Abstract:Low temperature is a major environmental factor that severely impairs plant growth and productivity. Watermelon ( Citrullus lanatus ) is a chilling-sensitive crop. Grafting of watermelon onto pumpkin rootstock is an effective technique to increase the chilling tolerance of watermelon when exposure to short-time chilling stress. However, the mechanism by which pumpkin rootstock increases chilling tolerance remains poorly understood. Under 10°C/5°C (day/night) chilling stress treatment, pumpkin-grafted watermelon seedlings showed higher chilling tolerance than self-grafted watermelon plants with significantly reduced lipid peroxidation and chilling injury (CI) index. Physiological analysis revealed that pumpkin rootstock grafting led to the notable accumulation of putrescine in watermelon seedlings under chilling conditions. Pre-treat foliar with 1 mM D-arginine (inhibitor of arginine decarboxylase, ADC) increased the electrolyte leakage (EL) of pumpkin-grafted watermelon leaves under chilling stress. This result can be ascribed to the decrease in transcript levels of ADC , ornithine decarboxylase , spermidine synthase , and polyamine oxidase genes involved in the synthesis and metabolism of polyamines. Transcriptome analysis showed that pumpkin rootstock improved chilling tolerance in watermelon seedlings by regulating differential gene expression under chilling stress. Pumpkin-grafted seedling reduced the number and expression level of differential genes in watermelon scion under chilling stress. It specifically increased the up-regulated expression of ADC ( Cla97C11G210580 ), a key gene in the polyamine metabolism pathway, and ultimately promoted the accumulation of putrescine. In conclusion, pumpkin rootstock grafting increased the chilling tolerance of watermelon through transcription adjustments, up regulating the expression level of ADC , and promoting the synthesis of putrescine, which ultimately improved the chilling tolerance of pumpkin-grafted watermelon plants.

Abstract:Salicylic acid (SA) has been proven to be a multifunctional signaling molecule that participates in the response of plants to abiotic stresses. In this study, we used cold-sensitive cucumber and cold-tolerant pumpkin as experimental materials to examine the roles of SA in root–shoot communication responses to aerial or/and root-zone chilling stress in own-root and hetero-root grafted cucumber and pumpkin plants. The results showed that pumpkin ( Cm ) rootstock enhanced the chilling tolerance of grafted cucumber, as evidenced by the observed lower levels of electrolyte leakage (EL), malondialdehyde (MDA), and higher photosynthetic rate (Pn) and gene expression of Rubisco activase (RCA). However, cucumber ( Cs ) rootstock decreased the chilling tolerance of grafted pumpkins. Cs/Cm plants showed an increase in the mRNA expression of C-repeat-binding factor ( CBF1 ), an inducer of CBF expression ( ICE1 ), and cold-responsive ( COR47 ) genes and CBF1 protein levels in leaves under 5/25 and 5/5°C stresses, or in roots under 25/5 and 5/5°C stresses, respectively, compared with the Cs/Cs . Chilling stress increased the endogenous SA content and the activity of phenylalanine ammonia-lyase (PAL), and the increase in SA content and activity of PAL in Cs/Cm plants was much higher than in Cs/Cs plants. Transcription profiling analysis revealed the key genes of SA biosynthesis, PAL , ICS , and SABP2 were upregulated, while SAMT , the key gene of SA degradation, was downregulated in Cs/Cm leaves, compared with Cs/Cs leaves under chilling stress. The accumulation of SA in the Cs/Cm leaves was mainly attributed to an increase in SA biosynthesis in leaves and that in transport from roots under aerial and root-zone chilling stress, respectively. In addition, exogenous SA significantly upregulated the expression level of cold-responsive ( COR ) genes, enhanced actual photochemical efficiency ( Φ PSII ), maximum photochemical efficiency ( F v / F m ), and Pn, while decreased EL, MDA, and CI in grafted cucumber. These results suggest that SA is involved in rootstock–scion communication and grafting-induced chilling tolerance by upregulating the expression of COR genes in cucumber plants under chilling stress.

