Abstract:Rapid and strong adhesion of hydrogel adhesives is required for instant wound closure and hemostasis. However, in situ hydrogel formation and sufficient adhesion at target tissue sites in biological environments are severely compromised by the presence of blood and body fluids. In this work, an underwater adhesive hydrogel (named SHCa) is fabricated with rapid in situ gelation, enhanced mechanical toughness, and robust underwater adhesion. The SHCa can undergo rapid UV irradiation-induced gelation under water within 5 s and adhere firmly to underwater surfaces for 6 months. The synergistic effects of crystalline β-sheet structures and dynamic energy-dissipating mechanisms enhance the mechanical toughness and cohesion, supporting the balance between adhesion and cohesion in wet environments. Importantly, the SHCa can achieve rapid in situ gelation and robust underwater adhesion at various tissue surfaces in highly dynamic fluid environments, substantially outperforming the commercially available tissue adhesives. The lap shear adhesion strength and wound closure strength of SHCa on blood-covered substrates are 7.24 and 12.68 times higher than those of cyanoacrylate glue, respectively. Its fast hemostasis and wound sealing performance are further demonstrated in in vivo animal models. The proposed hydrogel with strong underwater adhesion provides an effective tool for fast wound closure and hemostasis.

Abstract:Mussel-inspired catechol-based strategy has been widely used in the development of underwater adhesives. Nonetheless, the properties of the adhesives were still severely limited under harsh environments. A facile approach was proposed herein to prepare a double network hydrogel adhesive with low swelling rate and high strength in seawater, where the first network was polyacrylamide (PAM) and the second network was alginate (Alg). Meanwhile, polydopamine (PDA) nanoparticles, which were formed through self-polymerization as adhesion anchoring sites, distributed evenly throughout the double network hydrogel and effectively enhanced the adhesion capability of the hydrogel. The properties of the resulting hydrogel have been fully characterized. The optimal adhesion strength of the hydrogel adhesive in seawater was as high as 146.84 ± 7.78 kPa. Furthermore, the hydrogel also has excellent ability to promote the growth of zooxanthellae. Our studies provide useful insights into the rational design of underwater adhesives with high performances even beyond nature.

Abstract:Hydrogels, exhibiting wide application prospects in soft robotics, tissue engineering, implantable electronics, etc., upon functioning often require to adhere to substrates especially in wet environments despite that most hydrogels are nonsticky because of their high water content and hydrophilicity. Herein, a strategy to rapidly reverse the adhesion properties of hydrogels via surface anchoring very thin adhesive coatings is reported. Inspired by mussel adhesion, poly(vinyl alcohol) hydrogel is first coated with polymerized tannic acid that chelates with Fe3+, which serves as the interface bonding layer. Subsequently, a wet adhesive poly(dopamine methacrylamide‐co‐methoxyethyl acrylate) is firmly anchored to form the very thin adhesive coating (<10 µm) that can generate high adhesion strength both in the air and underwater. The adhesive coating also endows hydrogels with high water retention capacity under warm condition (50 °C) and is able to get on‐demand functionalization on the designated area to achieve asymmetric adhesion, static and dynamic control over wettability. The strategy demonstrates potentials for broad applications from biomedicine to wearable electronics. One kind of novel underwater adhesive hydrogels with robust underwater adhesion performance is engineered by anchoring mussel‐protein‐inspired copolymer onto poly(vinyl alcohol) hydrogel surface with tannic acid/Fe3+ as bridge‐bonding layer. The adhesive hydrogels demonstrate good adhesion strengths both in dry and wet environments, cyclic adhesion performance, good water retention capacity, along with highly sensitive conductivity as artificial electronic skins.

