Abstract:We report on a 16-year-old girl with a unique neuromuscular disorder characterised by progressive proximal muscle weakness and numerous tubular aggregates, intracytoplasmic, as well as intranuclear inclusions of the IBM type in her muscle biopsy. The clinical features of the presented case, as manifested by the early childhood onset of the disease, proximal weakness, lumbar hyperlordosis, and bilateral Achilles tendon contractures, were suggestive of congenital myopathy. To the best of our knowledge, the coexistence of tubular aggregates and tubulofilamentous inclusions of the IBM type in a child has never been described.
Molecular and Cytological Identification of Wheat-Thinopyrum intermedium Partial Amphiploid Line 92048 with Resistance to Stripe Rust and Fusarium Head Blight

Abstract:Thinopyrum intermedium (2n = 6x = 42, EeEeEbEbStSt or JJJsJsStSt) contains a large number of genes that are highly adaptable to the environment and immune to a variety of wheat diseases, such as powdery mildew, rust, and yellow dwarf, making it an important gene source for the genetic improvement of common wheat. Currently, an important issue plaguing wheat production and breeding is the spread of pests and illnesses. Breeding disease-resistant wheat varieties using disease-resistant genes is currently the most effective measure to solve this problem. Moreover, alien resistance genes often have a stronger disease-resistant effect than the resistance genes found in common wheat. In this study, the wheat-Th. intermedium partial amphiploid line 92048 was developed through hybridization between Th. intermedium and common wheat. The chromosome structure and composition of 92048 were analyzed using ND-FISH and molecular marker analysis. The results showed that the chromosome composition of 92048 (Octoploid Trititrigia) was 56 = 42W + 6J + 4Js + 4St. In addition, we found that 92048 was highly resistant to a mixture of stripe rust races (CYR32, CYR33, and CYR34) during the seedling stage and fusarium head blight (FHB) in the field during the adult plant stage, suggesting that the alien or wheat chromosomes in 92048 had disease-resistant gene(s) to stripe rust and FHB. There is a high probability that the gene(s) for resistance to stripe rust and FHB are from the alien chromosomes. Therefore, 92048 shows promise as a bridge material for transferring superior genes from Th. intermedium to common wheat and improving disease resistance in common wheat.

Abstract:Agropyron cristatum (2n = 4x = 28, PPPP), which harbours many high-yield and disease-resistance genes, is a promising donor for wheat improvement. Narrow genetic diversity and the trade-off between grain weight and grain number have become bottlenecks for increasing grain yield in wheat. In this study, a novel translocation line, WAT650l, was derived from the chromosome 6P addition line 4844–12, which can simultaneously increase both grain number per spike (GNS) and thousand-grain weight (TGW). Cytological analysis and molecular marker analysis revealed that WAT650l was a 5BL·5BS-6PL (bin 12–17) translocation line. Assessment of agronomic traits and analysis of the BC4F2 and BC5F2 populations suggested that the 6PL terminal chromosome segment in WAT650l resulted in increased grain number per spike (average increased by 14.07 grains), thousand-grain weight (average increased by 4.31 g), flag leaf length, plant height, spikelet number per spike and kernel number per spikelet during the two growing seasons of 2020–2021 and 2021–2022. Additionally, the increased GNS locus and high-TGW locus of WAT650l were mapped to the bins 16–17 and 12–13, respectively, on chromosome 6PL by genetic population analysis of three translocation lines. In , we provide a valuable germplasm resource for broadening the genetic base of wheat and overcoming the negative relationship between GNS and TGW in wheat breeding.