Abstract:Salicylic acid (SA) plays an important role in plant response to abiotic stresses. This study investigated the potential role of SA in alleviating the adverse effects of chilling stress on photosynthesis and growth in watermelon (Citrullus lanatus). Chilling stress induced the simultaneous accumulation of free and conjugated SA in watermelon plants, and the chilling-induced SA production was attributed to the phenylalanine ammonia-lyase pathway. Applying SA at moderate concentrations induced chilling tolerance, whereas inhibition of SA biosynthesis by L-ɑ-aminooxy-β-phenylpropionic acid (AOPP) increased the photooxidation of PS II under chilling stress in watermelon, resulting in reduced photosynthesis and growth. Chilling induced a transient increase in the ratios of reduced to oxidized glutathione and reduced ascorbate to dehydroascorbate. Then, the expression of antioxidant genes was upregulated, and the activities of antioxidant enzymes were enhanced. Furthermore, SA-induced chilling tolerance was associated with cellular glutathione and ascorbate homeostasis, which served as redox signals to regulate antioxidant metabolism under chilling stress. AOPP treatment stimulated the chilling-induced expression of cold-responsive genes, particularly via C-repeat binding factors CBF3 and CBF4. These results confirm the synergistic role of SA signaling and the CBF-dependent responsive pathway during chilling stress in watermelon.

Abstract:The purpose of this study is to determine suitable temperature for selecting germplasm with cold tolerance and to investigate the effects of exogenous SA on cold tolerance in watermelon. We found that low temperature (10°C) was the most suitable temperature to identify cold tolerance germplasm in watermelon. Moreover, we classified the capacities of four different watermelon germplasms, of which No. 9 was the germplasm with strongest cold tolerance and No. 2 was the one with chilling sensitiveness. The suitable exogenous SA (1.0mmol/L) enhanced cold tolerance in watermelon through activations of antioxidative capacity, such as, G-POD, ASA-POD, CAT, SOD and GR. The activities of antioxidative enzymes were more significantly increased in cold tolerance germplasm No. 9 than that in chilling sensitive germplasm No. 2 in watermelon after being treated with suitable exogenous SA (1.0mmol/L), although they were also been enhanced in No. 2. However, higher concentration of exogenous SA decreased the activity of antioxidative enzymes, consequently, weakened the capacity of cold tolerance in watermelon.
Salicylic Acid Biosynthesis Inhibitors Increase Chilling Injury to Maize (zea Mays L.) Seedlings

Abstract:Salicylic acid (SA) is involved in plant response to many abiotic stresses. This study investigated the potential role of SA in regulating chilling tolerance of maize (Zea mays L.) seedlings. The results showed that chilling stress induced transient accumulation of free SA in maize plants, and stimulated the enzymatic activities of phenylalanine ammonia-lyase (PAL) and benzoic acid-2-hydroxylase. However, seedlings pretreated with SA biosynthesis inhibitors (i.e., 2-aminoindane-2-phosphonic acid or 1-aminobenzotriazole) accumulated significantly less free and bound SA and suffered more severe cold injury. Together, these results indicated that the chilling-induced SA accumulation in maize seedlings was mainly attributed to the PAL pathway. SA application also induced chilling tolerance of maize seedlings; whereas, inhibition of SA biosynthesis significantly increased membrane peroxidation, decreased antioxidant enzymes activities, and resulted in growth retardation. Exogenous SA treatments reversed the effects of the SA inhibitors on chilling tolerance, indicating the important endogenous role of SA in inducing chilling tolerance of maize plants.
Relationship Between Endogenous Salicylic Acid and Antioxidant Enzyme Activities in Maize Seedlings under Chilling Stress

