Abstract:Antibiotics have attracted widespread attention around the world because they are ubiquitous in the environment and can lead to antibiotic-resistant microbes developing and pose ecotoxicological risks. In this study, we determined the spatiotemporal distributions of 39 antibiotics in 19 drinking water sources in Jiangsu area of the lower Yangtze River and attempted to identify the sources of the antibiotics and to prioritize the antibiotics. The total antibiotic concentrations in spring and fall were 234.56-6515.99 and 151.12-2562.59 ng/L, respectively. In spring, the total antibiotic concentration gradually increased from upstream to downstream. In fall, the antibiotic concentration did not markedly vary upstream to downstream (total concentrations 151.12-432.17 ng/L) excluding site S9 and S10. Analysis using a positive matrix factorization (PMF) model indicated that the antibiotics had four main sources. Pharmaceutical wastewater was the main source, contributing 34.1% and 41.2% of total antibiotics in spring and fall, respectively, and domestic wastewater was the second most important source, contributing 24.4% and 43% of total antibiotics in spring and fall, respectively. Pharmaceutical wastewater was the main source from midstream to downstream, but the other sources made different contributions in different areas because of the various ranges of human activities. An ecological risk assessment was performed. Stronger risks were posed by antibiotics in spring than fall, and fluoroquinolone antibiotics posed the strongest risks. Optimized risk quotients indicated that norfloxacin was a high-risk contaminant. An assessment of the risk of resistance development indicated that norfloxacin, ciprofloxacin, and enrofloxacin posed moderate to high risks of resistance development and should be prioritized for risk management. The results of this study are important reference data for identifying key sources of antibiotics and developing strategies to manage antibiotic contamination in similar areas.

Abstract:The potential adverse environmental and health-related impacts of antibiotics are becoming more and more concerning. China is globally the largest antibiotic producer and consumer, possibly resulting in the ubiquity and high detection levels of antibiotics in environmental compartments. Clear status on the concentration levels and spatial distribution of antibiotic contamination in China's environment is necessary to gain insight into the establishment of legal and regulatory frameworks. This study collects information from over 170 papers reporting the occurrence and distribution of antibiotics in China's environment. A total of 110 antibiotics were detected, and 28 priority antibiotics were ubiquitous in China in almost all compartments of the environment, excluding the atmosphere. Seven dominant antibiotics in all environment compartments were identified by cluster analysis, including tetracycline, oxytetracycline, chlortetracycline, ofloxacin, enrofloxacin, norfloxacin, and ciprofloxacin. Meanwhile, sulfamethoxazole, sulfadiazine, and sulfamethazine were also frequently found in aqueous phases. Among the main basins where antibiotics were detected, the Haihe River Basin had higher median antibiotic concentrations in surface water compared to other basins, while the Huaihe River Basin had higher median concentrations in sediment. The median values of antibiotic concentrations in the sources were as follows: animal manure, 39 μg/kg (microgram per kilogram); WWTP (wastewater treatment plant) sludge, 39 μg/kg; animal wastewater, 156 ng/L (nanogram per liter); WWTP effluent: 15 ng/L. These concentrations are 1 − 2 orders of magnitude higher than that of the receptors (soil, 2.1 μg/kg; sediment, 4.7 μg/kg; surface water, 8.1 ng/L; groundwater, 2.9 ng/L), whether in solid or aqueous phases. Based on the number of detected antibiotics in various environmental compartments, animal farms and WWTPs are the main sources of antibiotics, and surface water and sediment are the main receptors of antibiotics. Hierarchical clustering identified the two main pathways of antibiotic transfer in various environmental compartments, which are from animal wastewater/WWTP effluent to surface water/sediment and from animal manure/WWTP sludge to soil/groundwater.

