Abstract:The short- and long-term effects of phenol on anaerobic ammonium oxidation (Anammox) were evaluated. The short-term impact of phenol on Anammox activity was determined by a batch test, and an IC50 value of 678.2 mg L(-1) was calculated. Anammox granular sludge was equally seeded into two identical upflow anaerobic sludge blanket reactors (R0 and R1); synthetic wastewater without phenol was fed to R0 while with varied phenol was fed to R1 to study the long-term effects. The performance of R0 was stable, with a steadily rising nitrogen removal rate of 10.5-21.3 kg N m(-3)d ay(-1). However, the performance of R1 was significantly suppressed by an influent phenol concentration of 50 mg L(-1), and was recovered and stabilized by applying one or more control strategies. The phenol-mediated inhibition depressed the Anammox activity and biomass, and caused a change of stoichiometric ratios and granule characteristics.

Abstract:The rapid development of chemical industry has induced to the large amount of phenolic wastewater production. When the promising anaerobic ammonium oxidation (anammox) was employed to treat the industrial wastewater, phenolic compounds would possibly inhibit the microbial performance. Extracellular polymeric substances (EPSs) play an essential role in protecting cells from being intoxicated by phenolic compound while the distinct mechanism remains elusive. In this work, the interaction of phenol with anammox sludge EPSs and transmembrane ammonium transport (Amt) domain was explored at molecular level by using spectral method and molecular docking simulation. It was found that phenol statically quenched the fluorescent components of EPSs and the protein component dominated the interaction between EPSs and phenol. The overall interaction was an entropy-driven process with hydrophobic interaction as the main driving force, and the CO vibration responded preferentially. As phenol continued to penetrate into the cell surface, there were hydrogen bond, hydrophobic interaction force and π-π base-stacking forces between the Amt domain and phenol. The interaction between phenol and amino acid residues of the Amt domain would interfere the NH4+ transport and further affect the activity of anammox sludge. This work is beneficial for in-depth understanding the role of EPSs in protecting anammox sludge from inhibiting by phenolic pollutants.

Abstract:The response characteristic and performance stabilization of anammox process under the stress of the potential organic pollutants support the application of ammonia-nitrogen wastewater treatment. In the present study, nitrogen removal performance was significantly suppressed with the addition of 4-chlorophenol. The activity of anammox process was inhibited by 14.23% (0.1 mg/L), 20.54% (1 mg/L) and 78.15% (10 mg/L), respectively. Metagenomic analysis revealed a significant decrease in the abundance of KEGG pathways associated with carbohydrate and amino acid metabolism with increasing 4-chlorophenol concentration. Metabolic pathway profiles suggest that putrescine is down-regulated at high 4-chlorophenol stress due to inhibition of nitrogen metabolism processes, while it is up-regulated to reduce oxidative damage. In addition, the presence of 4-chlorophenol induced an enhancement of EPS and bacterial debris decomposition, and a partial conversion of 4-chlorophenol to p-nitrophenol. This study unravels the mechanism of effect on anammox consortia in response to 4-CP, which could provide supplementary to facilitate its full-scale application.
Long-term effect of phenol, quinoline, and pyridine on nitrite accumulation in the nitrification process: performance, microbial community, metagenomics and molecular docking analysis

Abstract:Phenol, quinoline, and pyridine, commonly found in industrial wastewater, disrupt the nitrification process, leading to nitrite accumulation. This study explores the potential mechanisms through which these biotoxic organic compounds affect nitrite accumulation, using metagenomic and molecular docking analyses. Despite increasing concentrations of these compounds from 40 to 160 mg/L, ammonia nitrogen removal was not hindered, and stable nitrite accumulation rates exceeding 90 % were maintained. Additionally, these compounds inhibited nitrite-oxidizing bacteria (NOB) and enriched ammonia-oxidizing bacteria (AOB) in situ. As the concentration of these compounds rose, protein (PN) and polysaccharide (PS) concentrations also increased, along with a higher PN/PS ratio. Metagenomic analysis further revealed an increase in hao relative abundance, while microbial community analysis showed increased Nitrosomonas abundance, which contributed to nitrite accumulation stability. Molecular docking indicated that these compounds have lower binding energy with hydroxylamine oxidoreductase (HAO) and nitrate reductase (NAR), theoretically supporting the observed sustained nitrite accumulation.
Effect of reactive oxygen species (ROS) produced by pyridine and quinoline on NH4+-N removal under phenol stress: The shift of nitrification pathway and its potential mechanisms

