Abstract:CBT cell line developed from a warm-water fish Colossoma Brachypomum,was used to evaluate the effects of cold stress on fish cells. Cell viability was measured by Thiazolyl blue (MTT) method and cellular ultrastructure was examined under transmission electron microscope. Externalization of phosphatidylserine(PS) and mitochondrial membrane potential(MMP) were monitored with confocal laser scanning microscope. Results showed that cell viability and MMP decreased in a time-dependent manner, and the early events of apoptosis such as chromatin condensation, margination and PS externalization occurred, when the cells were exposed to 10 degrees C. It suggested that low temperature (10 degrees C) could induce apoptosis in Colossoma Brachypomum CBT cells.

Abstract:Temperature stress affect a stress response of aquatic animals including physiological metabolism, immune defense and other metabolic processes, damaged their immune response even cause dead. In this study, the physiological and histological changes of Procambarus clarkii under the high temperature (30 degrees C) and the low temperature (10 degrees C) stress were investigated. With the increase of temperature, the pyruvate kinase activity in the hepatopancreas and gill tissues of crayfish was gradually increased, and the fatty acid synthase activity at 10 degrees C and 30 degrees C were significantly lower than the control group of 25 degrees C. Furthermore, lysozyme, catalase and superoxide dismutase in 10 degrees C and 30 degrees C were significantly lower than that of 25 degrees C. ELISA revealed that HSP90 activities was enhanced with the increasing temperature in hepatopancreas after 36h of treatment. HE sections showed that high temperature stress or hypothermia caused deformation of R cells (storage cells), rupture of B cells (secretory cells), and partial rupture of the basement membrane (BM) in hepatopancreas, as well as the deformation of epithelial cells and respiratory epithelial cells of BM in gill tissues. While the temperature stress (30 degrees C and 10 degrees C treatment) caused the deformation of epithelial cells and respiratory epithelial cells, making rupture of the BM, comparing with the group at 25 degrees C. The study provides the data about the response of crayfish to temperature stress at the molecular level, and benefit to develop the strategy to defend the damage in the crayfish aquaculture caused by the temperature neither in the different regions nor in the different season.
FT-IR Study of Chilling Injury of Fishes' Cells

Abstract:CBT cells from colossoma brachypomum and CP cells from eurythermic grass carp were induced with intimidated cold temperature(10℃ and 4℃ respectively) for 0-8 days.Using FT-IR technique,relative contents of the cells were determined,including DNA,RNA,protein,phosphates and glucose.In this way,the dynamic information and cell structure in the process of chilling injury can be well obtained.It was found that,for CBT cells,during the 1st-3rd days,the ratios of DNA/amide II,RNA/amide II,glucose/phosphate and glucose/phospholipids increased;in the 4th day,the four ratios decreased,increased during the 5th-7th days and decreased at the 8th day.The ratio of DNA/RNA decreased in the 1st-3rd days,increased at the 4th day,decreased in the 5th-7th days and increased at the 8th day.For CP cells,the ratios of DNA/amide II and RNA/amide II did not vary during 1st-3rd days,decreased at the 4th day and increased during the 5th-8th days.The ratios of glucose/phosphate and glucose/phospholipids increased steadily during the 0-2nd days,decreased during the 3rd-7th days,and increased at the 8th day.For RNA/DNA ratio,there are maximum values at the 1st,4th and 6th days,and minimum values at the 2nd and 8th days.For both kinds of cells,there are good linear relationships between the ratios of RNA/DNA and glucose/phosphate.The variation of those biochemical components is related to the apoptosis process induced by cold temperatures.
Effects Of Both Cold And Heat Stress On The Liver Of The Giant Spiny Frog (Quasipaa Spinosa): Stress Response And Histological Changes