Abstract:Antibiotics as emerging pollutants are frequently detected in surface water, raising concerns about the associated risk of antibiotic resistance genes (ARGs). Despite the widespread apprehension, there are still research gaps in the occurrence of antibiotic pollution in surface water and the associated ecological risks to aquatic organisms in China. Here, we established a dataset of antibiotic pollution in surface water in China during 2018-2022, which encompassed 3 368 concentration values of 128 antibiotics reported in 124 articles. Our analysis showed that antibiotic concentrations were predominantly in the ng/L-μg/L range, reaching up to 26 μg/L. Notably, sulfonamides (e.g., sulfamethoxazole) and quinolones (e.g., ciprofloxacin) were frequently reported at high concentrations. The pollution degree of antibiotics represented by sulfamethoxazole, ciprofloxacin, roxithromycin, and tetracycline exhibited no significant variation across different years but was lower in summer than that in spring and autumn. Additionally, distinct spatial distribution characteristics of the pollution were observed. According to calculation results of the aquatic ecological risk assessment model and the weighted frequency, we proposed a list of priority antibiotics including clarithromycin, erythromycin, sulfamethoxazole, ofloxacin, and oxytetracycline in surface water. Last but not least, this study points out the deficiencies in current research on the occurrence and ecological risks of antibiotics in surface water of China and provides viable screening strategies and monitoring recommendations in this context.
Antibiotics and microplastics in manure and surrounding soil of farms in the Loess Plateau: Occurrence and correlation

Abstract:The wide use of animal manure in farmland operations is a source of soil nutrients. However, the return of manure affected antibiotics and microplastics in the soil, thus the potential ecological risks cannot be overlooked. This study investigated the distribution of different antibiotics and microplastics and their correlation. It was found that multiple classes of veterinary antibiotics and microplastics could be detected simultaneously in most manure and soil. In manure, the average concentration of tetracycline antibiotics was higher than fluoroquinolones and sulfonamides. A much lower concentration of antibiotics was found in the soil samples relative to manure. The abundance of microplastics ranged from 21,333 to 88,333 n/kg in manure, and the average abundance was 50,583 ± 24,318 n/kg. The average abundance was 3056 ± 1746 n/kg in the soil. It confirmed that applying organic fertilizer to agricultural soil and the application of plastic mulch in farmlands introduced microplastics. Moreover, microplastics were found to be significantly correlated with antibiotics (r = 0.698, p < 0.001). The correlation between microplastics and antibiotics in soil was significantly weaker than that in manure. Farms could be the hotspot for the co-spread of microplastics and antibiotics. These findings highlighted the co-occurrence of antibiotics and microplastics in agricultural environments.

Abstract:Antibiotic residues are being highlighted around the world as emerging concerns in aquatic environments. Here, the occurrence of antibiotics in tributaries of the Xiaoqing River, China, as well as the spatial and temporal distributions, were reported. The total concentrations of 19 antibiotics in surface water and sediments ranged from 8.20–809.07 ng L–1 and from 39.57– 2,151.64 μg kg–1 dry weight (dw), respectively. Among all detected antibiotics, ofloxacin showed the highest concentration and detection frequency in both surface water (283.83 ng L–1, 100%) and sediment (1,827.18 μg kg–1 dw, 100%). The pollution levels of antibiotics in different seasons were in the following order: dry season > normal season > wet season, except for clarithromycin, sulfadiazine, sulfamerazine and sulfaquinoxaline. Domestic, aquaculture and livestock waste-water might be the main pollution sources of antibiotics in the tributaries of the Xiaoqing River. Moreover, the Pearson correlation coefficient showed significantly positive correlations between antibiotics and heavy metals (such as Cu, Zn and Pb), indicating that there may be a combination of contaminants. The environmental risk assessment indicated a high environmental risk of ofloxacin, clarithromycin, roxithromycin and sulfamethoxazole.

