Abstract:We demonstrated that pretreatment with chromium (Cr) significantly alters inflammatory responses of mice or macrophage cell lines. The mice were pretreated with 50 and 200 mg L(-1) of Cr dissolved in drinking water for 7 or 21 d, respectively. Then, the mice were challenged with lipopolysaccharide (LPS) or saline for 3 h. The body and liver weights significantly decreased after exposure to 200 mg L(-1) of Cr for both 7 and 21 d. Serious infiltration of inflammatory cells around the artery was found in the liver treated with 200 mg L(-1) of Cr for 7 and 21 d. The levels of tumor necrosis factor-α (TNFα) and interleukin-6 (IL6) in peritoneal macrophage significantly increased after the treatment with 200 mg L(-1) of Cr for 7 d. Moreover, LPS-induced increases in the serum levels and the transcriptional status of some cytokine genes were amplified by the Cr pretreatment. In the in vitro test, the RAW264.7 cell line was pretreated with Cr for 3, 6, 12, and 24 h, followed by stimulation with LPS (1 μg mL(-1)) for 6 h. LPS-induced the increases in TNFα, IL6, Interleukin-1α (IL1α), Interleukin-1β (IL1β), inducible nitric oxide synthase (iNOS), and cyclooxygenase-2 (COX2) mRNA levels were significantly promoted by the pretreatment with Cr for 3, 6, and 12 h, whereas they were weakened by the pre-exposure to Cr for 24 h in a concentration-dependent manner. In addition, LPS-induced the release of TNFα and IL6 in the medium was also significantly enhanced or suppressed by the different Cr pretreatment. The results suggested that Cr had the potential to induce immunotoxicity by altering the inflammatory responses.
Purified human plasma glycosaminoglycans reduced NF-κB activation, pro-inflammatory cytokine production and apoptosis in LPS-treated chondrocytes

Abstract:Introduction: There have been several cases reporting a significant increase in chondroitin sulphate plasma levels in patients with different types of disease, such as systemic lupus erythematosus, rheumatoid arthritis, and liver disease. At present, the precise role of chondroitin sulphate molecules in blood is unclear. Previous investigations have shown that the addition of purified human plasma glycosaminoglycans (GAGs), containing a high percentage of chondroitin-4-sulphate (C4S) was able to inhibit lipid peroxidation and to protect cells from reactive oxygen species damage, suggesting antioxidant activity. Starting from these reports, the aim of this study was to evaluate the effectiveness of GAG structures purified from normal human plasma in reducing inflammation using a model of lipopolysaccharide (LPS)-induced increase of pro-inflammatory cytokines in mouse articular chondrocyte cultures. Results: Chondrocyte stimulation with LPS (50 μg/ml) for 24 h enhanced gene expression of tumor necrosis factor alpha (TNF-α), interleukin-1 beta (IL-1β), interleukin 6 (IL-6), interferon gamma (IFN-γ), inducible nitric oxide synthase (iNOS) and increases in their related protein levels, as well as NF-κB activation, IκBα phosphorylation and apoptosis evaluated by the increase in caspase-3 expression and its related protein amount. LPS treatment also generated a high amount of nitric oxide (NO). The addition of different doses of purified human GAGs to LPSstimulated chondrocytes reduced inflammatory cytokines and iNOS both at mRNA and protein levels, blocked NF-κB activation and cytoplasmic IκBα phosphorylation, limited cell death by inhibiting apoptosis, and reduced NO concentrations. Conclusions: These results further support the hypothesis that plasma GAGs may function as immunomodulators and their increased release and degradation could be a biological response acting to modulate inflammation during disease.

