Abstract:In this study, the electrospinning technique was employed to create a nanofiber membrane by stretching an organic polymer into nanofibers under a high electric field. Metronidazole (MNZ) at a concentration of 3 wt% was loaded into a poly(lactic-co-glycolic acid) (PLGA) and silk fibroin (SF)-blended nanofiber membrane. This formulation aims to achieve effective and sustained drug release, enabling the eradication of bacteria for the efficient treatment of periodontitis. Results demonstrated that SF interacted with PLGA molecules, forming dense and uniform nanofibers with a diameter of 570 nm. Excessive SF molecules tended to aggregate, leading to an increased particle size, with the interaction between MNZ and SF contributing to adhesion. The composition of MNZ, SF, and PLGA formed a physical chimera without any chemical reactions. Moreover, as the SF content increased, the tensile properties of the membrane gradually improved. Concurrently, the in vitro degradation rate increased with higher SF content. Among the various groups tested, the 3 wt% MNZ/PLGA/SF 2:1 membrane exhibited superior drug release characteristics, with 71.76% release within 24 h. This formulation demonstrated excellent antibacterial properties, indicated by a bacterial inhibition diameter of 13.5 mm, noteworthy hydrophilicity with a contact angle of 44.3°, and favorable biocompatibility. The membrane holds significant application value in regenerative engineering and drug delivery systems, showcasing substantial potential for the treatment of periodontitis.

Abstract:Bioactive membranes with growth factors that mimic the physiological microenvironment of periodontal tissues have shown great potential for guided tissue regeneration (GTR). However, the loss of bioactivity and the uncontrolled drug release in conventional membranes limit their efficacy. In addition, membranes with local infection control ability would provide additional benefits to regeneration. In this work, nanoengineered membranes with growth factor and antibiotic delivery capabilities were developed to achieve dual functions for efficient GTR. Mesoporous silica nanoparticles (MSNs) with large pores with the ability to encapsulate and preserve the bioactivity of growth factors were used as the core, and antibiotics were loaded in shell nanofibers with coaxial electrospinning. A sustained release behavior of the growth factor was observed, and the nanoparticle-encapsulated core–shell nanofibers show better ontogenetic regeneration abilities toward bone marrow stromal cells, compared to current commercial Bio-Gide membranes. In addition, the drug-loaded nanocomposite core–shell nanofibers showed excellent antibacterial properties toward Gram-positive, Gram-negative, and multispecies oral bacteria. The MSNs-embedded core–shell nanofiber system provides an attractive strategy to deliver dual drugs for GTR with better performance.

Abstract:Antibiotic administration is an adjacent therapy to guided tissue regeneration (GTR) in the management of periodontitis. This is due to the major role of pathogen biofilm in aggravating periodontal defects. This study aimed to fabricate a GTR membrane for sustained delivery of doxycycline hydrochloride (DOX) while having a space-maintaining function. The membranes were prepared using a polymeric blend of polycaprolactone/polyvinyl alcohol/chitosan by the electrospinning technique. The obtained membranes were characterized in terms of physicochemical and biological properties. Nanofibers showed a mean diameter in the submicron range of < 450 nm while having uniform randomly aligned morphology. The obtained membranes showed high strength and flexibility. A prolonged in vitro release profile during 68 h was observed for manufactured formulations. The prepared membranes showed a cell viability of > 70% at different DOX concentrations. The formulations possessed antimicrobial efficacy against common pathogens responsible for periodontitis. In vivo evaluation also showed prolonged release of DOX for 14 days. The histopathological evaluation confirmed the biocompatibility of the GTR membrane. In conclusion, the developed nanofibrous DOX-loaded GTR membranes may have beneficial characteristics in favour of both sustained antibiotic delivery and periodontal regeneration by space-maintaining function without causing any irritation and tissue damage.

