Abstract:Psyllium seed mucilage is a gel forming therapeutically important dietary fiber polysaccharide. In present work, an attempt has been made to synthesize arabinoxylan psyllium mucilage-2-methacryloyl-oxyethyl trimethylammonium chloride copolymeric hydrogel by using high energy gamma radiation for drug delivery applications. Various thermal, mechanical, physicochemical, biomedical properties of the hydrogel were determined along with characterization with FTIR, XRD, TGA, DSC and AFM techniques. The release of antibiotic drug cefuroxime occurred in sustained manner with Fickian diffusion from the crosslinked polymer and release profile was best fitted in Higuchi kinetic model. It is inferred from the results of crosslinking density, simulated fluid sorption, hemolytic potential, antioxidant and mucoadhesion properties that the crosslinked psyllium mucilage could be explored as a potential drug delivery carrier for gastrointestinal tract.

Abstract:Keeping in view the therapeutic and gel forming role of the psyllium seed mucilage in pharmaceutical industries, herein this article, its potential has been explored for developing the drug delivery (DD) carrier. The main aim of the study was to modify the psyllium polysaccharides through grafting and crosslinking of 2-methacryloyloxyethyl trimethylammonium chloride onto psyllium to develop the hydrogels for use in DD application. Various thermal, mechanical, physiochemical, biomedical properties of the hydrogel were determined along with characterization with 13C NMR, FTIR, XRD, TGA, DSC and AFM. The release of antibiotic drug cefuroxime occurred in sustained manner and followed Fickian diffusion mechanism and was best fitted in Higuchi kinetic model. The networks showed mesh size in the range between 25.14 to 38.92 nm during the change in crosslinker content in copolymerization reaction. It has been inferred from the results that these hydrogels could be used as a potential carrier for drug delivery.

Abstract:The dietary fiber sterculia gum is a therapeutic important polysaccharide comprising of glucuronic–galacturonic acid. In the present work, it has been explored for the development of the crosslinked polymeric network hydrogels with phosphated polymers for use in drug delivery applications. Polymers were characterized by SEM, FTIR, XRD, 13 C NMR, TGA-DSC and swelling studies. SEM images of hydrogels showed porous structural heterogeneity. The incorporation of phosphate polymers enhanced the thermal stability of the gum. The swelling of sterculia and phosphate polymers are pH responsive and release of cefuroxime from the polymers occurred slowly with non-Fickian diffusion best fitted in first order kinetic model. The hydrogels showed haemolytic index value < 1 and mucoadhesion with intestinal mucosa = 0.498 ± 0.07 N. These results indicated that these cefuroxime loaded phosphate containing sterculia hydrogels can be explored for the drug delivery applications

[Oxprenolol in the treatment and prevention of cardiac arrhythmias].

Abstract:Authors have analyzed the antiarrhythmic effect of oxprenolol, a beta blocking agent, in the treatment of various types of arrhythmias and in the prophylaxis of recurrences of auricular flutter and fibrillation. The results obtained in a group of 68 cases of different arrhythmias may be summarized in the following way: a) the drug efficiently reduces the ventricular rate in sinus tachycardia and in atrial flutter and fibrillation with high ventricular rate, even if resistant to treatment with digitalis; b) in patients with asynchronous pacemaker oxprenolol leads to disappearance or an important reduction of competitive rhythms; c) in supraventricular paroxysmal tachycardias the results are positive in the majority of cases (while on the contrary in 2 cases of ventricular tachycardias the drug was not effective). A group of 116 cases with auricular flutter or fibrillation (in which sinus rhythm had been restored with quinidine or cardioversion has been analyzed to study the prophylactic activity of oxprenolol in these arrhythmias. The cases have been divided at random into two groups and have been treated with quinidine (g 0.80 p.d.) or with an association of oxprenolol (mg 60 p.d.) and quinidine (g 0,60 p.d.). The observation period varied from a minimum of 1 month to a maximum of 3 years and 3 months. The curves showing the percentage of persistance of sinus rhythm in the two groups were very similar and after 3 years and 3 months 100% of patients observed presented a recurrence of arrhythmias.