Abstract:Ambient temperature-associated stress can affect normal physiological functions in ectotherms. To assess the effects of cold or heat stress on amphibians, giant spiny frogs (Quasipaa spinosa) were acclimated at 22 degrees C followed by exposure to 5 degrees C or 30 degrees C for 0, 3, 6, 12, 24 and 48 h, respectively. Histological alterations, apoptotic index, generation of mitochondria! reactive oxygen species (ROS), antioxidant activity indices and stress-response gene expression in frog livers were subsequently determined. Results showed that many fat droplets appeared after 12 h of heat stress and the percentage of melanomacrophage centres significantly changed after 48 h at both stress conditions. Furthermore, the mitochondrial ROS levels were elevated in a time-dependent manner up to 6 h and 12 h in the cold and heat stress groups, respectively. The activities of superoxide dismutase, glutathione peroxidase and catalase were successively increased with increasing periods of cold or heat exposure, and their gene expression levels showed similar changes in both stress conditions. Most tested heat shock protein (HSP) genes were sensitive to temperature exposure, and the expression profiles of most apoptosis-related genes was significantly upregulated at 3 and 48 h under cold and heat stress, respectively. Apoptotic index at 48 h under cold stress was significantly higher than that under heat stress. Notably, lipid droplets, HSP30, HSP70 and HSP110 might be suitable bioindicators of heat stress. The results of these alterations at physiological, biochemical and molecular levels might contribute to a better understanding of the stress response of Q. spinosa, and perhaps amphibians more generally, under thermal stress.
Osmotic pressure-adaptive responses in the eye tissues of rainbow smelt (Osmerus mordax)

Abstract:Purpose: The rainbow smelt (Osmerus mordax), is a teleost fish, which avoids freezing by becoming virtually isosmotic with seawater. The effects that such massive changes in osmolarity have on both its visual system and its highly evolved and specialized circulation are not known. New knowledge about the osmotic adaptation of the rainbow smelt eye is highly relevant to the adaptation and survival of this species and to its ability to feed as a visual predator in the face of environmental pressures. Moreover, the molecular physiologic response of the smelt to osmotic stress might provide valuable insights into understanding and managing mammalian pathological hyperosmolarity conditions, such as diabetes. We undertook the present study to provide an initial assessment of gene expression in ocular vasculature during osmotic adaptation in rainbow smelt. Methods: Immunohistochemistry with species cross reactive antibodies was used to assess blood vessel protein expression in paraffin sections. Western blotting was used to further verify antibody specificity for orthologs of mammalian blood vessel proteins in rainbow smelt. Thermal hysteresis and the analysis of glycerol concentrations in vitreous fluid were used to assess the physiologic adaptive properties of cold stressed eyes. Results: Glycerol levels and osmotic pressure were significantly increased in the vitreal fluid of smelt maintained at <0.5 °C versus those maintained at 8-10 °C. Compared to the 8-10 °C adapted specimens, the rete mirabile blood vessels and connecting regions of the endothelial linings of the choroidal vessels of the <0.5 °C adapted specimens showed a higher expression level of Tubedown (Tbdn) protein, a marker of the endothelial transcellular permeability pathway. Expression of the zonula occludens protein ZO-1, a marker of the endothelial paracellular permeability pathway showed a reciprocal expression pattern and was downregulated in rete mirabile blood vessels and connecting regions in the endothelial linings of choroidal vessels in <0.5 °C adapted specimens. Smelt orthologs of the mammalian Tbdn and zoluna occludens protein 1 (ZO-1) proteins were also detected by western blotting using anti-mammalian antibodies raised against the same epitopes as those used for immunohistochemistry. Conclusions: This work provides the first evidence that molecules known to play a role in ocular vascular homeostasis are expressed and may be differentially regulated during anti-freezing cold adaptation in smelt eyes. We propose a hypothesis that in a state of cold-induced hyperosmolarity, changes in ZO-1 expression are associated with the passage of small solutes from the plasma space to ocular fluid, while changes in Tbdn expression regulate the passage of proteins between the ocular fluid and plasma space. This work also provides fundamental insight into the mechanisms underlying the adaptation of the blood-retinal barrier to metabolically relevant compounds such as glycerol.

