First report of Colletotrichum scovillei causing anthracnose fruit rot of pepper in Ohio

Abstract:In August 2019, the Ohio State University Vegetable Pathology laboratory received multiple bell and banana pepper fruits (Capsicum annuum, cvs. unknown) from Columbiana County, Ohio. The grower reported a disease incidence of 100% and severity of 70% in fruits across their pepper fields. Fruit lesions were brown, sunken, and covered with orange-colored sporulation. On banana peppers, the lesions mainly affected the blossom end of the fruits, while the lesions were distributed randomly on bell pepper fruits. Pieces of diseased tissue were cut from the fruit and surface sterilized in 0.5-0.6 % sodium hypochlorite, rinsed in sterile water, blotted dry, and placed on potato dextrose agar. All of the fungal cultures recovered were cottony, pale gray-green with shades of orange on the underside of the mycelial mat. Two representative isolates, SM209-19 (bell pepper) and SM210-19 (banana pepper), were grown on oatmeal agar to induce sporulation. Pink-orange concentric rings containing acervuli and conidia were present on the oatmeal agar plates after one week of growth at 22◦C (12-h dark/light). Conidia (n=29) were hyaline, aseptate, cylindrical in shape, and had an average length of 10.5 µm (std. dev. = 1.3 µm) and width of 4.1 µm (std. dev. = 0.6 µm) (Fig.1). DNA was extracted from both isolates using a DNeasy Plant Kit (Qiagen Inc, Germantown, MD), and partial sequences of the internal transcribed spacer (ITS) region, -tubulin 2 gene (TUB2), and glyceraldehyde-3-phosphate dehydrogenase gene (GDPH) were amplified by PCR with the following primers: ITS4/ITS5 (White et al. 1990), Bt1a/Bt1b (Glass et al. 1995), and GDF1/GDR1 (Guerber et al. 2003), and squenced. The ITS region of both isolates SM209-19 and SM210-19 (PP280815 and PP280816, respectively) showed 100% identity with C. scovillei (Cs) isolate LJTJ35 (KP748226). The partial sequences of GDPH, (PP320348, PP320349, respectively) showed 99% sequence identity with the Cs CBS 126528 (JQ948597) and 100% identity with Cs HP1 (MT826948) The partial sequences of TUB2 (PP472464 and PP472465, respectively) had 100% sequence similarity with the Cs HP1 and Cs CBS 126528 (MT826951, JQ949918 respectively). Pathogenicity was tested in a greenhouse experiment on blossoming bell pepper plants (cv. Carmen) by spraying 10 ml of 1 X 105 conidia/ml suspension onto flower blooms (nine plants per isolate). Control pepper plants were mock inoculated by spraying 10 mL of sterile deionized water. The plants were re-inoculated one week later. Brown, sunken lesions with orange sporulation developed on the fruits of inoculated plants 21 days after the initial inoculation (Fig. 2), while the mock-inoculated plants did not produce any symptoms. Culturing from symptomatic fruits on PDA, following the same method described above, produced fungal colonies with the same morphological traits previously described. C. scovillei causing anthracnose on pepper has been described in the US (Toporek et al. 2021), Brazil (Caires et al. 2014), China (Zhao et al. 2016), and different South Asia Countries (Khalimi et al. 2019). Open-field peppers are produced in Ohio on more than 5,400 acres, with a value of more than $53 million, with anthracnose being one of the most severe fungal diseases reducing yield. This newly reported Colletotichum species could represent a further threat for this crop. Further studies evaluating fungicide sensitivity and efficacy against this pathogen will be of crucial importance for disease management.

First Report of Fruit Rot Caused by Fusarium incarnatum-equiseti species complex on Greenhouse Bell Pepper in Malaysia

