Abstract:We report the first determination of lattice structure for classical (fatty acid and alcohol) monolayer systems on the surface of water, based on our observation of first- and second-order x-ray diffraction peaks. Monolayers of both heneicosanoic acid and heneicosanol in the highest-pressure phase pack into a distorted hexagonal lattice with orthorhombic cell dimensions of a\ensuremath{\simeq}7.5 t/rA\r{}, b\ensuremath{\simeq}5.0 i/rA\r{}. From the observed peak intensities, we have calculated the molecular tilt directions and magnitudes, using the assumption (reasonable given the dense packing) that the chains are parallel to each other and in the all-trans configuration. the temperature range 0--5 \ifmmode^\circ\else\textdegree\fi{}C, the molecules tilt towards the b axis, but at higher temperatures the tilt direction changes to the a axis.

Abstract:Fundamental studies of the supramolecular layer structures, dynamics and water permeation free energy of hexa-acyl-chain Lipid A and its analogue of tetra-acyl chains would be useful for polymer membranes design for endotoxin removal in water treatment, drug delivery and other biotechnologies. In this work, we studied their supramolecular bilayer by using molecular dynamics simulations and efficient free energy computations. Our simulation accuracy was verified by the agreement between the bilayer structural properties (structure factor, bilayer thickness, and the area per lipid) and lateral diffusion coefficient in our simulation and experimental measurements. More importantly, our simulation for the first time illustrated hexagonal compact packing of the hydrocarbon acyl chains within a leaflet of Lipid A membrane (at 298 K and water content of 40 wt %), which is consistent with experiments. In contrast, Lipid A analogue is found with less ordered ripple structures at the same condition. Our study also demonstrated slower dynamics and larger and broader free energy barrier (∼23 kJ/mol) for water permeation for Lipid A, compared with that of Lipid A analogue. Moreover, the analysis of dynamics showed that highly hydrated hydrophilic diglucosamine backbone is structurally stable, whereas the interdigitated hydrophobic acyl chain tails inside the membrane with faster dynamics screen the aqueous environment from the lipid interior and also reinforce the membrane's structural stability.

Abstract:We have used diffraction of synchrotron radiation to study Langmuir monolayers of several different materials. Lead octadecanoate (stearate) was seen to undergo first order melting, although there is no clear indication of this in its isotherm. 1-eicosanol, which has a “kink” in its isotherm, shows diffraction peaks of similar size both above and below this kink. Tetracosanoic acid shows, at high pressure, a second peak close to the first.

Abstract:We have used synchrotron radiation to study organic monolayers on water ("Langmuir films"). At high monolayer pressures, lead stearate [Pb${({\mathrm{C}}_{17}{\mathrm{H}}_{35}\mathrm{C}\mathrm{O}\mathrm{O})}_{2}$] shows a powder peak at 1.60 ${\mathrm{\AA{}}}^{\ensuremath{-}1}$, implying an area per unit cell of 17.8 ${\mathrm{\AA{}}}^{2}$ if the lattice is triangular. The correlation length is about 250 \AA{}. Lignoceric acid (${\mathrm{C}}_{23}$${\mathrm{H}}_{47}$COOH) shows a similar peak even though no heavy ions are attached. When the pressure is reduced, the peak in lead stearate does not observably move or broaden; below the "knee" in the isotherm, however, the peak height decreases slowly with increasing area, implying a first-order melting transition.

Abstract:The interactions of lanthanide ions and the Ln-DTPA (DTPA = diethylenetriaminepentaacetate) complex with di palmitoylphosphatidylcholine (DPPC) and dipalmitoylphosphatidylethanolamine (DPPE) bilayers are studied by 2D NOESY and FT-Raman spectroscopy. Proton NMR spectroscopic results show that lanthanide ions combine with phosphate groups in the polar region of the outer layer of DPPC liposomes, leading to the separation in chemical shift of the proton signal of N(CH3)(3) The conformational change of the O-C-C-N+ backbone from the gauche conformer to the trans one is not found; i.e., the orientation of the polar headgroup is still parallel to the surface of the bilayers. The Ln-DTPA complex at low concentration in a pH 7.4 solution localizes far away from bilayers and thereby has little effect on the structure of bilayers. The FT-Raman spectroscopic results indicate that lanthanide ions affect strongly the fluidity of acyl chains of DPPE bilayers while the Ln-DTPA complex affects it slightly.
Unusual phosphatidylcholine lipid phase behavior in the ionic liquid ethylammonium nitrate

