Abstract:In this study, secondary brewery wastewater (SBWW) supplemented with sugarcane molasses (SCM) was used for SBWW treatment with concomitant lipid and carotenoid production by the yeast Rhodosporidium toruloides NCYC 921. In order to improve the biomass production, ammonium sulfate, yeast extract and urea were tested as nitrogen sources. Urea was chosen as the best low-cost nitrogen source. A fed-batch cultivation was carried out with SBWW supplemented with 10 g L-1 of sugarcane molasses as carbon source, and 2 g L-1 of urea as nitrogen source. A maximum biomass concentration of 42.5 g L-1 was obtained at t = 126.5 h and the maximum biomass productivity was 0.55 g L-1 h-1 at t = 48.25 h. The maximum lipid content was 29.9 % w/w (DCW) at t = 94 h of cultivation and the maximum carotenoid content was 0.23 mg g-1 at 120 h of cultivation. Relatively to the SBWW treatment, after the batch phase, 45.8% of total Kjeldahl nitrogen removal, 81.7% of COD removal and 100% of sugar consumption were observed. Flow cytometry analysis revealed that 27.27% of the cells had injured membrane after the inoculation. This proportion was reduced to 10.37% at the end of the cultivation, indicating that cells adapted to the growth conditions.

Culture Strategies for Lipid Production Using Low-cost Carbon Sources by Rhodosporidium Toruloides

Abstract:The development of sustainable biofuels is inserted in the biorefinery concept through the use and valorization of raw material. Volatile fatty acids (VFAs) and rich-carbon wastes, present in different raw materials, have gradually been used as the carbon source for lipid production by oleaginous yeasts. In this context, it is essential to develop practical culture strategies for oleaginous yeast to use these wastes, acetic acid and the carob industry by-product. In this study, new approaches in the cultivation of the yeast Rhodosporidium toruloides were tested, in order to maximize the accumulation of biomass and lipids. Different concentrations of carob kibbles sugar rich extract and acetic acid waste were used, as low-cost carbon sources, to select the best culture strategy for the bioconversion of these wastes into microbial oils. Microbial oils were produced by R. toruloides using 3–18 g/l acetic acid and 9–70 g/l carob extract, with different carbon-to-nitrogen ratios (C/Ns). Batch and fed-batch cultures were compared in terms of biomass and lipid production and lipid productivity. The highest biomass of 19.14 ± 0.54 g/l was obtained in batch cultures with 70 g/l of carob extract. A lipid content of 20% (w/w) was achieved using 15 g/l of acetic acid as the carbon source in a fed-batch mode. To improve biomass production as well as lipid accumulation, the strategy used was the combination of both residues in fed-batch sequential mode of the cultures in a 2L STR bioreactor. This strategy improved the lipid yield to 40% (w/w). These results provide viable culture strategies for lipid accumulation by R. toruloides using diverse waste-derived VFA’s and sugars.

Abstract:Rhodosporidium toruloides is an unconventional yeast with a natural lipid pathway known to accumulate oil under suitable conditions, typically characterized by nutritional stress induced through the depletion of essential nutrients. The objective of this research was to evaluate microbial oil production by implementing an adaptation strategy in three strains of Rhodosporidium toruloides , all isolated from the Brazilian biome, to increase tolerance to degradation products resulting from pretreatment steps of sugarcane biomass. This is the first time these three strains have been tested for lipid accumulation and tolerance to biomass hydrolysate. The assays were performed to study the assimilation of glucose and xylose, as well as the performance of their mixture in synthetic medium. Subsequently, all non-adapted strains were tested using the previously obtained non-detoxified hemicellulosic hydrolysate. Due to their performance, a step of previous adaptation of the media was employed. Essentially, the strains were successive cultivated in increasing concentrations of hemicellulosic hydrolysate, incremented by 10% every 24 h until reaching 100% hydrolysate. Microbial oil production and yeast growth in the hemicellulosic hydrolysate were compared for the adapted and non-adapted strains. The results indicate that lipid accumulation was enhanced through fermentation of the adapted strain, resulting in improvements of 1.69, 2.51, and 2.84 times compared to the non-adapted strain for R. toruloides 2781, R. toruloides 2882, and R. toruloides 2896, respectively. Overall, the adapted strain R. toruloides 2896 showed the best performance, with a productivity of 16.6 ± 0.63 mg.L −1 .h −1 . This study introduces an improved approach to microbial oil production from biowaste and highlights the detrimental impact of toxic compounds in the medium, leading to the inhibition of cellular fermentative performance, especially noticeable in non-adapted strains. Conversely, adapted strains exhibited increased tolerance to specific toxic compounds in hemicellulosic hydrolysate, particularly towards acetic acid and components such as acid-soluble lignin. Notable constituents include syringaldehyde, gallic acid, 2,4-dihydroxybenzoic acid, and the furanic compound furfural. This heightened comprehension may yield insights that contribute to the advancement of knowledge in this field of study.

