Abstract:Dopamine (DA) is catecholamine neurotransmitters that play an important role in the central nervous system. In recent years people started to intentionally add DA to animal feed to enhance muscle development and increase their profit margin. Human consumption of the residual DA from animal tissues has been reported to be associated with the development of such diseases as Parkinson's disease, epilepsy, senile dementia, and schizophrenia and pose serious human health risks. These require development of rapid, cheap, and sensitive methods for detection of DA from animal tissue. Compared to other techniques that require access to expensive instruments, skilled human power, and tiresome routine procedures, colorimetric methods provide cheap and reliable options for detection of DA. Here we report a colorimetric method based on the peroxidase-mimic activity of Fe3O4@C@AgNPs for the detection of DA. A simple wet chemical method was employed to synthesize AgNPs on hydrophilic carbon coated Fe3O4. The produced nanocomposites were characterized by transmission electron microscopy (TEM), Fourier Transform infrared spectroscopy (FTIR), and surface-enhanced Raman spectroscopy (SERS). The detection of DA was done based on inhibition of the peroxidase-like activity of Fe3O4@C@AgNPs using 3, 3', 5, 5'-tetramethylbenzidine (TMB) as a substrate. In the presence of DA, however, the peroxidase-like activity started to decrease. The decrease in activity was concentration dependent showing a linear relationship in the range of 0.5-80 µM. In this linear range, the limit of detection (LOD) was computed and found to be as low as 0.12 µM. Therefore, we propose that the peroxidase-like activity of Fe3O4@C@AgNPs could be used for quantitative detection of DA from different samples.
High oxidase-mimic activity of Fe nanoparticles embedded in an N-rich porous carbon and their application for sensing of dopamine

Abstract:The N-doped porous carbon (NC) has been regarded as one of the promising support materials for nanoparticles (NPs) catalyst due to its inherent virtues such as porosity, large surface areas, and heteroatom incorporation. In this work, Fe/NC-800 hybrid was facilely prepared by uniform dispersion of in situ formed FeNPs onto NC-800 from carbonization of ZIF-8 at 800 °C for the first time. The optimized Fe/NC-800 catalyst was characterized by TEM, XPS and XRD. Compared with sole FeNPs and NC-800, the Fe/NC-800 catalyst exhibited an enhanced oxidase-like activity that could oxidize the colorless 3,3',5,5'-tetramethylbenzidine (TMB) to the heavy blue without extra oxidants such as H2O2. The possible reason for the enhanced oxidase-like activity of the Fe/NC-800 was discussed on the basis of the experiments of radical scavengers, indicating the importance of superoxide (O2•-) and singlet (1O2) in colorimetric reaction between TMB and Fe/NC-800 hybrid. Furthermore, the oxidase-like activity of Fe/NC-800 was significantly inhibited by dopamine (DA), leading to blue color fading. On this basis, a sensitive and selective colorimetric sensor was fabricated for the quantitative analysis of DA with a linear range of 0.01-40 μM and a low detection limit of 10 nM. The proposed colorimetric method was successfully applied to determine DA in human serum and injection samples, suggesting a promising application in biological analysis.

Abstract:Substantially, the unique enzyme-like bioactivity of nanoparticles is essential for the detection performance promotion of electrochemical sensors. Herein, the gold magnetic particles (Fe3O4@Au) were successfully formed by self-assembly method, its physical and chemical properties are characterized by Fourier transform infrared (FT-IR), Transmission electron microscopy (TEM) and Vibrating sample magnetometer (VSM) techniques. Fe3O4@Au was found to be able to exhibit impressive peroxidase-like activity and follow Michaelis-Menten kinetics with a good affinity towards H2O2 and TMB. A novel highly sensitive and selective enzyme-free enhanced electrochemical sensor based on the peroxidase-like activity of Fe3O4@Au was prepared for the detection of dopamine (DA). Electrochemical cyclic voltammetry (CV) was used to measure dopamine and the electrochemical detection system was optimized. The results showed that the established enzyme-free enhanced electrochemical sensor showed extremely high sensitivity towards dopamine under the optimized conditions, with a good linear relationship between the current response, and the concentration of dopamine in the range of 0.01–1 mmol/L with a detection limit of 0.0109 mg/L (S/N = 3). In addition, the established enzyme-free enhanced electrochemical sensor was successfully applied in the detection of dopamine in pork samples. In view of this, the proposed new enzyme-free electrochemical assay has great potential in the fields of food quality control and security detection.

