Abstract:Ten urinary biomarkers of organophosphate esters (OPEs) from six parent OPEs were analyzed in urine from adolescents students in Eastern China. Bis (1,3-dichloro-2-propyl) phosphate, urinary biomarker of tris (1,3-dichloro-2-propyl) phosphate (TDCIPP), was detected in all urine samples with the highest residual concentration of 6.2 ng/mL (median). Bis (1-chloro-propyl) hydroxyl-2-propyl phosphate, dibutyl phosphate, biomarkers of tris (1-chloro-2-propyl) phosphate (TCIPP), tri-n-butyl phosphate (TNBP), respectively, and tris (2-chloroethyl) phosphate (TCEP), were also frequently detected with median concentrations of 1.5, 2.6 and 0.3 ng/mL, respectively. Results indicate that exposure to OPEs, in particular to TDCIPP, TNBP, TCIPP and TCEP, was highly prevalent for adolescent students. The exposure pathways of OPEs were then evaluated according to the OPE internal body burdens (IBBs). Three pathways were identified as the main pathways for adolescents exposed to OPEs including dermal absorption, oral intake of food and dust and inhalation.
Association of exposure to organophosphate esters with increased blood pressure in children and adolescents

Abstract:Organophosphate esters (OPEs) are widely added to various industrial and consumer products, and are mainly used as flame retardants and plasticizers. Existing epidemiological studies suggest that OPE exposure may be linked to increased blood pressure (BP) and hypertension risk in adults. However, it remains unclear whether OPE exposure is associated with increased BP in children and adolescents. Here, we investigated the associations between OPE exposure and BP levels in 6-18-year-old children and adolescents from a cross-sectional study in Liuzhou, China. OPE metabolites were determined in spot urine samples (n = 1194) collected between April and May 2018. Three measurements of systolic and diastolic BP for each participant were averaged as study outcomes. Associations of OPE exposure with age-, sex- and height-standardized BP were assessed using linear regression models. We found that each natural log unit increment of bis(1,3-dichloro-2-propyl) phosphate (BDCIPP) was associated with a 0.06 standard deviation unit (95% confidant interval (CI): 0.01, 0.11) increase in systolic BP z-score. When conducting stratified analysis based on sex, age, and BMI category, BDCIPP was shown to be positively associated with systolic/diastolic BP z-score in females, but not in males. The associations between bis(2-butoxyethyl) phosphate (BBOEP) and systolic/diastolic BP z-score were pronounced in adolescents, but not in children. Moreover, a significant positive association between 1-hydroxy-2-propyl bis(1-chloro-2-propyl) phosphate (BCIPHIPP) and diastolic BP z-score was observed in obese subjects. The present study provides the first evidence that OPE exposure was related to increased BP in children and adolescents. Given the scarcity of high-quality evidence supporting these results, the health effects of OPEs are warrant investigation in well-designed prospective studies.
Exposure of children and mothers to organophosphate esters: Prediction by house dust and silicone wristbands

Abstract:Ubiquitous human exposure to organophosphorus tri-esters (tri-OPEs) has been reported worldwide. Previous studies investigated the feasibility of using house dust and wristbands to assess human OPE exposure. We hypothesized that these two approaches could differ in relative effectiveness in the characterization of children and adult exposure. In the participants recruited from Guangzhou, South China, urinary levels of major OPE metabolites, including diphenyl phosphate (DPHP) and bis(butoxyethyl) phosphate (BBOEP), were significantly higher in children than their mothers (median 6.6 versus 3.7 ng/mL and 0.11 versus 0.06 ng/mL, respectively). The associations of dust or wristband-associated OPEs with urinary metabolites exhibited chemical-specific patterns, which also differed between children and mothers. Significant and marginally significant associations were determined between dust concentrations of triphenyl phosphate (TPHP), tris(2-butoxyethyl) phosphate (TBOEP), trimethylphenyl phosphate (TMPP), or tris(1-chloro-2-propyl) phosphate (TCIPP) and their metabolites in children urine and between dust tris(1,3-dichloroisopropyl) phosphate (TDCIPP), TPHP or TMPP and urinary metabolites in mothers. By contrast, wristbands exhibited better efficiency of predicting internal exposure to TDCIPP. While both house dust and wristbands exhibited the potential as a convenient approach for assessing long-term OPE exposure, their feasibility requires better investigations via larger-scale studies and standardized sampling protocols.