Abstract:The wild species Thinopyrum intermedium (genome JJJSJSStSt) serves as a valuable germplasm resource providing novel diseases resistance and agronomically important genes for wheat improvement. Two wheat-Th. intermedium partial amphiploids, TAI7045 (2n = 56) and 78784 (2n = 56), exhibit high resistance to stripe rust and powdery mildew, and their chromosome constitutions have been characterized. With the aim to transfer novel resistance genes from Th. intermedium, the crosses of common wheat line MY11 with TAI7045 and 78784 were produced, and their individual F2-F5 progenies were characterized using sequential non-denaturing fluorescence in situ hybridization (ND-FISH) and molecular markers. We identified a set of wheat-Th. intermedium addition lines, involving the chromosomes 1St-JS, 2St, 2St-JS, 3St, 4J, 4St, 5St, 5J.St, 6JS.J, and 7JS. Above all, the stable wheat-Th. intermedium small segmental translocation lines with chromosomes 4DS.4DL-4StL-4DL-4JL and 4DS.4DL-4StL-4DL were selected. Combining data from specific marker amplification and resistance evaluation, we mapped the gene(s) for resistance to powdery mildew and stripe rust in the 233.56–329.88 Mb region of the long arm of the 4St chromosome from the reference Th. intermedium genome. The new wheat-Th. intermedium introgressions will be used as novel germplasm for breeding purposes.

Abstract:Thinopyrum intermedium (JJJ s J s StSt, 2 n = 6 x = 42), a member of tertiary gene pool of hexaploid wheat ( Triticum aestivum L., AABBDD, 2 n = 6 x = 42), provides several beneficial genes for wheat improvement. In this study, line CH51 was developed from the BC 1 F 8 progeny of a partial wheat- Th. intermedium amphiploid TAI8335 (2 n = 56) and wheat cultivar (cv.) Jintai 170. Somatic metaphase chromosome counting showed that CH51 had stable 42 chromosomes. Genomic in situ hybridization (GISH) analysis showed that CH51 had 40 wheat chromosomes and two Th. intermedium chromosomes involving translocation between J s - and St-genome chromosomes. Non-denaturing fluorescence in situ hybridization (ND-FISH) analysis revealed that CH51 lacked a pair of wheat chromosome 6B. Wheat 55K SNP array analysis verified that chromosome 6B had the highest percentage of missing SNP loci in both CH51 and Chinese Spring (CS) nullisomic 6B-tetrasomic 6D (CS-N6BT6D) and had the highest percentage of polymorphic SNP loci between CH51 and cv. Jintai 170. We identified that CH51 was a wheat- Th. intermedium T6StS.6J s L (6B) disomic substitution line. Disease resistance assessment showed that CH51 exhibited high levels of resistance to the prevalent Chinese leaf rust and stripe rust races in the field. Therefore, the newly developed line CH51 can be utilized as a potential germplasm in wheat disease resistance breeding.

Abstract:Wheat is one of the most important food and protein sources in the world and although, in recent years wheat breeders have achieved yield gains, they are not sufficient to meet the demands of an ever-growing population. Development of high yielding wheat varieties, resilient to abiotic and biotic stress resulting from climate change, has been limited by wheat’s narrow genetic base. In contrast to wheat, the wild relatives of wheat provide a vast reservoir of genetic variation for most, if not all, agronomic traits. Previous studies by the authors have shown the transfer of genetic variation from T. urartu into bread wheat. However, before the introgression lines can be exploited for trait analysis, they are required to have stable transmission of the introgressions to the next generation. In this work, we describe the generation of 86 doubled haploid (DH) wheat- T. urartu introgression lines that carry homozygous introgressions which are stably inherited. The DH lines were characterised using the Axiom ® Wheat Relative Genotyping Array and 151 KASP markers to identify 65 unique T. urartu introgressions in a bread wheat background. DH production has helped accelerate the breeding process and facilitated the early release of homozygous wheat- T. urartu introgression lines. Together with the KASP markers, this valuable resource could greatly advance identification of beneficial alleles that can be used in wheat improvement.

