Abstract:Mass spectrometry (MS) based methodology offers simple, fast and sensitive diagnosis. While it has become the predominate approach in biomolecular analysis, it has not been suitable for analyzing nucleic acid due to its low ionization efficiency. We report herein on a DNA assay based on monolayer-barcoded nanoparticles that were encoded with reporter mass molecules, which act as surrogate molecules for the matrix-assisted laser desorption/ionization time-of-flight MS (MALDI-TOF MS) identification of target DNA through mass spectrometry in combination with magnetic microprobes. This assay demonstrated high MS sensitivity, with the ability to detect target DNA at femtomolar (10−15M) levels. This inaugural effort using combined techniques is significant because it showed an extraordinary analytical capability for differentiating the single nucleotide polymorphism (SNP), which comprises the most abundant source of genetic variation in the human genome. We also report herein the feasibility of MS detection of two target DNAs that have the same mass but different nucleotide base composition, which classic MS methodology is inherently unable to differentiate.

Abstract:using the synthesized nickel particle/graphene (Ni/Gr) composite as a new matrix and adsorbent, a matrix-assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF MS) platform was constructed for the efficient analysis of various drugs, amino acids and other small biological molecules. Compared with the traditional matrix of 2, 5-dihydroxyphenyl acid (DHB) and a series of comparison materials (Gr, Ni-1/gr and Ni-5/gr), Ni-2/gr as MALDI matrix has the advantages of low background noise, high ionic strength, high signal-to-noise ratio and wide linear range (0.01 ~ 50 μmol/L, 0.2 ~ 50 μmol/l and 0.05 ~ 60 μmol/l) when analyzing the molecules of resveratrol, 6-gingerol and rutin in the positive ion mode. The detection limits (LODs) of resveratrol, 6-gingerol and rutin were respectively 0.0038, 0.09 and 0.02 μmol/L. Ni-2/gr complex has the characteristics of high specific surface area, rich mesoporous structure, a large number of sp2 structures, strong UV absorption and high saturation magnetization value (MS). It can be used as an adsorbent to magnetically enrich phenylalanine, tryptophan and tyrosine, and can also be used to magnetically enrich low concentration tryptophan in mouse serum samples.

Abstract:Matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI MSI) is widely applied in mapping macrobiomolecules in tissues, but it is still limited in profiling low-molecular-weight (MW) compounds (typically metabolites) due to ion interference and suppression by organic matrices. Here, we present a versatile "top-down" strategy for rational engineering of carbon material-based matrices, by which heteroatom-doped graphene quantum dots (HGQDs) were manufactured for LDI MS detection and imaging of small biomolecules. The HGQDs derived from parent materials inherited the π-conjugated networks and doping sites for promoting energy transfer and negative ion generation, while their extremely small size guaranteed the matrix uniformity and signal reproducibility in LDI MSI. Compared to other HGQDs, nitrogen-doped graphene quantum dots (NGQDs) exhibited superior capability of assisting LDI of various small molecules, including amino acids, fatty acids, saccharides, small peptides, nucleobases, anticancer drugs, and bisphenol pollutants. Density functional theory simulations also corroborated that the LDI efficiency was markedly raised by the proton-capturing pyridinic nitrogen species and compromised by the electron-deficient boron dopants. NGQDs-assisted LDI MS further enabled label-free investigation on enzyme kinetics using an ordinary short peptide as the substrate. Moreover, due to the high salt tolerance and signal reproducibility, the proposed negative-ion NGQDs-assisted LDI MSI was able to reveal the abundance and distribution of low-MW species in rat brain tissue and achieved the imaging of low-MW lipids in coronally sectioned rat brains subjected to traumatic brain injury. Our work offers a new route for customizing nanomaterial matrices toward LDI MSI of small biomolecules in biomedical and pathological research.