Abstract:The physiological problem of chronic inflammation and its associated pathologies attract ongoing attention with regard to methods for their control. Current systemic pharmacological treatments present problematic side effects. Thus, the possibility of new anti‐inflammatory compounds with differing mechanisms of action or biophysical properties is enticing. Cationic polymers, with their ability to act as carriers for other molecules or to form bio‐compatible materials, present one such possibility. Although not well described, several polycations such as chitosan and polyarginine, have displayed anti‐inflammatory properties. The present work shows the ubiquitous laboratory transfection reagent, polyethylenimine (PEI) and more specifically low molecular weight branched PEI as also possessing such properties. Using a RAW264.7 murine cell line macrophage as an inflammation model, it was found the B‐PEI 700 Da as being capable of reducing the production of several pro‐inflammatory molecules induced by the endotoxin lipopolysaccharide. Although further studies are required for elucidation of its mechanisms, the revelation that such a common lab reagent may present these effects has wide‐ranging implications, as well as an abundance of possibilities. Abstract:AMP-activated protein kinase (AMPK) is a serine/threonine protein kinase critically involved in the regulation of cellular energy homeostasis. Many studies have suggested that AMPK activation exert significant anti-inflammatory effects. In this study, we assessed the anti-inflammatory effect of AMPK activator from natural products, Broccoli seed (BS, Brassica oleracea var. italic) extract and Sweet tea (ST, Rubus Suavissmus S. Lee) extract, in RAW264.7 cells. AMPK assay Natural product samples treated purified AMPK enzyme (CycLex Co., Ltd. Japan) or cell lysates were added to the well of CycLex AMPK kinase assay kit, then performed. Cell Culture HepG2 and RAW264.7 cells were cultured in Dulbecco’s Modified Eagle Medium supplemented with buffer. ELISA assay of IL-6 RAW264.7 cells were incubated with LPS, with or without presence of natural products, IL-6 in the supernatant was determined with Mouse IL-6 ELISA Kit. Western Blotting Cells were treated with LPS in the presence of or absence of natural products for 6 h, then lysed with buffer and a protease inhibitor. Proteins separated and blotted onto PVDF, blocked and probed with antibodies against caspase-1, finally incubated with the goat anti-rabbit horseradish-peroxidase coupled secondary antibody. The target proteins were analyzed using the ChemiDoc XRS System and quantified by ImageJ. Natural products BS and ST were identified as AMPK activators with the established AMPK activity assay model. (Fig. 1) Macrophages were stimulated with LPS, while the secretion of IL-6 was decreased in the presence of BS or ST, the protective effect observed of BS on the LPS-induced secretion of IL-6 was 0.0625–0.125 mg/mL, ST at 0.25–0.5 mg/mL. (Fig. 2) Furthermore, BS and ST suppressed the LPS-induced expression of caspase-1 gene in dose-dependent manner. (Fig. 3–4) Caspase-1 is a cysteine protease that converts the inactive form of IL-1β to the active inflammatory cytokine. Our preliminary study showed that AMPK activators, BS and ST, reduced LPS-induced inflammation in macrophages by combined inhibition of IL-6 secretion and block of inflammatory gene caspase-1 expression in RAW264.7 macrophage cell line. This result implicates anti-inflammatory effect of BS and ST, which may be induced by activation of AMPK and then regulated via NLRP3 pathway and caspase-1 expression. [Effect of Lipopolysaccharide of Porphyromonas Gingivalis on Prostaglandin E2 Biosynthetic Pathway in Human Monocytic Cell Strain THP-1].

