Abstract:The unearthing of various forms of broad and narrow-spectrum antibiotics have helped mankind combat various forms of microbic infections ranging from protozoal to bacterial. The applicability of antibiotics has been decisive in a wide range of frontline therapeutic dealings. An extensive use of antibiotics and certain genetic modifications in microbes has paved the way for a new issue to be answered by the scientific community, an antibiotic resistance. An unparallel infections triggered by multidrug-resistant organisms (MDRO) account to be a major cause of mortality and morbidity, today this situation demands the development of alternative strategies of cure and medicine by the scientific community. Study shows the amplification of the effect of antibiotics may be a possible route to address antibiotic resistivity. The assortment of various surface properties of nanoparticles (NPs) coupled with their ability to efficiently bind with bacterial surfaces leading to the rupture and lysis of cell deliberates potential strategies to manage infections caused by MDROs.The present study scrutinizes the amplification of the antibacterial activity of ciprofloxacin (CIP) against the bacterial strains of Staphylococcus aureus , Escherichia coli , Pseudomonas and S treptococcus mutans on coupling with synthesized copper, silver and zinc oxide NPs (ZnO NPs) by disc diffusion technique. The current study describes the efficacious synthesis and characterization of ZnONP, copper Nps (CuNPs) and silver NPs (AgNPs) followed by the induced amplification of the antibacterial activity of CIP by coupling it with synthesized metal/metal oxide (M/MO NP). The synthesized M/MO NPs were characterized using XRD and FE-SEM studies and TEM. The results indicated that among the ZnONP, AgNP, the Cu NPs potentiate the bactericidal efficacy of CIP antibiotics.

Abstract:The advent of antibiotics plays a vital role in dealing with the challenges of various bacterial infections, which have saved the millions of patients from the brink of death. However, the abuse of antibiotics accelerated the development of bacterial drug-resistance, posing a serious threat to public health. To conquer the increasingly severe drug-resistance, the amphiphilic ciprofloxacined peptide-based polymer (PAC) was prepared, which could self-assemble into positively charged nanostructures (PAC-NPs) with a diameter of 300 nm and a Zeat potential of +28.6 mV in aqueous solution confirmed by SEM and DLS. The biological assays displayed that the PAC-NPs possessed broad-spectrum and excellent antibacterial effect against both Gram-positive and Gram-negative bacteria, with a MIC in the range of 1.0 ~ 4.0 μg/mL and more than 91% sterilization rate. Most importantly, the cationic PAC-NPs did not easily cause the antibiotic-resistance and showed low toxicity regardless of in vivo or vitro. The unique antibacterial mechanism systematically detected by different techniques revealed that the nanoparticles could completely destroy the morphology of bacterial cells, eventually, leading to the irreversible apoptosis of bacteria cells, which elucidated why the bacteria were less prone to develop resistance to PAC-NPs. This work provides a promising strategy by mimicking the structure of AMPs to construct novel bactericidal materials without inducing drug-resistance.

Soft nanoparticles as antimicrobial agents and carriers of microbiocides for enhanced inhibition activity

Abstract:Antibiotic resistance continues to pose significant health challenges. Considering severe limitations in the discovery and supply of new antibiotics, there is an unmet need to design alternative and more effective strategies for addressing this global issue. Use of polymeric nanoparticles with cationic shell surfaces offers a highly promising approach to coupling their inherent bactericidal action with sustained delivery of small lipophilic microbicides. We have utilized this platform for assembling multi-tasking soft core-shell nanoparticles from star polymers with the desired asymmetric arm composition. These stable nanoparticles with low critical micelle concentration imparted intrinsic antimicrobial potency due to high positive charge density in the corona, as well as the loading of active biocidal agents (such as curcumin and terbinafine) for potential dual and coadjuvant inhibition. This strategic combination allows for both immediate (direct contact) and extended (drug delivery) antibacterial activities for better therapeutic efficacy. Micellar nanoparticles with and without therapeutic cargo were highly efficient against both Escherichia coli (E. coli) and Bacillus subtilis (B. subtilis), representative Gram-negative and Gram-positive bacteria, respectively. Interestingly, we observed bacteria- and concentration-dependent effects, in which higher concentrations of charged nanoparticles were more effective against E. coli, whereas B. subtilis was inhibited only at lower concentrations. This work highlights a valuable platform to achieve combination therapy through nanoparticles with charged coronas and delivery of potent therapeutics to overcome antimicrobial resistance.
Metal–carbenicillin Framework-Based Nanoantibiotics with Enhanced Penetration and Highly Efficient Inhibition of MRSA