Abstract:Periodontitis is a prevalent chronic, destructive inflammatory disease affecting tooth‐supporting tissues in humans. Guided tissue regeneration strategies are widely utilized for periodontal tissue regeneration generally by using a periodontal membrane. The main role of these membranes is to establish a mechanical barrier that prevents the apical migration of the gingival epithelium and hence allowing the growth of periodontal ligament and bone tissue to selectively repopulate the root surface. Currently available membranes have limited bioactivity and regeneration potential. To address such challenges, an osteoconductive, antibacterial, and flexible poly(caprolactone) (PCL) composite membrane containing zinc oxide (ZnO) nanoparticles is developed. The membranes are fabricated through electrospinning of PCL and ZnO particles. The physical properties, mechanical characteristics, and in vitro degradation of the engineered membrane are studied in detail. Also, the osteoconductivity and antibacterial properties of the developed membrane are analyzed in vitro. Moreover, the functionality of the membrane is evaluated with a rat periodontal defect model. The results confirmed that the engineered membrane exerts both osteoconductive and antibacterial properties, demonstrating its great potential for periodontal tissue engineering. Abstract:Electrospinning technique has attracted considerable attention in fabrication of cellulose nanofibrils or nanocellulose membranes, in which polycaprolactone (PCL) could be used as a promising precursor to prepare various cellulose nanofibril membranes for periodontal tissue regeneration. Conventional bio-membranes and cellulose films used in guided tissue regeneration (GTR) can prevent the downgrowth of epithelial cells, fibroblasts, and connective tissue in the area of tooth root but have limitations related to osteogenic and antimicrobial properties. Cellulose nanofibrils can be used as an ideal drug delivery material to encapsulate and carry some drugs. In this study, magnesium oxide (MgO) nanoparticles-incorporated PCL/gelatin core-shell nanocellulose periodontal membranes were fabricated using coaxial electrospinning technique, which was termed as Coaxial-MgO. The membranes using single-nozzle electrospinning technique, namely Blending-MgO and Blending-Blank, were used as control. The morphology and physicochemical property of these nanocellulose membranes were characterized by scanning electron microscopy (SEM), energy-dispersive spectrum of X-ray (EDS), transmission electron microscopy (TEM), contact angle, and thermogravimetric analysis (TGA). The results showed that the incorporation of MgO nanoparticles barely affected the morphology and mechanical property of nanocellulose membranes. Coaxial-MgO with core-shell fiber structure had better hydrophilic property and sustainable release of magnesium ion (Mg 2+ ). CCK-8 cell proliferation and EdU staining demonstrated that Coaxial-MgO membranes showed better human periodontal ligament stem cells (hPDLSCs) proliferation rates compared with the other group due to its gelatin shell with great biocompatibility and hydrophilicity. SEM and immunofluorescence assay results illustrated that the Coaxial-MgO scaffold significantly enhanced hPDLSCs adhesion. In vitro osteogenic and antibacterial properties showed that Coaxial-MgO membrane enhanced alkaline phosphatase (ALP) activity, formation of mineralized nodules, osteogenic-related genes [ALP, collagen type 1 (COL1), runt-related transcription factor 2 (Runx2)], and high antibacterial properties toward Escherichia coli ( E. coli ) and Actinobacillus actinomycetemcomitans ( A. a ) when compared with controls. Our findings suggested that MgO nanoparticles-incorporated coaxial electrospinning PCL-derived nanocellulose periodontal membranes might have great prospects for periodontal tissue regeneration.