Abstract:OBJECTIVE:To investigate the effect of lipopolysaccharide of Porphyromonas gingivalis (Pg-LPS) on the bio-thythetic pathway of prostaglandin E2 (PGE2) and its difference from lipopolysaccharide of Escherichia coli (Ec-LPS).METHODS:Purified Pg-LPS and Ec-LPS were used to stimulate a human monocytic cell strain THP-1. PGE2 concentration was determined by an enzyme immunoassay kit. The release of tritium labeled arachidonic acid (AA) was detected by a liquid scintillation counter. Reverse transcription polymerase chain reaction and western blot were used to analyse the expression of cytosolic phospholipase A2 (cPLA2) enzyme, cyclooxygenase-2 (COX-2), and microsomal prostaglandin E synthase-1 (mPGES-1).RESULTS:The effect of Pg-LPS on induction of PGE2 and release of AA was significantly weaker than that of Ec-LPS (P < 0.05).Increased secretion of PGE2 was observed after stimulation with Pg-LPS for 6 h, which peak at 24 h at (221.40 +/- 29.46) ng/L; or with Ec-LPS for 1-48 h, at (161.80 +/- 17.31) approximately (379.80 +/- 37.35) ng/L. The highest levels of COX-2 and mPGES-1 were shown after 16 h treatment by Pg-LPS, or after 8 h and 16 h by Ec-LPS respectively.cPLA2 inhibitor AACOCF3 could lower the level of LPS-induced release of AA, while it did not influence the production of PGE2. COX-2 inhibitor NS-398 could remarkably reduce the concentration of PGE2.CONCLUSIONS:Pg-LPS showed delayed and weaker effect on PGE2 biosynthetic pathway than Ec-LPS. Pg-LPS-induced PGE2 synthesis was mainly due to enhanced expression of COX-2 and mPGES-1, whereas cPLA2 played an insignificant role.

Abstract:To determine the effect of Porphyromonas gingivalis lipopolysaccharide (LPS) on induced secretion of inflammatory cytokines by different cell lines.LPS of P. gingivalis strain ATCC33277 (Pg-LPS) was extracted with phenol-water method, and identified by Limulus test and infrared spectrum analysis. KB, HGF-1 and THP-1 cells were treated with Pg-LPS of different concentrations and time duration, a commercial LPS of E.coli strain O111:B4 (E-LPS) was used as the control. TNF-alpha, IL-1beta, IL-6 and IL-8 levels in culture supernatants were measured by quantitative ELISA.The minimal dosages of both Pg-LPS and E-LPS to solidify Limulus agents were 15 ng/ml and their infrared spectrums were similar. With the treatment of Pg-LPS or E-LPS, the TNF-alpha and IL-1beta levels secreted by HGF-1 cells were remarkably increased with a single perk (P<0.01) while a continuous enhancement of secretion by THP-1 cells was observed (P<0.01). Either Pg-LPS or E-LPS stimulated HGF-1 or THP-1 cells to continuously increase the secretion of IL-6 (P<0.01). Both Pg-LPS and E-LPS induced IL-8 secretion by THP-1 cells (P<0.01), but only Pg-LPS showed the similar effect on HGF-1 cells (P<0.01). Neither Pg-LPS nor E-LPS induced KB cells to secrete inflammatory cytokines.Pg-LPS can promote target cells to increase their secretion of inflammatory cytokines. KB cells can not be used as the target cell to determine inflammation-causing effect of LPS.
Choline Glycerophospholipid-Derived Prostaglandins Attenuate TNFα Gene Expression in Macrophages via a cPLA2α/COX-1 Pathway