Abstract:Abstract In view of the pharmacological importance of dietary fibre, psyllium, to cure the constipation and diverticulitis, in the present study, an attempt has been made to modify psyllium polysaccharide with PVP to develop the hydrogel meant for slow and controlled drug delivery systems. The polymer was characterized by SEMs, FTIR, XRD, TGA and swelling studies. Swelling of hydrogels and drug (ciprofloxacin) release profile from the drug loaded hydrogels were determined for the evaluation of the swelling/release mechanism. Biomedical properties; biocompatibility and mucoadhesion of the hydrogels, were also studied. Swelling of the hydrogels and release of drugs from drug loaded hydrogels occurred through non-Fickian diffusion mechanism. Here it is pertinent to mention that both psyllium husk polysaccharide and antibiotic drug ciprofloxacin are used for gastrointestinal tract (GIT) problem, especially in case of diverticulitis. Hence, degradation of the polymer matrix and release of drug may exert the synergic effect and the present drug delivery system may act with enhanced potential.

Abstract:In order to improve the pharmacokinetic deficiencies and bioavailability of therapeutic agent used in the upper gastro intestinal tract (GIT), herein this article gastro-retentive drug delivery system (DDS) were developed via green approach from medicinally important polysaccharides psyllium, moringa gum and alginate. The antibiotic drug ciprofloxacin was impregnated into DDS. These polymeric DDS were characterized by cryo-SEMs, FTIR, 13 C NMR and XRD analysis. The simulated gastric fluid uptake was reduced with rise in crosslinking during ionotropic gelation. The diffusion of ciprofloxacin was non-Fickian and it was fitted in Korsmeyer-Peppas model. The porous nature of the crosslinked network hydrogel beads was observed in cryo SEM images. XRD indicated the amorphous nature of the hydrogel beads. The haemolytic index value of beads was observed less than 5%. The results of the polymer interactions with blood indicated the non-hemolytic nature of the polymers with their suitability for controlled drug delivery applications. The antioxidant property indicated 64.03 ± 0.29% DPPH radicals scavenging assay of the beads. Overall, anti-ulcer activity of the DDS for gastric ulceration could be enhanced due to slow release of ciprofloxacin drug from drug delivery carrier which itself has antiulcer therapeutic potential due to composed dietary fibers psyllium-moringa gum-alginate.

Abstract:The work was intended to develop a novel acrylic acid grafted hydrogel by chemical crosslinking of xanthan gum and starch under microwave irradiation. The swelling capacity of hydrogel was found to be dependent upon pH. The maximum swelling capacity of hydrogel was recorded as 32.21 g/g under the optimized conditions. The swelling capacity of hydrogel was quite higher than most of the hydrogels containing xanthan gum mentioned in the literature. Various characterization techniques including FTIR, SEM, TGA and XRD confirmed successful synthesis of hydrogel with porous morphology and better thermal stability. Synthesized hydrogel was employed as an oral drug delivery vehicle. Releasing behavior of the hydrogels for the drugs aspirin and paracetamol was studied under specific physiological conditions. The drug release was significantly higher at pH 7.4 in comparison to acidic and neutral media. Synthesized hydrogel was found to be suitable for colon-specific drug delivery. Both aspirin and paracetamol followed non-Fickian diffusion mechanism at higher pH and Fickian mechanism at lower pH. Release profiles of both the drugs were best fitted in the first order model. Hydrogel was found to be non-cytotoxic to human fibroblast cells and biocompatible, with a low hemolytic ratio. Consequently results supported potential of this non-toxic, biofriendly and appropriately tailored polysaccharide based hydrogel to be used as drug delivery carrier for controlled and site-specific drug release.