Abstract:The circular bioeconomy concept relies on the exploitation of wastes as a feedstock of different biotechnological processes to obtain, as much as possible, a huge spectrum of biochemical components through a biorefinery platform. This work deals with the treatment of brewery effluent through the cultivation of Scenedesmus obliquus microalga and the use of the biomass in a complex biorefinery. The treatment proved efficient in the removal of nutrients (N, P and COD removals of 88, 30 and 71% respectively). Several compounds and products were obtained from the biomass, such as: (a) phenols (0.249–1.016 mg GAE mL−1) and flavonoids (0.05–0.167 mg CE mL−1) by subcritical water extraction (SWE) at 120 °C, the extraction efficiency being five times higher at 200 °C; (b) biohydrogen by dark fermentation (67.1 mL H2 gVS−1); (c) bio‐oil (64%), biochar (30%) and biogas (6%) by pyrolysis; and (d) enhanced capacity of germination/growth of wheat and barley seeds by S. obliquus culture and biomass (pellet, after centrifugation); better results were obtained with the biomass cultivated in brewery effluent (when compared with synthetic medium), and the biomass pellet was better than the whole culture; barley seeds treated with the pellet from the brewery effluent had the highest germination index (GI) of 85 compared with the control (tap water) GI of 35. The innovative study emphasis was on reducing microalgae production costs, providing environmental benefits in a biorefinery‐based S. obliquus platform. © 2019 Society of Chemical Industry and John Wiley & Sons, Ltd

Abstract:To use unicellular microalgae to remove waste nutrients from brewery wastewater while converting them into algal biomass has been explored but high-cost treatment and low-value biomass associated with current technologies have prevented this concept from further attempts. In this study, a filamentous microalga Tribonema aequale was introduced and the alga can grow vigorously in brewery wastewater and algal biomass concentration could be as high as 6.45 g L −1 which can be harvested by a cost-effective filtration method. The alga together with autochthonous bacteria removed majority of waste nutrients from brewery wastewater. Specifically, 85.39% total organic carbon (TOC), 79.53% total dissolved nitrogen (TN), 93.38% ammonia nitrogen (NH 3 –N) and 71.33% total dissolved phosphorus (TP) in brewery wastewater were rapidly removed by co-cultivation of T . aequale and autochthonous bacteria. Treated wastewater met the national wastewater discharge quality, and resulting algal biomass contained large amounts of high-value products chrysolaminarin, palmitoleic acid (PLA) and eicosapentaenoic acid (EPA). It is anticipated that reduced cost of algal harvesting coupled with value-added biomass could make T . aequale as a promising candidate for brewery wastewater treatment and resource utilization.

