Abstract:In order to overcome the depth limitation of light penetration in brain tissue, a promising approach in optogenetics is implanting multimode optical fibers for stimulation and fluorescence imaging. However, traditional fiber photometry can only realize stimulation or recording in a single brain region, so optogenetic stimulation and simultaneous recording at the implantation site cannot be achieved. And there is currently no scheme for synchronization of multiple brain regions, therefore the connection between brain regions cannot be strictly proved. To overcome these issues, we developed a multichannel fiber-based in vivo simultaneous neural stimulation and signal readout system. Different from traditional fiber photometry, our system uses the galvanometer scanning technology and a specially-made optical fiber bundle with 200-μm diameter to realize high-precision multichannel stimulation with up to 1-kHz frequency and simultaneous recording. Combined with the specific light-sensitive proteins such as ChrimsonR and genetically encoded calcium indicator GCamP6s, we realize simultaneous optogenetic stimulation and recording of fluorescence signals representing neural activity in up to 7 brain regions of a mouse in vivo. At the same time, we performed optogenetic stimulation in the SNc/VTA brain region of 3 mice expressing the ChrimsonR light-sensitive protein, and recorded the fluorescence signal of the corresponding site simultaneously. In addition, our scheme is also suitable for stimulation and simultaneous recording of multiple brain regions in a single mouse, as well as signal recording and regulation in social and other behaviors in multiple mice. Our system and experimental method allow acquisition and analysis of functional information about relevant brain regions in specific behaviors, thus providing important data and theoretical support for further understanding and treatment of related diseases.

Abstract:Recording and interrogating brain activities using optical methods have become emerging technologies in neuroscience. Traditional tools for optogenetic stimulation in the deep brain are mostly based on implantable fibers, imposing constraints on the animal movement. Recently developed microscale light-emitting diodes (micro-LEDs), which can be wirelessly operated, serve as injectable light sources that directly interact with neural systems. Here, we exploit a wirelessly controlled, implantable system for optogenetic studies in behaving animals. Thin-film indium gallium nitride (InGaN)-based blue micro-LEDs transferred onto flexible probes are injected into the animal brain and optically activate channelrhodopsin-2 expressing neurons. A customized circuit module with a battery is employed to modulate the micro-LED, which is remotely controlled at a distance up to 50mvia 2.4-GHz radio frequency communications. The systems are implemented on freely moving mice, and demonstrate optogenetic modulation of locomotive behaviors in vivo. Moreover, independent and synchronous control of multiple animals is accomplished with the communication unit in the design circuit. The proposed system provides the potential for advanced optical neural interfaces and offers solutions to study complicated animal behaviors in neuroscience research.

Abstract:Objective: Optogenetic manipulations of excitable cells enable activating or silencing specific types of neurons. By expressing two types of exogenous proteins, a single neuron can be depolarized using light of one wavelength and hyperpolarized with another. However, routing two distinct wavelengths into the same brain locality typically requires bulky optics that cannot be implanted on the head of a freely-moving animal. Methods: We developed a lens-free approach for constructing dual-color head-mounted, fiber-based optical units: any two wavelengths can be combined. Results: Here, each unit was comprised of one 450 nm and one 638 nm laser diode, yielding light power of 0.4 mW and 8 mW at the end of a 50 micrometer multimode fiber. To create a multi-color/multi-site optoelectronic device, a four-shank silicon probe mounted on a microdrive was equipped with two dual-color and two single-color units, for a total weight under 3 g. Devices were implanted in mice expressing the blue-light sensitive cation channel ChR2 and the red-light sensitive chloride pump Jaws in parvalbumin-immunoreactive (PV) inhibitory neurons. The combination of dual-color units with recording electrodes was free from electromagnetic interference, and device heating was under 7{\deg}C even after prolonged operation. Conclusion: Using these devices, the same cortical PV cell could be activated and silenced. This was achieved for multiple cells both in neocortex and hippocampus of freely-moving mice. Significance: This technology can be used for controlling spatially intermingled neurons that have distinct genetic profiles, and for controlling spike timing of cortical neurons during cognitive tasks.

