Abstract:The growing use of graphene-based nanomaterials (GBNs) for various applications increases the probability of their environmental releases and calls for a systematic assessment of their potential impacts on soil invertebrates that serve as an important link along terrestrial food chains. Here, we investigated the response of earthworms (Eisenia fetida) to three types of multi-layer graphenes (MLGs) (G1, G2 and G3 with 12-15 layers) with variable morphology (lateral sizes: 7.4 +/- 0.3, 6.4 +/- 0.1 and 2.8 +/- 0.1 mm; thicknesses: 5.0 +/- 0.1, 4.2 +/- 0.1 and 4.0 +/- 0.2 nm, respectively) and hydrophobicity ((O + N)/C ratios: 0.029, 0.044 and 0.075; contact angles: 122.8, 118.8 and 115.1 degrees, respectively). Exposure to these materials was conducted for 28 days (except for 48-h avoidance test) separately in potting or farm soil at 0.2% and 1% by weight. Earthworms avoided both soils when amended with 1% of the smaller and more hydrophilic MLGs (G2 and G3), leading to a decreased trend in worm cocoon formation. The smallest and most hydrophilic MLG (G3), which was easier to assimilate, also significantly inhibited the viability (20.2 -56.0%) and mitochondrial membrane potential (32.0-48.5%) of worm coelomocytes in both soils. In contrast, oxidative damage (indicated by lipid peroxides) was more pronounced upon exposure to more hydrophobic and larger graphenic materials (G1 and G2), which were attributed to facilitated adhesion to and disruption of worm membranes. These findings highlight the importance of MLG morphology and hydrophobicity in their potential toxicity and mode of action, as well as ecological risks associated with incidental and accidental releases. Contributions of Altered Permeability of Intestinal Barrier and Defecation Behavior to Toxicity Formation from Graphene Oxide in Nematode Caenorhabditis Elegans

Abstract:Graphene oxide (GO) has been extensively studied for potential biomedical applications. Meanwhile, potential GO toxicity arises in both biomedical applications and non-biomedical products where environmental exposures may occur. In the present study, we examined the potential adverse effects of GO and the underlying mechanism using nematode Caenorhabditis elegans as the assay system. We compared the in vivo effects of GO between acute exposure and prolonged exposure, and found that prolonged exposure to 0.5-100 mg L-1 of GO caused damage on functions of both primary (intestine) and secondary (neuron and reproductive organ) targeted organs. In the intestine, ROS production was significantly correlated with the formation of adverse effects on functions of both primary and secondary targeted organs. GO could be translocated into intestinal cells with loss of microvilli, and distributed to be adjacent to or surrounding mitochondria. Prolonged exposure to GO resulted in a hyper-permeable state of the intestinal barrier, an increase in mean defecation cycle length, and alteration of genes required for intestinal development and defecation behavior. Thus, our data suggest that prolonged exposure to GO may cause potential risk to environmental organisms after release into the environment. GO toxicity may be due to the combinational effects of oxidative stress in the intestinal barrier, enhanced permeability of the biological barrier, and suppressed defecation behavior in C. elegans.
The Effect of Water Hardness on the Toxicity of Graphene Oxide to Bacteria in Synthetic Surface Waters.