Abstract:Macrophages are professional antigen presenting cells with intense phagocytic activity, strategically distributed in tissues and cavities. These cells are capable of responding to a wide variety of innate inflammatory stimuli, many of which are signaled by lipid mediators. The distribution of arachidonic acid (AA) among glycerophospholipids and its subsequent release and conversion into eicosanoids in response to inflammatory stimuli such as zymosan, constitutes one of the most studied models. In this work, we used liquid and/or gas chromatography coupled to mass spectrometry to study the changes in the levels of membrane glycerophospholipids of mouse peritoneal macrophages and the implication of group IVA cytosolic phospholipase A2 (cPLA2α) in the process. In the experimental model used, we observed that the acute response of macrophages to zymosan stimulation involves solely the cyclooxygenase-1 (COX-1), which mediates the rapid synthesis of prostaglandins E2 and I2. Using pharmacological inhibition and antisense inhibition approaches, we established that cPLA2α is the enzyme responsible for AA mobilization. Zymosan stimulation strongly induced the hydrolysis of AA-containing choline glycerophospholipids (PC) and a unique phosphatidylinositol (PI) species, while the ethanolamine-containing glycerophospholipids remained constant or slightly increased. Double-labeling experiments with 3H- and 14C-labeled arachidonate unambiguously demonstrated that PC is the major, if not the exclusive source, of AA for prostaglandin E2 production, while both PC and PI appeared to contribute to prostaglandin I2 synthesis. Importantly, in this work we also show that the COX-1-derived prostaglandins produced during the early steps of macrophage activation restrict tumor necrosis factor-α production. Collectively, these findings suggest new approaches and targets to the selective inhibition of lipid mediator production in response to fungal infection.
Phloroglucinol possesses anti-inflammatory activities by regulating AMPK/Nrf2/HO-1 signaling pathway in LPS-stimulated RAW264.7 murine macrophages

Abstract:Inflammation is closely related to the pathogenesis of chronic illnesses. Secondary metabolites of marine seaweeds are recognized as reliable sources of bioactive compounds due to their health benefits besides their nutritional value. The objective of this study was to determine the potential anti-inflammatory effect of phloroglucinol (Phl) in RAW264.7 murine macrophages after lipopolysaccharides (LPS) stimulation. MTT, nitric oxide (NO), and DCFH-DA assays were conducted to determine cell viability, NO production, and reactive oxygen species (ROS) generation respectively. Pro-inflammatory cytokines and prostaglandin E 2 (PGE 2 ) levels were measured using ELISA assay kits. Protein expression levels were determined by western blot analysis. Phl treatment showed a promising anti-inflammatory effect by reducing NO production, secretion of pro-inflammatory cytokines (TNF-α, IL-1β, and IL-6), PGE 2 production, protein expression levels of inducible nitric oxide synthase (iNOS), and cyclooxygenase-2 (COX-2), and ROS generation in LPS-stimulated RAW264.7 murine macrophages. Phl treatment upregulated heme oxygenase-1 (HO-1) expression by inducing nuclear translocation of nuclear factor erythroid 2-related factor 2 (Nrf2) and activating AMPK. However, Zinc protoporphyrin (ZnPP), an inhibitor of HO-1, partially reversed these effects, including NO production, pro-inflammatory cytokine secretion, iNOS, COX-2 and HO-1 expression, and ROS generation. Phl has potential anti-inflammatory activities by regulating AMPK/Nrf2/HO-1 pathway in LPS-stimulated RAW264.7 murine macrophages.

In vitro characterization of lactic acid bacterial strains isolated from fermented foods with anti-inflammatory and dipeptidyl peptidase-IV inhibition potential

Abstract:Probiotics are known to stimulate, modulate, and regulate host immune response by regulating specific sets of genes and improve glucose homeostasis through regulating dipeptidyl peptidase (DPP-IV) activity, but the mechanism behind their protective role is not clearly understood. Therefore, the present study was designed to isolate indigenous lactic acid bacterial (LAB) strains from different fermented food samples, vegetables, and human infant feces exhibiting anti-inflammatory, antioxidant, and DPP-IV inhibitory activity. A total of thirty-six Gram-positive, catalase-negative, and rod-shaped bacteria were isolated and screened for their anti-inflammatory activity using lipopolysaccharide (LPS)-induced inflammation on the murine (RAW264.7) macrophages. Among all, sixteen strains exhibited more than 90% reduction in nitric oxide (NO) production by the LPS-treated RAW264.7 cells. Prioritized strains were characterized for their probiotic attributes as per the DBT-ICMR guidelines and showed desirable probiotic attributes in a species and strain-dependent manner. Accordingly, Lacticaseibacillus rhamnosus LAB3, Levilactobacillus brevis LAB20, Lactiplantibacillus plantarum LAB31, Pediococcus acidilactici LAB8, and Lactiplantibacillus plantarum LAB39 were prioritized. Furthermore, these strains when co-supplemented with LPS and treated on RAW264.7 cells inhibited the mitogen-activated protein kinases (MAPKs), i.e., p38 MAPK, ERK1/2, and SAPK/JNK, cyclooxygenase-2 (COX-2), relative to the LPS-alone-treated macrophages. LAB31 and LAB39 also showed 64 and 95% of DPP-IV inhibitory activity relative to the Lacticaseibacillus rhamnosus GG ATCC 53103, which was used as a reference strain in all the studies. Five prioritized strains ameliorated the LPS-induced inflammation by downregulating the JNK/MAPK pathway and could be employed as an alternative bio-therapeutic strategy in mitigating gut-associated inflammatory conditions. The potential mechanism of action of prioritized LAB strains in preventing the LPS-induced inflammation in RAW 264.7 macrophage cells.
Phosphatidylserine-Specific Phospholipase A1 Alleviates Lipopolysaccharide-Induced Macrophage Inflammation by Inhibiting MAPKs Activation.