An All-in-One Tannic Acid-Containing Hydrogel Adhesive with High Toughness, Notch Insensitivity, Self-Healability, Tailorable Topography, and Strong, Instant, and On-Demand Underwater Adhesion

Abstract:Hydrogels that are mechanically tough and capable of strong underwater adhesion can lead to a paradigm shift in the design of adhesives for a variety of biomedical applications. We hereby innovatively develop a facile but efficient strategy to prepare hydrogel adhesives with strong and instant underwater adhesion, on-demand detachment, high toughness, notch-insensitivity, self-healability, low swelling index, and tailorable surface topography. Specifically, a polymerization lyophilization conjugation fabrication method was proposed to introduce tannic acid (TA) into the covalent network consisting of polyethylene glycol diacrylate (PEGDA) of substantially high molecular weight. The presence of TA facilitated wet adhesion to various substrates by forming collectively strong noncovalent bonds and offering hydrophobicity to allow water repellence and also provided a reversible cross-link within the binary network to improve the mechanical performance of the gels. The long-chain PEGDA enhanced the efficacy and stability of TA conjugation and contributed to gel mechanics and adhesion by allowing chain diffusion and entanglement formation. Moreover, PEGDA/TA hydrogels were demonstrated to be biocompatible and capable of accelerating wound healing in a skin wound animal model as compared to commercial tissue adhesives and can be applied for the treatment of both epidermal and intracorporeal wounds. Our study provides new, critical insight into the design principle of all-in-one hydrogels with outstanding mechanical and adhesive properties and can potentially enhance the efficacy of hydrogel adhesives for wound healing.

Abstract:The non-invasive tissue adhesives with strong tissue adhesion and good biocompatibility are ideal for replacing traditional wound treatment methods such as sutures and needles. The self-healing hydrogels based on dynamic reversible crosslinking can recover their structure and function after damage, which is suitable for the application scenario of tissue adhesives. Herein, inspired by mussel adhesive proteins, we propose a facile strategy to achieve an injectable hydrogel (DACS hydrogel) by grafting dopamine (DOPA) onto hyaluronic acid (HA) and mixing it with carboxymethyl chitosan (CMCS) solution. The gelation time and rheological and swelling properties of the hydrogel can be controlled conveniently by adjusting the substitution degree of the catechol group and the concentration of raw materials. More importantly, the hydrogel exhibited rapid and highly efficient self-healing ability and excellent biodegradation and biocompatibility in vitro. Meanwhile, the hydrogel exhibited ~4-fold enhanced wet tissue adhesion strength (21.41 kPa) over the commercial fibrin glue. This kind of HA-based mussel biomimetic self-healing hydrogel is expected to be used as a multifunctional tissue adhesive material.

Abstract:The development of new tissue adhesives is still in great demand as the current available tissue adhesives cannot meet the requirements of sophisticated surgeries. One major limitation of current adhesives is difficult to balance the adhesive strength, the mechanical property and the responsive degradation rate. Herein, we report a new injectable and self-healable hydrogel, which can function as a multi-responsive tissue adhesive with on-demand degradation profile under different external stimuli. The adhesive hydrogel is composed of aldehyde functionalized hyaluronic acid (HA-CHO) and a disulfide containing crosslinker - 3,3'-dithiobis(propionic hydrazide) (DTPH). With the optimized formulation, the gelation can be formed within 1 min in situ. Multi responsive properties were introduced into this adhesive hydrogel, namely (1) Strain-responsive; (2) Reduction-responsive; (3) Oxidation-responsive; (4) Enzyme-responsive; and (5) UV-responsive properties. Bovine serum albumin (BSA)/Glutaraldehyde hydrogel - a commercial tissue adhesive BioGlue (R), was selected as the control. Abstract:Current medical adhesives based on cyanoacrylates typically exhibit cellular toxicity. In contrast, fibrin adhesives are non-toxic but have poor adhesive properties. To overcome these drawbacks we designed a simple and scalable adhesive precursor inspired by marine mussel adhesion that functioned with strong adhesion in wet conditions and with low cytotoxicity. Dopamine, an-amine derivative of an amino acid abundantly present in mussel adhesive proteins, was co-polymerised with a tri-functional vinyl monomer, to form a hyperbranched poly(β-amino ester) polymer termed poly(dopamine-co-acrylate) (PDA). A variety of molecular weights and crosslinking methods were analysed using an ex vivo porcine skin model and an almost 4 fold increase in wet adhesion strength was observed compared to TISSEEL® fibrin sealant. With a fast curing time, degradable properties and low cytotoxicity, PDA is highly attractive for medical purposes and could have a broad impact on surgeries where surgical tissue adhesives, sealants, and haemostatic agents are used.