Abstract:Mannosylerythritol lipids (MELs) are biosurfactants with excellent biochemical properties and a wide range of potential applications. However, most of the studies focusing on MELs high titre production have been relying in the use of vegetable oils with impact on the sustainability and process economy. Herein, we report for the first time MELs production using oils produced from microalgae. The bio-oil was extracted from Neochloris oleoabundans and evaluated for their use as sole carbon source or in a co-substrate strategy, using as an additional carbon source D-glucose, on Moesziomyces spp. cultures to support cell growth and induce the production of MELs. Both Moesziomyces antarcticus and M. aphidis were able to grow and produce MELs using algae-derived bio-oils as a carbon source. Using a medium containing as carbon sources 40 g/L of D-glucose and 20 g/L of bio-oils, Moesziomyces antarcticus and M. aphidis produced 12.47 ± 0.28 and 5.72 ± 2.32 g/L of MELs, respectively. Interestingly, there are no significant differences in productivity when using oils from microalgae or vegetable oils as carbon sources. The MELs productivities achieved were 1.78 ± 0.04 and 1.99 ± 0.12 g/L/day, respectively, for M. antarcticus fed with algae-derived or vegetable oils. These results open new perspectives for the production of MELs in systems combining different microorganisms.

Abstract:The distribution of radioactivity from N-acetyl-9-[114C]-2-aminofluorene and p-[14C]-dimethylaminoazobenzene administered i.v. or p.o. to 18-day-pregnant mice was observed by means of autoradiography of longitudinal section of the whole mouse. Radioactive substances derived from these carcinogens passed through the placenta and were distributed in the fetal organs including the kidney and intestine. Column, thin-layer, and paper chromatography revealed the presence, in while fetuses as well as in maternal livers, of N-acety-9-[14C]-2-aminofluorene. N-hydroxy-N-acetyl-9-[14C]-2-aminofluorene, ring-hydroxy-N-acetyl-9-[14C]-2-aminofluorene, and p-[14C]-dimethylaminoazobenzene. These results establish that N-acetyl-9-[14C]-2-aminofluorene and [14C]-p-dimethylaminoazobenzene are transported, metabolized, and excreted in the mouse fetus.
Yeast Lipids from Crude Glycerol Media and Utilization of Lipid Fermentation Wastewater as Maceration Water in Cultures of Edible and Medicinal Mushrooms

Abstract:Four wild "red" yeast strains (Rhodosporidium kratochvilovae FMCC Y70, R. toruloides NRRL Y-27013, R. toruloides NRRL Y-17902 and R. toruloides NRRL Y-6985) were cultured in shake flasks on industrial glycerol at an initial substrate (Gly0) concentration ≈ 50 g/L under nitrogen limitation. Strains NRRL Y-27013, NRRL Y-17902 and NRRL Y-6985 presented appreciable dry cell weight (DCW) and lipid synthesis (DCW up to 18–19 g/L containing lipids in quantities ≈ 47%, w/w). Strains NRRL Y-27013 and NRRL Y-6985 were further tested in higher Gly0 concentrations (≈90 g/L and ≈110 g/L) with the same initial nitrogen quantity as in the first ("screening") experiment. Both strains, despite the high Gly0 concentrations and C/N ratios (up to 120 moles/moles) imposed, presented significant DCW production (up to c. 29.0–29.5 g/L). Yeast biomass contained significant lipid (42–43%, w/w) and endopolysaccharide (up to 42%, w/w) quantities. Both lipids and endopolysaccharide quantities (in % w/w) noticeably increased as a response to the imposed nitrogen limitation. Lipids containing mainly oleic and palmitic acids constituted ideal candidates for biodiesel synthesis. Thereafter, the wastewaters derived from the lipid production process (lipid fermentation wastewaters—LFWs) were used as maceration waters in cultivations of edible and medicinal fungi, where novel (non-conventional) substrates were used in the performed cultures. CW (coffee residue + wheat straw), CB (coffee residue + beech wood shavings), OW (olive crop + wheat straw), OB (olive crop + beech wood shavings), RW (rice husk + wheat straw) and RB (rice husk + beech wood shavings) were soaked/sprayed with LFWs or tap water and utilized in the cultivation of Pleurotus, Ganoderma and Lentinula mushrooms. The impact of LFWs on the mycelial growth rate (mm/d) and biomass production was evaluated. The results show that regardless of the wetting method, the highest growth rates (6.2–6.6 mm/d) were noticed on RW and RB for Pleurotus eryngii and Ganoderma resinaceum, on OW, OB and RW for Ganoderma applanatum and on RW, OW and OB for Lentinula edodes. Nevertheless, high biomass production was obtained on substrates soaked with LFWs for Pleurotus ostreatus (RW: 443 mg/g d.w.), L. edodes (RB: 238 mg/g d.w.) and Ganoderma lucidum (RW: 450 mg/g d.w.). Overall, this study demonstrates the possibility of the industrial conversion of low-value agro-waste to mycelial mass and eventually to important food products.

