Abstract:The quantification of illicit drugs in wastewater has become a valuable tool for monitoring illicit drug abuse. The commonly utilized methods for detecting drugs in wastewater require a substantial sample volume, extended pretreatment durations, and intricate procedures. This study first employed polydopamine-coated magnetic nanocomposites as adsorbents for magnetic solid-phase extraction, combined with UPLC-MS/MS, to simultaneously determine the concentrations of eleven common illicit drugs in wastewater. The synthesis process for Fe3O4@PDA is straightforward and high-yield. Benefiting from the strong magnetic response, good dispersibility, and abundant binding sites of the prepared nanocomposites, the extraction of illicit drugs from wastewater could be achieved in just 15 min. The method exhibited satisfactory limits of quantitation (ranging from 5 to 10 ng/L), commendable accuracy (ranging from 90.59 % to 106.80 %), good precision (with RSDs below 10 %), and less sample consumption (only 1 mL). The efficacy of this method was successfully validated through its application to actual wastewater samples collected from ten wastewater treatment plants. The results indicated that morphine, codeine, methamphetamine, and ketamine were the predominant illicit drugs present in the samples. The method developed is able to meet the needs of common illicit drug monitoring and high-throughput analysis requirements.

Abstract:Concerns have been raised about synthetic cannabinoids (SCs), which are among the most often trafficked and used illegal substances. An analytical method that holds promise for determining illicit drug use in the general population is wastewater-based epidemiology (WBE). Unfortunately, the concentration of SCs in wastewater is often extremely low on account of their hydrophobic nature, thus presenting a significant obstacle to the accurate detection and quantification of SCs using WBE. In this study, we present a novel magnetic nanomaterial as amphiphilic adsorbents for pretreatment of wastewater using magnetic solid phase extraction (MSPE). Polydopamine-modified Fe3O4 nanoparticles were used as the magnetic core and further functionalized with poly(divinylbenzene-N-vinylpyrrolidone). Coupled with UHPLC-MS/MS analysis, an analytical method to simultaneously detect nine SCs at trace-level in wastewater was developed and validated, enriching 50 mL wastewater to 100 μL with limits of detection (LOD) being 0.005-0.5 ng/L, limits of quantification (LOQ) being 0.01-1.0 ng/L, recoveries ranging from 73.99 to 110.72%, and the intra- and inter-day precision's relative standard deviations less than 15%. In comparison to the time-consuming conventional column-based solid phase extraction, the entire MSPE procedure from sample pre-treatment to data acquisition could be finished in one hour, thus largely facilitating the WBE method for drug surveillance and control.

Abstract:Drug abuse is a social issue worldwide, and there is an increasing demand for on-site rapid detection of illicit drugs. In this study, a rapid and simple analytical method for the detection of 6-monoacetylmorphine (6-MAM), methamphetamine (MA), methylenedioxymethamphetamine (MDMA), ketamine (K), norketamine (NK), and cocaine (COC) in urine was developed. The developed method combines C18 pipette-tip based solid-phase extraction (C18 PT-SPE) with a miniaturized mass spectrometer (miniMS), exhibiting remarkable simplicity, high sensitivity, and strong reliability, compared with the conventional method. The optimal extraction and elution conditions for C18 PT-SPE were considered as 9 and 3 aspirating-dispensing cycles, respectively. The miniMS parameters including spray voltage, isolation potential, and collision-induced dissociation energy for the detection of these six illicit drugs were optimized using a nano-electrospray ionization method. The limit of detection (LOD), limit of quantification (LOQ), linear range, and linearity for the analysis of six illicit drugs in urine with the proposed C18 PT-SPE-miniMS method were determined. Except for the LOD of K and COC was determined as 0.5 and 0.25 ng mL-1 respectively, and the LOD of 6-MAM MA, MDMA, and NK was determined as 1 ng mL-1. This method enables rapid on-site detection, providing easier operation, lower cost, and better portability compared to conventional methods, making it a potential tool in drug crime investigation and forensic science.

