Abstract:We developed nested PCR protocols and performed a multiyear survey on the prevalence of several protozoan parasites in wild northern bobwhite (Colinus virginianus) and scaled quail (Callipepla squamata) in the Rolling Plains ecoregion of Texas and Oklahoma (i.e. fecal pellets, bird intestines and blood smears collected between 2010 and 2013). Coccidia, cryptosporidia, and microsporidia were detected in 46.2%, 11.7%, and 44.0% of the samples (n = 687), whereas histomona and hematozoa were undetected. Coccidia consisted of one major and two minor Eimeria species. Cryptosporidia were represented by a major unknown Cryptosporidium species and Cryptosporidium baileyi. Detected microsporidia species were highly diverse, in which only 11% were native avian parasites including Encephalitozoon hellem and Encephalitozoon cuniculi, whereas 33% were closely related to species from insects (e.g. Antonospora, Liebermannia, and Sporanauta). This survey suggests that coccidia infections are a significant risk factor in the health of wild quail while cryptosporidia and microsporidia may be much less significant than coccidiosis. In addition, the presence of E. hellem and E. cuniculi (known to cause opportunistic infections in humans) suggests that wild quail could serve as a reservoir for human microsporidian pathogens, and individuals with compromised or weakened immunity should probably take precautions while directly handling wild quail.
Susceptibility of wild turkeys (Meleagris gallopavo) to experimental West Nile virus infection

Abstract:Since the introduction of West Nile virus (WNV) to North America in 1999, WNV is estimated to have contributed to population-level declines in numerous avian species. However, the potential impacts of this virus on many free-ranging upland game bird species, including the wild turkey (Meleagris gallopavo), which is undergoing regional population declines, remain unknown. Herein, two age groups (∼5 to 6 weeks and ∼15 to 16 weeks post-hatch) of juvenile wild turkeys were subcutaneously inoculated with WNV, sampled daily from 1 to 7 days post-inoculation (dpi), and euthanized on 14 dpi. No clinical signs and minimal gross lesions were attributable to WNV infection. Peak viraemia titres were similar between age groups (<101.7 to 104.6 plaque-forming units [PFU]/ml), but the duration of viraemia was longer in the old group (3-4 days) than in the young group (0-3 days). Intermittent oral and/or cloacal viral shedding from 2 to 7 dpi was detected in both age groups. No infectious virus was detected in the heart, brain, kidney, skeletal muscle, spleen, and feathers from WNV-inoculated turkeys euthanized on 14 dpi. All WNV-inoculated birds seroconverted by 14 dpi, as well as two co-housed sham-inoculated birds. The most consistent microscopic lesions among all WNV-inoculated birds were mild lymphoplasmacytic myocarditis and encephalitis. Minimal immunohistochemical labelling was detected in tissues in addition to scant macrophages within the blood, spleen, and bone marrow. These data suggest WNV is unlikely to pose a significant risk to wild turkey populations, although the possibility remains that WNV may indirectly decrease fitness or predispose wild turkeys to other health stressors.RESEARCH HIGHLIGHTS Clinical disease was not observed in wild turkeys experimentally infected with WNV.Pathology attributed to WNV was mild and included brain and heart inflammation.Viraemias suggest WNV-infected wild turkeys do not play a role in WNV transmission.No age-associated differences in WNV clinical disease or pathology were observed.
Molecular characterization of quail bronchitis virus isolated from bobwhite quail in Minnesota