Abstract:BackgroundChanges in ambient temperature seriously affect physiological regulation and biochemical reactions in ectotherms. However, transient elevation in oceanic temperature occurs naturally during the day. Short-term elevation in the ambient temperature affects different physiological responses in marine fish, including cellular protein stability and osmotic balance of the internal environment. Since fish gills are vital osmoregulatory organ which directly contacts external environment, activation of cytoprotective responses to maintain gill cell viability and biological function is essential for fish survival under challenging environmental conditions. The purpose of this study was to investigate the short-term effects of elevated temperature on physiological regulation in the gills of a marine teleost, blue green damselfish (Chromis viridis).ResultsAs part of the stress response, plasma glucose levels were induced by short-term hyperthermic exposure (12 h). Furthermore, upregulation of the levels of gill heat shock proteins (HSPs) and ubiquitinated proteins was essential for preventing the accumulation of protein aggregations in branchial cells of C. viridis under hyperthermic stress. The specific activity of branchial Na+/K+-ATPase (NKA), however, significantly reduced while the amount of protein was similar between normal and high-temperature groups.ConclusionsThe present study provided the evidence to illustrate that activation of the branchial protein quality control mechanism to carry out cytoprotective response was involved in coping with thermal stress. However, plasma osmolality and muscle water content, respectively, that slightly but evidently increased and decreased might result from impaired osmoregulatory ability due to hyperthermia-decreased gill NKA activity.

Abstract:Goldfish and crucian carp at low temperature exhibit plasticity in gill morphology during exposure to hypoxia to enhance gas exchange. Hypoxia-induced changes in gill morphology and cellular ultrastructure of the high altitude scaleless carp from Lake Qinghai, China, were investigated to determine whether this is a general characteristic of cold water carp species. Fish were exposed to acute hypoxia (0.3 mg O-2 l(-1)) for 24 h followed by 12 h recovery in normoxic water (6 mg O-2 l(-1) at 3200 m altitude), with no mortality. Dramatic alterations in gill structure were initiated within 8 h of hypoxia and almost complete by 24 h, and included a gradual reduction of filament epithelial thickness (> 50%), elongation of respiratory lamellae, expansion of lamellar respiratory surface area (> 60%) and reduction in epithelial water-blood diffusion distance (< 50%). An increase in caspase 3 activity in gills occurred following 24 h exposure to hypoxia, indicating possible involvement of apoptosis in gill remodeling. Extensive gill mucous production during hypoxia may have been part of a general stress response or may have played a role in ion exchange and water balance. The large increase in lamellar surface area and reduction in diffusion distance presumably enhances gas transfer during hypoxia (especially in the presence of increased mucous production) but comes with an ionoregulatory cost, as indicated by a 10 and 15% reduction in plasma [Na+] and [Cl-], respectively, within 12-24 h of hypoxia. Within 12 h of hypoxia exposure, 'wavy-convex '-mitochondria rich cells (MRCs) with large apical crypts and numerous branched microvilli were transformed into small 'shallow-basin' cells with a flattened surface. As the apical membrane of MRCs is the site for active ion uptake from the water, a reduction in apical crypt surface area may have contributed to the progressive reduction in plasma [Na+] and [Cl-] observed during hypoxia. The changes in the macro-and ultra-structure of fish gills, and plasma [Na+] and [Cl-] during hypoxia were reversible, showing partial recovery by 12 h following return to normoxia. Although the large morphological changes in the gill observed in the scaleless carp support the hypothesis that gill remodeling during hypoxia is a general characteristic of cold water carp species, the reduced magnitude of the response in scaleless carp relative to goldfish and crucian carp may be a reflection of their more active lifestyle or because they reside in a moderately hypoxic environment at altitude.

Effects of Water Temperature on the Body Color and Expression of the Genes Related to Body Color Regulation in the Goldfish

Abstract:Melanin-concentrating hormone (MCH), melanocyte-stimulating hormone (MSH), and somatolactin (SL) in the hypothalamus-pituitary axis are associated with body color regulation in teleost fish. Although these hormones' production and secretion respond well to light environments, such as background color, little is known about the effects of different water temperatures. We investigated the effects of water temperature, 10°C, 20°C, and 30°C, on body color and the expression of these genes and corresponding receptor genes in goldfish. The body color in white background (WBG) becomes paler at the higher water temperature, although no difference was observed in black background (BBG). Brain mRNA contents of proMCH genes (pmch1 and pmch2) increased at 30°C and 20°C compared to 10°C in WBG, respectively. Apparent effects of background color and temperature on the pituitary mRNA contents of a POMC gene (pomc) were not observed. The pituitary mRNA contents of the SLα gene were almost double those on a WBG at any temperature, while those of the SLβ gene (slb) at 30°C tended to be higher than those at 10°C and 20°C on WBG and BBG. The scale mRNA contents of the MCH receptor gene (mchr2) in WBG were higher than those in BBG at 30°C. The highest scale mRNA contents of MSH receptor (mc1r and mc5r) on BBG were observed at 20°C, while the lowest respective mRNA levels were observed at 30°C on WBG. These results highlight the importance of temperature for the endocrinological regulation of body color, and darker background color may stabilize those endocrine functions.