Abstract:Graphene oxide (GO), used in a wide variety of applications, is increasingly being introduced into aquatic environments; this situation calls for research on GO toxicity to assess its environmental risks. In this study, the toxic effect of GO to E.coli was studied before and after its aggregation equilibrium in the synthetic surface waters (the soft water, moderately hard water, and hard water) to reveal the effects of GO aggregation and solution hardness. The cytotoxicity of GO increased with increasing solution hardness while decreased after GO aggregation. The 3 h 50% inhibitory concentration (IC50) values of dispersed GO in the soft water, moderately hard water, and hard water were 12.2 +/- 2.2, 8.5 +/- 1.5, and 4.0 +/- 1.0 mg/L, respectively. After 24 h shaking (aggregation equilibrium) in the synthetic surface waters, the dispersed GO aggregated and the 3 h IC50 values of GO aggregates in the three synthetic waters were 40.3 +/- 6.9, 15.9 +/- 2.2, and 7.5 +/- 1.5 mg/L, respectively. The dispersed GO sheets wrapped E. coli cells and cut the cell membrane, resulting in the disruption of cell membrane and the cell inactivation. With increasing water hardness, the heteroaggregation between GO sheets/aggregates and E. coli cells was enhanced, resulting in the increase of toxic effect. The GO aggregates could also entrap E.coli cells while exhibited limited effect on cell membrane disruption without sharp edges, thereby causing the lower toxic effect compared with the dispersed GO sheets. These outcomes shed new light on the assessment of ecological effects of GO.
Coexposure to Environmental Concentrations of Cis-Bifenthrin and Graphene Oxide: Adverse Effects on the Nervous System During Metamorphic Development of Xenopus Laevis

Abstract:Despite the great concerns associated with the combined biological effects of nanoparticles and insecticides, the current understanding of the corresponding ecological risks remains limited. Xenopus laevis (X. laevis) tadpoles were exposed to various concentrations of typical pyrethroid (cis-bifenthrin; cis-BF), either alone or in combination with graphene oxide (GO), for 21 days. The presence of GO resulted in increased bioconcentration of cis-BF and a higher 1S-enantiomer fraction. Exposure to cis-BF and GO caused further reduction in pre-metamorphic developmental rates and activated dopaminergic, noradrenergic, and serotonergic neurotransmitter systems. Reduced tadpole activity and levels of genomic DNA methylation at cytosine nucleotides (5hmC) were observed in the coexposure groups. These results indicate that GO enhance the bioconcentration of cis-BF and promote the conversion of its 1R-enantiomer to the 1S form, which lead to disruption of neurotransmitter systems as well as interference in metamorphic development.
Toxicity of Graphene Oxide and Multi-Walled Carbon Nanotubes Against Human Cells and Zebrafish

Abstract:Graphene possesses unique physical and chemical properties, which have inspired a wide range of potential biomedical applications. However, little is known about the adverse effects of graphene on the human body and ecological environment. The purpose of our work is to make assessment on the toxicity of graphene oxide (GO) against human cell line (human bone marrow neuroblastoma cell line and human epithelial carcinoma cell line) and zebrafish (Danio rerio) by comparing the toxic effects of GO with its sister, multi-walled carbon nanotubes (MWNTs). The results show that GO has a moderate toxicity to organisms since it can induce minor (about 20%) cell growth inhibition and slight hatching delay of zebrafish embryos at a dosage of 50 mg/L, but did not result in significant increase of apoptosis in embryo, while MWNTs exhibit acute toxicity leading to a strong inhibition of cell proliferation and serious morphological defects in developing embryos even at relatively low concentration of 25 mg/L. The distinctive toxicity of GO and MWNTs should be ascribed to the different models of interaction between nanomaterials and organisms, which arises from the different geometric structures of nanomaterials. Collectively, our work suggests that GO does actual toxicity to organisms posing potential environmental risks and the result is also shedding light on the geometrical structure-dependent toxicity of graphitic nanomaterials.
Exposure to Graphene Oxide at Environmental Concentrations Induces Thyroid Endocrine Disruption and Lipid Metabolic Disturbance in Xenopus Laevis.

