Abstract:Freshwater microalgae are primary producers and cosmopolitan species subjected to the effects of herbicides. In this work, the in vitro algal growth inhibitory effects of 11 agrochemicals (9 herbicides, 1 metabolite, and 1 safener) were quantified. Chemical compounds were applied singly and in specific mixtures. Three species were used in axenic condition: the green alga Desmodesmus subspicatus (Chodat), the diatoms Nitzschia palea (Kützing) W. Smith and Navicula pelliculosa (Kützing) Hilse. When exposed to single compounds, N. palea and N. pelliculosa were only sensitive to atrazine/desethylatrazine and the safener benoxacor (BE), respectively. D. subspicatus was equally sensitive to four herbicides including atrazine and its metabolite and significantly more sensitive to iodosulfuron-methyl-sodium (IODO). The mixture of these five compounds induced a significantly higher growth inhibition of about 1.5-fold than IODO alone, which could be attributed to the four other herbicides. The mixture of all compounds was twofold less toxic than IODO on D. subspicatus . A halogen atom is present in IODO as in the herbicides to which the safener BE − known to induce glutathione- S -transferases − is associated in agrochemical preparations. We then showed that IODO was less toxic when combined with non-toxic concentrations of BE. These results indicated that the toxicity of the most active herbicide studied was decreased by a non-herbicide compound present in agrochemical formulations of other herbicides. These results suggest the importance to take into account the chemistry and the mechanisms of action for each compound in a risk assessment approach of a complex mixture.
Metabolomic analysis of Scenedesmus obliquus reveals new insights into the phytotoxicity of imidazolium nitrate ionic liquids

Abstract:Due to the persistence of ionic liquids (ILs) in aquatic environments, it is necessary to reveal their ecological risk to aquatic organisms. Herein, the biotoxicity of two alkyl-methylimidazolium nitrate ILs ([C10mim]NO3 and [C12mim]NO3) against Scenedesmus obliquus were studied. Results showed that the growth inhibition of S. obliquus increased with increasing concentrations of ILs, maximum values of 94.61% at 4 mg/L of [C10mim]NO3 and 97.34% at 0.8 mg/L of [C12min]NO3 were observed. The fluorescence parameters of photosystem II, such as light quantum yield and electron transfer rate, showed a negative relationship with the exposure dose. [C12mim]NO3 had a more significant effect than [C10mim]NO3. Moreover, the redox homeostasis of algae was disrupted; the accumulation of reactive oxygen species, leading to obvious inhibition of superoxide dismutase and catalase activities was observed. A metabolomic analysis indicated that the contents of most metabolites were reduced significantly, and fructose and galactose decreased significantly by 42.3% and 88.6%, respectively, in the [C10mim]NO3 treatment compared to those in the control. The inhibition of amino acid biosynthesis and glyoxylate and dicarboxylate metabolism explained the more serious biotoxicity of [C12mim]NO3 than that of [C10mim]NO3. This study facilitates a better understanding of the environmental safety and ecological risks of ILs.
Evaluation of the toxicity of herbicide topramezone to Chlorella vulgaris: Oxidative stress, cell morphology and photosynthetic activity.

Abstract:Topramezone is a new, highly selective herbicide of pyrazole structure for the post-emergence control of broadleaf and grass weeds in corn. In this study, the effects of topramezone on C. vulgaris, especially in relation to the cell growth, oxidative stress, cell morphology and photosynthetic activity were assessed. Results showed that topramezone treatment was detrimental to C. vulgaris growth during the 24-96h of exposure. The changes in cells pigments content and relative transcript of photosynthesis-related genes, which implies that topramezone disrupted the photosynthetic system. Moreover, topramezone induced membrane permeability in a significant proportion of cells with a maximum damage rate of 40.40%, and morphology of cells was more complicated than the control group. TEM images also revealed that topramezone compromised the integrity of the cells. The data corroborated topramezone induced ROS triggered oxidative stress, leading to an increase of MDA. These results suggested that topramezone could have significant effects on growth and physiological functions in algae species, and we supposed that this herbicide affected all of these parameters and the observed effects can be explained by the generation of oxidative stress. This research helps to understand how topramezone affects C. vulgaris and provides a scientific basis for applications of topramezone in aquatic environment.
Enantioselective Toxicity of Chiral Herbicide Metolachlor to Microcystis Aeruginosa