Abstract:Zvi is the last name listed in my e-mail nickname directory. Whenever I alphabetize stored mail, Zvi’s messages appear at the end, with his most recent one appearing last. So, as it happens, the last message in my alphabetized list is a short note from Zvi’s secretary informing me that Zvi was too sick to come into his office and that she would take my message to his home. I regularly clean up my e-mail account. I erase old solicitations, unfunny jokes, and complaints from students about grades. Yet I cannot bring myself to erase this note from Zvi’s secretary, even though Zvi died over a year ago. The personal and professional have crossed in such an unexpected way. The personal observation is mildly embarrassing. E-mail has integrated itself into my life—so much so that it can become an object of sentimentality. The professional observation is of more lasting significance. E-mail has become the seed for significant changes in society. Zvi would have said that the spread of e-mail resembled the spread of agricultural improvements. This resemblance is not self-evident.
Development of N,S-doped carbon dots as a novel matrix for the analysis of small molecules by negative ion MALDI-TOF MS

Abstract:Recently, MALDI-TOF MS has emerged as a popular analytical tool with the advantages of easy sample preparation and good salt tolerance, as well as fast and high throughput data acquisition ability. However, for the existing matrices, they were limited in detecting low-molecular-weight analytes due to the matrix-related peaks and low reproducibility. Therefore, the development of new matrices is highly desirable. Here, a new matrix N, S-CDs, for the first time, was synthesized and used for small molecule analysis. Comprehensive assessments of N, S-CDs were conducted including structural characterization, comparisons with conventional CHCA, 9-AA and N-CDs matrices in detecting amino acids, peptides, nucleosides and anticancer drugs in different ion modes. The results showed that N, S-CDs could be a sensational matrix which had good signal intensity of the analytes without matrix-related interference, especially in negative ion mode. What's more, the N, S-CDs matrix displayed good salt tolerance and signal reproducibility. In addition, the N, S-CDs matrix was further applied for serum detection, quantitative determination of endogenous glucose, and the classification of different subtypes of breast cancer cell lines. The detection of various endogenous metabolites in serum analysis and significantly distinguishing between two cell lines indicated the ability of our new matrix to be applied in complex biological samples. Furthermore, the ionization mechanism of the N, S-CDs as a matrix was also discussed comparing with N-CDs in the transformation of structure and composition. This work creates a new application branch for N, S-CDs and provides an improved MALDI matrix for small molecule analysis.

Abstract:Matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) using conventional organic matrices for detection of small molecules has some limitations, such as heterogeneous analyte/matrix co-crystals, as well as interference of matrices in the low-molecular-weight range. In this work, a zeolite framework nanomaterial, TS-1, was applied as a MALDI matrix for the analysis of small molecules by MALDI-MS for the first time. To improve the signal intensity and reproducibility, TS-1 was modified with polydopamine (). Using as a matrix, organic substances in the low-molecular-weight region such as amino acids, nucleosides, peptides, oligosaccharides, and fatty acids can be detected by MALDI-MS in positive ion mode. Compared with traditional organic matrices like 2,5-dihydroxybenzoic acid (2,5-DHB) and α-cyano-4-hydroxycinnamic acid (CHCA), has the advantages including the formation of uniform sample spots, small background interference at low molecular weight, and better salt tolerance. Furthermore, this matrix was employed for the analysis of endogenous glucose in urine samples, and the level of glucose was quantified with a linear range of 0–10 mM (R2 > 0.98). The results demonstrated that has the potential to be used as an effective MALDI matrix for the analysis of small molecules in biological samples with excellent reproducibility and moderate sensitivity.This article has not yet been cited by other publications.

N-Doped Graphene: an Alternative Carbon-Based Matrix for Highly Efficient Detection of Small Molecules by Negative Ion Maldi-Tof Ms

Abstract:Gas-phase N-doped graphene (gNG) was synthesized by a modified thermal annealing method using gaseous melamine as nitrogen source and then for the first time applied as a matrix in negative ion matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) for small molecule analysis. Unlike the complicated adducts produced in positive ion mode, MS spectra obtained on gNG matrix in negative ion mode was only featured by deprotonated molecule ion peaks without matrix interference. By the gNG assisted desorption/ionization (D/I) process, some applications were carried out on a wide range of low-molecular weight (MW) analytes including amino acids, fatty acids, peptides, anabolic androgenic steroids as well as anticancer drugs, with an extraordinary laser desorption/ionization (LDI) efficiency over traditional α-cyano-4-hydroxycinnamic acid (CHCA) and other carbon-based materials in the negative ion detection mode. By comparison of a series of graphene-based matrixes, two main factors of matrix gNG were unveiled to play a decisive role in assisting negative ion D/I process: a well-ordered π-conjugated system for laser absorption and energy transfer; pyridinic-doped nitrogen species functioning as deprotonation sites for proton capture on negative ionization. The good salt tolerance and high sensitivity allowed further therapeutic monitoring of anticancer drug nilotinib in the spiked human serum, a real case of biology. Signal response was definitely obtained between 1 mM and 1 μM, meeting the demand of assessing drug level in the patient serum. This work creates a new application branch for nitrogen-doped graphene and provides an alternative solution for small molecule analysis.
Super-assembled sandwich-like Au@MSN@Ag nanomatrices for high-throughput and efficient detection of small biomolecules