Abstract:The development of flexible and porous materials to control antibacterial delivery is a pivotal endeavor in medical science. In this study, we aimed to produce long and defect-free fibers made of zein and hydroxypropyl methylcellulose acetate succinate (HPMCAS) to be used as a platform for the release of metronidazole (MDZ) and metronidazole benzoate (BMDZ) to be potentially used in periodontal treatment. Microfibers prepared via electrospinning under a 2:3 (w/w) zein to HPMCAS ratio, containing 0.5 % (w/w) poly(ethylene oxide) (PEO) and 1 % (w/w) cellulose nanofibril (CNF) were loaded with 40 % (w/w) MDZ, 40 % (w/w) BMDZ, or a combination of 20 % (w/w) of each drug. The addition of CNF improved the electrospinning process, resulting in long fibers with reduced MDZ and BMDZ surface crystallization. MDZ- and BMDZ-incorporated fibers were semicrystalline and displayed commendable compatibility among drugs, nanocellulose and polymeric chains. Release tests showed that zein/HPMCAS/PEO fibers without CNF and with 20 % (w/w) MDZ/ 20 % (w/w) BMDZ released the drug at a slower and more sustained rate compared to other samples over extended periods (up to 5 days), which is a favorable aspect concerning periodontitis treatment.

Abstract:This study developed Chitosan/Collagen film surfaces and drug‐loaded PVA/Amoxicillin nanofibers using solvent casting and electrospinning, respectively. The hybrid bilayer membranes exhibited over 90% cell viability and showed superior antimicrobial efficacy against Streptococcus mutans, suggesting potential for enhanced cell growth, controlled drug release, and improved regeneration of gingival tissues. Periodontal diseases, if untreated, can cause gum recession and tooth root exposure, resulting in infection and irreversible damage. Traditional treatments using autologous grafts are painful and often result in postoperative complications. Scaffolds offer a less invasive alternative, promoting cell proliferation and healing without additional surgery, thus enhancing comfort for patients and doctors. This study developed Chitosan (Chit)/Collagen (Col) film surfaces and drug‐loaded Polyvinyl Alcohol (PVA)/Amoxicillin (AMX) nanofibers using solvent casting and electrospinning methods, respectively. The surfaces are characterized by scanning electron microscopy (SEM), mechanical testing, Fourier Transform Infrared Spectroscopy (FTIR), and differential scanning calorimetry (DSC). Biocompatibility and antimicrobial properties are assessed using NIH/3T3 fibroblast cells and bacterial cultures. SEM images confirmed the structural integrity of AMX‐loaded 13% PVA nanofibers, while FTIR analysis validated the compositional integrity of PVA/AMX nanofibers and Chit/Col film hybrid surfaces. Cell studies showed over 90% viability for Chit/Col film + PVA/AMX nanofiber hybrid bilayer membranes, confirming their biocompatibility. The antimicrobial assessment indicated that the Chit/Col film + PVA/AMX (0.2%) nanofiber hybrid bilayer membrane exhibited superior efficacy against Streptococcus mutans. These findings suggest that this hybrid bilayer membrane can enhance cell growth, promote proliferation, and enable controlled drug release, offering significant promise for regeneration of gingival tissues.

Personalized and Defect-Specific Antibiotic-Laden Scaffolds for Periodontal Infection Ablation

Abstract:Periodontitis compromises the integrity and function of tooth-supporting structures. Although therapeutic approaches have been offered, predictable regeneration of periodontal tissues remains intangible, particularly in anatomically complex defects. In this work, personalized and defect-specific antibiotic-laden polymeric scaffolds containing metronidazole (MET), tetracycline (TCH), or their combination (MET/TCH) were created via electrospinning. An initial screening of the synthesized fibers comprising chemo-morphological analyses, cytocompatibility assessment, and antimicrobial validation against periodontopathogens was accomplished to determine the cell-friendly and anti-infective nature of the scaffolds. According to the cytocompatibility and antimicrobial data, the 1:3 MET/TCH formulation was used to obtain three-dimensional defect-specific scaffolds to treat periodontally compromised three-wall osseous defects in rats. Inflammatory cell response and new bone formation were assessed by histology. Micro-computerized tomography was performed to assess bone loss in the furcation area at 2 and 6 weeks post implantation. Chemo-morphological and cell compatibility analyses confirmed the synthesis of cytocompatible antibiotic-laden fibers with antimicrobial action. Importantly, the 1:3 MET/TCH defect-specific scaffolds led to increased new bone formation, lower bone loss, and reduced inflammatory response when compared to antibiotic-free scaffolds. Altogether, our results suggest that the fabrication of defect-specific antibiotic-laden scaffolds holds great potential toward the development of personalized (i.e., patient-specific medication) scaffolds to ablate infection while affording regenerative properties.

