Abstract:Granzyme B (GrB) is a serine protease released by natural killer cells and cytotoxic T lymphocytes during immune responses, which not only plays a role in tumor diagnosis but also provides valuable guidance during tumor treatment. In this work, we have designed a charge-switching peptide to fabricate an electrochemical biosensor for quantitative analysis of GrB. Specifically, the designed zwitterionic peptide is in an electrically neutral state before activation, and a door lock structure (proline) is constructed by utilizing the selectivity of carboxypeptidase A (CPA) to the carboxy-terminus of the peptide chain. The door lock is opened when the target is present, allowing CPA to hydrolyze the peptide. At this time, the peptide will convert from neutral to positive, triggering the assembly of a positively charged peptide layer on the electrode surface, resulting in a signal change. Studies have shown that the biosensor has good analytical performance, with a detection range of 0.01 pM–8 pM and a detection limit as low as 3.5 fM. Moreover, the developed biosensor has been effectively applied to the analysis of clinical samples, demonstrating its ability to monitor tumor progression and treatment with clinical applications.

Abstract:Temozolomide (TMZ)-resistant glioblastoma multiforme (GBM) cells would have abnormal redox status due to bio-thiols, like glutathione (GSH), which constitute the most crucial defense system that protects cells from therapeutic agents. Current strategies for GSH detection often require sophisticated instruments that may not be available in laboratories with fewer resources. Here, we circumvent this problem by introducing a lateral flow plasmonic biosensor (LFPB) based on gold-viral biomineralized nanoclusters (AuVCs) as nanozymes that enables the detection of a few molecules with the naked eye and quantified by an auto-analysis software. The GSH level controls the growth of gold nanoparticles (AuNPs) and generates coloured patterns with distinct tonality, which are then auto-analyzed to calculate the GSH concentrations by smartphone with an auto-analysis software. Under the optimized conditions, grayscale value plotted against GSH concentration exhibited a linear relationship within the range of 25–500 μM with a limit of detection (LoD) of 9.80 μM and highly positive correlation between detected GSH level and TMZ drug-resistance level in GBM cells. This excellent property allowed our approach to be used for on-site determination of GSH levels in a rapid (i.e., within 30 min), simple (i.e., auto-analysis software), and cost-effective process (i.e., instrument-free) for cancer precision therapy.

Abstract:A triple signal amplification strategy was designed for ultrasensitive immunosensing of cancer biomarker. This strategy was achieved using graphene to modify immunosensor surface for accelerating electron transfer, poly(styrene-co-acrylic acid) microbead (PSA) carried gold nanoparticles (AuNPs) as tracing tag to label signal antibody (Ab(2)) and AuNPs induced silver deposition for anodic stripping analysis. The immunosensor was constructed by covalently immobilizing capture antibody on chitosan/electrochemically reduced graphene oxide film modified glass carbon electrode. The in situ synthesis of AuNPs led to the loading of numerous AuNPs on PSA surface and convenient labeling of the tag to Ab(2). With a sandwich-type immunoreaction, the AuNPs/PSA labeled Ab(2) was captured on the surface of an immunosensor to further induce a silver deposition process. The electrochemical stripping signal of the deposited silver nanoparticles in KCl was used to monitor the immunoreaction. The triple signal amplification greatly enhanced the sensitivity for biomarker detection. The proposed method could detect carcinoembryonic antigen with a linear range of 0.5 pg mL(-1) to 0.5 ng mL(-1) and a detection limit down to 0.12 pg mL(-1). The immunosensor exhibited good stability and acceptable reproducibility and accuracy, indicating potential applications in clinical diagnostics.
Disposable Reagentless Electrochemical Immunosensor Array Based On A Biopolymer/Sol-Gel Membrane For Simultaneous Measurement Of Several Tumor Markers

