Abstract:Recent advances in video tracking technologies provide the tools for a sensitive and reproducible analysis of invertebrate activity under stressful conditions nurturing the field of behavioural ecotoxicology. This study aimed to evaluate behavioural responses of the freshwater planarian Dugesia subtentaculata exposed to a model compound, chlorantraniliprole (CAP). This compound is an anthranilic diamide insecticide and due to its neurotoxic action can, at low concentrations, impair behaviour of exposed organisms. Behavioural endpoints measured included feeding and locomotor activities. Feeding responses were based on planarian predatory behaviour using Chironomus riparius larvae as prey. Locomotion was measured by the traditional planarian locomotor velocity (pLMV) assay and additionally using an automated video tracking system using a Zebrabox(®) (Viewpoint, France) device. While feeding and pLMV were significantly impaired at 131.7μg/L CAP, the video tracking system showed that total distance covered by planarians was significantly reduced at concentrations as low as 26.2μg/L CAP. Our results show that more advanced automated video recording systems can be used in the development of sensitive bioassays allowing a reliable, time- and cost-effective quantification of behaviour in aquatic invertebrates. Due to their ecological relevance, behavioural responses should not be disregarded in risk assessment strategies and we advocate the suitability of planarians as suitable organisms for behavioural ecotoxicological studies.

Abstract:The toxicological study of pesticides at sub-lethal and environment-relevant concentrations has become increasingly crucial for human and environmental health. Toxic mechanisms of agrochemicals contribute to discovering green pesticides, assessing the hazards of pesticides comprehensively, and supporting legitimate regulatory decisions. However, the toxicological effects of difenoconazole are not yet fully understood despite being frequently detected in fruits, vegetables, waters, and soils and posing hazards to humans and the environment. This lack of knowledge could lead to flawed risk assessment and administrative oversight. Thus, the review aimed to provide some investigation perspectives for clarifying the toxicological effects of difenoconazole by synthesizing the toxic data of difenoconazole on various organisms, such as bees, Daphnia magna , fish, earthworms, mammals, and plants and summarizing the toxicological mechanisms of difenoconazole, especially in fish and mammals from peer-reviewed publications. Evidence revealed that difenoconazole caused multiple toxicological effects, including developmental toxicity, reproductive toxicity, endocrine disruption effects, neurotoxicity, and transgenerational toxicity. The toxic mechanisms involved in metabolic disturbance, oxidative stress, inflammation, apoptosis, and autophagy by activating reactive oxygen species-mediated signaling pathways and mitochondrial apoptosis routes, disturbing amino acids, lipid, and nucleotide metabolism, and regulating gene transcription and expression in mammals and fish. Based on the review, further studies better focus on the toxic differences of difenoconazole stereoisomers, the toxicological effects of transformation products of difenoconazole, and the mechanism of action of difenoconazole on sex-specific endocrine disruption effects, intestinal damage, and gut dysbacteriosis for its hazard assessment and management synthetically.

Abstract:The ability of aquatic organisms to sense the surrounding environment chemically and interpret these signals correctly is crucial to their survival and ecological niche. This study applied the Heterogenous Multi-Habitat Assay System - HeMHAS to evaluate the avoidance potential of Daphnia magna to detect fipronil-contaminated habitats in a connected landscape after a short (48 h), previous, forced exposure to an environmentally relevant concentration of the same insecticide. The swimming of daphnids was also analyzed by recording the total distance covered. D. magna preferred areas with less contamination, although the effect of fipronil on their swimming ability (a decrease) was observed for all the concentrations tested. The application of non-forced multi-compartment exposure methodologies is a recent trend and is ecologically relevant as it is based on how contamination can really produce changes in an organism's habitat selection. Finally, we consider the importance of more non-forced exposure approaches where Stress Ecology can be aggregated to improve systemic understanding of the risk that contaminants pose to aquatic ecosystems from a broader landscape perspective.