Abstract:The extensive pollution of antibiotics and antibiotic resistance genes (ARGs) in drinking water has aroused worldwide concern. Successive monitoring of these pollutants has noteworthy significance for drinking water safety. Accordingly, this study conducted successive monitoring of antibiotics and ARGs from 2015 to 2017 in a drinking water source in East China. The total antibiotic concentration ranged from 19.68 ng/L to 497.00 ng/L, and decreased slightly from 2015 to 2017. Eighteen out of forty-one ARG subtypes showing resistance to six antibiotic classes and one class I integrase gene intI1, were detected in the drinking water source at concentrations ranging from 6.5 × 104 copies/mL to 1.6 × 106 copies/mL. Importantly, the total ARG concentration increased on an annual basis from 2015 to 2017 with an average annual increment of 0.25 orders of magnitude, which was mainly attributed to the increase in specific ARG subtypes, such as sul1, sul2, sul3, tetA, qnrB, and ermB. Most ARGs was positively correlated with the intI1 genes (r = 0.47–0.55, P < 0.01). Furthermore, the variation of antibiotics and ARGs appeared to be related to the water indices, particularly of the values of COD, BOD5, NO2-N (P < 0.05). This study provides basic data on antibiotic and ARG pollution in the studied drinking water source. Importantly, the findings expound that although the residual antibiotics in this drinking water source decreased slightly from 2015 to 2017, while its biological effect, the antibiotic resistance, increased annually, which give a warning of the antibiotic resistance pollution in the drinking water source.

Abstract:With rapid improvements in industrialization and urbanization, antibiotics are now extensively used to prevent and treat human and animal diseases and husbandry and aquaculture. Some research has been conducted to assess the environmental distribution and risk level of antibiotics, but their distribution remains largely uncharacterized. Thus, this study investigated the distribution and abundance of 39 antibiotics belonging to five groups, and their associated risks in surface water around Luoma Lake in the north of Jiangsu province, China. Nineteen antibiotics were detected, at a detection frequency (DF) ranging from 2.27% to 100%. The total antibiotics (ΣABs) concentrations ranged from 34.91 to 825.93 ng/L, with a median concentration of 195.45 ng/L. Among these antibiotics, chlortetracycline (DF: 100%; median: 172.02 ng/L) was the dominant antibiotic, accounting for a median percentage of 91.0% of ΣABs concentrations. Spearman rank correlation method found a significant correlation between clindamycin (DF: 72.7%; median: 2.01 ng/L) and lincomycin (DF: 79.5%; median: 4.58 ng/L). The ecological risk quotient (RQ) values for two out of 44 sampling sites were higher than 1, indicating a high risk; 11.4% of the RQ values fell between 0.1 and 1, indicating a medium risk. Moreover, roxithromycin was found to be the dominant contributor to the ecological risk, accounting for a median of 79.7% of ΣABs. However, the total non-carcinogenic (<6.54 × 10−4) and carcinogenic risks (<1.64 × 10−7) of ΣABs were negligible at the detected concentrations.

Abstract:Drinking water reservoir is threatened globally due to anthropogenic contamination and thus in need of more attention. Here, we comprehensively investigated the occurrence, distribution and risk assessment of representative antibiotics in a large drinking water reservoir (Fengshuba Reservoir, China). The total concentrations of antibiotics in the water phase, porewater phase, sediment phase and soil phase (drawdown area) were in the ranges of 195-569 ng/L, 47.1-333 ng/L, 114-272 μg/kg and 2.84-77.2 μg/kg, respectively. The dominant antibiotic was CIP in both the water and porewater phases, while it was OTC in the sediment phase. For the water phase, seasonal factor consisting hydrologic condition and pattern of antibiotic use could influence the occurrence level and environmental fate of antibiotics. In contrast, exogenous particles derived from the soils that had used manures during the spring planting season coupled with heavy rainfall was responsible for the occurrence level and composition of antibiotics in the sediment phase. Moreover, Chl-α, NO3-, TP and EC were the most dominant factor influencing the antibiotic distributions in the water phase, porewater phase, sediment phase and soil phase, respectively. Pseudo-partitioning coefficients indicated that PENV and PENG might accumulate more easily into the sediments from the aqueous phase compared with other antibiotics. Risk assessments suggested that TC might pose high risks to the aquatic ecosystem, but the antibiotics presented no risk to the health of consumers. Generally, TC could be used as a promising indicator for evaluating the occurrence and potential risk of antibiotics in Fengshuba Reservoir.

