Abstract:In recent years, due to the frequently reported food safety events, people generally care about the food safety. According to former research, the application amount of organophosphorus pesticides and carbamate pesticides possesses about 70% of the total application amount of pesticides. So, it is necessary to develop fast, sensitive and accurate analysis of pesticide residues to assure the quality of agricultural products. However, the existing food quality supervision system is generally based on the instrument analysis technology, which is time-consuming and high-costing, and will delay the actual release of information for the food safety determination. So it cannot provide the rapid response to the consumer queries. Therefore, there is an urgent need to develop the rapid detection techniques for food safety. A novel disposable enzyme tablet was developed for rapid colorimetric detection of pesticide residues in agricultural products, which based on the inhibition of acetylcholinesterase (AChE) activity by pesticides and the AChE-catalyzed hydrolysis of chromogenic substrate, indoxyl acetate. As known, indoxyl acetate can be catalyzed to indole with the existence of AChE. Indole was oxidized in air quickly, turning to be blue-green, which could be easily distinguished by the naked eye. Firstly, the AChE was immobilized onto the Hybond N+nylon membrane. It was a crucial work to choose a suitable immobilization method. In our study, the physical absorption method seemed more easy-operative and suitable than the cross-linking since it rarely has no inference on the results. Besides, immobilization temperature, time and methods of drying were optimized in the immobilization process. The enzymatic tablets were immobilized under 4℃ for 30 min, then freeze-dried in a vacuum chamber, the recovery rate of enzyme activity was as high as 27.3%, which considered to be optimal. The other parameters such as substrate concentration, inhibition time and coloring time were optimized. The results were as follows: the best amount of acetate solution was 10 mmol/L, and the inhibition time and the color develop time were 15 and 10 min, respectively. In the measurement process, the color of the enzyme tablet was blue-green. Through analyzing the changes of color intensity, the limit of detection (LOD) of this assay system were 1μg/mL for omethoate, 0.05μg/mL for chlorpyrifos, 1.5 μg/mL for carbaryl, and 0.8 μg/mL for pirimicarb, all of them have attained the requirements of the maximum residue limits in food. Detection results of pesticide residues in real samples of grape and cabbage showed that the enzyme tablet had high sensitivity, high degree of accuracy, and good reproducibility, suggesting its great potential for practical application in rapid qualitative determination of organophosphorus (OP) and carbamate (CM) pesticide residues in agricultural products. Furthermore, this assay has long storage time at 60 days. In sum, this work provides a feasible way to develop fast, sensitive detection for pesticide residue.
A Lab-in-a-Syringe Device Integrated with a Smartphone Platform: Colorimetric and Fluorescent Dual-Mode Signals for On-Site Detection of Organophosphorus Pesticides

Abstract:Herein, a portable lab-in-a-syringe device integrated with a smartphone sensing platform was designed for rapid, visual quantitative determination of organophosphorus pesticides (OPs) via colorimetric and fluorescent signals. The device was chiefly made up of a conjugate pad labeled with cetyltrimethylammonium bromide-coated gold nanoparticles (CTAB-Au NPs) and a sensing pad modified by ratiometric probes (red-emission quantum 2 quantum dots, 2@gQDs probe), which was assembled through a disposable syringe and reusable plastic filter. In the detection system, thiocholine (Tch), the hydrolysis product of thioacetylcholine (ATch) by acetylcholinesterase (AchE), could trigger the aggregation of CTAB-Au NPs, resulting in a significant color change from red to purple. Then, CTAB-Au NPs flowed vertically upward and bound to the 2@gQDs probe on the sensing pad, reducing the fluorescence resonance energy transfer effect between CTAB-Au NPs and gQDs. Meanwhile, rQDs embedded in SiO2 NPs remained stable as internal reference fluorescence, achieving a color transition from red to green. Thus, based on the inhibition of AChE activity by OPs, a colorimetric and fluorescent dual-mode platform was constructed for on-site detection of OPs. Using glyphosate as a model, with the support of a color recognizer application (APP) on a smartphone, the ratio of red and green channel values could be utilized for accurate OP quantitative analysis ranging from 0 to 10 μM with a detection limit of 2.81 nM (recoveries, 90.8–122.4%; CV, 1.2–3.4%). Abstract:Owing to their large specific surface, favorable electrical conductivity, and excellent electrocatalytic capabilities, two-dimensional transition metal dichalcogenides have received considerable attention in the field of biosensors. On the basis of these properties, we developed a portable and disposable enzyme-based biosensor for paraoxon detection using a metallic MoS2 nanosheets modified screen-printed electrode (SPE). The exfoliated ultrathin metallic MoS2 nanosheets can accelerate the electron transfer on electrode surface and contribute to the immobilization of acetylcholinesterase (AChE) via the cross-linking of glutaraldehyde. Electrodeposited gold nanoparticles (AuNPs) on SPE were used to immobilize MoS2 nanosheets through the interaction between Au atoms on AuNPs and S atoms on MoS2. Using acetylcholine as the substrate, AChE can catalyze the formation of electroactive thiocholine and further generate the redox current. In the presence of paraoxon, the activity of AChE can be inhibited, making the related electrochemical signals weaken. Under the optimized conditions, this electrochemical biosensor exhibited a favorable linear relationship with the concentration of paraoxon from 1.0 to 1000 μg L-1, with the detection limit of 0.013 μg L-1. Furthermore, this developed biosensor was successfully applied to detect paraoxon in pretreated apple and pakchoi samples, which can provide a reliable method for the rapid analysis of organophosphorus pesticides in agricultural products.