Abstract:Due to the obvious minimal doses of drugs in biological matrices as well as the societal difficulties caused by methamphetamine usage, methamphetamine identification is critical in clinical and forensic laboratories. Because of their simple and inexpensive production procedure, as well as their excellent selectivity and sensitivity, polymeric carbon-based nanocomposites are strong contenders for the diffusive solid-phase extraction approach. The diffusive solid-phase extraction absorbent nanographene oxide polypyrrole composite was produced and used to recover methamphetamine from a complicated urine substrate. The generated NGPPC was fully characterized, and the significant extracting parameters have been explored using the one-parameter-at-a-time strategy. NGOPC is being used to extract methamphetamine using a urine medium with high efficiency. The NGPPC synthesizing procedure was easy, and the extraction method will demonstrate good repeatability. Moreover, the practical and efficient synthesis process stimulates the use of carbon-based compounds in various extraction procedures. As for detecting and quantifying equipment, HPLC monitors are being used. 300 mL methanol, 7 min extracting and desorption duration, 5000 mixing frequency, urinary pH value of 20, 40 mg adsorption, and 5 mL amount of urine were the optimal extraction variables. Following tracing the calibration graph, the method’s linear ranges were determined to be 40-600 ng/ml. The detection limits (LOD) and quantitation limits (LOQ), correspondingly, were 10 and 35.80 ng/mL. The proposed methodology seemed to have a detection range of 9 ng/mL. The suggested approach’s applicability in numerous characterization and medical facilities was proven by the examination of addicted subjects using the proposed technique. For successful extraction of methamphetamine using biological urine samples, the carbon-based adsorbent was being used as diffusive solid-phase extraction adsorption.

Abstract:A novel nano petal-shaped covalent organic frameworks modified magnetic polystyrene-divinylbenzene-glycidylmethacrylate (/DVB/GMA) microsphere has been synthesized. It is a perfect combination of high productivity of PS/DVB/GMA microspheres and excellent enrichment efficiency of COF particles, and the excellent properties of /DVB/GMA microspheres are characterized by scanning electron microscopy (SEM), Brunauer-Emmett-Teller (BET) and X-ray photoelectron spectroscopy (XPS). The sorbent can extract illicit drugs via the reverse-phase interactions provided by benzene ring on the polymer backbone and the hydrogen bonding interactions provided by functional group (-NH-) on the COF particles. Based on using /DVB/GMA as sorbents, an easiness-to-handle of magnetic dispersive solid phase extraction (Mag-dSPE) procedure is proposed for the simultaneous preconcentration of 12 illicit drugs from wastewater. The obtained results show high extraction efficiency of /DVB/GMA to illicit drugs with recoveries between 81.6-116%. Furthermore, a liquid chromatography-tandem quadrupole mass spectrometry (LC-MS/MS) method for the determination of 12 illicit drugs from wastewater at sub-ppt levels has been proposed and validated with the pretreatment of samples by Mag-dSPE. The limits of quantification (LOQs) for the 12 illicit drugs are between 0.40 ng/L and 4.90 ng/L. Validation results on linearity, specificity, trueness and precision, as well as on application to the analysis of 12 illicit drugs in ten real samples demonstrate the applicability to environment monitoring analysis.

Abstract:The social and environment effects of illicit drug abuse are a growing global concern. Wastewater-based epidemiology (WBE) has become a common monitoring tool for assessing drug consumption. In this study, mechanical carrier (MGO), skeletal framework (ZIF-67) and functional modification (DES) were together prepared a three-dimensional nanocomposite (DES/ZIF-MGO) as an adsorbent, and developed an ultrahigh performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method coupled with optimal MSPE for the determination of ultra-trace illicit drugs in wastewater. The method exhibited good linearity ( R 2 ≥ 0.9985) with low limit of detection (0.02–1.55 μg/L) and qualified recovery (92.1–100.9 %) for four illicit drugs (AM, METH, MDMA and MC). The characterization results showed that electrostatic, hydrogen bond, and π-π interaction contributed to the adsorption process. The drug consumption in a city was investigated using WBE. MC was not detected in any of the wastewater samples. The detection rates of METH and MDMA were 100 % and 82 %, respectively, while AM was found in about 18 % of the wastewater samples. Thus, based on advantages of the DES/ZIF-MGO nanocomposite, the proposed method is a rapid, sensitive and reliable method for the determination of trace illicit drugs in wastewater.