Abstract:The percutaneous penetration and exposure risk of organophosphate esters (OPEs) from children’s toys remains largely unknown. Percutaneous penetration of OPEs was evaluated by EPISkinTM model. Chlorinated OPEs (Cl-OPEs) and alkyl OPEs, except tris(2-ethylhexyl) phosphate, exhibited a fast absorption rate and good dermal penetration ability with cumulative absorptions of 57.6–127 % of dosed OPEs. Cumulative absorptions of OPEs through skin cells were inversely associated with their molecular weight and log octanol-water partition coefficient. Additionally, a quantitative structure-activity relationship model indicated that topological charge and steric features of OPEs were closely related to the transdermal permeability of these chemicals. With the clarification of the factors affecting the transdermal penetration of OPEs, the level and exposure risk of OPEs in actual toys were studied. The summation of 18 OPE concentrations in 199 toy samples collected from China ranged from 6.82 to 228,254 ng/g, of which Cl-OPEs presented the highest concentration. Concentrations of OPEs in toys exhibited clear type differences. Daily exposure to OPEs via dermal, hand-to-mouth contact, and mouthing was evaluated, and dermal contact was a significant route for children’s exposure to OPEs. Hazard quotients for noncarcinogenic risk assessment were below 1, indicating that the health risk of OPEs via toys was relatively low.

Abstract:Human exposure to OPEs is pervasive and should be of great concern due to associations with adverse health effects, especially in susceptible populations. In this study, body burdens and exposure pathways of OPEs were investigated for 76 healthy elderly people in Jinan, China based on the measured OPE and metabolite concentrations in human bio-samples (whole blood and urine) and paired environmental matrices (air and dust), as well as food frequency questionnaire. Eight of 16 OPEs and 5 of 11 metabolites were detected in > 50% of whole blood and urine samples, respectively. Tri(1-chloro-2-propyl) phosphate (TCIPP), tris(2-chloroethyl) phosphate (TCEP), tri-phenyl phosphate (TPHP), and 2-ethylhexyl di-phenyl phosphate (EHDPP) were frequently detected and abundant in whole blood, while their corresponding metabolites were detected at low frequencies or levels in urine. The reduced metabolic and/or excretory capacity of elderly people may be an important reason, implying a higher health risk to them. Fourteen OPEs had over 50% detection frequencies in indoor air and dust, while 6 di-esters in indoor dust. Tris(2-ethylhexyl) phosphate (TEHP) in indoor dust and tri-n-butyl phosphate (TnBP) in indoor air were positively correlated with paired levels in blood but not with their metabolites (BEHP and DnBP) in urine. Combined with the direct intakes of BEHP and DBP from dust, blood is indicated as more suitable biomarker for TEHP and TnBP exposure. High consumption frequencies of several foods were associated with higher blood concentrations of three OPEs and urinary levels of four di-OPEs, indicating the importance of dietary exposure pathway. Estimated daily total intakes of OPEs via inhalation, dust ingestion, and dermal absorption ranged from 2.78 to 42.0 ng/kg bw/day, which were far less than the reference dosage values. Further studies were warranted to explore the potential health effects of OPE exposure in the elderly populations.

Organophosphate ester and phthalate ester metabolites in urine from primiparas in Shenzhen, China: Implications for health risks