Abstract:Antifungals support modern medical and agricultural practices, and extensive screening environmental data are missing. This work aims to survey antifungals for medical and agricultural purposes in surface- and groundwater in Portugal. A passive sampling technique and a high-resolution chromatographic system were used. Antifungals applied in medical practice were not detected as only antifungals for agricultural purposes (fungicides). Thirty-nine different fungicide molecules were found; its detection frequency in surface- and groundwater was dominated by rabenzazole (61%, 92%) and ethoxyquin (54%, 77%); among the five fungicides with similar surface- and groundwater catching rates, ferimzone was the most polluting (54%, 54%); oxadixyl (61%), kresoxim-methyl (61%) and fenamidone (46%) were primarily designated surface water contaminants; for azoles, the occurrence in surface water ranged from a residual (10%) to a moderate detection rate for two compounds (31%—propiconazole and tebuconazole). Surprisingly, only 51% of detected fungicides are authorised. The frequently detected fungicides that are either not authorised (ethoxyquin and its impurity, furmecyclox, oxadixyl), without data in the EU (rabenzazole, ferimzone), or authorised at the national level (fenamidone), should be included in environmental monitoring programmes and followed as emerging (micro)contaminants. Policy makers should gather their efforts to allow the implementation of proper risk management and effective contamination control strategies to achieve Sustainable Development Goals.

Abstract:The integration of untargeted lipidomics approaches in ecotoxicology has emerged as a strategy to enhance the comprehensiveness of environmental risk assessment. Although current toxicity tests with soil microarthropods focus on species performance, that is, growth, reproduction, and survival, understanding the mechanisms of toxicity across all levels of biological organization, from molecule to community is essential for informed decision‐making. Our study focused on the impacts of sublethal concentrations of the insecticide teflubenzuron on the springtail Folsomia candida. Untargeted lipidomics was applied to link changes in growth, reproduction, and the overall stress response with lipid profile changes over various exposure durations. The accumulation of teflubenzuron in organisms exposed to the highest test concentration (0.035 mg a.s. kg–1 soil dry wt) significantly impacted reproductive output without compromising growth. The results suggested a resource allocation shift from reproduction to size maintenance. This hypothesis was supported by lipid shifts on day 7, at which point reductions in triacylglycerol and diacylglycerol content corresponded with decreased offspring production on day 21. The hypermetabolism of fatty acids and N‐acylethanolamines on days 2 and 7 of exposure indicated oxidative stress and inflammation in the animals in response to teflubenzuron bioaccumulation, as measured using high‐performance liquid chromatography–tandem mass spectrometry. Overall, the changes in lipid profiles in comparison with phenotypic adverse outcomes highlight the potential of lipid analysis as an early‐warning tool for reproductive disturbances caused by pesticides in F. candida. Abstract:Leaf litter decomposition is a key ecological process in freshwater ecosystems. Fungi, particularly aquatic hyphomycetes, play a major role in organic matter turnover and constitute a pivotal node in detrital food webs. The extensive use of antifungal formulations, which include agrochemicals and pharmaceuticals, is a threat to biodiversity and may affect non-target microbial and invertebrate decomposer communities. Using a laboratory approach, we assessed the effects of tebuconazole (agrochemical), clotrimazole and terbinafine (pharmaceuticals) on aquatic communities and on the decomposition of plant litter. Alder leaves were colonized by natural microbiota in a clean stream, and then exposed in microcosms to 8 concentrations of each fungicide (10 to 1280 μg L-1). Fungicides led to shifts in species dominance in all tested concentrations, but no effects on leaf decomposition were observed. In addition, tebuconazole and clotrimazole strongly reduced fungal biomass and reproduction, whilst terbinafine stimulated fungal reproduction at lower concentrations but had no measurable effects on fungal biomass. Subsequently, the indirect effects of the fungicides were assessed on the next trophic level (detritivore invertebrates), by evaluating leaf consumption by a specialist (Allogamus sp.) and a generalist (Chironomus riparius) species, when feeding on fungicide-preconditioned leaves. The feeding activity of C. riparius and Allogamus sp. was not affected, and as expected, specialists were more efficient than generalists in exploring leaves as a dietary resource. However, results indicated that these fungicides have direct negative effects on microbial decomposers, and thus may compromise ecosystem functions on the long term.
Toxicity of fungicides to terrestrial non-target fauna - Formulated products versus active ingredients (azoxystrobin, cyproconazole, prothioconazole, tebuconazole) - A case study with Enchytraeus crypticus (Oligochaeta)