Abstract:From 2008 to 2012, 4 separate cases of quail bronchitis virus infection were seen in bobwhite quail (Colinus virginianus) raised in Minnesota. The quail chicks ranged in age from 5 d to 8 wk and suffered from respiratory distress and elevated mortality. On necropsy, gross lesions consisted of mucus in trachea, congested lungs, caseous air sacculitis, accumulation of chalky white urates on internal organs, necrotic foci in liver, and enlarged spleen. Histologic examination revealed fibrinoheterophilic rhinitis, heterophilic bronchitis, heterophilic tracheitis, and interstitial pneumonia in addition to deciliation, desquamation, and necrosis of bronchial respiratory epithelium. Karyomegaly with basophilic intranuclear inclusions was also seen in affected epithelium. Severe epicarditis, pericarditis, myocarditis, multifocal necrotizing hepatitis, and splenitis were additional pathological findings. Quail bronchitis virus (QBV) was isolated from all four samples when inoculated in specific-pathogen-free (SPF) embryonated chicken eggs. The virus was confirmed by electron microscopy and polymerase chain reaction using fowl adenovirus (FAdV) hexon gene-specific primers. Nucleotide sequences of the four isolates showed 99.0% identity with CELO strain of fowl adenovirus A. Nine nucleotide substitutions were observed; 3 of these were nonsynonymous (A281G, C314T and G565C), leading to changes in deduced amino acid sequences (S94G, T105M and A189P, respectively). Based on partial sequence of the hexon gene, QBV isolates of this study clustered closely with fowl adenovirus A and were different from FAdV groups B through E and from adenoviruses of goose, duck, turkey, and pigeon. Further studies are indicated to determine the impact of nonsynonymous substitutions on host specific pathogenicity of these viruses.
An Outbreak of Fowl Aviadenovirus A-Associated Gizzard Erosion and Ulceration in Captive Bobwhite Quail (Colinus virginianus)

Abstract:A flock of captive bobwhite quail (Colinus virginianus) experienced loose droppings, depression, and increased mortality starting at 3 wk of age. Necropsy of the affected birds revealed intestines dilated with frothy and tan fluid. Irregular dark brown fissures within the koilin layer of the gizzard were found in 20%-30% of the birds. Histologically, gizzards showed multifocal koilin degeneration or fragmentation, degeneration and necrosis of the subjacent epithelial cells, and infiltration of macrophages, lymphocytes, and heterophils. Necrotic epithelial cells occasionally contained large, smudgy, basophilic intranuclear inclusion bodies with marginated nuclear chromatin. Adenoviral paracrystalline arrays composed of icosahedral virions (60-70 nm diameter) were seen on transmission electron microscopy in the nuclei of epithelial cells in the gizzard mucosa. Adenovirus was isolated from gizzard, liver, intestine, and trachea by inoculation of specific-pathogen-free embryonated chicken eggs. Homogenates of the gizzard, liver, and intestine were positive for the adenovirus hexon gene by PCR. Sequencing of PCR amplicons confirmed the virus as fowl aviadenovirus A. The study isolates showed more than 99% and 97% nucleotide identity with quail bronchitis virus and with aviadenoviruses from gizzard erosion and ulceration (GEU) in broilers, respectively. The viral isolates showed six substitutions (G1T, C174A, A229G, C513A, T579A, and G621C) of which two were nonsynonymous (G1T and A229G), resulting in a change in the translated amino acid as A1S and S77G, respectively. These results indicate that adenoviruses of the same type or species can cause different clinical presentations in quails, e.g., bronchitis or GEU.
Neurological lesions in chickens experimentally infected with virulent Newcastle disease virus isolates

Abstract:Distribution, character, and severity of lesions were evaluated in tissues from the central nervous system of chickens inoculated with 10 different Newcastle disease virus (NDV) isolates: CA 1083, Korea 97-147, Australia (all velogenic viscerotropic), Texas GB and Turkey North Dakota (both velogenic neurotropic), Nevada cormorant, Anhinga and Roakin (all mesogenic), and B1 and QV4 (lentogenic). Tissues for the present study included archived formalin-fixed, paraffin-embedded brain (all strains) plus spinal cord (two strains). Encephalitis was observed in all velogenic viscerotropic and velogenic neurotropic strains, and in some mesogenic strains. In general, the encephalitic lesions began at 5 days post infection, with more severe lesions occurring around 10 days post infection. At this time point, especially in the grey matter of the brain, cerebellum and spinal cord, there were neuronal necrosis, neuronal phagocytosis, and clusters of cells with microglial morphology. Axonal degeneration and demyelination was also observed. Immunohistochemistry (IHC) for viral nucleoprotein confirmed the presence of virus. In the areas of encephalomyelitis, IHC for CD3 revealed that many of the inflammatory cells were T lymphocytes. IHC using an antibody for glial fibrillar acid protein showed reactive astrogliosis, which was most pronounced at the later time points.