Abstract:Synthetic periodontal membranes have appeared as a promising treatment modality for periodontal disease. They provide a suitable site for attachment and growth environment for periodontal cells and exhibit high biocompatibility and high water retaining capacity. However, existing membranes can develop biofilms and induce inflammation at the periodontal pocket and gingival cells, hence limiting their effectiveness. A Polycaprolactone/sodium alginate-based core-shell membrane with Magnesium doped Zinc oxide (PCL/SA/MgZnO) nanoparticles (NPs) was developed, with initial burst release followed by sustained release of the antibacterial agent. The membrane was synthesized using casting and dip coating methods and characterized using Scanning Electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR). The membranes were then evaluated for their antibacterial potential against Enterococcus faecalis, Streptococcus mutans , saliva microcosm, and diabetic saliva microcosm. Furthermore, they were analyzed for water-retaining capacity as well as mechanical properties and biocompatibility. The membranes were finally tested in a novel alloxan-induced diabetic periodontal rodent model based on the coinfection of E. faecalis and S. mutans. The results showed the enhanced antibacterial potential of PCL/SA/MgZnO membranes against E. faecalis, S . mutans , saliva microcosm, and diabetic saliva microcosm. The PCL/SA/MgZnO exhibited better mechanical strength as compared to PCL/SA and PCL/SA/ZnO membranes. The histological findings showed reduced inflammation in diabetic rats with periodontitis treated with PCL/SA/MgZnO. Furthermore, periodontal regeneration was also observed in the X-ray radiographs of rats treated with PCL/SA/MgZnO membranes. The PCL/SA-based core-shell membranes with MgZnO are effective candidates for the treatment of periodontal biofilms and inflammation in high-risk patients such as diabetic patients.

Designing of Drug Delivery Systems to Improve the Antimicrobial Efficacy in the Periodontal Pocket Based on Biodegradable Polyesters

Abstract:Delivery systems for biologically active substances such as proanthocyanidins (PCANs), produced in the form of electrospun nonwoven through the electrospinning method, were designed using a polymeric blend of poly(L-lactide-co-glycolide) (PLGA)and poly[(R,S)-3-hydroxybutyrate] ((R,S)-PHB). The studies involved the structural and thermal characteristics of the developed electrospun three-dimensional fibre matrices unloaded and loaded with PCANs. In the next step, the hydrolytic degradation tests of these systems were performed. The release profile of PCANs from the electrospun nonwoven was determined with the aid of UV-VIS spectroscopy. Approximately 30% of the PCANs were released from the tested electrospun nonwoven during the initial 15-20 days of incubation. The chemical structure of water-soluble oligomers that were formed after the hydrolytic degradation of the developed delivery system was identified through electrospray ionization mass spectrometry. Oligomers of lactic acid and OLAGA oligocopolyester, as well as oligo-3-hydroxybutyrate terminated with hydroxyl and carboxyl end groups, were recognized as degradation products released into the water during the incubation time. It was also demonstrated that variations in the degradation rate of individual mat components influenced the degradation pattern and the number of formed oligomers. The obtained results suggest that the incorporation of proanthocyanidins into the system slowed down the hydrolytic degradation process of the poly(L-lactide-co-glycolide)/poly[(R,S)-3-hydroxybutyrate] three-dimensional fibre matrix. In addition, in vitro cytotoxicity and antimicrobial studies advocate the use of PCANs for biomedical applications with promising antimicrobial activity.
Preparation of Doxycycline Hydrochloride Guided Tissue Regeneration/Guided Bone Regeneration Membranes Through Electrostatic Spinning for the Treatment of Peri-Implantitis