Abstract:In Malaysia, bell pepper (Capsicum annuum var. grossum), also known as sweet pepper or paprika, is one of the highly imported vegetable crops. In 2021 alone, Malaysia imported nearly 74 thousand metric tons of its chilies, including bell peppers, from other countries (DOSM, 2022). Often, farmers grow the bell peppers in moderate to cool conditions within highland regions for local commercial purposes. In June 2022, the Malaysian Agricultural Research and Development Institute (MARDI) in Serdang, Selangor, conducted a research study to grow lowland bell peppers under a glasshouse rain protection system. A disease inspection carried out found fruit rot on approximately 30% of mature bell pepper fruits in the greenhouse. Symptoms appeared as firm and sunken black lesions covered with white to light pink spore masses on the outer surface, which eventually fell off. Infected fruit parts were disinfected with 10% hypochlorite (NaOCl) for 2 min, followed by double washing with sterile distilled water, air-dried, and placed onto potato dextrose agar (PDA). After 3 days of incubation, the fungal colonies that grew from the symptomatic tissue pieces were transferred onto new PDA to obtain pure cultures. The pure fungal colony appeared dense, whitish aerial mycelium that slowly became cream to pinkish-orange after 7 days of incubation at room temperature (25±2 °C). To examine the morphology features, the pure cultures were subbed onto carnation leaf agar (CLA) and incubated at 25±2°C for 14 days. Macroconidia were abundant, slightly curved with tapered apical cells, 3- to 5-septate, and ranged between 21.8 and 34.0 x 3.0 and 5.1 μm. Microconidia were single-celled, often 1-septate, and ranged between 10.0 and 12.6 x 2.1 and 3.4 μm. Chlamydospores were globose and in chains. The fungus was identified as Fusarium sp. according to Fusarium key by Leslie and Summerell (2006). PCR amplification and DNA sequencing were performed using primers EF1F/EF2R and ITS1/ITS4 (O'Donnell et al., 1998; White et al., 1990) to amplify the partial elongation factor 1-alpha (TEF1-α) gene and internal transcribed spacer region (ITS), respectively. The TEF1-α and ITS sequences of this isolate were deposited in GenBank as OQ672911 and OR349657. BLAST analysis with TEF1-α gene sequences revealed 99.74% and 99.33% sequence identity with F. pernambucanum (accession no. ON330424) and Fusarium isolate NRRL 25134 (accession no. JF740755), respectively; both belonged to the Fusarium incarnatum-equiseti species complex (FIESC). BLAST search of the TEF1-α sequence in the database of the International Mycological Association (www.mycobank.org) showed 99.18% identity with FIESC (NRRL 36548). The ITS sequences were 100% identical to those of F. incarnatum (MT563420, MT563419, and MT563418). Pathogenicity test was conducted on three unwounded and three wounded mature red bell pepper fruits (SP299 Red Masta variety). Two healthy bell peppers were used as controls for each treatment. Prior to inoculation, the fruits were surface-sterilized by dipping in 70% ethanol and rinsed twice with sterile distilled water. Unwounded fruits were inoculated with fungal mycelium disks (5 mm diameter), whereas control fruits were inoculated with sterile PDA agar disks. For wound method, 6 µl of spore suspension (1x106 spores/ml) was obtained from 7-day-old cultures and injected (1 mm depth) into the fruit wall using a sterile syringe needle. Control fruits were inoculated with sterile distilled water only. Each fruit was inoculated with the inoculum at three distinct spots and kept in a humid chamber at a temperature of 25±2 °C. The pathogenicity test was done twice. Five days post-inoculation, the control fruits showed no symptoms, whereas all inoculated wounded and non-wounded fruits developed necrotic lesions with white mycelium growing on the inoculation points. The pathogen was successfully re-isolated from the infected fruits and morphologically identified as FIESC, fulfilling Kochs postulates. It has been reported previously that the members of FIESC are responsible for the fruit rot of bell peppers under greenhouse conditions (Ramdial et al., 2016). To the best of our knowledge, this is the first report of FIESC causing fruit rot on greenhouse bell peppers in Malaysia. This fruit rot disease may impose significant constraints on bell pepper production in Malaysia; hence, effective strategies to control the pathogen and prevent disease dispersal should be implemented.

First report of Botryosphaeria dothidea as the causal agent of a new fruit rot disease of pepper in China