Abstract:The enantioselective effects of chiral herbicides on aquatic organisms have received increasing attention. As one kind of freshwater algae responsible for most algal blooms, Microcystis aeruginosa can produce hepatotoxic microcystin and cause serious health concerns for drinking water. Thus, the effects of chiral herbicides on M. aeruginosa are of vital significance but poorly understood, especially as the structures of chiral herbicides become more complex. In this study, the enantioselective effects of four metolachlor enantiomers based on carbon center and axis chirality on M. aeruginosa were investigated for the first time at an enantiomeric level. The results of the investigation into algal growth inhibition, chlorophyll a content, and cell integrity indicated that ( S)-metolachlor [( S)-Met] was significantly more toxic than any other isomer. The toxicity ranking of different enantiomers at the highest concentration (15 mg/L) against M. aeruginosa was ( S)-Met > (α R,1' S)-Met > (α S,1' S)-Met > (α S,1' R)-Met > (α R,1' R)-Met, with (α S,1' S)-Met and (α R,1' S)-Met displaying a synergistic effect. Additionally, the Fe distribution in M. aeruginosa presented distinct enantioselectivity, which may contribute to the enantioselective toxicity of metolachlor. Furthermore, metolachlor upregulated the expression of genes mcyD and mcyH in an enantioselective manner, indicating that this herbicide can potentially promote the synthesis and efflux of microcystin, thus aggravating agricultural water contamination to different extents. Overall, this study will help to understand the ecotoxicity of metolachlor at a deeper level and provide theoretical insights into the enantioselective behaviors of metolachlor.
Ecotoxicological evaluation of imazalil transformation products on Eisenia Andrei

Abstract:Data concerning the toxicity of the transformation products of some pesticides considered emerging contaminants are still incipient. This study aimed to evaluate acute (filter paper contact and avoidance test) and chronic (assays carried out in Red yellow Ultisoil) effects of the transformation products of the fungicide imazalil (IMZ) by heterogeneous photocatalysis (TiO2/UV) in Eisenia andrei. Some endpoints, i.e. biomass variation, disorder in reproduction and avoidance behavior, cytotoxicity (cell density and feasibility), and coelomic fluid (eleocytes and amoebocytes, immune cell) and antioxidant system (catalase (CAT) and glutathione S-transferase (GST)) changes were assessed. The studied degradation times (6, 18, 35 and 90 min) represented 70; 35; 10 and 0% of the initial IMZ concentration (5 mg L-1). No lethality and no significant difference in biomass variations were noted in the acute contact test. Decreased GST and increased CAT activities, as well as cell typing alterations and decreased cell viability and density detected at the 90-min degradation timepoint (PDT-90) indicate direct cytotoxic IMZ transformation product effects. In the avoidance test, no significant earthworm repulsion or attraction was observed. PDT-90 transformation products were responsible for losses in biomass and a reduction in the number of earthworm cocoons and juveniles present in soil. No cytotoxic effects were observed in the long term, although increased CAT and decreased GST activities were observed. Investigations on the relative toxicity of IMZ transformation products are relevant for future discussions regarding the environmental treatment, control and destination of this compound and its derivatives.
Impact of Microorganisms, Humidity, and Temperature on the Enantioselective Degradation of Imazethapyr in Two Soils.

Abstract:Imazethapyr (IM) is a chiral herbicide composed of an (-)-R-enantiomer and an (+)-S-enantiomer with differential herbicidal activity. In this study, the effects of microbial organisms, humidity, and temperature on the selective degradation of the (-)-R- and (+)-S-enantiomers of IM were determined in silty loam (SL) and clay loam (CL) soil with different pH values. The (-)-R-enantiomer of IM was preferentially degraded in two soils under different microorganism, humidity, and temperature conditions. The average half-lives of R-IM ranged from 43 to 66.1 days and were significantly shorter (P < 0.05) than those of S-IM, which ranged from 51.4 to 79.8 days. The enantiomer fraction (EF = (+)-S-enantiomer/((-)-R-enantiomer + (+)-S-enantiomer)) values were used to describe the enantioselectivity of degradation of IM were >0.5 (P < 0.05) in two unsterilized soils under different humidity and temperature conditions. The highest EF values were observed at unsterilized CL soil samples under 50% maximum water-holding capacity (MWHC) and 25 °C environmental conditions. The EF values of the IM enantiomers were significantly higher (P < 0.05) in CL soils (higher pH = 5.81) and were 0.581 (unsterilized) and 0.575 (50% MWHC; 25 °C) compared with those recorded in SL soil (lower pH = 4.85). In addition, this study revealed that microbial organisms preferentially utilized the more herbicidal active IM enantiomer.

