Abstract:The study was conducted to investigate the effects of replacing antibiotics using multi-enzyme preparations on growth performance, coefficient of total tract apparent digestibility, digestive enzyme activity, and antioxidant property in piglets. A total of 160 piglets ((21.35±0.22) kg) were randomly assigned to five dietary treatments: 1) basal diet supplemented with antibiotics (AC), 2) antibiotic diet supplemented with 0.5 g kg−1 multi-enzyme preparations (AC+0.5EP), 3) antibiotic diet supplemented with 1.5 g kg−1 multi-enzyme preparations (AC+1.5EP), 4) basal diet supplemented with a half dosage of antibiotics and 1.5 g kg−1 multi-enzyme preparations (AH+1.5EP), and 5) basal diet supplemented with 1.5 g kg−1 multi-enzyme preparations (BC+1.5EP). The results showed that AC+1.5EP significantly improved the feed efficiency, apparent digestibility of ether extract (EE) and crude ash (CA), lipase activity in pancreas and duodenum content, maltase and lactase activity in jejunum and ileum mucosa, glutathione peroxidase (GSH-Px) concentration in serum and liver, and decreased malondialdehyde (MDA) concentration in serum and liver compared with piglets receiving AC (P<0.05). Piglets receiving BC+1.5EP showed no significant difference in growth performance (P>0.05) but had lower MDA concentration than piglets receiving AC (P<0.05). The apparent digestibility of EE and crude fiber (CF), duodenal lipase activity, jejunum mucosa maltase, and ileum mucosa lactase activity of piglets receiving AH+1.5EP or BC+1.5EP were significantly improved compared with piglets receiving AC (P<0.05). These results indicated an additive growth promotion effect between antibiotics and multi-enzyme preparations on piglets, and the multi-enzyme preparations may be used as substitutes for antibiotics for improving piglet production performance and health status.
Antibiotic Resistance Profiles of Salmonella Recovered from Finishing Pigs and Slaughter Facilities in Henan, China.

Abstract:With the increase in commercial pig farming, there is a simultaneous increase in the use of antibiotics for prophylaxis as well as therapeutics in China. In this study, we evaluated the prevalence and resistance diversity of salmonellae isolated from feces of asymptomatic, live and slaughtered pigs. We analyzed 1,732 pig fecal samples collected over 8 months, at Henan province of China. The salmonellae were isolated and identified by PCR. They were serotyped using commercial antisera and assayed for the MIC of 16 antibiotics by broth microdilution method. The average prevalence of Salmonella was 19.4% (95% CI: 17.6–21.4). Large farms (herd size ≥1,000) were found to have a higher prevalence as compared to the small- and medium-scale farms (p < 0.0001). The prevalence of salmonellae in samples collected from the farms [11.77% (95% CI: 10.1–13.6)] and from the slaughterhouse [45.23% (95% CI: 40.3–50.30)] was statistically different (p < 0.0001). Uncommon serovars of Salmonella such as Agama and common serovars such as Derby and Typhimurium were isolated. High resistance (>80%) was recorded toward ciprofloxacin (100%), tetracycline (99.4%), doxycycline (97%), sulfamethoxazole (85.8%), ampicillin (81.6%), and amoxicillin (80.4%). Multidrug resistance (MDR) to four, five, and seven classes of antibiotics was recorded to be approximately 25% in the most prevalent serovar like Derby. We conclude that the presence of alarmingly high resistance, toward the critical antibiotics such as fluoroquinolones and beta-lactams, in large swine farms in China, should draw public attention. These results highlight the need for continued antibiotic stewardship programs for judicious use of critical antibiotics in animal health as well as for producing safe pork.