Abstract:Pepper is an important and widely cultivated economic vegetable in the world (Yin et al., 2021). In June 2021, approximately 25% to 33.3% of the pepper plants had rot disease symptoms in Zhuanghang Comprehensive Experimental Base (30.894829 °N, 121.391374 °E), Fengxian district, Shanghai city, China. Water-soaked spots appeared on fruits that increased in size and leading to smelly fruit decay. To isolate the pathogen, three pepper samples with severe symptoms were collected. The samples were surface disinfected with 70% ethanol for 30 sec, 10% chlorine bleach for 10 min, rinsing with sterile water for three times and the rot tissues were cut and dried on sterile filter paper. The dried paper was later placed on potato dextrose agar (PDA) medium and incubated at 28°C (Tang et al., 2021). After 2-3 days, four types of colonies with different colony appearances were observed, in which only one can induce fruit rot phenotype (data not shown). Four isolates were cultured for molecular identification in each type. ITS1/ITS4, T1/βt-2b and EF1-526F/EF1-1567R primers were used to amplify the internal transcribed spacer region (ITS), the beta-tubulin (TUB2) and the translation elongation factor I alpha (EF1-α) genes, respectively (Chen et al., 2018) and corresponding sequences from the isolates were analyzed with BLAST. Sequences of the isolate which can induce pepper decay were submitted to GenBank under the accession numbers of OM663701 (ITS), OM720127 (TUB2) and OM720128 (EF1-α). The results showed that the pathogen had 99% sequence homology to most strains of Botryosphaeria dothidea (B. dothidea) and displayed the highest sequence similarity to strain LBSX-1 (ITS: KF55123), strain JGT01 (TUB2: MW202404) and isolate CZA (EF1-α: MN025271). Based on molecular characterization, the isolate was identified as B. dothidea isolate SH01. A phylogenetic tree was constructed using Maximum Parsimony (MP) methods by MEGA7, and showed that SH01 was closely related to isolate CMW9075. To confirm the pathogenicity, five healthy pepper fruits were surface sterilized, 500μl of conidial suspension (1×103 conidia/ml) were injected into pepper (sterilized distilled water as control). Six days post inoculation (dpi), fruit rot symptoms appeared and the pepper decayed at 12 dpi. Four days post inoculation with mycelium plugs (from a 4-day-old culture on PDA, PDA plugs as control), hyphae were observed in the inoculation site and B. dothidea was re-isolated from the symptomatic areas, thus fulfilling Koch's postulates (Back et al., 2021, Chen et al., 2020). The pepper rotted severely at 7 dpi. The colonies of SH01 were pale to white and gradually turned into gray in 4-6 days. Conidia of the pathogen were unicellular, aseptate, hyaline and fusiform to fusoid, with dimensions of 19.7-23.5 μm × 3.8-5.2 μm (average = 21.9 μm × 4.8 μm, n = 50). Hyphae were transparent, branched and composed of multiple cells. The characteristic was consistent with the descriptions of B. dothidea (Vasic et al., 2013). B. dothidea belongs to Botryosphaeriaceae, causing widespread diseases in many plant species, commonly associated with cankers and dieback of woody plants and economic crops, such as plumcot trees (Back et al., 2021), eucalyptus (Yu et al., 2009) and soybeans (Chen et al., 2020) in China and Korea. Our findings reported for the first time that B. dothidea (SH01) can induce the pepper rot disease and future work on its pathogenesis may provide strategies for disease control against this fungus.
Identification of the Pathogen Inducing Fruit Soft Rot of Melon

Abstract:Soft rot with symptoms of initial brown spots on the surface and soft rot inside of melon (Cucumis melo L.) was often reported in Yiwu and other areas, Zhejiang province. The infected melon fruit corrupted consequently. In some areas this disease caused significant yield loss to melon industry. 4 isolates (M1 to M4) with two types of morphology of bacterial colonies were isolated and purified from infected melon fruit. The tests of hypersensitive reaction in tobacco (Nicotiana tabacum L.) and pathogenicity in the melon fruit for these isolates demonstrated that M1 and M3 were pathogenic, whereas M2 and M4 were not. Both M1 and M3 were identified to be an isolate of Erwinia carotovora subsp. carotovora based on their properties of morphology, flagella, gram reaction, host range and the sequence alignment of 16S rRNA gene. In addition, inoculation test showed that this pathogen could induce soft rot to tomato, cucumber, watermelon and pumpkin fruits.
First Report of Globisporangium ultimum Causing Pythium Damping-off and Root Rot on Pepper in Guizhou, China

Abstract:Pepper (Capsicum annuum L.) is a popular vegetable and condiment consumed around the world. In the Guizhou Province of China, peppers are the most commonly grown crop on 300,000 planted hectares. A variety of diseases routinely occur on peppers in this province, resulting in yield losses (Liu et al., 2022). Root rot is one of the most common symptoms and produces poor root growth and wilting of pepper. In April 2023, symptomatic pepper plants displaying stunting, dwarfism, wilting, and root browning were collected from five fields in Guizhou, with disease incidence ranging from 10% to 20%. The collected rotten roots were cleaned with sterilize distilled water and placed in selective V8 juice agar (V8A) medium (15% clarified V8 juice with 2.5 g/L CaCO3 and 2% agar) containing nystatin, ampicillin, rifampicin, and miconazole, and incubated at 25℃ for 1 to 2 days (Morita and Tojo, 2007). Eight isolates with similar colony morphology were transferred to V8A medium via hyphal tipping, and incubated at 25℃ in the dark. Colony and sexual structures were observed using a microscope. Mycelium was aseptate and formed white cottony colonies. Globose, intercalary, or terminal hyphal swellings were observed with a diameter of 20.5 to 25 µm (average: 22 µm), and aplerotic oospores had a diameter of 15 to 20 µm (average: 17.5 µm) with a wall thickness of approximately 2 µm. Three representative isolates HSLJ-3, LJG-1, and LJY-2 were chosen for further molecular identification. Sequences of the internal transcribed spacer (ITS) and mitochondrial cytochrome c oxidase subunit 1 (cox1) genes were identified using primer sets ITS4/ITS5 (White et al., 1990) and OomCoxI-Levup/OomCoxI-Levlo (Robideau et al., 2011), respectively. All sequences were deposited in GenBank (accession nos. OR554005, PP083310, and PP083420 for ITS, and OR529247, PP093821 and PP093822 for cox1). BLAST analysis revealed all ITS and cox1 sequences exhibited 100% identity with Globisporangium ultimum (Pythium ultimum) isolate BR850 (GenBank accession nos. HQ643892.1 and HQ708933.1 for ITS and cox1, respectively). Phylogenetic analysis was performed by the maximum-likelihood method on the CIPRES web portal (https://www.phylo.org/portal2/login!input.action, accessed on 9 January 2024). For pathogenicity tests, each isolate was cultured in V8A medium containing 50 autoclaved wheat seeds at 25℃ for 7 days. Budding pepper seedling (cv. Huaxi) was transplanted into a 0.4 L pot containing sterilized commercial potting mix (Seedling Cultivation Substrate, Hunan Xianghui Agricultural E-commerce Co., Ltd.) which was saturated with deionized water. Eight infected and non-infected wheat seeds were placed near the roots of five pepper seedlings, respectively. Plants were placed in an artificial climate chamber, with a 14 h photoperiod and approximately 75% relative humidity at 25℃. After 14 days, inoculated seedlings showed symptoms of stunting, wilting, and rotting roots similar to those observed in the field. No disease was observed on the non-inoculated control plants. The pathogen was isolated from infected pepper roots and confirmed as G. ultimum by morphological and molecular analyses as previously described. This is the first report of G. ultimum causing root rot on pepper in Guizhou, China. This finding is critical to the discover of treatment options for this pathogen, thereby improving management practices to reduce yield losses in pepper.