Abstract:Hypothesis: The forces that govern lipid self-assembly ionic liquids are similar to water, but their different balance can result in unexpected behaviour. Experiments: The self-assembly behaviour and phase equilibria of two phospholipids, 1,2-distearoyl-sn-glycero-3-phosphocholine (DSPC) and 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC), in the most common protic ionic liquid, ethylammonium nitrate (EAN) have been investigated as function of composition and temperature by small- and wide-angle X-ray scattering (SAXS/WAXS) and small-angle neutron scattering (SANS). Findings: Both lipids form unusual self-assembly structures and show complex and unexpected phase behaviour unlike that seen in water; DSPC undergoes a gel Lβ to crystalline Lc phase transition on warming, while POPC forms worm-like micelles L1 upon dilution. This surprising phase behaviour is attributed to the large size of the EAN ions that solvate the lipid headgroup compared to water changing amphiphile packing. Weaker H-bonding between EAN and lipid headgroups also contributes. These results provide new insight for the design of lipid based nanostructured materials in ionic liquids with atypical properties.

Abstract:The polar headgroup of dipalmitoylphosphatidylcholine (DPPC) molecule both in gas phase and aqueous solution is investigated by the hybrid quantum mechanical/molecular mechanical (QM/MM) method, in which the polar head of DPPC molecule and the bound water molecules are treated with density functional theory (DFT), while the apolar hydrocarbon chain of DPPC molecule is treated with MM method. It is demonstrated that the hybrid QM/MM method is both accurate and efficient to describe the conformations of DPPC headgroup. Folded structures of headgroup are found in gas phase calculations. In this work, both monohydration and polyhydration phenomena are investigated. In monohydration, different water association sites are studied. Both the hydration energy and the quantum properties of DPPC and water molecules are calculated at the DFT level of theory after geometry optimization. The binding force of monohydration is estimated by using the scan method. In polyhydration, more extended conformations are found and hydration energies in different polyhydration styles are estimated.

Abstract:Membrane proteins are frequently reconstituted in different detergents as a prerequisite to create a phospholipid environment reminiscent of their native environment. Different detergent characteristics such as their chain length and bond types could affect the structure and function of proteins. Yet, they are seldom taken into account when choosing a detergent for structural studies. Here, we explore the effect of different detergents and lipids with varying degrees of double- or single-bond composition on 1H-15N transverse relaxation optimized spectroscopy spectra of the outer membrane protein W (OmpW). We observed changes in nuclear magnetic resonance chemical shifts for OmpW reconstituted in micelles, bicelles, and nanodiscs, depending on their detergent/lipid composition. These results suggest that a careful evaluation of detergents is necessary, so as not to jeopardize the structure and function of the protein.

Abstract:The internal molecular structure of lipid-based formulations (LBFs) is poorly understood. In this work we aimed at establishing coarse-grained molecular dynamics simulations as a tool for rapid screening and investigation of the internal environment of these formulations. In order to study complex LBFs composed of different kinds of lipids we simulated a number of systems containing either medium-chain or long-chain lipids with varying proportions of tri-, di-, and monoglycerides. Structural and dynamic measurements and analyses identified that the internal environment in a mixture of lipids was locally ordered even in the absence of water, which might explain some of the previously reported effects on drug solubility in these systems. Further, phase changes occurring upon water dispersion are well captured with coarse-grained simulations. Based on these simulations we conclude that the coarse-grained methodology is a promising in silico approach for rapid screening of structures formed in complex formulations. More importantly it facilitates molecular understanding of interactions between excipients and water at a feasible time scale and, hence, opens up for future virtual drug formulation studies.