Abstract:Grassland birds have been declining substantially for the past several years. Habitat loss, degradation, and fragmentation as well as climate change are all thought to be the main drivers of the decline. However, as the declines continue to accelerate, it is becoming imperative to examine other factors that may contribute to population fluctuations. The nematodes Oxyspirura petrowi, Aulonocephalus pennula, and Physaloptera sp. are commonly found infecting northern bobwhite (Colinus virginianus), a game species of economic importance, and all 3 nematodes use insects as an intermediate host. Here we used polymerase chain reaction techniques to determine the occurrence of the 3 nematodes in 7 insect orders to uncover epidemiological patterns of the greatest potential for transmission to northern bobwhite. Insects were collected from March through September using sweep nets and pitfall traps. An R × C chi-squared test with Monte Carlo simulation was used to determine differences in the occurrence of the parasites across taxa and time. The results of the statistical analysis showed the nematodes are predominantly found in the order Orthoptera, and A. pennula and Physaloptera sp. showed epidemiological patterns in insects. However, no such pattern was observed with O. petrowi. An explanation for the lack of epidemiological pattern in O. petrowi is proposed and the diversity of known insect hosts of the 3 nematodes is increased.
COMPARATIVE PATHOLOGICAL STUDY OF EXPERIMENTAL INFECTION OF NEWCASTLE DISEASE VIRUS GENOTYPES VII IN CHICKENS AND DUCKS

Abstract:Newcastle disease virus (NDV) is a highly contagious virus infects a wide range of avian species including chicken and ducks. The disease can be fatal for many types of them. It was believed that ducks and geese act as natural reservoirs of NDV because of their natural resistance. Recently, NDV isolates were pathogenic for ducks. Therefore, our study aimed to investigate the pathogenicity of highly pathogenic NDV genotype VII in white Ross commercial broiler chickens and Mallard ducks on the basis of clinical, histological observation and immunohistochemistry findings of virus antigen through studying the sequential pathological lesions of both species at 2nd, 5th and 10th dpi. Both species were inoculated via intranasal route by NDV genotype VII (accession number MT887290). Clinical lesions and mortalities were evident mostly in chickens rather than in ducks. Respiratory, digestive and nervous signs were clearly manifested in chickens. Ducks showed low level of mortalities and the nervous signs were the most obvious symptoms. Microscopic findings of chickens showed severe pancreatitis, pneumonia, enteritis as well as pericarditis and brain malacia. Similar findings were noticed in ducks but with lower degree of severity. Brain lesions in ducks included encephalitis and perivascular reaction mostly within the cerebellar tissue. Immunohistochemistry reaction against NDV antigen showed marked immunostaining within epithelial cells, purkinje cells and inflammatory cells mostly the macrophages. The immunohistochemistry reaction was correlated with the pathological findings in both species. In conclusion, the mortality and lesions of NDV in both species were correlated with systemic alterations associated with pancreatitis, pneumonia and encephalopathy. Moreover, the obtained data revealed possible susceptibility of ducks to NDV genotype VII, which might be need forthcoming viral vaccination strategy for ducks against NDV.

Abstract:AbstractThe continued spread and occurrence of Newcastle disease virus (NDV) has posed potential threat to domestic poultry industry around the globe. Mainly, wild avian species has always been implicated for the natural reservoir for virus and spread of the disease. In the present study, we report the isolation of Newcastle disease virus (NDV/Peacock/India/2012) in necropsy brain tissue sample of wild peacock from North India. Complete genome of the virus was found to be 15,186 nucleotides (nts) with six genes in order of 3′-N–P-M-F-HN-L-5′, which was limited by 55-nts leader region at the 3′ end and a 114-nts trailer sequence at 5′ end. Sequence analysis of fusion protein revealed the dibasic amino acid cleavage site 112R-R-Q-K-R-F117, a characteristic motif of virulent virus. Phylogenetic analysis placed the isolate in genotype II of Newcastle disease virus showing the lowest mean percent divergence (6 %) with other genotype II counterparts. The isolate was characterized as mesogenic (intermediate pathotype) based on the mean death time (63 h) in embryonated chicken eggs and the intra-cerebral pathogenicity index (1.40) in day-old chicks. The report emphasizes the dynamic ecology of NDV strains circulating in a wild avian host during the outbreak of 2012 in North India. Further the genotypic and pathotypical characterizations of the isolate could help in development of homologous vaccine against NDV strain circulating in avian population.