Abstract:Novel Fe3O4@C@MnO2 composites were successfully synthesized for the first time via an interfacial reaction between magnetic porous carbon and KMnO4, in which the magnetic porous carbon was derived from the pyrolysis of Fe-MIL-88A under N2 atmosphere. Interestingly, the obtained Fe3O4@C@MnO2 composites were found to have triple-enzyme mimetic activity including peroxidase-like, catalase-like, and oxidase-like activity. As a peroxidase mimic, Fe3O4@C@MnO2 composites could catalyze the oxidation of TMB into a blue oxidized product by H2O2. As a catalase mimic, Fe3O4@C@MnO2 could catalyze the decomposition of H2O2 to generate O2 and H2O. As an oxidase mimic, Fe3O4@C@MnO2 could catalyze the direct oxidation of TMB to produce a blue oxidized product without H2O2. Reactive oxygen species measurements revealed that the oxidase-like activity originated from 1O2 and O2-∙and little∙OH generated by the dissolved oxygen, which was catalyzed by the Fe3O4@C@MnO2 in the TMB oxidation reaction. The oxidase-like activity of Fe3O4@C@MnO2 was investigated in detail. Under the optimized conditions, a rapid, sensitive, visual colorimetric method for dopamine detection was developed based on the inhibitory effect of dopamine on the oxidase-like activity. The proposed method allows for dopamine detection with a limit of detection of 0.034 μM and a linear range of 0.125-10 μM. This new colorimetric method was successfully used for the determination of dopamine in human blood samples.
Pt Nanoparticles Anchored on NH2-MIL-101 with Efficient Peroxidase-Like Activity for Colorimetric Detection of Dopamine

Abstract:Dopamine (DA) is an important catecholamine neurotransmitter that plays a highly relevant role in regulating the central nervous system, and abnormal DA content can cause many immune-related diseases. Hence, it is of significance to sensitively and specifically identify DA for clinical medicine. In this work, Pt/NH2-MIL-101 hybrid nanozymes with bimetallic catalytic centers were fabricated by forming coordinate bonds between Pt nanoparticles (Pt NPs) and –NH2 on metal–organic frameworks (MOF). The catalytic activity of Pt/NH2-MIL-101 was increased by 1.5 times via enlarging the exposure of more active sites and improving the activity of the active sites through the strategy of forming bimetallic catalytic centers. In the presence of DA, competing with 3, 3′, 5, 5′-tetramethylbenzidine (TMB) for the generated hydroxyl radicals (•OH), the blue oxidation state TMB (Ox-TMB) is reduced to colorless TMB, showing dramatic color changes. The Pt/NH2-MIL-101-based colorimetric assay enables the sensitive and robust detection of DA molecules with a detection limit of only 0.42 μM and has an observable potential in clinical applications.

Abstract:To reliably monitor dopamine (DA), a common neurotransmitter, a kind of nano CeO 2 /CuO modified carboxylated chitosan nanozyme (CeO 2 /CuO@CC) was constructed under microwave-assisted conditions. The proposed CeO 2 /CuO@CC exhibited superior synergistic- enhancement oxidase-like activity, with high specific surface area and superior stability. It could accelerate the oxidization of colorless 3,3′,5,5′-tetramethylbenzidine (TMB) into blue oxTMB in the air. What's more, the presence of trace DA could exclusively change the enzyme-like activity of CeO 2 /CuO@CC, accompanied with visual color changes from blue to colorless. Under the optimized conditions, there expressed double ratiometric linear relationships between lg( A 652 / A 285 )/lg( A 370/ A 285 ) and c DA , with a large linear range between 6.0 × 10 −9 and 1.0 × 10 −5 M of DA. When applied for visual detection of DA in real samples, the detection limit was low to 2.0 × 10 −9 M with RSD % less than 4.14 %. The mechanisms for enhanced oxidase-mimic activity of CeO 2 /CuO@CC and its selective detection of DA were proposed. This work will offer an insight to design efficient nanozyme for visual detection of bio-analytes in practice.