Abstract:Organophosphate esters (OPEs) and phthalate esters (PAEs) are extensively used as additives in commercial and household products. However, knowledge on human exposure to OPEs and PAEs remains limited in China. This study aimed to investigate OPE and PAE metabolites in urine samples of primiparas and to evaluate the cumulative risk of OPE and PAE exposure. A total of 8 OPE metabolites and 11 PAE metabolites were measured in urine samples of 84 primiparas from Shenzhen, China. The OPE metabolites were found in at least 72% of the urine samples with bis(2-chloroethyl) phosphate (BCEP) being the dominant analogue. Among the 11 PAE metabolites, mono-n-butyl phthalate (mBP) was the most abundant analogue and had a median concentration (139 μg/L) greater than those reported in urine samples from other countries and regions. A significant, positive correlation was found between Σ8OPEMs (the sum of 8 OPE metabolites) and body mass index (BMI). The urinary concentration of Σ11PAEMs (the sum of 11 PAE metabolites) was positively associated with the time of computer using by the primiparas. The estimated daily intakes (EDI) of tris(2-chlorethyl) phosphate (TCEP, the parent chemical of BCEP) and di-n-butyl phthalate (DnBP, the parent chemical of mBP) were determined to be 0.47 and 9.14 μg/kg bw/day, respectively. The 95th percentile EDI values for TCEP and DnBP both exceeded their corresponding reference doses. Twelve and fifty-five percentage of the primiparas were estimated to have HIRfD (hazard index corresponding to reference doses) and HITDI (hazard index corresponding to tolerable daily intake) values exceeding 1 for OPEs and PAEs, respectively, suggesting a relatively high exposure risk.

Abstract:Toxicologic studies reported that organophosphate esters (OPEs) may disrupt lipid metabolism, thus affecting serum lipid levels. However, epidemiological evidence regarding the association between OPEs and the risk of hyperlipidemia (HPL) as well as serum lipid levels is scarce. In the present study, our aim was to investigate the impact of individual and mixed OPE exposure on HPL. A total of 1981 Chinese adults were involved based on a cross-sectional design. Overall, we found a positive association between bis(1,3-dichloro-2-propyl) phosphate (BDCIPP) and the risk of HPL. Bis(1-chloro-2-propyl) phosphate (BCIPHIPP) showed a positive association with total cholesterol (TC) and low-density lipoprotein cholesterol (LDL-C). BDCIPP, diphenyl phosphate (DPHP), di-ocresyl phosphate and di-p-cresyl phosphate (Docp&Dpcp), and 4-hydroxyphenyl-diphenyl phosphate (4-OH-DPHP) exhibited a negative association with high-density lipoprotein cholesterol (HDL-C). In stratified analyses, BDCIPP and BCIPHIPP were significantly correlated with the increased risk of HPL in the age ≤ 45 group. Bis(2-butoxyethyl) phosphate (BBOEP) was in relationship with an elevated risk of HPL in the subgroup of BMI < 24 kg/m2. BDCIPP was also positively associated with HPL in men. Quantile-based g computation (qgcomp) and generalized weighted quantile sum regression (gWQS) models demonstrated a negative association between OPEs mixed exposure and HDL-c in the total population, as well as a positive effect of them on HPL in the subgroup of age ≤ 45 years, which is consistent with the individual analyses. Furthermore, joint effect analyses revealed that participants with detected BDCIPP urinary levels and unhealthy lifestyles had the highest risk of HPL. Our findings offer evidence supporting the correlation between exposure to OPE and the risk of HPL, necessitating further prospective studies for validation.
Organophosphate esters in a cohort of pregnant women: Variability and predictors of exposure

Abstract:Organophosphate esters (OPEs) are a group of chemicals used as flame retardants and plasticizers that replaced polybrominated diphenyl ethers in consumer products such as furniture and electronics. To characterize exposure to OPEs during fetal development, we measured urinary OPE metabolite concentrations in women twice during pregnancy (16 and 26 weeks' gestation) and at delivery (n = 357). We also previously quantified house dust OPE parent compound concentrations at 20 weeks' gestation (n = 317). Diphenyl phosphate (DPHP) had the highest geometric mean urinary concentrations (1.5-2.3 μg/g creatinine), followed by bis(1,3-dichloro-2-propyl) phosphate (BDCIPP; 0.75-0.99 μg/g creatinine), and bis(2-chloroethyl) phosphate (BCEP; 0.72-0.97 μg/g creatinine), while dibutyl phosphate (DNBP) had the lowest concentrations (0.25-0.28 μg/g creatinine). Urinary OPE metabolites were moderately correlated with each other at 26 weeks (rs: 0.23-0.38, p < 0.001) while the correlations at 16 weeks and delivery were slightly weaker. Intra-class correlations for urinary metabolites measured at three time points were poor (0.16-0.34), indicating high variability within individuals. Dust concentrations of OPE parent compounds were associated with BCEP, BDCIPP, and DPHP concentrations in urine at some but not all time points. In linear mixed models of urinary OPE metabolite concentrations, household size was inversely associated with BCEP concentrations, and being non-white was associated with lower BDCIPP and DPHP concentrations. Urine samples collected in the summer had the highest OPE metabolite concentrations. This study highlights the need to collect multiple urine samples during pregnancy to define exposure patterns and investigate potential periods of susceptibility.

