Abstract:Since their introduction in the 1840s, one of the largest mysteries of modern anesthesia are how general anesthetics create the state of reversible loss of consciousness. Increasing researchers have shown that neural pathways that regulate endogenous sleep-wake systems are also involved in general anesthesia. Recently, the Lateral Habenula (LHb) was considered as a hot spot for both natural sleep-wake and propofol-induced sedation; however, the role of the LHb and related pathways in the isoflurane-induced unconsciousness has yet to be identified. Here, using real-time calcium fiber photometry recordings in vivo, we found that isoflurane reversibly increased the activity of LHb glutamatergic neurons. Then, we selectively ablated LHb glutamatergic neurons in Vglut2-cre mice, which caused a longer induction time and less recovery time along with a decrease in delta-band power in mice under isoflurane anesthesia. Furthermore, using a chemogenetic approach to specifically activate LHb glutamatergic neurons shortened the induction time and prolonged the recovery time in mice under isoflurane anesthesia with an increase in delta-band power. In contrast, chemogenetic inhibition of LHb glutamatergic neurons was very similar to the effects of selective lesions of LHb glutamatergic neurons. Finally, optogenetic activation of LHb glutamatergic neurons or the synaptic terminals of LHb glutamatergic neurons in the rostromedial tegmental nucleus (RMTg) produced a hypnosis-promoting effect in isoflurane anesthesia with an increase in slow wave activity. Our results suggest that LHb glutamatergic neurons and pathway are vital in modulating isoflurane anesthesia.

Abstract:Two lines of mice have been genetically selected for differential sensitivity to ethanol. These lines have been designated long sleep (LS) and short sleep (SS) on the basis of their hypnotic response to the ethanol selection dose. Earlier studies of these mice suggested that this difference was limited to alcohols and did not extend to other classes of hypnotics. The present study examined hypnotic and hypothermic responses produced by pentobarbital in recent generations of these mice. Dose-dependent differences in sleep time and in hypothermia were found, with SS mice affected to a greater degree than LS mice. Pharmacokinetic studies showed that the half-life of pentobarbital disappearence from SS blood was twice that reported for SS mice of the 18th generation. The half-life in the LS line had not changed. The volumes of distribution and waking brain concentrations were identical in LS and SS mice. An altered rate of elimination (not differential CNS sensitivity) appeared to be the major factor responsible for the differences observed between these lines.

Abstract:The effects of L-theanine on barbiturate-induced sleeping tirade, subthreshold barbiturate-induced sleeping time and barbiturate-induced sleeping latency period in mice were investigate. Healthy ICR mice were randomly divided into four groups of 12 mice each: control group fed with distilled water and three treatment groups fed L-theanine at low-, medium-, and high- and the best effect was found in the medium-dose-treated mice. However, no direct hypnotic effect of L-theanine was observed while it was able to prolong the sleeping time induced by pentobarbital sodium [that of the medium-dose-treated mice reached (40.3±26.0) min] and to enhance the sleeping rate induced by barbital sodium at the threshold dose (that of the medium-dose- treated mice reached 33.3 %). L-Theanine also eoald significantly decrease (P < 0.05) the sleeping latency period induced by barbital sodium [that of the medium-dose-treated mice reached (30.32±4.8) min].

Abstract:A unified physiologically based pharmacokinetic (PB-PK) model was developed and used to describe the disposition of methylchloroform (1,1,1-trichloroethane, MC) in three different species (rats, mice, and humans) after four different routes of exposure (inhalation, intravenous injection, bolus gavage, and drinking water administration). Metabolism of MC followed Michaelis-Menten kinetics in each species. Vmax's were calculated from the allometric equation: Vmax = 0.419 BW0.7, and Km appeared to be identical in each species (5.75 mg equivalents/liter). Once the PB-PK model had been developed for young adult animals (1-3 months of age), it was used to study the disposition of MC in older rats and mice (approximately 18.5 months of age). Most of the changes in the pharmacokinetic behavior of MC in older rats could be simulated by increasing the size of the fat compartment in the PB-PK model from 7 to 18% of body weight. However, the pharmacokinetic behavior in older mice was more complex; increasing the size of the fat compartment in this species from 4 to 18% only accounted for part of the observed differences between old and young animals. An appropriate dose surrogate (average area under the liver concentration/time curve) was selected and the PB-PK model was used to make quantitative comparisons between "internal doses" of MC in long term animal studies and "internal doses" associated with human exposures to MC. Values of the dose surrogate in humans consuming 2 liters/day of water with typical levels of MC contamination (1-10 ppb) were four to six orders of magnitude lower than the dose surrogates in the rodent studies at levels of MC exposure which failed to produce adverse effects on the liver (875-1500 ppm, 6 hr/day, 5 days/week).