In July 2018, an unknown disease occurred on 5 to 10% of 5,000 postharvest Cuiguan pear fruit (P. pyrifolia) after 15 days in our laboratory storage room (20°C, 85 to 90% relative humidity) in NanChang City (28.68° N, 115.85° E), Jiangxi Province. Initial symptoms were shallow black spots on the fruit surface that later enlarged into circular brown lesions with water-soaked margins and a lesion diameter of 20 to 40 mm. White hyphae grew on the pear lesions and, slowly, infected fruit became soft and necrotic. To isolate the suspected pathogen, small sections (5 × 3 mm) of diseased fruit were sterilized with 75% ethanol for 30 s followed by 0.1% mercuric chloride solution for 5 min, rinsed three times with sterile water, and then transferred onto potato dextrose agar (PDA) and incubated at 28°C for 5 days. Finally, five isolates were obtained and purified on PDA. The isolates produced abundant aerial mycelium, which was initially white and then became purple over time at 28°C. The average growth of mycelium was 5.4 mm per day (n = 5). On carnation leaf agar (Fisher et al. 1982), macroconidia were three to five septa, moderately curved to straight, and 28.7 to 53.2 × 3.2 to 5.3 µm in size (38.4 × 4.2 µm on average, n = 25). On PDA, the microconidia were almost straight, aseptate, and 6.3 to 11.4 × 2.1 to 4.0 µm (8.3 × 2.9 µm on average, n = 25), and no chlamydospores were observed. The morphological features were consistent with Fusarium proliferatum (Leslie and Summerell 2006). In addition, molecular phylogenetic analyses were used to define the isolate identity. The universal primers ITS1/ITS4 (White et al. 1990) and F. proliferatum-specific primers PRO1/PRO2 (Mulè et al. 2004) were used to amplify the internal transcribed spacer (ITS)-5.8S ribosomal DNA region and calmodulin gene. The PCR amplicons (accession nos. MK158221 and MK144327) aligned with GenBank showed 100% similarity with F. proliferatum (accession nos. KU891521.1 and LT841258.1). Thus, the pathogen was identified as F. proliferatum based on morphological and molecular characteristics. Pathogenicity tests were conducted by inoculating nine disease-free pear fruit after disinfesting in 75% ethanol. A prepared spore suspension (10 μl, 1.0 × 106 spores/ml) was dropped on the wounds (a depth of 1 mm with a 1-mm diameter) on the pear surface, which were made by a sterilized needle. Sterile water (10 μl) was dropped on the wounded sites of nine pear fruits as negative controls, and 10 μl of a prepared spore suspension was dropped onto nine nonwounded control pears. All fruits were incubated in a growth chamber at 30/26°C (day/night, 12-h photoperiod, 90% relative humidity). After 3 days, the first symptom of shallow black spots was observed on nine wounded inoculated pears, and after 5 days mean lesion diameter was 19.8 mm, whereas the symptom of nonwounded pears was mild. The water controls remained healthy. To complete Koch’s postulates, F. proliferatum was again isolated from infected pears. To our knowledge, this is the first report of F. proliferatum causing rot on Cuiguan pear in China and around the world. Abstract:Bell pepper (Capsicum annuum) is a widely grown vegetable crop that is nutritious and flavorful and economically important for growers worldwide. A significant limiting factor in the postharvest storage and long-distance transport of peppers is gray mold caused by Botrytis cinerea. The pathogen is widespread in nature, highly aggressive, and able to cause disease at cool refrigerated temperatures during transport and storage. Fungicides have been relied on in the past to reduce bell pepper rots in storage; however, concern over residues on the fruit and environmental degradation have heightened the importance of natural and generally recognized as safe (GRAS) solutions that effectively limit disease. Essential oils, plant extracts, inorganic chemicals, biocontrols, defense activators, hot water treatments, and modified storage conditions have been tested to reduce losses from gray mold. Despite significant amounts of research on natural methods of control of B. cinerea postharvest, research specific to gray mold in peppers is limited. The objective of this review is to summarize the research conducted with environmentally friendly alternatives to chemical fungicides to control this important pathogen of peppers postharvest. To ensure a steady supply of healthy and nutritious produce, more research is needed on the development, use, and application of non-hazardous Botrytis control methods. Until an effective solution is found, using a combined approach including environmental controls, sanitation, and GRAS products remain paramount to limit Botrytis fruit rot of peppers postharvest.
Integrated proteomic and metabolomic analyses reveal significant changes in chloroplasts and mitochondria of pepper (Capsicum annuum L.) during Sclerotium rolfsii infection