Abstract:Objective: The nanofibre membrane with the antibacterial doxycycline (DOX) intermediate layer can direct tissue regeneration and have antibacterial properties. Through the use of electrospinning, we were able to create DOX-loaded guided tissue regeneration (GTR)/guided bone regeneration (GBR) membranes and assess their efficacy in the treatment of peri-implantitis. Methods: Electrostatic spinning was used to create DOX-containing poly(caprolactone) (PCL) nanofibres, and a scanning electron microscope (SEM) was used to examine the membranes' surface morphology. Results: The synthesised DOX-PCL nanofibres were found to have a smooth surface and a uniform diameter distribution, as revealed by scanning electron microscopy. When the percentage of DOX was raised from 15% to 25%, the fibre diameter shrank from 247.17 nm to 181.34 nm. According to differential calorimetry, the heat absorption peaks for the pre-electrospun PCL and 10% PCL electrospun membranes were most pronounced at 66 °C, while the peaks for the DOX powder showed at 171.5 °C. With the shift in drug loading, carbonization occurred at 223.5 degrees Celsius. PCL underwent a thermal reaction between 60 and 66 degrees Celsius. However, medication loading was strongly correlated with the level of suppression. Agar diffusion results showed that DOX-loaded nanofibre membranes inhibited the growth of Actinomyces (Aa) and Porphyromonas gingivalis; for both bacterial species, the diameter of the inhibition zone grew larger when more drug was added to the membranes. After 1 hour, 4 hours, 12 hours, 1 day, 7 days, and 28 days of the experiment, the cumulative drug release rates of CL+DOX were 19.14%, 36.16%, 44.37%, 59.52%, and 65.150%, respectively. After the initial steep drop during the first three days of the trial, the PCL-DOX release rate stabilised at around 1.61 percent per hour. Minimum concentration was 3.13 g/mL, while PCL-DOX release rate fell from 1.28 percent on Day 4 to 0.51 percent on Day 28. Conclusion: Combining the availability of natural polymeric materials with the reliability of manufactured polymeric materials, the three-layer nanofibre membranes use PCL-GE as a double surface layer and PCL-DOX as an intermediate layer. In addition, the physical insulating qualities of these membranes are maintained, which allows the fibre membrane to perform a sustained antibacterial action and prevents the initial, abrupt release of the medication in the tissue. Peri-implantitis may be treatable with the use of both pharmaceuticals and GTR/GBR technology.

Abstract:Background Periodontal disease is a chronic inflammatory condition that gradually deteriorates the supportive tissues of teeth, eventually leading to tooth loss. Mechanical debridement stands as the gold standard method for treating periodontitis. However, antimicrobial therapy is recommended for optimal results when used alongside mechanical debridement. Numerous studies have investigated local drug delivery as an adjunct to mechanical debridement of affected tooth surfaces. Ocimum sanctum exhibits anti-inflammatory, antioxidant, and antimicrobial properties. Similarly, curcumin, as documented in the literature, demonstrates a broad spectrum of anti-inflammatory and antimicrobial effects. Electrospinning has demonstrated itself to be a highly effective method for fabricating drug-loaded fibers. Electrospun nanofibers containing Ocimum sanctum and curcumin are expected to exhibit greater efficacy due to their increased surface area, facilitating the dispersion of larger quantities of drugs, and their ability to control drug release when employed as a local drug delivery system. This study aims to fabricate and characterize the properties of nanofiber membranes loaded with Ocimum sanctum and curcumin using the electrospinning technique. Methods About 50 mg each of Ocimum sanctum and curcumin were blended with 15% polyvinyl alcohol and 2% chitosan polymer in a 4:1 ratio and left to stir overnight. A 10 mL syringe was filled with this solution, and an 18 G blunt-end needle charged at 15.9 kV was used for extrusion. Continuous fibers were collected onto a collector plate positioned 12 cm from the center of the needle tip, at a flow rate of 0.005 mL/min. The morphology of the fabricated membrane was assessed through scanning electron microscopy (SEM), the strength of the material was assessed through tensile strength analysis using INSTRON, an Electropuls E3000 Universal Testing Machine (INSTRON, Norwood, MA), and the drug release pattern was analyzed using Jasco V-730 UV-visible spectrophotometer (Jasco, Easton, MD). Results The morphology of this nanofiber showed a random distribution of fibers with no bead formation. The average diameter of the membrane was 383±102 nm, and the tensile strength of this material was 1.87 MPa. The drug release pattern showed an initial burst release of Ocimum sanctum, followed by a controlled release in subsequent hours. However, curcumin showed very little drug release because of its solubility. Conclusion In summary, the Ocimum sanctum and curcumin-loaded nanofibers exhibited robust tensile strength, a controlled drug release profile, and uniform drug distribution within the nanofiber membrane. Consequently, it can be concluded that curcumin nanofibers and electrospun Ocimum sanctum serve as valuable agents for local drug delivery in the treatment of periodontitis.