Abstract:The detection of illicit drugs in wastewater can effectively monitor and evaluate the trend of illicit drug abuse. A novel mixed-mode cation exchange magnetic sorbent Fe3O4 @poly(ST/DVB/MA-COOH) was prepared and firstly applied as magnetically dispersed solid phase extraction material to efficiently, rapidly, and selectively extract 21 illicit drugs from wastewater. The selectivity of the sorbent was mainly attributed to the electrostatic interaction. The effects of Fe3O4 @poly(ST/DVB/MA-COOH) preparation and extraction conditions on the adsorption performance were thoroughly discussed. Among the 21 illicit drugs, the absolute extraction recovery values for 19 illicit drugs were greater than 80 % and the entire adsorption process could be achieved in one minute. Subsequently, the Fe3O4 @poly(ST/DVB/MA-COOH) sorbent combined with UHPLC-MS/MS was used to establish a quantitative method for the effectively extracted 19 illicit drugs in wastewater. The method had a good determination coefficient in the range of 0.2-200 ng/L and the limits of detection of the method were 0.03-0.67 ng/L. The spiked recovery values were in the range of 87.0-119.6 %. Finally, the method was successfully applied to the detection of 19 illicit drugs in wastewater samples and also compared with the commonly used SPE method. The obtained results indicate that Fe3O4 @poly(ST/DVB/MA-COOH) has great advantages in the detection of illicit drugs in wastewater.

Abstract:Herein, a magnetic borate‐functionalized MXene composite with multiple boronic affinity sites was fabricated by embedding Fe 3 O 4 nanoparticles with 4‐formylphenylboronic acid functionalized Ti 3 C 2 T x nanosheets and served as sorbent for the simultaneous extraction of catecholamines (CAs) in urine samples. The morphology and structure of the magnetic materials were investigated using scanning microscopy, vibrating sample magnetometer, X‐ray photoelectron spectrometer, and X‐ray diffraction. The introduction of polyethyleneimine can amplify the bonded boronic acid groups, thereby effectively improving the adsorption capacities for CAs based on the multiple interactions of boronic affinity, hydrogen bonding, and metal coordination. The adsorption performance was investigated using the kinetics and isotherms models, and the main parameters that influence the extraction efficiency were optimized. Under the most favorable magnetic solid‐phase extraction condition, a sensitive method for the analysis of CAs in urine samples was developed by combining magnetic solid‐phase extraction conditions with high‐performance liquid chromatography detection. The findings illustrated that the proposed approach possessed a wide linearity range of 0.05–250 ng/mL with an acceptable correlation coefficient (R 2 ≥ 0.9984) and detection limits of 0.010–0.015 ng/mL for the target CAs. The research not only provides a notable composite with multiple boronic affinity sites but also offers an effective and feasible measure for the detection of CAs in biological samples.

Automatic analytical approach for the determination of 12 illicit drugs and nicotine metabolites in wastewater using on-line SPE-UHPLC-MS/MS

Abstract:In this study, we developed a novel on-line solid phase extraction (SPE)-ultra-high-performance liquid chromatography tandem mass spectrometry (UHPLC-MS/MS)-based analytical method for simultaneously quantifying 12 illicit drugs and metabolites (methamphetamine, amphetamine, morphine, codeine, 6-monoacetylmorphine, benzoylecgonine, 3,4-methylenedioxymethamphetamine, 3,4-methylenedioxyamphetamine, cocaine, ketamine, norketamine, and methcathinone) and cotinine (COT) in wastewater samples. The analysis was performed by loading 2 mL of the sample onto an Oasis hydrophilic-lipophilic balance cartridge and using a cleanup step (5% methanol) to eliminate interference with a total run time of 13 min. The isotope-labeled internal standard method was used to quantify the target substances and correct for unavoidable losses and matrix effects during the on-line SPE process. Typical analytical characteristics used for method validation were sensitivity, linearity, precision, repeatability, recovery, and matrix effects. The limit of detection (LOD) and limit of quantification (LOQ) of each target were set at 0.20 ng/L and 0.50 ng/L, respectively. The linearity was between 0.5 ng/L and 250 ng/L, except for that of COT. The intra- and inter-day precisions were <10.45% and 25.64%, respectively, and the relative recovery ranged from 83.74% to 162.26%. The method was used to analyze various wastewater samples from 33 cities in China, and the results were compared with the experimental results of identical samples analyzed using off-line SPE. The difference rate was between 19.91% and -20.44%, and the error range could be considered acceptable. These findings showed that on-line SPE is a suitable alternative to off-line SPE for the analysis of illicit drugs in samples.
A Novel Biosensor Based on Au@Ag Core-Shell Nanoparticles for Sensitive Detection of Methylamphetamine with Surface Enhanced Raman Scattering.