Abstract:The development of effective therapies to control methicillin-resistant Staphylococcus aureus (MRSA) infections is challenging because antibiotics can be degraded by the production of certain enzymes, for example, β-lactamases. Additionally, the antibiotics themselves fail to penetrate the full depth of biofilms formed from extracellular polymers. Nanoparticle-based carriers can deliver antibiotics with better biofilm penetration, thus combating bacterial resistance. In this study, we describe a general approach for the construction of β-lactam antibiotics and β-lactamase inhibitors co-delivery of nanoantibiotics based on metal–carbenicillin framework-coated mesoporous silica nanoparticles (MSN) to overcome MRSA. Carbenicillin, a β-lactam antibiotic, was used as an organic ligand that coordinates with Fe3+ to form a metal–carbenicillin framework to block the pores of the MSN. Furthermore, these β-lactamase inhibitor-loaded nanoantibiotics were stable under physiological conditions and could synchronously release antibiotic molecules and inhibitors at the bacterial infection site to achieve a better elimination of antibiotic resistant bacterial strains and biofilms. We confirmed that these β-lactamase inhibitor-loaded nanoantibiotics had better penetration depth into biofilms and an obvious effect on the inhibition of MRSA both in vitro and in vivo.
Novel copper-containing ferrite nanoparticles exert lethality to MRSA by disrupting MRSA cell membrane permeability, depleting intracellular iron ions, and upregulating ROS levels

Abstract:Objective: The widespread use of antibiotics has inevitably led to the emergence of multidrug-resistant bacterial strains, such as methicillin-resistant Staphylococcus aureus (MRSA), making treatment of this infection a serious challenge. This study aimed to explore new treatment strategies for MRSA infection. Methods: The structure of Fe3O4 NPs with limited antibacterial activity was optimized, and the Fe2+ ↔ Fe3+ electronic coupling was eliminated by replacing 1/2 Fe2+ with Cu2+. A new type of copper-containing ferrite nanoparticles (hereinafter referred to as Cu@Fe NPs) that fully retained oxidation-reduction activity was synthesized. First, the ultrastructure of Cu@Fe NPs was examined. Then, antibacterial activity was determined by testing the minimum inhibitory concentration (MIC) and safety for use as an antibiotic agent. Next, the mechanisms underlying the antibacterial effects of Cu@Fe NPs were investigated. Finally, mice models of systemic and localized MRSA infections was established for in vivo validation. Results: It was found that Cu@Fe NPs exhibited excellent antibacterial activity against MRSA with MIC of 1 μg/mL. It effectively inhibited the development of MRSA resistance and disrupted the bacterial biofilms. More importantly, the cell membranes of MRSA exposed to Cu@Fe NPs underwent significant rupture and leakage of the cell contents. Cu@Fe NPs also significantly reduced the iron ions required for bacterial growth and contributed to excessive intracellular accumulation of exogenous reactive oxygen species (ROS). Therefore, these findings may important for its antibacterial effect. Furthermore, Cu@Fe NPs treatment led to a significant reduction in colony forming units within intra-abdominal organs, such as the liver, spleen, kidney, and lung, in mice with systemic MRSA infection, but not for damaged skin in those with localized MRSA infection. Conclusion: The synthesized nanoparticles has an excellent drug safety profile, confers high resistant to MRSA, and can effectively inhibit the progression of drug resistance. It also has the potential to exert anti-MRSA infection effects systemically in vivo. In addition, our study revealed a unique multifaceted antibacterial mode of Cu@Fe NPs: (1) an increase in cell membrane permeability, (2) depletion of Fe ions in cells, (3) generation of ROS in cells. Overall, Cu@Fe NPs may be potential therapeutic agents for MRSA infections.