Abstract:The overuse and misuse of antibiotics have resulted in the pollution of antibiotics and antibiotic resistance genes (ARGs) in municipal wastewater treatment plants (WWTPs), posing threats to ecological security and human health. Thus, a comprehensive investigation was conducted to assess the occurrence, removal efficiency, and ecological risk of antibiotics, along with the diversity, abundance, and co-occurrence of ARGs, and their correlations in 13 WWTPs along the Yangtze River Basin. Among 35 target antibiotics, 23 antibiotics within 6 categories were detected in all the samples. Amoxicillin (AMO), ofloxacin (OFL), and pefloxacin (PEF) were predominant in influents, while AMO exhibited dominance with the highest concentration of 1409 ng/L in effluents. Although antibiotic removal performance varied among different WWTPs, a significant decrease in each antibiotic category and overall antibiotics was observed in effluents compared with that in influents ( p < 0.05). Remarkably, ecological risk assessment revealed high risks associated with AMO and ciprofloxacin (CIP) and medium risks linked to several antibiotics, notably including OFL, roxithromycin (ROX), clarithromycin (CLA), and tetracycline (TC). Furthermore, 96 ARG subtypes within 12 resistance types were detected in this study, and the total absolute abundance and diversity of ARGs were significantly decreased from influents to effluents ( p < 0.05). Enrichment of 38 ARGs (e.g., bla NDM , erm A, vat A, mex A, and dfrA 25) in effluents indicated potential health risks. Various mobile genetic elements (MGEs), exhibited significant correlations with a majority of ARGs in both influents and effluents, such as int I1, tnp A1, tnp A5, and tp 614, underscoring the important role of MGEs in contributing to the ARG dissemination. Many antibiotics displayed lower correlations with corresponding ARGs, but exhibited higher correlations with other ARGs, suggesting complex selective pressures influencing ARG propagation. Overall, the incomplete elimination of antibiotics and ARGs in WWTPs is likely to pose adverse impacts on aquatic ecosystems in the Yangtze River Basin.

Abstract:Antibiotics are widely used in humans and animals, but their transformation from surface water to groundwater and the impact of land uses on them remain unclear. In this study, 14 antibiotics were systematically surveyed in a complex agricultural area in Central China. Results indicated that the selected antibiotic concentrations in surface waters were higher in winter (average: 32.7 ng/L) than in summer (average: 17.9 ng/L), while the seasonal variation in groundwaters showed an opposite trend (2.2 ng/L in dry winter vs. 8.0 ng/L in summer). Macrolides were the predominant antibiotics in this area, with a detected frequency of over 90%. A significant correlation between surface water and groundwater antibiotics was only observed in winter (R2 = 0.58). This study further confirmed the impact of land uses on these contaminants, with optimal buffer radii of 2500 m in winter and 500 m in summer. Risk assessment indicated that clarithromycin posed high risks in this area. Overall, this study identified the spatiotemporal variability of antibiotics in a typical agricultural area in Central China and revealed the impact of land uses on antibiotic pollution in aquatic environments.