Abstract:SUMMARY Salicylic acid (SA) can induce multiple stress tolerance in plants. This study investigated the relationship between SA and antioxidant enzyme activities in maize seedlings under chilling stress. Changes of endogenous SA, antioxidant enzyme activities and malondialdehyde (MDA) concentrations were assessed in two different chilling-tolerant maize inbred lines (Huang C and Mo17) under chilling stress. The results showed that both endogenous free and bound salicylic acid contents increased in roots and leaves of both lines. MDA concentrations also increased significantly in roots and leaves of both lines after chilling stress. In addition, in Huang C, chilling stress increased the activities of four antioxidant enzymes, ascorbate peroxidase (APX), catalase (CAT), glutathione reductase (GR) and peroxidase, while in Mo17, only CAT and APX increased. Furthermore, a regression analysis was conducted between SA and MDA concentrations or antioxidant enzyme activities under chilling stress. The results indicated that MDA concentrations were positively correlated with total SA contents in roots ( r = 0.9776, p = 0.0224) and bound SA in leaves ( r = 0.9974, p = 0.0458), respectively. Total SA contents had positive correlations with APX activities both in roots ( r = 0.9993, p = 0.002) and leaves ( r = 0.9630, p = 0.037) and GR in leaves ( r = 0.9298, p = 0.0221). Together, these results suggested that chilling stress improved the biosynthesis of endogenous SA, and lipid peroxidation and antioxidant enzyme activities could be indicated by endogenous SA contents of maize seedlings under chilling stress. Furthermore, increased activities of antioxidant enzymes, especially in roots, may contribute to the chilling tolerance of maize seedlings.
Salicylic acid analogues with biological activity may induce chilling tolerance of maize (Zea mays) seeds

Abstract:One kind of biologically active salicylic acid (SA) analogue (acetylsalicylic acid, ASA) and two inactive compounds (4-aminosalicylic acid and 4-aminobenzoic acid), along with SA were chosen to evaluate their role in inducing chilling tolerance of two different chilling-tolerant maize (Zea mays L.) inbred lines. These compounds were applied as seed treatments or as a hydroponic application. The results showed that four compounds had no significant effect on germination of maize seeds; however, SA or ASA soaking treatments significantly increased the root length, shoot height, and shoot and root dry weights of seedlings grown under chilling stress. Hydroponic applications of SA or ASA significantly alleviated the accumulation of malondialdehyde, hydrogen peroxide, and superoxide radicals in roots and leaves of both lines under chilling stress, and the applications also increased the photosynthetic pigments, including chlorophyll a, chlorophyll b, and carotenoids. However, 4-aminosalicylic acid and 4-aminobenzoic acid applications had no significant effect in ameliorating the growth inhibition of seedlings under chilling stress. This study showed that SA and ASA significantly induced the chilling tolerance of maize; however, 4-aminosalicylic acid and 4-aminobenzoic acid were not effective in inducing tolerance to chilling stress. The results suggest that only SA analogues with biological activity may have the ability to induce chilling tolerance of maize.
Salicylic Acid Improves Chilling Tolerance via CsNPR1–CsICE1 interaction in Grafted Cucumbers

Abstract:Abstract Salicylic acid (SA) plays a role in regulating of grafting-induced cold tolerance. However, the molecular mechanism behind it is still unknown. Here, we established that the phenylalanine ammonia-lyase (PAL) pathway-dependent elevate in SA content in grafted cucumber leaves was not only synthesized in the leaves but also transported from the roots under chilling stress. RNAi-CsPAL with low SA content as rootstock reduced SA accumulation in grafted seedling leaves while decreasing rootstock-induced cold tolerance, as evidenced by higher electrolyte leakage (EL), hydrogen peroxide (H2O2), and superoxide anion (O2·−) contents and lower expression of cold-responsive genes (CsICE1, CsDREB1A, CsDREB1B, and CsCOR47), whereas OE-CsPAL with high SA content as rootstock improved the cold tolerance of grafted plants in comparison with the wild type (WT). In addition, CsNPR1 was significantly upregulated in grafted cucumber under chilling stress, with exogenous and endogenous overexpressed SA inducing its transcriptional expression and protein stability, which exhibited higher expression in grafted plants than in self-root plants. While CsNPR1-overexpression (OE-CsNPR1) seedlings as scions were more tolerant to chilling stress than WT seedlings, CsNPR1-suppression (Anti-CsNPR1) seedlings as scions were more vulnerable to chilling stress. Notably, CsNPR1–CsICE1 interactions alleviated ROS accumulation and activated the expression of CsDREB1A, CsDREB1B, CsCOR47, CsCOR15, CsCOR413, and CsKIN1 to enhance SA-mediated chilling tolerance in grafted cucumber. Overall, our findings reveal that SA enhances chilling tolerance in grafted cucumbers via the model of the CsNPR1–CsICE1 transcriptional regulatory cascade.