Abstract:In the fight against antibiotic resistance, which is rising to dangerously high levels worldwide, new strategies based on antibiotic-conjugated biocompatible polymers bound to magnetic nanoparticles that allow the drug to be manipulated and delivered to a specific target are being proposed. Here, we report the direct surface engineering of nontoxic iron oxide nanoparticles (IONs) using biocompatible dextran (Dex) covalently linked to β-cyclodextrin (β-CD) with the ability to form non-covalent complexes with silver-sulfamethazine (SMT-Ag). To achieve a good interaction of β-CD-modified dextran with the surface of the nanoparticles, it was functionalized with diphosphonic acid (DPA) that provides strong binding to Fe atoms. The synthesized polymers and nanoparticles were characterized by various methods, such as nuclear magnetic resonance (NMR), Fourier transform infrared (FTIR) and ultraviolet–visible (UV–Vis) spectroscopies, transmission electron microscopy (TEM), thermogravimetric analysis (TGA), atomic absorption spectroscopy (AAS), dynamic light scattering (DLS), etc. The resulting magnetic ION@DPA-Dex-β-CD-SMT-Ag nanoparticles were colloidally stable in water and contained 24 μg of antibiotic per mg of the particles. When tested for in vitro antimicrobial activity on Gram-positive (Staphylococcus aureus) and Gram-negative (Escherichia coli) bacteria and fungi (yeast Candida albicans and mold Aspergillus niger), the particles showed promising potential.

Abstract:Applications of different types of magnetic nanoparticles for biomedical purposes started a long time back. The concept of surface functionalization of the iron oxide nanoparticles with antibiotics is a novel technique which paves the path for further application of these nanoparticles by virtue of their property of superparamagnetism. In this paper, we have synthesized novel iron oxide nanoparticles surface functionalized with Gentamicin. The average size of the particles, concluded from the HR-TEM images, came to be around 14 nm and 10 nm for unmodified and modified nanoparticles, respectively. The magnetization curve M(H) obtained for these nanoparticles are typical of superparamagnetic nature and having almost zero values of coercivity and remanance. The release properties of the drug coated nanoparticles were studied; obtaining an S shaped profile, indicating the initial burst effect followed by gradual sustained release. In vitro investigations against various gram positive and gram negative strains viz Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa and Bacillus subtilis indicated significant antibacterial efficiency of the drug-nanoparticle conjugate. The MIC values indicated that a small amount like 0.2 mg ml−1 of drug capped particles induce about 98% bacterial death. The novelty of the work lies in the drug capping of the nanoparticles, which retains the superparamagnetic nature of the iron oxide nanoparticles and the medical properties of the drug simultaneously, which is found to extremely blood compatible.

Abstract:The Treponema pallidum haemagglutination (TPHA) test was carried out on 274 sera known to show biological false positive reactions to reagin tests for syphilis. The Treponema pallidum immunization (TPI) and fluorescent treponemal antibody absorption (FTA-ABS) tests were non-reactive on all these sera. Thirty-one or 11·3% showed reactive results in the TPHA test. Sera from 267 people who had lepromatous leprosy were also tested in the TPHA test. Fourteen sera were reactive in the TPHA, TPI, and FTA-ABS tests and were from people who had both syphilis and leprosy. Biological false positive reactions were shown by 26 of the leprosy sera, of which three or 11·5% were also reactive in the TPHA test. A further four sera in the leprosy group were reactive only in the TPHA test. The possible cause of false reactive TPHA test results is discussed. It was concluded that where reagin and TPHA tests are reactive in a person who has no history or clinical signs of syphilis, the serum should be referred for TPI and FTA-ABS testing.