Abstract:The skeletal muscle is a highly plastic tissue. Its ability to respond to external stimuli and challenges allows it to face the functional needs of the organism. In the goldfish Carassius auratus, a model of hypoxia resistance, exposure to reduced oxygen is accompanied by an improvement of the swimming performance, relying on a sustained contractile behavior of the skeletal muscle. At the moment, limited information is available on the mechanisms underlying these responses. We here evaluated the effects of short- (4 days) and long- (20 days) term exposure to moderate water hypoxia on the goldfish white skeletal muscle, focusing on oxidative status and mitochondrial dynamics. No differences in lipid peroxidation, measured as 2-thiobarbituric acid-reacting substances (TBARS), and oxidatively modified proteins (OMP) were detected in animals exposed to hypoxia with respect to their normoxic counterparts. Exposure to short-term hypoxia was characterized by an enhanced SOD activity and expression, paralleled by increased levels of Nrf2, a regulator of the antioxidant cell response, and HSP70, a chaperone also acting as a redox sensor. The expression of markers of mitochondrial biogenesis (TFAM) and abundance (VDAC) and of the mtDNA/nDNA ratio was similar under normoxia and under both short- and long-term hypoxia, thus excluding a rearrangement of the mitochondrial apparatus. Only an increase of PGC1α (a transcription factor involved in mitochondrial dynamics) was detected after 20 days of hypoxia. Our results revealed novel aspects of the molecular mechanisms that in the goldfish skeletal muscle may sustain the response to hypoxia, thus contributing to adequate tissue function to organism requirements.

The interactive effects of exercise and gill remodeling in goldfish (Carassius auratus)

Abstract:Gill remodeling in goldfish (Carassius auratus) is accomplished by the appearance or retraction of a mass of cells (termed the interlamellar cell mass or ILCM) between adjacent lamellae. Given the presumed effects of gill remodeling on diffusing capacity, the goals of the current study were (1) to determine the consequences of increased aerobic O(2) demand (swimming) on gill remodelling and (2) to assess the consequences of the presence or absence of the ILCM on aerobic swimming capacity. Fish acclimated to 7 °C exhibited a marked increase in the ILCM which occupied, on average, 70.0 ± 4.1% of the total interlamellar channel area in comparison to an average ILCM area of only 28.3 ± 0.9% in fish acclimated to 25 °C. Incrementally increasing swimming velocity in fish at 7 °C to achieve a maximum aerobic swimming speed (U (CRIT)) within approximately 3 h resulted in a marked loss of the ILCM area to 44.8 ± 3.5%. Fish acclimated to 7 °C were subjected to 35 min swimming trials at 30, 60 or 80% U (CRIT) revealing that significant loss of the ILCM occurred at swimming speeds exceeding 60% U (CRIT). Prior exposure of cold water-acclimated fish to hypoxia to induce shedding of the ILCM did not affect swimming performance when assessed under normoxic conditions (control fish U (CRIT) = 2.34 ± 0.30 body lengths s(-1); previously hypoxic fish U (CRIT) = 2.99 ± 0.14 body lengths s(-1)) or the capacity to raise rates of O(2) consumption with increasing swimming speeds. Because shedding of ILCM during U (CRIT) trials complicated the interpretation of experiments designed to evaluate the impact of the ILCM on swimming performance, additional experiments using a more rapid 'ramp' protocol were performed to generate swimming scores. Neither prior hypoxia exposure nor a previous swim to U (CRIT) (both protocols are known to cause loss of the ILCM) affected swimming scores (the total distance swum during ramp U (CRIT) trials). However, partitioning all data based on the extent of ILCM coverage upon cessation of the swimming trial revealed that fish with less than 40% ILCM coverage exhibited a significantly greater swimming score (539 ± 86 m) than fish with greater than 50% ILCM coverage (285 ± 70 m). Thus, while loss of the ILCM at swimming speeds exceeding 60% U (CRIT) confounds the interpretation of experiments designed to assess the impact of the ILCM on swimming performance, we suggest that the shedding of the ILCM, in itself, coupled with improved swimming scores in fish exhibiting low ILCM coverage (<40%), provide evidence that the ILCM in goldfish acclimated to cold water (7 °C) is indeed an impediment to aerobic swimming capacity.
Modulation of Antioxidant Enzymes, Heat Shock Protein, and Intestinal Microbiota of Large Yellow Croaker (Larimichthys crocea) Under Acute Cold Stress