Abstract:Bioinspired green underwater adhesive hydrogel is prepared by a stepwise immersion method with gelatin hydrogel in tannic acid solution without any polymerization. Underwater adhesion is realized by treating the hydrogel with urea solutions or heating with the adhesion strength adjustable at a wide range of 100–103 kPa. The hydrogel shows multiple environmental adaptability in different liquids and on different substrates. Underwater adhesion of hydrogels is essential for their applications in liquid environment. However, green raw materials, facile preparation process and multiple environmental adaptability are still issues that need to be improved. Inspired by the protein‐based underwater bio‐adhesion phenomena in nature, an effective stepwise immersion method is developed without any polymerization to prepare a green underwater adhesive hydrogel by combining gelatin and tannic acid (TA), two common materials from nature yet difficult to obtain their homogenous compound. By facile treatments in urea solution or heating, the hydrogels can realize stable immediate adhesion on various substrates and in different liquids in addition to water with a wide range of adjustable adhesion strength of 100–103 kPa. The evolution of the hydrogel network structure and the adhesion mechanism are systematically studied. Furthermore, several typical portable products based on the adhesive hydrogels are demonstrated to show the commercialization possibility. This work presented a novel, facile, and distinctive strategy for designing environmentally friendly, multiple adaptive adhesives.

Abstract:Hydrogels with fascinating adhesion have been demonstrated great potential in various applications. However, most hydrogels lose their adhesion in wet or underwater environments due to the influence of interfacial water. Inspired by mussel, an underwater adhesive hydrogel was facilely fabricated by introducing electrostatic interactions, which consisted of poly (acrylic acid) (PAA), quaternized xylan (QAX) and tannic acid (TA). In this hydrogel, -COO − from PAA, -N + (CH 3 ) 3 from QAX and catechol group from TA resembled amino acids with negative and positive charges and 3,4-dihydroxyphenylalanine units in mussel, which endowed the hydrogels with great underwater adhesion through multiple interactions. Notably, acrylic acid (AA) played a key role in the dispersion of the system. QAX, a biomass derived from plants with excellent properties, worked with PAA to construct hydrogel networks. The resultant hydrogels exhibited excellent mechanical properties including remarkable stretchability (>4000 %) and compressibility. Moreover, the hydrogels had superior UV-blocking (~99.96 %), and showed good adhesion both in air and underwater. The hydrogels can be exploited as a wearable sensor to monitor human motions and even subtle motions, which have the potential to be explored in human health monitoring.

Abstract:To date, the robust and durable adhesion capability of hydrogel adhesives in wet environments remains a huge challenge. Herein, a physicochemically double-network crosslinked hydrogel matrix was prepared by mixing acrylic acid (AAc), chitosan (CS) and tannic acid (TA) as the main components and the subsequent in situ polymerization of AAc. The abundant reactive sites on the surface of the hydrogel matrix facilitate rapid, strong and repeatable adhesion to different surfaces of engineering solids and biological tissues in an aquatic environment. The formation of amide covalent bonds resulting from the addition of the bridging agent further expands the long-term application of the hydrogel in tissue repair, and the constructed hydrogel-tissue adhesive interface still has robust adhesion energy after soaking in a physiological environment for up to one month. Moreover, the hydrogel showed fantastic hemostatic performance due to its characteristics of platelet adhesion and high burst pressure. Overall, the persistent adhesion and excellent cytocompatibility of the hydrogel adhesive make it potentially applicable in medical adhesives.