Abstract:The goal of this study is to describe the current use of antibiotics in the European pig industry based on an extensive literature review. To achieve this, an overview of results from national (n = 15) and multi-country (n = 2) cross-sectional and longitudinal (n = 2) surveys, which describe antimicrobial use in pigs, is presented. Results are further linked to the outcome of the European Surveillance of Veterinary Antimicrobial Consumption (ESVAC) project. Overall, it was found that weaned piglets received the most antibiotics, followed by suckling piglets resulting in over 80% of the treatments being administered to animals before 10 weeks of age. Furthermore, it was observed that antibiotic use (ABU) was significantly associated across age categories, indicating that farms with a high use in piglets also used more antibiotics in their finishers. This may, among other things, be explained by farmers' habits and behavior. However, above all, the studies showed surprisingly large differences in ABU between the countries. These differences may be related to the differences in disease prevalence and/or differences in the level of biosecurity. However, they may also reflect variations in rules and regulations between countries and/or a difference in attitude towards ABU of farmers and veterinarians that are not necessarily linked to the true animal health situation. Furthermore, it was observed that already a substantial proportion of the European pig production is able to successfully raise pigs without any group treatments, indicating that it is possible to rear pigs without systematic use of antibiotics. Based on the ESVAC data, a decline of 43.2% was observed in sales of antibiotics for animals in Europe between 2011 and 2020. To enable efficient antimicrobial quantification and stewardship, 15 European countries have already established systems for herd level monitoring ABU in pigs.

Abstract:Abstract China has a long history of pig rearing, and it currently raises and consumes approximately half of the pigs in the world. Major improvements have been made in pig farming in China in the last four decades with the growing application of new livestock farming technologies. Among the new improvements, the use of antibiotics in pig farming is a common but not well‐documented practise. In order to understand the behaviour of the farmers regarding antibiotic use in pig farming, we conducted a household survey in four townships of L County in Yunnan Province, China, during August 2014 and April 2015. In this survey, 404 farmer households were interviewed using a questionnaire. Among the farmers interviewed, 89% reported easy access to antibiotics, 83.7% reported experience of self‐purchasing antibiotics, and 40.3% reported that they often used antibiotics in pig farming mainly for the prevention and treatment of pig diseases. These farmers identified 20 antibiotics that they had used in pig farming 6 months before the survey. Of these, 11 and 8 antibiotics have been categorised under ‘critically important’ and ‘highly important’ antimicrobial groups, respectively, by the World Health Organization (WHO), and 12 and 8 have been categorised under the ‘Watch’ and ‘Access’ groups, respectively, as per the 2019 WHO AWaRe classification of antibiotics. Factors associated with the behaviour of self‐purchasing antibiotics included types of farms, sources of antibiotics, and previous experiences of pig diseases: those who were smallholders, buying antibiotics from veterinary drugstores and village vets, and whose pigs had suffered diseases previously were more likely to self‐purchase antibiotics for their pigs. Farmers who cleaned their pigsties less frequently and those whose pigs had suffered from diseases used antibiotics more frequently as compared to their peer farmers.

Abstract:Pigs reared on many farms are mass-medicated for short periods with antibiotics through their drinking water to control bacterial pathogen loads and, if a disease outbreak occurs, to treat pigs until clinical signs are eliminated. Farm managers are responsible for conducting in-water antibiotic dosing events, but little is known about their dosing practices. We surveyed managers of 25 medium to large single-site and multi-site pig farming enterprises across eastern and southern Australia, using a mixed methods approach (online questionnaire followed by a one-on-one semi-structured interview). We found wide variation in the antibiotics administered, the choice and use of dosing equipment, the methods for performing dosing calculations and preparing antibiotic stock solutions, the commencement time and duration of each daily dosing event, and the frequency of administration of metaphylaxis. Farm managers lacked data on pigs’ daily water usage patterns and wastage and the understanding of pharmacology and population pharmacometrics necessary to optimize in-water dosing calculations and regimens and control major sources of between-animal variability in systemic exposure of pigs to antibiotics. There is considerable scope to increase the effectiveness of in-water dosing and reduce antibiotic use (and cost) on pig farms by providing farm managers with measurement systems, technical guidelines, and training programs.