Abstract:Tragacanth gum (TG) is generally recognized as safe by the Food and Drug Administration. The present article discusses the design of ciprofloxacin loaded TG based hydrogels for use in drug delivery especially to improve the pharmacotherapy of diverticulitis. The polymers were characterized by SEMs, FTIR, 13C NMR, XRD, TGA, DSC, gel strength and swelling studies. The polymer network parameters, mucoadhesion, gel strength, drug release mechanism and kinetic model were also determined. The release of drug occurred through non-Fickian diffusion mechanism and best fitted in the Korsmeyer-Peppas model. The pH of the swelling medium has also exerted a strong effect on polymer network structure and mechanical strength. These hydrogels have been observed pH responsive and mucoadhesive in nature and could be utilized for site specific drug delivery.
Designing protein-polysaccharides based bioactive copolymeric network by supra-molecular interactions for sustained drug delivery

Abstract:Keeping in view recent advancements in designing biomaterials from bioactive polysaccharides, the present work deals with the design of protein (gelatin)-polysaccharide (tragacanth gum) based bioactive copolymeric network by supramolecular interactions and covalent linkage for sustained drug delivery (DD) applications. The network copolymeric structure was characterized by field emission-scanning electron micrographs (FE-SEM), electron dispersion X-ray analysis (EDAX), Fourier transform infrared spectroscopy (FTIR), 13C-nuclear magnetic resonance (NMR), and X-ray diffraction (XRD). FESEM and XRD analyses revealed the heterogeneous morphology of copolymers with an amorphous nature. 13C NMR and FTIR spectra demonstrated the incorporation of poly(acrylamide) (AAm) into network structure by copolymerization reaction. Diffusion of anticancer drug 5-flurouracil (5-FU) occurred in a sustained manner with the Fickian mechanism and was best fitted in first order kinetic model. Polymer-blood interactions revealed the non-hemolytic character of hydrogels. An antioxidant assay evaluated their antioxidant property (26.61 ± 0.85 % of free radical scavenging). Copolymers exhibited mucoadhesiveness during polymer-mucous membrane interactions and required 113.33 ± 5.68 mN detachment forces. Furthermore, the combination of polysaccharide-gelatin has enhanced supramolecular interactions and improved physiological and biomedical properties of network hydrogels. Overall, these properties revealed the suitability of copolymeric hydrogels for drug delivery applications.

Abstract:The present work deals with the evaluation of the physiochemical and biomedical properties of hydrogels derived from copolymerization of tragacanth gum (TG) and gelatin for use in drug delivery (DD) applications. Copolymers were characterized by field emission-scanning electron micrographs (FE-SEM), electron dispersion X-ray analysis (EDAX), Fourier transform infrared spectroscopy (FTIR), 13 C-nuclear magnetic resonance (NMR), thermo-gravimetric analysis (TGA), differential scanning calorimetry (DSC) and X-ray diffraction (XRD) analysis. FE-SEM revealed heterogeneous morphology and XRD analysis demonstrated an amorphous nature with short range pattern of polymer chains within the copolymers. The release of the drug ofloxacin occurred through a non-Fickian diffusion mechanism and the release profile was best described by the Korsmeyer-Peppas kinetic model. The hydrogels exhibited blood compatibility and demonstrated a thrombogenicity value of 75.63 ± 1.98 % during polymer-blood interactions. Polymers revealed mucoadhesive character during polymer-mucous membrane interactions and required 119 ± 8.54 mN detachment forces to detach from the biological membrane. The copolymers illustrated the antioxidant properties as evidenced by 2, 2′-diphenylpicrylhydrazyl (DPPH) assay which demonstrated a 65.71 ± 3.68 % free radical inhibition. Swelling properties analysis demonstrated that by change in monomer and cross linker content during the reaction increased the crosslinking of the network. These results suggest that the pore size of network hydrogels could be controlled as per the requirement of DD systems. The copolymers were prepared at optimized reaction conditions using 14.54 × 10 −1 molL −1 of acrylic acid monomer and 25.0 × 10 −3 molL −1 of crosslinker NNMBA. The optimized hydrogels exhibited a crosslink density of 2.227 × 10 −4 molcm −3 and a mesh size of 7.966 nm. Additionally, the molecular weight between two neighboring crosslinks in the hydrogels was determined to be 5332.209 gmol −1 .The results indicated that the combination of protein-polysaccharide has led to the development of hydrogels suitable for potential applications in sustained drug delivery.

