Abstract:dine treatment before stavudine therapy was initiated. These patients had a mean MCV of 103.9 fL (range, 81.3–118.8 fL) at initiation of therapy and a statistically insignificant mean increase in MCV of 2.2 fL (range, 10.3–20.2 fL; P 5 .11) at least 3 months after stavudine was substituted. Stavudine was used in conjunction with lamivudine therapy for 79 (87%) of the patients. Of 12 patients taking stavudine without lamivudine therapy, 10 (83%) had an MCV of .95.0 fL and five (42%) had an MCV of .100.0 fL. Lamivudine monotherapy (as used in phase 2/3 trials) is uncommonly associated with macrocytosis (only at doses at least twice as high as those currently administered). Macrocytosis induced by zidovudine is often associated with anemia. Anemia has not typically been associated with stavudine therapy. In our patients, there was actually a small, but statistically significant, increase in hemoglobin levels with stavudine therapy. This increase was accentuated in patients whose treatment was switched from zidovudine who had an initial hemoglobin concentration of ,12 g/dL. The mean baseline hemoglobin level for all 91 patients was 13.4 g/dL (range, 8.4–17.2 g/dL), and the mean hemoglobin level after at least 3 months of stavudine treatment was 14.2 g/dL (range, 10.1–17.1 g/dL; P , .05). For those patients for whom stavudine was substituted for zidovudine treatment, the mean baseline hemoglobin level was 12.9 g/dL (range, 5.5–17.2 g/dL), and the mean hemoglobin concentration after at least 3 months of therapy was 14.1 g/dL (range, 11.7–17.1 g/dL; P , .05). As treatment of HIV infection has evolved, ever more complex regimens with five or more drugs in combination are being used. There have been limited clinical trials of many of the drugs before widespread use, and toxicities are discovered only after significant use outside of these trials. Unexplained macrocytosis in an HIVinfected patient receiving combination therapy may lead to an extensive workup for bone marrow or gastrointestinal disease. Alternately, macrocytosis may be attributed to a new agent as a yet undescribed toxic effect. Fortunately, macrocytosis associated with stavudine, at the currently recommended doses, is not associated with anemia. We conclude that the finding of isolated macrocytosis, without anemia, in a patient being treated with stavudine may be due primarily to stavudine itself and may not warrant further evaluation.
Fermentation of the Brown Seaweed Alaria esculenta by a Lactic Acid Bacteria Consortium Able to Utilize Mannitol and Laminari-Oligosaccharides

Abstract:The brown seaweed Alaria esculenta is the second most cultivated species in Europe, and it is therefore of interest to expand its application by developing food products. In this study, a lactic acid bacteria consortium (LAB consortium) consisting of three Lactiplantibacillus plantarum strains (relative abundance ~94%) and a minor amount of a Levilactobacillus brevis strain (relative abundance ~6%) was investigated for its ability to ferment carbohydrates available in brown seaweed. The consortium demonstrated the ability to ferment glucose, mannitol, galactose, mannose, and xylose, of which glucose and mannitol were the most favored substrates. No growth was observed on fucose, mannuronic and guluronic acid. The consortium used different pathways for carbohydrate utilization and produced lactic acid as the main metabolite. In glucose fermentation, only lactic acid was produced, but using mannitol as a carbohydrate source resulted in the co-production of lactic acid, ethanol, and succinate. Xylose fermentation resulted in acetate production. The consortium was also able to utilize laminari-oligosaccharides (DP2-4), obtained after enzymatic hydrolysis of laminarin, and produced lactic acid as a metabolite. The consortium could grow directly on A. esculenta, resulting in a pH decrease to 3.8 after 7 days of fermentation. Incubation of the same seaweed in corresponding conditions without inoculation resulted in spoilage of the seaweed by endogenous bacteria.