Abstract:Infection by Sclerotium rolfsii will cause serious disease and lead to significant economic losses in chili pepper. In this study, the response of pepper during S. rolfsii infection was explored by electron microscopy, physiological determination and integrated proteome and metabolome analyses. Our results showed that the stomata of pepper stems were important portals for S. rolfsii infection. The plant cell morphology was significantly changed at the time of the fungal hyphae just contacting (T1) or surrounding (T2) the pepper. The chlorophyll, carotenoid, and MDA contents and the activities of POD, SOD, and CAT were markedly upregulated at T1 and T2. Approximately 4129 proteins and 823 metabolites were clearly identified in proteome and metabolome analyses, respectively. A change in 396 proteins and 54 metabolites in pepper stem tissues was observed at T1 compared with 438 proteins and 53 metabolites at T2. The proteins and metabolites related to photosynthesis and antioxidant systems in chloroplasts and mitochondria were disproportionally affected by S. rolfsii infection, impacting carbohydrate and amino acid metabolism. This study provided new insights into the response mechanism in pepper stems during S. rolfsii infection, which can guide future work on fungal disease resistance breeding in pepper.

Abstract:Eight-days-old mouse embryos were transferred to the subcutaneous tissue of the dorsal skin of host mice. A high rate of embryos developed into hemorrhagic (HN) or nonhemorrhagic nodules (NHN). The latter had trophoblastic cells as well as embryoblastic derivatives whereas HN contained almost only trophoblastic cells. At least two kinds of trophoblastic cells were present in NHN: small cells and large cells similar to trophoblastic giant cells. In HN most trophoblastic cells arranged themselves into a network whose meshes contained host blood. Although embryoblastic derivatives such as endoderm and Reichert's membrane were apposed to host connective tissue cells, trophoblastic cells were always surrounded by collagen fibrils or by a layer of an amorphous material.

Abstract:Phytophthora blight, caused by Phytophthora capsici , is a devastating disease threatening the bell pepper industry. This pathogen affects all parts of plants including causing rot, blight, significant harm and losses the Capsicum species. In the present study, symptoms of blight and damping off were noted during two years of field surveys, February–November 2021–2022. Seventy-two isolates were obtained from rhizosphere soil samples of the infected bell pepper ( Capsicum annuum ) plants. Phytophthora capsici Leonian was identified on the basis of morphological and cultural characteristics and culture being deposited in the Indian Type Culture Collection (ITCC) in New Delhi. The 18S rRNA ITS region was undertaken to confirm species identity. A phylogenetic tree confirmed that the obtained ITS region sequences were identical to Phytophthora capsici isolates. This is the first report of Phytophthora blight on bell peppers from the Haryana region of India.

Abstract:Phytoplasma infects a wide variety of crops, causing considerable economic losses. About half of the vegetable crops damaged by phytoplasma belong to the Solanaceae family including tomato, eggplant and pepper which play an important role in the agriculture economics of Moldova. Our previous research confirmed the presence of ‘Candidatus Phytoplasma solani’ (16SrXII-A subgroup) in tomatoes and also identified insect vectors. In this communication, we present for the first time in Moldova the results of molecular diagnosis of association of ‘Ca. P. solani’ in 4% of the analyzed sweet pepper samples. ‘Ca. P. asteris’ group was absent in the pepper field.