Abstract:Stewardship of antibiotics used in livestock production has come under increasing scrutiny, from both the animal welfare point of view and due to concerns that antibiotic use in livestock may pose a risk to human health through selection pressure to drive development of antibiotic resistant strains of bacteria. Despite this concern, however, antibiotic consumption in the sheep sector is currently poorly described. This study determines the range and quantities of antibiotics used in the Northern Irish (NI) sheep flock as well as exploring drivers for their use. A mixed-methods approach was utilised, with an anonymous online scoping survey, analysis of the medicine records from 52 NI sheep farms and semi-structured interviews undertaken with 27 farmers. Eighteen farmers contributed both records and participated in interviews. Veterinary medicine records were derived from two sources: on-farm medicine books (seven) or veterinary practice sales data (51). As six of these farmers provided information from both sources a total of 52 unique farms participated. Overall, antibiotic use in sheep on the 52 farms sampled was low, with a median value of 11.35 mgPCU-1 (mean 13.63 mgPCU-1, sd 10.7; range 0-45.29 mgPCU-1), with all farms below 50 mgkg-1. Critically important antibiotics accounted for 0.21% of all antibiotics purchased. Lameness was the main driver of antibiotic use identified by this study. Others included a range of prophylactic treatments such as oral antibiotics to prevent watery mouth, injectable antibiotics to prevent abortion and following assisted lambing. Farmers acknowledged some of these uses had become habitual over time. The veterinary medicine sales records demonstrated significant sales of antibiotics not authorised for use in sheep, on an ongoing, rather than case-by-case, basis. Farmers were positive about their veterinarian's ability and knowledge to improve flock welfare and productivity, but were unwilling to pay for this advice. However, veterinarians may have facilitated weak medicine stewardship through a failure to adequately challenge farmers seeking antibiotics. Farmers did not maintain accurate or up-to-date on farm medicine or production records in the majority of cases. Despite this lack of on-farm recording, veterinary sales records can be studied in consultation with farmers to provide veterinarians with a farm-specific insight into antibiotic use and related attitudes and behaviours. Farmers and veterinarians can then identify areas and behaviours to target collaboratively, improving antibiotic and wider medicine stewardship, whilst simultaneously improving flock health and productivity.

Abstract:OBJECTIVE: To describe how various antimicrobials are used in commercial pig herds in Australia and for what disease conditions.PROCEDURE: Managers of large pig herds (> 200 sows) across Australia and their veterinarians participated in an internet-based survey in 2006. Questions were asked about herd management, the occurrence of bacterial diseases and the type and frequency of antimicrobial use. An antimicrobial usage index for each herd was derived as a summary of the risk of selection for antimicrobial resistance. Relationships between responses were explored with univariate and multivariate analysis.RESULTS: Responses were received for 197 herds estimated to represent at least 51% of all large pig herds in Australia. Most piggeries relied on drugs of low importance in human medicine (e.g. tetracyclines, penicillins and sulfonamides). For the two drugs of high importance in human medicine that can be legally prescribed to pigs in Australia, ceftiofur use was reported in 25% of herds and virginiamycin in none. Infections attributed to Lawsonia, Mycoplasma and Escherichia coli motivated the most use of antimicrobials. No useful association was found between management factors and the antimicrobial use index.CONCLUSION: Most antimicrobial use in the Australian pig industry is based on drugs of low importance to public health. Enhanced control of E. coli infections without reliance on antimicrobials would further reduce the risk of selecting for antimicrobial resistance relevant to public health. The amount of variation in the usage index between herds suggests that antimicrobial use should be constantly reviewed on a herd by herd basis.