Abstract:Assessing the interactions between environmental pollutants and these mixtures is of paramount significance in understanding their negative effects on aquatic ecosystems. However, existing research often lacks comprehensive investigations into the physiological and biochemical mechanisms underlying these interactions. This study aimed to reveal the toxic mechanisms of cyproconazole (CYP), imazalil (IMA), and prochloraz (PRO) and corresponding these mixtures on Auxenochlorella pyrenoidosa by analyzing the interactions at physiological and biochemical levels. Higher concentrations of CYP, IMA, and PRO and these mixtures resulted in a reduction in chlorophyll (Chl) content and increased total protein (TP) suppression, and malondialdehyde (MDA) content exhibited a negative correlation with algal growth. The activity of catalase (CAT) and superoxide dismutase (SOD) decreased with increasing azole fungicides and their mixture concentrations, correlating positively with growth inhibition. Azole fungicides induced dose-dependent apoptosis in A. pyrenoidosa , with higher apoptosis rates indicative of greater pollutant toxicity. The results revealed concentration-dependent toxicity effects, with antagonistic interactions at low concentrations and synergistic effects at high concentrations within the CYP-IMA mixtures. These interactions were closely linked to the interactions observed in Chl- a , carotenoid (Car), CAT, and cellular apoptosis. The antagonistic effects of CYP-PRO mixtures on A. pyrenoidosa growth inhibition can be attributed to the antagonism observed in Chl- a , Chl- b , Car, TP, CAT, SOD, and cellular apoptosis. This study emphasized the importance of gaining a comprehensive understanding of the physiological and biochemical interactions within algal cells, which may help understand the potential mechanism of toxic interaction.

Abstract:Quizalofop-Ethyl (QE) is a widely used phenoxy herbicide. It contains two enantiomers, and only its R-enantiomer quizalofop-p-ethyl (QpE) has herbicidal activity. Cyanobacterial blooms caused by Microcystis aeruginosa pose a threat to human health and aquatic environments, but the potential effects of chiral herbicides on M. aeruginosa are rarely studied. In this study, we carried out experiments using 0, 0.5, 1, 2, and 5 mg/L of QE and QpE to explore the effects on the growth, metabolism, synthesis, and release of microcystin-LR (MC-LR) in M. aeruginosa, which is the dominant species of algal blooms and indicator organism of water contamination. The biomass and chlorophyll a and protein content were consistent, showing that QpE at high concentrations (2 and 5 mg/L) showed greater acute toxicity than QE. On the expression of photosynthesis-related genes psaB and psbD1, QpE at 2 and 5 mg/L showed a stronger inhibition effect than QE. The oxidative stress caused by QE and QpE indicated that 144 h of exposure increased the concentration of MDA and enhanced the activities of superoxide dismutase and catalase. The synthesis of intracellular MC-LR in M. aeruginosa was stimulated by 144 h of QpE and QE exposure, and QpE showed a stronger stimulation effect than QE at high concentrations. The content of extracellular MC-LR increased after 144 h of QpE exposure and remained stable under QE exposure compared with that in the control group. Moreover, the expression of MC-LR-related gene mcyH was upregulated by QpE (144 h exposure) but downregulated by QE. These results are helpful to understand the toxicological effects of chiral herbicides on nontarget aquatic organisms, thus providing a theoretical basis for the rational use and the ecological risk assessment of chiral pesticides.