First Report of Powdery Mildew of Greenhouse Pepper Caused by Leveillula taurica in Canada

Abstract:In 1999, powdery mildew on cvs. Oberon and Triple 4 of greenhouse pepper (Capsicum annum L.) and cv. FireFlame of chili pepper was reported simultaneously in two commercial greenhouses at two separate locations, Leamington and Vineland, geographically separated by 290 km, in southern Ontario. Losses of 10 to 15% each in the 2 and 3.1 ha greenhouse pepper operations were noted. The greenhouse pepper industry in Canada consists of 89.4 ha with sales of $43.6 million (Canadian). Lesions appeared as a white, powdery coating on the abaxial leaf surface only, generally on the lower foliage of pepper plants, while diffuse chlorotic spots were present at corresponding locations on the adaxial surface. In chili pepper, this chlorosis was restricted to interveinal tissue, causing the leaves to have a somewhat netted appearance. In some cases, pale yellow spots appeared on the adaxial portion of the affected foliage during later stages of disease development. No cleistothecia were observed. Pyriform and cylindrical, hyaline, single-celled conidia were present in equal numbers. Both conidia had a network of crests and granules containing tiny, thornlike projections on the surface. These projections were also evident on the lower half of conidiophores. Pyriform conidia ranged in length from 61.6 to 84.0 μm (mean = 70.8, SE = 0.7) and in width from 14.0 to 25.8 μm (mean = 21.1, SE = 0.4, n = 50), with a mean length to width ratio of 3.4. Cylindrical conidia ranged in length from 54.9 to 80.1 μm (mean = 66.3, SE = 0.9) and in width from 15.7 to 24.1 μm (mean = 18.6, SE = 0.3, n = 50), with a mean length to width ratio of 3.1. Short conidial chains borne on conidiophores consisted of a pyriform conidium first followed by cylindrical conidium. Stomatal penetration and extensive endophytic mycelial growth in the mesophyll layer were evident in foliar tissue cleared with glacial acetic acid:EtOH (1:2) and stained with lactophenol cotton blue. To confirm pathogenicity, conidia from infected pepper leaves were dusted onto the water-misted abaxial leaf surface of 41-day-old greenhouse pepper cvs. Cubico and Edison, and onto the similarly treated adaxial leaf surface of 58-day-old field pepper cv. Renegade. Plants were enclosed in plastic bags for 24 h on a bench. Control plants were treated identically but not inoculated. Inoculated plants developed foliar powdery mildew symptoms, including sporulation similar to that of naturally infected plants. Occurrence of the fungus on the abaxial surface of pepper foliage, presence of endophytic mycelium, and the morphological characteristics of the imperfect state confirm the identity of the fungus. First Report of Chili Anthracnose Caused by Colletotrichum sojae in South Korea

Abstract:Chili (Capsicum annuum L.) is an economically important crop worldwide, valued for its culinary uses. In South Korea, anthracnose caused by Colletotrichum spp. including C. truncatum, C. gloeosporioides, C. coccodes, C. acutatum, and C. scovillei incurs on substantial economic loss (Kim et al. 2008; Oo and Oh 2020). In August 2022, somewhat different types of symptoms that was not typical on chilli fruits were observed in a field in Yereonggwang (GPS: 35.2579° N, 126.4742° E), South Korea. The disease symptoms appeared as sunken, necrotic lesions with dense black spore masses forming in concentric rings. The estimated disease incidence the 0.2 ha field showing up to 1% of fruits affected. To isolate the pathogen, six symptomatic chilli fruits were collected. Small pieces (5 mm²) were cut from the margins of the lesions, surface-sterilized in 70% ethanol for 30 sec, followed by 1% sodium hypochlorite for 1 minute, and then rinsed three times in sterile distilled water. The tissue pieces were placed on potato dextrose agar (PDA) plates and incubated at 25°C in the dark. After 3 to 5 days, emerging fungal colonies were sub-cultured to obtain pure isolates. A total of five isolates were obtained and initially identified as Colletotrichum spp. based on morphological characteristics. Seven-day old colonies were initially white, turning light orange with age on PDA. Setae (observed on lesion) were dark brown, verruculose and septate. Conidia were cylindrical, hyaline, and measured 14.8 to 19.9 × 4.2 to 6.5 µm (mean 16.7 × 5.6 μm, n = 70) in size; appressoria were brown to dark brown and irregularly shaped. These morphological characteristics of the isolates agree with those reported for the morphology of C. sojae by Damm et al. (2019). To confirm the identity of the isolates, DNA was extracted and specific gene regions were amplified and sequenced using the following primer sets: ITS (ITS1 and ITS4), GAPDH (GDF1 and GDR1), ACT (ACT-512F and ACT-783R), TUB (T1 and Bt2b), HIS3 (CYLH3F and CYLH3R), and CHS-1 (CHS-79F and CHS-345R). The resulting sequences were deposited in the NCBI GenBank with accession numbers (LC830742 to LC830766). Maximum likelihood phylogenetic analysis using combine sequences of ITS, GAPDH, ACT, TUB, HIS3 and CHS-1 in MEGA X confirmed the isolates as C. sojae, marking the first report of this pathogen on chilli in South Korea, previously known to infect soybean. Pathogenicity tests were conducted on wound and nonwounded healthy and mature-green chili fruits (cv. Bicksita) to confirm the pathogenicity of the isolated C. sojae. The fruits were surface-sterilized using 70% ethanol and then rinsed with sterile distilled water. The fruits were wounded using a sterile needle to facilitate infection. A conidial suspension (1x106 conidia/mL) was prepared from 7-day-old PDA cultures. Each fruit was inoculated by placing a 10 µL drop of the conidial suspension onto the wounded and nonwounded sites (4 to 5) of the wound and unwound fruits, respectively. Control fruits were inoculated with sterile water. A total of 40 fruits per treatment were used and the experiment repeated twice. The fruits were placed in plastic box lined with moist paper towels to maintain high humidity and incubated at 25°C. Anthracnose symptoms developed on the inoculated fruits within 7 days, while control and unwounded fruits remained symptom-free. Colletotrichum sojae was successfully reisolated from the symptomatic fruits, fulfilling Koch's postulates and confirming its role as the causal agent of the disease. Colletotrichum sojae is known to infect Fabaceae species worldwide such as Glycine max, Medicago sativa, Phaseolus vulgaris, Atractylodes ovata and Vigna unguiculata (Damm et al. 2019; Talhinhas and Baroncelli 2021), Atractylodes ovata in South Korea (Hassan et al. 2021) and chili pepper in China (Zhanget al. 2023). The first report of C. sojae causing chili anthracnose in South Korea represents a new challenge for chili growers. Integrated disease management strategies need to be developed and implemented to mitigate its impact.