Abstract:Despite the high usage of pesticides in current agricultural practices, its effects to humans and to the environment (non-target species) are a continuous concern. Soil dwelling organisms are among the first in line of exposure to pesticides, however their risks are often based on the pure active ingredient (a.i.) and not on the commercial formulated products (FPs) actually applied in the fields. In the present study, we investigated the effects of two fungicide FPs versus its a.i. (s): Amistar® XTRA and the respective a.i. (s) azoxystrobin and cyproconazole, and Prosaro® 250 EC and the respective a.i. (s) prothioconazole and tebuconazole, to the non-target soil oligochaete Enchytraeus crypticus. The standard Enchytraeid Reproduction Test was used to assess effects on survival and reproduction. Results showed that Amistar was more toxic than Prosaro, particularly for reproduction (EC50 = 161 mg Amistar/kg soil, EC50 = 350 mg Prosaro/kg soil). For both FPs, reproductive effects were mainly related to one of its a.i. (s) (azoxystrobin [EC50 = 37 mg azosxystrobin/kg soil] for Amistar, and tebuconazole [EC50 = 41 mg tebuconazole/kg soil] for Prosaro), while lethal effects were not predicted by the toxicity of its a.i. (s) (particularly in the case of Prosaro, which was more toxic than its a.i. (s)). These findings highlight the need to further explore the toxicity data of the FPs compared to the a.i. (s), aiming to predict a more realistic environmental hazard of pesticides.
Insights into the effects of difenoconazole on the livers in male mice at the biochemical and transcriptomic levels

Abstract:Difenoconazole (DFZ) is a broad-spectrum triazole fungicide, that is extensively used in agriculture. Studies have shown that residues of DFZ and other fungicides have toxic effects on nontarget organisms. However, its hepatoxicity in mammals remains unclear. Here, we characterized the toxic hepatic effects in male C57BL/6 mice exposed to 30 and 100 mg/kg bw DFZ for 14 and 56 days, respectively. The results revealed that DFZ could increase the relative liver weights, however, the relative fat and spleen weights decreased. More importantly, DFZ exposure changed the hepatic morphology and induced hepatic oxidative stress. Gene expression analysis suggested that DFZ could induce a glycolipid metabolism disorder. Moreover, hepatic transcriptomic analysis revealed the effects of DFZ exposure on the transcriptional levels of various genes, and enrichment analysis of differentially expressed genes (DEGs) showed that energy metabolism and immune-associated pathways were mainly affected. We validated the results from transcriptomic analysis and found that some key genes related to energy metabolism were affected. In addition, flow cytometry showed that the CD3+/CD4+ and CD3+ /CD8+ levels declined in the spleen of mice. Taken together, these findings combined with transcriptome analysis highlighted that DFZ caused different endpoints in the liver, which could provide more evidence for investigating the toxic effects of DFZ in mammals.

Abstract:One of the commonest pesticides used in agriculture is the fungicide carbendazim, which can cause deleterious effects on the physiology and behaviour of acutely and chronically exposed animals. However, few studies have evaluated the effects of carbendazim on fish behaviour and our main objective was to evaluate behavioural changes on adult zebrafish ( Danio rerio ) exposed to this fungicide. We studied 177 fishes, divided into control groups (not exposed) and treatment groups, exposed to a concentration of 120 μg/L for 7, 14, 21 and 28. Behavioural data were collected using the scan sampling, with instantaneous recording every 30 s. We evaluated and compared fish behaviours and aquarium occupancy between treatments and controls using Generalized Linear Mixed Models. The results indicate that carbendazim initially elicited slow swimming and later fast swimming, decreased aggressiveness, and caused behavioural changes indicative of distress, like abnormal swimming and the gulping of air in water surface. In the wild, this behavioural change can result in an increased predation risk and consequent decrease or extinction of populations, revealing important ecological issues for animal communities living in polluted water bodies.