Abstract:We describe a novel biosensing strategy for sensitive detection of methylamphetamine (MAMP) based on surface enhanced Raman scattering (SERS) by the mediation of spacing between 4-mercaptobenzoic acid (4-MBA) labeled Au@Ag core-shell nanoparticles (Au@Ag). To achieve a favorable SERS substrate, Au@Ag shell-core nanoparticle was synthesized with seeds growth method and well characterized by SEM, TEM and UV-vis spectrometer. The uniform Au@Ag shows an excellent dispersion ability for SERS detection. Under the optimized conditions, the novel biosensor shows a good logarithm linear correlation with the concentration of MAMP ranging from 0.5 ppb to 40 ppb (R-2 = 0.986), with a limit of detection at 0.16 ppb of MAMP (3 sigma). Furthermore, our biosensors hold an excellent selectivity, demonstrated by the negligible interference from the detection of other illicit drugs and metabolites. The concentrations determined with our biosensor from spiked MAMP in human urine sample fell within the same range with the results from mass spectrometry. This indicates that our sensor has a clear potential for the rapid detection of illicit drug in real samples.
Red blood cell storage age – what we know from clinical trials

Abstract:Red blood cell (RBC) transfusion has become one of the most commonly utilized life-saving therapies in medicine with approximately 12–16 million units transfused in the US per year [1,2]. Since storage solutions were developed 100 years ago, they have been modified to allow for increased storage duration, primarily to increase availability, especially in remote areas, such as during combat operations [3]. Currently in the US and Europe, RBCs can be stored up to 42–56 days based upon at least 75% of radiochromium-labeled cells remaining in the circulation at 24 h after infusion and hemolysis less than 1% [4,5]. As RBCs are stored over time, the metabolic and biochemical changes that occur have been coined ‘the storage lesion.’ To further improve the availability of RBCs, it is standard for blood distributors to transfer RBCs that are close to expiration at community hospitals to medical centers where utilization rates are higher, and for hospital blood banks to issue the ‘first-in, first-out’ inventory to reduce RBC outdating and waste [2,6]. A consequence of these policies focused on inventory management is that patients with increased severity of illness are transfused RBCs of increased storage duration compared to less acutely ill patients. It is important to recognize that as storage duration for RBC units has increased for logistical purposes, there has not been a commensurate evaluation of clinical efficacy or safety. As a result of the preferential allocation of older RBCs to sicker patients, there has been concern regarding the quality of RBCs after prolonged storage related to both efficacy and safety. This has led to a significant amount of research into the clinical relevance of the storage lesion with laboratory, preclinical, and clinical studies [4,7–9]. The results of these studies suggested biologic plausibility that the RBC storage lesion was potentially able to affect outcomes, especially in critically ill populations. These studies though are limited due to potential bias and higher-quality studies were needed to more accurately determine if RBC storage age affects outcomes. Consequently, a few randomized controlled trials (RCTs) have been completed, and a few are still underway. Abstract:Abstract: Amphetamines, as psychoactive drugs, are extensively abused in society and cause serious mental and physical disorders among young people. Furthermore, the extremely euphoric and excited sense of stimulant consumption leads to dramatic social problems. Determination of various analytes and related metabolites in the complex biological matrices at trace levels has made sample preparation an indispensable part of forensic sciences. According to the problems above, providing high sensitivity, solving some analytical problems like matrix effects in LCMS-MS, and needing a cleaner extract are remarkable aspects of novel sample preparation methods in drug analysis. Application of nanotechnology and carbon-based nanocomposites seems to bring the above properties in developed and novel sample preparation methods. This review will try to provide an overview of different carbonic nano adsorbents used in sample preparation methods of amphetamines and discuss their superiority over the other nanomaterials.