Abstract:The co-cultivation of red yeasts and microalgae works with the idea of the natural transport of gases. The microalgae produce oxygen, which stimulates yeast growth, while CO2 produced by yeast is beneficial for algae growth. Both microorganisms can then produce lipids. The present pilot study aimed to evaluate the ability of selected microalgae and carotenogenic yeast strains to grow and metabolize in co-culture. The effect of media composition on growth and metabolic activity of red yeast strains was assessed simultaneously with microalgae mixotrophy. Cultivation was transferred from small-scale co-cultivation in Erlenmeyer flasks to aerated bottles with different inoculation ratios and, finally, to a 3L bioreactor. Among red yeasts, the strain R. kratochvilovae CCY 20-2-26 was selected because of the highest biomass production on BBM medium. Glycerol is a more suitable carbon source in the BBM medium and urea was proposed as a compromise. From the tested microalgae, Desmodesmus sp. were found as the most suitable for co-cultivations with R. kratochvilovae. In all co-cultures, linear biomass growth was found (144 h), and the yield was in the range of 8.78–11.12 g/L of dry biomass. Lipids increased to a final value of 29.62–31.61%. The FA profile was quite stable with the UFA portion at about 80%. Around 1.98–2.49 mg/g CDW of carotenoids with torularhodine as the major pigment were produced, ubiquinone production reached 5.41–6.09 mg/g, and ergosterol yield was 6.69 mg/g. Chlorophyll production was very low at 2.11 mg/g. Pilot experiments have confirmed that carotenogenic yeasts and microalgae are capable of symbiotic co-existence with a positive impact om biomass growth and lipid metabolites yields.

Abstract:Even though biofuel production from microalgae has become more and more attractive in recent years, it is limited especially by the high cost of microalgae cultivation. However, microalgae can be grown in wastewater in order to reduce their production cost and, at the same time, the polluting impact of wastewaters. Winery wastewaters, which are abundantly released from the wine making process, have a large pollution impact related to their high loads of total solids, chemical oxygen demand (COD) and polyphenol concentration. In this research work a co-culture of Chlorella vulgaris and Arthrospira platensis was used to treat three different winery wastewaters from different steps of the wine production process, in order to produce low-cost biomass intended for biofuel production. Growth of the co-culture and reduction of wastewater pollutant impact were followed by daily determinations of biomass concentration, COD and polyphenol content. The highest productivities of biomass (0.66 gDry Weight/L·day) and lipids (7.10 ± 0.22 gLipid/100 L·day) were obtained using 20% of second washing winery wastewater after 4 days of treatment. Moreover, COD and polyphenol content of the three different wastewaters were reduced by the co-culture by more than 92% and 50%, respectively. These results suggest that winery wastewaters can be used successfully for the growth of A. platensis and C. vulgaris co-culture in order to obtain inexpensive biomass for energy production purposes.