Abstract:This paper explores the theoretical economic outcome of management changes that result in different levels of antimicrobial use (AMU) in two types of UK pig farm. A static farm economic pig production model (FEPM) was used on a representative 'Top-third' most profitable farm and a representative 'Mid-range' profitable farm. Three AMU theoretical management scenarios were investigated; (a) management changes leading to a reduction of AMU by 35% (AMU35); (b) more extensive management changes leading to a reduction of AMU by 95% (AMU95); and (c) implementing depopulation of the herd (AMU Depop). A sensitivity analysis was conducted to determine the effect of increases or decreases in pig revenue and feed price on farm gross margin under these scenarios. Over a single year, the AMU35 scenario was estimated to have a small positive impact (+3%) on both farm types. The other two AMU reduction scenarios had higher AMU reduction on farms but required higher variable cost and hence they resulted in lower farm profitability. There was a substantial reduction (up to -50%) in farm gross margin under these two AMU reduction scenarios in the modeled short-term time-period. The impact of the alternative AMU scenarios was slightly higher on a farm representing the 'Top-third' farm type, reducing farm gross margin further by 7% compared to the 'Mid-range' farm. Nevertheless, both farm types stay profitable under all three AMU scenarios. The results showed that in the modeled short-term implementing management changes that result in a reduction of on-farm AMU by 35% had a good economic outcome. In practice, the other two scenarios would be considered as longer-term strategies. Although both require higher initial costs to implement, the improved biosecurity and hygiene will benefit from lower disease occurrence for a longer term. Farm gross margins were, however, found to be highly sensitive to changes on market prices especially increasing feed prices. An increase of more than 15% in feed price moved a profitable farm into a loss-making farm. It will be economically challenging for uptakes of these, or similar, AMU reduction scenarios on farms if the market prices become un-favorable to pig farmers.
Antibiotic Inclusion in the Diet Did Not Alter the Standardized Ileal Digestible Tryptophan to Lysine Ratio for Growing Pigs

Abstract:Two 21-d experiments were conducted to determine the optimum standardized ileal digestible (SID) Trp:Lys ratio for growing pigs; 1 experiment fed diets supplemented with or without an antibiotic. The primary response variables in both experiments were ADG, ADFI, G:F, and plasma urea N (PUN) concentrations with the optimum SID Trp:Lys ratio detected using broken-line analysis. Experiment 1 evaluated the optimum SID Trp:Lys ratio in growing pigs fed diets supplemented with an antibiotic. This experiment used 120 crossbred pigs that were blocked by sex and initial BW (24.13 ± 2.72 kg) and allotted to 6 SID Trp:Lys ratios in 4 replicates. Dietary treatments were formulated by the addition of crystalline Trp to create 6 SID Trp:Lys ratios (13.08%, 14.06%, 15.04%, 17.00%, 18.95%, and 20.91%) with a constant SID Lys level of 0.655%. As SID Trp:Lys ratios increased, ADG, ADFI, and G:F increased, and PUN concentrations decreased linearly ( < 0.05) and quadratically ( < 0.05). Linear broken-line analysis yielded optimum SID Trp:Lys ratios of 17.93% ( < 0.001) and 16.17% ( = 0.009) for ADG and PUN, respectively, resulting in a mean optimum SID Trp:Lys ratio of 17.05%. Experiment 2 evaluated the optimum SID Trp:Lys ratio in growing pigs fed diets supplemented with or without an antibiotic. It used a total of 324 crossbred pigs (initial BW: 30.81 ± 3.56 kg) that were allotted to 6 SID Trp:Lys ratios in 6 replicates. Dietary treatments were formulated by the addition of crystalline Trp to create 6 SID Trp:Lys ratios (12.52%, 14.86%, 17.20%, 19.54%, 21.88%, and 24.22%) with a constant SID Lys level of 0.67%. As SID Trp:Lys ratios increased, ADG, ADFI, and G:F increased, and PUN concentrations decreased linearly ( < 0.001) and quadratically ( < 0.001) regardless of antibiotic inclusion. There were no differences by the antibiotic treatment in ADG, ADFI, G:F, or PUN concentrations ( > 0.49) and no interactions between antibiotics and Trp:Lys ratios ( > 0.29). When the data for all pigs were pooled for the various Trp:Lys ratios, the optimum SID Trp:Lys ratios for ADG and PUN based on linear broken-line analysis were 14.58% ( < 0.001) and 14.54% ( < 0.001), respectively, resulting in an optimum SID Trp:Lys ratio of 14.56% as the mean of the determined optima for ADG and PUN responses. These results demonstrate that the optimum SID Trp:Lys ratio for 30- to 50-kg growing pigs is not impacted by the dietary inclusion of an antibiotic as long as the diets are formulated on an SID AA basis.
Trends in Antimicrobial Usage on Swiss Pig Farms from 2018 to 2021: Based on an Electronic Treatment Journal