Abstract:Graphene oxide (GO) has become a topic of increasing concern for its environmental and health risks. However, the potential toxic effects of GO on wildlife remain limited. The present study chose the Xenopus laevis tadpole as a model to assess the thyroid endocrine disruption as well as the lipid metabolic disturbance of GO. Tadpoles at the 51 stage were exposed to GO (0, 0.01, 0.1, and 1 mg/L) for 21 days, when tadpoles were undergoing an extremely complicated phase of morphological changes and growth. GO treatment showed obvious developmental toxicity, such as shortened snout-to-vent length (SVL) and hind limb length (HLL), decreased body weight, and delayed developmental stage. Exposure to GO also induced obvious decreases in whole-body triiodothyronine (T3) and thyroxin (T4) concentrations. The mRNA expression of genes related to the hypothalamic-pituitary-thyroid (HPT) axis also changed significantly. Furthermore, we observed significant decline in the fatty acids and triglycerides (TGs) concomitantly with changes in the expression of genes involved in the synthesis and metabolism of lipids in GO exposure groups. In contrast, high-density lipoprotein (HDL) and total bile acid levels increased remarkably, but cholesterol and low-density lipoprotein (LDH) levels showed no obvious changes. Taken together, the results revealed for the first time that GO could induce thyroid endocrine disruption and produce obvious disturbance effect on lipid synthesis and metabolism. Abstract:The mass production of graphene oxide (GO) unavoidably elevates the chance of human exposure, as well as the possibility of release into the environment with high stability, raising public concern as to its potential toxicological risks and the implications for humans and ecosystems. Therefore, a thorough assessment of GO toxicity, including its potential reliance on key physicochemical factors, which is lacking in the literature, is of high significance and importance. In this study, GO toxicity, and its dependence on oxidation level, elemental composition, and size, were comprehensively assessed. A newly established quantitative toxicogenomic-based toxicity testing approach, combined with conventional phenotypic bioassays, were employed. The toxicogenomic assay utilized a GFP-fused yeast reporter library covering key cellular toxicity pathways. The results reveal that, indeed, the elemental composition and size do exert impacts on GO toxicity, while the oxidation level exhibits no significant effects. The UV-treated GO, with significantly higher carbon-carbon groups and carboxyl groups, showed a higher toxicity level, especially in the protein and chemical stress categories. With the decrease in size, the toxicity level of the sonicated GOs tended to increase. It is proposed that the covering and subsequent internalization of GO sheets might be the main mode of action in yeast cells.

Abstract:With the extensive application of graphene oxide (GO), its leakage and release into wastewater treatment plants become inevitable. However, the toxicity of graphene oxide (GO) on nitrification process and the underlying mechanisms still remain unclear. In this study, the toxic effects of GO at concentration of 10 and 100 mg/L in 4 h and 10 days were evaluated with sealed reactors operated in sequencing batch mode. In the initial 4 h, both GO concentrations showed no negative effect on nitrogen conversion. However, the exposure to 100 mg/L GO significantly weakened the NH+ 4-N and NO- 2-N conversion capabilities and intensified the nitrous oxide (N2O) generation after 10 days. Extracellular polymeric substance (EPS) analysis suggested that 100 mg/L GO decreased the protein content of the nitrifying activated sludge. Moreover, reactive oxygen species (ROS) level was promoted by 100 mg/L GO owing to the impaired endogenous antioxidant enzymes including superoxide dismutase (SOD) and catalase (CAT), which caused oxidative stress to bacteria. Finally, quantitative PCR results confirmed that nitrite-oxidizing bacteria (NOB) and complete ammonia oxidizing bacteria (CAOB) were more sensitive to GO, which was the primary cause for the significant promotion of N2O generation in the high GO concentration. This study offered new insights in the toxicity of GO on nitrification and N2O generation in the terms of dose and exposure time.
Microbial response to the chronic toxicity effect of graphene and graphene oxide nanomaterials within aerobic granular sludge systems