Abstract:The continuous stimulation of periodontitis leads to a decrease in the number of stem cells within the lesion area and significantly impairing their regenerative capacity. Therefore, it is crucial to promote stem cell homing and regulate the local immune microenvironment to suppress inflammation for the regeneration of periodontitis-related tissue defects. Here, we fabricated a novel multifunctional bilayer nanofibrous membrane using electrospinning technology. The dense poly(caprolactone) (PCL) nanofibers served as the barrier layer to resist epithelial invasion, while the polyvinyl alcohol/chitooligosaccharides (PVA/COS) composite nanofiber membrane loaded with calcium beta-hydroxy-beta-methylbutyrate (HMB-Ca) acted as the functional layer. Material characterization tests revealed that the bilayer nanofibrous membrane presented desirable mechanical strength, stability, and excellent cytocompatibility. In vitro , PCL@PVA/COS/HMB-Ca (P@PCH) can not only directly promote rBMSCs migration and differentiation, but also induce macrophage toward pro-healing (M2) phenotype-polarization with increasing the secretion of anti-inflammatory and pro-healing cytokines, thus providing a favorable osteoimmune environment for stem cells recruitment and osteogenic differentiation. In vivo , the P@PCH membrane effectively recruited host MSCs to the defect area, alleviated inflammatory infiltration, and accelerated bone defects repair. Collectively, our data indicated that the P@PCH nanocomposite membrane might be a promising biomaterial candidate for guided tissue regeneration in periodontal applications.

Abstract:Nanodrug delivery systems for the treatment of periodontitis remain a significant clinical challenge. Herein, combined with the pathological heterogeneity of periodontitis revealed by bioinformatics analysis, this study develops a novel cell membrane‐coated nanoparticle, which leverages the excellent natural properties of cell membrane‐coated nanotechnology and the versatility of nanoparticle cores, provides a promising strategy for the clinical treatment of periodontitis. Nanodrug delivery systems (NDDSs) for the treatment of periodontitis remain a significant clinical challenge. The low biocompatibility, singular effect, and lack of disease specificity of conventional nanoparticles limit their biomedical application. Recent studies have highlighted the pivotal role of cell membrane‐coated nanotechnology in anti‐inflammatory strategies due to its improved biocompatibility and superior biointerface properties. Herein, combined with the pathological heterogeneity of periodontitis revealed by bioinformatics analysis, this study develops a novel cell membrane‐coated nanoparticle composed of two elements: minocycline‐loaded polydopamine nanoparticles (PM) and cRGD‐modified cell membrane (RCM). The in vitro results indicate that PM@RCM rescues impaired human periodontal ligament stem cells through antioxidant, anti‐ferroptosis, anti‐inflammatory, and pro‐osteogenic effects, and exhibits favorable antibacterial bioactivity. The in vivo studies further reveal that PM@RCM promotes periodontal tissue regeneration and remodeled periodontal homeostasis in periodontitis mice. Collectively, these findings highlight the unique pathological changes in periodontitis. Moreover, the novel NDDS developed in this study, which leverages the excellent natural properties of cell membrane‐coated nanotechnology and the versatility of nanoparticle cores, provides a promising strategy for the clinical treatment of periodontitis.