Abstract:The experiment was conducted in a sealed, temperature-controlled room. The initial water temperature of the treatment group was 12°C, lowered by 1°C a day to 8°C using a chiller, and maintained at that temperature for 3 days. The water temperature was then increased by 1°C per day to 12°C using a heater. The control group were the fish that kept at 12°C at the beginning of the experiment; T1 represents the group that sampled on the first day when the water temperature reached 8 from 12°C; T2 represents the group that sampled when the fish had been kept at 8°C for 3 days; T3 represents the group sampled when the water temperature was increased to 12 from 8°C. The antioxidant abilities of the liver, muscle, and intestine of large yellow croaker at different temperatures and times were determined, and the expression of HSP70 and HSP90 in the liver as well as the changes in intestinal microbiota were investigated. The results showed that the activities of SOD, CAT, GSH-Px, and MDA content increased significantly when the water temperature decreased from 12 to 8°C, significantly higher than those in the control group. The gene expression levels of HSP70 and HSP90 in the liver of large yellow croaker significantly increased under low temperature stress and were significantly higher than those in the control group. The diversity and abundance of intestinal microbiota of large yellow croaker were altered when the water temperature was maintained at 8°C, although the change was not obvious. Proteobacteria, Firmicutes, and Bacteroidetes were the dominate phyla in both the 12 and 8°C groups, and the temperature variation did not change the functional pathways of the microbiota. In conclusion, the acute temperature variation affected the antioxidant ability and heat shock protein expression of large yellow croaker but did not affect the diversity or abundance of the intestinal microbiota. These results will provide a theoretical basis for studying the mechanisms of large yellow croaker overwintering and adaptation to low temperatures.

Effects of saline immersion on the physiological alterations of grass goldfish (Carassius auratus) during subsequent recovery in freshwater

Abstract:The present work aims to evaluate the tolerance, osmoregulation, metabolism, and antioxidant ability of saline water immersed grass goldfish (Carassius auratus) during the recovery in freshwater. Grass goldfish (38.15 ± 5.48g) acclimated in freshwater were immersed by salinities (0‰, 20‰ and 30‰) for different time durations (10, 20, 30 and 60 min); and the physiological responses were measured during freshwater recovery. The blood osmolalities were not significantly different at any group fish, while whereas the decline of Na+ concentration and the ratio of Na+/Cl- as well as the rise of Cl- concentration was observed in saline treated fish. Soon after freshwater recovery, the transcription of NKA-α and NKA-β mRNA in gills of salinity 20 immersed fish elevated significantly and then decreased, whereas no obvious changes were detected in salinity 30 treated fish. Till 24h post freshwater recovery, gill Na+/K+-ATPase activities in saline treated fish were lower than control group except for the fish immersed by salinity 20 for 10-30 min. At 24h of recovery, cortisol levels in salinity 20 immersed fish were lower than salinity 30 treated fish, but remained higher than control. As for serum lactic acid, fish treated by salinity 20 for 10 or 20 min did not show any fluctuation. However, higher lactic acid contents were detected in all other five salinity treated groups during recovery. Generally, at 24 h of recovery, salinity 20 treated fish exhibited higher SOD and CAT activities than fish immersed by salinity 30. In summary, grass goldfish could survive by immersion in salinity 20 less than 60 min or salinity 30 less than 30min, even though immersion by salinity 20 could minimize the negative effects.
Environmental Temperature Variation Affects Brain Lipid Composition in Adult Zebrafish (Danio rerio)