Abstract:Core-shell nanostructures (NSs) are a type of nanozyme broadly used in various fields due to their significant physicochemical advantages. In the present study, the CoFe 2 O 4 @ZIF-8@ZIF-67 core–shell were fabricated by a consecutive two-step synthesis method, including hydrothermal for CoFe 2 O 4 metal spinel and epitaxial solvothermal growth of ZIFs bilayers on the metal spinel NSs. The core–shell surface was modified using ATP inspired by the active site of the natural peroxidase enzyme. The nanocomposites were characterized by XRD, FT-IR, SEM, EDX, and Dot-mapping techniques. The peroxidase-like activity of the ATP/CoFe 2 O 4 @ZIF-8@ZIF-67 was assessed by a colorimetric method. Modified nanocomposite displayed desirable peroxidase mimetic activity and could enzymatically generate hydroxyl radicals from H 2 O 2 and oxidize TMB. The high catalytic activity of ATP-modified nanocomposite and its ability to oxidize TMB provides a rapid and sensitive platform for the colorimetric detection of dopamine. Dopamine consumes hydroxyl radicals, prevents TMB oxidation, and causes a color change in the detection solution, which the naked eye can recognize. The presented colorimetric test provided an excellent LOD for dopamine (0.4 μM) with a good linear range (0.2–18 μM) and selectivity, which was prosperously applied to dopamine assay in serum blood samples.

Abstract:Urchin-like peroxidase mimics 5,10,15,20-tetrakis (4-carboxyl phenyl) porphyrin–functionalized CuCo2O4 nanospheres (Por-CuCo2O4) has been fabricated as an excellent visual biosensor. X-ray diffractometry (XRD), scanning electron microscopy (SEM), and X-ray photoelectron spectroscopy (XPS) have been employed to characterize the composition, morphologies, and elemental analysis of the as-synthesized Por-CuCo2O4. The catalytic activity of Por-CuCo2O4 was evaluated by the chromogenic substrate 3,3′,5,5′-tetramethylbenzidine (TMB) with the aid of H2O2, which exhibited a visual blue change with an absorption maximum at 652 nm for only 10 s. The peroxidase-like behaviors of Por-CuCo2O4 conformed to the Michaelis-Menten equation. Electrochemistry, radical scavenger, and fluorescence probe experiments verified that electron transfer, •O2− radicals, and holes (h+) are the important factors during the catalytic oxidation of TMB. Based on the inhibition of dopamine (DA) on TMB oxidation, the Por-CuCo2O4-based colorimetric biosensor has been successfully constructed for sensitive determination of DA witha detection limit (LOD) of 0.94 μΜ. In addition, colorimetry was validated to detect DA in serum samples with high sensitivity and good selectivity.

Abstract:As a central neurotransmitter, DA (dopamine) plays a vital part in human metabolism, and its accurate detection is of great significance in disease diagnosis. In this work, we used Cu/Co bimetallic metal-organic frameworks (MOFs) as templates and gold nanoparticles (AuNPs) to construct novel nanocomposite coral-like CuCo2O4@AuNPs with strong peroxidase activity and electrochemical response. The coral-like CuCo2O4@AuNPs showed excellent peroxidase activity, and the Km value was as low as 0.358 mM. In the presence of H2O2, the colorless substrate 3,3',5,5', -tetramethylbenzidine (TMB) can be catalytically oxidized into a blue product. Simultaneously, coral-like CuCo2O4@AuNPs, as an electroactive substance, possess strong electrocatalytic activity, which enhances the electron-transfer rate and promotes excellent current response. In the presence of DA, coral-like CuCo2O4@AuNPs can catalyze the oxidation of DA to dopaquinone, which further enhances the electrochemical signal. In addition, DA captures hydroxyl radicals and inhibits the oxidation of TMB, resulting in an obvious color change (blue turns colorless) and realizing colorimetric detection with the naked eye. On this basis, we successfully established a dual-mode colorimetric/electrochemical sensor using coral-like CuCo2O4@AuNP nanocomposites as a dual-signal probe. Combining colorimetric and electrochemical detection, the sensor achieved a wide linear range (0-1 mM) and a low detection limit (0.07 μM) for DA concentration. It was also successfully used for the detection of DA in human serum and urine with good results. In summary, this work provides an intuitive, economical, sensitive, and promising platform for DA detection.