Abstract:Pyridine and quinoline are typical nitrogenous heterocyclic compounds with different structures that are found in coking wastewater. However, neither the corresponding mechanism nor its effect on the degradation of NH4+-N under phenol stress is known. In this study, the effects of pyridine and quinoline degradation on NH4+-N removal under phenol stress were evaluated using three lab-scale sequencing batch reactors. The average NH4+-N removal efficiencies of the reactors were 99.46 %, 88.86 %, and 98.64 %. With the increased concentration of pyridine and quinoline, NH4+-N and NO3--N accumulated to 58.37 mg/L and 141.37 mg/L, respectively, due to the lack of an electron donor and anaerobic environment. The addition of pyridine and quinoline significantly improved antioxidant response and altered the nitrification pathway. The nitrification process shifted from the mediation of amo and hao to the mediation of Ncd2 due to oxidative stress induced by pyridine and quinoline. Furthermore, oxidative stress interferes with the metabolism of carbon sources, resulting in decreased biomass. These results provide a new perspective for coking wastewater treatment processes.
The Anammox Process at Typical Feast-Famine States: Reactor Performance, Sludge Activity and Microbial Community

Abstract:Anaerobic ammonium oxidation (Anammox) is a chemolithotrophic bioprocess which has been widely applied in the treatment of different concentrations of ammonium-containing wastewaters. However, there is less attention on the problem that the instantaneous growth rate (or metabolic rate) and equilibrium growth rate were un-synchronous for anammox bacteria due to their long generation time and self-immobilization of the granular sludge which could lead to the inaccurate estimation. In this study, the anammox process was firstly divided to four typical feast-famine (starvation, satiation, tolerance and poison) states based on the combination of both off-site and in-situ anammox reaction kinetics. Then, four respective lab-scale bioreactors were operated at each state for over a year to achieve stable anammox performance. The results showed that the nitrogen removal rates of bioreactors were 0.53, 2.24, 9.30 and 12.96 kg N/(m(3).d); and the specific anammox activities of granular sludge were 188.94 (48%), 313.29 (79%), 397.50 (100%) and 198.60 (50%) mg N/(g VSS.d) which could reflect the reactivity of each feast-famine state. The stable microbial communities of bioreactors were cultured and analyzed, whose species diversity went down with the decrease of Shannon and ACE index. The relative abundance of anammox bacteria increased from 11% to 57% from starvation to poison state. Candidatus Brocadia/Nitrospira, Candidatus Kuenenia and Brocadiaceae unclassified were revealed to be the distinctive functional bacteria, which could serve as the indicator of each state. The setting up of the typical feast-famine states could be regarded as the landmark to help the design, control and optimization of anammox process.

Abstract:The specialized wastewater treatment plants for the chemical industry are rapidly developed in China and many other countries. But there is a common bottleneck in that the toxic pollutants in chemical wastewater often cause shock impacts on biological nitrogen removal systems, which directly affects the stability and cost of operation. As the research on nitrification inhibition characteristics is not sufficient till now, there is a great lack of theoretical guidance on the control of the inhibition. This study investigated the response of nitrifying activated sludge to chlorophenols (CPs) inhibition in terms of metabolism disorder and oxidative stress. At the initial stage of reaction (i.e., 1 h), reactive oxygen species (ROS)-induced membrane damage which might account for declining nitrification performance. Simultaneously excessive extracellular polymeric substances (EPS) were secreted to alleviate oxidative stress injury and protected microorganisms to some extent. In particular tyrosine-like substances in LB-EPS with a Fmax increase of 242.30% were confirmed to efficiently resist phenols inhibition. Thus, as the inhibition proceeded, metabolism disorder replaced oxidative stress as the main cause of nitrification inhibition. The affected metabolic processes include weakened enzyme catalysis, restricted electron transport and lessened energy generation. At 4 h, nitrifying production of sludge amended with 5 mg/L chlorophenols was 89.27 ± 9.51%-98.15 ± 9.60% lower than blank, the inhibition could be attributed to comprehensively affected metabolism. The structural equation modeling indicated that phenols restricted nitrification enzymes and bacterial electron transport efficiency which was critical to nitrification performance. Moreover, the lessened energy generation weakens enzyme activity to further suppress nitrification. These findings enriched our knowledge of nitrifiers' responses to CPs inhibition and provided the basis for addressing nitrification inhibition.

