Abstract:The use of DNA as programmable building blocks to achieve highly-definable nanostructures, termed as DNA nanotechnology, has offered great opportunities to control matter at a very small scale. Fast growing researches along this direction has led to the emergence of several significant branches of DNA nanotechnology. This review intends to summarize the recent developments in this area. Several topics will be discussed with a focus on the works from the authors' group, including (1) DNA self-assembly and DNA-directed self-assembly, (2) DNA nanomachines, (3) DNA-based electronics, (4) DNA assisted nanofabrications, and (5) DNA computations.

This review provides a comprehensive overview of the latest advancements in the synthesis of DNA amphiphiles and their assembly into well-defined nanostructures. It explores the diverse applications of these nanostructures across various medical domains, including targeted drug delivery, innovative immunotherapies, and gene-silencing techniques. Moreover, the review delves into the current challenges and future prospects of this rapidly evolving field, highlighting the potential of DNA hybrid materials to revolutionize medical treatments and diagnostics. Abstract:Over the past decades, deoxyribonucleic acid (DNA), as a versatile building block, has been widely employed to construct functionalized nanostructures. Among the diverse types of materials, DNA related nanostructures have gained growing attention due to their intrinsic programmability, favorable biocompatibility, and strong molecular recognition capability. The conventional construction strategy for building DNA structures is based on Watson–Crick base-pairing rules, which are mainly driven by the hydrogen bonding of bases. However, hydrogen bonding-based DNA nanostructures cannot meet the requirements of specific morphology and multifunctionality. Currently, various functional elements have been introduced to expand the synthetic methodologies for constructing the DNA hybrid nanostructures, including small molecules, peptide polymers, organic ligands and transition metal ions. Besides, the potential applications for these DNA hybrid nanostructures have also been explored. It has been demonstrated that DNA hybrid structures with various properties can be extensively applied in the fields of magnetic resonance, luminescence imaging, biomedical detection, and drug delivery systems. In this review, we highlight the pioneering contributions to the methodologies of DNA-based nanostructure assembly. Furthermore, the recent advances in drug delivery systems and biomedical diagnosis based on DNA hybrid nanostructures are briefly summarized.

Abstract:Nucleic acids hold great promise for bottom-up construction of nanostructures via programmable self-assembly. Especially, the emerging of advanced sequence design principles and the maturation of chemical synthesis of nucleic acids together have led to the rapid development of structural DNA/RNA nanotechnology. Diverse nucleic acids-based nano objects and patterns have been constructed with near-atomic resolutions and with controllable sizes and geometries. The monodispersed distribution of objects, the up-to-sub-millimeter scalability of patterns, and the excellent feasibility of carrying other materials with spatial and temporal resolutions have made DNA/RNA assemblies extremely unique in molecular engineering. In this review, we summarize recent advances in nucleic acids-based (mainly DNA-based) near-atomic fabrication by focusing on state-of-the-art design techniques, toolkits for DNA/RNA nano-engineering and related applications in a range of areas.

However, an electronic publishing environment should: (1) support the whole life cycle of publication; (2) be a highly integrated environment; (3) have a hypermedia publishing capability; and (4) be easily extended to deal with new media and support new publishing activities. An electronic publishing environment called MUCH (Many Using and Creating Hypermedia) has been developed that supports collaborative authoring, reuse, formating and printing, management, hypermedia publishing and delivery, and interchange. MUCH is based on public domain tools, such as Andrew, and has been used in the authoring of many lengthy documents. © 1994 John Wiley & Sons, Inc.

Abstract:Abstract— –When washed cells of histidine‐grown Pseudomonas putida were incubated at 8°C in darkness, urocanase activity diminished after several days; most of this lost activity could be restored by treatment of cell extracts with near‐u.v. light. Sunlight and daylight were also effective for activation. Non‐irradiated extract, when added to the active preparation, did not inhibit the enzyme activity. Heated and boiled extracts with or without irradiation did not catalyze the urocanase reaction nor did they change the activity of an activated extract when added to it. Photoactivation of cell extracts did not require oxygen, was not dependent on temperature and was not prevented by dialysis. Urocanase purified by gel electrophoresis was capable of light activation. It is suggested that the photoreceptor is closely associated with urocanase since it is not separated from the enzyme by dialysis or electrophoresis.