Abstract:The environmental load by isoxaflutole and its formulated herbicide products has increasingly become apparent because, after the ban of atrazine, isoxaflutole has become its replacement active ingredient (a.i.). Obtaining information regarding the fate of this a.i. in environmental matrices and its ecotoxicological effects on aquatic organisms is essential for the risk assessment of the herbicide. In this study, the effects of Merlin Flexx- and Merlin WG75 formulated isoxaflutole-based herbicide products and two selected additives (cyprosulfamide safener and 1,2-benzisothiazol-3(2H)-one antimicrobial agent) were investigated on Raphidocelis subcapitata in growth inhibition assays. In ecotoxicological tests, two conventional (optical density and chlorophyll-a content) and two induced fluorescence-based (Fv*/Fp: efficiency of the photosystem PSII and Rfd* changes in the observed ratio of fluorescence decrease) endpoints were determined by UV-spectrophotometer and by our FluoroMeter Module, respectively. Furthermore, dissipation of isoxaflutole alone and in its formulated products was examined by an HPLC-UV method. In ecotoxicological assays, the fluorescence-based Rfd* was observed as the most sensitive endpoint. In this study, the effects of the safener cyprosulfamide and the antimicrobial agent 1,2-benzisothiazol-3(2H)-one on R. subcapitata is firstly reported. The results indicated that the isoxaflutole-equivalent toxicity of the mixture of the isoxaflutole–safener–antimicrobial agent triggered lower toxicity (EC50 = 2.81 ± 0.22 mg/L) compared to the individual effect of the a.i. (EC50 = 0.02 ± 0.00 mg/L). The Merlin Flexx formulation (EC50 = 27.04 ± 1.41 mg/L) was found to be approximately 50-fold less toxic than Merlin WG75, which can be explained by the different chemical characteristics and quantity of additives in them. The additives influenced the dissipation of the a.i. in Z8 medium, as the DT50 value decreased by approximately 1.2- and 3.5-fold under light and dark conditions, respectively.

Abstract:Ivermectin (IVM) is a macrocyclic lactone widely used to control endo- and ectoparasites in livestock. After administration, it is only partially metabolized by the animal and therefore excreted in its original form. IVM can enter water bodies through groundwater, runoff, soil erosion, and direct deposition. Once in aquatic and wetland environments, due to its chemical characteristics, it can persist for a long time, increasing its environmental risk. Macrophytes are in frequent contact with this drug, resulting in chronic exposure and leading to an accumulation process. The objective of this study is to evaluate the uptake of IVM in S. minima , and its phytotoxicity potential. Bioassays were performed by exposing S. minima to different concentrations of IVM, 5 mg/L, 10 mg/L, and 30 mg/L. After 10 days, the accumulation of the compound in fronds, roots, and effluent was measured. Morphological parameters and photosynthetic pigments were evaluated. IVM was found in fronds and roots of S. minima after exposure. The percentage removal of the drug in effluent was significant, up to 66%. Only the highest concentration evaluated caused phytotoxic effects. S. minima proved to be a promising species for IVM removal processes, especially in wetlands subject to intensive livestock farming activities. Of interest is its applicability in wetlands subjected to intensive livestock farming.

Abstract:Perfluorooctanoic acid (PFOA) and microcystin-LR (MCLR) are pervasive pollutants in surface waters that induce significant toxic effects on aquatic organisms. However, the combined environmental risk of PFOA and MCLR remains unclear. To assess the toxic effects of PFOA and MCLR on submerged macrophytes and biofilms, Vallisneria natans was exposed to different concentrations of PFOA and MCLR (0.01, 0.1, 1.0 and 10.0 μg L −1 ). Vallisneria natans was sensitive to high concentrations of MCLR (10 μg L −1 ): plants exposed to 10 μg L −1 of MCLR measured a biomass of 3.46 g, which was significantly lower than the 8.71 g of the control group. Additionally, antagonistic interactive effects were observed in plants exposed to combined PFOA and MCLR. Exposure to these pollutants adversely affected photosynthesis of the plants and triggered peroxidation that promoted peroxidase, superoxide dismutase and catalase activities, and increased malondialdehyde and glutathione concentrations. The total chlorophyll content was lower in the highest concentration of the combined treatment group (0.443 mg g −1 ) than in the control group (0.534 mg g −1 ). Peroxidase activity increased from 662.63 U mg −1 Pr to 1193.45 U mg −1 Pr with increasing PFOA concentrations. Metabolomics indicated that the stress tolerance of Vallisneria natans was improved via altered fatty acid metabolism, hormone metabolism and carbon metabolism. Furthermore, PFOA and MCLR influenced the abundance and structure of the microbial community in the biofilms of Vallisneria natans . The increased contents of autoinducer peptide and N -acylated homoserine lactone signaling molecules indicated that these pollutants altered the formation and function of the biofilm. These results expand our understanding of the combined effects of PFOA and MCLR in aquatic ecosystems.