Abstract:Abstract Physical and chemical technologies have been continuously progressing advances in neuroscience research. The development of research tools for closed-loop control and monitoring neural activities in behaving animals is highly desirable. In this paper, we introduce a wirelessly operated, miniaturized microprobe system for optical interrogation and neurochemical sensing in the deep brain. Via epitaxial liftoff and transfer printing, microscale light-emitting diodes (micro-LEDs) as light sources and poly(3,4-ethylenedioxythiophene) polystyrene sulfonate (PEDOT:PSS)-coated diamond films as electrochemical sensors are vertically assembled to form implantable optoelectrochemical probes for real-time optogenetic stimulation and dopamine detection capabilities. A customized, lightweight circuit module is employed for untethered, remote signal control, and data acquisition. After the probe is injected into the ventral tegmental area (VTA) of freely behaving mice, in vivo experiments clearly demonstrate the utilities of the multifunctional optoelectrochemical microprobe system for optogenetic interference of place preferences and detection of dopamine release. The presented options for material and device integrations provide a practical route to simultaneous optical control and electrochemical sensing of complex nervous systems.

Abstract:Electrophysiology and optogenetics are pivotal in neuroscience for probing and modulating neural activities, playing a vital role in unraveling the complexities of brain functionality. Despite their importance, the efficacy of existing devices is hampered by insufficient functional integration, pronounced foreign body reactions, and physical constraints that impede natural animal behaviors. Here, we develop a multifunctional, ultraflexible neural probe designed for simultaneous electrophysiological monitoring and optical neural modulation, along with mechanical properties that are conducive to flexibility and compliance. By integrating a wireless neural signal acquisition and stimulation circuit, we achieved wireless recording of brain signals in mice and wireless optogenetic control over their locomotor behavior. The multifunctional ultraflexible probe presented in this study holds substantial promise for closed-loop brain-machine interfaces and deepening our understanding of neural circuit functions. This innovative approach addresses the aforementioned limitations by a comprehensive solution for in vivo neural interrogation and manipulation, marking a significant advancement in the tools available for neuroscience research.

Abstract:Optogenetic nanoprobes are an important technique in optogenetics, utilized to deliver precise light stimulation to neurons in organisms, aiding neuroscientists in investigating the working mechanisms of the brain. Optogenetic nanoprobes have the potential to be used in the diagnosis and treatment of neurological disorders. To fulfill stimulation requirements such as stimulation intensity, stimulation range, stimulation patterns, and temporal and spatial resolution, researchers have developed probes capable of performing various optical functions. To fulfill functional requirements such as in-situ electrophysiological recording and delivery of chemical or biological molecules, scientists have developed multifunctional probes. In order to overcome the disadvantages of traditional optoelectronic devices, as they are rigid and easy to cause severe damage to organisms, flexible optical neural probes have been invented. This type of probe causes minimal harm to the organism during implantation and maintains a consistent level of light illumination, ensuring a prolonged life span. This paper provides an overview and the prospects for different types and functions of optogenetic probes, as well as their flexible technologies.

Abstract:Implantable devices and systems have been emerging as powerful tools for neuroscience research and medical applications. Here we report a wireless, dual-channel optoelectronic system for functional optogenetic interrogation of superior colliculus (SC), a layered structure pertinent to defensive behaviors, in rodents. Specifically, a flexible and injectable probe comprises two thin-film microscale light-emitting diodes (micro-LEDs) at different depths, providing spatially resolved optical illuminations within the tissue. Under remote control, these micro-LEDs interrogate the intermediate layer and the deep layer of the SC (ILSC and DLSC) of the same mice, and deterministically evoke distinct freezing and flight behaviors, respectively. Furthermore, the system allows synchronized optical stimulations in both regions, and we discover that the flight response dominates animals' behaviors in our experiments. In addition, c-Fos immunostaining results further elucidate the functional hierarchy of the SC. These demonstrations provide a viable route to unraveling complex brain structures and functions.