Abstract:Co-cultivation of microalgae and bacteria during municipal wastewater treatment can boost carbon and nutrient recovery as a result of their synergistic interactions. The symbiotic relationships between the locally isolated microalga Chlorella vulgaris and the bacterium Rhizobium sp., co-isolated from municipal wastewater, were investigated batchwise under photoautotrophic, heterotrophic, and mixotrophic conditions in a synthetic municipal wastewater medium. During photoautotrophic growth in BG11 medium, photosynthetic algal oxygenation and organic carbon production supported bacterial activity but no significant beneficial effects on microalgal growth were observed. In synthetic wastewater, a twofold higher biomass concentration was achieved in the axenic algal culture compared with the co-culture under heterotrophic conditions, suggesting a competition for nutrients. A comparable carbon removal was observed in all cultures (83–79% TOC), but a faster nitrogen consumption (59% TN) and complete phosphate assimilation (100% TP) was only achieved in the co-culture. A positive synergistic relationship was found under mixotrophic conditions, clearly supported by an in situ O2/CO2 exchange between the microorganisms. This mutualism led to a threefold higher biomass production with a 13-fold higher fatty acid content compared with the axenic algal culture, as well as a superior wastewater treatment performance (+ 58% TOC, + 41% TN and + 44% TP). The co-cultivation of C. vulgaris and Rhizobium is therefore suggested as a potential microbial consortium for a cost-efficient biomass generation during municipal wastewater reclamation, especially under mixotrophic conditions.

Abstract:The cultivation of oleaginous yeast on second-generation feedstocks is an attractive alternative for edible lipid production. Despite many studies in this area addressing single aspects of this bioprocess, one of the major bottlenecks is the integration and optimization of the multiple unit operations while demonstrating this on a larger scale. In this investigation, a microwave-assisted hydrothermal process was developed to solubilize distillers' dried grains with solubles (DDGS). The optimal MW process was run semicontinuously and produced a fermentable oligosaccharide-rich stream with negligible furan-based content. An overliming step and reverse-osmosis stage were demonstrated, increasing the solubilized carbohydrate content to over 60 g/L, suitably concentrated for further bioprocessing. The oleaginous yeast, Metschnikowia pulcherrima, was used to ferment the material and was demonstrated to metabolize Up to 75% of the oligosaccharide pool and produced 41% lipid content at a concentration of 14 g/L in a 27 day semicontinuous process.

Abstract:In this study, the growth of yeast and yeast-like fungi in the liquid digestate from vegetable wastes was investigated in order to remove nutrients and organic pollutants, and for their application as co-culture members with green microalgae. The studied yeast strains were characterized for their assimilative and enzymatic profiles as well as temperature requirements. In the first experimental stage, the growth dynamics of each strain were determined, allowing to select the best yeasts for further studies. In the subsequent stage, the ability of selectants to remove organic pollutants was assessed. Different cultivation media containing respectively 1:3, 1:1, 3:1 vol ratio of liquid digestate and the basal minimal medium were used. Among all tested yeast strains, Rhodotorula mucilaginosa DSM 70825 showed the most promising results, demonstrating the highest potential for removing organic substrates and nutrients. Depending on the medium, this strain achieved 50–80% sCOD, 45–60% tVFAs, 21–45% TN, 33–52% PO 4 3− reduction rates. Similar results were obtained for the strain Candida sp. OR687571. The high nutrient and organics removal efficiency by these yeasts could likely be linked to their ability to assimilate xylose (being the main source of carbon in the liquid digestate). In culture media containing liquid digestate, both yeast strains achieved good viability and proliferation potential. In the liquid digestate medium, R. mucilaginosa and Candida sp. showed vitality at the level of 51.5% and 45.0%, respectively. These strains seem to be a good starting material for developing effective digestate treatment strategies involving monocultures and/or consortia with other yeasts or green microalgae.

Abstract:Rhodosporidium toruloides has emerged as an excellent option for microbial lipid production due to its ability to accumulate up to 70% of lipids per cell dry weight, consume multiple substrates such as glucose and xylose, and tolerate toxic compounds. Despite the potential of Rhodosporidium toruloides for high lipid yields, achieving these remains is a significant hurdle. A comprehensive review is essential to thoroughly evaluate the advancements in processes and technologies to enhance lipid production in R. toruloides. The review covers various strategies for enhancing lipid production like co-culture, adaptive evolution, carbon flux analysis, as well as different modes of fermentation. This review will help researchers to better understand the recent developments in technologies for sustainable and scalable lipid production from R. toruloides and simultaneously emphasize the need for developing an efficient and sustainable bioprocess.