Hormesis, the Individual and Combined Phytotoxicity of the Components of Glyphosate-Based Formulations on Algal Growth and Photosynthetic Activity

Abstract:The occurrence of the market-leading glyphosate active ingredient in surface waters is a globally observed phenomenon. Although co-formulants in pesticide formulations were considered inactive components from the aspects of the required main biological effect of the pesticide, several studies have proven the high individual toxicity of formulating agents, as well as the enhanced combined toxicity of the active ingredients and other components. Since the majority of active ingredients are present in the form of chemical mixtures in our environment, the possible combined toxicity between active ingredients and co-formulants is particularly important. To assess the individual and combined phytotoxicity of the components, glyphosate was tested in the form of pure active ingredient (glyphosate isopropylammonium salt) and herbicide formulations (Roundup Classic and Medallon Premium) formulated with a mixture of polyethoxylated tallow amines (POEA) or alkyl polyglucosides (APG), respectively. The order of acute toxicity was as follows for Roundup Classic: glyphosate < herbicide formulation < POEA. However, the following order was demonstrated for Medallon Premium: herbicide formulation < glyphosate < APG. Increased photosynthetic activity was detected after the exposure to the formulation (1.5–5.8 mg glyphosate/L and 0.5–2.2 mg POEA/L) and its components individually (glyphosate: 13–27.2 mg/L, POEA: 0.6–4.8 mg/L), which indicates hormetic effects. However, decreased photosynthetic activity was detected at higher concentrations of POEA (19.2 mg/L) and Roundup Classic (11.6–50.6 mg glyphosate/L). Differences were demonstrated in the sensitivity of the selected algae species and, in addition to the individual and combined toxicity of the components presented in the glyphosate-based herbicides. Both of the observed inhibitory and stimulating effects can adversely affect the aquatic ecosystems and water quality of surface waters.

Abstract:Keratinocytes are a rich source of IL-1, a cytokine which stimulates prostaglandin synthesis in many cell types. The effects on arachidonic acid metabolism of this cytokine were therefore studied in cultured adult human keratinocytes. Exogenous IL-1 increased basal cellular prostaglandin synthesis (particularly PGE2) threefold. Increased PGE2 synthesis in response to IL-1 was inhibited by cycloheximide, suggesting a requirement for new protein synthesis. Irradiation of the keratinocytes with low-dose ultraviolet light B (UVB) resulted in the release of increased quantities of both IL-1 and PGE2. The amount of IL-1 released was sufficient to increase PGE2 synthesis when exogenously added to unstimulated cells, suggesting a causal relationship. The time course of accumulation of IL-1 and PGE2 in the medium of irradiated keratinocytes was also consistent with a cause-effect relationship. No feedback inhibition of IL-1 release by the increased PGE2 was detected as demonstrated by the observation that IL-1 production in response to UVB was not augmented by treatment with indomethacin or blunted by the exogenous addition of PGE2. These data suggest that keratinocyte IL-1 may be partially responsible for induction of keratinocyte PGE2 synthesis after UVB irradiation.