Abstract:Nanomaterials present in wastewater can pose a significant threat to aerobic granular sludge (AGS) systems. Herein, we found that compared to graphene nanomaterials (G-NMs), the long-term presence (95 days) of graphene oxide nanomaterials (GO-NMs) resulted in an increased proliferation of filamentous bacteria, poorer sedimentation performance (SVI30 of 74.1 mL/g) and smaller average particle size (1224.4 µm) of the AGS. In particular, the GO-NMs posed a more significant inhibitory effect to the total nitrogen removal efficiency of AGS (decreased by 14.3 %), especially for the denitrification process. The substantial accumulation of GO-NMs within the sludge matrix resulted in a higher level of reactive oxygen species in AGS compared to G-NMs, thereby inducing lactate dehydrogenase release, and enhancing superoxide oxidase and catalase activities. Such excessive oxidative stress could potentially result in a significant reduction in the activity of nitrogen metabolism enzymes (e.g., nitrate reductase and nitrite reductase) and the expression of key functional genes (e.g., nirS and nirK). Altogether, compared to G-NMs, prolonged exposure to GO-NMs had a more significant chronic toxicity effect on AGS systems. These findings implied that the presence of G-NMs and GO-NMs is a hidden danger to biological nitrogen removal and should receive more attention.
Effect of graphene oxide on conformation and activity of catalase

Abstract:To explore the biocompatibility of graphene oxide (GO) with proteins, we investigated its effects on conformation and activity of catalase, by using molecular spectroscopic techniques such as UV–visible absorption, fluorescence, circular dichorism (CD) and fourier transform infrared (FT-IR) spectroscopy, and conducting in vitro enzymatic activity assay. CD combined with FT-IR and UV–visible spectra measurements indicated that GO induced the decrease of α-helical content and the increase of β-sheet structure in catalase, resulting in the loosening and unfolding of the protein skeleton. The internal hydrophobic amino acids (mainly tryptophan) in the protein were exposed, and their characteristic fluorescence was distinctly quenched, according to synchronous fluorescence and fluorescence emission together with UV–visible spectra analyses. The results showed that the tertiary and secondary structure of catalase is changed by GO. The conformational changes of the protein were also found to show a dependence on concentration and time of GO treatment. Moreover, the activity assay indicated that catalase activity is inhibited by GO. The study demonstrated that GO could cause negative effects on proteins, and the biosafety of its application may worth further investigation.
Toxicological Effects of TiO2 and ZnO Nanoparticles in Soil on Earthworm Eisenia Fetida

Abstract:Nanoparticles (NPs) of TiO2 and ZnO are receiving increasing attention due to their widespread applications. To evaluate their toxicities to the earthworm Eisenia fetida (Savigny, 1826) in soil, artificial soil systems containing distilled water, 0.1, 0.5, 1.0 or 5.0gkg−1 of NPs were prepared and earthworms were exposed for 7 days. Contents of Zn and Ti in earthworm, activities of antioxidant enzymes, DNA damage to earthworm, activity of cellulase and damage to mitochondria of gut cells were investigated after acute toxicity test. The results from response of the antioxidant system combined with DNA damage endpoint (comet assay) indicated that TiO2 and ZnO NPs could induce significant damage to earthworms when doses were greater than 1.0gkg−1. We found that Ti and Zn, especially Zn, were bioaccumulated, and that mitochondria were damaged at the highest dose in soil (5.0gkg−1). The activity of cellulase was significantly inhibited when organisms were exposed to 5.0gkg−1 of ZnO NPs. Our study demonstrates that both TiO2 and ZnO NPs exert harmful effects to E. fetida when their levels are higher than 1.0gkg−1 in soil and that toxicity of ZnO NPs was higher than TiO2.

Abstract:Simultaneous exposure to multiple pollutants has received great concerns considering that the interactions between pollutants can alter the environment fate and bioavailability of pollutants with potentially deleterious effects. Graphene oxide (GO) has been widely used in many areas including environmental remediation, biology and agriculture. However, researchers have largely ignored the combined toxicity of GO with coexisting toxicants. Cis-bifenthrin (cis-BF), a typical synthetic pyrethroid insecticide, was frequently detected in the environment, which raised the possibility of interaction between cis-BF and GO. Our study investigated the toxic effects of cis-BF alone or combined with GO on the lipid homeostasis in Xenopus laevis. Tadpoles at 51 stage were exposed to cis-BF (0, 12, 60 and 300 ng/L) or in their combination with GO (0.1 mg/L) for 21 days. Coexposure to cis-BF and GO deteriorated the lipid homeostasis disruption in tadpoles. The upor down-regulation of lipogenesis genes expression and enzymes activity were amplified in the coexposure groups. Furthermore, the presence of GO enhanced the deleterious impacts of cis-BF on the hepatic function in tadpoles. This study uniquely shows that GO promotes the lipotoxicity and hepatic function deficit caused by cis-BF exposure in frog.
Biocompatibility of graphene oxide intravenously administrated in mice—effects of dose, size and exposure protocols