Abstract:BACKGROUND: A reagentless sensor array for simultaneous multianalyte testing (SMAT) may enable accurate diagnosis and be applicable for point-of-care testing. We developed a disposable reagentless immunosensor array for simple immunoassay of panels of tumor markers.METHODS: We carried out SMAT with a direct capture format, in which colloidal gold nanoparticles with bound horseradish peroxidase (HRP)-labeled antibodies were immobilized on screen-printed carbon electrodes with biopolymer/sol-gel to trap their corresponding antigens from sample solution. Upon formation of immunocomplex, the direct electrochemical signal of the HRP decreased owing to increasing spatial blocking, and the analytes could be simultaneously determined by monitoring the signal changes.RESULTS: The proposed reagentless immunosensor array allowed simultaneous detection of carcinoma antigen 153, carcinoma antigen 125, carbohydrate antigen 199, and carcinoembryonic antigen in clinical serum samples in the ranges of 0.4-140 kU/L, 0.5-330 kU/L, 0.8-190 kU/L, and 0.1-44 mu g/L, respectively, with detection limits of 0.2 kU/L, 0.5 kU/L, 0.3 kU/L, and 0.1 mu g/L corresponding to the signals 3 SD above the mean of a zero standard. The interassay imprecision of the arrays was <9.5%, and they were stable for 35 days. The positivity detection rate of panels of tumor markers was >95.5% for 95 cases of cancer-positive sera.CONCLUSIONS: The immunosensor array provides a SMAT with short analytical time, small sampling volume, no need for substrate, and, no between-electrode cross-talk. This method not only proved the capability of the array in point-of-care testing, but also allowed simultaneous testing of several tumor markers. (C) 2008 American Association for Clinical Chemistry.

Abstract:A novel tracer, glucose oxidase-functionalized nanocomposite, was designed to label the signal antibodies for ultrasensitive multiplexed measurement of tumor markers using a disposable immunosensor array. The immunosensor array was constructed by coating layer-by-layer colloidal Prussian blue (PB), gold nanoparticles, and capture antibodies on screen-printed carbon electrodes. The preparation of glucose oxidase-functionalized nanocomposites and the labeling of antibody were performed by one-pot assembly of glucose oxidase and antibody on gold nanoparticles attached carbon nanotubes. The PB immobilized on immunosensor surface acted as a mediator to catalyze the reduction of H2O2 produced in the enzymatic cycle. Both the high-content glucose oxidase and carbon nanotubes in the tracer amplified the detectable signal for the sandwich-type immunoassay. Using carcinoembryonic antigen and alpha-fetoprotein as model analytes, the simultaneous multiplexed immunoassay method using the immunosensor array and the designed tracer showed linear ranges of 3 orders of magnitude with the detection limits down to 1.4 and 2.2 pg/mL, respectively. The assay results of serum samples with the proposed method were in an acceptable agreement with the reference values. The dual signal amplification of glucose oxidase-functionalized nanocomposites provided a promising ultrasensitive simultaneous multiplexed immunoassay approach for clinical applications.

Abstract:Inverted follicular keratosis (IFK) is a solitary benign epithelial tumor of infundibular hair follicle, generally observed in middle-aged and older individuals. An elderly male presented with a rough & raised pink colored lesion on the bald area of parietal scalp since 4 months. Fine needle aspiration (FNA) followed by histopathologic examination of the lesion was done. The FNA smears revealed features suggestive of intra-epidermal inclusion cyst while the final diagnosis of IFK was established on histopathological examination. The present article describes a correlation between the cytopathological features and histomorphology of IFK. The cytopathological features of IFK have not been described in the literature yet, hence this article provides a chance to study cytopathological features of this rare lesion.

Abstract:To improve long-term graft patient outcomes and develop more effective antirejection therapies, noninvasive monitoring of acute cellular rejection (ACR) after organ transplantation is urgently needed. As a biomarker of ACR, Granzyme B (GrB) is expected to be applied in the noninvasive monitoring of ACR. Herein, we have developed a method for detecting the GrB activity based on the target-initiated great change in electrochemical steric hindrance by designing a nanoprobe. The nanoprobe is prepared by conjugating a specific peptide, which is responsive to GrB cleavage activity, to gold nanoparticles (AuNPs). Meanwhile, a piece of DNA sequence with G-quadruplex (G4) is attached at the distal end of the peptide. Upon exposure to GrB, the peptide substrate is cleaved to eliminate the steric hindrance between inter-nanoprobes as well as nanoprobe and DNA tetrahedron (TDN), allowing the released DNA strand to hybridize with TDN, giving sensitive signal output. The method can also be used to detect GrB activity in complex biological settings, so it has a great potential for monitoring GrB activity in the blood or urine of graft patients.