Abstract:Recently, various efforts have been made to explore the potential of natural polysaccharides derived from sea weeds to promote sustainable development. Herein, carrageenan (CG), a polysaccharide extracted from red sea algae, was utilized to design network structures as hydrogels, aimed at significant applications in drug delivery (DD) systems. Hydrogels were designed by graft copolymerization reaction of poly(bis [2-methacryloyloxy] ethyl phosphate [poly(BMEP)] and poly(acrylic acid) [poly(AAc)] onto CG in the presence of a crosslinking agent. Hydrogels were developed by covalent linkage by graft copolymerization and supramolecular interactions, existing in the copolymers. Copolymers were characterized by Atomic force microscopy (AFM), Fourier transform infrared spectroscopy (FTIR), 13C-nuclear magnetic resonance (NMR), and X-ray diffraction (XRD) instrumentations. The drug diffusion exhibited a sustained pattern due to polymer-drug interactions. The drug release followed non-Fickian diffusion mechanism and the release profile was most accurately depicted by first order kinetic model. The biocompatible nature of the copolymer was demonstrated from the hemolytic index value signifying minimal adverse interactions with blood component upon exposure. A protein adsorption test was performed using bovine serum albumin (BSA), exhibiting 8.15 ± 0.26 % albumin adsorption. Polymers exhibited mucoadhesive character, evidenced by their requirement of a detachment force measuring 195 ± 4.72 mN for separation from the membrane during interactions with the mucosal surface. The hydrogels exhibited antioxidant properties, evidenced by 2, 2'-Diphenylpicrylhydrazyl (DPPH) assay, revealing copolymer capable of scavenging 58.21 ± 2.26 % of free radical. The hydrogel revealed antimicrobial activity against P. aeruginosa and S. aureus bacteria, a property further enhanced in hydrogels with the drug doxycycline. These findings suggest suitability of these hydrogels for biomedical applications, with a significant emphasis on drug delivery.

Abstract:In this comprehensive investigation, a novel pH-responsive hydrogel system comprising mimosa seed mucilage (MSM), β-cyclodextrin (β-CD), and methacrylic acid (MAA) was developed via free radical polymerization technique to promote controlled drug delivery. The hydrogel synthesis involved strategic variations in polymer, monomer, and crosslinker content in fine-tuning its drug-release properties. The resultant hydrogel exhibited remarkable pH sensitivity, selectively liberating the model drug (Capecitabine = CAP) under basic conditions while significantly reducing release in an acidic environment. Morphological, thermal, and structural analyses proved that CAP has a porous texture, high stability, and an amorphous nature. In vitro drug release experiments showcased a sustained and controlled release profile. Optimum release (85.33 %) results were recorded over 24 h at pH 7.4 in the case of MMB9. Pharmacokinetic evaluation in healthy male rabbits confirmed bioavailability enhancement and sustained release capabilities. Furthermore, rigorous toxicity evaluations and histopathological analyses ensured the safety and biocompatibility of the hydrogel. This pH-triggered drug delivery system can be a promising carrier system for drugs involving frequent administrations.

Abstract:The dietary fibers psyllium-sterculia gum have therapeutic roles in diarrhea, therefore, herein this work, hydrogels were prepared for use as drug delivery devices (DDD) for ornidazole. Grafted copolymers were characterized with SEM, FTIR, EDAX and swelling studies. The drug release was observed in sustained manner with Fickian diffusion. Overall, these hydrogels can act as DDD with enhanced potential due to therapeutic role of psyllium-sterculia and slow release of ornidazole.