A mega study of antibiotics contamination in Eastern aquatic ecosystems of China: occurrence, interphase transfer processes, ecotoxicological risks, and source modeling

Abstract:Understanding the occurrence, sources, transfer mechanisms, fugacity, and ecotoxicological risks of antibiotics play a pivotal role in improving the sustainability and ecological health of freshwater ecosystems. Therefore, in order to determine the levels of antibiotics, water and sediment samples were collected from multiple Eastern freshwater ecosystems (EFEs) of China, including Luoma Lake (LML), Yuqiao Reservoir (YQR), Songhua Lake (SHL), Dahuofang Reservoir (DHR), and Xiaoxingkai Lake (XKL), and were analyzed using Ultra Performance Liquid Chromatography/Tandem Mass Spectrometry (UPLC-MS/MS). EFEs regions are particularly interesting due to higher urban density, industrialization, and diverse land use in China. The findings revealed that a collective total of 15 antibiotics categorized into four families, which included sulfonamides (SAs), fluoroquinolones (FQs), tetracyclines (TCs), and macrolides (MLs), exhibited high detection frequencies, indicating widespread antibiotic contamination. The pollution levels in the water phase were in the order of LML > DHR > XKL > SHL > YQR. The sum concentration of individual antibiotics for each water body ranged from not detected (ND) to 57.48 ng/L (LML), ND to 12.25 ng/L (YQR), ND to 57.7 ng/L (SHL), ND to 40.50 ng/L (DHR), and ND to 26.30 ng/L (XKL) in the water phase. Similarly, in the sediment phase, the sum concentration of individual antibiotics ranged from ND to 15.35 ng/g, ND to 198.75 ng/g, ND to 1233.34 ng/g, ND to 388.44 ng/g, and ND to 862.19 ng/g, for LML, YQR, SHL, DHR, and XKL, respectively. Interphase fugacity (ffsw) and partition coefficient (Kd) indicated dominant resuspension of antibiotics from sediment to water, causing secondary pollution in EFEs. Two groups of antibiotics, namely MLs (erythromycin, azithromycin, and roxithromycin) and FQs (ofloxacin and enrofloxacin), showed a medium-high level of adsorption tendency on sediment. Source modeling (PMF5.0) identified wastewater treatment plants, sewage, hospitals, aquaculture, and agriculture as the major antibiotic pollution sources in EFEs, contributing between 6% and 80% to different aquatic bodies. Finally, the ecological risk posed by antibiotics ranged from medium to high in EFEs. This study offers valuable insights into the levels, transfer mechanisms, and risks associated with antibiotics in EFEs, enabling the formulation of large-scale policies for pollution control.
Study on the Distribution Characteristics and Risk Assessment of Antibiotics and Resistance Genes in Water Sources of Wuhan

Abstract:In contemporary society, the improper use of antibiotics leads to their persistent presence in the ecological environment. Due to the diverse physical and chemical properties of antibiotics, their spatial and temporal distribution in the environment varies. Moreover, antibiotics can stimulate the emergence of antibiotic resistance genes (ARGs), which complicates the monitoring and regulation of antibiotics and poses a significant threat to both aquatic and terrestrial environments. This study investigated the distribution of 15 antibiotics and 11 typical ARGs across four categories at 19 sites of drinking water sources in Wuhan, China. The findings revealed that the concentration of antibiotics during the dry season (nd~61,883 ng/L) was significantly higher compared to both the normal water season (nd~49,883 ng/L) and the wet season (nd~28,686 ng/L). Sulfamethoxazole (SMX), sulfamethoxazole (SMD), sulfadiazine (SD), and roxithromycin (RTM) were the predominant antibiotics in the target water environments. The study indicated that most of the antibiotics analyzed posed little to no risk to aquatic organisms. The primary ARGs detected in the surface water of the study area were sul1, qnrD, and tetO. Furthermore, some ARGs showed a negative correlation with their respective antibiotics. Additional research is necessary to evaluate the impact of these emerging pollutants (antibiotics and ARGs) on the safety of high-quality drinking water for residents in Wuhan City.