Abstract:The global spread of multi-drug-resistant (MDR) bacteria is rapidly increasing due to antibiotic overuse, posing a major public health threat and causing millions of deaths annually. The present study explored the potential of nanocarriers for delivering novel and alternative antibacterial agents using nanotechnology-based approaches to address the challenge of MDR bacteria. The purpose was to enhance the solubility, stability, and targeted delivery of berberine (BER) and its synthetic derivative NR16 using Styrene- co -Maleic Acid (SMA) nanoparticles. Characterization of the nanoparticles, including dynamic light scattering (DLS) analysis, TEM, and UV/Vis absorption spectroscopy, confirmed their suitability and high stability for passive drug delivery. Antibacterial and antifungal activities were evaluated against a panel of pathogens, revealing significant inhibitory effects on Gram-positive strains; particularly BER, SMA-BER, and NR16 were active against MRSA, MSSA, VR, and VS E. faecalis , and S. epidermidis . Additionally, SMA-BER and SMA-NR16 showed promising activity against biofilm formation of S. epidermidis ; while the two free drugs contributed to S. epidermidis biofilm disruption activity. Hemolysis tests and in vitro studies on human embryonic kidney cells (HEK-293) confirmed the safety profiles of the nanoparticles and free drugs. Overall, this research highlighted the potential of nanotechnology in developing effective antibacterial agents with reduced toxicity, addressing the growing threat of MDR bacterial infections.

Abstract:Silver nanoparticles (AgNPs) have demonstrated numerous physicochemical, biological, and functional properties suitable for biomedical applications, including antibacterial and drug carrier properties. In the present study, the antibiotic, ciprofloxacin (CIP), was loaded onto AgNPs, which were synthesized via the chemical reduction method, thereby enhancing CIP’s antibacterial activity against Gram-negative (Acinetobacter baumannii and Serratia marcescens) and Gram-positive (Staphylococcus aureus) bacterial strains. Polyethylene glycol–400 (PEG) was used to prepare an AgNPs-PEG conjugate with enhanced stability and to act as the linker between CIP and AgNPs, to produce the novel nanocomposite, AgNPs-PEG-CIP. The prepared AgNPs and their conjugates were characterized by ultraviolet-visible spectrophotometry, Fourier-transform infrared spectroscopy, X-ray diffraction, field emission scanning electron microscopy with energy-dispersive X-ray spectroscopy, transmission electron microscopy, zeta potential analysis, and dynamic light scattering techniques. The inhibitory activity of AgNPs and their conjugates on the growths of pathogenic bacteria was assessed using the well-diffusion method. The results showed the enhanced antibacterial effects of AgNPs-CIP compared to CIP alone. The AgNPs-PEG-CIP nanocomposite showed excellent inhibitory effects against bacterial isolates, with its inhibition zones diameters reaching 39, 36, and 40 mm in S. aureus, A. baumannii, and S. marcescens, respectively. The minimum inhibitory concentration and minimum bactericidal concentration of fogNPs and their conjugates and their antibiofilm effects were also determined. The antioxidant potentials of AgNPs and their conjugates, tested via their 1,1-diphenyl-2-picryl-hydrazyl (DPPH) scavenging ability, showed that the activity increased with increasing AgNPs concentration and the addition of the PEG and/or CIP. Overall, according to the results obtained in the present study, the new nanocomposite, AgNPs-PEG-CIP, showed the highest antibacterial, antibiofilm, and antioxidant activity against the pathogenic bacteria tested, compared to CIP alone. The preparation has high clinical potential for prospective use as an antibacterial agent.