Abstract:Graphene oxide (GO) shows great promise in in vivo drug delivery and therapy applications. However, several reports have reported an in vivo toxicity of GO. In this study, we found that the toxicity of GO intravenously injected into mice could be tuned by dose, size and exposure protocols of GO. The exposure to a single dose of 2.1 mg kg(-1) (single-high-dose exposure) small size GO or large size GO caused macrophage nodule formation in the lungs of the mice, and the exposure to seven repeated doses of 0.3 mg kg(-1) (multiple-low-dose exposure) large size GO also induced small macrophage nodule formation, serious lymphocyte infiltration around the bronchioles in the lungs of the mice, and even death of the mice. Nephritic inflammatory reactions were also observed after the multiple-low-dose exposure to large size GO. However, no obvious lung toxicity but hepatic inflammatory infiltration was observed after the exposure to multiple-low-dose small size GO. GO accumulation in the macrophage nodules was verified by Raman mapping. These findings will benefit the applications of GO in the future, especially in biomedical fields.
Toxicity of Zinc Oxide Nanoparticles in the Earthworm, Eisenia Fetida and Subcellular Fractionation of Zn

Abstract:The extensive use of nanoparticles (NPs) in a variety of applications has raised great concerns about their environmental fate and biological effects. This study examined the impact of dissolved organic matter (DOM) and salts on ZnO NP dispersion/solubility and toxicity to the earthworm Eisenia fetida. To be able to better evaluate the toxicity of NPs, exposure in agar and on filter paper was proposed for enabling a comparison of the importance of different uptake routes. A dose-related increase in mortality was observed in earthworms exposed in agar with almost 100% mortality after 96 h exposure to the highest concentration (1000 mg ZnO/kg agar). Scanning electron microscopy (SEM) showed that the addition of salts enhanced the aggregation of ZnO NPs in agar and consequently affected the dissolution behavior and biological availability of the particles. On filter paper, mortality was the highest at the lowest exposure concentration (50 mg ZnO/L) and seemed to decrease with increasing exposure levels. TEM images of ZnO showed that the solubility and morphology of NPs were changed dramatically upon the addition of humic acids (HA). The subcellular distribution pattern of Zn in earthworms after 96 h exposure in agar and on filter paper showed that the Zn taken up via dietary ZnO particles (from agar) was mainly found in organelles and the cytosol while the Zn accumulated as soluble Zn from filter paper was mainly distributed in cell membranes and tissues. Antioxidant enzymatic activities (SOD, CAT, and GSH-px) were investigated in the worms surviving the toxicity tests. A slight increase of SOD activities was observed at the lowest exposure dose of ZnO (50mg/kg), followed by a decrease at 100mg/kg in the agar cubes. Activities of both CAT and GSH-Px enzymes were not significantly influenced in the worms exposed to agar, although a slight decrease at 500 and 1000 mg ZnO/kg agar was observed. A similar change trend of SOD activities was observed for the earthworms on filter paper, but a significant decrease began at a higher ZnO NP concentration of 500 mg ZnO/L. The use of soil extracts instead of deionized water (DW) to simulate a realistic exposure system significantly reduced the toxicity of the ZnO NPs on filter paper, which increases the predictive power of filter paper toxicity tests for the environmental risk assessment of NPs.