Abstract:The synergistic metabolism by anammox cultures and nitrate-reducers for anaerobic PAH biodegradation is largely unknown, including whether anammox culture and which kind of anammox bacterium can perform nitrogen metabolism in the anaerobic PAH biodegradation processes, the inhibitory effect of PAH on anammox activity and nitrite on PAH-degrading nitrate-reducers activity, and the synergistic metabolic processes. Herein, an anammox culture that can eliminate nitrite accumulation and decrease inorganic carbon emission during anaerobic phenanthrene (a model of PAH in this study) biodegradation, the synergistic mechanism for phen-anthrene biodegradation by a nitrate-reducer and such anammox culture, and the inhibition effect of phenan-threne on such anammox culture and nitrite on a phenanthrene-degrading nitrate-reducer were newly discussed. The results showed that nitrite largely accumulated during anaerobic phenanthrene biodegradation (nitrate accumulation is a common phenomenon for the biodegradation of refractory matter, including PAHs, by nitrate -reducers) by a nitrate-reducer, PheN2, which mineralizes phenanthrene to inorganic carbon, and nitrite was verified as an inhibiting factor for further biodegradation. Anaerobic phenanthrene biodegradation rates and nitrite concentrations (0-7 mM) appeared to have a negative linear correlation. The anammox culture that mainly contained Candidatus Kuenenia was newly found to efficiently reduce nitrite accumulation and inorganic carbon emissions and significantly promote biodegradation efficiency by similar to 1.94-fold. Our results showed that phenanthrene absorbed in and on anammox cells had a more direct relationship with the inhibitory effect on anammox activity than phenanthrene in the environment, and 15.2 mg/gVSS phenanthrene absorbed in and on the cells (4 mM concentration in the culture) showed nearly complete inhibition of anammox culture in this study. In addition, few (less than 2% abundance) anammox bacteria were found to be enough for the removal of nitrite produced from anaerobic phenanthrene biodegradation. In an ideal world, co-pollutants of ammonia, nitrate, phenanthrene, and nitrite could be converted to nitrogen gas and biomass by the synergistic metabolism of anammox cultures and nitrate reducers. Our study reveals a new synergistic process that may exist in our environments for PAH elimination by an anammox culture and a nitrate-reducer, which provides a new strategy for the bioremediation of PAH-polluted anoxic zones.
Anaerobic Degradation of Phenol in Wastewater at Ambient Temperature

Abstract:Treating a synthetic wastewater containing phenol as the sole substrate at 26°C, an upflow anaerobic sludge blanket reactor was able to remove over 98% of phenol up to 1,260 mg/l in wastewater with 12 h of hydraulic retention time, corresponding to 6.0 g-COD/(l·day). Results showed that benzoate was the key intermediate of phenol degradation. Conversion of benzoate to methane was suppressed by the presence of phenol. Based on DNA cloning analysis, the sludge was composed of five groups of microorganisms. Desulfotomaculum and Clostridium were likely responsible for the conversion of phenol to benzoate, which was further degraded by Syntrophus to acetate and H2/CO2. Methanogens lastly converted acetate and H2/CO2 to methane. The role of epsilon-Proteobacteria was, however, unclear.

