Abstract:NaClO, as a common membrane cleaning agent were introduced from the permeate side across membrane, which could diffuse into MBR membrane pool. Microplastics would also accumulate in MBR membrane pool during wastewater treatment, thus, NaClO and MPs are co-exposed to activated sludge. This study investigated the effects of PET-MPs (0, 50, 100 and 300 mu g/g-ss) on activated sludge with or without NaClO stress. The presence of PET-MPs had no significant impact on nitrification and denitrification rates, but when exposed to NaClO, the increasing level of PET-MPs (i.e., from 0 to 300 mu g/g-ss) inhibited the nitrification (94.85 % to 65.90 %) and denitrification (88.68 % to 60.09 %) rates. For microbial metabolism (key enzymes) evaluated, the key enzymes related to organic matter biodegradation, nitrification as well as denitrification were both inhibited with PET- MPs level rise under NaClO stress. The generation of reactive oxygen species (ROS) and the release of lactate dehydrogenase (LDH) increased significantly indicating NaClO caused the toxicity of MPs to activated sludge by enhancing the oxidative stress level. Correspondingly, a variation in abundance related to membrane pollution, nitrification, and denitrification after exposure to PET-MPs with NaClO stress accompanied these adverse effects.
Photochlorination-induced degradation of microplastics and interaction with Cr(VI) and amlodipine

Abstract:Wastewater treatment plants (WWTPs) are the important source of microplastics (MPs) in the environment, and disinfection processes bear high potential to degrade MPs. This study investigated the physicochemical degradation, dissolved organic products and interaction with co-existed pollutants (heavy metal and pharmaceutical) on polyethylene (PE), polypropylene (PP) and polystyrene (PS) MPs during simulated disinfection processes. Compared to photo or chlorination, photochlorination significantly resulted in the physicochemical degradation, including morphology alteration, fragmentation, and chemical oxidation on PP and PS MPs, but showed relatively low effect on PE, indicating the different resistance among polymers to disinfected treatment. Photochlorination also caused the formation of chain-scission organic compounds and even chlorinated products from MPs (e.g. C11H19O4Cl for PP and monochlorophenol, dichlorophenol, chloroacetophenone and chlorobenzoic acid for PS), which may form disinfection byproducts to induce healthy risk. The adsorption potentials of MPs for Cr(VI) or amlodipine were enhanced by photochlorination since the cracking and formed oxygen functional groups enhanced the pore filling and surface precipitation of Cr(VI), and the electrostatic attraction and hydrogen bonding with amlodipine. The findings indicated the physicochemical degradation of MPs and the combined pollution with co-existed pollutants, highlighting the health risks of MP-derived organic products during the disinfection treatments (even in normal dosage) in WWTPs.

Abstract:Microplastics (MPs) in drinking water are predominantly < 10 µm. The leaching of MPs derived dissolved organic matters (MP-DOM) from 5 µm polypropylene MPs (PP-MPs) and polystyrene MPs (PS-MPs) and the formation of MP-DOM derived disinfection byproducts during chlorination disinfection were first investigated. Comparably, PS-MPs are more vulnerable to chlorination and the primary attacks are on para C in aromatic side-chains via electrophilic Cl-substitution and oxidation by two-electron transfer. The O/C and Cl/C ratio of polystyrene MPs was linear and exponential versus initial available Cl2 concentrations, respectively. The significant PS-DOM leaching was observed with initial available Cl2 of 4.0 mg/L (USEPA recommended upper dose). As the initial available Cl2 concentration increased to 8.0 mg/L, the adsorption of chloro-phenolic-components of 200 Daltons in PS-DOM by 5 µm PS-MPs was observed for the first time. Trichloromethane (TCM) was identified as the dominant disinfection byproduct with a formation potential of 60.3 ± 7.8 and 73.7 ± 9.8 μg/mg for PS-DOM and PP-DOM, respectively. The derived TCM could adsorb onto PS-MPs followed the pseudo-second-order kinetic and Langmuir isotherm models. Extreme chlorination could reduce the maximal adsorption capacity of TCM on 5 µm PS-MPs from 196.68 ± 48.66 to 146.02 ± 32.98 μg/g. Thus, PS-MPs act as precursors and carriers of TCM in chlorination.
Effect of microplastics on sodium hypochlorite disinfection and changes in its toxicity on zebrafish