Abstract:Graphene oxide (GO) possesses versatile applicability and high hydrophilicity, thus may have frequent contact with aquatic organisms. However, the ecological risks of GO in aquatic ecosystems remain largely unexplored currently. This study evaluated the comprehensive toxicological effects of GO on Daphnia magna, a key species in fresh water ecosystem. The results revealed nonsevere acute toxicities, including immobility (72 h EC50: 443 mg/L) and mortality (72 h LC50: 45.4 mg/L), of GO on D. magna. To understand the underlying mechanism of GO exposure, changes in superoxide dismutase (SOD) and lipid peroxidation (LPO) of D. magna exposed to GO were correlated, which revealed elevated GO-mediated oxidative stress and damages, especially in the long-time and high-dose exposure groups. The observations of in vivo fluorescence labeled with 2', 7'-dichlorofluorescin further demonstrated that reactive oxygen species were concentrated in daphnia guts, which corresponded with the high bioaccumulation level (5 mg/L, 24 h body burden: 107.9 g/kg) of GO in daphnia guts. However, deputation of GO from daphnia was not difficult. Daphnia almost released all GO within 24 h after it was transferred to clean water. These results hence suggest that GO could accumulate and induce significant oxidative stress in the gut of D. magna, while D. daphnia can also relieve the acute toxicity by depurating GO. Abstract:Nanomaterials of graphene and its derivatives have been widely applied in recent years, but whose impacts on the environment and health are still not well understood. In the present study, the potential adverse effects of graphite (G), graphite oxide nanoplatelets (GO) and graphene quantum dots (GQDs) on the motor nervous system were investigated using nematode Caenorhabditis elegans as the assay system. After being characterized using TEM, SEM, XPS and PLE, three nanomaterials were chronically exposed to C. elegans for 6 days. In total, 50-100 mg l(-1) GO caused a significant reduction in the survival rate, but G and GDDs showed low lethality on nematodes. After chronic exposure of sub-lethal dosages, three nanomaterials were observed to distribute primarily in the pharynx and intestine; but GQDs were widespread in nematode body. Three graphene-based nanomaterials resulted in significant declines in locomotor frequency of body bending, head thrashing and pharynx pumping. In addition, mean speed, bending angle-frequency and wavelength of the crawling movement were significantly reduced after exposure. Using transgenic nematodes, we found high concentrations of graphene-based nanomaterials induced down-expression of dat-1::GFP and eat-4::GFP, but no significant changes in unc-47::GFP. This indicates that graphene-based nanomaterials can lead to damages in the dopaminergic and glutamatergic neurons. The present data suggest that chronic exposure of graphene-based nanomaterials may cause neurotoxicity risks of inducing behavioral deficits and neural damage. Metabolism of Phenol and Cresols by Mutants of Pseudomonas putida

Abstract:Mutant strains of Pseudomonas putida strain U have been obtained which are deficient in enzymes of the degradative pathways of phenol and cresols. Mutant strains deficient in catechol 2, 3-oxygenase accumulated the appropriate catechol derivative from cresols. A mutant strain which would not grow on either phenol or a cresol was shown to be deficient in both 2-hydroxymuconic semialdehyde hydrolase and a nicotinamide adenine dinucleotide, oxidized form, (NAD+)-dependent aldehyde dehydrogenase. When this strain was grown in the presence of phenol or a cresol, the appropriate product of meta fission of these compounds accumulated in the growth medium. A partial revertant of this mutant strain, which was able to grow on ortho- and meta-cresol but not para-cresol, was shown to have regained only the hydrolase activity. This strain was used to show that the products of meta ring fission of the cresols and phenol are metabolized as follows: (i) ortho- and meta-cresol exclusively by a hydrolase; (ii) para-cresol exclusively by a NAD+-dependent aldehyde dehydrogenase; (iii) phenol by both a NAD+-dependent dehydrogenase and a hydrolase in the approximate ratio of 5 to 1. This conclusion is supported by the substrate specificity and enzymatic activity of the hydrolase and NAD+-dependent aldehyde dehydrogenase enzymes of the wild-type strain. The results are discussed in terms of the physiological significance of the pathway. Properties of some of the mutant strains isolated are discussed.
Effects of multi-walled carbon nanotubes in soil on earthworm growth and reproduction, enzymatic activities, and metabolomics