Abstract:Researchers have worked for many decades to master the rules of biomolecular design that would allow artificial biopolymer complexes to self-assemble and function similarly to the diverse biochemical constructs displayed in natural biological systems. The rules of nucleic acid assembly (dominated by Watson–Crick base-pairing) have been less difficult to understand and manipulate than the more complicated rules of protein folding. Therefore, nucleic acid nanotechnology has advanced more quickly than <i>de novo</i> protein design, and recent years have seen amazing progress in DNA and RNA design. By combining structural motifs with aptamers that act as affinity handles and add powerful molecular recognition capabilities, nucleic acid-based self-assemblies represent a diverse toolbox for use by bioengineers to create molecules with potentially revolutionary biological activities. In this review, we focus on the development of self-assembling nucleic acid nanostructures that are functionalized with nucleic acid aptamers and their great potential in wide ranging application areas.This article has not yet been cited by other publications.

Abstract:DNA origami nanotechnology has provided predictable static nanoarchitectures and dynamic nanodevices with rationally designed geometries, precise spatial addressability, and marked biocompatibility. Multiple functional elements, such as peptides, aptamers, nanoparticles, fluorescence probes, and proteins, etc. can be easily integrated into DNA origami templates with nanoscale precision, leading to a variety of promising applications. Triggered by chemical/physical stimuli, dynamic DNA origami nanodevices can switch between defined conformations or translocate autonomously, providing powerful tools for intelligent biosensing and drug delivery. In this minireview, we summarize the recent progress of dynamic DNA origami nanodevices with desired reconfigurability and feasibility to perform multiple biological tasks. We introduce varieties of DNA nanodevices that can be controlled by different molecular triggers and external stimuli. Subsequently, we highlight the recent advances in employing DNA nanodevices as biosensors and drug delivery vehicles. At last, future possibilities and perspectives are also discussed.

Abstract:The predictable nature of DNA interactions enables the programmable assembly of highly advanced 2D and 3D DNA structures of nanoscale dimensions. The access to ever larger and more complex structures has been achieved through decades of work on developing structural design principles. Concurrently, an increased focus has emerged on the applications of DNA nanostructures. In its nature, DNA is chemically inert and nanostructures based on unmodified DNA mostly lack function. However, functionality can be obtained through chemical modification of DNA nanostructures and the opportunities are endless. In this review, we discuss methodology for chemical functionalization of DNA nanostructures and provide examples of how this is being used to create functional nanodevices and make DNA nanostructures more applicable. We aim to encourage researchers to adopt chemical modifications as part of their work in DNA nanotechnology and inspire chemists to address current challenges and opportunities within the field.

Owing to their appealing properties such as precision in bottom-up fabrication, enhanced stability and cellular internalization, designed nucleic acid nanodevices (NANDs) have been widely explored in biomedical research. Compared with pristine nucleic acids, chemically modified NANDs exhibit a wider spectrum of applicability. In this Focus Review, we highlight recent advances of NANDs functionalized with small molecules, peptides/proteins or inorganic materials, and elucidate their promising future for biomedical application.