Abstract:Wheat-Agropyron cristatum 7P disomic addition line Ⅱ-5-1, derived from the distant hybridization between A. cristatum (2n = 4x = 28, PPPP) and the common wheat cv. Fukuhokomugi (Fukuho), displays numerous desirable agronomic traits, including enhanced thousand-grain weight, smaller flag leaf, and enhanced tolerance to drought. In order to transfer these traits into common wheat, Ⅱ-5-1 was induced by 60Co-γ ray, leading to the creation of 18 translocation lines and three deletion lines. Genomic in situ hybridization (GISH) and fluorescence in situ hybridization (FISH) indicated that multiple wheat chromosomes were involved in the translocation events, including chromosome 2A, 3A, 5A, 7A, 3B, 5B, 7B, 3D and 7D. A. cristatum 7P chromosome was divided into 15 chromosomal bins with fifty-five sequence-tagged site (STS) markers specific to A. cristatum 7P chromosome. Seven and eight chromosomal bins were located on 7PS and 7PL, respectively. The above-mentioned translocation and deletion lines each contained different, yet overlapping 7P chromosomal fragments, covering the entire A. cristatum 7P chromosome. Three translocation lines (7PT-13, 7PT-14 and 7PT-17) and three deletion lines (del-1, del-2 and del-3), which contained the common chromosomal bins 7PS1-3, displayed higher thousand-grain weigh than Fukuho, suggesting that potential genes conferring high thousand-grain weigh might be located on these chromosomal bins. Therefore, wheat-A. cristatum 7P translocation lines with elite traits will be useful as novel germplasms for wheat genetic improvement.

Abstract:By backcrossing to the clonal stocks of perennial F1's between common wheat (including an amphidiploid, timopheevi-squarrosa) and Agropyron glaucum with wheat pollens, 11 mature BF1 plants were obtained. Among female stocks used, there were some clones derived from F1's which failed to multiply their chromosomes in spite of their being treated with colchicine in the first year. As a result of such careless selection of material, 7 out of 11 BF1 plants showed the abnormalities of either 2n chromosome numbers or meiotic chromosome behaviours. Namely, some possessed more than 63 somatic chromosomes, surpassing theoretical 2n numbers to be expected in BF1's and others showed the formation of aneuploid cells (chromosome mosaics), syncytes, telocentrics or a much quantity of multivalent chromosomes. Normal plants assumed to be derived from normal F1's were only four, one of which, the plant No. 4, showed simple associations having its modal configuration in 25II+11I (30%). Meiotic chromosome behaviour and fertility were in good agreement. Normal four plants showed relatively high fertility in pollens and seeds. The chromosome numbers of 4 BF1 plants obtained from F1 (Martin×A. glaucum)×Fultz No. 1 in 1958 were 48, 51, 58 and 59 respectively. No plant was to be found among them having more than 63 chromosomes, although every one of them showed a lot of multivalents.Culms of BF1's were tall, ears approaching vulgare-type but long and lax. The tillering-ability was so great that young tillers sprouted constantly throughout the summer like a perennial, but withered up and died after overwinter. An individual of plant No. 11, which possessed telocentric chromosomes, was dwarf, had a few tillers, and ended its life before winter. They all were highly resistant to the most virulent race group 21B of leaf-rust, Puccinia triticina Eriks.

Abstract:Background. To breed drought-resistant cultivars of common wheat1 (Triticum aestivum L.), it is important to use the gene pools of its relatives, including tall wheatgrass Thinopyrum ponticum (Podpěra) Z.-W. Liu & R.- C. Wang (= Agropyron elongatum (Host) Beauv.). Materials and methods. The introgressive lines of spring common wheat with T. ponticum genetic material and standard cultivars were studied in the field in the southern forest-steppe of Western Siberia using generally recognized methods. The ecological plasticity of cultivars and introgressive lines by grain yield and yield components was calculated according to the method of S. A. Eberhart and W. A. Russell. During the research period, there was a prolonged drought in 2012, and irregular short severe droughts occurred in 2013, 2014, and 2017. Results. An analysis of the ecological plasticity of standard cultivars adapted to the regional conditions showed that cv. ‘Pamyati Azieva’ corresponded to the extensive type, and cvs. ‘Duet’, and ‘Erythrospermum 59’ corresponded to the intensive type. Under drought conditions, the grain yield of cv. ‘Pamyati Azieva’ was determined by the stable development of productive tiller number, seed number and grain yield per main ear, but plasticity in 1000 grain weight was observed. Cvs. ‘Duet’ and ‘Erythrospermum 59’ showed ecological plasticity due to the adaptive development of two or three yield components. Introgressive lines exceeded the standard cultivars in grain yield (1.1–2.2 times) in dry seasons. Five lines were similar to cv. ‘Pamyati Azieva’ in plasticity and stability, and under drought conditions they demonstrated high and stable development of three or four yield components. The intensive lines formed their yield due to the compensatory development of three yield components in different combinations. Conclusion. Introgressive lines with T. ponticum genetic material are valuable for breeding spring common wheat cultivars with various drought-adaptation mechanisms.