Abstract:The interaction of FAA or TNF with radiation was studied in the murine KHT sarcoma. When used alone both agents showed a dose- and time-dependent toxicity towards the tumour cells and significantly reduced tumour blood flow within 1 h of treatment. When used in combination with radiation, both TNF and FAA caused an increase in the fraction of hypoxic cells in the KHT tumour. This was assessed by an in vivo/in vitro clonogenic assay and by a comparison with the radioprotection provided by clamping tumours prior to and during irradiation. When TNF was given at a dose of 2.5 X 10(5) U/kg an increase in tumour hypoxia was seen after 30 min. Close to 100% radiobiological hypoxia was reached by 1 h after treatment, lasting for up to 16 h. Doses of TNF below 0.25 X 10(5) U/kg did not induce levels of hypoxia comparable to clamping when administered 3 h prior to irradiation. Similarly, FAA produced a rapid increase in tumour hypoxia: a dose of 200 mg/kg induced close to 100% radiobiological hypoxia when give 1 h prior to irradiation. Complete tumour hypoxia was still apparent 18 h after treatment with FAA. Administered doses of FAA below 100 mg/kg did not produce close to 100% radiobiological hypoxia when administered 3 h prior to irradiation.

Abstract:Dopamine is one of the most important neurotransmitters and plays a crucial role in various neurological, renal, and cardiovascular systems. However, the abnormal levels of dopamine mainly point to Parkinson's, Alzheimer's, cardiovascular diseases, etc. Hydroxyapatite (HAp), owing to its catalytic nature, nanoporous structure, easy synthesis, and biocompatibility, is a promising matrix material. These characteristics make HAp a material of choice for doping metals such as cobalt. The synthesized cobalt-doped hydroxyapatite (Co-HAp) was used as a colorimetric sensing platform for dopamine. The successful synthesis of the platform was confirmed by characterization with FTIR, SEM, EDX, XRD, TGA, etc. The platform demonstrated intrinsic peroxidase-like activity in the presence of H 2 O 2 , resulting in the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB). The proposed sensor detected dopamine in a linear range of 0.9–35 μM, a limit of detection of 0.51 μM, limit of quantification of 1.7 μM, and an R 2 of 0.993. The optimization of the proposed sensor was done with different parameters, such as the amount of mimic enzyme, H 2 O 2 , pH, TMB concentration, and time. The proposed sensor showed the best response at 5 mg of the mimic enzyme, pH 5, 12 mM TMB, and 8 mM H 2 O 2 , with a short response time of only 2 min. The fabricated platform was successfully applied to detect dopamine in physiological solutions.

Abstract:Under visible light irradiation, photoelectrons react with oxygen molecules adsorbed on the surface of CeO2@ZIF‐8/Cu‐CDs nanozymes to form superoxide free radicals and hydroxyl free radicals, which catalyse the oxidation of dopamine to produce red compounds. Therefore, a new method for the rapid detection of dopamine with high sensitivity and high selectivity was established. This study reports a sensitive and selective colorimetric approach for the analysis of dopamine (DA) based on CeO2@ZIF‐8/Cu‐CDs laccase‐like nanozymes activity. The CeO2@ZIF‐8/Cu‐CDs was synthesized using cerium oxide (CeO2) and copper‐doped carbon dots (Cu‐CDs) with 2‐methylimidazole by a facilely hydrothermal approach. The CeO2@ZIF‐8/Cu‐CDs exhibited excellent laccase‐like nanozymes activity and can oxidize the colorless substrate (DA) to red product with 4‐aminoantipyrine as the chromogenic agent. The Michaelis–Menten constant (Km) and the maximal velocity (Vmax) of CeO2@ZIF‐8/Cu‐CDs are 0.20 mM and 1.48 μM/min, respectively. The detection method has a linear range of 0.05–7.5 μg/mL and a detection limit as low as 8.5 ng/mL with good reproducibility. The developed colorimetric sensor was applied to rapid and precise quantitative evaluation of DA levels in serum and urine samples. This study presents a new approach for detecting biological molecules by utilizing the controlled regulation of nanozymes' laccase‐like activity.