Abstract:This study delves deeper into the impact of environmental temperature variations on the nervous system in teleost fish. Previous research has demonstrated that exposing adult zebrafish (Danio rerio) to 18 °C and 34 °C for 4 or 21 days induces behavioural changes compared to fish kept at a control temperature of 26 °C, suggesting alterations in the nervous system. Subsequent studies revealed that these temperature conditions also modify brain protein expression, indicating potential neurotoxic effects. The primary aim of this work was to investigate the effects of prolonged exposure (21 days) to 18 °C or 34 °C on the brain lipidomes of adult zebrafish compared to a control temperature. Analysis of the brain lipidome highlighted significant alteration in the relative abundances of specific lipid molecules at 18 °C and 34 °C, confirming distinct effects induced by both tested temperatures. Exposure to 18 °C resulted in an increase in levels of phospholipids, such as phosphatidylethanolamine, alongside a general reduction in levels of sphingolipids, including sphingomyelin. Conversely, exposure to 34 °C produced more pronounced effects, with increases in levels of phosphatidylethanolamine and those of various sphingolipids such as ceramide, gangliosides, and sphingomyelin, alongside a reduction in levels of ether phospholipids, including lysophosphatidylethanolamine ether, phosphatidylethanolamine ether, and phosphatidylglycerol ether, as well as levels of glycolipids like monogalactosyldiacylglycerol. These results, when integrated with existing proteomic and behavioural data, offer new insights into the effects of thermal variations on the nervous system in teleost fish. Specifically, our proteomic and lipidomic findings suggest that elevated temperatures may disrupt mitochondrial function, increase neuronal susceptibility to oxidative stress and cytotoxicity, alter axonal myelination, impair nerve impulse transmission, hinder synapse function and neurotransmitter release, and potentially lead to increased neuronal death. These findings are particularly relevant in the fields of cell biology, neurobiology, and ecotoxicology, especially in the context of global warming.
Hypoxia increases intracellular calcium in glutamate-activated horizontal cells of goldfish retina via mitochondrial K ATP channels and intracellular stores

Abstract:Central neurons of the common goldfish ( Carassius auratus ) are exceptional in their capacity to survive Ca 2+ -induced excitotoxicity and cell death during hypoxia. Horizontal cells (HCs) are inhibitory interneurons of the retina that are tonically depolarized by the neurotransmitter, glutamate, yet preserve intracellular Ca 2+ homeostasis. In HCs isolated from goldfish, and in the absence of glutamatergic input, intracellular Ca 2+ concentration ([Ca 2+ ] i ) is protected from prolonged exposure to hypoxia by mitochondrial ATP-dependent K + (mK ATP ) channel activity. In the present study, we investigated the effects of hypoxia upon [Ca 2+ ] i in isolated HCs during tonic activation by glutamate to better predict the effects of hypoxia in the active retina. Dynamic changes in [Ca 2+ ] i were measured using the ratiometric Ca 2+ indicator, Fura-2. Application of 100 μM glutamate during hypoxia (P O2 = 25 mmHg) produced a 1.3-fold greater rise in [Ca 2+ ] i compared to the same glutamate stimulus during normoxia. The hypoxia-dependent increase in [Ca 2+ ] i was abolished by application of 5-hydroxydecanoic acid, which renders mK ATP channels inactive. Extracellular Ca 2+ did not contribute to the elevated [Ca 2+ ] i observed during hypoxia, as the effect persisted in Ca 2+ -free solution and during application of verapamil, an L-type Ca 2+ channel blocker. By contrast, inhibition of the mitochondrial Ca 2+ uniporter or ryanodine receptors (with ruthenium red or ryanodine, respectively) abolished the hypoxia-dependent rise in [Ca 2+ ] i . This study reports an mK ATP -dependent rise in [Ca 2+ ] i during hypoxia in HCs activated by glutamate, and suggests roles for the mitochondria and intracellular Ca 2+ stores in regulating this mechanism.