Abstract:Agropyron cristatum (L.) Gaertn. (2n = 4x = 28, PPPP), which is a wild relative of common wheat and has a P genome, has many excellent characteristics (for example, it is disease-resistant, stress-resistant, and high yielding) and has important utilization values for the genetic improvement of wheat. In this study, the wheat– A. cristatum 3P addition line I-27 and 3P(3B) substitution line I-15 were identified for the first time via in situ hybridization (ISH) and specific molecular marker (EST-STS) from wheat– A. cristatum -derived lines, which are a representative and important material for the utilization of the excellent genes from the A. cristatum chromosome 3P for wheat genetic improvement. In terms of agronomic traits, the tiller number and spike length of the 3P addition line I-27 were significantly higher than those of Fukuhokomugi. The spike length and spikelet number per spike of the 3P substitution line I-15 were significantly greater than those of Fukuho. Additionally, the wheat– A. cristatum 3P substitution line I-15 was resistant to wheat leaf rust. The generation of the 3P addition line I-27 and the 3P substitution line I-15 was essential for the transfer of the excellent genes of the A. cristatum 3P chromosome into common wheat, and resulted in the wheat– A. cristatum addition line containing a complete set of genetic research materials, laying the foundation for comparative studies of the P genome and subsequent fine mapping.

Abstract:Enrichment of grains with anthocyanins is considered a feasible approach to improving the nutritional properties of bread wheat. Here, two black-grained substitution lines with either 4B or 4D chromosomes substituted by wheatgrass (Thinopyrum ponticum) 4Th were created via marker-assisted combining the anthocyanin biosynthesis regulatory genes Pp-1, Pp3 (Purple pericarp), and Ba1 (Blue aleurone) in the genetics background of cv. Saratovskaya 29. The black grains manifested the simultaneous accumulation of anthocyanins in aleurone, and the pericarp layers resulted in the highest total anthocyanin content (TAC), which amounted to approximately the sum of TACs of the purple and blue grains. The lines with substitution 4Th(4B) had long trichomes on the leaves, whereas the lines with substitution 4Th(4D) did not differ from parental cv. Saratovskaya 29. Despite the chromosome substitution having a positive effect on the main spike length, the other yield-related traits (grain weight per main spike, the spike number per plant, and the 1000-grain weight) were decreased in the lines with chromosome substitution. The developed lines together with the parental ones constitute a valuable model for comparative studies and genetic stock for breeding commercial wheat cultivars featuring high levels of anthocyanins in grains.

[Early treatment discontinuation in tuberculosis hospital].

Abstract:By analyzing the data on patients (n = 17295) who were discharged from 5 regional tuberculosis hospitals of Central Russia, the authors defined and assessed the major factors showing a steady-state strong correlation with early treatment termination through a patient's fault. Descriptive and univariate analyses and multifactor modeling were made to determine interfering factors and systematic errors (logistic regression modeling). For inpatient treatment at a tuberculosis hospital, a logistic regression model was used to develop a predictive decision rule that would determine the probability of treatment discontinuation, the quality control of the predictive rule on the basis of the bootstrap method. The rule makes it possible to form high and very high discontinuation risk groups in order to organize target measures to enhance the efficiency of tuberculosis control.

Abstract:Abstract Diseases adversely affect grain yield of crop plants. Leaf rust is a major disease of wheat. As race-specific resistance breaks down, introduction of newer sources of resistance often from older accessions is necessary. Linkage drag from the donor accessions adversely impacts grain yield. As a result, CIMMYT breeding effort has shifted to using durable resistance, also referred to as race non-specific host resistance, which allows slow rusting but maintains grain yield. One of the key genes for durable resistance is Lr67 and the resistant form of this gene is absent in CIMMYT elite lines. Hence, we have initiated efforts to convert the susceptible copy of Lr67 into its resistant form directly in these elite lines using gene editing. This would eliminate backcrossing and thus save time as well as eliminate linkage drag that would accompany the resistant copy of the gene if it were to be introgressed from an older accession. As first steps, we have isolated and sequenced the genomic copies from each of the A, B, and D genome of the Lr67 gene from three elite lines and an experimental line. Identification of more than 50 single nucleotide polymorphisms (SNPs) in the open reading frames (ORF) among these lines would be useful in designing the guide RNA molecules with precision. Further, we have streamlined genetic transformation of these elite lines, a prerequisite step for gene editing.
The use of wheatgrass (Thinopyrum intermedium) in breeding