Abstract:Objective To investigate the effects of melatonin on choline acetyltransferase (ChAT) in rat hippocampus after isoflurane anesthesia. Methods Sixty male SD rats weighing 390 - 440 g were randomized into 5 groups (n = 12 each): control group (group C), 1% isoflurane group (group Ⅰ), 1% isoflurane + melatonin group (group IM) , 2% isoflurane group (group J) and 2% isoflurane + melatonin group (group JM) . In IM and JM groups, melatonin 10 mg/kg was administered intraperitoneally once a day for 7 consecutive days, while equal volume of normal saline was given intraperitoneally instead of melatonin in C, I and J groups. Groups Ⅰ and IM inhaled 1% isoflurane and groups J and JM 2% isoflurane for 4 h on 7th day. All the rats underwent Morris water maze test on the day after anesthesia for assessment of learning and memory ability (escape latency and probe time) . The training test was performed 4 times a day for S days. Six rats randomly selected from each group were sacrificed the end of the test. The blood samples were collected for detection of plasma melatonin level by ELISA.The brain tissues were removed for determination of the expression and activity of ChAT in hippocampus by Western blot or colorimetric assay. The left rats were selected and sacrificed for determination of the number of ChAT positive neurons in hippocampal CA1 region and entate gyrus by immunofluorescence. Results The plasma melatonin level and expression and activity of ChAT were significantly lower in group I than in group C ( P ＜ 0.01) . The escape latency was significantly longer, the probe time was significantly shorter, and the plasma melatonin level and expression and activity of ChAT were significantly lower in group J than in group C ( P ＜ 0.05 or 0.01) . The escape latency was significantly shorter, the probe time was significantly longer, and the plasma melatonin level and expression and activity of ChAT were significantly higher in group IM than in group Ⅰ ( P ＜ 0.05 or 0.01). The escape latency was significantly shorter and the plasma melatonin level and ChAT activity were significantly higher in group JM than in group J ( P ＜ 0.05 or 0.01) . The results of immunofluorescent staining showed that the number of ChAT positive neurons in hippocampal CA1 region and dentate gyrus wag consistent with the changes in the measured ChAT expression. Conclusion Melatonin can reduce isoflurane-mediated inhibition of ChAT expression and activity and thus improve spatial memory impaired by isoflurane anesthesia in rats.\r\n\r\nKey words: \r\nMelatonin; Isoflurane; Hippocampus; Choline O-acetyltransferase

Abstract:The relationship between the duration of pentobarbital induced sleep and respiratory oxygen tension was investigated in mice and cats. Pentobarbital, when administered to cats in a high dose (70 mg/kg), resulted in irregular respiration, and prolonged biologic half life. And found that the characteristic curve of disappearance of pentobarbital obtained from a high dose administration of pentobarbital were caused by low activity of liver microsomal oxidative enzyme and not by the tissue accumulation. The low activity was assumed to be caused by insufficiency of oxygen required by oxidative enzymes.

Abstract:In a 6- to 8-month dose-response toxicity study, methapyrilene hydrochloride (HCl) was administered in the feed to Fischer-344N rats and B6C3F1 mice at concentrations of 125, 250, 500, 1000, or 2000 ppm. After 26 weeks, rats fed the higher dose levels of methapyrilene HCl developed cholangiocellular carcinomas in addition to severe hepatotoxic lesions. After 31 weeks, mice exhibited only mild hepatotoxicity from ingesting methapyrilene HCl, even at the higher dose levels. The duration of sodium pentobarbital-induced sleeping time (PEN-induced ST) is determined by the rate of hepatic metabolism of PEN and can therefore be used as an in vivo measurement of the level of hepatic detoxifying enzymes. A subsequent study was undertaken with PEN-induced ST to investigate differences in the effect of methapyrilene HCl on hepatic detoxifying enzymes between the two species; in this study, prior treatment with methapyrilene HCl significantly increased PEN-induced ST of rats but not of mice, indicating that methapyrilene HCl had a suppressive effect on detoxifying enzymes in rats but not in mice. In other groups in this study, phenobarbital, a known inducer of hepatic detoxification enzymes, was administered simultaneously with methapyrilene HCl to both rats and mice. Methapyrilene HCl plus phenobarbital exerted a synergistic effect of enzyme induction in mice. In rats, the phenobarbital-induced hepatic enzymes increased the rate of hepatic detoxification of methapyrilene HCl, as measured by a reduction of PEN-induced ST as compared with that in control rats or rats receiving methapyrilene HCl alone. The difference in the effects of methapyrilene HCl on hepatic detoxifying mechanisms of rats and mice, as indicated by the metabolism of PEN, may explain the difference between the two species in their susceptibility to the hepatocarcinogenic effects of methapyrilene HCl.