Abstract:Neurotransmitters are essential chemical messengers required for proper brain function, and any changes in their concentrations can lead to neuronal diseases. Therefore, sensitive and selective detection is crucial. This study presents a fast and simple colorimetric method for dopamine detection using three reagent solutions: AgNP and MPA, Ag/Au nanocomposite, and mercaptophenylacetic acid. TEM images showed a narrow distribution of Ag and Au nanoparticles with average sizes of 20 nm and 13 nm, respectively, with gold nanoparticles bound to the edges of silver nanoparticles. A paper-based biosensor was created using manual wax printing for the colorimetric detection of dopamine. Visual detection onsite showed color changes with both the silver nanoparticles and mercaptophenylacetic acid mixture and the silver–gold nanoparticle composite. Electrochemical detection using a glassy carbon electrode modified with 8 mM mercaptophenylacetic acid demonstrated high selectivity and sensitivity towards dopamine, with a peak in the range of 0.7–0.9 V. Interferences were minimized, ensuring high sensitivity and selective detection of dopamine.

The outcome of staged management of complicated diverticular disease of the sigmoid colon.

Abstract:In a retrospective study, 83 consecutive patients with indication for a staged treatment of complicated diverticular disease were reviewed. There was a considerable overall morbidity rate following construction of the colostomy, colonic resection, and colostomy closure. However, only 58% underwent a resection of the diseased segment, and even fewer (46%) obtained an eventual restoration of the colon continuity. This was primarily due to an unacceptable high mortality rate when patients with general peritonitis were managed by a proximal colostomy and drainage only. Subsequently, advanced age and associated major illness in patients with a planned three-stage management were the most important factors, determining the deficient completion of the staged treatment. Due to the permanence of many transverse colostomies but also to the fact that serious anastomotic complications, including fatal consequences, occurred despite a defunctioning colostomy, one should re-evaluate the need for a proximal colostomy in patients at high risk, considering resection and an end sigmoid colostomy which is the superior permanent stoma if reconstruction fails to come.

Herein, Co3O4 nanoplates with a uniform nanostructure were prepared by the thermolysis of cobalt hydroxide at different temperatures, and the influence of the annealing temperature on the performance of the mimetic enzyme also was reported for the first time. The results demonstrated that Co3O4 nanoplates obtained at an annealing temperature of 200 °C possessed strong oxidase activity and efficiently catalyzed the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) without the addition of hydrogen peroxide to generate the blue color product ox-TMB. Once the annealing temperature was increased to 500 °C and 800 °C, the oxidase activity of Co3O4 decreased rapidly, and was even inactivated. This might be attributed to the relatively large specific surface area of Co3O4 annealed at 200 °C. Besides this, based on the TMB-Co3O4 nanoplate system, a colorimetric analysis method was developed to detect dopamine with a limit of 0.82 μmol/L in a linear range from 1.6 μmol/L to 20 μmol/L.

Abstract:The prevalence of psychiatric disorders diagnosed according to DSM-III in adolescents in the general population is not known. The authors address this issue in a community sample of 150 adolescents 14-16 years of age. Structured interviews as well as other instruments were used to collect data. Twenty-eight (18.7%) of the 150 adolescents were identified as having a psychiatric disorder. These 28 adolescents viewed their parents as less caring, had lower self-esteem, and resolved their conflicts through verbal aggression and physical violence more often than did the adolescents who did not have a psychiatric disorder. The authors make recommendations regarding the use of structured interviews in future research.
Fluorescence detection of dopamine based on the peroxidase-like activity of Fe3O4-MWCNTs@Hemin