Combating Antibiotic Resistance through the Synergistic Effects of Mesoporous Silica-Based Hierarchical Nanocomposites

Abstract:The enormous influence of bacterial resistance to antibiotics has led researchers toward the development of various advanced antibacterial modalities. In this vein, nanotechnology-based devices have garnered interest owing to their excellent morphological as well as physicochemical features, resulting in augmented therapeutic efficacy. Herein, to overcome the multidrug resistance (MDR) in bacteria, we demonstrate the fabrication of a versatile design based on the copper-doped mesoporous silica nanoparticles (Cu-MSNs). Indeed, the impregnated Cu species in the siliceous frameworks of MSNs establish pH-responsive coordination interactions with the guest molecules, tetracycline (TET), which not only enhance their loading efficiency but also assist in their release in the acidic environment precisely. Subsequently, the ultrasmall silver nanoparticles-stabilized polyethyleneimine (PEI-SNP) layer is coated over Cu-MSNs. The released silver ions from the surface-deposited SNPs are capable of sensitizing the resistant strains through establishing the interactions with the biomembranes, and facilitate the generation of toxic free radicals, damaging the bacterial components. In addition to SNPs, Cu species impregnated in MSN frameworks synergistically act through the production of free radicals by participating in the Fenton-like reaction. Various physical characterization techniques for confirming the synthesis and successful surface modification of functional nanomaterials, as well as different antibacterial tests performed against MDR bacterial strains, are highly commendable. Remarkably, this versatile formulation has shown no significant toxic effects on normal mammalian fibroblast cells accounting for its high biocompatibility. Together, these biocompatible MSN-based trio-hybrids with synergistic efficacy and pH-responsive delivery of antibiotics potentially allow for efficient combat against MDR in bacteria.

Antibacterial and antibiofilm efficacy of Ag NPs, Ni NPs and Al2O3 NPs singly and in combination against multidrug-resistant Klebsiella pneumoniae isolates

Abstract:Background: Although traditional antibiotic therapy provided an effective approach to combat pathogenic bacteria, the long-term and widespread use of antibiotic results in the evolution of multidrug-resistant bacteria. Recent progress in nanotechnology offers an alternative opportunity to discover and develop novel antibacterial agents. Methods: A total of 51 K. pneumoniae strains were collected from several specimens of hospitalized patients and identified by two parallel methods (biochemical tests and Vitek-2 system). The antibiotic sensitivity of isolates was evaluated by disk diffusion antibiogram and Vitek-2 system. The biofilms formation ability of antibiotic-resistant strains was examined by microtiter plate and tube methods based on crystal violet staining. The molecular technique was used to determine key genes responsible for biofilms formation of clinical isolates. The antibacterial and antibiofilm activities of Ag NPs, Ni NPs, Al2O3 NPs singly (NPs) and in combination (cNPs) were investigated against selected strains using standard methods. Moreover, the cytotoxicity of NPs was evaluated on mouse neural crest-derived (Neuro-2A) cell line. Results: The results of bacterial studies revealed that more than 80 % of the isolates were resistant to commonly used antibiotics and about 95 % of them were able to form biofilms. Moreover, the presence of fimA and mrkA genes were determined in all biofilm-producing strains. The results of antibacterial and antibiofilm activities of NPs and cNPs demonstrated the lower MIC and MBEC values for Al2O3 NPs singly as well as for Ag/Ni cNPs and Ag/Al2O3 cNPs in combination, respectively. Overall, the inhibitory effects of cNPs were superior to NPs against all strains. Furthermore, the results of the checkerboard assays showed that Ag NPs act synergistically with two other NPs against multidrug-resistant Klebsiella pneumoniae (MDR-K. pneumoniae) isolates. The in vitro cytotoxicity assay revealed no significant toxicity of NPs against Neuro-2A cells. Conclusion: In the present study, the combination of Ag NPs, Ni NPs, and Al2O3 NPs were used against MDR-K. pneumoniae strains and antibacterial and antibiofilm activities were observed for Ag/Ni cNPs and Ag/Al2O3 cNPs.