Effect of Cold Stress on Growth, Physiological Characteristics, and Calvin-Cycle-Related Gene Expression of Grafted Watermelon Seedlings of Different Gourd Rootstocks

Abstract:Recently, grafting has been used to improve abiotic stress resistance in crops. Here, using watermelon ‘Zaojia 8424’ (Citrullus lanatus) as scions, three different gourds (Lagenaria siceraria, 0526, 2505, and 1226) as rootstocks, and non-grafted plants as controls (different plants were abbreviated as 0526, 2505, 1226, and 8424), the effect of cold stress on various physiological and molecular parameters was investigated. The results demonstrate that the improved cold tolerance of gourd-grafted watermelon was associated with higher chlorophyll and proline content, and lower malondialdehyde (MDA) content, compared to 8424 under cold stress. Furthermore, grafted watermelons accumulated fewer reactive oxygen species (ROS), accompanied by enhanced antioxidant activity and a higher expression of enzymes related to the Calvin cycle. In conclusion, watermelons with 2505 and 0526 rootstocks were more resilient compared to 1226 and 8424. These results confirm that using tolerant rootstocks may be an efficient adaptation strategy for improving abiotic stress tolerance in watermelon.

Comparative transcriptome analysis of grafting to improve chilling tolerance of cucumber

Abstract:Grafting with pumpkin as rootstock could improve chilling tolerance of cucumber; however, the underlying mechanism of grafting-induced chilling tolerance remains unclear. Here, we analyzed the difference of physiological and transcriptional level between own-rooted (Cs/Cs) and hetero-grafted (Cs/Cm) cucumber seedlings under chilling stress. The results showed that grafting with pumpkin significantly alleviated the chilling injury as evidenced by slightly symptoms, lower contents of electrolyte leakage (EL), malondialdehyde (MDA), hydrogen peroxide (H2O2), and superoxide anion (O2-) and higher relative water content in Cs/Cm seedlings compared with Cs/Cs seedlings under chilling stress. RNA-seq data showed that grafting induced more DGEs at 8 °C/5 °C compared with 25 °C/18 °C. In accordance with the increase of the activities of antioxidant enzymes (SOD, POD, CAT, APX), grafting upregulated the expression of the regulated redox-related genes such as GST, SOD, and APX. Moreover, grafting increased the expression of genes participated in central carbon metabolism to promote the conversion and decomposition of sugar, which provided more energy for the growth of Cs/Cm seedlings under chilling stress. In addition, grafting regulated the genes involved in the intracellular signal transduction pathways such as calcium signal (CAML, CML, and CDPK) and inositol phospholipid signal (PLC), as well as changed the gene expression of plant hormone signal transduction pathways (ARF, GAI, ABF, and PYR/PYL). These results provide a physiological and transcriptional basis for the molecular mechanism of grafting-induced chilling tolerance of cucumber seedlings.
Methyl jasmonate mediates melatonin-induced cold tolerance of grafted watermelon plants