Abstract:Kelp (Laminaria japonica) is an important marine resource with low cost and rich nutrition. However, its fishy odor has compromised consumer acceptance. In this study, the effects of fermentation with Lactobacillus plantarum FSB7, Pediococcus pentosaceus CICC 21862 and Saccharomyces cerevisiae SK1.008 on fishy notes in kelp was studied using gas chromatography-mass spectrometry (GC-MS), gas chromatography-ion mobility spectrometry (GC-IMS) and odor activity values (OAVs). Forty-four volatile organic compounds (VOCs) were identified in unfermented kelp, most of which were aldehydes, followed by alkanes, alcohols and ketones. Among them were 19 volatile compounds with OAV greater than one. Substances containing α,β-unsaturated carbonyl structure (1-Octen-3-one, (E,Z)-2,6-nonadienal, (E,E)-2,4-decadienal, etc.) are the main contributors to kelp fishy odor. The number of VOCs in kelp samples fermented by L. plantarum, P. pentosaceus and S. cerevisiae were decreased to 22, 24 and 34, respectively. GC-IMS shows that the fingerprint of the S. cerevisiae fermented sample had the most obvious changes. The disappearance of 1-octen-3-one and a 91% decrease in unsaturated aldehydes indicate that S. cerevisiae was the most effective, while L. plantarum and P. pentosaceus only reached 43–55%. The decrease in kelp fishy notes was related to the decrease in α,β-unsaturated carbonyl groups. The experimental results show that odor reduction with fermentation is feasible.

Abstract:Wet anaerobic storage of algal biomass is a promising preservation approach that can ensure a continuous supply of these feedstocks to biorefineries year-round. An effective solution to preservation must ensure minimal dry matter loss and a change in biochemical composition during storage. Therefore, the objective of this study is to investigate the preservation of Nannochloropsis gaditana biomass through wet anaerobic storage and its impact on biomass quality. Prior to storage, the algae sample is inoculated with two different strains of lactic acid bacteria and thereafter stored for 30 and 180 days. Each inoculant limited the dry matter loss to <10% (dry basis) after the storage duration. Final pH values (4.3–4.8) indicate that the biomass samples are properly ensiled, achieving the acidic conditions necessary for preservation. Compositional analysis of the biomass after storage shows a reduction in carbohydrate content, a relative increase in lipid content, and no significant change in the protein fraction. Glucose and galactose were the most prevalent sugar monomers. The low dry matter loss and minimal compositional change indicate that wet anaerobic storage is an effective means of preserving algal biomass and ensuring a constant supply of algal biomass feedstock to a biorefinery.

Abstract:Sugar kelp ( Saccharina latissima ) is a native European brown macroalga with the potential to become a vital part of the green transition of the food industry. Knowledge of the sugar kelp shelf-life is essential to designing the food supply chain to ensure safe and high-quality food. Establishing a single-compound quality index (SCQI) of freshness would be useful for the industry. However, information is currently lacking on how different post-harvest treatments affect the shelf-life of sugar kelp, even though it is important knowledge for manufacturers, authorities and consumers. The objective of this study was to establish the shelf-life of refrigerated sugar kelp following five post-harvest treatments and evaluate the effect of these treatments on changes in quality attributes (sensory, microbial, chemical and physical) during storage to select the SCQI. The post-harvest treatments included washing in sea water, washing in potable water, blanching for 2 min in sea water or potable water and untreated sugar kelp. Based on sensory analysis, the refrigerated (+ 2.8°C) shelf-lives for sugar kelp from all treatments were seven to 9 days. The end of the sensory shelf-life correlated with the development of >7 log (CFU g −1 ) aerobic viable counts, suggesting this attribute can be used as a SCQI to evaluate the shelf-life of sugar kelp. The microbiota was dominated by putative spoilage organisms from the Pseudoalteromonadaceae and Psychromonadaceae families. Untreated and washed sugar kelp continued to respire and consume carbohydrates up to 5 days post-harvest, indicating respiration rates may be used to determine freshness of non-blanched kelp. Favorable organoleptic properties, e.g., sweetness and umami, decreased during storage and coincided with a reduction in water-soluble mannitol and free glutamic acid. Both blanching treatments changed texture and color and reduced iodine and vitamin C contents while retaining components such as fucoxanthin, chlorophyll a and β-carotene. This study provides crucial documentation of quality changes during the post-harvest storage period of sugar kelp, including information about sugar kelp spoilage and nutrient changes, which would facilitate the development of best practices for manufacturers using sugar kelp in their production of food.