Abstract:Mesocestoides spp. have a cosmopolitan distribution with zoonotic potential. Mesocestoides tetrathyridia were found under the pericardial sac, on the surface of the crop, and in the peritoneal cavity of a hunter-harvested Northern Bobwhite ( Colinus virginianus) and a Scaled Quail ( Callipepla squamata) collected during the 2016-17 quail hunting season in northwest and southern Texas, US respectively. Molecular analysis indicated that the tetrathyridia from the birds likely belonged to an undescribed species and are identical to pretetrathyridium stages recently found in Scincella lateralis skinks in Oklahoma. This is the first report of Mesocestoides from North American birds.

Abstract:Background Oxyspirura petrowi appears to be emerging as a nematode parasite that could negatively impact Northern Bobwhite quail individuals and populations within Texas and other regions of the United States. Despite this eye worm's potential importance in the conservation of wild quail, little is known about the general biology and genome composition of O . petrowi . To fill the knowledge gap, we performed a small scale random genome sequence survey, sequenced its 18S rRNA and the intergenic region between the 18S and 28S rRNA genes, studied its phylogenetic affinity, and developed a PCR protocol for the detection of this eye worm. Results We have generated ~240 kb of genome sequence data derived from 348 clones by a random genome survey of an O . petrowi genomic library. The eye worm genome is AT-rich (i.e., 62.2% AT-content), and contains a high number of microsatellite sequences. The discovered genes encode a wide-range of proteins including hypothetical proteins, enzymes, nematode-specific proteins. Phylogenetic analysis based on 18S rRNA sequences indicate that the Spiruroidea is paraphyletic, in which Oxyspirura and its closely related species are sisters to the filarial nematodes. We have also developed a PCR protocol based on the ITS2 sequence that allows sensitive and specific detection of eye worm DNA in feces. Using this newly developed protocol, we have determined that ~28% to 33% of the fecal samples collected from Northern Bobwhites and Scaled Quail in Texas in the spring of 2013 are O . petrowi positive. Conclusions The O . petrowi genome is rich in microsatellite sequences that may be used in future genotyping and molecular fingerprinting analysis. This eye worm is evolutionarily close to the filarial nematodes, implying that therapeutic strategies for filariasis such as Loa loa would be referential in developing treatments for the Thelazoidea parasites. Our qPCR-based survey has confirmed that O . petrowi infection is of potential concern to quail managers in Texas.
Sequential histopathological changes of the lower respiratory and gastrointestinal tracts in chickens infected with velogenic Newcastle disease virus

Abstract:Velogenic Newcastle disease is a highly contagious and fatal disease that affects domestic poultry, and is clinically manifested by haemorrhagic gastroenteritis and respiratory distress. This study investigated the sequential histopathogic changes in the lungs, proventriculus, and intestines of broiler chickens infected with velogenic Newcastle disease virus (vNDV). Fifty broiler chickens (6-weeks-old) were randomly assigned into two groups of 25 each: UC – unvaccinated with Newcastle disease (ND) vaccines and intranasallly inoculated with vNDV, UU – unvaccinated uninfected. At 4, 5 and 6 days post-infection (d.p.i) tissue samples were collected from birds in each group for histopathology. The lungs showed hyperaemia, haemorrhages, inflammatory oedema and cellular infiltrations in the secondary bronchi, parabronchi (atria, infundibular, air and blood capillaries of the exchange tissue), depletion of bronchial-asociated lymphoid tissues, loss of cilia and hyperplasia of surface epithelium of bronchi and parabronchi at 4 d.p.i. The lesions progressed to collapsed air capillaries with fibroplasia and fibrosis of the area at 6 d.p.i. Similar inflammatory changes were observed in the proventriculus, but accompanied by degeneration and attenuation of surface epithelium of proventiculus and proventricular glands at 4 d.p.i., and severe necrosis and disintegration of the proventricular glands at 6 d.p.i. The intestine showed severe inflammatory changes in the mucosa, submucosa and serosa and depletion of gut-associated lymphoid tissues at 4 d.p.i., and ulcerative enteritis at 5 d.p.i. The lumen was filled with necrotic debris at 6 d.p.i. This study reinforces the concept making pathologic animal experiments important tools for a complete characterization of strains in susceptible animals, as this information will be helpful in the accurate diagnosis of velogenic Newcastle disease in chickens.
Experimental Velogenic Viscerotropic Newcastle Disease Virus Infection in Chickens Immunologically Impaired by Treatment with Cyclophosphamide