Bioremediation of brewery wastewater using Arthrospira sp.: Preliminary assessment of biomass as a biofertilizer toward circular economy

Abstract:Microalgae have been pointed out as promising agents for the bioremediation of wastewater from numerous sources. One of the main advantages of such technology is the production of valuable biomass to be exploited in other processes, increasing the sustainability and circularity of the system. In this study, untreated brewery wastewater was used as culture medium for Arthrospira sp. on a pilot-scale setup positioned within the production site of Nova Rio brewery (Rio de Janeiro, Brazil). The resulting biomass was then preliminarily assessed as a biofertilizer for barley ( Hordeum vulgare ) growth. First, the optimal concentration for the inoculum was determined in lab-scale cultures that were subsequentially scaled up to start a 300-L cultivation of Arthrospira sp. in a 1.55-m 2 open raceway pond. After 100 days of semi-continuous cultivation with a dilution of 0.35 on feeding days, the resulting dry biomass was finally tested as a biofertilizer in different degrees of substitution of a commercial mineral fertilizer. Using an optimal inoculum concentration of 0.5 g L -1 , the onsite pilot-scale cultivation of Arthrospira sp. using Nova Rio's brewery effluent reached an average biomass productivity of 10.68 ± 5.13 g m -2 day -1 . Nitrate, ammonium, and sulfate were successfully removed according to the Brazilian regulations (56.8, 100, and 78.7% removal, respectively). Preliminary biofertilization assays showed that 50% and 100% substitution of commercial fertilizer with raw Arthrospira sp. biomass resulted in faster barley growth (~60%) when compared to solely commercial fertilizer, indicating a promising area for further research and investment.

Co-cultivation of microalgae and bacteria for optimal bioenergy feedstock production in wastewater by using response surface methodology

Abstract:This work uses response surface methodology (RSM) to study the co-cultivation of symbiotic indigenous wastewater microalgae and bacteria under different conditions (inoculum ratio of bacteria to microalgae, CO2, light intensity, and harvest time) for optimal bioenergy feedstock production. The findings of this study demonstrate that the symbiotic microalgae-bacteria culture not only increases total microalgal biomass and lipid productivity, but also enlarges microalgal cell size and stimulates lipid accumulation. Meanwhile, inoculum ratio of bacteria to microalgae, light intensity, CO2, and harvest time significantly affect biomass and lipid productivity. CO2 concentration and harvest time have significant interactive effect on lipid productivity. The response of microalgal biomass and lipid productivity varies significantly from 2.1 × 105 to 1.9 × 107 cells/mL and 2.8 × 102 to 3.7 × 1012 Total Fluorescent Units/mL respectively. Conditions for optimum biomass and oil accumulation are 100% of inoculation ratio (bacteria/microalgae), 3.6% of CO2 (v/v), 205.8 µmol/m2/s of light intensity, and 10.6 days of harvest time. This work provides a systematic methodology with RSM to explore the benefits of symbiotic microalgae-bacteria culture, and to optimize various cultivation parameters within complex wastewater environments for practical applications of integrated wastewater-microalgae systems for cost-efficient bioenergy production.

Abstract:High-performance thin-layer chromatography (HPTLC) has advantages for the analysis of cyclosporine (CsA) and its metabolites in peripheral blood not shared by the radioimmunoassay (RIA) or high-performance liquid chromatography (HPLC) methods. While separation by HPTLC and HPLC is based on relative hydrophobicity, HPTLC (unlike HPLC) is capable of concurrent multisample analysis without expensive instrumentation. As distinguished from RIA, HPTLC detects CsA and metabolites as separate components, and does not use radioactive reagents. A novel rhodamine B/alpha-cyclodextrin stain was developed and the characteristic retention factors (Rf values), as determined by the ratio of the migration distance of a component in relation to the solvent front, were determined for the mobile phase heptane: pyridine: ethyl acetate (100: 75: 1, v/v) on aminopropyl-bonded silica gel HPTLC plates: 0.65, cyclosporin D (CsD); 0.60, CsA; 0.50, dihydrocyclosporin C (dhCsC); 0.42, metabolites M-21 and M-17; 0.40, M-1; 0.35, M-E; 0.25, M-D; and 0.22, M-A. Metabolite M-18 showed migration similar to that of M-17 using a mobile phase of heptane: pyridine: acetonitrile (5:2:1, v/v) in the 0.60-0.50 range. The metabolite profiles were obtained in 8 patients receiving the drug for the first time. The HPTLC analytical technique identifies CsA and its metabolites in peripheral blood and offers advantages for pharmacologic monitoring of transplant patients.
Xylose Metabolism and the Effect of Oxidative Stress on Lipid and Carotenoid Production in Rhodotorula toruloides: Insights for Future Biorefinery