Abstract:(1) Background: The aim of this retrospective observational study was to observe the trends in antimicrobial usage (AMU) from 2018 to 2021 in Swiss pigs based on an electronic treatment journal used nationwide by farmers. Thus, for the first time, standardized, longitudinal comparisons of AMU between the years could be analyzed, as well as the influence of targeted interventions, on farms with higher consumption. (2) Methods: The data was evaluated by different indicators, such as the amount of active ingredient in kilograms, treatment days per farm (ATI) and treatment incidence (TI) based either on animal-defined daily doses (TIADD) or used daily doses (TIUDD). Calculations were performed across the following five age categories: suckling piglets, weaners, fattening pigs, and gestating and lactating sows, and the proportions of antimicrobial classes were evaluated for each age category. (3) Results: The highest amount of the active ingredient was administered to the group of fattening pigs, while the suckling piglets received the lowest amount of the active ingredient. In 2021, there was a significant decrease in active ingredient consumption per pig, but a significant increase in ATI, TIADD and TIUDD compared to 2018. The largest proportion of AMU was attributed to penicillins each year, followed by sulfonamides and tetracyclines. The "Highest Priority Critically Important Antimicrobials" represented a proportion of overall usage, declining from 5.2% in 2018 to 3.1% in 2021, while polypeptides were the most used class of critical antimicrobials. Interventions on high-usage farms showed that some farms decreased their AMU in the following year while others did not. (4) Conclusions: This study reveals a decrease in the overall usage measured in kilograms per pig of antimicrobials in Swiss pigs between 2019 and 2021 through the monitoring of AMU, but, at the same time, there was an increase in treatment days or incidence per farm. Critical antimicrobials can be reduced regardless of the indicator. The significance and quality of interventions should be investigated in future studies.

Adaptation and Resistance of Microbial Communities to Antibiotic Contamination in the Surrounding Environment of a Sustainable Pig Farm

Abstract:An investigation was conducted in the vicinity of a sustainable pig farm to assess the presence of antibiotics through SPE-HPLC/MS/MS, microbial communities via Illumina high-throughput sequencing, and antibiotic resistance genes using SmartChip technology. The study revealed that tetracyclines were the predominant antibiotics detected in the soil and sediment surrounding the pig farm, with residual concentrations ranging from 33.3 to 1244.2 μg∙kg−1. The most prevalent phyla identified at various sampling sites were Firmicutes, Chloroflexi, Proteobacteria, Actinobacteria, and Acidobacteria. A total of 188 antibiotic resistance genes (ARGs) and 9 mobile genetic elements were found in the sediment, with aminoglycoside (particularly aadA2-03), sulfonamide (specifically sul2), and tetracycline (particularly tetX) resistance genes being the most frequently observed. The presence of tetracycline residue was observed to influence the composition of the microbial community, whereas no significant association was found between antibiotics and ARGs. Examination of the correlation between ARGs and bacteria at the phylum level demonstrated that Cyanobacteria, Acidobacteria, Planctomycetota, and Gemmatimonadota were the predominant phyla associated with ARG presence near an intensive pig farm. Notably, Cyanobacteria may function as a continual reservoir and/or shelter for ARGs, thereby potentially contributing to the dissemination of ARGs in the sediment environment in close proximity to a pig farm. This study presents evidence of the ecological risks posed by antibiotics in a pig farm-cropland system, highlighting the connection between microbial community structure and ARGs. Therefore, the issue of antibiotic residues must be factored into the sustainability of animal husbandry practices.
Linking Animal Welfare and Antibiotic Use in Pig Farming—A Review