Abstract:The presence of microplastics (MPs) in water may affect the efficacy of the disinfection process and induce toxicity changes to MPs themselves during disinfection. Therefore, this study evaluated the two-way effects of polyethylene microplastic (MP) particles in water and wastewater during sodium hypochlorite (NaClO) disinfection. On the one hand, it has been confirmed that the presence of MPs reduced the disinfection efficiency of NaClO. The required CT (concentration of the disinfection × contact time) for a 2-4-log inactivation of Escherichia coli (E. coli) in different water samples was in the order of deionized water < turbid water (1 NTU) < water with MPs (1 mg/L) < turbid water (10 NTU). On the other hand, although exposure to MPs did induce significant changes in the activities of superoxide dismutase and glutathione, compared to pristine MPs, the MPs treated by NaClO at current conditions (0.3 and 3.0 mg/L for 30 min) did not show significant changes in their toxicity on zebrafish, at an MP exposure concentration of 1 mg/L. There was no significant difference in the survival rate and weight growth rate, neither as in the activities of the oxidative stress-related enzymes (superoxide dismutase, catalase, glutathione, glutathione peroxidase, and glutathione s-transferase) in both gut and muscle tissues of the zebrafish, between exposure to the pristine and NaClO-treated MPs. It is indicated that NaClO disinfection commonly applied for water and wastewater treatment would not pose a serious concern to effluent safety in the presence of mild levels of MPs.
Comparison of chlorination resistance of biodegradable microplastics and conventional microplastics during the disinfection process in water treatments

Abstract:Nowadays, microplastics (MPs) widely exist in the environment, and water treatment plants are important sources of MPs. Chlorine is widely used in the disinfection process in water treatment plants and has strong oxidation, however, the chemical and physical properties changes of MPs during chlorination were unclear. Thus, in this study, based on the actual used chlorine concentrations, different chlorination conditions were simulated to study the variation of MPs after chlorination. Meanwhile, the produced disinfection by-products were monitored. The results showed that under high chlorination concentration conditions, functional groups of polyethylene (PE), polystyrene (PS), and polylactic acid (PLA) changed, while no peak shift or change of poly (butyleneadipate-co-terephthalate) (PBAT) could be detected. Moreover, after chlorination, partial yellowing and cracks appeared on PS, PLA, and PBAT, while PE remained white and showed little morphological changes. Besides, chlorination led to the narrowing of the cold crystallization peak and melting peak of PLA, while chlorination had little influence on the crystal structure of PE and PBAT. Furthermore, the reaction between PLA and chlorine mostly produced more trichloromethane than other types of MPs. Consequently, when chlorine concentrations were in the range of 2.5 to 5000 mg/L, the chlorination resistance was PBAT/PE > PLA > PS. Specifically, PBAT had the strongest chlorination resistance in terms of chemical properties, while PE had the strongest chlorination resistance in terms of physical properties. Therefore, the degradability of biodegradable MPs is not higher than that of conventional MPs in all cases. Moreover, it should be noted that most changes occurred only in high chlorine concentrations. Thus, neither conventional MPs nor biodegradable MPs can be completely degraded during the chlorination process in water treatments.

Abstract:Limited studies to-date have assessed the impacts of disinfectants (chlorine and ultraviolet (UV)/H2O2) on microplastics (MPs), with most employing dosages and fluences that are much higher than typically applied during drinking water treatment. Formation of hydroxyl, carbonyl, and carbon-chlorine bonds on the surface of MP particles, as well as the potential for fragmentation and changes to surface roughness have been linked to an increase in toxicity. Additional knowledge regarding physicochemical changes to MPs under conditions representative of drinking water disinfection will aid our understanding of potential risk. In this study, changes to virgin low-density polyethylene (LDPE) and high-density polyethylene (HDPE) fragments (2 - 125 μm) were examined following exposure to 6 mg/L free chlorine over a two-week period. Relative changes to hydroxyl, carbonyl, carbon-oxygen, and carbon-chlorine indices were determined using Attenuated Total Reflectance Fourier Transform Spectroscopy (ATR-FTIR). Following exposure, particle size distributions remained consistent, indicating that MPs did not fragment as reported in earlier studies by others. Neither polymer type nor pH level exhibited an impact on fragmentation or bond indices that was statistically significant (P>0.05). These findings suggest that neither chlorination nor UV/H2O2 adversely impact LDPE or HDPE.