Abstract:The periodontal tissue regeneration strategy based on guided tissue regeneration (GTR) membranes is an effective therapy for periodontal defects. Traditional GTR membranes, however, primarily serve as physical barriers and lack antimicrobial and osteogenic functions. Herein, we developed a multifunctional nanofiber membrane with zeolitic imidazolate framework-8 nanoparticles (ZIF-8 NPs) loaded in a hydrophilic gelatin layer. The release of Zn2+ from the ZIF-8 NPs effectively promoted bone tissue repair and simultaneously enabled GTR membranes with >99 antibacterial efficacies against Escherichia coli and Staphylococcus aureus. Additionally, the incorporation of gelatin enhances cellular adhesion and growth. Furthermore, in vivo studies revealed significant bone regeneration, with increased trabecular number and reduced separation. Owing to its multiple functions, excellent biocompatibility and desirable mechanical properties, this membrane has considerable potential in the field of periodontal therapy.
Preparing gelatin-containing polycaprolactone / polylactic acid nanofibrous membranes for periodontal tissue regeneration using side-by-side electrospinning technology

Abstract:Journal of Biomaterials Applications, Ahead of Print. Electrospinning technology has recently attracted increased attention in the biomedical field, and preparing various cellulose nanofibril membranes for periodontal tissue regeneration has unique advantages. However, the characteristics of using a single material tend to make it challenging to satisfy the requirements for a periodontal barrier film, and the production of composite fibrous membranes frequently impacts the quality of the final fiber membrane due to the influence of miscibility between different materials. In this study, nanofibrous membranes composed of polylactic acid (PLA) and polycaprolactone (PCL) fibers were fabricated using side-by-side electrospinning. Different concentrations of gelatin were added to the fiber membranes to improve their hydrophilic properties. The morphological structure of the different films as well as their composition, wettability and mechanical characteristics were examined. The results show that PCL/PLA dual-fibrous composite membranes with an appropriate amount of gelatin ensures sufficient mechanical strength while obtaining improved hydrophilic properties. The viability of L929 fibroblasts was evaluated using CCK-8 assays, and cell adhesion on the scaffolds was confirmed by scanning electron microscopy and by immunofluorescence assays. The results demonstrated that none of the fibrous membranes were toxic to cells and the addition of gelatin improved cell adhesion to those membranes. Based on our findings, adding 30% gelatin to the membrane may be the most appropriate content for periodontal tissue regeneration, considering the scaffold's mechanical qualities, hydrophilic properties and biocompatibility. In addition, the PCL-gelatin/PLA-gelatin dual-fibrous membranes prepared using side-by-side electrospinning technology have potential applications for tissue engineering.

Abstract:Barrier membranes have been used for the treatment of alveolar bone loss caused by periodontal diseases or trauma. However, an optimal barrier membrane must satisfy multiple requirements simultaneously, which are challenging to combine into a single material. We herein report the design of a bioinspired membrane consisting of three functional layers. The primary layer is composed of clay nanosheets and chitin, which form a nacre-inspired laminated structure. A calcium phosphate mineral layer is deposited on the inner surface of the nacre-inspired layer, while a poly(lactic acid) layer is coated on the outer surface. The composite membrane integrates good mechanical strength and deformability because of the nacre-inspired structure, facilitating operations during the implant surgery. The mineral layer induces the osteogenic differentiation of bone marrow mesenchymal stem cells and increases the stiffness of the membrane, which is an important factor for the regeneration process. The poly(lactic acid) layer can prevent unwanted mineralization on the outer surface of the membrane in oral environments. Cell experiments reveal that the membrane exhibits good biocompatibility and anti-infiltration capability toward connective tissue/epithelium cells. Furthermore, in vitro analyses show that the membrane does not degrade too fast, allowing enough time for bone regeneration. In vivo experiments prove that the membrane can effectively induce better bone regeneration and higher trabecular bone density in alveolar bone defects. This study demonstrates the potential of this bioinspired triple-layered membrane with hierarchical structures as a promising barrier material for periodontal guided tissue regeneration.