Abstract:Abstract Root–shoot communication has a critical role in plant adaptation to environmental stress. Grafting is widely applied to enhance the abiotic stress tolerance of many horticultural crop species; however, the signal transduction mechanism involved in this tolerance remains unknown. Here, we show that pumpkin- or figleaf gourd rootstock-enhanced cold tolerance of watermelon shoots is accompanied by increases in the accumulation of melatonin, methyl jasmonate (MeJA), and hydrogen peroxide (H 2 O 2 ). Increased melatonin levels in leaves were associated with both increased melatonin in rootstocks and MeJA-induced melatonin biosynthesis in leaves of plants under cold stress. Exogenous melatonin increased the accumulation of MeJA and H 2 O 2 and enhanced cold tolerance, while inhibition of melatonin accumulation attenuated rootstock-induced MeJA and H 2 O 2 accumulation and cold tolerance. MeJA application induced H 2 O 2 accumulation and cold tolerance, but inhibition of JA biosynthesis abolished rootstock- or melatonin-induced H 2 O 2 accumulation and cold tolerance. Additionally, inhibition of H 2 O 2 production attenuated MeJA-induced tolerance to cold stress. Taken together, our results suggest that melatonin is involved in grafting-induced cold tolerance by inducing the accumulation of MeJA and H 2 O 2 . MeJA subsequently increases melatonin accumulation, forming a self-amplifying feedback loop that leads to increased H 2 O 2 accumulation and cold tolerance. This study reveals a novel regulatory mechanism of rootstock-induced cold tolerance.

The Synergistic Priming Effect of Exogenous Salicylic Acid and H2O2 on Chilling Tolerance Enhancement during Maize (Zea mays L.) Seed Germination

Abstract:Chilling stress is an important constraint for maize seedling establishment in the field. To examine the role of salicylic acid (SA) and hydrogen peroxide (H2O2) in response to chilling stress, we investigated the effects of seed priming with SA, H2O2, and SA+H2O2 combination on maize resistance under chilling stress (13°C). Priming with SA, H2O2, and especially SA+H2O2 shortened seed germination time and enhanced seed vigor and seedling growth as compared with hydropriming and non-priming treatments under low temperature. Meanwhile, SA+H2O2 priming notably increased the endogenous H2O2 and SA content, antioxidant enzymes activities and their corresponding genes ZmPAL, ZmSOD4, ZmAPX2, ZmCAT2, and ZmGR expression levels. The α-amylase activity was enhanced to mobilize starch to supply metabolites such as soluble sugar and energy for seed germination under chilling stress. In addition, the SA+H2O2 combination positively up-regulated expressions of gibberellic acid (GA) biosynthesis genes ZmGA20ox1 and ZmGA3ox2, and down-regulated GA catabolism gene ZmGA2ox1 expression; while it promoted GA signaling transduction genes expressions of ZmGID1 and ZmGID2 and decreased the level of seed germination inhibitor gene ZmRGL2. The abscisic acid (ABA) catabolism gene ZmCYP707A2 and the expressions of ZmCPK11 and ZmSnRK2.1 encoding response receptors in ABA signaling pathway were all up-regulated. These results strongly suggested that priming with SA and H2O2 synergistically promoted hormones metabolism and signal transduction, and enhanced energy supply and antioxidant enzymes activities under chilling stress, which were closely relevant with chilling injury alleviation and chilling-tolerance improvement in maize seed. Highlights:Seed germination and seedling growth were significantly improved under chilling stress by priming with SA+H2O2 combination, which was closely relevant with the change of reactive oxygen species, metabolites and energy supply, hormones metabolism and regulation.
Grafting Delays Watermel on Fruit Ripening by Altering Gene Expression of ABA Centric Phytohormone Signaling

Abstract:Grafting cultivation is implemented worldwide mainly to resist abiotic and biotic stresses and is an effective method to improve watermelon production. However, grafting may affect fruit development and quality. In our experiment, pumpkin-grafted (PG) watermelon fruits developed slower and the ripening period was extended compared to self-grafted (SG) fruits. We found that the concentrations of abscisic acid (ABA) among endogenous phytohormones were dramatically reduced by pumpkin grafting. In order to understand these changes at the gene expression level, we performed a comprehensive analysis of the fruit flesh transcriptomes between PG and SG during fruit development and ripening. A total of 1,675 and 4,102 differentially expressed genes (DEGs) were identified between PG and SG. Further functional enrichment analysis revealed that these DEGs were associated with carbohydrate biosynthesis, phytohormone signaling transmission, and cell wall metabolism categories. ABA centric phytohormone signaling and fruit quality-related genes including ABA receptor, PP2C proteins, AP2-EREBP transcription factors, sucrose transporter, and carotenoid isomerase were co-expressed with fruit ripening. These results provide the valuable resource for understanding the mechanism of pumpkin grafting effect on watermelon fruit ripening and quality development.