Abstract:Small biomolecules (m/z < 500) are the material basis of organisms and participate in life activities, but their comprehensive and accurate detection in complex samples remains a challenge. Matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) is a powerful detection tool for molecular analysis with high throughput. The development of a new matrix is essential to improve the efficiency of the MALDI-MS for molecular compound detection. In this work, the sandwich-like gold nanoparticles@mesoporous silica nanocomposite@silver nanoparticles (Au@MSN@Ag) nanospheres were prepared by layer-by-layer super-assembly strategy, and can be used as a novel matrix for the quantitative detection and enrichment of small biomolecules by LDI-MS. The sandwich-like nanospheres form a unique plasma resonant cavity that effectively absorbs the laser energy, while the homogeneous mesoporous structure of MSN can lock the analyte, which is essential for efficient LDI of small molecules. Compared to traditional matrices, Au@MSN@Ag shows the advantages of low background, wide application range, high sensitivity, super high salt and protein tolerance, and good stability. For example, the detection limit of glucose was as low as 5 fmol, and showed a good linear relationship in the range of 1–750 µg/mL. Au@MSN@Ag assisted LDI-MS allows the enrichment and detection of small molecules in traditional Chinese medicine (TCM) without derivatization and purification, classification of herbs using the accurate quantitative results oligosaccharides, and identification of gelatin by amino acid content. This research could help in designing more efficient nanostructure matrices and further explored the application of LDI-MS.

Abstract:This paper reports a facile synthesis of molybdenum disulfide nanosheets/silver nanoparticles (MoS2/Ag) hybrid and its use as an effective matrix in negative ion matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). The nanohybrid exerts a strong synergistic effect, leading to high performance detection of small molecule analytes including amino acids, peptides, fatty acids and drugs. The enhancement of laser desorption/ionization (LDI) efficiency is largely attributed to the high surface roughness and large surface area for analyte adsorption, better dispersibility, increased thermal conductivity and enhanced UV energy absorption as compared to pure MoS2. Moreover, both Ag nanoparticles and the edge of the MoS2 layers function as deprotonation sites for proton capture, facilitating the charging process in negative ion mode and promoting formation of negative ions. As a result, the MoS2/Ag nanohybrid proves to be a highly attractive matrix in MALDI-TOF MS, with desired features such as high desorption/ionization efficiency, low fragmentation interference, high salt tolerance, and no sweet-spots for mass signal. These characteristic properties allowed for simultaneous analysis of eight different drugs and quantification of acetylsalicylic acid in the spiked human serum. This work demonstrates for the first time the fabrication and application of a novel MoS2/Ag hybrid, and provides a new platform for use in the rapid and high throughput analysis of small molecules by mass spectrometry.
Graphene As A Novel Matrix for the Analysis of Small Molecules by Maldi-Tof Ms

Abstract:Graphene was utilized for the first time as a matrix for the analysis of low molecular weight compounds using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). Polar compounds including amino acids, polyamines, anticancer drugs, and nucleosides could be successfully analyzed. Additionally, nonpolar compounds including steroids could be detected with high resolution and sensitivity. Compared with a conventional matrix, graphene exhibited a high desorption/ionization efficiency for nonpolar compounds. The graphene matrix functions as a substrate to trap analytes, and it transfers energy to the analytes upon laser irradiation, which allows for the analytes to be readily desorbed/ionized and interference of intrinsic matrix ions to be eliminated. The use of graphene as a matrix avoided the fragmentation of analytes and provided good reproducibility and a high salt tolerance, underscoring the potential application of graphene as a matrix for MALDI MS analysis of practical samples in complex sample matrixes. We also demonstrated that the use of graphene as an adsorbent for the solid-phase extraction of squalene could improve greatly the detection limit. This work not only opens a new field for applications of graphene, but also offers a new technique for high-speed analysis of low molecular weight compounds in areas such as metabolism research and natural product characterization.
One-step synthesis of N, B-codoped carbon nanofiber as a novel matrix for high-throughput and efficient laser desorption/ionization mass spectrometry analysis