Abstract:Fungicides play an important role in crop protection, but they have also been shown to adversely affect non-target organisms, including those living in the aquatic environment. The aim of the present study is to combine experimental and computational approaches to evaluate the effects of flutriafol, metconazole, myclobutanil, tebuconazole, tetraconazole and triticonazole on aquatic model organisms and to obtain information on the effects of these fungicides on Lemna minor, a freshwater plant, at the molecular level. The EC50 (the half-maximum effective concentration) values for the growth inhibition of Lemna minor in the presence of the investigated fungicides show that metconazole (EC50 = 0.132 mg/L) and tetraconazole (EC50 = 0.539 mg/L) are highly toxic, tebuconazole (EC50 = 1.552 mg/L), flutriafol (EC50 = 3.428 mg/L) and myclobutanil (EC50 = 9.134 mg/L) are moderately toxic, and triticonazole (EC50 = 11.631 mg/L) is slightly toxic to this plant. The results obtained with the computational tools TEST, ADMETLab2.0 and admetSAR2.0 also show that metconazole and tetraconazole are toxic to other aquatic organisms: Pimephales promelas, Daphnia magna and Tetrahymena pyriformis. A molecular docking study shows that triazole fungicides can affect photosynthesis in Lemna minor because they strongly bind to C43 (binding energies between −7.44 kcal/mol and −7.99 kcal/mol) and C47 proteins (binding energies between −7.44 kcal/mol and −8.28 kcal/mol) in the reaction center of photosystem II, inhibiting the binding of chlorophyll a to these enzymes. In addition, they can also inhibit glutathione S-transferase, an enzyme involved in the cellular detoxification of Lemna minor.

Abstract:Predation presents specific behavioral characteristics for each species, and the interaction between prey and predator influences the structuring of the food web. Concerning insects, predation can be affected in different ways, such as exposure to chemical stressors, e.g., pesticides. Therefore, analyses were carried out of the effects of exposure to insecticide fipronil and the herbicide 2,4-D on predation, parameters of food selectivity, and the swimming behavior of two neotropical predatory aquatic insects of the families Belostomatidae (giant water bugs) and larvae of Libellulidae (dragonfly). These predatory insects were exposed for 24 h to a commercial formulation of the chlorophenoxy herbicide, 2,4-D at nominal concentrations of 200, 300, 700, and 1400 μg L −1 , and to a commercial formulation of the phenylpyrazole insecticide, fipronil at nominal concentrations of 10, 70, 140, and 250 μg L −1 . In a control treatment, the insects were placed in clean, unspiked water. At the end of the exposure, the maximum swimming speed of the predators was evaluated. Afterward, the predators were placed in clean water in a shared environment for 24 h with several prey species, including the cladoceran Ceriodaphnia silvestrii , larvae of the insect Chironomus sancticaroli , the amphipod Hyalella meinerti , the ostracod Strandesia trispinosa , and the oligochaete Allonais inaequalis for 24 h. After this period, the consumed prey was counted. The results reveal that predators from both families changed prey consumption compared with organisms from the control treatment, marked by a decrease after exposure to fipronil and an increase in consumption caused by 2,4-D. In addition, there were changes in the food preferences of both predators, especially when exposed to the insecticide. Exposure to fipronil decreased the swimming speed of Belostomatidae individuals, possibly due to its neurotoxic effect. Exposure to the insecticide and the herbicide altered prey intake by predators, which could negatively influence the complex prey–predator relationship and the functioning of aquatic ecosystems in contaminated areas.