Abstract:Guided tissue regeneration (GTR) membranes have great potential to promote periodontal tissue regeneration and reestablishment. However, the regeneration potential and microbial infection resistance of current GTR membranes still need to be improved. Here, a bi-layered nanofibrous membrane on the basis of poly (lactic-co-glycolic acid) (PLGA)/gelatin with osteogenic and antibacterial functions was fabricated for periodontal tissue regeneration. The antimicrobial layer (AL) of the bi-layered nanofibrous membrane was composed of nanofibrous PLGA/gelatin nanofibers loaded with nano-silver (nAg), while the osteoconductive layer (OL) of the nanofibrous membrane consisted of PLGA/gelatin nanofibers loaded with nano-hydroxyapatite (nHA). The bi-layered nanofibrous membrane was examined by scanning electron microscopy (SEM), energy dispersive spectrometer (EDS), transmission electron microscope (TEM), Fourier transform infrared spectroscopy (FTIR), X-ray photoelectron spectrometry (XPS) and X-ray diffractometry (XRD). The results showed that nHA and nAg particles were well evenly loaded or embedded in PLGA/gelatin nanofibers. The cell culture experiments suggested that the bi-layered nanofibrous membrane possessed good cytocompatibility and the OL of the bi-layered nanofibrous membrane possessed an enhanced osteogenic capacity for human osteoblast-like cells (MG63), which was verified by the good cell viability and the increased alkaline phosphatase (ALP) activity, respectively. The results of in vitro antimicrobial study displayed that the AL of the bi-layered nanofibrous membrane possessed an effective antibacterial capability. In conclusion, the prepared bi-layered nanofibrous membrane with osteogenic and antibacterial functions may have great potential for periodontal tissue regeneration and reestablishment. Abstract:Presently, guided bone regeneration (GBR) stands as a preeminent strategy for rectifying periodontal and maxillofacial bone imperfections. Nonetheless, the current GBR membranes encounter limitations in terms of antibacterial efficacy and osteogenic potential, impeding their overall effectiveness. To address this, our study endeavors to engineer an innovative GBR membrane with dual attributes of osteoinductiveness and antibacterial activity. This novel asymmetric nanocomposite integrates an intricate structure comprising a compact chitosan hydrogel layer embedded with tetracycline (TCH) and a porous polycaprolactone fibrous layer incorporating chitosan nanoparticles loaded with icariin (IcA@CNps). In the initial phase, IcA@CNps, exhibiting a balanced CNps to IcA ratio of 1:2, an average dimension of 80.94 ± 33.78 nm, and a zeta potential of −19.3 mV, were synthesized and affixed onto nanofibers. This integration facilitated the controlled release of icariin over a span of 22 days. Subsequently, the asymmetric nanocomposite GBR membrane (referred to as GBR + ) was meticulously crafted. This involved the application of a TCH hydrogel laden with a drug concentration of 120 μg/ml onto the IcA@CNps-coated nanofibrous layer through an intermediary hydrogel stratum. Impressive results were witnessed during in vitro trials against Staphylococcus aureus , showcasing the remarkable antibacterial prowess of the GBR + membrane. Notably, human alveolar bone marrow stem cells (aBMSCs) cultivated on the GBR + membrane exhibited favorable cell adhesion and proliferation rates, devoid of any cytotoxic manifestations. Gene expression analyses further unveiled elevated expressions of SPP1, BMP2 , and RUNX2 in the proximity of the fabricated membrane, indicating the promotion of bone formation pathways. Consequently, this asymmetric GBR membrane presents a promising therapeutic avenue for bolstering bone regeneration, underscored by its dual action of fostering bone growth while concurrently displaying antibacterial attributes.