Abstract:On-site quantitative detection of organophosphorus pesticides (OPs) is crucial for safeguarding food and public safety. This study presents a novel acetylcholinesterase (AChE)-mediated paper-based Au3+-etching of gold nanobipyramids (AuNBPs) system. The system employs a long-term storable AuNBPs-deposited nylon membrane embedded within a portable and temperature-controlled paper-based analytical device. This system, coupled with a colorimeter-based quantitative method, enables the development of a practical paper-based multicolor sensor (PMS) for on-site quantitative detection of three common OPs (paraoxon, dichlorvos, and trichlorfon). In the absence of OPs, AChE hydrolyzes acetylthiocholine to thiocholine, which reduces Au3+ to Au+. The presence of OPs inhibits AChE activity, thereby preserving Au3+ to etch AuNBPs on nylon membranes, accompanied by multicolor changes. These color changes can be simply quantified by measuring the a∗ parameter of the CIELab color space using a portable colorimeter. Under optimal conditions, the PMS displayed eight OPs-corresponding color changes with a minimum detectable concentration of 1.0-10 μg/L (visual observation) and limits of detection of 0.8-7.2 μg/L (colorimeter) and 0.2-3.4 μg/L (UV-vis spectrometry). The PMS successfully determined the OPs in vegetable and rice samples with recoveries of 89.0-109 % and RSDs (n = 5) of <6 %. These results were consistent with those obtained using the HPLC-MS method. The PMS demonstrates excellent portability, AuNBPs stability, detection sensitivity, and reproducibility, making it a promising tool for the on-site quantitative detection of OPs residues in food. Furthermore, the paper-based etching system coupled with the colorimeter-based quantitative method provides a valuable reference to develop practical PMSs for various targets in diverse fields.

Abstract:An uncomplicated method for the detection of organophosphorous pesticide characterized by a thione moiety (P = S) was developed, and chlorpyrifos was used as an example. It was based on the detection of thiocholine generated by acetylcholinesterase. Organophosphorothionates slightly inhibited acetylcholinesterase, and P = S in chlorpyrifos was oxidized into P = O to improve the toxicity. Optimized amount of n -bromosuccinimide (4 ppm) and ascorbic acid (20 ppm) were used for the conversion. The influence of the excessive ascorbic acid and the high overpotential of the thiocholine was addressed by using a screen-printed silver electrode. According to cyclic voltammetry and amperometry, oxidization of thiocholine occurred on the electrode at a low potential while the ascorbic acid was not oxidized. The working potential was minimized to 0 V, and a detection limit of 2.5 ppb for chlorpyrifos was obtained. This method was further used to detect chlorpyrifos in Fuji apple and cucumber.