Abstract:The effects of typical organic compounds including easily degradable organic matters sodium acetate, yeast and methanol, and refractory organic matter (ROM) humic acid on anaerobic ammonium oxidation (anammox) systems in short-term and medium-term exposure time were studied. During short-term experiments, nitrogen removal activity (NRA) was inhibited at sodium acetate level of 150 mg L-1 total organic carbon (TOC) and methanol level of 30-150 mg L-1 TOC, but humic acid and yeast (≤150 mg L-1 TOC) enhanced nitrogen removal in anammox systems. The greatest NRA of 30.10 mg TN g-1 VSS h-1 was recorded at yeast level of 90 mg L-1 TOC. In medium-term experiments, organics significantly inhibited the nitrogen removal ability. As a ROM, humic acid enhanced sludge aggregation and biological diversity, but decreased the bioactivity and extracellular polymeric substances levels. Due to the endogenous denitrification, the relative abundance of anammox bacteria (AnAOB) was decreased. Candidatus Kuenenia is still dominant in sludge with methanol and humid acid, but AnAOB are not dominant due to the addition of sodium acetate and yeast. This research would be beneficial for the full-scale application of the anammox process in treating real wastewater with organics and ammonia.

Abstract:Some industrial wastewaters contain high amounts of toxic nitrogen-containing heterocyclic compounds, which may inhibit the efficiency of biological treatment. This work systematically investigated how exogenous pyridine affected the anaerobic ammonia oxidation (anammox) system and discussed the microscopic response mechanisms based on genes and enzymes. The anammox efficiency was not seriously inhibited by pyridine less than 50 mg/L. Bacteria secreted more extracellular polymeric substances to resist pyridine stress. After 6 days stress with 80 mg/L pyridine, the nitrogen removal rate of anammox system lost 47.7%. Long-term stress of pyridine reduced anammox bacteria by 7.26% and the expression of functional genes by 45%. Pyridine could actively bind to hydrazine synthase and ammonium transporter. This work fills a research gap in the ongoing threat of pyridines to anammox, and has guiding value for the application of anammox process in the treatment of ammonia-rich wastewater containing pyridine.

Abstract:The effects of phenol on aerobic granular sludge including extracellular polymeric substances (EPS) and microbial community were investigated for low strength and salinity wastewater treatment. Elevated phenol over 20 mg/L stimulated biological phosphorus removal mainly via co-metabolism with nearly complete phenol degradation, whereas resulted in significant accumulation of nitrate around 4 mg/L. Aerobic granules kept structural stability via enhancing production of extracellular polymeric substances (EPS), especially folds of polysaccharides (PS) and varying functional groups identified through EEM, FTIR and XPS spectral characterizations at increasing phenol loads. Illumina MiSeq sequencing results indicated that elevated phenol decreased the bacterial diversity and richness, and caused remarkable variations in structural and compositions of microbial population. Multiple halophilic bacteria including Stappia, Luteococcus, and Formosa laid the biological basis for stability of aerobic granules and efficient biological nutrients and phenol removal. Redundancy analysis (RDA) suggested the key role of phenol in shaping the relative abundances and predominant genera. This study proved that aerobic granular sludge was feasible for low-saline and phenol-laden low-strength wastewater treatment.

Abstract:In the context of the ever more increasing number of cyber attacks targeted against Industrial Control Systems, the protection of data (e.g., process variables) commonly exchanged between the system's components (e.g., between Programmable Logic Controllers — PLCs, and Human Machine Interfaces — HMIs), is of greatest importance. In spite of their modest computational resources, we show that, when compared to other computer systems (e.g., traditional PCs), it is possible to enhance the control programs running within PLCs with Message Authentication Codes (MACs) constructions. These enable to authenticate the exchanged data, therefore providing a means to simultaneously verify both the data integrity and the authentication of process variables. This paper presents PLC-specific details for several MAC implementations and assesses their efficiency, in terms of execution time, as tested on Phoenix Contact's ILC 350 PN controller.

Abstract:This work compared the efficiencies and internal reasons for aniline removal in a bioaugmented sequencing batch reactor at elevated dissolved oxygen (DO) concentrations. Nearly complete aniline removal was achieved while medium DO of 2.65 mg/L was optimal for subsequent nitrogen removal via heterotrophic nitrification-aerobic denitrification. Apart from the highest bacterial diversity richness, favorable DO condition largely enriched putatively aniline degrader, nitrifiers and denitrifiers. Further evidence from qPCR confirmed that moderate DO significantly stimulated the functional genes including aniline degrading gene tad, nitrifying genes amoA, hao, and denitrifying genes nirS and napA, respectively. This study indicates that the significant enrichment of key microorganisms and effective functional genes under optimal DO is the inner mechanism for reliable aniline degradation and subsequent nitrogen removal in the activated sludge reactor.