Abstract:Calreticulin is a ubiquitously expressed protein, which has been implicated in a large number of cellular functions, including calcium storage and signaling, protein folding, and cell attachment. To examine the role of calreticulin during in vivo development, mice deficient in calreticulin were generated by targeted inactivation of the calreticulin gene. Calreticulin-deficient mutants die in utero, mostly in late gestation. Half of these embryos had decreased cardiac cell mass, associated with increased apoptosis of cardiac myocytes. In vitro differentiation cultures of calreticulin-deficient embryonic stem cells resulted in fewer embryoid bodies with contractile activity than cultures derived from calreticulin +/- stem cells (P < 0.001). Sixteen percent of the mutants exhibited exencephaly secondary to a defect in neural tube closure. Embryos surviving until Embryonic Day 16.5 had omphalocele. Lack of calreticulin did not influence survival of embryonic fibroblasts under various endoplasmic reticulum stress conditions. However, calreticulin did influence cell migration in a calcium- and substrate-dependent manner. We conclude that calreticulin is not essential during the early stages of embryonic development, but is important for the development of heart and brain and for ventral body wall closure. The observed abnormalities are compatible with a role of calreticulin in the modulation of cellular calcium signaling.
Spatial occurrence and composition profile of organophosphate esters (OPEs) in farmland soils from different regions of China: Implications for human exposure

Abstract:The environmental load of organophosphate ester (OPE) flame retardants has caused a series of problems due to their extensive use. The soil matrix, as an ultimate sink for organic pollution, plays a vital part in the fate of OPEs in the environment. In this study, the spatial occurrence, composition profile and health risk of 13 OPE species in farmland soils from four provinces of China were characterized. Excluding tris(2,3-dibromopropyl) phosphate (TDBPP) and ethylhexyl diphenyl phosphate (EHDPP), the remaining eleven OPEs had a high detection frequency (DF) ranging from 60% to 100%. The range of total OPE (ΣOPE) concentrations were 62.3-394 ng/g dry weight (dw), with a median of 228 ng/g dw. Among these OPEs, tris(2-ethylhexyl) phosphate (TEHP) with a median of 143 ng/g dw) was the predominant species, followed by tricresyl phosphate (TCP; median of 20.1 ng/g dw) and tris(2-chloroethyl) phosphate (TCEP; median of 17.9 ng/g dw). In terms of geographical distribution, significantly lower OPEs levels were found in samples from Heilongjiang (159±47.0 ng/g dw) than in those of Guangxi (264±66.0 ng/g dw), Henan (252±74.5 ng/g dw) and Hubei (242±52.8 ng/g dw) provinces. Principal component analysis and Spearman's correlations were used to reveal potential sources of OPEs in the different provincial regions. Health risk exposure to OPEs in farmland soils was at an acceptable level (<1.20×10−5 for non-carcinogenic risk to children as the most sensitive age group; and <6.47×10−10 for carcinogenic risk to adults as the most sensitive age group) at the present detected concentrations. However, TCEP and TEHP, the predominant risk contributors, should be paid more attention.