Abstract:The relation of the physiological releases of growth hormone-releasing hormone (GHRH) and growth hormone (GH) into the circulation in various conditions was investigated using a sensitive and specific radioimmunoassay for plasma GHRH. The mean fasting plasma level of immunoreactive (IR)-GHRH in 72 normal adults was 10.3 +/- 0.5 (mean +/- SEM) pg/ml and there was no significant sex difference in the level. The concentrations of IR-GHRH in plasma from the umbilical artery and umbilical vein were 107.3 +/- 20.5 pg/ml and 33.6 +/- 3.8 pg/ml, respectively, and a marked arterio-venous gradient was observed in all 12 individuals examined. The plasma level of IR-GHRH in the maternal vein was significantly lower than that in the cord blood, but was similar to that in non-pregnant women. In normal adults, although there was no apparent fluctuation in the level of plasma IR-GHRH or of plasma GH during bed rest, a significant increase of plasma IR-GHRH was detected followed by, or synchronized with the surge of plasma GH after oral administration of L-dopa. In contrast, on L-arginine infusion, no proportional elevation of plasma IR-GHRH with increase in plasma GH was observed. During and after intravenous infusion of somatostatin, the circulating IR-GHRH level did not increase, but on stopping the infusion there was an immediate and marked rebound surge of GH. We conclude that 1) the elevated IR-GHRH in the cord blood plasma originates from the fetus and may have a primary role in enhancing secretion of GH which promotes growth in early human life, and 2) the participations of GHRH in the mechanisms of GH secretion seen after administrations of L-dopa, L-arginine and somatostatin are different.

Abstract:On-site quantitative analysis of pesticides is important for food safety. Colorimetric gold nanobipyramids (AuNBPs) sensors are powerful methods for on-site detection. However, a single quantitative method and the instability of AuNBPs in solution limit the practicability of those sensors. Here, a paper-based multicolor AuNBPs sensor involved a colorimeter-assisted method for quantifying color was developed for quantitative detection of 2,4-dichlorophenoxyacetic acid (2,4-D), a common herbicide. The novelty of this study lies in developing a general paper-based quantitative on-site method (PQOM) for colorimetric AuNBPs sensors. Firstly, a paper-based analytical device (PAD) consisting of a nylon membrane, absorbent cotton layers, and two acrylic plates was fabricated to deposit AuNBPs. We demonstrated the PAD could improve the stability of AuNBPs and the detection sensitivity of AuNBPs sensors. Then, a handheld colorimeter was first used to quantify the color change of AuNBPs on the PAD based on the CIELab color space. Finally, as proof of concept, the PQOM was successfully employed to quantify 2,4-D by combining with an alkaline phosphatase-mediated AuNBPs growth method. In this method, 2,4-D specifically inhibited alkaline phosphatase activity to suppress the generation of L-ascorbic acid, thereby mediating AuNBPs growth. The developed sensor exhibited seven 2,4-D concentration-related colors and detected as low as 50 ng mL(-1) 2,4-D by naked-eye observation and 18 ng mL(-1) 2,4-D by a colorimeter. It was applied to detect 2,4-D in the spiked rice and apple samples with good recovery rates (91.8-112.0%) and a relative standard deviation (n = 5) < 5%. The success of this study provides a sensing platform for quantifying 2,4-D on site.
Photothermal effect-enhanced peroxidase-like performance for sensitive detection of organophosphorus pesticides on a visual test strip

Abstract:Pesticides pose a serious risk to public health, even in trace amounts. The sensitive, rapid, visual, and on-site detection of pesticides in a real sample remains a challenge. Herein, we report a visual and rapid organophosphorus pesticide (OP) sensing device with excellent sensitivity, ease of application, and specificity based on a photothermal effect-based peroxidase (POD) mimetic. The ultrathin MIL-101-NH 2 (Fe) (Fe-MOF) shell-coated Au nanobipyramide (NBP) not only possesses POD-like activity, but also exhibits excellent photothermal effect under near-infrared (NIR) light. The results reveal that the excellent photo-to-thermal conversion of the Au NBPs can rapidly heat the MIL-101-NH 2 (Fe) shell, which subsequently induces a markedly enhanced POD-like activity. Using paraoxon-ethyl as model OP, the combination of the OP inhibition effect on the biocatalytic activity of acetylcholinesterase with the inhibition effect of thiocholine on the colorimetric assays of the POD-mimicking Fe-MOF leads to a significant improved selectivity for OP sensing. In particular, the NIR light irradiation increased the OP detection sensitivity by 100 times. Importantly, based on this detection strategy, a visual test strip with high sensitivity was developed and enabled real-time detection through a simple color analysis software on a smartphone. The successful detection of OP residues in fruit peel using this test strip further confirmed its application potential in real sample analysis.