Abstract:Considering the large volumes of treated water and incomplete elimination of pollutants, wastewater treatment plants (WWTPs) remain a considerable source of microplastics (MPs). Chlorine, the most frequently used disinfectant in WWTPs, has a strong oxidizing impact on MPs. However, little is documented, to date, about the impact of chlorination on the transformation of MPs and the subsequent environmental behaviors of the chlorinated MPs when released into the aquatic environment. This study explored the response of the physicochemical properties of specific thermoplastics, namely polyurethane (TPU) MPs and polystyrene (PS) MPs, to chlorination and their emerging pollutant [tetracycline (TC)] adsorption behavior in aqueous solution. The results indicated that the O/C ratio of the MP surface did not significantly change, and that there were increases in the O-containing functional groups of the TPU and PS MPs, after chlorination. The surface area of the chlorinated TPU MPs increased by 45 %, and that of the chlorinated PS increased by 21 %, compared with the pristine ones, which contributed to the TC adsorption. The adsorption isotherm fitting parameters suggested that the chlorinated TPU fitted the multilayer adsorption, and the chlorinated PS was inclined to the monolayer adsorption. The relative abundance of the O-containing functional groups, on the TPU surface, led to the release of CHCl3 molecules, and the clear surface irregularities and fissures occurred after chlorine treatment. No fissures were found on the surface of the chlorinated PS MPs. The hydrophobicity and electrostatic adsorption were proved to be the major impacts on the TC adsorption of the chlorinated MPs, and the subsequently formed hydrogen bonds led to the stronger adsorption capacity of the chlorinated TPU than the chlorinated PS MPs.

Abstract:Given the widespread presence of Pseudomonas aeruginosa in water and its threat to human health, the metabolic changes in Pseudomonas aeruginosa when exposed to polystyrene microplastics (PS-MPs) exposure were studied, focusing on molecular level. Through non-targeted metabolomics, a total of 64 differential metabolites were screened out under positive ion mode and 44 under negative ion mode. The content of bacterial metabolites changed significantly, primarily involving lipids, nucleotides, amino acids, and organic acids. Heightened intracellular oxidative damage led to a decrease in lipid molecules and nucleotide-related metabolites. The down-regulation of amino acid metabolites, such as L-Glutamic and L-Proline, highlighted disruptions in cellular energy metabolism and the impaired ability to synthesize proteins as a defense against oxidation. The impact of PS-MPs on organic acid metabolism was evident in the inhibition of pyruvate and citrate, thereby disrupting the cells' normal participation in energy cycles. The integration of Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis revealed that PS-MPs mainly caused changes in metabolic pathways, including ABC transporters, Aminoacyl-tRNA biosynthesis, Purine metabolism, Glycerophospholipid metabolism and TCA cycle in Pseudomonas aeruginosa . Most of the differential metabolites enriched in these pathways were down-regulated, demonstrating that PS-MPs hindered the expression of metabolic pathways, ultimately impairing the ability of cells to synthesize proteins, DNA, and RNA. This disruption affected cell proliferation and information transduction, thus hampering energy circulation and inhibiting cell growth. Findings of this study supplemented the toxic effects of microplastics and the defense mechanisms of microorganisms, in turn safeguarding drinking water safety and human health.