Abstract:Increased production and environmental release of multi-walled carbon nanotubes (MWCNTs) increase soil exposure and potential risk to earthworms. However, MWCNT toxicity to earthworms remains unclear, with some studies identifying negative effects and others negligible effects. In this study, to determine whether exposure to MWCNTs negatively affects earthworms and to elucidate possible mechanisms of toxicity, earthworms were exposed to sublethal soil concentrations of MWCNTs (10, 50, and 100 mg/kg) for 28 days. Earthworm growth and reproduction, activities of cytochrome P450 (CYP) isoforms (1A2, 2C9, and 3A4) and antioxidant enzymes (superoxide dismutase (SOD), catalase (CAT), and glutathione-s-transferase (GST)), and metabolomics were determined. Effects of MWCNTs on earthworms depended on exposure concentration. Exposure to MWCNTs did not significantly affect growth and reproduction of individual earthworms. Exposure to 50 mg/kg MWCNTs significantly increased activities of CYP2C9, CYP3A4, SOD, CAT, and GST but clearly reduced levels of L-aspartate, L-asparagine, and glutamine. With exposure to 100 mg/kg MWCNTs, toxic effects on earthworms were observed, with significant inhibition in activities of CYP isoenzymes and SOD, significant reductions in L-aspartate, L-asparagine, glutamine, and tryptophan, and simultaneous accumulations of citrate, isocitrate, fumarate, 2-oxoglutarate, pyruvate, D-galactose, carbamoyl phosphate, formyl anthranilate, hypoxanthine, and xanthine. Results suggest that toxicity of MWCNTs to earthworms is associated with reduced detoxification capacity, excessive oxidative stress, and disturbance of multiple metabolic pathways, including amino acids metabolism, the tricarboxylic acid cycle, pyruvate metabolism, D-galactose metabolism, and purine metabolism. The study provides new insights to better understand and predict the toxicity of MWCNTs in soil.

Abstract:Recently, attempts have been made to apply graphene oxide (GO) in the field of biology and medicine, such as DNA sensing and drug delivery with some necessary modifications. Therefore, the toxicity of GO must be evaluated before it is applied further in biomedicine. In this paper, the cytotoxicity and genotoxicity of GO to human lung fibroblast (HLF) cells have been assessed with methyl thiazolyl tetrazolium (MTT), sub-G1 measurement and comet assays, and the mechanism of its toxicity has been explored. Various modifications of GO have been made to help us determine the factors which could affect the toxicity of GO. The results indicated that cytotoxicity and genotoxicity of GO to HLF cells were concentration dependent, and the genotoxicity induced by GO was more severe than the cytotoxicity to HLF cells. Oxidative stress mediated by GO might explain the reason of its toxic effect. Furthermore, the electronic charge on the surface of GO would play a very important role in the toxicity of GO to HLF cells. Copyright (C) 2013 John Wiley & Sons, Ltd.

Abstract:Graphene and its derivatives have attracted great research interest for their potential applications in electronics, energy, materials and biomedical areas. However, little information of their toxicity and biocompatibility is available. Herein, we performed a comprehensive study on the toxicity of graphene oxide (GO) by examining the influences of GO on the morphology, viability, mortality and membrane integrity of A549 cells. The results suggest that GO does not enter A549 cell and has no obvious cytotoxicity. But GO can cause a dose-dependent oxidative stress in cell and induce a slight loss of cell viability at high concentration. These effects are dose and size related, and should be considered in the development of bio-applications of GO. Overall, GO is a pretty safe material at cellular level, which is confirmed by the favorable cell growth on GO film.