Abstract:Significant progress has been made in tumor immunotherapy that uses the human immune response to kill and remove tumor cells. However, overreactive immune response could lead to various autoimmune diseases and acute rejection. Accurate and specific monitoring of immune responses in these processes could help select appropriate therapies and regimens for the patient and could reduce the risk of side effects. Granzyme B (GzmB) is an ideal biomarker for immune response, and its peptide substrate could be coupled with fluorescent dyes or contrast agents for the synthesis of imaging probes activated by GzmB. These small molecules and nanoprobes based on PET, bioluminescence imaging, or fluorescence imaging have proved to be highly GzmB specific and accuracy. This review summarizes the design of different GzmB-responsive imaging probes and their applications in monitoring of tumor immunotherapy and overreactive immune response. Abstract:A GOx/HRP@ZIF-90 nanomaterial is proposed by coating GOx and HRP in ZIF-90 using a bio-simulated mineralization method to improve the tolerance of the enzyme to the external environment. In the detection process, the ZIF-90 is turned on under mild conditions by the competitive reaction of ATP with Zn2+ and imidazole-2-carboxaldehyde (2-ICA), and the electrical signal of the system is amplified by the enzyme cascade reaction of GOx and HRP. Finally, based on the signal amplification strategy of the competitive reaction between Zn2+ and ATP to construct a "signal on" mode, electrochemical immunosensor of GOx-HRP enzyme–linked cascade reaction was prepared. The proposed electrochemical immunosensor shows an excellent analytical performance when detecting CA-125, with good selectivity and stability, with a detection range of 0.1 pg mL−1–40 ng mL−1 and a detection limit of 0.05 pg mL−1. The test has been performed using chronoamperometry under a constant voltage of −0.4 V. The immunosensor also shows an excellent performance when analysing human blood samples. The recovery of the immunosensor is 97.94–101.8%, with a relative standard deviation of 3.7–6.1%. The proposed sensor provides a novel idea for clinical use of GOx and HRP enzymes and a new method for the clinical detection of tumor markers.

Abstract:A nonenzymatic electrochemical immunosensor based on palladium nanoparticles conjugated reduced graphene nanosheets (Pd-GS) for sensitive detection of cancer biomarker alpha-fetoprotein (AFP) is described. Primary antibody-AFP (Ab(1)) was immobilized onto the surface of reduced graphene nanosheets (rGO) through an amidation reaction between the carboxylic acid group of the rGO and the available amine groups of Ab(1). Pd-GS which was prepared by one-spot synthesis is employed to immobilize secondary antibody (Ab(2)). The resulting Pd-GS-Ab(2) conjugate was used as a label for the immunosensor to detect AFP. The amplified immunoassay exhibits high sensitivity, wide linear rang (0.01-10 ng/mL), low detection limit (3.0 pg/mL), acceptable stability and reproducibility. And such immunosensor also shows good recovery in the assay results for AFP in human serum samples.

Abstract:Tumor markers play an important role in the early diagnosis and therapeutic effect monitoring of tumors. Combined detection of multiple tumor markers is a realistic way of improving the sensitivity and specificity of cancer diagnosis. To achieve this, we studied and designed a giant magneto resistance (GMR) multi-biomarker immunoassay biosensor that can simultaneously detect twelve kinds of tumor markers by integrating a GMR sensor chip, a microfluidic device, a magnetic nano-beads label, and a double antibody sandwich immunoassay method. As a proof of concept, the proposed immunosensor was utilized to detect 12 tumor markers (AFP, CEA, CYFRA21-1, NSE, SCC, PG I, PG II, CA19-9, total PSA, free PSA, free-β-hCG, Tg) and to screen patients with lung cancer, liver cancer, digestive tract cancer, prostatic cancer, etc. The immunosensor showed excellent sensitivity, accuracy, precision and stability. Designed as a POCT device, the immunosensor also allows for portability, able to perform rapid detection wherever necessary. As a multi-analyte assay, it provides significant advantages over single-analyte tests in terms of cost per test, labor and convenience. The system's ability to simultaneously measure the concentration of multiple markers in serum samples with excellent sensitivity and accuracy allows the immunosensor to be used for early tumor diagnosis.
Porous Silicon-Based Aptasensors: Toward Cancer Protein Biomarker Detection