Abstract:Macrophages are a key in innate immune responses and play vital roles in homeostasis and inflammatory diseases. Phosphatidylserine-specific phospholipase A1 (PS-PLA1) is a specific phospholipase which hydrolyzes fatty acid from the sn-1 position of phosphatidylserine (PS) to produce lysophosphatidylserine (lysoPS). Both PS and lysoPS are associated with activation of immune cells including macrophages. However, the effect of PS-PLA1 on macrophage inflammation remains unclear. The purpose of this study is to evaluate the role of PS-PLA1 in lipopolysaccharide (LPS)-induced macrophage inflammation. Alterations of PS-PLA1 expression in LPS-stimulated RAW264.7 macrophages were investigated via Western blot. PS-PLA1 stable knockdown and overexpression RAW264.7 cell lines were generated by infecting cells with appropriate lentiviral vectors, respectively. PS-PLA1 expression was found to be dramatically upregulated in RAW264.7 macrophages after LPS stimulation. PS-PLA1 knockdown promotes while PS-PLA1 overexpression ameliorates the release of tumor necrosis factor (TNF)-α, interleukin (IL)-1β and nitric oxide from RAW264.7 cells and M1 macrophage polarization. Additionally, PS-PLA1 knockdown facilitates phosphorylation of p38, extracellular signal-regulated kinase (ERK) and c-Jun N-terminal kinase (JNK), while PS-PLA1 overexpression attenuates their phosphorylation. Moreover, mitogen-activated protein kinase (MAPK) inhibitors block the release of TNF-α and IL-1β in PS-PLA1 knockdown RAW264.7 cells after LPS stimulation. These findings suggest PS-PLA1 ameliorates LPS-induced macrophage inflammation by inhibiting MAPKs activation, and PS-PLA1 might be considered as a target for modulating macrophage inflammation.

Abstract:Obesity-induced adipose chronic inflammation is closely related to the development of insulin resistance and T2DM. Tripeptides l-valyl-l-prolyl-l-proline (VPP) and l-isoleucyl-l-prolyl-L-proline (IPP) derived from bovine casein have been reported to prevent inflammatory changes and mitigate insulin resistance in adipocytes. In this study, we aimed to investigate the influence of casein hydrolysates (CH) containing VPP and IPP on a high fat diet (HFD)-induced obese mice and cytokine TNF-α-induced adipocytes. Our data showed that CH alleviated chronic inflammation both in vivo and in vitro. 4% CH suppressed HFD-induced systemic inflammatory factors, hypertrophic white adipocytes, and macrophage infiltration. More importantly, CH was able to improve adipocyte dysfunction induced by TNF-α by increasing the expression of CCAAT/enhancer binding protein α (C/EBP-α) rather than peroxisome proliferator-activated receptor γ (PPAR-γ). Furthermore, CH also dose-dependently suppressed mitogen-activated protein kinase (MAPK)-c-Jun N-terminal kinase (JNK) phosphorylation and enhanced the phosphorylation of Erk 1/2, but not nuclear factor-kappa B (NF-κB) p65 phosphorylation, in TNF-α-induced 3T3-L1 cells. These results indicated that CH could ameliorate adipose chronic inflammation through the MAPK pathway. Altogether, our findings suggested that 4% CH supplementation for 6 weeks exerted a protective role in preventing obesity-related inflammation and adipose dysfunction.