Abstract:AIM To verify whether arterial-phase contrast-enhanced ultrasonography (CEUS) of tumor parenchymal tissue is useful for evaluation of anti-angiogenesis agents. METHODS Rabbits with liver tumor were subjected to CEUS, and images of the nodular maximal diameter in vascular phase were recorded. Image analysis was performed to plot the time intensity curve (TIC) at the tumor parenchyma, which set the diameter of the region of interest of intensity measurement. The TIC was calculated to obtain the time to peak intensity (TPI) and the magnitude of PI. Rabbits were randomly assigned to a treatment group with sorafenib and a control group. Two weeks later, the same ultrasound examination was repeated followed by pathological testing to assess the effect of sorafenib on the liver tumor. RESULTS In four rabbits in the treatment group, the rate of change of tumor size was decreased compared with that of the control (the rate 2.3 vs 7.9, P = 0.02). The TPI of the treatment group elongated significantly (the rate 3.1 vs 1.1, P = 0.07 for SonoVue, 2.0 vs 0.88, P = 0.09 for Sonazoid). The magnitude of PI showed no significant changes. In pathological examination, capillary diameters in the treatment group were significantly smaller than those in the control group (26.4 vs 42.8 μm, P = 0.013). CONCLUSION Analysis of the TIC in the arterial phase of tumor tissue could evaluate the efficacy of anti-angiogenesis drug treatment in liver tumor.
Cydonia oblonga-Seed-Mucilage-Based pH-Sensitive Graft Copolymer for Controlled Drug Delivery-In Vitro and In Vivo Evaluation

Abstract:The primary objective of this study was to assess the potential utility of quince seed mucilage as an excipient within a graft copolymer for the development of an oral-controlled drug delivery system. The Cydonia oblonga-mucilage-based graft copolymer was synthesized via a free radical polymerization method, employing potassium per sulfate (KPS) as the initiator and N, N-methylene bisacrylamide (MBA) as the crosslinker. Various concentrations of monomers, namely acrylic acid (AA) and methacrylic acid (MAA), were used in the graft copolymerization process. Metoprolol tartarate was then incorporated into this graft copolymer matrix, and the resultant drug delivery system was subjected to comprehensive characterization using techniques such as Fourier-transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM). The swelling behavior of the drug delivery system was evaluated under different pH conditions, and in vitro drug release studies were conducted. Furthermore, pharmacokinetic parameters including the area under the curve (AUC), maximum plasma concentration (Cmax), time to reach Cmax (Tmax), and half-life (t1/2) were determined for metoprolol-loaded hydrogel formulations in rabbit plasma, and these results were compared with those obtained from a commercially available product. The key findings from the study include observations that higher concentrations of acrylic acid (AA) and Cydonia oblonga mucilage (CM) in the graft copolymer enhanced swelling, while the opposite trend was noted at elevated concentrations of methacrylic acid (MAA) and N, N-methylene bisacrylamide (MBA). FTIR analysis confirmed the formation of the graft copolymer and established the compatibility between the drug and the polymer. SEM imaging revealed a porous structure in the prepared formulations. Additionally, the swelling behavior and drug release profiles indicated a pH-sensitive pattern. The pharmacokinetic assessment revealed sustained release patterns of metoprolol from the hydrogel network system. Notably, the drug-loaded formulation exhibited a higher Cmax (156.48 ng/mL) compared to the marketed metoprolol product (96 ng/mL), and the AUC of the hydrogel-loaded metoprolol was 2.3 times greater than that of the marketed formulation. In conclusion, this study underscores the potential of quince seed mucilage as an intelligent material for graft-copolymer-based oral-controlled release drug delivery systems.

Using N, N methylene bisacrylamide as a crosslinker and ammonium persulfate as an initiator, the formed hydrogel was evaluated using an in vitro and in vivo toxicity study of pectin–acrylamide fabricated hydrogels. Fourier transform infrared confirmed the fabrication of stable pectin/acrylamide hydrogel. Swelling studies have shown that swelling is preferably increased by increasing the acrylamide content of hydrogel structures in solutions of higher pH 7.5. For the formulated formulations, porosity and sol–gel fraction were also calculated. With a rise in pectin and acrylamide content, porosity and gel fraction increased, while porosity decreased and gel fraction improved with an increase in N, N methylene bisacrylamide content. At different pH strengths of 1.2, 5.5 and 7.5, the drug release profile was also analysed. With the increasing pH of the dissolution medium, maximum drug release was seen. By fitting experimental data to model equations and determining the corresponding parameters, the release mechanisms were studied. The maximum drug release of 90% was observed in the A6 formulation. The findings showed that the kinetics of drug release from the hydrogels in the respective buffer solutions were mainly due to Fickian diffusion. The in vivo toxicity studies confirmed the safety of the prepared hydrogels and showed no toxicity.