Abstract:Introduction Mice are the most common animal model used for in vivo studies relevant to human disease. The numerous mouse phenotypes available, created by genetic manipulation, enable the linking of specific genes to normal or abnormal molecular and organ functions. Isoflurane is a very common type of anesthesia used in many laboratories performing metabolism and diabetes research. The study of diabetes and obesity focuses on the glucose level as one of the main variables. Moreover, microPET imaging techniques specifically focus on blood glucose biodistribution 1 2 3 4. The handling and preparation of mice (stress, body temperature, and anesthesia) are very important factors that can impact on glucose levels. Pomplun et al. 5 have previously reported that blood glucose levels in wild type C57Bl/6 mice were significantly elevated immediately following isoflurane anesthesia while Fueger et al. 6 reported that isoflurane anesthesia (2% in 100% oxygen) mildly increased blood glucose levels in severe combined immunodeficient (SCID) male mice 5 6. Other investigators have reported development of hypoglycemia during isoflurane anesthesia 1 2 7. Since we routinely use isoflurane, a common type of inhalation anesthesia, in our imaging studies we examined its impact on blood glucose levels in male and female mice. Mice exhibited an elevation in blood glucose levels immediately after the period of anesthesia. This effect was significant for male mice. Subsequently, glucose readings declined significantly, and eventually attained a reading very similar to the initial reading, pre-anesthesia.

Abstract:The dose and route dependency of the metabolism and the toxicity of chloroform (CHCl3) and carbon tetrachloride (CCl4) was investigated in ethanol-treated rats. Rats that had been kept on either an ethanol (containing ethanol at 2 g/rat/day) or a control (containing no ethanol) liquid diet for 3 weeks were challenged with CHCl3or CCl4by inhalation (0, 50, or 500 ppm × 6 hr), or by po or ip administration (0, 0.105, or 1.675 mmol/kg). Ethanol consumption, which increased thein vitrometabolism of both compounds six- to sevenfold, affected the metabolism and toxicity of CCl4differently from those of CHCl3. Ethanol increased the metabolism and toxicity of CHCl3at 500 ppm only, whereas it increased the metabolism and toxicity of CCl4at either 50 or 500 ppm. In addition, the effect of ethanol consumption differed between CHCl3and CCl4depending on the route of administration (po or ip) and the dose (0.105 or 1.675 mmol/kg). For the po route, ethanol increased the metabolism and toxicity of both compounds at either dose. For the ip route, however, ethanol increased the metabolism and toxicity of the high dose of CHCl3only, but of both CCl4doses. The dose- and route-dependent differences in the effect of enzyme induction on the metabolism and toxicity between CHCl3and CCl4can be explained by a supposition that the hepatic blood flow rate-limits the metabolism of a low dose of CHCl3(perfusion-limited metabolism), whereas the metabolic capacity of the liver limits the metabolism of CCl4(capacity-limited metabolism) irrespective of dose.
Impacts of Different Anesthetic Agents on Left Ventricular Systolic Function in Mice Assessed by Echocardiography

Abstract:The present experiments were performed to study the effects and time trends of different anesthetic agents on the left ventricular (LV) systolic function and heart rate by high-resolution echocardiography in mice. Ten male C57BL/6J mice were submitted to echocardiography imaging separated by 72-hour intervals under the following conditions: 1) conscious mice, 2) mice anesthetized with isoflurane (ISO, inhaled), 3) mice anesthetized with tribromoethanol (TBE, intraperitoneal), 4) mice anesthetized with chloral hydrate (CH, intraperitoneal), and 5) mice anesthetized with pentobarbital sodium (PS, intraperitoneal). The effect of ISO, TBE, CH, and PS on LV systolic function was measured at 0, 1, 2, 3, 4, 6, 8, and 10 min after anesthesia. The results showed that LV systolic function and heart rate (HR) of anesthetized mice were reduced significantly (P<0.05), compared with results in the same mice studied in the conscious state. In addition, the results indicated that the anesthetic with the least effect on LV function was CH, and followed by TBE, PS, ISO. We conclude that different anesthetic agents always depressed the HR and LV systolic function of mice, and, furthermore, the effects and time trends of different anesthetics on LV function are different. In echocardiographic experiments, we should choose proper anesthetic agents according to the experimental requirements.

