATA-3 (Th1/Th2 switcher). Results showed that coadministration of OVA with paclitaxel induced significantly higher IgG, IgG1, IgG2a, IgG2b, IgG3 and IgM responses than when OVA was used alone. In addition, up-regulated T-bet/GATA-3 together with significantly increased mRNA expression of IL-4, IL-10, IFN-γ and IL-12 by splenocytes, as well as the proliferative responses of splenocytes to Con A, LPS and OVA were observed in paclitaxel-adjuvanted groups. Incubation of a murine macrophage-like cell line with paclitaxel significantly increased TNF-α and -10 released from the cells and expression of microRNAs such as miR-155, miR-147, miR-146a and miR-132. Therefore, paclitaxel activated both Th1 and Th2 responses. Considering its unique adjuvant effect demonstrated in this study and a safe record clinically used as an antineoplastic agent, paclitaxel could be an ideal adjuvant candidate when mixed Th1/Th2 immune responses are needed.
Optimization of in Vitro Immunization Protocol to Produce Antigen Specific Human Monoclonal Antibody by Demonstrating the Role of IL-10

Abstract:We have previously reported that in vitro immunization (IVI) protocol enables antigen specific antibody production from Leu-Leu-Ome (LLME)-treated human peripheral blood lymphocytes (PBL) by stimulating with antigen in the presence of IL-2, IL-4 and muramyl dipeptide (MDP). In the course of our studies to optimize the antibody production, we evaluated the cytokine production profiles in the IVI protocol. IL-10 was produced in non-treated PBL, which was thought to be one of reasons for inability of non-treated PBL to produce antibody, and also produced in LLME-treated PBL 1 wk after the antigen sensitization, where antibody production was strongly enhanced. Furthermore, IL-10 added to the LLME-treated PBL before the antigen sensitization, antibody production decreased. However, IL-10 added to the LLME-treated 13131, at the antigen sensitization augmented antibody production level. These results strongly suggest that IL-10 might play an important role in the activation and/or inactivation of T cells, B cells and antigen presenting cells at the antigen sensitization, where IL-10 might alter the cellular milieu, resulting in the suppression or activation of antibody production. In the present study, we tried to further evaluate the function of IL-10 in antibody production in the IVI protocol. Results showed that IL-10 added before the antigen sensitization targets CD8 T cells, which might lead to elicit the suppressive function of T cells, while IL-10 added at the antigen sensitization elicits Th2 type responses via suppressing IFN-gamma and augmenting IL-4 production.
Combination of Microneedles and MF59 Adjuvant As a Simple Approach to Enhance Transcutaneous Immunization.

Abstract:MF59, an oil-in-water nanoemulsion, has been used in licensed seasonal influenza vaccines for many years. Administration of such vaccines by injection is associated with pain and safety issues. Here, we evaluated the potential of administering MF59 via a transcutaneous route with antigen loading (either encapsulated into or mixed with MF59) to intact or microneedle-pretreated skin. In addition to commercial MF59, we also prepared a nanoemulsion to encapsulate hydrophilic antigens by mimicking the formulation and preparation technique of MF59. The nanoemulsion was prepared using a water-in-oil-in-water emulsion method, and was similar to MF59 in composition, particle size, and morphology. Compared with the intact skin group, the microneedle-pretreated group showed significant enhanced antigen penetration. In vivo transcutaneous immunization analysis showed that the MF59-adjuvant influenza vaccine elicited approximately 3-5 times higher hemagglutination inhibition titers than the influenza solution alone in BALB/c mice after microneedle pretreatment. The intact skin group showed negative immune results at the same dose, suggesting that microneedle pretreatment was critical for efficient delivery of antigens, to obtain strong immune responses. Furthermore, the loading method (encapsulation or mixing with the vehicle) did not affect the dermal penetration or transcutaneous immunization of antigens on microneedle-pretreated skin. Moreover, in vitro cellular assays showed that MF59 facilitated the maturation of dendritic cells and enhanced antigen uptake by antigen-presenting cells. In conclusion, the combination of microneedle pretreatment and mixing of MF59 with antigen provides a simple approach to enhance transcutaneous immunization.