Abstract:This study investigated whether lymphocytic depletion following chemical bursectomy influenced the severity of infection and development of lesions in chickens challenged with velogenic viscerotropic Newcastle disease virus (vvNDV). Cockerel chickens treated with cyclophosphamide on days 2, 3 and 4 post-hatch showed loss of weight, atrophy and lymphocytic depletion in the bursa of Fabricius and spleen. At 6 weeks of age, the chickens were assigned to four groups- Bursectomized intramuscularly vvNDV inoculated (BI), bursectomized uninfected (BU), non-bursectomized infected (NBI) and non-bursectomized uninfected (NBU) chickens. The BI and NBI chickens showed significant (P < .05) loss of weight than their uninfected controls. Depression, anorexia, greenish diarrhea, listlessness, tremor, and oculo-nasal discharges were observed in both infected groups, but were more severe and frequent in the NBI than in the BI chickens. Total mortalities were 100% and 95.5% for the NBI and BI chickens, respectively (P > .05). Lesions in both infected groups included atrophy of the bursa, spleen and thymus. Hemorrhages in the proventricular mucosa, intestines and cecal tonsils, as well as congestion and enlargement of the kidneys were significantly (P < .05) more severe and frequent in NBI than BI chickens. Histopathology showed necrosis and depletion of lymphocytes in the three lymphoid organs in both infected groups with more severity in the NBI than BI chickens. These results show that depletion of lymphocytes by treatment with cyclophosphamide may influence the severity of infection and development of lesions in vvNDV infection in cockerel chickens.
The Susceptibility of Chickens to Zika Virus: A Comprehensive Study on Age-Dependent Infection Dynamics and Host Responses

Abstract:Zika virus (ZIKV) remains a public health concern, with epidemics in endemic regions and sporadic outbreaks in new areas posing significant threats. Several mosquito-borne flaviviruses that can cause human illness, including West Nile, Usutu, and St. Louis encephalitis, have associations with birds. However, the susceptibility of chickens to ZIKV and their role in viral epidemiology is not currently known. We investigated the susceptibility of chickens to experimental ZIKV infection using chickens ranging from 1-day-old chicks to 6-week-old birds. ZIKV caused no clinical signs in chickens of all age groups tested. Viral RNA was detected in the blood and tissues during the first 5 days post-inoculation in 1-day and 4-day-old chicks inoculated with a high viral dose, but ZIKV was undetectable in 6-week-old birds at all timepoints. Minimal antibody responses were observed in 6-week-old birds, and while present in younger chicks, they waned by 28 days post-infection. Innate immune responses varied significantly between age groups. Robust type I interferon and inflammasome responses were measured in older chickens, while limited innate immune activation was observed in younger chicks. Signal transducer and activator of transcription 2 (STAT2) is a major driver of host restriction to ZIKV, and chicken STAT2 is distinct from human STAT2, potentially contributing to the observed resistance to ZIKV infection. The rapid clearance of the virus in older chickens coincided with an effective innate immune response, highlighting age-dependent susceptibility. Our study indicates that chickens are not susceptible to productive ZIKV infection and are unlikely to play a role in the ZIKV epidemiology.

Abstract:Avian bornavirus (ABV) is known to infect at least 80 avian species and is associated with avian bornaviral ganglioneuritis (ABG). Avian bornaviral ganglioneuritis is characterized by a lymphoplasmacytic infiltration of the nervous tissue, mainly affecting the nerves that supply the gastrointestinal tract of birds. This disease is diagnosed commonly in psittacines under human care and has been demonstrated in wild bird species; however, its occurrence in raptors is largely unknown. Because of the commonality of ABV in the pet bird population, there is concern about the spread of this virus to other companion avian species, such as falconry birds, as well as wildlife. This prospective study used reverse-transcription quantitative PCR (RT-qPCR) to survey free-ranging Colorado and Wyoming, US, raptor populations for ABV. Quantitative PCR was performed on mixed conjunctival-choanal-cloacal swabs collected from live birds (n=139). In dead birds, a combination of mixed swabs (n=265) and tissue samples of the brain (n=258), heart (n=162), adrenal glands (n=162), liver (n=162), kidney (n=139), spinal cord (n=139), and brachial plexus (n=139) were evaluated. All 1,565 swab and tissue samples RT-qPCR results from the 404 birds evaluated were negative. Based on these results and a lack of clinical signs suggestive of ABG, ABV is likely not a prevalent pathogen in Colorado and Wyoming raptor populations at this time.