Abstract:In this study, high performance hydrogels with strong underwater adhesion were constructed through a “mussel feet-rock” inspired strategy, in which a random copolymer containing side-chain positive charges and catechol groups (PMD) simulated the adhesive proteins of mussel, and poly(acrylic acid) (PAA) mimicked the rocky surface. PMD and PAA chains could noncovalently cross-link together through hydrogen bonding, charge interaction, and chain entanglement. Catechol groups further provide the hydrogel strong adhesion ability. Taking advantage of this strategy, the obtained PAA/PMD hydrogels featured the best stretchability among reported hydrogels and intensive repeatable adhesion behavior, and further exhibited unusual wet-enhanced tensile and shear strength together with durable underwater adhesion. These findings are valuable for the development of mussel-inspired adhesives and high performance hydrogels.

Abstract:An ideal hydrogel for biomedical engineering should mimic the intrinsic properties of natural tissue, especially high toughness and self-healing ability, in order to withstand cyclic loading and repair skin and muscle damage. In addition, excellent cell affinity and tissue adhesiveness enable integration with the surrounding tissue after implantation. Inspired by the natural mussel adhesive mechanism, we designed a polydopamine–polyacrylamide (PDA–PAM) single network hydrogel by preventing the overoxidation of dopamine to maintain enough free catechol groups in the hydrogel. Therefore, the hydrogel possesses super stretchability, high toughness, stimuli-free self-healing ability, cell affinity and tissue adhesiveness. More remarkably, the current hydrogel can repeatedly be adhered on/stripped from a variety of surfaces for many cycles without loss of adhesion strength. Furthermore, the hydrogel can serve as an excellent platform to host various nano-building blocks, in which multiple functionalities are integrated to achieve versatile potential applications, such as magnetic and electrical therapies.

Abstract:<p>Long-lasting and reusable adhesive hydrogels are highly desirable in biomedical and relevant applications, however, its design still remains challenge. Here, a series of plant-inspired adhesive hydrogels were prepared based on Ag/Tannic acid-Cellulose nanofibers (Ag/TA-CNF) triggered reversible quinone/catechol chemistry, which mimicked the long-lasting reductive/oxidative balance in mussels. The dynamic redox system generated catechol groups inner the hydrogel continuously, imparting hydrogels with high and repeatable adhesiveness. Besides, the hydrogel still maintained its high adhesiveness after storing at extreme temperatures for 30 days. Furthermore, to broaden the biomedical applications of the hydrogels, the pre-gel solution with optimal composition was cast onto the surface of vaccarin-loaded electrospun nanofibers to form the bilayer nanocomposite hydrogel () <em>in situ</em>. The with the intrinsic properties of the hydrogel layer (e.g. stretchable, adhesive, antioxidant, antifreezing, antidrying, photothermal and antibacterial) exhibited enhanced mechanical properties, sustained drug release and good cytocompatibility, which could be an attractive candidate for wound healing material. Taken together, this study may inspire new aspects for designing reusable and long-lasting adhesive hydrogels according to dynamic catechol chemistry.</p>

Abstract:Inspired by mussel foot proteins, catechol derivatives with a long aliphatic side chain (≈10 atoms in length) are synthesized to prepare tough wet/underwater tissue adhesives. The adhesive with excellent wet tissue adhesion and on‐demand detachment has potential applications in emergency healthcare and the field of sensors and wearable devices. Wet adhesion is critical in cases of wound closure, but it is usually deterred by the hydration layer on tissues. Inspired by dopamine‐mediated underwater adhesion in mussel foot proteins, wet tissue adhesives containing catechol with 2–3 carbons side chains are reported mostly. To make wet adhesion of this type of adhesives much tougher, catechol derivatives with a long aliphatic side chain (≈10 atoms length) are synthesized. Then, a series of strong wet tissue adhesive hydrogels are prepared through photoinduced copolymerization of acrylic acid with synthetic monomers. The adhesive hydrogel has a high cohesion strength, that is, tensile strength and strain, and toughness of ≈1800 kPa, ≈540%, and ≈4100 kJ m−3, respectively. Its interfacial toughness on wet and underwater porcine skin is respectively ≈1300 and ≈1100 J m−2, and its adhesion strength to wet porcine skin is ≈153 kPa. These values are much higher than those of dopamine‐based adhesives in the same conditions, demonstrating that the long aliphatic side chain on catechol can greatly improve the wet tissue‐adhesion. Additionally, the tough interfacial adhesion can be broken on demand with 5 wt.% aqueous urea solution. This adhesive hydrogel is highly promising in safe wound closure.