Abstract:It is of significance to analyze various small molecules with high efficiency using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). In this work, we synthesized N and B dual-doped 3D carbon (N, B-C) nanofiber as a promising MALDI matrix through a facile impregnation and thermal pyrolysis process. Compared to the control C nanofiber, the N and B codoping could enhance surface area, functional groups, defective and active sites, which was conducive to adsorb the laser and enrich the desorption/ionization of analytes. For the first time, the N, B-C nanofiber was utilized as a novel matrix for MALDI-TOF MS analysis of various small molecules both in the positive-ion and negative-ion modes. This matrix showed high signal to noise (S/N) ratio, excellent salt-tolerance, and homogeneous ion distribution for the analysis of amino acids, saccharides, polymers and surfactant, which was obviously superior to the traditional α-cyano-4-hydroxycinnamic acid (CHCA) and the control C nanofiber matrix. Noticeably, the wide linear ranges (0.05–5 mM) of glucose was achieved by MALDI-TOF MS with N, B-C nanofiber matrix. Meanwhile, the MALDI-TOF MS imaging could observe the change of different content glucose based on the ion peak of [M+Na]+. Furthermore, N, B-C nanofiber-assisted LDI-TOF MS has also been applied to identify and analyze the saccharides in the natural honey.

Abstract:Within the past two decades, the escalation of research output in nanotechnology fields has boosted the development of novel nanoparticles and nanostructured substrates for use as matrices in surface assisted laser desorption/ionization mass spectrometry (SALDI-MS). The application of nanomaterials as matrices, rather than organic matrices, offers remarkable characteristics that allow the analysis of small molecules with fewer matrix interfering peaks, and share higher detection sensitivity, specificity, and reproducibility. The technological advancement of SALDI-MS has in turn, propelled the application of the analytical technique in the field of biomedical analysis. In this review, the properties and fabrication methods of nanostructured substrates in SALDI-MS such as metallic-, carbon-, and silicon-based nanostructures, quantum dots, metal-organic frameworks, and covalent-organic frameworks are described. Additionally, the latest progress (most within 5 years) of biomedical applications in small molecule, large biomolecule, and MS imaging analysis including metabolite profiling, drug monitoring, bacteria identification, disease diagnosis, and therapeutic evaluation are demonstrated. Key parameters that govern nanomaterial's SALDI efficiency in biomolecule analysis are also discussed. Finally, perspectives of the future development are given to provide a better advancement and promote practical application in clinical MS.

A single atom cobalt anchored MXene bifunctional platform for rapid, label-free and high-throughput biomarker analysis and tissue imaging

Abstract:Few of single-atom materials have been served as platform to analyze small molecules for surface assisted laser desorption/ionization mass spectrometry (SALDI-MS). Herein, a novel single Co atom-anchored MXene (Co–N–Ti 3 C 2 ) is prepared to achieve enhanced SALDI-MS and mass spectrometry imaging (MSI) performance for the first time. The Co–N–Ti 3 C 2 films were prepared by a simple in situ self-assembly strategy to generate an efficient SALDI-MS platform. Compared to typical inorganic/organic matrices, Co–N–Ti 3 C 2 films exhibit superior performance in small molecules detection with ultra-high sensitivity (LOD at amol level), excellent repeatability (CV <4%), clean background and wide analyte coverage, enabling accurate quantitative analysis of various low-concentration metabolites from 1 μL biofluid in seconds. Its usage efficiently enhanced SALDI-MS detection of various small-molecule biomarkers such as amino acids, succinic acid , itaconic acid, arachidonic acid , citrulline, prostaglandin E2 , creatinine, uric acid , glutamine, D-mannose, cholesterol and inositol in positive ion mode. The blood glucose level in humans was successfully determined from a linearity concentration range (0.25–10 mM). Notably, the Co–N–Ti 3 C 2 assisted SALDI-MSI enables study the spatial distribution of small molecules covering the range central to metabolomics at a high resolution on a tissue section. Furthermore, Co–N–Ti 3 C 2 platform revealed a specific peak profile that distinguishes osteoarthritis (OA) from rheumatoid arthritis (RA) tissue. Density functional theory theoretical investigation revealed that single Co atoms anchored on Ti 3 C 2 could highly enhanced the ionization ability of metabolites, resulting in high-sensitivity and heterogeneous metabolome coverage.