Abstract:Microplastics (MPs) are persistent environmental contaminants. The toxic effects of MPs on aquatic organisms have raised increasing concerns, but their toxic effects on aquatic phytoplankton has not been thoroughly investigated. In the present study, the toxic effects of two sizes MPs (1 μm and 5 μm) on Chlorella pyrenoidosa at 2, 10, 50 mg/L were explored for 1, 5, 10 days. The growth ratio, photosynthetic pigments content, extracellular polymeric substances content, soluble protein content, MDA content and relative expression of genes related to photosynthesis and energy metabolism were measured. These results indicated that 1 μm MP could significantly inhibit the growth of C. pyrenoidosa. Compared with the control group, 1 μm MP significantly reduced the photosynthetic pigment content, induced oxidative stress and disrupted the cell membrane integrity of C. pyrenoidosa. At the molecular level, 1 μm MP altered the transcript levels of genes related to photosynthesis and energy metabolism. Scanning electron microscopy and fluorescent images showed that MPs aggregation with C. pyrenoidosa may be the main reason for the toxic effects of MPs. These results will provide new insight into the toxicity of different MPs on aquatic phytoplankton, and evaluate the risks caused by MPs in aquatic environments.

Abstract:Chlorination on microplastic (MP) biofilms was comprehensively investigated with respect to disinfection efficiency, morphology, and core microbiome. The experiments were performed under various conditions: i) MP particles; polypropylene (PP) and polystyrene (PS), ii) MP biofilms; Escherichia coli for single-species and river water microorganisms for multiple-species, iii) different chlorine concentrations, and iv) different chlorine exposure periods. As a result, chlorination effectively inactivated the MP biofilm microorganisms. The disinfection efficiency increased with increasing the free chlorination concentration and exposure periods for both single- and multiple-species MP biofilms. The multiple-species MP biofilms were inactivated 1.3-6.0 times less than single-species MP biofilms. In addition, the PP-MP biofilms were more vulnerable to chlorination than the PS-MP biofilms. Morphology analysis verified that chlorination detached most MP biofilms, while a small part still remained. Interestingly, chlorination strongly changed the biofilm microbiome on MPs; the relative abundance of some microbes increased after the chlorination, suggesting they could be regarded as chlorine-resistant bacteria. Some potential pathogens were also remained on the MP particles after the chlorination. Notably, chlorination was effective in inactivating the MP biofilms. Further research should be performed to evaluate the impacts of residual MP biofilms on the environment.

Abstract:Widely distributed nanoplastics have become a threat to water environment. Information regarding their conversion behaviors and risks under oxidation conditions is still lacking. In this study, conversion of polystyrene nanoplastics (PSNPs) in water induced by electrical plasma was investigated, and its risks were evaluated in terms of Scenedesmus obliquus growth, Human alveolar cell A549 toxicity, and chlorinated disinfection byproducts (DBPs) formation. Attacks of 1O2 and ·OH resulted in rough surface, smaller particle size of PSNPs (particles larger than 200 nm disappeared, and it was in the range of 50–150 nm), sequence breakages in functional groups, and generation of hydroxylated and carboxylated intermediates. The plasma-treated PSNPs exhibited slightly weaker toxicity for Scenedesmus obliquus growth when PSNPs concentration was 30 mg L−1, but stronger toxicity for A549 (relative viability of A549 decreased from approximately 40% to 6.9% after 15 min treatment). The hydroxylated and carboxylated intermediates were important precursors of DBPs, and the formation potentials and toxicities of haloacetic acid DBPs increased with the plasma treatment. The relevant roadmaps among haloacetic acid DBPs and conversion intermediates were figured out. Overall, the conversion of PSNPs under oxidation exposure may pose a greater threat to DBPs formation, which deserved more attention.