Abstract:Adhesion to many kinds of surfaces, including biological tissues, is important in many fields but has been proved to be extremely challenging. Furthermore, peeling from strong adhesion is needed in many conditions, but is sometimes painful. Herein, a mussel inspired hydrogel is developed to achieve both strong adhesion and trigger-detachment. The former is actualized by electrostatic interactions, covalent bonds, and physical interpenetration, while the latter is triggered, on-demand, through combining a thixotropic supramolecular network and polymer double network. The results of the experiments show that the hydrogel can adhere to various material surfaces and tissues. Moreover, triggered by shear force, non-covalent interactions of the supramolecular network are destroyed. This adhesion can be peeled easily. The possible mechanism involved is discussed and proved. This work will bring new insight into electronic engineering and tissue repair like skin care for premature infants and burn victims.
An Instant Underwater Tissue Adhesive Composed of Catechin-Chondroitin Sulfate and Cholesterol-Polyethyleneimine

Abstract:Due to the safety issue and poor underwater adhesion of current commercially available bioadhesives, they are hard to apply to in vivo physiological environments and more diverse medical use conditions. In this study, a novel and facile bioadhesive for underwater medical applications are designed based on the coacervation of electrostatic interactions and hydrophobic interactions, with the introduction of catechin as a provider of catechol moieties for adhesion to surrounding tissues. The orange-colored bio-adhesive, named PcC, is generated within seconds by mixing catechin-modified chondroitin sulfate and cholesterol chloroformate-modified polyethyleneimine with agitation. In vitro mechanical measurements prove that this novel PcC bio-adhesive is superior in underwater adhesion performance when applied to cartilage. Animal experiments in a rat mastectomy model and rat cartilage graft implantation model demonstrate its potential for diverse medical purposes, such as closing surgical incisions, reducing the formation of seroma, and tissue adhesive applied in orthopedic or cartilage surgery.
An Integrated Asymmetric Wet Adhesive Hydrogel with Gradient Charge Distribution Induced by Electrostatic Field

Abstract:The asymmetric wet adhesive hydrogel with gradient charge distribution is prepared by using a new process of hydrogels prepared by electrostatic field. Catechol‐cation enrichment provides strong underwater adhesion at the bottom of the hydrogel, while the other side can effectively prevent unnecessary adhesion after surgery. Adhesive hydrogels have been applied in biomedical field as an alternative to surgical sutures. However, there still exist rigorous challenges in rough underwater adhesion and asymmetric adhesion of hydrogels, especially applied in wound healing and organ repair in vivo. Herein, a strategy is proposed to prepare integrated hydrogels with asymmetric underwater adhesion capability by forming the asymmetric electrical interface under an electrostatic field. The synergistic effects between catechol and amine and complex coacervation are used to improve the underwater adhesion of hydrogel. Furthermore, by applying an electrostatic field, the cations and anions in solution of monomers are separated to form asymmetric adhesion interfaces. The hydrogel exhibits obvious asymmetric underwater adhesion ability on porcine skin with a strong adhesive strength of 97 kPa on cation side and 25 kPa on the anion side. Animal experiment outcomes reveal that only one side of the asymmetric hydrogel could adhere firmly to the rat liver and the rabbit stomach, while the other side could not effectively prevent postoperative tissue adhesion. The asymmetric distribution of adhesive molecules induced by electrostatic fields will provide a new alternative for designing and adjusting asymmetric adhesives after surgery.