Abstract:A multicolor enzyme-linked immunosorbent sensor was constructed to detect organophosphates pesticides (OPs) on base of TMB2+-mediated AuNRs etching. As the etching progresses, a rainbow-like color responses was generated. The naked eye limit of detection (LOD) of the sensing system is 0.25 ng mL-1. In particular, the longitudinal localized surface plasmon resonances (LSPR) peak shift of AuNRs is linearly correlated with the logarithm of OPs concentration in range of 104 to 5×106 pg mL-1 for parathion-methyl and fenitrothion, 5×104 to 107 pg mL-1 for fenthion, respectively. Compared with reported methods, this sensor displayed satisfactory linear ranges and detection limits. Average recoveries of this method ranged from 73.28% to 98.08% and relative standard deviations (RSDs) were below 8.0% as for food samples analysis without complicated pretreatments. Remarkably, the developed multicolor enzyme-linked immunosorbent sensor is capable of providing accurate and reliable assay for OPs in complex food samples.

Abstract:Ultrasmall Au nanoparticles (UsAuNPs) in the size range of 4.0-7.0 nm was successfully immobilized on the surface of 2D metalloporphyrinic metal-organic framework nanosheets (2D MOF). Firstly, The obtained hybrid nanomaterial, UsAuNPs/2D MOF, was fully characterized by TEM, HRTEM, element mapping images and XPS. Then, the peroxidase-like activity of UsAuNPs/2D MOF was comparatively studied with other hybrid nanozyme to explore the influence of AuNPs size on peroxidase-like activity. Further, UsAuNPs/2D MOF with outstanding peroxidase-like activity was selected to form ternary cascade enzyme reaction with acetylcholinesterase (AChE) and choline oxidase (ChOx). Based on the inhibitory effect of organophosphorus pesticides on AChE, a fast and sensitive colorimetric method was established for trichlorfon detection with the advantages of simple operation, low detection limit (1.7 μM), good linear range (1.7-42.4 μM) and high accuracy (recovery rate of 96.6-105.3%). Finally, this method was applied to visual analysis of trichlorfon concentration in tomatoes, cucumbers and eggplants.
A dual-signal sensor for the analysis of parathion-methyl using silver nanoparticles modified with graphitic carbon nitride

Abstract:A g-C3N4/AgNPs sensor could be used for the determination of organophosphorus pesticides (OPs).The method exhibits ultra-sensitivity and high-selectivity toward OPs.Double-signal OPs probe was developed with low cost, simple interpretation and easy handling.A highly sensitive and selective method was developed for both UV–vis spectrophotometric and fluorimetric determination of organophosphorus pesticides (OPs). This method uses silver nanoparticles (AgNPs) modified with graphitic carbon nitride (g-C3N4). The AgNPs reduce the fluorescence intensity of g-C3N4. Acetylthiocholine (ATCh) can be catalytically hydrolyzed by AChE to form thiocholine, which induces aggregation of the AgNPs. This aggregation leads to the recovery of the blue fluorescence of g-C3N4, with excitation/emission peaks at 310/460 nm. This fluorescence intensity can be reduced again in the presence of OPs because of the inhibitory effect of OPs on the activity of AChE. The degree of reduction was found to be proportional to the concentration of the OPs, and the limit of fluorometric detection was 0.0324 μg/L (S/N = 3). In addition, the absorption of the g-C3N4/AgNPs at 390 nm decreased because of the aggregation of the AgNPs, but was recovered in presence of OPs because of the inhibition of enzyme activity by OPs. Abstract:It is of great significance to on-site detection of organophosphate pesticides (OPs) for pollution monitoring and poisoning estimation. Herein, we developed a portable hydrogel test kit for on-site detection of OPs, which is based on the integration of agarose hydrogel with dual-emission coordination polymers (CPs) nanocomposite comprised of Ru(bpy)32+ and zinc (II)-based CPs (ZnCPs) loaded with thioflavin T (ThT). Different from Ru(bpy)32+ with stable fluorescence in acidic environment, ThT@ZnCPs is highly sensitive to H+, which destroys the structure of ZnCPs as a host and quenches ThT@ZnCPs fluorescence. The distinct fluorescence behaviors of Ru(bpy)32+ and ThT@ZnCPs in acidic environment enable the hydrogel test kit to exhibit ratiometric fluorescence responses to acetylcholinesterase (AChE), which hydrolyzes acetylcholine to acetic acid and provides H+. On this basis, combining the inhibition effect of OPs to AChE activity, a ratiometric fluorescence method for OPs detection was established with the hydrogel test kit, and satisfactory results have been achieved in buffered aqueous solutions and apple juice samples. Attractively, by employing smartphone as a signal readout, on-site quantitation of OPs was accomplished with the features of easy to use, portability and low cost, demonstrating a great promising for point-of-care testing in food safety monitoring.