Dose and Route Dependency of Metabolism and Toxicity of Chloroform in Ethanol-Treated Rats

Abstract:The effects of a single dose of ethanol on the metabolism and toxicity of chloroform administered to rats per os (p.o.), intraperitoneally (i.p.), or by inhalation (inh) at different doses were investigated. Rats that had been given either ethanol (2 g/kg) or vehicle (water) alone at 4 p.m. on the previous day were challenged with chloroform at 10 a.m. p.o. (0, 0.1, 0.2, or 0.4 g/kg), i.p. (0, 0.1, 0.2, or 0.4 g/kg), or inh (for 6 h each at 0, 50, 100, or 500 ppm). The ethanol treatment, which had no influence on the intake of food and water, increased chloroform metabolism in vitro about 1.5-fold with no significant influence on liver glutathione content. The treatment had a dose-dependent effect o nthe metabolism and toxicity of chloroform, and the effect differed depending on the route of administration. Compared at the same dose level, the area under the curve (AUC) of blood chloroform concentration was invariably smaller following p.o. than i.p. administration. In accordance with this, chloroform administered p.o. caused more deleterious hepatic damage than the same amount of chloroform administered i.p. Although ethanol treatment had no significant influence on the AUC at any dose by any route of administration, the toxicity of p.o.-administered chloroform was significantly higher in ethanol-treated rats than in control rats at a dose as low as 0.1 g/kg, whereas no significant difference was observed in toxicity between both groups of rats at such a low dose administered i.p. When rats were exposed inh to air containing chloroform vapor, ethanol consumption had no effect on hepatotoxicity until the exposure concentration was raised to 500 ppm, a finding which suggests that a single dose of ethanol (2 g/kg) affects the toxicokinetics of inhaled chloroform in rats only at a concentration as high as 500 ppm.
Validation of the anesthetic effect of a mixture of remimazolam, medetomidine, and butorphanol in three mouse strains

Abstract:Proper administration of anesthesia is indispensable for the ethical treatment of lab animals in biomedical research. Therefore, selecting an effective anesthesia protocol is pivotal for the design and success of experiments. Hence, continuous development and refinement of anesthetic agents are imperative to improve research outcomes and elevate animal welfare. "Balanced anesthesia" involves using multiple drugs to optimize efficacy while minimizing side effects. The medetomidine, midazolam, and butorphanol, called MMB, and medetomidine, alfaxalone, and butorphanol, called MAB, are popular in Japan. However, the drawbacks of midazolam, including its extended recovery time, and the narrow safety margin of MAB, have prompted research for suitable alternatives. This study replaced midazolam in the MMB combination with remimazolam (RMZ), which is noted for its ultra-short half-life. The resulting combination, called MRB, was effective inproviding a wider safety margin compared to MAB while maintaining an anesthesia depth equivalent level to that of MMB in mice. Notably, MRB consistently exhibited better recovery scores after antagonist administration in contrast to MMB. Furthermore, the re-sedation phenomenon observed with MMB was not observed with MRB. The rapid metabolism of RMZ enables reliable anesthesia induction, circumventing the complications linked to MAB. Overall, MRB excelled in providing extended surgical anesthesia and swift post-antagonist recovery. These results highlight the potential of RMZ for broader animal research applications.