Abstract:A novel Fe3O4-MWCNTs@Hemin nanocomposite was synthesized using hemin and Fe3O4 with multi-walled carbon nanotubes (MWCNTs) by one-step hydrothermal methods. The as-prepared Fe3O4-MWCNTs@Hemin nanocomposites exhibited excellent peroxidase-like activities in the activation of H2O2. The mechanisms, kinetics, and catalytic performances of Fe3O4-MWCNTs@Hemin were systematically studied. Fe3O4-MWCNTs@Hemin can oxidize dopamine (DA) to dopaquinone in the presence of H2O2, and the intermediate products dopaquinone can further react with β-naphthol to generate a highly fluorescent derivative at 415 nm excitation wavelength. Therefore, an innovative fluorescence platform for the detection of DA was developed. The fluorescence intensity increased linearly with DA concentration in the range 0.33 to 107 μM, with a low detection limit of 0.14 μM. Due to the excellent activity, substrate universality, fast response, high selectivity, and sensitivity of Fe3O4-MWCNTs@Hemin, the proposed fluorescence method was used to analyze complex biological blood samples with a satisfactory result. It demonstrated the significant potential for developing effective and dependable fluorescent analytical platforms for preserving human health.

Abstract:Prussian blue (PB) is a typical peroxidase mimic with simple preparation, low cost and high eco-friendliness, but it still has drawbacks of poor stability (e.g., decomposition in aqueous dispersions) and intrinsic optical interference (e.g., high extinction coefficient in a wide wavelength range) in colorimetric assays. Herein, we used nitrocellulose (NC) membranes as synthesis hosts of PB nanoparticles (NPs) to develop a new type of three-dimensional (3D) porous nanozyme pads. By means of an in-situ synthesis routine, PB NPs were uniformly grown on surfaces of the fiber scaffolds with desirable stability, which also avoided the signal interference from PB NPs by easily handling the pads in a quantitative solid state. The pads showed significant peroxidase-mimicking activity toward the oxidation of 3,3',5,5'-tetramethylbenzidine (TMB) with the output of colorimetric signals. Based on the reduction of oxidized TMB (oxTMB) by dopamine (DA), the pads were exploited for simple and quantitative colorimetric detection of DA with a limit of detection (LOD) of 0.59 μM and a satisfactory accuracy for analysis of real human urine samples. This is the first attempt of exploiting NC membranes as the synthesis hosts to develop the nanozyme pads, which solves the above drawbacks of traditional PB-based peroxidase mimics and has promise for various colorimetric bioanalysis, given the structural benefits of NC membranes and their broad applications in biosensors.

Abstract:The Fe2+/Fe3+ redox couple is effective for voltammetric detection of trace dopamine (DA). However, achieving adequate concentrations with high electroactive surface area (ECSA), DA affinity, and fast interfacial charge transfer is challenging. Consequently, most reported Fe‐based sensors have a high nanomolar range detection limit (LOD). Herein, we address these limitations by manipulating the phase and morphology of FeOOH/Fe2O3 heterojunctions anchored on sp2‐carbon. FeOOH/Fe2O3 is synthesized by variable temperature aging of unique Fe5H9O15/Fe2O3@sp2‐carbon colloidal nanoparticles, which form via chelation between biomass‐derived carbon nanodots (CNDs) and Fe2+ ions. At 27°C and 120°C, Fe5H9O15/Fe2O3@sp2‐carbon transforms into β‐FeOOH/Fe2O3 nanoparticles and α‐FeOOH/Fe2O3 nanosheet, respectively. The β‐FeOOH/Fe2O3 interface exhibits higher eg orbital electron occupancy than α‐FeOOH/Fe2O3, thereby facilitating oxygen adsorption and the generation of Fe2+/Fe3+ sites near the polarization potential of DA. This facilitates interfacial electron transfer between Fe3+ and DA. Moreover, its nanoparticle morphology enhances ECSA and DA adsorption compared to α‐FeOOH/Fe2O3 nanosheets. With a LOD of ~3.11 nM, β‐FeOOH/Fe2O3 surpasses the lower threshold in humans (~10 nM) and matches noble‐metal sensors. Furthermore, it exhibits selective detection of DA over 10 biochemicals in urine. Therefore, the β‐FeOOH/Fe2O3@sp2‐C platform holds promise as a low‐cost, easy‐to‐synthesize, and practical voltammetric DA monitor.