Profiles, drivers, and prioritization of antibiotics in China's major rivers

Abstract:Through a systematic review of literature references from 2007 to 2022, we compiled a comprehensive national dataset comprising over 67,000 records and covering information on 129 antibiotics detected in the surface water and sediments of China's major rivers. Our analysis revealed notably high antibiotic concentrations in the Liaohe and Yellow Rivers. Among the antibiotics examined, sulfonamides, quinolones, and tetracyclines exhibited relatively high median concentrations in river water. Regional distribution analysis highlighted increased antibiotic levels in Shandong and Tianjin compared to other areas. Partial least squares path modeling revealed that animal production and pollution discharge positively influenced antibiotic levels in river water, whereas natural and socioeconomic factors had negative impacts. Based on the ecological risk assessment, we formulated a prioritized national list of antibiotics, with sulfonamides having the largest number of entries, followed by quinolones. Importantly, our analysis revealed a declining trend in antibiotic concentrations and the associated risk levels across China during the study period. This study not only enhances our understanding of antibiotic distribution in China's water systems, but also contributes to the development of a scientifically sound approach for prioritizing antibiotics. Ultimately, these findings will inform targeted antibiotic management and control strategies. Antibiotic Residues of Drinking-Water and Its Human Exposure Risk Assessment in Rural Eastern China.

Abstract:Trace levels of antibiotics were frequently found in drinking-water, leading a growing concern that drinking-water is an important exposure source to antibiotics in humans. In this study, we investigated antibiotics in tap water and well water in two rural residential areas in Eastern China to assess the related human health exposure risks in drinking-water. Twenty-seven antibiotics were analyzed using ultra performance liquid chro-matography coupled with triple quadrupole tandem mass spectrometry (UPLC-MS/MS). The average daily dose (ADD) and the health risk quotient (HRQ) for exposure to antibiotics in humans were evaluated using 10000 times of Monte Carlo simulations. Ten antibiotics were detected in drinking-water samples, with the maximum concentrations of antibiotic mixture of 8.29 ng/L in tap water and 2.95 ng/L in well water, respectively. Mac-rolides and sulfonamides were the predominant contaminants and showed the seasonality. Azithromycin had the highest detection frequencies (79.71-100%), followed by roxithromycin (25.71-100%) and erythromycin (21.43-86.96%). The estimated ADD and HRQ for human exposure to antibiotic mixture through drinking-water was less than 0.01 mu g/kg/day and 0.01, respectively, which varied over sites, water types, seasons and sex. Ingestion route was more important than dermal contact route (10-6 to 10-4 mu g/kg/day magnitude vs. 10-11 to 10-8 mu g/ kg/day magnitude). Macrolides also contributed mainly to health exposure risks to antibiotics through drinking-water, whose HRQ accounted for 46% to 67% of the total HRQs. Although the individual antibiotic and their combined effects contributed to acceptable health risks for human, the long-term exposure patterns to low-dose antibiotics in drinking-water should not be ignored.

Abstract:The pervasive use of antibiotics has raised substantial environmental concerns, especially regarding their temporal and spatial distribution across diverse water systems. This study addressed the gap in comprehensive research on antibiotic contamination during different hydrological periods, focusing on the Jilin section of the Songliao Basin in Northeast China, an area with severe winter ice cover. The study examined the occurrence, distribution, influencing factors, and potential ecological risks of prevalent antibiotic contaminants. Findings revealed antibiotic concentrations ranging from 239.64 to 965.81 ng/L, with antibiotic resistance genes (ARGs) at 5.22 × 10-2 16S rRNA-1 and antibiotic-resistant bacteria (ARB) up to 5.76 log10 CFU/mL. Ecological risk assessments identified significant risks to algae from oxytetracycline, erythromycin, and amoxicillin. Redundancy analysis and co-occurrence networks with ordinary least squares (OLS) demonstrated that the dispersion of ARGs and ARB is significantly influenced by environmental factors such as total organic carbon (TOC), total phosphorus (TP), total nitrogen (TN), fluoride (F⁻), and nitrate (NO₃⁻). These elements, along with mobile genetic elements (MGEs), play crucial roles in ARG patterns (R2 = 0.94, p ≤ 0.01). This investigation offers foundational insights into antibiotic pollution dynamics in cold climates, supporting the development of targeted mitigation strategies for aquatic systems.