Abstract:The anterior gradient homologue-2 (AGR2) protein is an attractive biomarker for various types of cancer. In pancreatic cancer, it is secreted to the pancreatic juice by premalignant lesions, which would be an ideal stage for diagnosis. Thus, designing assays for the sensitive detection of AGR2 would be highly valuable for the potential early diagnosis of pancreatic and other types of cancer. Herein, we present a biosensor for label-free AGR2 detection and investigate approaches for enhancing the aptasensor sensitivity by accelerating the target mass transfer rate and reducing the system noise. The biosensor is based on a nanostructured porous silicon thin film that is decorated with anti-AGR2 aptamers, where real-time monitoring of the reflectance changes enables the detection and quantification of AGR2, as well as the study of the diffusion and target-aptamer binding kinetics. The aptasensor is highly selective for AGR2 and can detect the protein in simulated pancreatic juice, where its concentration is outnumbered by orders of magnitude by numerous proteins. The aptasensor's analytical performance is characterized with a linear detection range of 0.05-2 mg mL-1, an apparent dissociation constant of 21 ± 1 μM, and a limit of detection of 9.2 μg mL-1 (0.2 μM), which is attributed to mass transfer limitations. To improve the latter, we applied different strategies to increase the diffusion flux to and within the nanostructure, such as the application of isotachophoresis for the preconcentration of AGR2 on the aptasensor, mixing, or integration with microchannels. By combining these approaches with a new signal processing technique that employs Morlet wavelet filtering and phase analysis, we achieve a limit of detection of 15 nM without compromising the biosensor's selectivity and specificity.

Abstract:We report on a novel label-free biosensing interface based on multifunctional polymeric bionanocomposites (PBNCs) and its application for sensitive detection of protein based on the analyte-induced suppression of enzymatic catalysis in PBNCs. Thrombin and its aptamer are adopted as a model system to construct an amperometric aptasensor. First, polydopamine-based PBNCs with glucose oxidase (GOx) entrapped at high load/activity and Au nanoparticles (AuNPs) dispersed in high abundance on the surface were prepared through a chemical/biochemical synthesis method, as proven by UV-vis spectrophotometry, digital imaging, and transmission electron microscopy. Then, the PBNCs were cast-coated onto an Au electrode. The PBNC-modified Au electrode presented a high chronoamperometric sensitivity of 113 ± 2 μA cm(-2) mM(-1) to glucose, as well as a high capability of immobilizing the aptamer through the surficial AuNPs to fabricate a label-free aptasensing interface. The binding of thrombin to the aptasensor surface significantly hindered the mass-transfer of the enzymatic substrates/products and thus suppressed the enzymatic catalysis efficiency, which produced obvious signal change through measuring the electrooxidation of enzymatically generated H(2)O(2). The thus-prepared aptasensor could detect thrombin with a broad detection range (1-100 nM), a detection limit down to 0.1 nM, and satisfactory specificity. The developed aptasensing method may find broad applications in the fields of clinical diagnosis, environmental protection, and food safety.
A Disposable Multianalyte Electrochemical Immunosensor Array for Automated Simultaneous Determination of Tumor Markers