Abstract:Localization of Newcastle disease viral nucleoprotein and pathological lesions was evaluated in tissues of 55 indigenous ducks (45 experimentally infected and 10 sentinel ones). In addition, ten Newcastle disease infected chickens were used to ensure that the virus inoculum administered to the ducks produced the disease in chickens, the susceptible hosts. Ducks were killed on day 1, 4, 8 and 14 post-infection. Post-mortem examination was done with six tissues (liver, spleen, lung, caecal tonsils, kidneys and brain) being collected from each bird. The tissues were preserved in 10% neutral formalin for 24 h. They were then transferred to 70% ethanol for histology and immunohistochemical staining. Airsacculitis, necrotic splenic foci, congested intestines, lymphoid depleted caecal tonsils and focal infiltrations by mononuclear cells were the main pathological lesions in infected ducks. Over 28.9% of the infected ducks had Newcastle disease viral nucleoprotein in macrophage-like large mononuclear cells in the caecal tonsils and kidney tubular epithelium. The viral antigens were located in the cytoplasm and nucleolus of the cells. The other organs had no detectable viral antigens. This study shows that the kidneys and caecal tonsils are the likely predilection sites for the virus in ducks. They thus need to be considered as diagnostic indicators for the viral carriage in ducks.

Abstract:This study was conducted to determine how reducing the parasite burden in a definitive host may affect the prevalence in intermediate hosts. Here we used the eyeworm Oxyspirura petrowi and cecal worm Aulonocephalus pennula as model species. Northern bobwhite quail (Colinus virginianus) were provided an anthelmintic medicated feed in wild systems because of convincing evidence that these parasites were suppressing their populations. Eyeworm and cecal worm prevalence were measured in Orthopterans, which act as intermediate hosts, using polymerase chain reaction. Individuals were collected from a control site, a site treated for 2 yr, and a site treated for 5 yr. Orthopteran community composition was significantly different among the sites; however, an interaction between subfamily and site was not significant for the eyeworm. There was a significant reduction in eyeworm-infected Orthopterans on the 5-yr site compared with the other 2, suggesting that treatment of a definitive host may indeed affect the prevalence of eyeworms during other life-cycle stages. There was an interaction between the Orthopteran subfamily and the site for the cecal worm, so results were analyzed within each subfamily. A significant reduction in the prevalence of cecal worms was only found in the Cyrtacanthacridinae subfamily on the 5-yr site when compared with the other sites. However, the greatest prevalence in the Gomphocerinae and Oedipodinae subfamilies across all 3 sites was 4.1%. This indicates an unknown degree of cecal worm host specificity. Therefore, conclusions could not be made through the simple assessment of prevalence.

Abstract:West Nile virus (WNV) can lead to fatal diseases in raptor species. Unfortunately, there is no vaccine which has been designed specifically for use in breeding stocks of falcons. Therefore the immunogenicity and protective capacity of two commercially available WNV vaccines, both approved for use in horses, were evaluated in large falcons. One vaccine contained adjuvanted inactivated WNV lineage 1 immunogens, while the second represented a canarypox recombinant live virus vector vaccine. The efficacy of different vaccination regimes for these two vaccines was assessed serologically and by challenging the falcons with a WNV strain of homologous lineage 1. Our studies show that the recombinant vaccine conveys a slightly better protection than the inactivated vaccine, but moderate (recombinant vaccine) or weak (inactivated vaccine) side effects were observed at the injection sites. Using the recommended 2-dose regimen, both vaccines elicited only sub-optimal antibody responses and gave only partial protection following WNV challenge. Better results were obtained for both vaccines after a third dose, i.e. alleviation of clinical signs, absence of fatalities and reduction of virus shedding and viraemia. Therefore the consequences of WNV infections in falcons can be clearly alleviated by vaccination, especially if the amended triple administration scheme is used, although side effects at the vaccination site must be accepted.