Anaerobic Treatment of Phenol in Wastewater under Thermophilic Condition.

Abstract:Over 99% of phenol was effectively degraded in an upflow anaerobic sludge blanket (UASB) reactor at 55°C with 40h of hydraulic retention time (HRT) for a wastewater containing 630mg/L of phenol, corresponding to 1500mg/L of chemical oxygen demand (COD) and a loading rate of 0.9 g-COD/L/d. The maximum specific methanogenic activity (SMA) of the phenol-degrading sludge was 0.09 g-CH4-COD/g-volatile suspended solids (VSS)/d. Based on 16S rDNA analysis, a total of 21 operational taxonomy units (OTUs) were found in the sludge, of which eight (42.6% of the total population) were related to the sequences in the GenBank with similarity of over 97%, and 13 (79.6%) were affiliated with the known thermophilic species. Additional SMA data and phylogenetic analysis suggest that the degradation pathway of phenol for thermophilic sludge was likely via caproate, instead of benzoate as for the mesophilic sludge.

Abstract:The feasibility of a simultaneous nitrification, denitrification and fermentation process (SNDF) under electric stirrer agitation conditions was verified in a single reactor. Enhanced activated sludge for phenol degradation and denitrification in pharmaceutical phenol-containing wastewater under low dissolved oxygen conditions, additional inoculation with Comamonas sp. BGH and optimisation of co-metabolites were investigated. At a hydraulic residence time (HRT) of 28 h, 15 mg/L of substrate as strain BGH co-metabolised substrate degraded 650 ± 50 mg/L phenol almost completely and was accompanied by an incremental increase in the quantity of strain BGH. Strain BGH showed enhanced phenol degradation. Under trisodium citrate co-metabolism, strain BGH combined with activated sludge treated phenol wastewater and degraded NO 2 − -N from 50 ± 5 to 0 mg/L in only 7 h. The removal efficiency of this group for phenol, chemical oxygen demand (COD) and TN was 99.67%, 90.25% and 98.71%, respectively, at an HRT of 32 h. The bioaugmentation effect not only promotes the degradation of pollutants, but also increases the abundance of dominant bacteria in activated sludge. Illumina MiSeq sequencing research showed that strain BGH promoted the growth of dominant genera ( Acidaminobacter , Raineyella , Pseudarcobacter ) and increased their relative abundance in the activated sludge system. These genera are resistant to toxicity and organic matter degradation. This paper provides some reference for the activated sludge to degrade high phenol pharmaceutical wastewater under the action of biological enhancement.

Abstract:Thiocyanate (SCN − ) and phenol, as reducible inorganic and organic toxic compounds, coexist in coal and petrochemical wastewater. However, the kinetics and microbial mechanisms of them as electron donors for mixotrophic denitrification have not been investigated. In this study, thiocyanate autotrophic denitrification (TAD) sludge was used to explore the kinetics and microbial mechanisms of mixotrophic denitrification using phenol and SCN − as electron donors. The batch experimental results showed that TAD sludge can efficiently use SCN − as electron donor for denitrification. Low-concentration phenol to TAD sludge presented no significantly affect SCN − degradation and only slightly inhibited denitrification. Higher 300 mg/L phenol significantly hampered both processes. However, the sludge gradually acclimated to phenol in continuous-flow reactor, achieving stable mixotrophic denitrification. Metagenomic analysis revealed Thiobacillus (sole SCNase-harboring genus) and Ottowia as dominant players. Thiobacillus , responsible for SCN − degradation, had complete sulfur oxidation and denitrification enzyme system. Ottowia , responsible for phenol degradation, also had some sulfur oxidation and denitrification genes. Under the leadership of Thiobacillus and Ottowia , various microorganisms collaborated to utilize SCN − and phenol for mixotrophic denitrification, which result in SCN − , phenol, and NO 3 − -N removed simultaneously. This study provides new insights into the denitrification in toxic wastewater and establishes a metabolic model for it.