Abstract:The development of effective strategies against multidrug-resistant (MDR) pathogens is an urgent need in modern medicine. Nanoantibiotics (nABs) offer a new hope in countering the surge of MDR-pathogens. The aim of the current study was to evaluate the antibacterial activity of two attractive nABs, TiO 2 NPs and ZnO NPs, and their performance in improving the antimicrobial activity of defined antibiotics (amoxicillin-clavulanic acid, amox-clav) against MDR-pathogens. The nABs were synthesized using a green method. The physicochemical characteristics of the synthesized nanoparticles were determined using standard methods. The results showed the formation of pure anatase TiO 2 NPs and hexagonal ZnO NPs with an average particle size of 38.65 nm and 57.87 nm, respectively. The values of zeta potential indicated the high stability of the samples. At 8 mg/mL, both nABs exhibited 100% antioxidant activity, while ZnO showed significantly higher activity at lower concentrations. The antibiofilm assay showed that both nABs could inhibit the formation of biofilms of Acinetobacter baumannii 80 and Escherichia coli 27G (MDR-isolates). However, ZnO NPs showed superior antibiofilm activity (100%) against E. coli 27G. The MIC values were determined to be 8 (1), 2 (2), and 4 (4) mg/mL for amox-clav, TiO 2 NPs, and ZnO NPs against A. baumannii 80 ( E. coli 27G), respectively. The results showed that both nABs had synergistically enhanced antibacterial performance in combination with amox-clav. Specifically, an 8-fold reduction in MIC values of antibiotics was observed when they were combined with nABs. These findings highlight the potential of TiO 2 NPs and ZnO NPs as effective nanoantibiotics against MDR-pathogens. The synergistic effect observed when combining nABs with antibiotics suggests a promising approach for combating antibiotic resistance. Further research and development in this area could lead to the development of more effective treatment strategies against MDR infections.

Abstract:Background: The increase in nosocomial multidrug resistance and biofilm-forming bacterial infections led to the search for new alternative antimicrobial strategies other than traditional antibiotics. Silver nanoparticles [AgNP] could be a viable treatment due to their wide range of functions, rapid lethality, and minimal resistance potential. The primary aim of this study is to prepare silver nanoparticles and explore their antibacterial activity against biofilms. background: The number of deaths and diseases is expected to increase significantly due to nosocomial multidrug resistance and biofilm-forming bacterial infections. The World Health Organization supports researchers in identifying an alternative antimicrobial that works differently than traditional antibiotics. Silver nanoparticles could be a viable treatment because of their broad spectrum of activity, fast killing power, and low resistance value. Methods: AgNPs with specific physicochemical properties such as size, shape, and surface chemistry were prepared using a chemical reduction technique, and then characterized by DLS, SEM, and FTIR. The activity of AgNPs was tested alone and in combination with some antibiotics against MDR Gram-negative and Gram-positive planktonic bacterial cells and their biofilms. Finally, mammalian cell cytotoxicity and hemolytic activity were tested using VERO and human erythrocytes. Results: The findings of this study illustrate the success of the chemical reduction method in preparing AgNPs. Results showed that AgNPs have MIC values against planktonic organisms ranging from 0.0625 to 0.125 mg/mL, with the greatest potency against gram-negative bacteria. It also effectively destroyed biofilm-forming cells, with minimal biofilm eradication concentrations [MBEC] ranging from 0.125 to 0.25 mg/ml. AgNPs also had lower toxicity profiles for the MTT test when compared to hemolysis to erythrocytes. Synergistic effect was found between AgNPs and certain antibiotics, where the MIC was dramatically reduced, down to less than 0.00195 mg/ml in some cases. Conclusion: The present findings encourage the development of alternative therapies with high efficacy and low toxicity.