Abstract:The release of dissolved organic matter (DOM) from microplastics (MPs) in the water environment under physical, chemical, and biological transformation has been widely concerned. However, the impacts of released DOM on the subsequent generation of chlorinated disinfection by-products (DBPs) are still unknown. In this study, the leaching behaviors of organic matter from four types of MPs by plasma treatment were investigated, with polystyrene (PS), polyvinyl chloride (PVC), polypropylene (PP), and polyethylene (PE) as the model MPs. The plasma treatment led to cracks, pits, and loose porous surfaces of MPs, and the C/O ratios of these four MPs significantly decreased. Simultaneously, the long-chain alkyl groups of the four MPs were weakened, and O-containing functional groups were introduced after the plasma treatment. As a result, the dissolved organic carbon (DOC) released from PP, PVC, PS, and PE after 120 min of plasma treatment were 4.35, 4.33, 5.09, and 14.0 mg L−1, respectively. The released conjugated organic compounds contained aromatic hydrocarbons and double bonds or hydroxyl groups, which were soluble microbial byproduct-like and humic acid-like substances. Trichloromethane (TCM) and dichloroacetic acid (DCAA) were the main DBPs in trihalomethanes (THMs) and haloacetic acids (HAAs). The DBPs in the four MPs filtrates were mainly HAAs, which reached 811.2, 780.2, 1234.7, and 420.7 μg L−1 in the PVC, PP, PE, and PS filtrates after 120 min of plasma treatment, respectively. Correlations among DBPs, UV–visible and fluorescence components of these four MPs were established. Overall, the released byproducts of MPs had the potential to be the precursors of DBPs, and thus their risks deserve more attention.

Abstract:The 10 μm polystyrene and polyethylene-terephthalate microplastics (MPs), prevalent in finished drink water, were employed to investigate the effect of normal dosage UVC-based advanced-oxidation-processes (UVC-AOPs) on the interaction between MPs and their derived disinfection-byproducts (DBPs) during subsequent chlorination-disinfection, in the presence of Br - , for the first time. The results indicated that UVC/H 2 O 2 caused higher leaching of microplastic-derived dissolved-organic-matter (MP-DOM), with smaller and narrower molecular-weight-distribution than UVC and UVC/peroxymonosulfate (UVC/PMS). The trihalomethanes (as dominant DBPs) molar-formation-potentials (THMs-MFPs) for MP-DOM leached in different UVC-AOPs followed the order of UVC/H 2 O 2 >UVC/PMS>UVC. The adsorption of formed THMs, especially Br-THMs, back on MPs was observed in all MPs suspensions with or without UVC-AOPs pre-treatment. The Cl-THMs adsorption by MPs is more sensitive to UVC-AOPs than Br-THMs. The adsorption experiments showed that UVC-AOPs reduce the capacity but increase the rate of THMs adsorption by MPs, suggesting the halogen and hydrogen bonding forces governed the THMs adsorption rate while hydrophobic interaction determines their adsorption capacity. The UVC-AOPs pre-treatment sharply increased the total yield of THMs via both indirectly inducing MP-DOM leaching and directly increasing the THMs-MFPs of MPs by oxidation. 21.36–41.96% of formed THMs adsorbed back on the UVC-AOPs-pretreated MPs, which might increase the toxicity of MPs.

Abstract:Using current water treatment systems, significant amounts of microplastics (MPs) are passing through and being released into the aquatic environment. However, we do not clearly know what effects disinfection processes have had on these particles. In this study, we applied inductively coupled plasma-mass spectrometry (ICP-MS) operating in time-resolved analysis (TRA) mode for quantifying changes in the chlorine (Cl) content of MPs under a variety of water treatment scenarios. Our results illustrated that time-resolved ICP-MS offers a potential method for sensitive and direct analysis of Cl content, including Cl mass and chlorine association (%Cl/C), of discrete particles in the MP suspension by the fast sequential measurements of signals from 35Cl1H2 and 12C1H. Our research, across various water treatment scenarios, also showed that polystyrene (PS) MPs exhibited greater reactivity to Cl disinfectant after being pre-disinfected with UV light and in mildly acidic to neutral pH environments. It is noteworthy that about half of the particles in MP suspension exposed to 10 mg Cl2/L, a typical Cl dose applied in water treatment, were chlorinated, and had a Cl content comparable to that of particles subjected to extreme conditions. Of even greater concern is the fact that our cell viability tests revealed that chlorinated MPs induced considerably higher rates of cell death in both human A549 and Caco-2 cells, and that the effects were Cl dose- and polymer type-dependent. Overall, this study demonstrates the potential of time-resolved ICP-MS as a valuable technique for quantifying the Cl content of MP particles, which is crucial to assessing the fate and transformation of MPs in our water supply and treatment systems.