Abstract:Objective To investigate the effects of chitosan/pCDNA3.1 + cytokine rcsponse modifier A (CrmA) nanoparticles on interkin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) expression of chondrocytes.Methods Rabbit chondrocytes were isolated and cultured,and treated with PBS,10 mg/L chitosan (CS) or chitosan/pCDNA3.1 + CrmA nanoparticles,respectively for 6 h.Then 10 μ g/L IL-1β was added into the culture medium.After 48 h,the mRNA expression of IL-13 and TNF-α in chondrocytes was detected by using real-time polymerase chain reaction.Results In chitosan/pCDNA3.1 + CrmA-treated group (0.55 ±0.08),the mRNA expression of IL-1β in chondrocytes was significantly suppressed as compared with corresponding samples of chitosan-treated group (0.69 ± 0.06,P ＜ 0.01) and PBS-treated group (0.99 ±0.04,P ＜ 0.01).There was significant difference in the I L-1 β expression between chitosantreated group and PBS-treated group (P ＜ 0.01).The TNF-α mRNA expression of chondrocytes in chitosan/pCDNA3.1 + CrmA-treated group (0.57 ± 0.07) was lower than that in chitosan-treated group (0.71 ± 0.05,P ＜ 0.01) and PBS-treated group (0.99 ± 0.06,P ＜ 0.01).Significant difference of TNF-α expression between chitoson treated group and PBS treated group was observed (P ＜ 0.01).Conclusion CrmA mediated by chitosan could significantly suppress the mRNA expression of IL-1β and TNF-α of chondrocytes induced bv interkin-1 β.chitosan/pCDNA3.1 * CrmA nanoparticles maybe a potential candidate for therapy of osteoarthritis.

Abstract:The bioactive peptides hydrolyzed from bone collagen have been found to possess health-promoting effects by regulating chronic diseases such as arthritis and hypertension. In the current study, the anti-inflammatory effect of bovine bone gelatin peptides (GP) was evaluated in 264.7 macrophages cells and followed by animal trials to investigate their interference on inflammatory cytokines and gut microbiota compositions in dextran sodium sulfate (DSS)-induced C57BL/6 mice. The GP was demonstrated to alleviate the extra secretion of interleukin-6 (IL-6), nitric oxide (NO) and tumor necrosis factor-α(TNF-α) in lipopolysaccharide (LPS)-induced RAW264.7 cells. In DSS-induced colitis mice, the gavage of GP was demonstrated to ameliorate the IBD symptoms of weight loss, hematochezia and inflammatory infiltration in intestinal tissues. In serum, the proinflammatory cytokines (TNF-α,IL-6, MCP-1, IL-1β) were suppressed along with the decreasing effect on toll-like receptor 4 and cyclooxygenase-2 by GP treatment. In the analysis of gut microbiota, the GP was checked to modulate the abundance of Akkermansia, Parasutterella, Peptococcus, Bifidobacterium and Saccharibacteria. The above results imply that GP could attenuate DSS-induced colitis by suppressing the inflammatory cytokines and regulating the gut microbiota.

[Extremely low birthweight infants in iceland. Survival and disability.].