Abstract:Lactobacillus plantarum (widespread member of the genus Lactobacillus ) is one of the most studied species extensively used in food industry as probiotic microorganism and/or microbial starter. The exploitation of Lb. plantarum strains with their long history in food fermentation forms an emerging field and design of added-value foods. Lb. plantarum strains were also used to produce new functional (traditional/novel) foods and beverages with improved nutritional and technological features. Lb. plantarum strains were identified from many traditional foods and characterized for their systematics and molecular taxonomy, enzyme systems ( α -amylase, esterase, lipase, α -glucosidase, β -glucosidase, enolase, phosphoketolase, lactase dehydrogenase, etc.), and bioactive compounds (bacteriocin, dipeptides, and other preservative compounds). This review emphasizes that the Lb. plantarum strains with their probiotic properties can have great effects against harmful microflora (foodborne pathogens) to increase safety and shelf-life of fermented foods.

Abstract:Seaweed, a valuable marine resource widely cultivated worldwide, can be vulnerable to stress and microbiome alterations, resulting in the decay of seaweeds and substantial economic losses. To investigate the seaweed-microbiome interaction, our study aimed to isolate marine bacteria and fungi that can cause Ice–Ice disease and evaluate their enzymatic characteristics for potential application in bioethanol production from seaweed biomass. Three red seaweed species ( Gracilaria edulis , Kappaphycus alvarezii , and Eucheuma cottonii ) were obtained for our study and placed in separate culture tanks. Among the 18 isolated marine microbial species, 12 tested positive for agar and carrageenan activity: six exhibited both activities, three displayed only agar activity, and three only carrageenan activity. DNA sequencing of the positive microbes identified ten bacteria and two yeast species. The 3,5-Dinitrosalicylic acid (DNSA) assay results revealed that the identified bacterial Caldibacillus kokeshiiformis strain FJAT-47861 exhibited the highest carrageenase activity (0.76 units/ml), while the yeast Pichia fermentans strain PM79 demonstrated the highest agarase activity (0.52 units/ml). Notably, Pichia fermentans strain PM79 exhibited the highest overall agarase and carrageenase activity, averaging 0.63 units/ml. The average carrageenase activity of all six positive microbes was 1.5 times higher than their agarase activity. These findings suggest that the 12 isolated microbes hold potential for bioethanol production from macroalgae, as their agarase and carrageenase activity indicates their ability to break down seaweed cell wall carbohydrates, causing ice–ice disease. Moreover, these results provide exciting prospects for harnessing the bioconversion capabilities of these microbes, paving the way for sustainable and efficient bioethanol production from seaweed resources.

Abstract:Spanish mackerel is an important economic fish species in China, but corruption undermines its overall economic value. Fermentation serves a crucial technique for preserving perishable fish in developing countries.This study aimed to examine the dynamic changes in the gel properties, physicochemical properties, microbial diversity, and volatile flavor compounds (VFCs) of Spanish mackerel sausages inoculated with Lactiplantibacillus plantarum CY1-2 during fermentation. The correlation between key microorganisms and VFCs was explored via two-way orthogonal partial least squares analysis. The results showed that after 40 h of fermentation, compared with the control group (without starter), the group inoculated with strainCY1-2 exhibited a 207.7% increase in gel strength, 54.2% decrease in TVB-N levels, 37.49% increase in the total amount of free amino acids, and a 34.12% and 58.73% increase in the DPPH and ABTS radical scavenging rate, respectively. In total, using gas chromatography coupled with mass spectrometry, 36 VFCs were detected in fermented sausages, including 9 aldehydes, 9 hydrocarbons, 7 alcohols, 4 acids, 2 ketones, and 5 esters. High-throughput sequencing demonstrated that the bacterial profiles were altered in sausages inoculated with strain CY1-2 during fermentation. Enhydrobacter dominated initially but was quickly replaced by Macrococcus after 8 h of fermentation, while Lactobacillus became the dominant genus after 40 h. Correlation analysis revealed that Lactobacillus and Staphylococcus played important roles in the production of VFCs in the fermented sausages. Notably, Lactobacillus was positively associated with 2-undecanone, pentadecane, and hexanal. This study confirmed that strainCY1-2 inoculation could enable the production of high-quality fermented fish sausages.