A Tomato Fruit Rot Caused by Trichothecium roseum in Brazil

Abstract:Fruit rots caused by distinct fungal pathogens are commonly observed on tomatoes (Solanum lycopersicum L.) throughout all major production areas in Brazil. Samples of fruits displaying white mycelial growth associated with a profuse salmon-color sporulation were collected in greenhouse-grown tomatoes in Brasília-DF in February 2011. The isolated fungus displayed pink-to-white colonies containing several conidiophores with conidia. Mycelia displayed hyaline hyphae as much as 4 μm in diameter; conidiophores were simple or branched, 112 to 300 (360) μm long, and 2 to 4 μm wide. Conidia were produced in basipetal chains (frequently clustered), were ellipsoidal to pyriform with oblique and prominent truncate basal scars, two-celled, hyaline, and (14-) 16 to 26 (-28) × (6-) 7 to 10 (-12) μm. These characteristics allocated the specimen to Trichothecium roseum (Pers.). Koch's postulates were fulfilled for one fungal isolate by either spraying 10 intact fruits or by placing a drop of a spore suspension (adjusted to 105 conidia/ml) into three to five wounds created on 10 mature fruits of each of two tomato cultivars (Santa Clara and Dominador) by puncturing each fruit with a sterile needle. Five fruits of each cultivar were treated with sterile water as the mock-inoculated control treatment. Identical symptoms to those of the original fruit were observed only in the T. roseum-inoculated samples 5 to 7 days after using both inoculation procedures. Total DNA was extracted from a pure colony of the fungus growing on potato dextrose agar medium and used as template in PCR assays with the internal transcribed spacer (ITS)-4 (5'-TCCTCCGCTTATTGATATGC-3') and ITS-5 (5'-GGAAGTAAAAGTCGTAACAAGG-3') primer pair (2). A single amplicon of approximately 630 bp was observed and directly sequenced. Sequence analysis of the Brazilian isolate (GenBank No. JN081877) indicated identity levels of 99% with T. roseum isolates reported on Leucadendron xanthoconus in South Africa (GenBank No. EU552162) and isolates from strawberry fruits in South Korea (GenBank No. HM355750). However, phylogenetic analysis was unable to discriminate isolates of T. roseum from Passalora (GenBank No. EF432764) and Fusarium (GenBank No. GU183369) isolates, confirming the low genetic variability of the ITS region in Hypocreales (3). T. roseum has been reported to be infecting greenhouse tomatoes in the United States (4) and causing postharvest disease of tomatoes in Argentina (1). First Report of Cladosporium tenuissimum causing spot diseases on leaves and fruits of cucurbits in Brazil