Abstract:The presence of microplastics in surface waters poses potential hazards to both organisms and human health. Traditional water treatment processes have proven to be inadequate in effectively removing microplastics. Besides, previous studies have often utilized disinfectant dosages that surpass those commonly employed in drinking water treatment plants (DWTPs), and the biotoxicity changes induced by microplastics after disinfection remain largely unexplored. To address these knowledge gaps, our study aimed to investigate the impact of practical disinfectant dosages on the leaching of organic compounds from microplastics and the formation of disinfection byproducts (DBPs). Furthermore, the resulting acute and genetic toxicity was evaluated through bioassays. Our findings revealed an elevated release of dissolved organic matter (DOM) derived from microplastics. Additionally, both acute and genetic toxicity increased following various disinfection processes. Specifically, chlorination exhibited the highest level of toxicity in short-term disinfection, while the pretreatment of ozonation followed by residual chlorination demonstrated the highest toxicity in the long-term disinfection, which was correlated with the concentration of CHCl3 formed. These findings will advance our understanding of the potential risks associated with microplastics during various disinfection processes, with the ultimate goal of safeguarding water supplies.

Abstract:Microplastics (MPs) and heavy metals are ubiquitous pollutants in the aquatic environment. In this study, the sorption behavior of two typical MPs (PVC and PE) to bivalent zinc ions (Zn(II)) and their combined toxic effects on Chlorella vulgaris were systemically studied. The growth inhibition rate, the activities of photosynthesis and antioxidant enzymes (SOD and CAT), the cell membrane integrity and the cell apoptosis rate were employed to evaluate the toxicity. Our result showed that PVC and PE have different adsorption capacities for Zn(II), and the combined exposure to Zn(II) and MPs had distinct patterns on the inhibition of the cell growth and induction of oxidative stress. Under our experimental concentrations, PE and Zn(II) showed a synergistic effect, while PVC and Zn(II) exhibited an antagonistic effect. Finally, an action mechanism was proposed to explain the experimental phenomena. This study demonstrated that flow cytometry can be a powerful tool to study the toxic effect of MP composites, and MPs can not only allow a free ride for the water contaminants, but also remarkably alter their toxic effects on phytoplankton. These effects deserve further consideration during evaluation of ecological risks of MPs in the water environment.
Oxidative degradation and possible interactions of coexisting decabromodiphenyl ether (BDE-209) on polystyrene microplastics in UV/chlorine process

Abstract:This work was to investigate the transformation of coexisting decabromodiphenyl ether (BDE-209) on microplastics and their possible interactions in UV/chlorine process. Compared with pristine microplastics, the highly aged polystyrene (PS) showed an inhibitory effect on degradation of BDE-209. Increasing initial concentration of BDE-209 on PS inhibited degradation, while the chlorine concentration and pH did not affect the final degradation efficiency. Moreover, the presence of NO 3 − , SO 4 2− , HCO 3 − and HA in water was unfavorable for BDE-209 degradation. According to the experimental and calculation results, the contribution to the degradation of BDE-209 was ranked as direct photolysis > HO• > •Cl in the UV/ chlorine system. Chlorination products released by PS during UV/chlorination were detected. Four possible reaction pathways of BDE-209 were proposed, which mainly involved debromination, hydroxylation, chlorine substitution, cleavage of ether bond, and intramolecular elimination of HBr. It was worth noting that PS microplastics not only inhibited the degradation of BDE-209, but also affected the type and abundance of its transformation products. Meanwhile, interaction products of PS and BDE-209 were determined, which was attributed to reactions of PS-derived radicals with •Br/•C 6 Br 5 and •Cl. Results of toxicity evaluation showed that the introduction of carbon-halogen bonds, especially C-Br bond, increased the toxicity of chain scission products of PS. This work provides some new insights into transformation, interaction, and associated ecological risks of coexisting microplastics and surface adsorbed contaminants in the UV/chlorine process of drinking water treatment plants (DWTPs) and wastewater treatment plants (WWTPs).