Abstract:This study investigated the effects of herbicide exposure on Navicula sp. (MASCC-0035) algae, focusing on growth density, chlorophyll content, antioxidant system, and lipid metabolism. Navicula cultures were exposed to different concentrations of atrazine (ATZ), glyphosate (Gly), and acetochlor (ACT) for 96 h. Results showed a significant decrease in cell numbers, with higher herbicide concentrations having the most noticeable impacts. For instance, Gly-G2 had reduced cell populations by 21.00% at 96 h. Chlorophyll content varied, with Gly having a greater impact on chlorophyll a compared to ATZ and ACT. Herbicide exposure also affected the antioxidant system, altering levels of soluble sugar, soluble protein, and reactive oxygen species (ROS). Higher herbicide rates increased soluble sugar content (e.g., ATZ, Gly, and ACT-G2 had increased by 14.03%, 19.88%, and 19.83%, respectively, at 72 h) but decreased soluble protein content, notably in Gly-G2 by 11.40%, indicating cellular stress. Lipid metabolism analysis revealed complex responses, with changes in free proline, fatty acids, and lipase content, each herbicide exerting distinct effects. These findings highlight the multifaceted impacts of herbicide exposure on Navicula algae, emphasizing the need for further research to understand ecological implications and develop mitigation strategies for aquatic ecosystems.

Abstract:Pesticides are chemical substances that have become essential for global food production. However, their extensive use has resulted in significant environmental and health costs. Among the most widely used are diazinon and methamidophos, an insecticide and acaricide, respectively. These chemicals are classified as highly hazardous with high toxicity upon contact, ingestion, and inhalation. They are known to cause mutagenic effects, cancer, damage to the endocrine and nervous systems of humans, and their high solubility makes them likely to be found in soil, water, air, and certain foods. In this research, we evaluated the use of the microalga Chlorella vulgaris for phycoremediation of these two pesticides. The first step was to establish and optimize the growth parameters of C. vulgaris to maintain the microalgae in a state of reproduction. Favorable results were obtained at pH 7.0, a dilution ratio of 1:2 for the culture medium, and a photoperiod of 12 h of light and 12 h of darkness. Subsequently, the Minimum Inhibitory Concentration (MIC) and the Half-Maximal Effective Concentration (EC 50 ) of the pesticides diazinon and methamidophos were evaluated. The microalga exhibited a high tolerance rate, with concentrations exceeding those naturally found in the environment. Based on these results, the conditions for assessing the biodegradation of both pesticides by the microalga were established. The outcome showed degradation percentages of 96 % for diazinon (25 mg L −1 ) and 100 % for methamidophos (500 mg L −1 ). In addition, the analysis of intermediate metabolites during the biodegradation process of both pesticides revealed the formation of less toxic compounds compared to the precursors and possibly complete mineralization as well; this given the positive response in terms of inorganic phosphate generation during the degradation. Finally, we propose biodegradation pathways for methamidophos and diazinon based on the detected metabolites and available information on biodegradation by certain bacteria.

Abstract:Microcystis aeruginosa is one of the most predominant freshwater bloom-forming cyanobacterium found globally which is capable of producing toxic secondary metabolites including microcystins that might intoxicate animals and humans when contaminated water or food is ingested. Salvinia auriculata Aubl is one of the plants that might possess bioactive compounds capable of controlling growth and reproduction of M. aeruginosa . The present study aimed to determine the presence of bioactive compounds in S. auriculata extracts and determine alterations occurred in growth and reproduction of M. aeruginosa when exposed to these plant extracts. In addition, this investigation aimed to examine the influence of S. auriculata on antioxidant enzymes detected in M. aeruginosa . The results obtained demonstrated that the aqueous and ethanolic extracts of S. auriculata presented potential for control of cyanobacteria populations, exhibiting algicidal action on M. aeruginosa as well as interfering in antioxidant enzymes activities and parameters associated with oxidative stress. Phytochemical analyses demonstrated the presence of polyphenols and flavonoids content in both extracts. In addition, application of S. auriculata extracts did not produce cytogenotoxicity and/or mutagenicity utilizing Allium cepa test. Therefore, further studies are needed in order to identify and characterize the compounds responsible for these effects on M. aeruginosa and provide information regarding the possible application of S. auriculata in the treatment of drinking water.