Abstract:Cucurbitaceae crops are widely cultivated in the Northeast region of Brazil, which is the biggest producer of melon and watermelon in the country (Oliveira, 2020). Between November and December 2020 leaves of pumpkins (Cucurbita maxima L.) and watermelon (Citrullus lanatus L.), and leaves and fruits of melon plants (Cucumis melo L.) were collected with moderate to severe necrotic, irregular, and brown lesions from farms in the state of Rio Grande do Norte, Brazil. Fragments of diseased tissues were cut into small pieces and surface disinfested in 70% ethanol for 30 seconds, then in 2% sodium hypochlorite for 1 minute, and washed in sterile distilled water. Disinfested pieces of tissue were plated on potato dextrose agar (PDA) and incubated for seven days in the dark at 28 ± 2 °C. A total of 12 fungal isolates (four from pumpkins, one from watermelon, and seven from melons) were isolated from leaves and symptomatic fruits. All isolates in this study shared similar morphological characteristics. The colonies were dark gray to olive green in color with a velvety texture and surrounded by gray-white hyphae. The conidiophores were erect, tall, dark, and irregularly branched at the apex containing dark conidia, with 0 to 3 septa, variable in shape and size, forming chains that were often branched, globose, or subglobose with 3 to 4.5 μm in diameter. DNA from each isolate was extracted using the SDS method (Smith et al., 2001) and submitted to PCR amplification of the ITS and TEF1α regions with the primers ITS1/ITS4 (White et al. 1990) and EF1-728F/EF1-986R (Carbone and Kohn 1999), respectively. The amplicons were sequenced and deposited in GenBank: ITS (OP493545-OP493556) and TEF1α (OP536836-OP536847). Blastn analysis of the ITS and TEF1α partial sequences revealed that all 12 isolates belong to the species Cladosporium tenuissimum, with 100% nucleotide similarity with sequences of many C. tenuissimum isolates deposited in GenBank. A phylogenetic tree was constructed using the Maximum Parsimony Analysis, with the concatenated sequences (ITS-TEF1α) on MEGAX software (version 11.0.8) (Tamura et al, 2018). All 12 isolates clustered in the same clade and were closely related to isolates A2PP5, A3I1, and XCHK2 with the respective accession numbers KU605789.1, KU605790.1, and MG873071.1 from GenBank, with 99% bootstrap support. The pathogenicity of the 12 isolates was evaluated in pumpkin and melon plants in a greenhouse. Spore suspensions (10 6 conidia/ml -1) were sprayed on the leaves of healthy seedlings until runoff, only water was sprayed on control plants as the mock, and five seedlings of each crop (melon and pumpkin) were inoculated in each treatment. All plants were covered with plastic bags for two days. Spots, similar to those observed on diseased plants in the field, developed on the inoculated leaves (after seven days from the inoculation day, no symptoms were observed on plants from the mock treatment) and the fungal morphology was identical to that observed on the originally diseased leaves, fulfilling Koch's postulate. The pathogenicity test was repeated and yielded the same results. The fact that all 12 isolates were pathogenic on pumpkin and melon leaves, indicates that many Cucurbits are susceptible to C. tenuissimum infection. Many growers in the region are reporting similar symptoms in their melon plantations and it appears that the disease incidence is getting more severe year after year, based on growers's reports. Therefore, more research needs to be conducted to determine the epidemiology and the extension of the economic impact caused by this pathogen to Cucurbits to develop strategies for disease control. To the best of our knowledge, this is the first report of C. tenuissimum causing disease in Cucurbits in Brazil.
Strength Changes Associated with Elongation in Distal Biceps Tendon Repair

Abstract:Objectives: Operative repair of distal biceps tendon ruptures have shown successful outcomes. However, little is know about the amount of tendon or repair site lengthening after repair and how this may affect strength. The purpose of our study was to evaluate distal biceps tendon repair via radiostereometric analysis and correlate with final strength changes. Methods: Eleven patients with distal biceps ruptures were recruited. During repair, two 2-mm tantalum beads with laser-etched holes were sutured to the distal biceps tendon. Beads were evaluated via CT scans immediately post-operatively and at 16 weeks. X-rays were obtained at time 0 and then at 4, 8, and 16 weeks. Seven patients were available for final strength testing at minimum of 9 months postoperative. Results: The mean amount of tendon lengthening after surgery was 22.8 mm (range 11.2 - 30.9 mm, p < 0.05). The greatest change in lengthening was noted between time 0 and week 4 (mean: 11.3 mm, p<0.05), with the least amount of lengthening between week 8 and week 16 (mean: 2.6 mm, p<0.05). DASH averaged 8.2. Strength testing performed at final follow-up (n=7) showed 78% supination and 88% flexion strength compared to contralateral side (33% with dominant arm repaired). Supination strength loss showed the greatest decreases with increased lengthening from 0-4 weeks (Correlation coefficient: -0.39). Final ultrasound evaluations found all tendons to be in continuity. Conclusion: This study’s findings suggest that all patients undergoing distal biceps tendon repair have significant lengthening after surgery. The greatest amount of lengthening was noted in the early post-operative period, in which the greater the amount of lengthening trended toward a greater loss of supination strength. Further studies will focus on post-operative protocols as they may have an impact on early tendon lengthening and could be adjusted to minimize this consequence on long-term supination strength.