Abstract:ABSTRACT BMS-200475 was recently shown to have potent antiviral activity against hepatitis B virus (50% effective concentration = 3.7 nM; 50% cytotoxic concentration = 30 μM). In metabolic studies in both HepG2 and hepatitis B virus-transfected 2.2.15 human hepatoma cell lines, the metabolism was similar, the primary products being the di- and triphosphates. The accumulation of triphosphate was rapid and detectable down to a 5 nM concentration of added drug. When cells were labeled at 25 μM, the intracellular triphosphate concentration attained 30 pmol/106 cells (∼30 μM). The intracellular half-life of the triphosphate was about 15 h. Compared with five other nucleoside analogs of medical interest (lamivudine, penciclovir, ganciclovir, acyclovir, and lobucavir), BMS-200475 was most efficiently phosphorylated to the triphosphate in HepG2 cells.
Immune Adjuvant Targeting Micelles Allow Efficient Dendritic Cell Migration to Lymph Nodes for Enhanced Cellular Immunity.

Abstract:Cellular immunity is essential for the effectiveness of vaccines against cancer. After capture of vaccines, dendritic cells (DCs) have to migrate to lymph nodes via chemokine receptor type 7 (CCR7). Subsequently, DCs present cytosolic antigens via major histocompatibility complex class I (MHC I) molecules to induce cellular immunity. However, various vaccines fail to induce potent cellular immunity due to insufficient MHC I-restricted antigen presentation and limitations of immune adjuvants. Hence, we constructed novel immune adjuvant targeting micelles (M-COSA) to targeted codeliver antigen ovalbumin (OVA) and plasmid DNA encoding CCR7 (CCR7 pDNA) to the cytosol of DCs, thus promoting DC migration to lymph nodes to boost MHC I-restricted antigen presentation. M-COSA exhibited adjuvant activity and demonstrated more efficient DC cellular uptake compared with COSA. M-COSA/OVA/pDNA increased costimulatory molecule expression and cytokine secretion, resulting in DC activation and maturation. Moreover, antigens and pDNA, which were encapsulated in micelles, escaped from the endosome into the cytoplasm to achieve MHC I-restricted antigen presentation and increase CCR7 expression. The number of CD8+ T cells, which was positively correlated with tumor rejection, was notably increased and tumor growth was dramatically suppressed after vaccination with M-COSA/OVA/pDNA. In summary, M-COSA/OVA/pDNA micelles, which allow DC targeting and efficient DC migration to lymph nodes to enhance cellular immunity, exhibit effective tumor inhibition and lay the foundation for novel vaccine design.
Immune Modulation by Adjuvants Combined with Diphtheria Toxoid Administered Topically in BALB/c Mice After Microneedle Array Pretreatment

Abstract:PurposeIn this study, modulation of the immune response against diphtheria toxoid (DT) by various adjuvants in transcutaneous immunization (TCI) with microneedle array pretreatment was investigated.MethodsTCI was performed on BALB/c mice with or without microneedle array pretreatment using DT as a model antigen co-administrated with lipopolysaccharide (LPS), Quil A, CpG oligo deoxynucleotide (CpG) or cholera toxin (CT) as adjuvant. The immunogenicity was evaluated by measuring serum IgG subtype titers and neutralizing antibody titers.ResultsTCI with microneedle array pretreatment resulted in a 1,000-fold increase of DT-specific serum IgG levels as compared to TCI. The immune response was further improved by co-administration of adjuvants, showing a progressive increase in serum IgG titers when adjuvanted with LPS, Quil A, CpG and CT. IgG titers of the CT-adjuvanted group reached levels comparable to those obtained after DT-alum subcutaneous injection. The IgG1/IgG2a ratio of DT-specific antibodies decreased in the following sequence: plain DT, Quil A, CT and CpG, suggesting that the immune response was skewed towards the Th1 direction.ConclusionsThe potency and the quality of the immune response against DT administered by microneedle array mediated TCI can be modulated by co-administration of adjuvants.