Abstract:Wheatgrass (Th. intermedium) has been traditionally used in wheat breeding for obtaining wheat-wheatgrass hybrids and varieties with introgressions of new genes for economically valuable traits. However, in the 1980s in the United States wheatgrass was selected from among perennial plant species as having promise for domestication and the development of dual-purpose varieties for grain (as an alternative to perennial wheat) and hay. The result of this work was the creation of the wheatgrass varieties Kernza (The Land Institute, Kansas) and MN-Clearwater (University of Minnesota, Minnesota). In Omsk State Agrarian University, the variety Sova was developed by mass selection of the most winter-hardy biotypes with their subsequent combination from the population of wheatgrass obtained from The Land Institute. The average grain yield of the variety Sova is 9.2 dt/ha, green mass is 210.0 dt/ ha, and hay is 71.0 dt/ha. Wheatgrass is a crop with a large production potential, benef icial environmental properties, and valuable grain for functional food. Many publications show the advantages of growing the Kernza variety compared to annual crops in reducing groundwater nitrate contamination, increasing soil carbon sequestration, and reducing energy and economic costs. However, breeding programs for domestication of perennial crops are very limited in Russia. This paper presents an overview of main tasks faced by breeders, aimed at enhancing the yield and cultivating wheatgrass eff iciency as a perennial grain and fodder crop. To address them, both traditional and modern biotechnological and molecular cytogenetic approaches are used. The most important task is to transfer target genes of Th. intermedium to modern wheat varieties and decrease the level of chromatin carrying undesirable genes of the wild relative. The f irst consensus map of wheatgrass containing 10,029 markers was obtained, which is important for searching for genes and their introgressions to the wheat genome. The results of research on the nutritional and technological properties of wheatgrass grain for the development of food products as well as the differences in the quality of wheatgrass grain and wheat grain are presented.

Abstract:BackgroundAmphidiploids generated by distant hybridization are commonly used as genetic bridge to transfer desirable genes from wild wheat species into cultivated wheat. This method is typically used to enhance the resistance of wheat to biotic or abiotic stresses, and to increase crop yield and quality. Tetraploid Thinopyrum elongatum exhibits strong adaptability, resistance to stripe rust and Fusarium head blight, and tolerance to salt, drought, and cold.ResultsIn the present study, we produced hybrid derivatives by crossing and backcrossing the Triticum durum–Th. elongatum partial amphidiploid (Trititrigia 8801, 2n = 6× = 42, AABBEE) with wheat cultivars common to the Sichuan Basin. By means of cytogenetic and disease resistance analyses, we identified progeny harboring alien chromosomes and measured their resistance to stripe rust. Hybrid progenies possessed chromosome numbers ranging from 40 to 47 (mean = 42.72), with 40.0% possessing 42 chromosomes. Genomic in situ hybridization revealed that the number of alien chromosomes ranged from 1 to 11. Out of the 50 of analyzed lines, five represented chromosome addition (2n = 44 = 42 W + 2E) and other five were chromosome substitution lines (2n = 42 = 40 W + 2E). Importantly, a single chromosome derived from wheat–Th. elongatum intergenomic Robertsonian translocations chromosome was occurred in 12 lines. Compared with the wheat parental cultivars (‘CN16’ and ‘SM482’), the majority (70%) of the derivative lines were highly resistant to strains of stripe rust pathogen known to be prevalent in China.ConclusionThe findings suggest that these hybrid-derivative lines with stripe rust resistance could potentially be used as germplasm sources for further wheat improvement.