Unsound interpretation of findings on a measurement bias in antidepressant drug research

Abstract:Isacsson and Adler (1) noted that the Hamilton Depression Rating Scale (HDRS) has limited reliability in assessing low levels of depression. The authors draw two conclusions on this assumption: (i) The precision of ratings decreases as the patient improves and (ii) improvements starting at lower levels of depression will be systematically underestimated compared with improvements starting at higher levels of depression severity. It is not a novel fact that the HDRS is an imperfect measure of depression severity. As the HDRS is considered to be the gold standard for the assessment of depression severity, it is, nevertheless, important to underline the deficits in its reliability; thus, I agree with the authors call for better instruments to assess the severity of depression. Where I got lost, however, is how these methodological restrictions of the HDRS lead to the authors conclusion that randomized controlled clinical trials (RCTs) underestimate the efficacy of antidepressants in less severe depression? The results only highlight that the reliability decreases with decreasing levels of depression severity. This could mean both that RCTs underestimate or overestimate the efficacy of antidepressant drugs in less severe depression. Similarly, the fact that improvements starting at lower levels of depression will be less reliable than improvements starting at higher levels simply indicates that we do have a measurement problem in RCTs using the HDRS to assess subthreshold to mild depression [NICE nomenclature of HDRS cut-off scores (2)] respectively mild to moderate depression [American Psychiatric Association (APA) nomenclature (2)]. As both patient groups (verum and placebo) have low levels of depression at baseline and both patient groups might improve over time, the measurement bias tells us nothing about whether the effects of antidepressant drugs are underestimated or overestimated or whether trials using the HDRS for the assessment of lower levels of depression are just faced with a large variance of results because of the measurement bias. Thus, the only finding of the article by Isacsson and Adler (1) is that the HDRS is a rather imperfect gold standard for the assessment of depression severity. How comes that the authors conclude that the efficacy of antidepressants is underestimated? There are three possible explanations I can think of: (i) A lack of statistical competency leading to the misinterpretation of results. However, this seems rather unlikely, as the article is on a high methodological level; (ii) the authors believe in the efficacy of antidepressant drugs and therefore tend to interpret results in favor of their beliefs. Beliefs prior to a study are known to have an impact on posterior interpretations of the findings (3); (iii) M. Adler stated that he has received...fees for speaking from Eli Lilly, Bristol Meyers Squibb, Sanofi-Aventis and for speaking and preparing manuscripts from AstraZeneca, which, too, could explain the unsound interpretation of results in favor of antidepressant drugs. To date, there is no evidence that antidepressant drugs are significantly superior to placebo in patients with subthreshold to mild depression (= APA mild to moderate depression) (4, 5). Consequently, antidepressant drugs should not routinely be used as first-line treatment for (persistent) subthreshold depressive symptoms or mild depression because of the poor risk–benefit ratio (4, 5). Forthcoming RCTs on antidepressant drug treatments based on psychometrically better assessment instruments might change this recommendation, while the study of Isacsson and Adler (1) is inappropriate to do so, despite its full-bodied title.
Mussel Foot Protein-Inspired Adhesive Tapes with Tunable Underwater Adhesion

Abstract:Instant and strong adhesion to underwater adherends is a big challenge due to the continuous interference of water. Mussel foot protein-bioinspired catechol-based adhesives have garnered great interest in addressing this issue. Herein, a novel self-made catecholic compound with a long aliphatic chain was utilized to prepare thin (∼0.07 mm) and optically transparent (>80%) wet/underwater adhesive tapes by UV-initiated polymerization. Its adhesion activity was water-triggered, fast (<1 min), and strong (adhesion strength to porcine skin: ∼1.99 MPa; interfacial toughness: ∼610 J/m2, burst pressure: ∼1950 mmHg). The effect of the catechol/phenol group and positively charged moiety on the wet/underwater adhesion to abiotic/biotic substrates was investigated. On the wet/underwater adherends, the tape with catechol groups presented much higher interfacial toughness, adhesion strength, and burst pressure than the analogous tape with phenol groups. The tape with both the catechol group and cationic polyelectrolyte chitosan had a more impressive improvement in its adhesion to wet/underwater biological tissues than to abiotic substrates. Therefore, catechol and a positive moiety in the tape would synergistically enhance its wet/underwater adhesion to various substrates, especially to biological tissues. The instant, strong, and noncytotoxic tape may provide applications in underwater adhesion for sealing and wound closure.