Abstract:Matrix assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) is a novel soft ionization bio-mass spectrometry technology emerging in the 1980s, which can realize rapid detection of non-volatile, highly polar, and thermally unstable macromolecules. However, the analysis of small molecular compounds has been a major problem for MALDI-TOF MS all the time. In the MALDI analysis process based on traditional matrices, large numbers of interference peaks in the low molecular weight area and "sweet spots" phenomenon are produced, so the detection method needs to be further optimized. The promotion of matrix means the improvement of MALDI performance. In recent years, many new nanomaterial-based matrices have been successfully applied to the analysis of small molecular compounds, which makes MALDI applicable to a wider range of detection and useful in more fields such as pharmacy and environmental science. In this paper, the newly developed MALDI matrix categories in recent years are reviewed initially. Meanwhile, the potential applications, advantages and disadvantages of various matrices are analyzed. Finally, the future development prospects of nanomaterial-based matrices are also prospected.

A series of glycosylated matrices including 3-glycosylaminoquinoline (3-GAQ), 6-glycosylaminoquinoline (6-GAQ), and 1-amino-5-glycosylaminoquinoline (GDAN) were synthesized by connecting glucose with the existing amine matrices. Compared with their parent matrices and the existing matrix (1,5-diaminonaphathelene, 1,5-DAN), the glycosylated matrices exhibited remarkably-improved sensitivity, higher signal reproducibility (RSD < 9%) in detecting metabolites, demonstrating the effectiveness of the glycosylation strategy. Among them, 6-GAQ exhibited the best performance. Using 6-GAQ, the detection limit of citric acid reached the low fmol range, and the calibration curve of citric acid had ideal linearity (R2 > 0.99), proving that 6-GAQ was capable of accurate quantitative analysis of metabolites. Furthermore, 6-GAQ was used for the imaging of metabolites in the mouse kidney section, showing higher sensitivity and lower background noise than the commonly-used matrices, 9-aminoquinoline (9-AA), and 1,5-DAN. More importantly, 6-GAQ can selectively detect the hydrophilic metabolites, especially the hydrophilic lipids in the mouse kidney. Overall, 6-GAQ is an ideal matrix potentially applied in the imaging and quantitative analysis of hydrophilic small molecules in complex samples.
Efficient Analysis of Small Molecules via Laser Desorption/Ionization Time-of-Flight Mass Spectrometry (LDI–TOF MS) Using Gold Nanoshells with Nanogaps

Abstract:Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI–TOF MS) is a commonly used technique for analyzing large biomolecules. However, the utilization of organic matrices limits the small-molecule analysis because of the interferences in the low-mass region and the reproducibility issues. To overcome these limitations, a surface-assisted laser desorption/ionization (SALDI), which utilizes nanostructured metallic surfaces, has been developed. Herein, a novel approach for SALDI–MS was proposed using silica@gold core–shell hybrid materials with a nanogap-rich shell (SiO2@Au NGS), which is an emerging material due to its excellent heat-generating capabilities. The gold shell thickness was controlled by adjusting the concentration of gold precursor for the growth of gold nanoparticles. SALDI-MS measurements were performed on a layer formed by drop-casting a mixture of SiO2@Au NGS and analytes. At the optimized process, the gold shell thickness was observed to be 17.2 nm, which showed the highest absorbance. Based on the enhanced SALDI capability, SiO2@Au NGS was utilized to detect various small molecules, including amino acids, sugars, and flavonoids, and the ionization softness was confirmed with a survival yield upon fragmentation. The limits of detection, reproducibility, and salt tolerance of SiO2@Au NGS demonstrate its potential as an effective and reliable SALDI material for small-molecule analyses.