Abstract:BACKGROUND:Automated and convenient multianalyte detection with high throughput is increasingly needed in clinical diagnosis. We developed a disposable 4-by-2 array for programmed simultaneous amperometric immunoassay of 4 tumor markers.METHODS:We used a screen-printed technique, 1-step immobilization method, and flow injection technique. We immobilized carcinoembryonic antigen, alpha-fetoprotein, beta-human choriogonadotropin, and carcinoma antigen 125 as model analytes in a redox mediator-grafted, biopolymer-modified, screen-printed carbon electrode array to capture corresponding horseradish peroxidase-labeled antibodies in competitive immunoreactions. The simultaneous multianalyte immunoassay was automatically carried out to amperometrically monitor the mediator-catalyzed enzymatic response to hydrogen peroxide, which decreased in proportion to the concentrations of analytes in samples.RESULTS:The multianalyte immunosensor array had a throughput of 60 samples/h and allowed simultaneous detection of carcinoembryonic antigen, alpha-fetoprotein, beta-human choriogonadotropin, and carcinoma antigen 125 in clinical serum samples with concentrations up to 188 microg/L, 250 microg/L, 266 IU/L, and 334 kIU/L, respectively. The detection limits (limits of the blank, mean of blank plus 3 SD) were 1.1 micro/L, 1.7 microg/L, 1.2 IU/L, and 1.7 kIU/L. The inter- and intraassay imprecision (CVs) of the immunosensor arrays were <7.8% and <9.0%, respectively. The immunosensor arrays were stable for 28 days.CONCLUSIONS:This newly constructed immunosensor array provides a simple, automated, simultaneous multianalyte immunoassay with high throughput, short analytical time, and sufficiently low detection limits for clinical application. This method offers the capability of miniaturizing the multianalyte detection device.

Abstract:AbstractThe detection of tumor markers is very important in early cancer diagnosis; however, tumor markers are usually present at very low concentrations, especially in the early stages of tumor development. Surface plasmon resonance (SPR) is widely used to detect biomolecular interactions; it has inherent advantages of being high-throughput, real-time, and label-free technique. However, its sensitivity needs essential improvement for practical applications. In this study, we developed a signal amplification strategy using antibody-quantum dot (QD) conjugates for the sensitive and quantitative detection of α-fetoprotein (AFP), carcinoembryonic antigen (CEA) and cytokeratin fragment 21-1 (CYFRA 21-1) in clinical samples. The use of a dual signal amplification strategy using AuNP-antibody and antibody-QD conjugates increased the signal amplification by 50-folds. The constructed SPR biosensor showed a detection limit as low as 0.1 ng/mL for AFP, CEA, and CYFRA 21-1. Moreover, the results obtained using this SPR biosensor were consistent with those obtained using the electrochemiluminescence method. Thus, the constructed SPR biosensor provides a highly sensitive and specific approach for the detection of tumor markers. Abstract:An enzyme electrocatalytic immunosensor based on functional composite nanofibers for sensitive detection of tumor suppressor protein p53 (AGp53) is developed. The carboxylated multi-walled carbon nanotube (MWCNT) doped nylon 6 (PA6) composite nanofibers (MWCNT-PA6) were prepared using electrospinning, which served as the nanosized backbone for thionine (TH) electropolymerization. The functional composite nanofibers (MWCNT-PA6-PTH) served as supporting scaffolds not only promotes electron transfer, but also increases the surface area to introduce a large amount of AGp53 capture antibody (Ab1), thus increasing the detection sensitivity. After a sandwich immunoreaction, AGp53 and horseradish peroxidase (HRP)-labeled detection antibody (HRP@Ab2) captured onto the electrode surface produced an amplified electrocatalytic response by the reduction of enzymatically oxidized polythionine (PTH) in the presence of hydrogen peroxide (H2O2). The increase of response current was proportional to the AGp53 concentration in the range of 2pgmL−1 to 2ngmL−1 and the detection limit was down to 1pgmL−1. This study provides new insight into the design of advanced electrospun nanofiber-based immunosensor for detection and analysis of a variety of proteins. It holds promise for the diagnosis of cancer development and monitoring of patient therapy.