Abstract:Human exposure to organophosphate esters (OPEs) is more pervasive in industrial areas manufacturing OPE-related products. OPE exposure is of great concern due to its associations with adverse health effects, while studies on OPE exposure in industrial districts are scarce. This study aimed to assess human exposure to OPEs in a typical industrial area producing large amounts of OPE-related products in Shenzhen, China. Tris (2-chloroethyl)-phosphate (TCEP), tris (2-chloroisopropyl) phosphate (TCPP) and other common OPEs were analyzed in urine (n = 30) and plasma (n = 21) samples. Moreover, we measured five OPE metabolites (mOPEs) in plasma samples (n = 21). The results show that TCPP and TCEP are dominant compounds, with moderate to high levels compared with those reported in urine and plasma samples from other regions. In addition, di-n-butyl phosphate (DnBP) and diethyl phosphite (DEP) were frequently detected in plasma samples and could be considered as biomarkers. Risk assessment revealed a moderate to high potential health risk from TCEP exposure. Our results provide basic data for human exposure to OPEs in industrial areas and call for the prevention and mitigation of industrial chlorinated OPE pollution.

Abstract:Sixty-eight paired samples of urban surface dust and soil as well as four samples of atmospheric dustfall were collected from the arid city of Urumqi in Northwest China. Thirteen organophosphate esters (OPEs) in these samples were analyzed for the characteristics, sources, bio-accessibility, and health risks of OPEs. The studied OPEs were widely detected in the urban surface dust, soil, and dustfall, with Σ13OPEs (total concentration of 13 OPEs) of 1362, 164.0, and 1367 ng/g, respectively, dominated by tris(2-chloroethyle) phosphate (TCEP), tri(2-chloroisopropyl) phosphate (TCiPP), tri(1, 3-dichloroisopropyl) phosphate (TDCiPP) and tris(2-butoxyethyl) phosphate (TBOEP), TBOEP and tri(2-ethylhexyl) phosphate (TEHP), and TCEP, TCiPP, TBOEP, triphenyl phosphate and TEHP, respectively. The low and high frequency magnetic susceptibility of surface dust and urban soil might indicate the pollution of OPEs in them. Elevated levels of the Σ13OPEs in the surface dust and urban soil were found in the west, south, and northeast of Urumqi city. The total deposition flux of dustfall-bound 13 OPEs ranged from 86.5 to 143 ng/m 2 /day, with a mean of 105 ng/m 2 /day. OPEs in the surface dust and urban soil were associated with the emissions of indoor and outdoor products containing OPEs, the dry and wet deposition of atmosphere, and the emissions of traffic. Trimethyl phosphate, triethyl phosphate, tripropyl phosphate, tri-isobutyl phosphate, TCEP, TCiPP, TDCiPP, and TBOEP in surface dust and urban soil had relatively high bio-accessibility. The bio-accessibility of OPEs was mainly affected by the physio-chemical properties of OPEs. The non-cancer and cancer risks of human exposure to OPEs in surface dust and urban soil were relatively low or negligible. The current research results may provide scientific supports for prevention and control of pollution and risks of OPEs.

Abstract:Organophosphate esters (OPEs) are an emerging class of chemicals used in a variety of consumer products as flame retardants, plasticizers, and additives. While prior epidemiologic studies suggest that OPEs may impact respiratory health, results remain inconclusive. We examined associations between urinary biomarkers of OPEs and symptoms of respiratory morbidity in a panel study of 147 predominantly Black school-aged children with asthma living in Baltimore City, Maryland. The study consisted of up to four seasonal, week-long, in-home visits where urine samples and self-reported asthma symptoms were collected on days 4 and 7 (nsamples = 438). We quantified concentrations of nine urinary OPE biomarkers: bis(2-chloroethyl) phosphate (BCEtp), bis(1-chloro-2-propyl) phosphate (BCPP), bis(1,3-dichloro-2-propyl) phosphate (BDCIPP), di-n-butyl phosphate (DBuP), di-benzyl phosphate (DBzP), di-o-cresylphosphate (DOCP), di-p-cresylphosphate (DPCP), di-(2-propylheptyl) phthalate (DPHP), and 2,3,4,5-tetrabromo benzoic acid (TBBA). We estimated prevalence odds ratios (POR) of respiratory morbidity symptoms using logistic regression with generalized estimating equations to account for our repeated measure design. We assessed BDCIPP and DPHP as continuous (log2) concentrations and dichotomized exposure of BCEtP, DBuP, and DPCP (detect vs non-detect) based on their lower detection frequencies. We adjusted models for season, visit day, age, gender, caregiver education, health insurance type, exposure to household smoking, atopy, and PM2.5. Higher DPHP concentrations were significantly associated with odds of daytime symptoms (POR: 1.26; 95% CI: 1.04-1.53; p = 0.02) where daytime symptoms consisted of trouble breathing due to asthma, reporting bother caused by asthma, and/or limitation in activities due to asthma. DBuP detection was associated with use of rescue medication on the day of sample collection (POR: 2.36; 95% CI: 1.05-5.29; p = 0.04). We also observed several consistent, albeit non-significant (p > 0.05), positive associations for BCEtP and DPCP and respiratory morbidity measures. This is the first study to evaluate the relationship between OPE biomarkers and respiratory morbidity symptoms in children with asthma, and findings suggest that further studies are warranted to confirm whether these associations are causal.