Abstract:Microplastics (MPs) and nanoplastics (NPs) have been generally regarded as emerging pollutants and received worldwide attention in recent years. Water and food consumption are the primary pathways for human exposure to MPs/NPs, thus gastrointestinal tracts may be susceptible to their toxicity. Although the recent report has indicated the presence of MPs/NPs in multiple human organs, little is known about their gastric effects. Therefore, this study focused on the adverse effects of polystyrene microplastics (PS-MPs) on gastric epithelium in vivo and in vitro. Surface-enhanced Raman spectroscopy (SERS) revealed the distribution of PS-MPs was associated with their particle sizes, and predominantly concentrated in gastric tissues. Gastric barrier injury and mitochondrial damage were observed in rats after exposure to PS-MPs. Compared with the larger ones, polystyrene nanoplastics (PS-NPs) more significantly reduced the activity of antioxidant enzymes while enhancing the level of MDA, 8-OhdG and γ-H2AX. Meanwhile, PS-MPs caused upregulation of β-catenin/YAP through redox-dependent regulation of nucleoredoxin (NXN) and dishevelled (Dvl). These findings supported the size-dependent effects of PS-MPs on oxidative stress and DNA damage. Moreover, the redox-dependent activation of the β-catenin/YAP cascade suggested a novel toxic mechanism for PS-MPs and implied the potential carcinogenic effects.

Abstract:Microplastics have emerged as environmental hazards in recent years. This study was intended to prove the toxic effects of microplastics on the male reproductive system and further elucidate its mechanism. C57bl/6 mice were exposed to ultrapure water or different doses (0.25, 0.5 and 1 mg/d) of 5μm polystyrene microplastics (PS-MPs) for 4 weeks, and the GC-1 mouse spermatogonium was treated with different concentrations of PS-MPs. The results showed that sperm count and motility were decreased, and sperm deformity rate was increased after exposure to PS-MPs. The morphology of testes in PS-MPs groups exhibited pathological changes, such as abnormal development of spermatogenic tubules, and inhibited spermatogonium function. Furthermore, the fluorescence intensity of TUNEL staining and the BAX/BCL2 ratio were increased. Exposure to PS-MPs resulted in impaired mitochondrial morphology of spermatogonium, decreased activity of GSH-px and SOD, and increased the MDA level. In vitro, after treatment with PS-MPs, the cell apoptosis rate of spermatogonium was significantly increased, mitochondrial membrane potential was decreased, mitochondrial morphology was damaged, and exposure to PS-MPs increased mitochondrial reactive oxygen species, inducing an oxidative stress state in spermatogonia. In summary, PS-MPs induced a decrease in sperm quality by activating spermatogonium mitochondrial oxidative stress and apoptosis, offering novel insights into mitigating the reproductive toxicity of microplastics.

Abstract:Microplastics (MPs) are substances that pose a risk to both human life and the environment. Their types and production are increasing year on year, and their potential to cause environmental pollution is a worldwide concern. Conventional water treatment processes, particularly coagulation and sedimentation, are not effective at removing all MPs. It is therefore important to assess the morphological changes in the MPs, i.e., the thermoplastic polyurethane (TPU) and polyethylene (PE), during ozonation and the dissolved organic carbon leaching as well as chloroform formation in the subsequent chlorination. The results show that the appearance and surface chemistry of the MPs changed during the ozonation process, most notably for TPU. The trichloromethane (CHCl3) generation during chlorination was 0.168 and 0.152 μmol/L for TPU and PE, respectively, and the ozone pretreatment significantly increased the CHCl3 yield of TPU, while it had a weak effect on PE. Additional disinfection byproducts (DBPs), including CHCl2Br, CHClBr2, and CHBr3, were produced in the presence of bromide ions in the water column, and the total amount of DBPs produced by PE, PE-O, TPU, and TPU-O was significantly increased to 0.787, 0.814, 0.931, and 1.391 μmol/L, respectively. The study provides useful information for the environmental risk assessment of two representative MPs, i.e., TPU and MPs, in disinfection procedures for drinking water.