Abstract:Carbofuran (CF) is a carbamate class pesticide, widely used in agriculture for pest control in crops. On the other hand, CF is also detected as a contaminant in food and water sources. This pesticide has high toxicity in non-target organisms, and its presence in the environment poses a threat to the ecosystem. Research has revealed that this pesticide acts as an inhibitor of acetylcholinesterase (AChE), inducing an accumulation of acetylcholine in the brain. Consequently, this leads to the emergence of a cholinergic syndrome and various detrimental effects on human health. Nonetheless, our understanding of CF impact on the central nervous system remains elusive. Therefore, this study aimed to investigate the effects of CF on behavioral and neurochemical parameters in adult zebrafish. The animals were exposed for 96 hours to different concentrations of CF (5, 50, and 500 µg/L) and subjected to behavioral assessments in the novel tank test (NTT) and social preference test (SPT). Subsequently, they were euthanized, and their brains were used to evaluate neurochemical markers associated with oxidative stress and AChE levels. In the NTT and SPT, CF did not alter the evaluated behavioral parameters. Furthermore, CF did not affect the levels of AChE, non-protein sulfhydryl groups, and thiobarbituric acid reactive species in the zebrafish brain. However, further research is needed regarding the environmental exposure of this compound to non-target organisms.

Abstract:To establish meaningful policy directives for sustainable agrochemical use, we require baseline knowledge of the impacts of agrochemicals on non-target organisms. The widespread use of the herbicide glyphosate has resulted in the global presence of its metabolite, aminomethylphosphonic acid (AMPA). AMPA is commonly found in water bodies, including freshwater systems. We investigated the effects of AMPA exposure on the survivorship, regenerative abilities, and locomotion of the brown planarian (Girardia tigrina), a water-dwelling flatworm commonly found in freshwater ecosystems. In a series of experiments, we bisected and then exposed planarians to realistic field doses of AMPA for seven days and then fourteen days. For the 14-day experiment, we exposed planarians to two concentrations consistent with the high and low ranges of concentrations observed in water systems. Compared to the control group, we found that planarians exposed to AMPA for fourteen days (un-bisected for the first seven days and recovering from bisection for the subsequent seven) exhibited slower regeneration from the tail segment. Our findings highlight the potential ecological impacts of AMPA contamination on planarian populations. Quantifying the effects of AMPA exposure on planarians contributes to our understanding of the ecological consequences of our current and common agricultural practices on our freshwater ecosystems.

Effects of difenoconazole on hepatotoxicity, lipid metabolism and gut microbiota in zebrafish (Danio rerio)

Abstract:In current study, larvae and adult zebrafish were exposed to difenoconazole to assess its effect on hepatotoxicity, lipid metabolism and gut microbiota. Results demonstrated that difenoconazole could induce hepatotoxicity in zebrafish larvae and adult, 0.400, 1.00, 2.00 mg/L difenoconazole caused yolk retention, yolk sac edema or liver degeneration after embryos exposure for 120 h, hepatocyte vacuolization and neoplasm necrosis were observed in adult liver after 0.400 mg/L difenoconazole exposure for 21 d. RNA sequencing showed that the 41 and 567 differentially expressed genes in zebrafish larvae and liver induced by 0.400 mg/L difenoconazole, were concentrated in pathways related to protein digestion and absorption, pancreatic secretion, steroid biosynthesis, and different metabolic pathways including galactose or sugar metabolism. Difenoconazole exposure caused lipid accumulation in larval yolk sac, and the elevated triglyceride (TG), malondialdehyde (MDA) and reactive oxygen species (ROS) levels in larvae and liver, which further confirmed the lipid metabolism disorders induced by difenoconazole. The results further showed that difenoconazole increased the abundance of gut microbiota such as Firmicutes, Aeromonas, Enterobacteriaceae and Bacteroides, further suggested that gut microbiota might participate in lipid metabolism and hepatotoxicity during zebrafish development. These findings advanced the field of the difenoconazole-induced developmental toxicity in larvae and adult zebrafish, and the imbalance of gut microbiota provided the plausible mode of action for the liver damage and disordered lipid metabolism in zebrafish.
Fipronil Risk Assessment in Tropical Soils: Relevant Concentrations Cause High Risk to the Edaphic Community