Abstract:We developed T1AL·1PS and T1AS·1PL Robertsonian translocations by breakage-fusion mechanism based on wheat-A. cristatum 1P(1A) substitution line with smaller leaf area, shorter plant height, and other excellent agronomic traits Agropyron cristatum, a wild relative of wheat, is a valuable germplasm resource for improving wheat genetic diversity and yield. Our previous study confirmed that the A. cristatum chromosome 1P carries alien genes that reduce plant height and leaf size in wheat. Here, we developed T1AL·1PS and T1AS·1PL Robertsonian translocations (RobTs) by breakage-fusion mechanism based on wheat-A. cristatum 1P (1A) substitution line II-3-1c. Combining molecular markers and cytological analysis, we identified 16 spontaneous RobTs from 911 F2 individuals derived from the cross of Jimai22 and II-3-1c. Fluorescence in situ hybridization (FISH) was applied to detect the fusion structures of the centromeres in wheat and A. cristatum chromosomes. Resequencing results indicated that the chromosomal junction point was located at the physical position of Triticum aestivum chromosome 1A (212.5 Mb) and A. cristatum chromosome 1P (230 Mb). Genomic in situ hybridization (GISH) in pollen mother cells showed that the produced translocation lines could form stable ring bivalent. Introducing chromosome 1PS translocation fragment into wheat significantly increased the number of fertile tillers, grain number per spike, and grain weight and reduced the flag leaf area. However, introducing chromosome 1PL translocation fragment into wheat significantly reduced flag leaf area and plant height with a negative effect on yield components. The pre-breeding of two spontaneous RobTs T1AL·1PS and T1AS·1PL was important for wheat architecture improvement.

Abstract:Agropyron cristatum (L.) Gaertn. (2n = 4x = 28, PPPP), one of the most important wild relatives of wheat, harbors many desirable genes for wheat genetic improvement. Development of wheat-A. cristatum translocation lines with superior agronomic traits facilitates wheat genetic improvement. In this study, 5106-DS was identified to be a wheat-A. cristatum 6P (6D) disomic substitution line using cytogenetic identification and molecular markers analysis, which displayed higher thousand-grain weight than its wheat parent Triticum aestivum cv. Fukuhokomugi (2n = 6x = 42, AABBDD). Analysis of its backcross populations indicated that there might be genes conferring increased grain weight and width on the chromosome 6P of 5106-DS. In the backcross population, we found three plants as Robertsonian translocation lines, created by chromosome centric breakage-fusion. Among them, there are one T6DS·6PL and two T6PS·6DL Robertsonian translocation lines. Additionally, the centromeres of these three translocation lines were determined to be fused centromeres of 6D and 6P using the probes pAcCR1 and pCCS1. The development of Robertsonian translocation lines would promote the utilization of A. cristatum chromosome 6P in wheat improvement.

Abstract:Rye ( Secale cereale L.), a naturally cross-pollinating relative of wheat, is a tertiary gene donor and of substantial value in wheat improvement. Wheat powdery mildew is caused by Blumeria graminis f. sp. tritici ( Bgt ), which seriously affects yield and quality worldwide. Identifying and transferring new, effective resistance genes against powdery mildew from rye is important for wheat breeding. The current study developed a wheat-rye line YT2 resistant to powdery mildew by crossing, backcrossing, and self-pollination for multiple generations between octoploid triticale 09R2-100 and common wheat cultivar Shixin 616. YT2 was confirmed to be a 6R disomic addition and T1RS⋅1BL translocation line by genomic in situ hybridization (GISH), multicolor fluorescence in situ hybridization (mc-FISH), multicolor-GISH (mc-GISH), and molecular marker analyses. Disease responses to different Bgt isolates and genetic analysis showed that the powdery mildew resistance gene of YT2 was derived from the rye chromosome 6R of 09R2-100, which differed from the previously reported Pm genes from rye including Pm20 on 6RL. Resistance phenotype of different translocation lines and deletion lines derived from YT2 combined with newly developed 6RL-specific markers analysis suggested that the powdery mildew resistance gene of YT2 was localized to the region in chromosome 6RL: 890.09–967.51 Mb and flanked by markers XM189 and X4M19 , corresponding to the reference genome of Weining rye. Therefore, YT2 could be used as a promising bridging parent for wheat disease resistance improvement.