Effect of P-Amino-Diphenyl Ethers on Hepatic Microsomal Cytochrome P450

Abstract:The present paper aims to investigate whether p-amino-2',4'-dichlorodiphenyl ether and p-amino-4'-methyldiphenyl ether are inhibitors as well as inducers of P450. Mice were given daily intraperitoneal (ip) injections of p-amino-2',4'-dichlorodiphenyl ether (0.25 mmol/kg) or p-amino-4'-methyldiphenyl ether (0.25 mmol/kg) for 4 days and tested at 24 h and 48 h after the last dose injection. The results showed the mice pentobarbital sleeping time was shorter and the P450 content of hepatic microsome increased significantly in the group pretreated with p-amino-4'-methyldiphenyl ether when compared with the control group, while in mice pretreated with p-amino-2',4'-dichlorodiphenyl ether the hepatic microsome P450 content increased but the pentobarbital sleeping time was extended in clear contrast to the control group. The sleeping time of the phenobarbital group (80 mg/kg daily ip injection for 4 days) was shortened at 24 h after the last injection with increased P450 content of hepatic microsome, but it showed no difference at 48 h. The zoxazolamine-paralysis times of mice treated with p-amino-2',4'-dichlorodiphenyl ether were longer than those of the control mice, while the same dose of zoxazolamine did not lead to paralysis in mice pretreated with BNF. p-Amino-2',4'-dichlorodiphenyl ether and p-amino-4'-methyldiphenyl ether inhibited the activity of 7-ethoxyresorufin O-deethylase from rat hepatic microsome induced by BNF in vitro by 70.0% and 50.1% respectively. These results suggest that p-amino-2',4'-dichlorodiphenyl ether and p-amino-4'-methyldiphenyl ether are inhibitors as well as inducers of P450.

Abstract:Chloroform is a prevalent toxic environmental pollutant in urban settings, posing risks to human health through exposure via various mediums such as air and tap water. The gut microbiota plays a pivotal role in maintaining host health. However, there is a paucity of research elucidating the impact of chloroform exposure on the gut microbiota. In this investigation, 18 SPF Kunming female mice were stratified into three groups (n = 6) and subjected to oral gavage with chloroform doses equivalent to 0, 50, and 150 mg/kg of body weight over 30 days. Our findings demonstrate that subchronic chloroform exposure significantly perturbs hematological parameters in mice and induces histopathological alterations in cecal tissues, consequently engendering marked disparities in the functional composition of cecal microbiota and metabolic equilibrium of cecal contents. Ultimately, our investigation revealed a statistically robust correlation, exhibiting a high degree of significance, between the intestinal microbiome composition and the metabolites that were differentially expressed consequent to chloroform exposure.

Determination of the Pharmacokinetics and Tissue Distribution of Methyl 3,4-Dihydroxybenzoate (MDHB) in Mice Using Liquid Chromatography-Tandem Mass Spectrometry

Abstract:Background and objectives: Methyl 3,4-dihydroxybenzoate (MDHB) has the potential to prevent neurodegenerative diseases (NDDs). The present work aims to reveal the pharmacokinetics and tissue distribution characteristics of MDHB. Methods: The pharmacokinetics and tissue distribution of MDHB were analyzed using LC-MS/MS after a single intragastric administration (50 to 450 mg/kg) in mice, and samples were collected from five animals at specific time points. Results: Pharmacokinetic parameters of MDHB following intragastric administrations were: the time to peak concentration (Tmax) ranged from 0.033 to 0.07 h, the peak concentration (Cmax) ranged from 12,379.158 to 109798.712 μg/l, the elimination half-life (t1/2z) ranged from 0.153 to 1.291 h, the area under the curve (AUC0-∞) ranged from 640.654 to 20,241.081 μg/l × h, the mean residence time (MRT0-∞) ranged from 0.071 to 0.206 h, the apparent volume of distribution (Vz/F) ranged from 17.538 to 45.244 l/kg, and the systemic clearance (Clz/F) ranged from 22.541 to 80.807 l/h/kg. The oral bioavailability of MDHB was 23%. The maximum MDHB content was detected in the stomach, and the minimum content was observed in the testes; the peak content in the brain was 15,666.93 ng/g. Conclusions: The pharmacokinetic characteristics of MDHB include fast absorption, high systemic clearance, a short half-life and an oral bioavailability of 23%. Additionally, MDHB permeates the blood-brain barrier (BBB) and is rapidly distributed to all organs. The identification of the pharmacokinetics of MDHB following its oral administration will contribute to further preclinical and clinical studies of its effects.
Evaluating Intrahepatic and Intraperitoneal Sodium Pentobarbital or Ethanol for Mouse Euthanasia