Abstract:A portable kit was demonstrated for rapid and reliable surface-enhanced Raman scattering (SERS) detection of drugs in human urine. This kit contains two sealed reagent tubes, a packet of standardized SERS substrates, and a mini Raman device. A 3 min pretreatment for separating amphetamines from human urine was developed with an extraction rate of >80% examined by ultraperformance liquid chromatography (UPLC). Simultaneously, highly reproducible two-dimensional (2D) gold nanorod (GNR) arrays were assembled by the use of methoxymercaptopoly(ethylene glycol) (mPEG-SH) capping. Thirty batches of GNR arrays produced the 1001 cm(-1) intensity of methamphetamine (MA) molecules with a relative standard deviation (RSD) of 7.9%, and a 21 × 21 μm(2) area mapping on a 2D GNR array produced a statistical RSD of <10%, implying an excellent reproducibility and uniformity. The detection limit of amphetamines in human urine was at least 0.1 ppm. Moreover, the portable kit was successfully used for detecting MA, 3,4-methylenedioxymethamphetamine (MDMA), and methcathinone (MC) in 30 volunteers' urine samples with various clinical natures, and the dual-analyte detection of MA and MDMA implied a good capability of multiplex analysis. UPLC examination and the SERS recovery test clearly indicated that our pretreatment procedure was sufficient to lower the high background signals caused by complex components in urine and demonstrated the practicability and the resistance to false positives, which is a vital problem for law enforcement applications. The excellent performance of our portable kit promises a great prospective toward a rapid, reliable, and on-spot analyzer, especially for public safety and healthcare.

Abstract:The author investigated how elderly women who recently moved to senior centers reacted in comparison to same-age persons who were either better off or worse off than were the former. The author expected 2 individual difference variables related to uncertainty about oneself—sense of control and social comparison orientation—to interact with social comparison direction to produce affect. Contrary to this expectation, the author observed only main effects of social comparison direction on affect. The results indicated that downward comparison produced more negative affect (and less positive affect) than did upward comparison, independently of participants' individual differences. The results provide some useful information on the social psychology of aging.
Tannic acid-directed synthesis of magnetic and boronic acid-functionalized metal-organic frameworks for selective extraction and quantification of catecholamines in human urine

Abstract:A¬†novel magnetic borate-functionalized metal-organic framework nanocomposite was designed and fabricated for selective enrichment of catecholamines from human urine. Firstly, the polytannic acid (PTA) layer with natural low-cost and ecofriendly polyphenol tannic acid as the organic ligand and Fe3+ as the cross-linker was coated onto the surface of Fe3O4. Then, the borate-functionalized metal-organic framework (MIL-100(Fe)-B) with 5-boronobenzene-1,3-dicarboxylic acid as a ligand fragment was modified onto the PTA-coated Fe3O4 through a metal-ligand-fragment coassembly strategy. The obtained smart porous adsorbent Fe3O4@PTA@MIL-100(Fe)-B was confirmed by means of several characterization methods and then applied as an effective magnetic solid phase extraction (MSPE) sorbent for specific extraction of trace catecholamines in human urine. The Plackett‚ÄďBurman design was used for screening the variables significantly affecting the extraction efficiency. Then, the significant factors were further investigated by the Box‚ÄďBehnken design to determine the optimal extraction conditions. Under the optimal conditions, a method for selective MSPE combined with high-performance liquid chromatography with a fluorescence detector for the quantitation of catecholamines in human urine was developed and validated. With the proposed method, the linearity range was from 0.500 to 500¬†ng¬†mL‚ąí1 for norepinephrine and epinephrine and from 1.00 to 500¬†ng¬†mL‚ąí1 for dopamine. The detection limits were 0.050, 0.11, and 0.20¬†ng¬†mL‚ąí1 for norepinephrine, epinephrine, and dopamine, respectively. The recoveries from spiking experiments varied from 91.5 to 108% with relative standard deviations (RSDs) of 0.80‚Äď4.8%. The established method is¬†rapid, sensitive, accurate, inexpensive, and ecofriendly and was successfully applied to the determination of the target catecholamines in human urine samples.