Abstract:We report here the first AFM studies of phase-separated monolayer Langmuir-Blodgett films composed of a mixture of L-alpha-dipalmitoylphosphocholine (DPPC) and peptide bond bridged dl-alpha-phenylalanine-tetraphenylporphyrin (PTPP) under different surface pressures. The shapes of DPPC domains as well as microstructures within domains were also directly observed by AFM.

Abstract:Phospholipids are essential building blocks of biological membranes. Despite of vast amount of accurate experimental data the atomistic resolution structures sampled by the glycerol backbone and choline headgroup in phoshatidylcholine bilayers are not known. Atomistic resolution molecular dynamics simulation model would automatically resolve the structures giving an interpretation of experimental results, if the model would reproduce the experimental data. In this work we compare the C-H bond vector order parameters for glycerol backbone and choline headgroup between 14 different atomistic resolution models and experiments in fully hydrated lipid bilayer. The current models are not accurately enough to resolve the structure. However, closer inspection of three best performing models (CHARMM36, GAFFlipid and MacRog) suggest that improvements in the sampled dihedral angle distributions would potentilly lead to the model which would resolve the structure. Despite of the inaccuracy in the fully hydrated structures, the response to the dehydration, i.e. P-N vector tilting more parallel to membrane normal, is qualitatively correct in all models. The CHARMM36 and MacRog models describe the interactions between lipids and cholesterol better than Berger/Höltje model. Abstract:We studied interfacial properties of a series of methyl and ethyl esters of enantioenriched syn-2,3-dihydroxy fatty acids with different chain lengths at the air–water interface, using a Langmuir type film balance and a Brewster angle microscope (BAM). After analyzing their surface pressure (Π)–area (A) isotherms, we inferred that these molecules existed as an E conformation in the liquid-expanded (LE) phase of monolayers, and the E conformation of molecules changed into a Z conformation during the LE–LC transition in a monolayer. BAM images evidenced the formation of elongated LC aggregates. This is possibly induced by the intermolecular hydrogen bonds, leading to the anisotropic growth of LC domains, on the basis of the FT-IR spectroscopy data. The enthalpy change of the LE–LC phase transition is considered to result from the three types of intermolecular interactions at the air–water interface during compression of these amphiphiles. These findings are discussed in terms of various physical factors that influenced intermolecular interactions and macroscopic aggregations of these amphiphiles.

Abstract:The Langmuir-Blodgett bilayer films of phosphatidic acid(DPPA) have been investigated by using atomic force microscopy in both large and nano-scale. Large scale AFM image shows that the bilayer of DPPA is a plane film with some defects which enable us to detect the thickness of bilayer by a trace of section analysis across the defects structure, Molecularly resolved images show that the polar head groups (phosphate groups) are packed in an ordered hexagonal organization with a long range orientational and positional ordering, Intermolecular hydrogen bonding interactions between adjacent phosphate groups are first visualized and local supramolecular structures are first observed in polar region of DPPA bilayer. Some phosphate groups in polar region linked with each other through intermolecular hydrogen bonding interaction to form local supramolecular structure with a diameter of 1.5 nm in size.
Keck Speckle Imaging of the White Dwarf G29-38: No Brown Dwarf Companion Detected

Abstract:The white dwarf Giclas 29-38 has attracted much attention due to its large infrared excess and the suggestion that excess might be due to a companion brown dwarf. We observed this object using speckle interferometry at the Keck telescope, obtaining diffraction-limited resolution (55 milliarcseconds) at K band, and found it unresolved. Assuming the entire K band excess is due to a single point-like companion, we place an upper limit on the binary separation of 30 milliarcseconds, or 0.42 AU at the star's distance of 14.1 pc. This result, combined with astroseismological data and other images of G29-38, supports the hypothesis that the source of the near-infrared excess is not a cool companion but a dust cloud.