Abstract:Background: The misuse of antibiotics leads to a global increase in antibiotic resistance. Therefore, it is imperative to search for alternative compounds to conventional antibiotics. ZnO nanoparticles (Zn NP) are one of these alternatives because they are an effective option to overcome biofilm bacterial cells and a novel way to overcome multidrug resistance in bacteria. The current research study aims to characterize the efficacy of ZnO nanoparticles alone and in combination with other antibacterial drugs against bacterial biofilms. Methods: ZnO NPs were prepared by co-precipitation method, and their anti-biofilm and antibacterial activities alone or combined with four types of broad-spectrum antibacterial (Norfloxacin, Colistin, Doxycycline, and Ampicillin) were evaluated against E. coli and S. aureus bacterial strains. Finally, the cytotoxicity and the hemolytic activity were evaluated. Results: ZnO NPs were prepared, and results showed that their size was around 10 nm with a spherical shape and a zeta potential of -21.9. In addition, ZnO NPs were found to have a strong antibacterial effect against Gram-positive and Gram-negative microorganisms, with a minimum inhibitory concentration (MIC) of 62.5 and 125 μg/mL, respectively. Additionally, they could eradicate biofilmforming microorganisms at a concentration of 125 μg/m. ZnO NPs were found to be non-toxic to erythrocyte cells. Still, some toxicity was observed for Vero cells at effective concentration ranges needed to inhibit bacterial growth and eradicate biofilm-forming organisms. When combined with different antibacterial, ZnO NP demonstrated synergistic and additive effects with colistin, and the MIC and MBEC of the combination decreased significantly to 0.976 μg/mL against planktonic and biofilm strains of MDR Gram-positive bacteria, resulting in significantly reduced toxicity. Conclusion: The findings of this study encourage the development of alternative therapies with high efficacy and low toxicity. ZnO nanoparticles have demonstrated promising results in overcoming multi-drug resistant bacteria and biofilms, and their combination with colistin has shown a significant reduction in toxicity. Further studies are needed to investigate the potential of ZnO nanoparticles as a viable alternative to conventional antibiotics.

Until now, the effect of design patterns on software quality attributes has been studied by many researchers. However, the results are not the expected ones, in the sense that several studies suggest that there are cases when a design pattern is not the optimum way of designing a system. In this paper, we present the findings of a systematic literature review that aims at cataloging published design solutions, referenced as alternative design solutions, which are equivalent to design patterns and can be used when a design pattern instance is not the optimum design solution for a specific design problem.

Abstract:Antibacterial resistance is considered to be one of the major causes for mortality in coming years. In recent years green nanotechnology played a key role in addressing this problem. Biocompatible metal nanoparticles have gained popularity owing to their excellent therapeutic effects and minimal side effects. Method: We report the synthesis of AgNPs and their amoxicillin conjugates (Ag-amoxi) using Micromeria biflora crude flavonoid extracts. The physicochemical properties of the synthesized NPs and Ag-amoxi conjugates were systematically evaluated using scanning electron microscopy (SEM), energy dispersive X-ray (EDX) and X-ray diffraction (XRD) analysis, Fourier transform infrared (FTIR), and UV-visible (UV-Vis) spectroscopic techniques. Results: The average sizes of AgNPs and Ag-amoxi conjugates were 45 and 62 nm, respectively. We have also explored the antibacterial, antioxidant, anti-inflammatory, and analgesic properties of the AgNPs and Ag-amoxi conjugates through in vivo and in vitro analysis. The Ag-amoxi conjugates showed better antibacterial potential against Streptococcus Pneumoniae (S.P), Staphylococcus aureus (S.A), Pseudomonas aeruginosa (P.A), and Methicillin resistance Staphylococcus aureus (MRSA) strain both the drug and AgNPs. Similarly, in vivo anti-inflammatory studies revealed that both Ag-amoxi (68 %) and AgNPs (64 %) had strong anti-inflammatory effects, with (***p < 0.001) significance at a dose of 10 mg kg-1 body weight as compared to standard, amoxicillin (45 %), and flavonoids extract (48 %) at a dose of 100 mg kg-1. The findings of the antinociceptive activities (writhing and hot plate tests) demonstrated that the Ag-amoxi conjugates produced fewer writhing (15 in 20 s) and a shorter latency time of 22 s as compared to vehicle-treated (tramadol) animals, amoxicillin, and P.E at much lower doses. In vitro antioxidant studies revealed that the Ag-amoxi conjugate has the potential to be used as an antioxidant with an IC50 value of 43.58, compared with AgNPs (46.34), amoxicillin (58.17), compared to the standard of ascorbic acid (34.14). Conclusion: These results reveals that these biologically inspired AgNPs and Ag-amoxi conjugate could be used to improve antibiotic efficiency and could play a critical role in addressing the multidrug resistance problem in coming years.
Development of Folic Acid Functionalized Carbon Nanotubes for Efficient Delivery of Curcumin and Quercetin Against Pathogenic Bacteria