Effects of Imazapyr on Spartina alterniflora and Soil Bacterial Communities in a Mangrove Wetland

Abstract:The invasion of Spartina alterniflora (S. alterniflora) has caused serious damage to coastal wetland ecosystems in China, especially the mangrove wetlands in South China. This study aimed to validate the effect of imazapyr on S. alterniflora and soil. The controlled experiment was conducted in May 2021 at the Zhangjiangkou Mangrove Wetland Reserve. In the experiment, 25% (W) imazapyr was used, and six treatments were set up: 3035, 6070, and 9105 mL/acre 25% imazapyr and 1299, 2604, and 5202 mL/acre of AGE 809 + 6070 mL/acre 25% imazapyr. The results showed no side effects on mangrove plants in the spraying area. The highest control efficiency (95.9%) was given by 2604 mL/acre of AGE 809 + 6070 mL/acre 25% imazapyr. The residues of imazapyr in different soils were reduced to 0.10–0.59 mg/kg. The sequencing results showed no significant difference in the overall bacterial communities under different treatments (p > 0.05). The soil bacterial diversity in the samples with adjuvant was higher than that in the samples without adjuvant, while the abundance values were the opposite. There were 10 main communities (>0.3%) at phylum level in all soil samples, among which Proteobacteria, Bacteroidetes, Acidobacteria, Chloflexi, and Actinobacteria were the dominant communities, and the latter four’s abundance changed significantly (p < 0.05). There were significant abundance differences between the groups of oligotrophic and eutrophic bacteria. The redundancy analysis and Monte Carlo tests showed that the total organic carbon (TOC), total phosphorus (TP), available phosphorus (AP), ammonia nitrogen, and total nitrogen were the main factors affecting soil bacterial diversity. At the same time, TOC, AP, and TP were the most critical factors affecting the overall characteristics of soil bacterial communities in different treatments, while soil residues had no significant effect on bacteria. This might be due to the addition and degradation of imazapyr and the coverage of S. alterniflora. The best recommendation is 2604 mL/acre of AGE 809 + 6070 mL/acre 25% imazapyr to be applied in China’s mangrove wetland reserves and coastal wetlands.

Abstract:Greater exposure to herbicide increases the likelihood of harmful effects in humans and the environment. Glufosinate, a non-selective herbicide, inhibits glutamine synthetase (GS) and thus blocks ammonium assimilation in plants. In the present study, the aquatic unicellular alga Chlorella vulgaris was chosen to assess the effects of acute glufosinate toxicity. We observed physiological changes during 12–96h of exposure, and gene transcription during 6–48h of exposure. Exposure to glufosinate increased malondialdehyde content by up to 2.73 times compared with the control, suggesting that there was some oxidative damage. Electron microscopy also showed that there were some chloroplast abnormalities in response to glufosinate. The activities of the antioxidant enzymes superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) also increased markedly in the presence of glufosinate. Maximum activities of SOD, POD, and CAT were 2.90, 2.91, and 2.48 times that of the control, respectively. These elevated activities may help alleviate oxidative damage. A real-time polymerase chain reaction (PCR) assay showed changes in transcript abundances of three photosynthetic genes, psaB, psbC, and rbcL. The results showed that glufosinate reduced the transcript abundances of the three genes after 12h exposure. The lowest abundances of psaB, psbC and rbcL transcripts in response to glufosinate exposure were 38%, 16% and 43% of those of the control, respectively. Our results demonstrate that glufosinate affects the activities of antioxidant enzymes, disrupts chloroplast ultrastructure, and reduces transcription of photosynthesis-related genes in C. vulgaris.

Abstract:We investigated the phytotoxic effects of volatile and water soluble chemicals produced by the shrub Artemisia herba-alba Asso. We conducted a number of germination and early seedling growth bioassays on species that co-exist with A. herba-alba in natural semiarid plant communities (Salsola vermiculata L. Lygeum spartum L. Pinus halepensis Mill. and A. herba-alba itself). In addition, we assessed the phytotoxic effects of a mixture of phenols that were identified in the aqueous extract of A. herba-alba on the germination of such species. We found that volatile chemicals inhibited the germination of P. halepensis seeds, promoted the growth of P. halepensis seedlings and reduced the root biomass of S. vermiculata seedlings, while water soluble chemicals promoted the growth of L. spartum and P. halepensis seedlings. On the other hand, both volatile and water soluble compounds inhibited the germination of A. herba-alba seeds, highlighting its strong autotoxic nature. We did not find a phytotoxic effect of phenols identified (catechol, protocatechuic and vanillic acids) on species that co-exists with A. herba-alba, although they had a moderate autotoxic effect. Our results suggest that both the volatile and water soluble chemicals might be involved in chemical interactions among plants in semiarid environments.