Abstract:Transcranial brain stimulation is a promising technology for safe modulation of brain function without invasive procedures. Recent advances in transcranial optogenetic techniques with external light sources, using upconversion particles and highly sensitive opsins, have shown promise for precise neuromodulation with improved spatial resolution in deeper brain regions. However, these methods have not yet been used to selectively excite or inhibit specific neural populations in multiple brain regions. In this study, we created a wireless transcranial optogenetic brain modulator that combines highly sensitive opsins and upconversion particles and allows for precise bimodal neuromodulation of multiple brain regions without optical crosstalk. We demonstrate the feasibility of our approach in freely behaving mice. Furthermore, we demonstrate its usefulness in studies of complex behaviors and brain dysfunction by controlling extorting behavior in mice in food competition tests and alleviating the symptoms of Parkinson's disease. Our approach has potential applications in the study of neural circuits and development of treatments for various brain disorders.

Abstract:Connecting neural circuit output to behaviour can be facilitated by the precise chemical manipulation of specific cell populations1,2. Engineered receptors exclusively activated by designer small molecules enable manipulation of specific neural pathways3,4. However, their application to studies of behaviour has thus far been hampered by a trade-off between the low temporal resolution of systemic injection versus the invasiveness of implanted cannulae or infusion pumps2. Here, we developed a remotely controlled chemomagnetic modulation—a nanomaterials-based technique that permits the pharmacological interrogation of targeted neural populations in freely moving subjects. The heat dissipated by magnetic nanoparticles (MNPs) in the presence of alternating magnetic fields (AMFs) triggers small-molecule release from thermally sensitive lipid vesicles with a 20 s latency. Coupled with the chemogenetic activation of engineered receptors, this technique permits the control of specific neurons with temporal and spatial precision. The delivery of chemomagnetic particles to the ventral tegmental area (VTA) allows the remote modulation of motivated behaviour in mice. Furthermore, this chemomagnetic approach activates endogenous circuits by enabling the regulated release of receptor ligands. Applied to an endogenous dopamine receptor D1 (DRD1) agonist in the nucleus accumbens (NAc), a brain area involved in mediating social interactions, chemomagnetic modulation increases sociability in mice. By offering a temporally precise control of specified ligand–receptor interactions in neurons, this approach may facilitate molecular neuroscience studies in behaving organisms. Controlled delivery of neuromodulators in the brain might improve the understanding of the molecular basis of behaviour. In this letter, magnetic liposomes injected in deep brain regions release small molecules under remote magnetic stimulation, activating specific neuronal circuits in freely moving mice.

Abstract:The optogenetic probe has proven to be a valuable tool in neuroscience, owing to its specific stimulus properties. Mechanically flexible optogenetic probes are crucial for chronic and deep brain stimulation, minimizing tissue damage, and improving long-term stability and reliability. However, the current implantation of flexible polymer probes often requires the use of additional auxiliary tools, which complicate the surgical process and may cause severe inflammatory responses. In this paper, we propose a flexible polymer waveguide optogenetic probe designed to be inserted directly and precisely into the target brain area without additional assistance through geometric and mechanical optimization of the probe. It also exhibits superior long-term performance in minimizing the inflammatory response after implantation. We eventually succeed in modulating mouse locomotor speed by brain secondary motor cortex (M2) stimulation. This study provides a reference for the future development of flexible waveguide optogenetic probes for reliable and long-term implantation.

Abstract:Bidirectional optogenetic manipulation enables specific neural function dissection and animal behaviour regulation with high spatial–temporal resolution. It relies on the respective activation of two or more visible-light responsive optogenetic sensors, which inevitably induce signal crosstalk due to their spectral overlap, low photoactivation efficiency and potentially high biotoxicity. Herein, a strategy that combines dual-NIR-excited orthogonal emissive upconversion nanoparticles (OUCNPs) with a single dual-colour sensor, BiPOLES, is demonstrated to achieve bidirectional, crosstalk-free NIR manipulation of motor behaviour in vivo . Core@shell-structured OUCNPs with Tm 3+ and Er 3+ dopants in isolated layers exhibit orthogonal blue and red emissions in response to excitation at 808 and 980 nm, respectively. The OUCNPs subsequently activate BiPOLES-expressing excitatory cholinergic motor neurons in C. elegans , leading to significant inhibition and excitation of motor neurons and body bends, respectively. Importantly, these OUCNPs exhibit negligible toxicity toward neural development, motor function and reproduction. Such an OUCNP–BiPOLES system not only greatly facilitates independent, bidirectional NIR activation of a specific neuronal population and functional dissection, but also greatly simplifies the bidirectional NIR optogenetics toolset, thus endowing it with great potential for flexible upconversion optogenetic manipulation.