Abstract:Developing simple, efficient, and inexpensive method for trace amount organophosphorus pesticides' (OPs) detection with high sensitivity and specificity is of significant importance for guaranteeing food safety. Herein, an Ag/Au bimetallic nanoparticle-based acetylcholinesterase (AChE) surface-enhanced Raman scattering (SERS) biosensor was constructed for in situ simple and sensitive detection of pesticide residues in food. The principle of this biosensor exploited 4-mercaptophenylboronic acid (4-MPBA)-modified Ag/Au bimetallic nanoprobes as SERS signal probe to improve sensitivity and stability. The combination of AChE and choline oxidase (CHO) can hydrolyze acetylcholine (ATCh) to generate H 2 O 2 . The product of H 2 O 2 selectively oxidizes the boronate ester of 4-MPBA, decreasing the Raman intensity of the B-O symmetric stretching. In the presence of OPs, it could inhibit the production of H 2 O 2 by destroying the AChE activity, so the reduction of the SERS signal was also alleviated. Based on the principle, an Ag/Au bimetallic nanoparticle-based AChE SERS sensor was established without any complicated pretreatments. Benefiting from the synergistic effects of Ag/Au bimetallic hybrids, a linear detection range from 5×10 −9 to 5×10 −4 M was achieved with a limit of detection down to 1.7×10 −9 M using parathion-methyl (PM) as the representative model of OPs. Moreover, the SERS biosensor uses readily available reagents and is simple to implement. Importantly, the proposed SERS biosensor was used to quantitatively analyze OP residues in apple peels. The levels of OPs detected in real samples by this method were consistent with those obtained using gas chromatography–mass spectrometry (GC–MS), suggesting the proposed assay has great potential applications for OPs in situ detection in food safety fields.
Stimulus Response of GQD-Sensitized Tb/GMP ICP Nanoparticles with Dual-Responsive Ratiometric Fluorescence: Toward Point-of-Use Analysis of Acetylcholinesterase and Organophosphorus Pesticide Poisoning with Acetylcholinesterase as a Biomarker

Abstract:In this study, by rationally designing the stimulus response of graphene quantum dot (GQD)-sensitized terbium/guanine monophosphate (Tb/GMP) infinite coordination polymer (ICP) nanoparticles, we have constructed a smartphone-based colorimetric assay with ratiometric fluorescence, which could be applied for the detection of acetylcholinesterase (AChE) and organophosphorus pesticides (OPs) directly. First, GQDs with abundant functional groups were chosen as the guest, which not only could be used as one of the signal readouts but also served as the antenna ligand to further sensitize the fluorescence of the host Tb/GMP. Upon being excited at 330 nm, the green fluorescence of the Tb/GMP host is highly enhanced, while the blue fluorescence of GQDs is suppressed due to the confinement of the ICP host. With the presence of thiocholine (TCh), an enzymatic product hydrolyzed from acetylthiocholine (ATCh) by AChE, the competitive coordination of Tb3+ between GMP and TCh results in the collapse of the ICP network and thereby the release of GQDs into the solution; thus, the fluorescence of Tb/GMP turns off and the fluorescence of GQDs turns on. The dual-responsive ratiometric fluorescent intensity change leads to the corresponding green-to-blue fluorescent color change obviously, which constitutes a novel mechanism for the colorimetric analysis of AChE. Moreover, when OPs are subsequently introduced, the activity of AChE is blocked, thus preventing the stimulus response of GQD@Tb/GMP ICP nanoparticles, leading to the fluorescent color change from greenish-blue to green, which could also be employed for OP detection. Benefitting from the high sensitivity, good reliability, and the obvious color changes, the method demonstrated here is a promising candidate to realize smartphone-based point-of-use applications, which is of great importance for timely clinical diagnosis and treatment of OPs related to health issues with AChE as an exposure biomarker in less industrialized countries, in remote settings, or even in home care services.