Abstract:Previous study showed that CTB (Cholera toxin subunit B) can be used as a genetic adjuvant to enhance the systemic immune responses. To further investigate whether it can also be used as a genetic adjuvant to improve mucosal immune responses, we constructed DNA and recombinant Tiantan vaccinia (rTTV) vaccines expressing OVA-CTB fusion antigen. Female C57BL/6 mice were immunized with an intranasal DNA priming/intramuscular rTTV boosting regimen. OVA specific T-cell responses were measured by IFN-γ ELISPOT and specific antibody responses were determined by ELISA. Compared to the nonadjuvant group (pSV-OVA intranasal priming/rTTV-OVA intramuscular boosting), pSV-OVA-CTB intranasal priming/rTTV-OVA-CTB intramuscular boosting group significantly improved the magnitudes of T-cell responses at spleen (1562 ± 567 SFCs/10(6) splenocytes versus 330 ± 182 SFCs/10(6) splenocytes, P < 0.01), mesenteric LN (96 ± 83 SFCs/10(6) lymphocytes versus 1 ± 2 SFCs/10(6) lymphocytes, P < 0.05), draining LNs of respiratory tract (109 ± 60 SFCs/10(6) lymphocytes versus 2 ± 2 SFCs/10(6) lymphocytes, P < 0.01) and female genital tract (89 ± 48 SFCs/10(6) lymphocytes versus 23 ± 21 SFCs/10(6) lymphocytes, P < 0.01). These results collectively demonstrated that fusion-expressed CTB could act as a potent adjuvant to improve both systemic and mucosal T-cell responses.
Fatty Acid-Mimetic Micelles for Dual Delivery of Antigens and Imidazoquinoline Adjuvants

Abstract:Vaccine design has undergone a shift towards the use of purified protein subunit vaccines, which offer increased safety and greater control over antigen specificity, but at the expense of immunogenicity. Here we report the development of a new polymer-based vaccine delivery platform engineered to enhance immunity through the co-delivery of protein antigens and the Toll-like receptor 7 (TLR7) agonist imiquimod (IMQ). Owing to the preferential solubility of IMQ in fatty acids, a series of block copolymer micelles with a fatty acid-mimetic core comprising lauryl methacrylate (LMA) and methacrylic acid (MAA), and a poly(ethylene glycol) methyl ether methacrylate (PEGMA) corona decorated with pyridyl disulfide ethyl methacrylate (PDSM) moieties for antigen conjugation were synthesized via reversible addition-fragmentation chain transfer (RAFT) polymerization. Carriers composed of 50 mole% LMA (LMA50) demonstrated the highest IMQ loading (2.2 w/w%) and significantly enhanced the immunostimulatory capacity of IMQ to induce dendritic cell maturation and proinflammatory cytokine production. Conjugation of a model antigen, ovalbumin (OVA), to the corona of IMQ-loaded LMA50 micelles enhanced in vitro antigen uptake and cross-presentation on MHC class I (MHC-I). A single intranasal (IN) immunization of mice with carriers co-loaded with IMQ and OVA elicited significantly higher pulmonary and systemic CD8+ T cell responses and increased serum IgG titer relative to a soluble formulation of antigen and adjuvant. Collectively, these data demonstrate that rationally designed fatty acid-mimetic micelles enhance intracellular antigen and IMQ delivery and have potential as synthetic vectors for enhancing the immunogenicity of subunit vaccines.
Iontophoresis‐Driven Microneedle Arrays Delivering Transgenic Outer Membrane Vesicles in Program That Stimulates Transcutaneous Vaccination for Cancer Immunotherapy

Abstract:Transdermal delivery of antigen and chemokine proteins that activates the maturation of skin dendritic cells (DCs) and direct the migration of activated DCs to lymph and spleen is an important alternative to conventional vaccines. However, stratum corneum forms a barrier to skin penetration. The poor cellular uptake of free antigens and chemokines also limits transcutaneous immunization efficacy. In this work, a pair of iontophoresis‐driven microneedle patches is constructed, of which, two kinds of outer membrane vesicles (OMVs) derived from Escherichia coli transformed by plasmid encoding gp100 (IPMN‐G) and chemokine ligand 21 (IPMN‐C) are incorporated within microneedles, respectively. The topical application of IPMN‐G and IPMN‐C shows the effectiveness of transdermally delivering gp100 and CCL21 secreting vesicles to skin DCs. With iontophoresis as a driving generator, the release and uptake of transgenic OMVs in target cells are significantly enhanced, with transcutaneous immunization initiated. The in vivo applications of IPMN‐G and IPMN‐C with a 12 h interval retard the progression and prevent the occurrence of tumor spheroids. IPMN‐GC is shown as a promising triplatform in engineering transgenic OMV‐incorporated microneedles, driven by iontophoresis into a transcutaneous vaccine, providing a noninvasive system for the transdermal delivery of antigen and chemokine proteins for transcutaneous vaccination‐meditated immunotherapy.