Abstract:<p>The revolutionary idea that DNA can play a non-biological role in a material world has indeed become today's reality. Although much attentions have recently been paid to "structural DNA nanotechnology" centered on DNA origami, due to the need for DNA computation, hybridization and modification that are usually not easily accessed, reproduced, or manipulated, preparation of nanostructures using DNA origami may have yet set a threshold for researcher to advance their fields with functional nanomaterials. It is suggested that, consisting of a few nucleotides (base pairs), or readily obtained by a simple procedure, non-origami DNA itself has sufficiently rich chemical functionality, and afford sequence-programmed growth of hybrid nano-architectures made of DNA and interactive materials with base-resolution control, and that the potential of non-origami DNA in building functional nanostructures has been underestimated. We have investigated an impressive number of fundamental and application-orientated studies in the late few years, and been preparing the review of the state of research in various subdisciplines of functional nanostructures based on non-origami DNA, which involve fundamental interactions of DNA-inorganic species, DNA nanofabrication, electronic nanodevices, functional metal nanoparticles and chiral nanostructures as well. Applications for these nanostructures have been covered in fields ranging from plasmonic biosensing and electrical nanodevices to optical bioimaging and photocatalysis. In this review, we will start with the fundamentals in DNA-based nanofabrication, including structure, characteristics and handling of DNA. Then, the capability of morphological control by simple DNA molecules, the resultant nanostructures and novel properties will be discussed and compared. Particularly, we emphasized the potential of single DNA molecules in templating the growth of nanostructures with different compositions, such as electrical polymer/metal nanowires, plasmonic nanostructures, fluorescent quantum dots and chiral silica nanostructures. Furthermore, the advanced applications of the non-origami DNA-based functional nanostructures in a range of fields will be figured out, including nanoelectronics, therapeutics and photocatalysis. Finally, conclusions will be drawn, and perspectives will be given. This review should be important for the development of DNA nanotechnology and DNA-based nanomaterials in the next decade, and creation of new nanomaterials based on simple DNA molecules, and their expanded applications as well.</p>

Abstract:Abstract DNA nanostructures are among the most fascinating self-assembled nanostructures in diverse areas of science and technology, because of their nanoscale precision in biomolecule and nanoparticle organization. The implementation of dynamic and spatial regulation in structural morphology and hierarchical assembly upon specific external stimuli will greatly expand their applications in biocomputation, clinical diagnosis, and cancer therapy. Recently, noncanonical nucleic acids, particularly DNA triplexes, i-motifs, and G-quadruplexes, have become powerful tools for biosensing and mechanical switching. Developments in incorporating stimuli-responsive noncanonical nucleic acids into DNA nanostructures provide a promising approach to regulating the spatial organization and hierarchical assembly of DNA nanostructures. In this review, we briefly introduce recent progress in constructing DNA nanostructures with dynamic regulation of the structural transformation and programmable assembly pathways at the nanometer scale by noncanonical nucleic acids and discuss their potential applications and challenges.

Abstract:Deoxyribonucleic acid (DNA) nanotechnology, a frontier in biomedical engineering, is an emerging field that has enabled the engineering of molecular-scale DNA materials with applications in biomedicine such as bioimaging, biodetection, and drug delivery over the past decades. The programmability of DNA nanostructures allows the precise engineering of DNA nanocarriers with controllable shapes, sizes, surface chemistries, and functions to deliver therapeutic and functional payloads to target cells with higher efficiency and enhanced specificity. Programmability and control over design also allow the creation of dynamic devices, such as DNA nanorobots, that can react to external stimuli and execute programmed tasks. This review focuses on the current findings and progress in the field, mainly on the employment of DNA nanostructures such as DNA origami nanorobots, DNA nanotubes, DNA tetrahedra, DNA boxes, and DNA nanoflowers in the biomedical field for therapeutic purposes. We will also discuss the fate of DNA nanostructures in living cells, the major obstacles to overcome, that is, the stability of DNA nanostructures in biomedical applications, and the opportunities for DNA nanostructure-based drug delivery in the future.

Abstract:Other than carrying the code of life, in the past three decades, DNA has played an important role in weaving two- and even three-dimensional nanostructures by taking advantage of its programmable sequences and precise recognition properties. The highly specific base-pairing interactions of DNA molecules not only allow for the building of static DNA nanostructures, but also enable the construction of machine-like devices capable of responding to external stimuli, such as pH, small molecules, light, electricity, and temperature, thus performing tasks otherwise beyond our reach. This review summarizes DNA-based switchable devices and materials, particularly where the responsiveness of DNA is caused by changes in secondary structures, and more importantly, where such stimuli-responsive processes are reversible. This review covers DNA-based devices and materials from smart surfaces and responsive nanopores/nanochannels to DNA hydrogels, which are expected to be designed with even more intelligence in the future.