Abstract:OBJECTIVE In recent years advances in medical care and technology have increased newborn survival rate, both fullterm and preterm. This is reflected in a low Perinatal Mortality Rate in Iceland. Survival of extremely low birthweight infants (ELBW with BW<1000g) has also increased, especially since the availability of surfactant therapy for Respiratory Distress Syndrome of Prematurity. The purpose of this geographically defined national study was to evaluate survival and longterm outcome of ELBW children in Iceland. MATERIAL AND METHODS Information on all births in Iceland 1982-95 was collected from the National Birth Registry and Statistics Iceland with information on ELBW infants weighing 500-999g born in two periods 1982-90 and 1991-95, before and after the use of surfactant became routine therapy. Information on disability was obtained from records at the State Social Security Institute. Comparison was made between the two groups of ELBW infants. RESULTS In 1982-90 the proportion of ELBW infants was 0.3% of all births (116 of 38.378) and longterm survival at five years of age was 19 of 87 liveborn children or 22%. In 1991-95 ELBW infants were 0.5% of all births (102 of 22.261) and longterm survival was 35 of 67 liveborn children or 52%. Of the 19 ELBW children born in 1982-90 three are considered handicapped (16%) and 6 of 35 ELBW children born in 1991-95 (17%). CONCLUSIONS The study shows that at the same time that proportionally more children are of extreme low birthweight, the survival of ELBW infants has increased from 22% in 1982-90 to 52% in 1991-95. The proportion of ELBW children with disability is not increased significantly between the two periods.
Casein Oligochitosan-Glycation by Transglutaminase Enhances the Anti-Inflammatory Potential of Casein Hydrolysates to the Lipopolysaccharide-Stimulated IEC-6 Cells

Abstract:In this study, milk protein casein was glycated by oligochitosan through the catalysis of transglutaminase (TGase) and then hydrolyzed by trypsin. The obtained glycated casein hydrolysates (GCNH) were assessed for their anti-inflammatory activities, using the lipopolysaccharide (LPS)-stimulated rat intestinal epithelial cells (IEC-6) as cell models and the casein hydrolysates (CNH) without TGase catalysis as controls. The results showed that GCNH had oligochitosan incorporation and thus possessed a glucosamine content of 5.74 g/kg protein. In general, GCNH at dose levels of 25–100 μg/mL could elevate IEC-6 cell growth, and at dose levels of 25–50 μg/mL, they were also able to alleviate the LPS-induced cytotoxicity by increasing cell viability efficiently. Although LPS caused clear inflammation in the LPS-stimulated cells, GCNH were capable of reducing the secretion of three pro-inflammatory mediators including interleukin-1β (IL-1β), IL-6, and tumor necrosis factor-α, or promoting the secretion of two anti-inflammatory mediators like IL-10 and transforming growth factor-β, demonstrating their anti-inflammatory activities to the stimulated cells. Moreover, GCNH also could down-regulate the expression of three inflammation-related proteins including TLR4, p-p38, and p-p65 in the stimulated cells, and thus possessed a capacity to suppress the phosphorylation of p38 and p65 proteins as well as to inactivate the NF-κB and MAPK signaling pathways. Additionally, a higher GCNH dose level consistently led to higher anti-inflammatory effect in the cells, while GCNH were always more potent than CNH at performing anti-inflammatory function targets. It is thus suggested that the TGase-catalyzed casein oligochitosan-glycation could enhance the anti-inflammatory activities of casein hydrolysates efficiently. TGase-catalyzed protein glycation thus might enhance the healthcare function of protein ingredients in the body.