Abstract:A highly sensitive and efficient home-use method is impressive and promising in the self-monitoring of chronic prostatitis (CP). Herein, we developed a glucose oxidase-zeolitic imidazolate framework-8-antibody composite (GOx@ZIF-8@Ab 2 ) to achieve highly sensitive point-of-care testing (POCT) of urinary prostatic exosomal protein (PSEP) by combining it with a personal glucometer (PGM). The developed GOx@ZIF-8@Ab 2 was prepared through a simple but effective biomineralization reaction and a strong binding affinity between Zn 2+ and Fc region of the antibody. This bifunctional material GOx@ZIF-8@Ab 2 could not only specifically recognize the PSEP via immunoreaction but also wrap a large number of GOx molecules. Following the sandwich immunoreaction, the prepared GOx@ZIF-8@Ab 2 facilitated hydrolysis of glucose in a one-to-many fashion, amplifying the detection signal and greatly improving the sensitivity of PGM detection. In comparison to those of the conventional enzyme-linked immunosorbent assay (ELISA), the detection time of the developed technique was shortened to 45 minutes and its limit of detection (0.23 pg mL −1 ) was reduced by 4–5 orders of magnitude. This technique was successfully utilized for detecting PSEP in human urine samples with a recovery rate of 96–116%. Owing to its rapid, ultrasensitive, specific and portable features, the as-designed platform based on GOx@ZIF-8@Ab 2 and the PGM device holds great promise as a home-use POCT platform for self-monitoring of CP and other chronic diseases.

Abstract:An innovative electrochemical immunosensing platform was designed for the sensitive monitoring of lung cancer biomarker (pro-gastrin-releasing peptide; ProGRP) by using platinum nanoparticles encapsulated inside dendrimers (PtDEN) as enzymatic mimics for the signal amplification. PtDEN nanocomposites were prepared through a simple chemical reduction method with the assistance of NaBH4. Thereafter, PtDEN-labeled anti-ProGRP secondary antibody was launched for the detection of target analyte with a sandwich-type assay format on anti-ProGRP capture antibody-modified screen-printed carbon electrode. Accompanying formation of immunocomplex, the labeled PtDENs electrochemically oxidized 3,3',5,5'-tetramethylbenzidine (TMB) in the presence of hydrogen peroxide to produce a well-defined voltammetric signal within the applied potentials. Thanks to the high-efficient catalytic efficiency of platinum nanoparticles and high-loading ability of dendrimer, improved analytical features were acquired with PtDENs relative to platinum nanoparticles alone. Using PtDENs labeling strategy, the properties and factors influencing the analytical performance of electrochemical immunosensor were studied in detail. The strong bioconjugation of antibodies with the PtDENs caused a good repeatability and intermediate precision down to 7.64%. Under optimum conditions, the electrochemical immunosensor exhibited a dynamic linear range of 0.001 – 10 ng mL-1 ProGRP with a detection limit of 0.86 pg mL-1. Good selectivity and relatively long-term stability (>6 months) were achieved for target ProGRP. Significantly, the acceptable accuracy was gotten for analysis of ProGRP in human serum specimens referring to commercially available human ProGRP enzyme-linked immunosorbent assay (ELISA) method.

Novel Polymeric Bionanocomposites with Catalytic Pt Nanoparticles Label Immobilized for High Performance Amperometric Immunoassay

Abstract:We report on the aqueous preparation of novel polymeric bionanocomposites (PBNCs) with polydopamine (PDA) as an efficient matrix to support antibody and uniformly dispersed abundant Pt nanoparticles (PtNPs) for high performance sandwich-type amperometric immunoassay. We prepare three kinds of PBNCs (PBNCs 1, 2, and 3) via chemical polymerization synthesis (1) and further adsorption (2) or glutaldehyde (GA) covalent crosslinking (3) of antibody. Transmission electron microscopy, atomic force microscopy, and quartz crystal microbalance are used to characterize the PBNCs. The results show that PDA is an excellent matrix to support antibody and PtNPs due to its high biocompatibility, adsorbability, and processibility, and the thus-prepared PBNCs are of high immuno-recognition efficiency and high catalytic activity toward H(2)O(2) reduction. Then, we examine the applicability of the prepared PBNCs for sandwich-type amperometric immunosensing using a model immuno-pair of human immunoglobulin G (hIgG) and anti-hIgG, and obtain the signal by detecting the electro-reduction of H(2)O(2) catalyzed by the PtNPs label. The constructed immunosensors using PBNCs 1, 2 and 3 exhibit detection limits of 0.068, 0.037, and 0.018 ng mL(-1), respectively, being competitive with or better than the reported analogues. Our immunosensors also present good reproducibility, stability, regeneration ability, specificity, and satisfactory feasibility for target assay in clinical human serum samples. The suggested protocol for the preparation of PBNCs with high signal-transduction ability is expected to find wide bioassay applications.