Abstract:Water temperature plays a crucial role in the growth, survival, and reproduction of fish species, as they make up the majority of aquatic fauna. In this study, the effects of low temperature were studied on the functional state of juvenile golden pompano ( Trachinotus ovatus ) under low-temperature stress. The study was conducted at 28°C in the control group and 18°C in the cold group for 14 d to determine the intestinal tissue, digestive and antioxidant enzyme activities, and metabolites of juvenile fish. The results showed that: (1) the swelling degree of the muscle layer deepened and was congested with a longer low-temperature stress period. The folds were sparse, from slight swelling to shedding and deformation. The intestinal mucosa was necrotic and had vacuoles, and the number gradually increased. Serious erosion of the villi occurred. (2) The specific activities of digestive enzymes showed a downward trend. (3) The intestinal superoxide dismutase (SOD) activity, malondialdehyde (MDA) content, and glutathione peroxidase (GSH-Px) activity showed an upward trend. The intestinal catalase (CAT) activity showed a downward trend. (4) Compared with the control group, there were 28 metabolites in the cold group showing significant differences, among which Z, 11Z, 14Z-eicosatrienoic acid, stearic acid, and adrenic acid showed an upward trend. In contrast, spermidine and uracil showed a downward trend. Among the enriched metabolic pathways, the main differential pathways were unsaturated fatty acid biosynthesis, fatty acid biosynthesis, linoleic acid metabolism, pyrimidine metabolism, and β-alanine metabolism. According to metabolomic analysis, under low-temperature stress, the fish body improved the synthesis of unsaturated fatty acids and saturated fatty acids to adapt to a low-temperature environment and consumed spermidine to improve its immune ability to clear the peroxide generated by the synthesis of unsaturated fatty acids in the body so that the cells were protected from oxidative damage. After 14 days, low-temperature stress affected metabolites and enzyme activity indices in juvenile golden pompano. Low-temperature stress causes changes in intestinal antioxidants and digestive enzymes and damage intestinal tissues. As a result of this exploration of how low temperatures affect the juvenile golden pompano, the foundation is laid for future studies, such as the molecular mechanisms of low-temperature adaptation in fish species.

Abstract:Dysfunction of AML1/Runx1, a transcription factor, plays a crucial role in the development of many types of leukemia. Additional events are often required for AML1 dysfunction to induce full-blown leukemia; however, a mechanistic basis of their cooperation is still elusive. Here, we investigated the effect of AML1 deficiency on the development of MLL-ENL leukemia in mice. Aml1 excised bone marrow cells lead to MLL-ENL leukemia with shorter duration than Aml1 intact cells in vivo. Although the number of MLL-ENL leukemia-initiating cells is not affected by loss of AML1, the proliferation of leukemic cells is enhanced in Aml1excised MLL-ENL leukemic mice. We found that the enhanced proliferation is the result of repression of p19ARF that is directly regulated by AML1 in MLL-ENL leukemic cells. We also found that downregulation of p19ARF induces the accelerated onset of MLL-ENL leukemia, suggesting that p19ARF is a major target of AML1 in MLL-ENL leukemia. Abstract:Although there are studies in the literature on the effects of different coloured light-emitting diodes (LEDs) on different organisms, there is limited information on how these effects change with temperature increase. In this study, the effects of blue, green, red and white LED lights on the early development process of zebrafish (Danio rerio (Hamilton, 1822)) were comprehensively investigated. In addition, to simulate global warming, it was examined how a one-degree temperature increase affects this process. For this purpose, zebrafish embryos, which were placed at 4 hpf (hours post fertilization) in an incubator whose interior was divided into four areas, were kept at three different temperatures (28, 29 and 30 °C) for 120 h. The group kept in a dark environment was chosen as the control. The temperature of the control group was also increased at the same rate as the other groups. The results showed that at the end of the exposure period, temperature and light colour caused an increase in body malformations. Histopathological damage and immunopositive signals of HSP 70 and 8-OHdG biomarkers in larval brains, increase in free oxygen radicals, apoptotic cells and lipid accumulation throughout the body, increase in locomotor activity, decrease in heart rate and blood flow, and significant changes in more than thirty metabolite levels were detected. In addition, it has been determined that many metabolic pathways are affected, especially glutathione, vitamin B6 and pyrimidine metabolism. Moreover, it has been observed that a one-degree temperature increase worsens this negative effect. It was concluded that blue light was the closest light to the control group and was less harmful than other light colours. The study revealed that blue light produced results that were most similar to those seen in the control group.