Abstract:A closed‐loop optogenetic modulation system (CLOMs) for ultra‐fast, ‐accurate, and ‐stable manipulation of neural activities in behaving mice is developed. The CLOMs is composed of multimodal fiber probes and a closed‐loop algorithm, and it is successfully applied for identification and intervention of seizures with high efficiency and long‐term stability, which has promising applications in brain science, neuroscience, and neuromodulation therapies. The precise manipulation of neural activity is of great significance to reveal the function of complex neural circuits in brain, especially the mechanism of neurological disorders such as epilepsy, Parkinson's, and Alzheimer's disease. In the past decades, optogenetics has become a powerful tool to investigate the function of neural circuits due to its unique advantages of cell specificity and high spatiotemporal resolution. However, to date, most optogenetic manipulation has been conducted in an open‐loop fashion. The closed‐loop neural modulation is still an important task. Here, a closed‐loop optogenetic modulation system is developed for ultra‐fast, ‐accurate, and ‐stable manipulation of neural activities in behaving mice. Specifically, this system is mediated by a functional fiber probe with excellent low optical loss (0.052 dB cm−1 at 589 nm) and electrical impedance (5.40 MΩ μm−2 at 1 kHz). In vivo electrophysiology and behavioral experiments demonstrated that the system can be successfully applied for the identification and intervention of seizures in epilepsy mice with high efficiency. It is envision that the closed‐loop manipulation system has promising applications in brain science, neuroscience, and neuromodulation therapies.

Abstract:Advanced technologies for controlled delivery of light to targeted locations in biological tissues are essential to neuroscience research that applies optogenetics in animal models. Fully implantable, miniaturized devices with wireless control and power-harvesting strategies offer an appealing set of attributes in this context, particularly for studies that are incompatible with conventional fiber-optic approaches or battery-powered head stages. Limited programmable control and narrow options in illumination profiles constrain the use of existing devices. The results reported here overcome these drawbacks via two platforms, both with real-time user programmability over multiple independent light sources, in head-mounted and back-mounted designs. Engineering studies of the optoelectronic and thermal properties of these systems define their capabilities and key design considerations. Neuroscience applications demonstrate that induction of interbrain neuronal synchrony in the medial prefrontal cortex shapes social interaction within groups of mice, highlighting the power of real-time subject-specific programmability of the wireless optogenetic platforms introduced here.

Abstract:Genetically encoded photoswitches have enabled spatial and temporal control of cellular events to achieve tailored functions in living cells, but their applications to probe the structure-function relations of signaling proteins are still underexplored. We illustrate herein the incorporation of various blue light-responsive photoreceptors into modular domains of the stromal interaction molecule 1 (STIM1) to manipulate protein activity and faithfully recapitulate STIM1-mediated signaling events. Capitalizing on these optogenetic tools, we identify the molecular determinants required to mediate protein oligomerization, intramolecular conformational switch, and protein-target interactions. In parallel, we have applied these synthetic devices to enable light-inducible gating of calcium channels, conformational switch, dynamic protein-microtubule interactions and assembly of membrane contact sites in a reversible manner. Our optogenetic engineering approach can be broadly applied to aid the mechanistic dissection of cell signaling, as well as non-invasive interrogation of physiological processes with high precision.
Modulating Dopamine Release By Optogenetics In Transgenic Mice Reveals Terminal Dopaminergic Dynamics