Abstract:Stevioside (250 mg capsules) was given three times daily to 10 healthy subjects. Steviol glucuronide (steviol 19-O-beta-D-glucopyranosiduronic acid; MM, 494.58; melting point, 198-199 degrees C) was characterized in the 24 h urine as the only excretion product of oral stevioside by MS, NMR, IR, and UV spectroscopy. This is the first report on the unambiguous identification of steviol glucuronide in human urine.

Abstract:Electronic waste (e-waste) is a well-known source of plastic additives in the environment. However, the e-waste-related occupational exposure to organophosphite antioxidants (OPAs) and the relevant oxidation products—novel organophosphate esters (NOPEs)—via different pathways is still unknown. In this study, six OPAs and three NOPEs were measured in 116 dust and 43 hand-wipe samples from an e-waste dismantling area in Central China. The median concentrations of ΣOPAs and ΣNOPEs were 188 and 13,900 ng·g−1 in workshop dust and 5,250 ng·m−2 and 53,600 ng·m−2 on workers' hands, respectively. The increasing concentrations of dust in the form of triphenyl phosphate (TPHP) (p < 0.01) and tris(2,4-di-tert-butylphenyl) phosphate (AO168 = O) (p < 0.05) were strongly associated with the corresponding concentration on workers' hands. Furthermore, men had significantly lower levels of NOPEs on their hands than did women (p < 0.01). Moreover, the hand wipe levels of AO168 = O (41,600 ng·m−2) was significantly higher than that of the typical OPE (TPHP, 7370 ng·m−2), and the hand-to-mouth contact (ΣOPAs, 9.48 ng·kg bw-1·day−1; ΣNOPEs, 109 ng·kg bw-1·day−1) was a more significant and integrated pathway than dust ingestion (ΣOPAs, 0.10 ng·kg bw-1·day−1; ΣNOPEs, 5.01 ng·kg bw-1·day−1) of e-waste related occupational exposure to these "new" chemicals.

Abstract:Organophosphate esters (OPE) are flame retardants and plasticizers used in a wide range of consumer products. Despite their widespread use, few studies have characterized pediatric exposures. We assessed variability and predictors of OPE exposures in a cohort panel study of 179 predominantly Black school-aged children with asthma in Baltimore City, MD. The study design included up to four seasonal week-long in-home study visits with urine sample collection on days 4 and 7 of each visit (nsamples = 618). We quantified concentrations of 9 urinary OPE biomarkers: bis(2-chloroethyl) phosphate (BCEtp), bis(1-chloro-2-propyl) phosphate, bis(1,3-dichloro-2-propyl) phosphate (BDCPP), di-benzyl phosphate (DBuP), di-benzyl phosphate, di-o-cresylphosphate, di-p-cresylphosphate (DPCP), di-(2-propylheptyl) phthalate (DPHP), 2,3,4,5-tetrabromo benzoic acid. We assessed potential predictors of exposure, including demographic factors, household characteristics, and cleaning behaviors. We calculated Spearman/tetrachoric correlations and intraclass correlation coefficients (ICCs) to examine within-week and seasonal intra-individual variability, respectively. We assessed OPE predictors using linear models for continuous log2 concentrations (BDCPP and DPHP) and logistic models for odds of detection (BCEtP, DBuP, DPCP), with generalized estimating equations to account for repeated measures. For all OPEs, we observed moderate within-week correlations (rs: 0.31-0.63) and weak to moderate seasonal reliability (ICC: 0.18-0.38). BDCPP and DPHP concentrations were higher in the summer compared to other seasons. DPHP concentrations were lower among males than females (%diff: -53.5%; 95% CI: -62.7, -42.0) and among participants spending >12 h/day indoors compared to ≤12 h (%diff: -20.7%; 95% CI: -32.2, -7.3). BDCPP concentrations were lower among children aged 8-10 years compared to 5-7 years (%diff: -39.1%; 95% CI: -55.9, -15.9) and higher among children riding in a vehicle on the day of sample collection compared to those who had not (%diff: 28.5%; 95% CI: 3.4, 59.8). This study is the first to characterize within-week and seasonal variability and identify predictors of OPE biomarkers among Black school-aged children, a historically understudied population.