Abstract:Drug-resistant bacteria pose an alarming risk to public health globally. Specifically, infections caused by multidrug-resistant (MDR) Gram-positive bacteria (e.g., Staphylococcus aureus ) are an increasing global health concerns. Herein, we report the first comparative evaluation of folic acid functionalized carbon nanotubes (FA-OMC) loaded with curcumin and quercetin to enhance their efficacies against gram positive MDR S. aureus and E. coli . The carbon nanotubes were oxidized and underwent successive carbodimide coupling and passive drug entrapments to achieve the drug loaded nanocomposites, which were further characterized with TGA, PXRD, SEM, AFM, FTIR, and DLS. The drug release kinetics as well as the cytotoxic evaluation and antibacterial studies were also evaluated. The results revealed that our drug loaded nanocomposites showed high drug entrapment, wrecked layered in morphology, nano range in size, possess negative zeta potential, low PDI values, higher thermal stability and partial crystallinity. FTIR analysis of the drug loaded nanocomposites shows the possible interaction between drug and nanotubes. The developed formulations (Cur-FA-OMC and Que-FA-OMC) revealed extended drug release and higher biocompatibility against tested NIH/3T3 cells. The antimicrobial assessment of drug loaded nanoparticles against gram positive bacteria showed significant bactericidal efficacy as compared to drug alone and their morphological disruption were also confirmed by AFM microscopy. The outcomes of our study suggests that our developed drug loaded nanoformulations could be a viable option for reversing MDR bacteria. More research needed in future to consolidate these findings.

Abstract:Antimicrobial resistance (AMR), caused by microbial infections, has become a major contributor to morbid rates of mortality worldwide and a serious threat to public health. The exponential increase in resistant pathogen strains including Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) poses significant hurdles in the health sector due to their greater resistance to traditional treatments and medicines. Efforts to tackle infectious diseases caused by resistant microbes have prompted the development of novel antibacterial agents. Herein, we present selenium and copper oxide monometallic nanoparticles (Se-MMNPs and CuO-MMNPs), characterized using various techniques and evaluated for their antibacterial potential via disc diffusion, determination of minimum inhibitory concentration (MIC), antibiofilm, and killing kinetic action. Dynamic light scattering (DLS), scanning electron microscopy (SEM/EDX), and X-ray diffraction (XRD) techniques confirmed the size-distribution, spherical-shape, stability, elemental composition, and structural aspects of the synthesized nanoparticles. The MIC values of Se-MMNPs and CuO-MMNPs against S. aureus and E. coli were determined to be 125 μg/mL and 100 μg/mL, respectively. Time–kill kinetics studies revealed that CuO-MMNPs efficiently mitigate the growth of S. aureus and E. coli within 3 and 3.5 h while Se-MMNPs took 4 and 5 h, respectively. Moreover, CuO-MMNPs demonstrated better inhibition compared to Se-MMNPs. Overall, the proposed materials exhibited promising antibacterial activity against S. aureus and E. coli pathogens.