Abstract:Highly oriented solid-supported lipid membranes in stacks of controlled number $N \simeq 16$ (oligo-membranes) have been prepared by spin-coating using the uncharged lipid model system 1,2-dimyristoyl-sn-glycero-3-phosphocholine (DMPC). The samples have been immersed in aqueous polymer solutions for control of osmotic pressure and have been studied by X-ray reflectivity. The bilayer structure and fluctuations have been determined by modelling the data over the full q-range. Thermal fluctuations are described using the continuous smectic Hamiltonian with the appropriate boundary conditions at the substrate and at the free surface of the stack. The resulting fluctuation amplitudes and the pressure-distance relation are discussed in view of the inter-bilayer potential.

Abstract:In the present study, an amphiphile 2,4-dihydroxy-N-octadecylbenzylideneamine with novel structure was designed and synthesized. The aggregation structure of this amphiphile in LB films was investigated through FTIR spectroscopic techniques. The surface pressure-dependent infrared spectra of LB films have revealed some important insights on the bonded properties of the headgroup for this amphiphile. It was found that the types of H-bond formed among/between the Schiff base headgroups are determined by the deposition pressure. Intermolecular H-bond is prominent in Z-type LB film deposited at a surface pressure of ≤30 mN/m, while intramolecular H-bond formed between 2-hydroxyl group and the azomethine nitrogen is more favorable in Y-type LB film fabricated at a surface pressure of ≥40 mN/m. The variable-temperature infrared spectra of LB films further provide powerful evidence for the conversion between intermolecular and intramolecular H-bond, depending on the surface pressure.

Abstract:The presented studies were aimed at determining the interactions in model membranes (Langmuir monolayers) created of phospholipids (PL) isolated from Legionella gormanii bacteria cultured with (PL + choline) or without (PL − choline) choline and to describe the impact of an antimicrobial peptide, human cathelicidin LL-37, on PL's monolayer behavior. The addition of choline to the growth medium influenced the mutual proportions of phospholipids extracted from L. gormanii. Four classes of phospholipids—phosphatidylcholine (PC), phosphatidylethanolamine (PE), phosphatidylglycerol (PG), cardiolipin (CL), and their mixtures—were used to register compression isotherms with or without the LL-37 peptide in the subphase. Based on them the excess area (Ae), excess (ΔGe), and total (ΔGm) Gibbs energy of mixing were determined. The thermodynamic analyses revealed that the PL − choline monolayer showed greater repulsive forces between molecules in comparison to the ideal system, while the PL + choline monolayer was characterized by greater attraction. The LL-37 peptide affected the strength of interactions between phospholipids' molecules and reduced the monolayers stability. Accordingly, the changes in interactions in the model membranes allowed us to determine the difference in their susceptibility to the LL-37 peptide depending on the choline supplementation of bacterial culture.

Thermodynamics of Wetting, Prewetting and Surface Phase Transitions with Surface Binding

Abstract:In living cells, protein-rich condensates can wet the cell membrane and surfaces of membrane-bound organelles. Interestingly, many phase-separating proteins also bind to membranes leading to a molecular layer of bound molecules. Here we investigate how binding to membranes affects wetting, prewetting and surface phase transitions. We derive a thermodynamic theory for a three-dimensional bulk in the presence of a two-dimensional, flat membrane. At phase coexistence, we find that membrane binding facilitates complete wetting and thus lowers the wetting angle. Moreover, below the saturation concentration, binding facilitates the formation of a thick layer at the membrane and thereby shifts the prewetting phase transition far below the saturation concentration. The distinction between bound and unbound molecules near the surface leads to a large variety of surface states {and complex surface phase diagrams with a rich topology of phase transitions. Our work suggests that surface phase transitions combined with molecular binding represent a versatile mechanism to control the formation of protein-rich domains at intra-cellular surfaces.

Abstract:A Reversed Duckweed polymeric Langmuir-Blodgett (LB) film was characterized by Fourier transform infrared (FTIR) spectroscopy. It has a well-ordered structure which is similar to that of the monomer-octadecylacrylamide-LB film, which provides proof that the flexible hydrophilic network does not interfere the side-chain alignment. The phase transition behavior was studied by using temperature dependent FTIR and small-angle X-ray diffraction. The LB film shows a unique thermal transition behavior, which undergoes a three-phase transition processes: from a highly ordered structure transition into a melted disordered state through two Langmuir-Blodgett "liquid crystal" states.