Abstract:A novel dual-signal fluorescent sensor was developed for detecting organophosphorus pesticides (OPs). It relies on the catalytic activities of acetylcholinesterase (AChE) and choline oxidase (ChOx) to generate hydrogen peroxide (H 2 O 2 ) through the conversion of acetylcholine (ACh) to choline·H 2 O 2 then oxidizes ferrocene-modified tetraphenylethylene (TPE-Fc) to its oxidized state (TPE-Fc + ), resulting in enhanced cyan fluorescence due to aggregation. Simultaneously, ferrocene oxidation generates hydroxyl radicals (•OH), causing a decrease in orange fluorescence of glutathione-synthesized gold nanoclusters (GSH-AuNCs). The presence of OPs restricts AChE activity, reducing H 2 O 2 production. Increasing OPs concentration leads to decreased cyan fluorescence and increased orange fluorescence, enabling visual OPs detection. The sensor has a linear dynamic range of 10–2000 ng/mL with a detection limit of 2.05 ng/mL. Smartphone-based color identification and a WeChat mini program were utilized for rapid OPs analysis with successful outcomes.

Abstract:The residues of organophosphorus pesticides in agricultural production and soil constitute a significant threat to both human health and ecosystems, underscoring the critical importance of developing effective detection methods for these pesticides. Here, we introduce an innovative method for detecting trichlorfon that integrates hydrogen peroxide (H 2 O 2 )-induced etching of silver-coated gold nanostars (AuNS@Ag) with an enzymatic reaction cascade. By precisely controlling the thickness of the silver layer and the morphology of the gold nanoparticle core, we enhance the sensitivity of the H 2 O 2 etching process. In this detection system, acetylcholinesterase (AChE) catalyzes the hydrolysis of acetylcholine chloride (ACh) into choline, which is then oxidized by choline oxidase (ChOx) to produce H 2 O 2 . The generated H 2 O 2 subsequently etches the silver layer of AuNS@Ag, leading to a pronounced red-shift in the localized surface plasmon resonance (LSPR) wavelength, from 590 nm to 735 nm. However, the presence of trichlorfon inhibits the activity of AChE, resulting in a reduced production of H 2 O 2 and a subsequent attenuation of the LSPR wavelength shift. Under optimized experimental conditions, this method enables the linear detection of trichlorfon within a concentration range of 0.1–5.0 μg mL −1 and has been successfully applied to detect trichlorfon in cabbage and tap water samples, showcasing its promising potential in food safety and environmental monitoring.

Abstract:Given the importance of food safety, it is highly desirable to develop a convenient, low-cost, and practical sensor for organophosphorus pesticides (OPs) detection. Here, a fluorescent paper analytical device (FPAD) based on aggregation-induced emission (AIE) nanoparticles (PTDNPs-0.10) and two-dimension MnO2 nanoflakes (2D-MnNFs) was developed for instrument-free and naked-eye analysis of OPs. PTDNP-MnNFs composites were obtained through 2D-MnNFs and PTDNPs-0.10 by electrostatic interaction. and the fluorescence emission of PTDNPs-0.10 was quenched through fluorescence resonance energy transfer (FRET). When acetylcholinesterase (AChE) was present, acetylthiocholine (ATCh) was catalytically hydrolyzed into thiocholine, which reduced MnO2 of PTDNP-MnNFs into Mn2+, subsequently blocking the FRET and enhancing the fluorescence. Upon the addition of OP, AChE activity was depressed and thus the FRET between 2D-MnNFs and PTDNPs-0.10 was not affected, resulting in a slight change in fluorescence. On the basis of the variation in fluorescence intensity, highly sensitive detection of OP was readily achieved with a detection limit of 0.027 ng/mL; on the basis of the variation in brightness of FPAD, instrument-free and visual detection of OP was realized using a smartphone with a detection limit of 0.73 ng/mL. The application of FPAD has significantly simplified the detection procedure and decreased the test cost, supplying a new approach for on-site detection of OPs.