Abstract:Although diet is regarded as a major exposure source of organophosphate esters (OPEs), the dietary survey of OPEs in China has been limited. Based on the sixth Chinese Total Diet Study (TDS) conducted during 2016-2019 in 24 of 34 provinces in China, 14 OPEs were detected in 96 food composites from four animal-origin food categories. Twelve OPEs were detected in more than 80% of the samples and 2-ethylhexyl diphenyl phosphate (EHDPP) presented the highest median concentration (1.63 ng/g wet weight (ww)). The most contaminated food composite was meat, with a median ∑14OPEs of 13.6 ng/g ww, followed by aquatic food (11.5 ng/g ww), egg (7.63 ng/g ww), and milk (3.51 ng/g ww). The contribution of the meat group was close to or even greater than 50% in the estimated dietary intake (EDI) of OPEs. The average (range) EDI of the ∑14OPEs via animal food consumption for a Chinese "standard man" was 34.4 (6.18-73.3) ng/kg bodyweight (bw)/day. The geographical distribution showed higher EDI in southern coastal provinces compared to the northern inland provinces. Nevertheless, the highest EDI of ∑14OPEs from animal food was still more than 10 times lower than the reference dose. This is the first national survey of OPEs in foods from China.
Young infants' exposure to organophosphate esters: Breast milk as a potential source of exposure

Abstract:Organophosphate esters (OPEs) are applied as both flame retardants and plasticizers to a variety of consumer items such as home furnishings, construction materials, and children's products. While some assessments have characterized exposure among toddlers and young children, little is known about the OPE exposure among infants, who are a vulnerable population due to their rapid development. Here, we collected spot urine samples from 6-week-old (n = 100) and 12-month-old infants (n = 63), with about half of the infants evaluated at both ages (n = 52), to characterize OPE exposure and determine what factors contributed to higher exposures. Five of six OPE metabolites analyzed were detected frequently (>70%). Diphenyl phosphate was detected in every urine sample, while bis(2-chloro-isopropyl) phosphate (BCIPP) was the most abundant metabolite measured overall. Concentrations of bis(1-chloro-2-propyl) 1-hydroxy-2-propyl phosphate (BCIPHIPP) and BCIPP [i.e., metabolites of tris(2-chloro-isopropyl) phosphate (TCIPP)] were significantly greater among 6-week-old infants compared to 12-month-olds, while levels of other OPE metabolites were not statistically different in the first year of life. OPE metabolites were generally correlated with one another in samples collected at each age (rs = 0.25-0.75; p < 0.05), and except BCIPHIPP, concentrations of the same metabolite were correlated over time (rs = 0.41-0.53; p < 0.05). Breastfeeding at 6 weeks of age and owning a larger number of children's products were associated with increased concentrations of urinary BDCIPP. Infants who were currently receiving breast milk had higher levels of TCIPP metabolites; urinary BCIPP concentrations were 6.2 times higher in infants receiving breast milk at 6 weeks of age, and BCIPHIPP levels were 2.2 times higher for 12-month-old infants receiving breast milk (10β = 7.2; 95% CI: 1.6-32.1 and 10β = 3.2; 95% CI: 1.2-8.1, respectively). Differences in the predominant TCIPP metabolite associated with breastfeeding may suggest differences in metabolism with age. Cumulatively, our results suggest levels of OPE exposure may be higher for infants than other age groups, including toddlers and older children.