Abstract:Preventative measures, such as biosecurity and vaccinations, are essential but not sufficient to ensure high standards of health in pig production systems. Restrictive, barren housing and many widely used management practices that cause pain and stress predispose high-performance pigs reared in intensive systems to disease. In this context, antibiotics are used as part of the infrastructure that sustains health and high levels of production in pig farms. Antimicrobial resistance (AMR) is a global emergency affecting human and animal health, and the use of antibiotics (AMU) in intensive livestock farming is considered an important risk factor for the emergence and spread of resistant bacteria from animals to humans. Tackling the issue of AMR demands profound changes in AMU, e.g., reducing their use for prophylaxis and ending it for growth promotion. In support of such recommendations, we revise the link between animal welfare and AMU and argue that it is crucial to sustainably reduce AMU while ensuring that pigs can live happy lives. In support of such recommendations, we aimed to revise the link between animal welfare and AMU in pigs by analysing stress factors related to housing and management and their impact on pig welfare. In particular, we reviewed critical management practices that increase stress and, therefore, pigs’ susceptibility to disease and reduce the quality of life of pigs. We also reviewed some alternatives that can be adopted in pig farms to improve animal welfare and that go beyond the reduction in stress. By minimising environmental and management stressors, pigs can become more immunocompetent and prepared to overcome pathogenic challenges. This outcome can contribute to reducing AMU and the risk of AMR while simultaneously improving the quality of life of pigs and, ultimately, maintaining the pig industry’s social license.

Overall assessment of antibiotic substitutes for pigs: a set of meta-analyses

Abstract:Background: Antibiotic growth promoters are widely used to improve weight gain. However, the abuse of antibiotics can have many negative effects on people. Developing alternatives to antibiotics is an urgent need in livestock production. We aimed to perform a meta-analysis and network meta-analysis (NMA) to investigate the effects of feed additives as potential antibiotic substitutes (ASs) on bacteriostasis, growth performance, intestinal morphology and immunity. Furthermore, the primary, secondary, and tertiary ASs were defined by comparing their results with the results of antibiotics. Results: Among 16,309 identified studies, 37 were summarized to study the bacteriostasis effects of feed additives, and 89 were included in the meta-analysis and NMA (10,228 pigs). We summarized 268 associations of 57 interventions with 32 bacteria. The order of bacteriostasis effects was as follows: antimicrobial peptides (AMPs) ≈ antibiotics>organic acids>plant extracts>oligosaccharides. We detected associations of 11 feed additives and 11 outcomes. Compared with a basal diet, plant extract, AMPs, probiotics, microelements, organic acids, bacteriophages, lysozyme, zymin, and oligosaccharides significantly improved growth performance (P < 0.05); organic acids, probiotics, microelements, lysozyme, and AMPs remarkably increased the villus height:crypt depth ratio (V/C) (P < 0.05); and plant extracts, zymin, microelements, probiotics, and organic acids notably improved immunity (P < 0.05). The optimal AMP, bacteriophage, lysozyme, microelements, oligosaccharides, organic acids, plants, plant extracts, probiotics, and zymin doses were 0.100%, 0.150%, 0.012%, 0.010%, 0.050%, 0.750%, 0.20%, 0.040%, 0.180%, and 0.100%, respectively. Compared with antibiotics, all investigated feed additives exhibited no significant difference in effects on growth performance, IgG, and diarrhoea index/rate (P > 0.05); AMPs and microelements significantly increased V/C (P < 0.05); and zymin significantly improved lymphocyte levels (P < 0.05). Furthermore, linear weighting sum models were used to comprehensively estimate the overall impact of each feed additive on pig growth and health. Conclusions: Our findings suggest that AMPs and plant extracts can be used as primary ASs for weaned piglets and growing pigs, respectively. Bacteriophages, zymin, plants, probiotics, oligosaccharides, lysozyme, and microelements can be regarded as secondary ASs. Nucleotides and organic acids can be considered as tertiary ASs. Future studies should further assess the alternative effects of combinational feed additives.
High Heritabilities for Antibiotic Usage Show Potential to Breed for Disease Resistance in Finishing Pigs