Abstract:The blooming field of structural DNA nanotechnology harnessing the material properties of nucleic acids has attracted widespread interest. The exploitation of the precise and programmable Watson- Crick base pairing of DNA or RNA has led to the development of exquisite nucleic acid nanostructures from one to three dimensions. The advances of computer-aided tools facilitate automated design of DNA nanostructures with various sizes and shapes. Especially, the construction of shell or skeleton DNA frameworks, or more recently dubbed "framework nucleic acids" (FNAs) provides a means to organize molecules or nanoparticles with nanometer precision. The intrinsic biological properties and tailorable functionalities of FNAs hold great promise for physical, chemical, and biological applications. This Perspective highlights state-of-the-art design and construction, of precisely assembled FNAs, and outlines the challenges and opportunities for exploiting the structural potential of FNAs for translational applications.

Abstract:Apart from providing the material for genetic information storage, DNA has also been regarded as a useful building block in the field of nanotechnology, which is known as DNA nanotechnology. Based on the Watson-Crick base-pairing and the famous double helical structure, a library of DNA 2D or 3D nanostructures has been designed, constructed and characterized, by either tile-based or origami-based self-assembly. More significantly, other molecules/nanomaterials can be organized on those DNA ensembles to achieve highly ordered arrays or specific patterns in nanometer spatial resolution. In this review, we summarize recent progress in creating DNA nanoarchitectures, and discuss potential applications in DNA-based nanofabrication.

Abstract:The two doses, 5 and 7 mg/kg/day, were applied for periods of 14 or 28 days, with a 21 day break between two 14 days treatments in the latter case. Controls were only treated with physiological saline. A decrease in corporal weight and rectal temperature, as well as a decrease in ingestion and faeces together with urine increment were observed in the experimental animals during the first period of treatment. Normal levels were recovered during the break time, but the previous pattern reappeared during the second period of treatment. Suprarenals, liver, kidneys and spleen increased in some experimental groups when compared with controls whereas the thymus presented no significant differences. The studied organs showed signs of impairment.

Abstract:DNA nanotechnology is driven by the precision and programmable properties of DNA, which serves as a reliable template to guide the self-assembly of various materials, such as metal nanoparticles and polymers. By integrating DNA nanotechnology into the synthesis of plasmonic nanomaterials, unprecedented precision in structural control is achieved, facilitating the fabrication of pre-designed nanostructures with modulated optical functionalities. In this comprehensive review, we explore various synthesis techniques ( e.g. , DNA linker-based assembly, DNA origami-based organization, and DNA-mediated growth), focusing on advanced DNA-assembled plasmonic nanoarchitecture (DAPNA). The article highlights the applications of DAPNA in localized surface plasmon resonance (LSPR) and surface-enhanced Raman scattering (SERS) based biosensors for diagnostic purposes. The review not only addresses the limitations and solutions in DAPNA manufacturing technology but also explores potential applications in biosensing, tissue engineering, gene editing, immunotherapy, and other emerging fields.

Abstract:With the advent of DNA nanotechnology, DNA-based biomaterials have emerged as a unique class of materials at the center of various biological advances. Owing to DNA's high modification capacity via programmable Watson–Crick base-pairing, DNA structures of desired design with increased complexity have been designed. However, the lack of inherent functionality, along with low mechanical properties, high synthesis costs, and poor stability, limited the adaptability of DNA-based material toward complex biological systems. To impart functionality, DNA-based hybrid biomaterials synthesized by conjugating DNA with functional materials have been realized. Today, DNA-based hybrid materials have attracted significant attention in biological engineering with broad applicative prospects in biomedicine, clinical diagnosis, and nanodevices. Here, we summarize the recent advances in DNA-based hybrid materials with an in-depth understanding of general molecular design principles, functionalities, and applications. Finally, the challenges and prospects associated with DNA-based hybrid materials are discussed at the end of the review.

DNA nanotechnology has been rapidly developed and has branched into several significantly different directions.