Abstract:The present study aims to develop and characterize polyethylene glycol (PEG) and xanthan-gum (XG) based interpenetrating network (IPN) for the controlled release of the drug. Natural and synthetic polymers were used in combination via free radical copolymerization to synthesize series of PEG/XG-co-poly-AA IPN hydrogels. Xanthan-gum and PEG (polymers) have been cross-linked with acrylic acid as a monomer, in presence of ammonium peroxo disulfate /sodium hydrogen sulfite as initiators. For cross-linking, ethylene glycol dimethacrylate was employed. Venlafaxine HCl was incorporated in formulated hydrogel network as an experimental drug. FTIR, XRD, TGA, DCS, Sol–gel fraction, in vitro swelling, drug release, and in vivo toxicity analysis were performed to characterize the developed network. FTIR confirmed the formation of polymeric network and ensured physical entanglement of the drug within the matrix. TGA and DSC thermograms indicated that prepared hydrogels were more stable than individual constituents. Effective response for in vitro swelling and release was quantified at pH 1.2 and 7.4. No considerable toxicity was observed. It is concluded that an optimum drug delivery system can be designed to achieve desired characteristics by using a combination of polymers and altering the concentration of formulation ingredients.Graphic abstract

H∞ robust design of PID controllers for arbitrary-order LTI systems with time delay

Abstract:For a given arbitrary-order LTI (linear time-invariant) plant with time delay, we propose a directly parametric design method of the H∞ PID controller in an analytical manner. The design problem of the PID controllers satisfying the H∞ norm requirement is first cast into simultaneous stabilization problem of a family of complex quasipolynomials and the characteristic equation. Then, the linear programming characterization of the PID controllers that can ensure the stability of the complex quasipolynomials are developed on the basis of the extended Hermite-Biehler Theorem.Finally, The admissible set of the H∞ PID controllers is presented by combining such the results with the stabilizing set of the PID controllers. The results reveal that the set of the integral and derivative gains is a union of convex sets for a fixed proportional gain. The proposed scheme works without any approximation and enable one to find a set of the PID parameters satisfying the H∞ norm requirement conveniently.

Abstract:The objective of this work was to evaluate grafted soy protein isolate (SPI) for pharmaceutical applications. The present work reports the microwave-assisted preparation of soy protein isolategrafted[acrylic acid-co-4-(4-hydroxyphenyl)butanoic acid] [SPI-g-(AA-co-HPBA)] hydrogel via graft copolymerization using N,N-methylene-bis-acrylamide and potassium persulphate as the cross-linker and initiator, respectively. The chemical and physical properties of the synthesized polymeric hydrogels were analyzed by Fourier transform infrared spectroscopy, liquid chromatography–mass spectrometry (LCMS), nuclear magnetic resonance 1H-NMR, X-ray diffraction (XRD), transmission electron microscopy (TEM), scanning electron microscopy (SEM), and thermogravimetric analysis (TGA). The SEM, TEM, and XRD analyses have confirmed the formation of hydrogel SPI-g-(AA-co-HPBA) with the network structure having a layered and crystalline surface. The SPI-g-(AA-co-HPBA) hydrogel was investigated for the sustained and controlled drug delivery system for the release of model drug ciprofloxacin at basic pH for its utilization against bacterial infection in oral cavity. The drug release profile for SPI-g-(AA-co-HPBA) hydrogels was studied using LCMS at the ppb level at pH = 7.4. The synthesized hydrogel was found to be noncytotoxic, polycrystalline in nature with a network structure having good porosity, increased thermal stability, and pH-responsive behavior. The hydrogel has potential to be used as the vehicle for controlled drug delivery in oral cavity bacterial infections.This article has not yet been cited by other publications.