Abstract:The use of antimicrobials in animal production is under public debate, mainly due to the risk of transfer of resistance to pathogenic bacteria in humans. Therefore, measures have been taken during the last few decades to reduce antibiotic usage in animals, for instance, by national monitoring programmes and by improving animal health management. Although some initiatives exist in molecular genetic selection, quantitative genetic selection of animals towards decreased antibiotic usage is an underexplored area to reduce antibiotic usage. However, this strategy could yield cumulative effects. In this study, we derived new phenotypes from on-farm parenteral antibiotic records from commercially grown crossbred finishing pigs used in the progeny test of Piétrain terminal sires to investigate the heritability of antibiotics usage. Parenteral antibiotic records, production parameters and pedigree records of 2238 full-sib pens from two experimental farms in Belgium between 2014 and 2020 were analysed. Heritability estimates were moderate (18–44%) for phenotypes derived from all antibiotic treatments, and low (1–15%) for phenotypes derived from treatments against respiratory diseases only. Moreover, genetic correlations between these new phenotypes and mortality were low to moderate (0.08–0.60) and no strong adverse genetic correlations with production traits were found. The high heritabilities and favourable genetic correlations suggest these new phenotypes, derived from on-farm antibiotics records, to be promising for inclusion in future pig breeding programs to breed for a decrease in antibiotics usage.
Exploration of aerosolization of indoor bacterial communities in relation to antibiotic usage on swine farms

Abstract:Airborne bacteria present on swine farms can adversely affect the health of pigs and livestock workers. However, many studies have intensively investigated the characterization of microbiota in pig feces and aerosol samples without considering the stepwise aerosolization process and the impact of antibiotic usage. This study aimed to comprehensively explore the bacterial communities of feces, surfaces, and bioaerosols during the aerosolization process and assess variations in bacterial resistance to antibiotic usage on swine farms. In this study, 32 samples were collected from six swine farms and categorized as high- or low-antibiotic-use farms based on the European Union's defined daily dose for animals (DDDvet). Fecal samples had significantly higher alpha diversities, followed by aerosol and surface samples (p <0.05). However, no alpha diversity indices showed significant differences according to antibiotic usage. Beta-diversity analyses revealed significant differences between bioaerosols and fecal samples depending on antibiotic usage. Notably, Staphylococcus spp., Corynebacteria spp., and Enterococcus spp. were significantly more prevalent in bioaerosols from high-antibiotic-use farms than in fecal samples (p <0.05), indicating a potentially high level of antibiotic-resistant bacteria and the spread of antibiotic resistance among pigs and workers. For source tracking analysis, over 50 % of the aerosol samples contained bacteria from fecal and surface sources, contributing to the aerosol microbiome. These findings highlight that the aerosolization of fecal and surface particles within swine farms distinctly affects airborne bacterial communities. Our results showed that different antibiotic uses notably affected indoor bacterial communities and antibiotic resistance during stepwise aerosolization in swine farms.

Abstract:The purpose of the study was to develop a blueprint using financial documentation to describe and quantify vaccine and antibiotic usage (ABU). This method was piloted in a commercial pig farm in South Africa, with the ultimate hope to serve as a tool in a future species-specific vaccine and ABU surveillance system. Data collection was based on templates from the European Surveillance of Veterinary Antimicrobial Consumption (ESVAC) network and the World Organisation for Animal Health (WOAH). Invoices from 2016 to 2018 were used as the main data source. In addition, monthly statement of accounts were used to check for missing invoices. An inventory check was done to ensure that the correct antibiotic concentrations were used in subsequent calculations. Livestock counts and slaughter statistics were also collected to be used as denominator data. Cost calculations for the procurement of antibiotics and vaccines were also done. The study showed that veterinary medicinal products were purchased only from a single veterinary practice. A total of 291 invoices were issued over 3 years, of which 2.75% (8/291) were missing and could therefore not be used in quantification. Tetracyclines (453.65 ± 25.49 kg and 135.16 ± 3.31 mg/kg), followed by quinoxalines (258.33 ± 8.04 kg and 77.07 ± 3.93 mg/kg) were used in the highest amounts, both in terms of weight (kg) and adjusted for animal biomass (mg/kg). Vaccines used on the farm targeted seven different diseases, namely enzootic pneumonia, erysipelas, ileitis, infectious infertility, leptospirosis, neonatal pig diarrhea and porcine circovirus disease. An average of 103 546 vaccine dosages was purchased for ZAR1 302,727 ($ 84,620 1 ) per year, whereas the average cost for the procurement of antibiotics was ZAR 907,372 ($ 69,561) per year. The study showed that invoices and monthly statement of accounts, in combination with an inventory check and on-farm production statistics, are useful data sources to quantify vaccine and ABU in the absence of veterinary prescriptions. In addition, vaccinating pigs were more expensive than administering antibiotics.