Abstract:Application of matrix-assisted laser-desorption/ionization mass spectrometry (MALDI MS) to analyze small molecules have some limitations, due to the inhomogeneous analyte/matrix co-crystallization and interference of matrix-related peaks in low m/z region. In this work, carbon dots (CDs) were for the first time applied as a binary matrix with 9-Aminoacridine (9AA) in MALDI MS for small molecules analysis. By 9AA/CDs assisted desorption/ionization (D/I) process, a wide range of small molecules, including nucleosides, amino acids, oligosaccharides, peptides, and anticancer drugs with a higher sensitivity were demonstrated in the positive ion mode. A detection limit down to 5 fmol was achieved for cytidine. 9AA/CDs matrix also exhibited excellent reproducibility compared with 9AA matrix. Moreover, by exploring the ionization mechanism of the matrix, the influence factors might be attributed to the four parts: (1) the strong UV absorption of 9AA/CDs due to their π-conjugated network; (2) the carboxyl groups modified on the CDs surface act as protonation sites for proton transfer in positive ion mode; (3) the thin layer crystal of 9AA/CDs could reach a high surface temperature more easily and lower transfer energy for LDI MS; (4) CDs could serve as a matrix additive to suppress 9AA ionization. Furthermore, this matrix was allowed for the analysis of glucose as well as nucleosides in human urine, and the level of cytidine was quantified with a linear range of 0.05–5 mM (R2 > 0.99). Therefore, the 9AA/CDs matrix was proven to be an effective MALDI matrix for the analysis of small molecules with improved sensitivity and reproducibility. This work provides an alternative solution for small molecules detection that can be further used in complex samples analysis.Graphical Abstractᅟ

Abstract:Crystals of the Escherichia coli replication terminator protein (Tus) complexed with its binding site DNA were obtained by a microdialysis method using PEG 4000. They belong to the tetragonal space group P41212 or P43212 with the unit cell parameter: a = 68.1 Å, c = 230.7 Å and contain one protein‐DNA complex in an asymmetric unit. The native data set has been collected to 2.7 Å resolution.

Abstract:Graphdiyne (GD) is a new kind of carbon nanomaterial which has carbon–carbon triple bonds to form a layered structure. Here, we report the application of GD as the matrix for small molecule analysis in laser desorption ionization mass spectrometry (LDI MS). The GD matrix displayed two advantages: little background in the low mass range and good molecular ion signal in negative ion mode for many small molecules, e.g., fatty acids, amino acids, peptides, and drugs can be obtained in negative ion mode. By comparing the signal intensity of tetraphenylborate and juglone with and without GD existing, it was found that GD can enhance both of the desorption efficiency and ionization efficiency in LDI. Composite PVK/SLGO As Matrix for MALDI-TOF MS Detection of Small Molecules in Dual-Ion Mode

Abstract:Currently, most matrices developed for matrix-assisted laser desorption/ionization mass spectrometry (MALDI-TOF MS) for small-molecule detection are only suitable for the positive or negative ion mode and not the dual-ion mode, except for carbon-based nanomaterials. The lone-pair electrons on the N atom in poly n-vinylcarbazole (PVK) can serve as a Lewis base with strong electron-donation effects, which is favorable for negative ion mode detection. The surface of single-layer graphene oxide (SLGO) contains many oxygen atoms in carboxyl and hydroxyl groups that act as Lewis acids and thereby provides favorable protonation sites for positive ion mode detection. In this study, composite PVK/SLGO was prepared by combining the advantages of amorphous PVK and SLGO. PVK/SLGO was tested as a novel matrix for positive- and negative-ion-mode MALDI-TOF MS for the analysis of amino acids, nucleic acid bases, environmental endocrine disruptors, antibiotics, and various small molecules. PVK/SLGO was compared with PVK, SLGO, and commercially available matrices of 9-aminoacridine (9-AA) and α-cyano-4-hydroxycinnamic acid (CHCA). The PVK/SLGO matrix was demonstrated to be suitable for the positive and negative ion modes, exhibiting high signal intensity and detection sensitivity without background interference. The limits of detection of the aforementioned molecules ranged from 0.1 to 0.0001 and 0.01 to 0.0001 mg/mL in the positive and negative ion modes, respectively. The quantitative determination of enrofloxacin in milk was realized using an internal standard method with a linear range of 0.0001-0.1 mg/mL (R 2 = 0.9991). Furthermore, the PVK/SLGO matrix exhibited high salt tolerance (up to 1000 mmol/L) and stability over 28 consecutive days. Studies regarding its ionization mechanism revealed that the good performance originates from the combined materials acting synergistically. This study provides a foundation for developing bimodal composite matrices and further expands the scope of PVK/SLGO applications.