Abstract:BACKGROUND: In biological treatment of coking wastewater, phenol may decrease the treatment efficiency because of its high concentration and toxicity to microorganisms. Bioaugmentation has been regarded as a good improvement of the traditional biological treatment using isolated degrading strains. In this study, two phenol degrading strains, Pseudomonas sp. PCT01 and PTS02, were isolated and investigated for degradation ability and application to real coking wastewater treatment. RESULTS: Complete phenol degradation was achieved after 18 h inoculation in medium containing 229-461 mg L-1 of phenol for both strains. The presence of phenol, pyridine and other compounds in mixed substrate or in coking wastewater prolonged the degradation to 20-32 h with an initial phenol concentration of 160-280 mg L-1. The study of degradation kinetics yielded a two-stage model to describe the effect of the initial phenol concentration and inhibitory compounds on phenol degradation. The highest degradation rate constant of the second stage, 1.25 h(-1) for PCT01 and 0.75 h(-1) for PTS02, was obtained at low phenol concentration in a single substrate. CONCLUSION: It was found that both strains could degrade phenol effectively and maintain their phenol degradation ability in coking wastewater, and therefore could be used for bioaugmentation treatment of coking wastewater. Revealing the Comprehensive Impact of Organic Compounds on the Partial Nitrification-Anammox System During Incineration Leachate Treatment: Metabolic Hierarchy and Adaptation

Abstract:Organics, as widespread pollutants in high-strength ammonia wastewater, typically exert adverse effects on the performance of partial nitrification-anammox (PNA) systems. However, the in-depth knowledge on how microbial consortia respond to these disturbances remains limited. In this study, we unveiled the evolution of complex organic matter flow and its impact on the metabolic hierarchy and adaptation of microbial consortia, employing multi-omics approaches, i.e., 16S amplicon sequencing, metagenomics, and metabolomics. In a two-stage PNA system sequentially treating synthetic wastewater and incineration leachate over 230 days, partial nitrification stayed stable (nitrite accumulation > 97%) while anammox efficiency dropped (nitrogen removal decreased from 86% to 78%). The phenomenon was revealed to be correlated with the evolution of dissolved organic matter (DOM) and xenobiotic organic compounds (XOCs). In the PN stage, ammonia-oxidizing bacteria (AOB) exhibited excellent adaptability through active metabolic regulation after treating leachate. Numerous heterotrophs proliferated to utilize DOM and XOCs, triggering a “boom” state evident in the glycerophospholipid metabolism. However, in the anammox stage, the competition between carbon fixation and central carbon metabolism within autotrophs and heterotrophs became evident. Increased biosynthesis costs inhibited the central metabolism (specific anammox activity decreased by 66%) and the Wood-Ljungdahl pathway of anammox bacteria (AnAOB) in the presence of recalcitrant organics. Additionally, the degradation of organics was limited, exhibiting a “bust” state. This study revealed the metabolic adaption and susceptibility of AOB and AnAOB in response to organics from the leachate, demonstrating the applicability of the two-stage configuration for treating high-strength wastewater containing abundant and diverse organics.

Abstract:The aim of this work is to study the synergistic effect of Stenotrophomonas sp. N5 and Advenella sp. B9 co-culture (COC) on enhancement of phenol biodegradation. These two strains utilizing phenol as sole carbon and energy source were isolated from phenol-containing coking wastewater. The results of biodegradation experiment showed the COC of N5 and B9 has stronger capability to degrade phenol than either of mono-culture (MOC). Growth kinetics studies indicated inhibitory effect of phenol on COC was reduced by the interaction of N5 and B9 in COC. The RNA-Seq results demonstrated that phenol biodegradation was enhanced by metabolic division of labor (DOL) in COC based on the expression of key genes for phenol degradation. GO enrichment analysis of differentially expressed genes (DEGs) indicated DEGs between COC and MOC degradation systems are mainly concentrated in the synthesis of cell components, microbial growth and metabolism, and catalytic activity. The expression of 3 transcriptional factors (LysR, Two-component system response regulator, and TetR families) which can regulate degradation of aromatic compounds, was identified beneficial to phenol degradation.