Abstract:The spread of antimicrobial resistance (AMR) in animal husbandry is usually attributed to the use of antibiotics and poor hygiene and biosecurity. We therefore conducted experimental trials to improve hygiene management in weaned pig houses and assessed the impact on the spread. For each of the two groups examined, the experimental group (EG) and the control group (CG), three replicate batches of piglets from the same pig breeder, kept in pre-cleaned flat decks, were analyzed. In the flat decks of the experimental groups, the hygiene conditions (cleaning, disinfection, dust removal and fly control) were improved, while regular hygiene measures were carried out in the control groups. The occurrence and spread of AMR were determined in Escherichia coli (E. coli; resistance indicator) using cultivation-dependent (CFU) and -independent (qPCR) methods as well as whole genome sequencing of isolates in samples of various origins, including feces, flies, feed, dust and swabs. Surprisingly, there were no significant differences (p > 0.05) in the prevalence of resistant E. coli between the flat decks managed with conventional techniques and those managed with improved techniques. Selective cultivation delivered ampicillin- and sulfonamide-resistant E. coli proportions of up to 100% and 1.2%, respectively. While 0.5% E. coli resistant to cefotaxime and no ciprofloxacin resistance were detected. There was a significant difference (p < 0.01) in the abundance of the blaTEM-1 gene in fecal samples between EG and CG groups. The colonization of piglets with resistant pathogens before arrival, the movement of flies in the barn and the treatment of bacterial infections with antibiotics obscured the effects of hygiene improvement. Biocide tolerance tests showed no development of resistance to the farm regular disinfectant. Managing hygiene alone was insufficient for reducing antimicrobial resistances in piglet rearing. We conclude that the complex factors contributing to the presence and distribution of AMR in piglet barns underscore the necessity for a comprehensive management strategy.

Abstract:Swine farms are one of the important sources of antibiotics in the environment. In this study, 42 samples of compound feed and feces of swine collected at different growth stages from intensive farms were evaluated for the occurrence and concentrations of three tetracyclines (TCs, namely oxytetracycline, chlortetracycline, and doxycycline) and three sulfonamides (SAs, namely sulfadiazine, sulfadimidine, and sulfamethoxazole). To check for other additional sources of antibiotic administration, ratios (R) of the measured and the predicted levels of each antibiotic excreted via feces were also estimated. Our results showed that the maximum concentration of TCs was 376,210 μg kg−1 and 541,020 μg kg−1 in the feeds and feces, respectively, both for oxytetracycline. In contrast, the highest concentration of SAs were 16.98 μg kg−1 for sulfadimidine in the feeds and 14.70 μg kg−1 for sulfadiazine in the feces. The concentrations of ΣTCs (sum of the three tetracyclines) in swine feeds and feces were found to be 1–4 orders of magnitude higher than those of ΣSAs (sum of the three sulfonamides). Approximately 36% of the R values were found to be greater than one, indicating other sources of administration such as injection and/or oral administration (via drinking water) may also contribute to the presence of antibiotics in feces. Most of the higher R values were found in starter pigs, which were generally administrated with antibiotics by multiple routes to prevent disease and promote swine growth. Our study suggests that comprehensive measures may be undertaken to control antibiotic use in intensive swine farms.