Abstract:Survival of northern bobwhite chicks is an important parameter influencing population dynamics. We determined cause‐specific mortality and apparent daily survival of bobwhite chicks (n = 170) during 2018 and 2019 at Tall Timbers Research Station, Tallahassee, Florida, USA. Avian predation comprised the majority (n = 21, 36%) of mortality followed by snakes (n = 17, 29%), mammals (n = 16, 28%), and research‐induced mortality (n = 4, 7%). Daily survival of chicks increased with age and reached an asymptote at approximately 70 days; late hatches (after 15 July) survived better than early hatches. Survival of northern bobwhite (Colinus virginianus) chicks is an important parameter influencing population dynamics. We assigned cause‐specific mortality to 58 chicks and unknown fates to 45 chicks. Of those with assigned fates, avian predation comprised the majority (n = 21, 36%) followed by snakes (n = 17, 29%), mammals (n = 16, 28%), and research‐induced mortality (n = 4, 7%). Corn snakes (Pantherophis guttatus) were the primary snake species (13 of 17 snake predations) responsible for predation followed by northern cottonmouths (Agkistrodon piscivorus; n = 3) and 1 eastern diamondback rattlesnake (Crotalus adamanateus). We determined diel timing of mortalities during 2019. All corn snake predations during 2019 (n = 8) occurred during roosting (2100–0700 hrs), whereas the eastern diamondback mortality was a diurnal event; no cottonmouths depredated chicks during 2019. Nine of 11 mammalian events occurred during roosting. Two avian events occurred during 2019, both during daylight hours. Survival was best predicted by the model including the effects of chick age, hatch timing, year, and their interactions. Daily survival of chicks increased with age and reached an asymptote at approximately 70 days; late hatches (after 15 July) survived better than early hatches (βLateHatch×ChickAge = 0.059, 95% CI =−0.003–0.121) and survival for late hatches was marginally lower in 2019 (βLateHatch×ChickAge×Year19 = −0.058, 95% CI = −0.137–0.022). Management focused on increasing survival during the first 2 months of life and early nesting season may substantially benefit bobwhite population abundance. Additionally, roosting appeared to be a vulnerable period for bobwhite chicks.

Abstract:ABSTRACT Newcastle disease virus (NDV) can infect more than 200 species of birds, and most reports have focused on poultry. The pathogenic ecology of NDV in wild birds remains largely unknown. Thus, an investigation of NDV in domestic wild birds and pigeons was conducted. Thirty-nine NDV strains were characterized from 3,549 oropharyngeal and cloacal swab samples collected from 2016 to 2019 in Guangxi, China. Among them, 3 NDV strains belonged to class I genotype 1.1.2, and 36 strains belonged to class II, including 6 strains belonging to genotype I, 1 strain belonging to genotype II, 4 strains belonging to genotype VI.2.1.1.2.1, 23 strains belonging to genotype VI.2.1.1.2.2, and 2 strains belonging to genotype XII. Phylogenetic analysis revealed that genotype VI NDVs circulate among multiple species of resident wild birds and may spread back to pigeons. In addition, resident wild birds are important for short-distance NDVs transmission. The prevalence of NDVs in wild birds indicates that more restrictive biosecurity measures and ongoing epidemiological investigations are needed. IMPORTANCE Surveillance of Newcastle disease viruses (NDVs) has been conducted primarily in poultry, but their prevalence in wild birds remains largely unknown. Increasing our understanding of the ecology and evolution of NDVs in different species of birds will help us develop better prevention and control strategies. In this study, large-scale epidemiological sampling of resident wild birds in Guangxi from 2016 to 2019 was performed. The results demonstrated that different genotypes, especially genotype VI NDVs, circulated among resident wild birds. Our findings highlight the potential threat to the pigeon industry and public health.

Diagnostic Detection of H7N3 Low Pathogenicity Avian Influenza in a Commercial Game Bird Flock

Abstract:Low pathogenicity avian influenza (LPAI) H7N3 was diagnosed in a flock of commercially raised Japanese quail (Coturnix japonica) breeder hens. The birds were submitted with an initial complaint of a drop in egg production and watery droppings. Initial PCR testing of the flock for avian influenza (AI) was negative; however, mortality started increasing drastically, prompting further examination and the submission of more birds to the diagnostic laboratory. On antemortem examination, the birds appeared lethargic, with ruffled feathers, and had labored breathing. Gross examination revealed poor body condition, moderate dehydration, splenomegaly, ovarian regression, and airsacculitis. Several hens produced thin-shelled and shell-less eggs. Microscopically, lymphoplasmacytic encephalitis with neuronal cell necrosis, fibrinoheterophilic pneumonia, and splenic lymphoid depletion with amyloid deposition were seen. AI virus was detected in a follow-up submission and characterized as LPAI H7N3. Quail are a species of interest in the pathobiology of avian influenza viruses, underlined by the ability to serve as an amplification host.