Abstract:Grafting is an important agricultural technique widely used to improve plant growth, yield, and adaptation to either biotic or abiotic stresses. However, the molecular mechanisms underlying grafting-induced physiological processes remain unclear. Watermelon (Citrullus lanatus L.) is an important horticultural crop worldwide. Grafting technique is commonly used in watermelon production for improving its tolerance to stresses, especially to the soil-borne fusarium wilt disease. In the present study, we used high-throughput sequencing to perform a genome-wide transcript analysis of scions from watermelon grafted onto bottle gourd and squash rootstocks. Our transcriptome and digital gene expression (DGE) profiling data provided insights into the molecular aspects of gene regulation in grafted watermelon. Compared with self-grafted watermelon, there were 787 and 3485 genes differentially expressed in watermelon grafted onto bottle gourd and squash rootstocks, respectively. These genes were associated with primary and secondary metabolism, hormone signaling, transcription factors, transporters, and response to stimuli. Grafting led to changes in expression of these genes, suggesting that they may play important roles in mediating the physiological processes of grafted seedlings. The potential roles of the grafting-responsive mRNAs in diverse biological and metabolic processes were discussed. Obviously, the data obtained in this study provide an excellent resource for unraveling the mechanisms of candidate genes function in diverse biological processes and in environmental adaptation in a graft system.
Exogenous Salicylic Acid Alleviates Salt Stress by Improving Leaf Photosynthesis and Root System Architecture in Cucumber Seedlings

Abstract:Exogenous salicylic acid (SA) can alleviate salt stress in plants. Previous researches have been focused on SA's role in leaf morphological and physiological characteristics, however, little information is available on its role in both leaf photosynthesis and root system architecture (RSA). A hydroponic study was conducted to analyze biomass, leaf photosynthesis, RSA parameters and expression of RSA genes in cucumber seedlings grown under four treatments, including control (CK), 50 mM NaCl (NaCl), 0.3 mM SA (SA) and 50 mM NaCl plus 0.3 mM SA (NaCl + SA). The results showed that compared with CK, both NaCl and SA treatments severely reduced relative growth rate (RGR) and leaf photosynthetic parameters (F-v/F-m, Phi(II) and Pn). However, compared with NaCl treatment, NaCl + SA treatment significantly increased RGR, leaf photosynthetic (F-v/F-m, Phi(II) and Pn) and RSA (total root length, root surface area, number and total length of primary and secondary LR) parameters. In addition, compared with CK, although both NaCl and SA treatments down-regulated expression of GL2 gene and up-regulated expression of RHD2 gene, NaCl + SA treatment notably up-regulated expression of NAC1, NAC2, GL2, EXP and down-regulated expression of RHD2. It is concluded that exogenous SA alleviated salt stress via effectively promoting leaf photosynthesis and RSA in cucumber seedlings. SA-mediated increment in expression of RSA genes, total length and number of primary and secondary LR could contribute to the improved RSA.
Bottle Gourd Rootstock-Grafting Affects Nitrogen Metabolism in NaCl-stressed Watermelon Leaves and Enhances Short-Term Salt Tolerance.