Abstract:Pesticide residue is an important problem in environmental pollution, and colorimetric methods are suitable for on-site fast assay of pesticides. Analytes-triggered aptamer desorption from gold nanoparticles (AuNPs) has been widely used to develop label-free colorimetric assays. But the direct interactions between pesticides and AuNPs have not been considered previously, leading to one possible misinterpretation of analysis results. In this work, we studied the direct adsorption of pesticides on AuNPs, and further performed a comprehensive analysis of aptamer’s role. Here the colorimetric response was independent on DNA sequences, and a similar aggregation of AuNPs was shown for all the tested DNA sequences. Our study demonstrated a misinterpretation in the previous assays, while pesticides adsorption by AuNPs was the main mechanism for these colorimetric assays. Finally, we designed another alternative sensor array based on different parameters (pH, ionic strength and [DNA]) for accurate discrimination of four pesticides including acetamiprid, carbendazim, pymetrozine and thiabendazole. Due to the different functional groups in these four pesticides, the fingerprints of AuNPs’ colorimetric responses allow intuitively identifying different pesticides for environmental or food application.

Abstract:Rapid and sensitive detection methods are in urgent demand for the screening of extensively used organophosphorus pesticides and highly toxic nerve agents for their neurotoxicity. In this study, we developed a novel Fe(3)O(4) magnetic nanoparticle (MNP) peroxidase mimetic-based colorimetric method for the rapid detection of organophosphorus pesticides and nerve agents. The detection assay is composed of MNPs, acetylcholinesterase (AChE), and choline oxidase (CHO). The enzymes AChE and CHO catalyze the formation of H(2)O(2) in the presence of acetylcholine, which then activates MNPs to catalyze the oxidation of colorimetric substrates to produce a color reaction. After incubation with the organophosphorus neurotoxins, the enzymatic activity of AChE was inhibited and produced less H(2)O(2), resulting in a decreased catalytic oxidation of colorimetric substrates over MNP peroxidase mimetics, accompanied by a drop in color intensity. Three organophosphorus compounds were tested on the assay: acephate and methyl-paraoxon as representative organophosphorus pesticides and the nerve agent Sarin. The novel assay displayed substantial color change after incubation in organophosphorus neurotoxins in a concentration-dependent manner. As low as 1 nM Sarin, 10 nM methyl-paraoxon, and 5 μM acephate are easily detected by the novel assay. In conclusion, by employing the peroxidase-mimicking activity of MNPs, the developed colorimetric assay has the potential of becoming a screening tool for the rapid and sensitive assessment of the neurotoxicity of an overwhelming number of organophosphate compounds.

Abstract:Accurately detect the residues of organophosphate pesticides (OPs) in food and environment is critical to our daily lives. In this study, we developed a novel acetylcholinesterase (AChE) biosensor based on Au–Tb alloy nanospheres (NSs) for rapid and sensitive detection of OPs for the first time. Au–Tb alloy NSs that with good conductivity and biocompatibility were produced with a mild hydrothermal. Under optimal conditions, the AChE biosensor was obtained by a simple assembly process, with a big linear range (10−13–10−7 M) and the limit of detection was 2.51 × 10−14 M for the determination of methyl parathion. Moreover, the determination of methyl parathion with the prepared biosensor presented a high sensitivity, outstanding repeatability and superior stability compared with other reported biosensors. Through the determination of tap water and Yanming lake samples, it was proved that the modified biosensor with satisfactory recoveries (96.76%–108.6%), and are realizable in the determination of OPs in real samples.