Abstract:Widely used in agricultural fields, fipronil acts in the arthropods' nervous systems. Although advances have been noticed, little is known about the effects of the insecticide to non-target soil organisms, especially considering specific parameters from tropical environments. This is the first study considering ecologically relevant tropical conditions to assess the environmental risk of fipronil to the terrestrial ecosystem. The present study aimed to estimate fipronil's ecological risk to soil organisms through a tiered approach. At the screening level (TIER I) data were obtained from both the literature and laboratory experiments. For the intermediate level (TIER II), species sensitivity distributions (SSD) curves were generated based on ecotoxicological data from six species of soil invertebrates in two soil types. To estimate the risk, the Toxicity-Exposure Ratios (TER) were calculated at each tier considering the related toxicity data and the fipronil concentrations, which were measured and predicted in the field. Results showed that ecologically relevant concentrations of fipronil lead to a high risk for the edaphic community (above 100 times higher than the established safety value). The high risks were observed for both tiers, although at TIER II (SSD approach) it was at least 2.5 times higher than at TIER I. It was also discussed here that the pesticide mode of action is the factor most influencing toxicity; the estimated risk considering artificial soil was comparable to that with the natural soil; and estimating risk using only one species (as performed in the tropics) may underestimate the risk to the edaphic community.

Abstract:The search for new methods in the toxicology field has increased the use of early life stages of zebrafish ( Danio rerio ) as a versatile organism model. Here, we use early stages of zebrafish to evaluate glyphosate as pure active ingredient and within a commercial formulation in terms of oxidative stress. Biomarkers involved in the oxidative status were evaluated along with other markers of neurotoxicity, genotoxicity, cytotoxicity, energy balance and motor performance, and the selected tools were evaluated by its sensitivity in determining early-warning events. Zebrafish embryos exposed to glyphosate active ingredient and glyphosate-based formulation were under oxidative stress, but only the commercial formulation delayed the embryogenesis, affected the cholinergic neurotransmission and induced DNA damage. Both altered the motor performance of larvae at very low concentrations, becoming larvae hypoactive. The energy balance was also impaired, as embryos under oxidative stress had lower lipids reserves. Although data suggest that glyphosate-based formulation has higher toxicity than the active ingredient itself, the most sensitive biomarkers detected early-warning effects at very low concentrations of the active ingredient. Biochemical biomarkers of defense system and oxidative damage were the most sensitive tools, detecting pro-oxidant responses at very low concentrations, along with markers of motor performance that showed high sensitivity and high throughput, suitable for detecting early effects linked to neurotoxicity. Alterations on morphology during embryogenesis showed the lowest sensitivity, thus morphological alterations appeared after several alterations at biochemical levels. Tools evaluating DNA damage and cell proliferation showed mid-sensitivity, but low throughput, thus they could be used as complementary markers.

Abstract:Current standard toxicity tests on nontarget soil invertebrates mainly focus on the endpoints survival and reproduction. Such results are likely insufficient to predict effects at higher organizational levels, for example, the population level. We assessed the effects of exposure to the pesticide teflubenzuron on the collembolan Folsomia candida, by performing a full life‐cycle experiment exposing single individuals via contaminated food (uncontaminated control and 0.2, 0.32, 0.48, 0.72, 1.08, and 1.6 mg/kg dry yeast). Several life‐history traits were considered by following the growth and development of newly hatched individuals over a period of 65 days. We assessed survival, body length, time to first oviposition, cumulative egg production, and hatchability of eggs. A two‐stage model was applied to calculate the population growth rate (λ) combined with elasticity analysis to reveal the relative sensitivity of λ to the effects of teflubenzuron on each life‐history parameter. Body length was the least sensitive life‐history parameter (median effective concentration = 1.10 mg teflubenzuron/kg dry yeast) followed by time to first oviposition (0.96 mg/kg), survival (median lethal concentration = 0.87 mg/kg), cumulative egg production (0.32 mg/kg), and egg hatchability (0.27 mg/kg). Population growth decreased with increasing concentrations of teflubenzuron (λ = 1.162/day in control to 1.005/day in 0.72 mg/kg dry yeast, with populations going extinct at 1.08 and 1.6 mg/kg dry yeast). Elasticity analysis showed that changes in juvenile survival had a greater impact on the population growth rate compared with the other life‐history traits. Our study provides a comprehensive overview of individual‐level effects of long‐term exposure to teflubenzuron and integrates these effects to assess the potential risk to collembolan populations.