Abstract:Chalcones (1,3-diaryl-2-propen-1-ones), a subfamily of flavonoid, are widely known to possess potent anti-inflammatory and anti-oxidant properties. In this study, we investigated the effect of 3-(4-Hydroxyphenyl)-1-(thio3-(4-Hydroxyphenyl phen-2-yl)prop-2-en-1-one (TI-I-175), a synthetic chalcone derivative, on endotoxin-induced expression of monocyte chemoattractant protein-1 (MCP-1), one of the key chemokines that regulates migration and infiltration of immune cells, and its potential mechanisms. TII-175 potently inhibited MCP-1 mRNA expression stimulated by lipopolysaccharide (LPS) in RAW 264.7 macrophages without significant effect on cell viability. Treatment of cells with TI-I-175 markedly prevented LPS-induced transcriptional activation of activator protein-1 (AP-1) as measured by luciferase reporter assay, while nuclear factor-κB (NF-κB) activity was not inhibited by TI-I-175, implying that TI-I-175 suppressed MCP-1 expression probably via regulation of AP-1. In addition, TI-I-175 treatment significantly inhibited LPS-induced Akt phosphorylation and led to a significant decrease in reactive oxygen species (ROS) production by LPS, which act as up-stream signaling events required for AP-1 activation in RAW 264.7 macrophages. Taken together, these results indicate that TI-I-175 suppresses MCP-1 gene expression in LPS-stimulated RAW 264.7 macrophages via suppression of ROS production and Akt activation.

Abstract:The aim of this study was to evaluate the effects of chitosan and quaternized chitosan (HTCC) modulate IL-1β and TNF-α in LPS-stimulated human periodontal ligament cells (HPDLCs). MTT assay revealed that chitosan stimulated the proliferation of HPDLCs. However, HTCC inhibited the proliferation of HPDLCs at concentrations of 1000 and 100 μg/mL more than the control, especially after 5d (P<0.001). ELISA assay exhibited that chitosan inhibited the production of IL-1β and TNF-α at 24, 48 and 72 h. IL-1β and TNF-α secreted by HPDLCs with LPS and treated with 1000 μg/mL of HTCC significantly increased compared to both the control and the chitosan group (P<0.001). The bioactive role for bFGF in modulating the responses of HPDLCs cells to LPS via inhibiting IL-1β and TNF-α production was demonstrated. All results were necessary to enhance our understanding of the biomedical properties of chitosan and HTCC for modulation of pro-inflammatory mediators.
Second scatter contribution to dose in a cobalt-60 beam.

Abstract:Semi-analytic and direct numerical integration approaches have been used to study the contribution of doubly scattered photons to dose at a point P in a homogeneous water medium irradiated by a cobalt-60 point source. The semi-analytic analysis was used for an infinite field radius to calculate the variation of second scatter dose with depth, the angular dependence of the second scatter dose, and the spectra. Direct numerical integration was used to calculate the second scatter dose at a depth of 10 cm for various finite field radii ranging from 0 to 25 cm. Maps were made of the locations of scattering sites that were important to the second scatter dose. These calculations show that the second scatter contribution is generally less than the first scatter dose with the exception of a few sites remote from the point of interest. In many ways, the pattern of dose deposition by second scatter is similar to first scatter with less pronounced features. In other respects, the second scatter appears more isotropic. The implications of these calculations for approximate radiotherapy dose calculations are discussed.
Immunomodulating effects of casein hydrolysates on murine peritoneal macrophages

Abstract:showed that the precursor form (i.e. full-length) of casein did not enhance phagocytosis activity, nitric oxide (NO) release, or cytolysis activity by lipopolysaccharide (LPS)-stimulated or un-stimulated mouse macrophages, while casein alcalase hydrolysate could enhance mouse macrophage activity. Casein hydrolysate could stimulate the phagocytosis activity of mouse peritoneal macrophages with or without the presence of LPS. It could induce the NO release and tumoricidal activity to Jurkat cells by LPS-stimulated mouse macrophages, but casein hydrolysate alone could not stimulate NO release, neither increase the tumoricidal activity to Jurkat cells of resting peritoneal macrophages. This study also showed that casein alcalase hydrolysate had dose-dependent effect on mouse macrophage activity, with the dose of 0.1 mg/ml had the best effect. Casein-derived peptide can have important implications on the modulation of murine peritoneal macrophages.