Abstract:Intraperitoneal (IP) injection of sodium pentobarbital (PB) is an accepted method of euthanasia for mice. However, this method has important drawbacks, including the potential for pain or misinjection. The objective of this prospective, randomized, blinded study was to determine whether intrahepatic (IH) injection of PB is more effective than IP delivery for mouse euthanasia. Secondary objectives were to: 1) determine whether IP ethanol (ET) is a suitable alternative to PB and 2) study the effect of isoflurane anesthesia on euthanasia with either PB or ET. Eighty adult CD1 mice were randomly assigned to 6 different treatment groups, were euthanized by using IP or IH injections of either PB or ET, and were either anesthetized or conscious before injection. Variables of interest were: 1) misinjection rates (based on necropsy evaluation), 2) time from injection to apnea and 3) time to cessation of heartbeat (CHB). The misinjection rate for IH injections was 93% (28/30). Two successful IH injections resulted in death within 4 s, but this method cannot be recommended due to the possibility for intrathoracic injection ( n = 4). In nonanesthetized mice, time to apnea and CHB was significantly shorter with IP ET (apnea: 72.5 s [median], CHB: 115 s) than with IP PB (apnea: 136 s, CHB: 176 s). Anesthesia at time of injection was associated with a shorter CHB time for IP PB. These data show the difficulty in achieving successful IH injections in mice, but confirm that IP ET is a viable and potentially superior alternative to IP PB. Lastly, anesthesia can shorten time to death after IP injection of PB.
Reticular telangiectatic erythema: a reactive clinicopathological entity related to the presence of foreign body

Abstract:Editor Reticular telangiectatic erythema is a cutaneous reaction which has been reported presenting near or at the site of an implanted medical device. We present a patient who developed postsurgical reticular telangiectatic erythema over a breast related to the persistence of a non-absorbable suture thread. A 61-year-old woman with history of hepatitis C and cirrhosis underwent lumpectomy for breast cancer. She refused radiation therapy or any other adjuvant treatment. One month later she developed an erythema with reticulated telangiectasia over the surgical scar which progressively spread around until covering almost the entire breast skin (Fig. 1a). When we assessed the patient, the lesion had 1 month of evolution and had been treated for 3 weeks with topical corticosteroids and antibiotics with no response and progression. Cutaneous erythema was asymptomatic and showed neither swelling, nor warmth nor tenderness in the exploration. Surgical scar was indurated on palpation but without signs that suggested infection and the patient had no fever. A skin biopsy was obtained and the histological study showed dermal telangiectasia, mild lymphocytic infiltrate and epidermal atrophy (Fig. 2). Alcian blue staining revealed no mucin deposition. These clinicopathological findings suggested the diagnosis of reticular telangiectatic erythema. Cutaneous lesion remained for two more months but in a follow-up visit the persistence of a long non-absorbable thread of intradermal suture was detected. This persistent suture thread was removed and reticular telangiectatic erythema disappeared completely after 2 weeks (Fig. 1b). Patch test was performed with the same non-absorbable suture thread as is and was negative. Negative patch test results supported the diagnosis of reticular telangiectatic erythema, excluding erythema arising from an allergic contact dermatitis. Reticular telangiectatic erythema is characterized by the appearance of erythematous, blanchable reticular telangiectatic area in the skin. It has been described mostly associated with implantable cardiac devices and other implantable medical devices with electronic components. This fact has led to some authors to hypothesize that its origin could be the heat and electric or magnetic fields generated by these devices. However, it has also been reported following implantation of articular prosthesis. Moreover, Morgan et al. reported in 2005 three cases of a postsurgical sternal erythema with clinical and histopathological findings remarkably similar to our patient. Later the same authors and others considered that these were actually reticular telangiectatic erythema probably related to sternal wires. (a)
Differential effects of pentobarbital and ethanol in mice

Abstract:The duration of the loss of righting reflex (RR) after ethanol, 4 g/kg, intraperitoneally (i.p.), was significantly longer in “long-sleep” (LS) than in “short-sleep” (SS) mice. This effect was shown to be correlated with differences in brain sensitivities to ethanol. In contrast, pentobarbital sodium (PB), 50 mg/kg, i.p., produced a significantly longer loss of RR in SS than in LS mice. The PB concentrations in the brain were the same in both mouse strains at the time of RR recovery suggesting equal sensitivities of the central nervous systems to PB. The rates of disappearance of PB from the blood were the same in both strains, but the apparent volume of distribution of PB in the LS strain was greater than in SS mice.In addition, C57BL/6J mice were found to be more sensitive than DBA/2J mice to PB, 50 mg/kg. In contrast, C57BL mice are known to be less sensitive than the DBA strain to ethanol. The PB concentration in the brain of DBA mice at the recovery of the RR was significantly greater than in C57BL mice. The apparent volumes of distribution of PB were not different in the two strains, but the rate of disappearance of PB from the blood of C57BL mice was significantly greater than for the DBA strain. In conclusion, factors which govern the brain sensitivities of selected mouse strains to ethanol and pentobarbital may not be equivalent.