Abstract:Dopamine (DA) release and uptake dynamics in the nucleus accumbens (NAc) have important implications for neurological diseases and mammalian animal behaviors. We demonstrate here the use of cell-typespecific optogenetic targeting in conjunction with fast-scan cyclic voltammetry applied to brain slices prepared from specifically tailored transgenic mice, which conditionally express channelrhodopsin-2 (ChR2) through dopamine transporter (DAT)-Cre. Terminal dopaminergic dynamics and the direct manipulation of induced DA release level by controlling light intensity, pulse width, and the shape of stimulation waveforms were studied. Effective cell terminal-targeting optogenetic induction of DA release at physiological levels in NAc is demonstrated and discussed. It was found that delivering more light energy by increasing stimulation intensity and length is not the only way to control DA release; the temporal shape of the stimulus waveform at light onset is also critically related to induced DA concentrations. In addition, DA uptake dynamics as well as the recovery of the presynaptic releasable DA pool are studied and modeled. More broadly, our experimental findings provide important further evidence for effectively applying optogenetics to induce neurotransmitter release in the behaviorally relevant region of the brain in a highly cell-type selective context. Abstract:In vivo pharmacology and optogenetics hold tremendous promise for dissection of neural circuits, cellular signaling, and manipulating neurophysiological systems in awake, behaving animals. Existing neural interface technologies, such as metal cannulas connected to external drug supplies for pharmacological infusions and tethered fiber optics for optogenetics, are not ideal for minimally invasive, untethered studies on freely behaving animals. Here, we introduce wireless optofluidic neural probes that combine ultrathin, soft microfluidic drug delivery with cellular-scale inorganic light-emitting diode (μ-ILED) arrays. These probes are orders of magnitude smaller than cannulas and allow wireless, programmed spatiotemporal control of fluid delivery and photostimulation. We demonstrate these devices in freely moving animals to modify gene expression, deliver peptide ligands, and provide concurrent photostimulation with antagonist drug delivery to manipulate mesoaccumbens reward-related behavior. The minimally invasive operation of these probes forecasts utility in other organ systems and species, with potential for broad application in biomedical science, engineering, and medicine.

Abstract:While offering high-precision control of neural circuits, optogenetics is hampered by the necessity to implant fiber-optic waveguides in order to deliver photons to genetically engineered light-gated neurons in the brain. Unlike laser light, X-rays freely pass biological barriers. Abstract:The use of optogenetic stimulation to evoke neuronal activity in targeted neural populations—enabled by opsins with fast kinetics, high sensitivity and cell-type and subcellular specificity—is a powerful tool in neuroscience. However, to interface with the opsins, deep-brain light delivery systems are required that match the scale of the spatial and temporal control offered by the molecular actuators. Here we show that organic light-emitting diodes can be combined with complementary metal–oxide–semiconductor technology to create bright, actively multiplexed emissive elements. We create implantable shanks in which 1,024 individually addressable organic light-emitting diode pixels with a 24.5 μm pitch are integrated with active complementary metal–oxide–semiconductor drive and control circuitry. This integration is enabled by controlled electrode conditioning, monolithic deposition of the organic light-emitting diodes and optimized thin-film encapsulation. The resulting probes can be used to access brain regions as deep as 5 mm and selectively activate individual neurons with millisecond-level precision in mice.

Abstract:Information is transmitted between brain regions through the release of neurotransmitters from long-range projecting axons. Understanding how the activity of such long-range connections contributes to behavior requires efficient methods for reversibly manipulating their function. Chemogenetic and optogenetic tools, acting through endogenous G-protein-coupled receptor pathways, can be used to modulate synaptic transmission, but existing tools are limited in sensitivity, spatiotemporal precision or spectral multiplexing capabilities. Here we systematically evaluated multiple bistable opsins for optogenetic applications and found that the Platynereis dumerilii ciliary opsin ( Pd CO) is an efficient, versatile, light-activated bistable G-protein-coupled receptor that can suppress synaptic transmission in mammalian neurons with high temporal precision in vivo. Pd CO has useful biophysical properties that enable spectral multiplexing with other optogenetic actuators and reporters. We demonstrate that Pd CO can be used to conduct reversible loss-of-function experiments in long-range projections of behaving animals, thereby enabling detailed synapse-specific functional circuit mapping.