Abstract:The use of cell factories to convert sugars from lignocellulosic biomass into chemicals in which oleochemicals and food additives, such as carotenoids, is essential for the shift toward sustainable processes. Rhodotorula toruloides is a yeast that naturally metabolises a wide range of substrates, including lignocellulosic hydrolysates, and converts them into lipids and carotenoids. In this study, xylose, the main component of hemicellulose, was used as the sole substrate for R. toruloides, and a detailed physiology characterisation combined with absolute proteomics and genome-scale metabolic models was carried out to understand the regulation of lipid and carotenoid production. To improve these productions, oxidative stress was induced by hydrogen peroxide and light irradiation and further enhanced by adaptive laboratory evolution. Based on the online measurements of growth and CO2 excretion, three distinct growth phases were identified during batch cultivations. Majority of the intracellular flux estimations showed similar trends with the measured protein levels and demonstrated improved NADPH regeneration, phosphoketolase activity and reduced β-oxidation, correlating with increasing lipid yields. Light irradiation resulted in 70% higher carotenoid and 40% higher lipid content compared to the optimal growth conditions. The presence of hydrogen peroxide did not affect the carotenoid production but culminated in the highest lipid content of 0.65 g/gDCW. The adapted strain showed improved fitness and 2.3-fold higher carotenoid content than the parental strain. This work presents a holistic view of xylose conversion into microbial oil and carotenoids by R. toruloides, in a process toward renewable and cost-effective production of these molecules.

Abstract:BackgroundMicrobial lipids (triacylglycerols, TAG) have received large attention for a sustainable production of oleochemicals and biofuels. Rhodosporidium toruloides can accumulate lipids up to 70% of its cell mass under certain conditions. However, our understanding of lipid production in this yeast is still much limited, especially for growth with mixed substrates at the level of metabolic network. In this work, the potentials of several important carbon sources for TAG production in R.toruloides are first comparatively studied in silico by means of elementary mode analysis followed by experimental validation.ResultsA simplified metabolic network of R.toruloides was reconstructed based on a combination of genome and proteome annotations. Optimal metabolic space was studied using elementary mode analysis for growth on glycerol, glucose, xylose and arabinose or in mixtures. The in silico model predictions of growth and lipid production are in agreement with experimental results. Both the in silico and experimental studies revealed that glycerol is an attractive substrate for lipid synthesis in R. toruloides either alone or in blend with sugars. A lipid yield as high as 0.53 (C-mol TAG/C-mol) has been experimentally obtained for growth on glycerol, compared to a theoretical maximum of 0.63 (C-mol TAG/C-mol). The lipid yield on glucose is much lower (0.29 (experimental) vs. 0.58 (predicted) C-mol TAG/C-mol). The blend of glucose with glycerol decreased the lipid yield on substrate but can significantly increase the overall volumetric productivity. Experimental studies revealed catabolite repression of glycerol by the presence of glucose for the first time. Significant influence of oxygen concentration on the yield and composition of lipids were observed which have not been quantitatively studied before.ConclusionsThis study provides for the first time a simplified metabolic model of R.toruloides and its detailed in silico analysis for growth on different carbon sources for their potential of TAG synthesis. Experimental studies revealed the phenomenon of catabolite repression of glycerol by glucose and the importance of oxygen supply on the yield and composition of lipids. More systematic studies are needed to understand the mechanisms which should help to further optimize the lipid production in this strain of industrial interest.