Abstract:The plant growth, nitrogen absorption, and assimilation in watermelon (Citrullus lanatus [Thunb.] Mansf.) were investigated in self-grafted and grafted seedlings using the salt-tolerant bottle gourd rootstock Chaofeng Kangshengwang (Lagenaria siceraria Standl.) exposed to 100mM NaCl for 3d. The biomass and NO3− uptake rate were significantly increased by rootstock while these values were remarkably decreased by salt stress. However, compared with self-grafted plants, rootstock-grafted plants showed higher salt tolerance with higher biomass and NO3− uptake rate under salt stress. Salinity induced strong accumulation of nitrate, ammonium and protein contents and a significant decrease of nitrogen content and the activities of nitrate reductase (NR), nitrite reductase (NiR), glutamine synthetase (GS), and glutamate synthase (GOGAT) in leaves of self-grafted seedlings. In contrast, salt stress caused a remarkable decrease in nitrate content and the activities of GS and GOGAT, and a significant increase of ammonium, protein, and nitrogen contents and NR activity, in leaves of rootstock-grafted seedlings. Compared with that of self-grafted seedlings, the ammonium content in leaves of rootstock-grafted seedlings was much lower under salt stress. Glutamate dehydrogenase (GDH) activity was notably enhanced in leaves of rootstock-grafted seedlings, whereas it was significantly inhibited in leaves of self-grafted seedlings, under salinity stress. Three GDH isozymes were isolated by native gel electrophoresis and their expressions were greatly enhanced in leaves of rootstock-grafted seedlings than those of self-grafted seedlings under both normal and salt-stress conditions. These results indicated that the salt tolerance of rootstock-grafted seedlings might (be enhanced) owing to the higher nitrogen absorption and the higher activities of enzymes for nitrogen assimilation induced by the rootstock. Furthermore, the detoxification of ammonium by GDH when the GS/GOGAT pathway was inhibited under salt stress might play an important role in the release of salt stress in rootstock-grafted seedlings.
Identification of Down-regulated Genes Modulated by an Alternative Oxidase Pathway under Cold Stress Conditions in Watermelon Plants

Abstract:This study investigated the cold responses of watermelon plants modulated by the alternative oxidase (AOX) pathway. Salicylhydroxamic acid was used in watermelon (Citrullus lanatus) plants in order to chemically inhibit the AOX pathway. After exposure to cold temperature (10°C) for 24 h, the SHAM-treated plants showed more serious cold-induced damage compared to non-treated plants. To investigate how the AOX pathway modulated the watermelon plants’ responses to cold, a polymerase chain reaction (PCR)-based suppression subtractive hybridization technique was used to isolate genes differently expressed between cold-stressed leaves of SHAM-treated and non-treated watermelon germplasm “IVSM No. 9”. After sequencing 400 randomly chosen cDNA clones, 314 uniquely expressed sequence tags (ESTs) were obtained. A total of 114 ESTs encoded unknown proteins or had no similarity in the database. Identified genes with significant protein homology mainly participated in stress and defense responses, metabolism and energetic processes, regulation of gene expression, signal transduction, protein fate and synthesis, cellular transport and others. These results indicated that watermelon plants could not respond appropriately to cold stress and suffer greater damage with an inhibited AOX pathway, thus, the AOX pathway had an important role in defining the plants cold responses.
Rootstock–scion exchanging mRNAs participate in the pathways of amino acid and fatty acid metabolism in cucumber under early chilling stress

Abstract:Abstract Cucumber (Cucumis sativus L.) often experiences chilling stress that limits its growth and productivity. Grafting is widely used to improve abiotic stress resistance by exploiting a vigorous root system, suggesting there exists systemic signals communication between distant organs. mRNAs are reported to be evolving fortification strategies involving long-distance signaling when plants suffer from chilling stress. However, the potential function of mobile mRNAs in alleviating chilling stress in grafted cucumber is still unknown. Here, the physiological changes, mobile mRNA profiles, and transcriptomic and metabolomic changes in above- and underground tissues of all graft combinations of cucumber and pumpkin responding to chilling stress were established and analyzed comprehensively. The co-relationship between the cluster of chilling-induced pumpkin mobile mRNAs with differentially expressed genes and differentially intensive metabolites revealed that four key chilling-induced pumpkin mobile mRNAs were highly related to glycine, serine, and threonine synthesis and fatty acid β-oxidative degradation metabolism in cucumber tissues of heterografts. The verification of mobile mRNAs, potential transport of metabolites, and exogenous application of key metabolites of the glycerophospholipid metabolism pathway in cucumber seedlings confirmed the role of mobile mRNAs in regulating chilling responses in grafted cucumber. Our results build a link between the long-distance mRNAs of chilling-tolerant pumpkin and the fatty acid β-oxidative degradation metabolism of chilling-sensitive cucumber. They also help to uncover the mechanism of signaling interaction between scion and stock as a means of achieving chilling tolerance in grafted cucumber.