Abstract:In this study, a novel enrichment technique based on magnetic core-mesoporous shell microspheres with C8-modified interior pore-walls (C8-Fe3O4@mSiO2) was successfully developed for the determination of diazepam in rat plasma by LC-MS. Due to the unique properties of the synthesized C8-Fe3O4@mSiO2 microspheres (C8-modified magnetic mesoporous microsphere), small drug molecules like diazepam can enter the mesopore channels and be efficiently absorbed through hydrophobic interaction by interior C8-groups (Octyl functional groups). Large molecules like proteins are excluded from the mesopore channels as a result of size exclusion effect, leading to direct extraction of drug molecules from protein-rich biosmaples such as plasma without any other pretreatment procedure. Moreover, diazepam adsorbed C8-Fe3O4@mSiO2 microspheres could be simply and rapidly isolated through placing a magnet on the bottom of container, and then diazepam could be easily eluted from C8-Fe3O4@mSiO2 microspheres for further LC-MS analysis. Extraction conditions such as amounts of C8-Fe3O4@mSiO2 microspheres added, adsorption time, elution solvent and elution time were investigated. Method validations including linear range, the limit of detection, precision, and recovery were also studied. The results indicated that the proposed method based on C8-Fe3O4@mSiO2 microspheres was simple and accurate for the analysis of diazepam in the rat plasma. And it will provide new ideas for analyzing plasma concentration and pharmacokinetics of similar drugs.
Application of Magnetic Materials Combined with Echo® Mass Spectrometry System in Analysis of Illegal Drugs in Sewage

Abstract:The aim of this study is to solve the problems of the complicated pretreatment and high analytical cost in the detection technology of trace drugs and their metabolites in municipal wastewater. A high-performance magnetic sorbent was fsynthesized for the enrichment of trace drugs and their metabolites in wastewater to develop a magnetic solid-phase extraction pretreatment combined with the acoustic ejection mass spectrometry (AEMS) analytical method. The magnetic nanospheres were successfully prepared by magnetic nanoparticles modified with divinylbenzene and vinylpyrrolidone. The results showed that the linear dynamic range of 17 drugs was 1–500 ng/mL, the recovery was 44–100%, the matrix effect was more than 51%, the quantification limit was 1–2 ng/mL, and the MS measurement was fast. It can be seen that the developed magnetic solid-phase extraction (MSPE) method is a good solution to the problems of the complicated pretreatment and analytical cost in the analysis of drugs in wastewater. The developed magnetic material and acoustic excitation pretreatment coupled with mass spectrometry analysis method can realize the low-cost, efficient enrichment, and fast analysis of different kinds of drug molecules in urban sewage.

Abstract:Wastewater-based epidemiology (WBE) has become an objective and updated surveillance strategy for monitoring and estimating consumption trends of psychoactive substances (PSs) in the population. Firstly, magnetic shrimp shell biochar-based adsorbent (DZMBC) was synthesized and employed for extraction trace PSs from municipal wastewater. Proper pyrolysis temperature and increased KOH activator content favored the pore structure and surface area, thus facilitating extraction. DZMBC delivered exceptional extraction performance such as pH stability, anti-interference property, fast magnetic separation ability, reusability, and reproducibility. Then, a method based on magnetic solid-phase extraction (MSPE) followed by ultra-high-performance liquid chromatography–tandem mass spectrometry (UPLC–MS/MS) was developed, validated, and utilized for the quantitative determination of five PSs in real wastewater samples. Methodological validation results indicated a favorable linearity, low method limits of detection (1.00–4.75 ng/L), and good precisions (intra-day and inter-day relative standard deviations < 4.8%). Finally, an objective snapshot of Chongqing drug use and consumption pattern was obtained. Methamphetamine (MAMP) and 3,4-methylenedioxymethamphetamine (MDMA) were the prevalent illegal drugs in local. Both concentrations and per capita consumption of MDMA displayed a change ( P < 0.05) between July and September, while no statistical differences were observed for each week.