Abstract:BACKGROUND: The non-dairy sector is growing, fermented alternatives to dairy are sparse. Adapted starter cultures to substituting raw materials needs to be developed.OBJECTIVE: Aims of this study were to isolate, identify, and phenotypically characterize lactic acid bacteria (LAB) that inhabit Swedish legumes, and assess properties necessary for selecting strains with the ability to ferment a bean beverage and with potential health beneficial properties.DESIGN: Isolates of presumed LAB were obtained from legumes collected at Öland, Sweden. Strain diversity was assessed by repetitive polymerase chain reaction (rep-PCR). The strains were identified using matrix-assisted laser desorption/ionization-time-of-flight mass spectrometry (MALDI-TOF MS). Species belonging to Enterococcus were predominant along with Pediococcus and closely related Bacillus. Strains were tested for tolerance to low pH, phenol, and bile as well as their bile salt hydrolase (BSH) activity. In addition, Enterococcus strains were tested for antibiotic resistance, and Pediococcus strains for their ability to ferment a bean beverage.RESULTS: From the 25 strains characterized, five were found resistant to low pH, bile, and phenol, suggesting that they can survive a passage through the gastrointestinal tract (GIT) and hence potentially exert beneficial effects in the host. These are suggested for further investigation on specific host-beneficial properties. Two of these, belonging to Pediococcus pentosaceus, were able to ferment a bean beverage without any added nutrients, indicating that the Pediococcus strains are well adapted to the bean substrate. One of the P. pentosaceus strains were also able to markedly improve the reduction of phytate by the phytase-producing yeast strain Pichia kudriavzevii TY1322 during co-fermentation as well as increase the final cell count of the yeast strain.CONCLUSION: Strain isolation and characterization performed in this study aids in selecting starter cultures for legume fermentation. Nutritional properties can be improved by co-fermentation with yeast indicating that novel nutritious fermented non-dairy products could be developed.

Abstract:Fermented Asian foods have recently gained popularity, crossing from Asian communities to mainstream markets, in many western countries. However, less attention has been paid to the safety of these foods. In South-East Asia, fermented products are still produced following traditional methods. Therefore, consumers are not confident in their microbial safety. The challenges awaiting fermentation in South-East Asia are thus to improve safety and quality in a sustainable system producing tasty and typical fermented products. A possible solution could be the use of starter cultures able to increase the safety of food stuffs. Starters could also help to standardize the production process and reduce ripening times. The current review addresses the role of lactic acid bacteria on the microbiological and chemical safety of Asian legume based fermented products. In particular, their role in the reduction of anti nutritional compounds (e.g. phytates) and protein allergenicity is discussed. Moreover, starters can inihibit the development of amino acid-decarboxylating microbes preventing the accumulation of biogenic amines, they can also be useful to reduce the accumulation of mycotoxins and inhibit pathogens' development. Finally, their role in the degradation of pesticides is analyzed.

Prevention of microbes-induced spoilage in sodium chloride-free cucumber fermentations employing preservatives

Abstract:This study evaluated preservatives to stabilize sodium chloride (NaCl)-free-cucumber fermentations. The brining of air-purged laboratory cucumber fermentations with 100.0 mM calcium chloride (CaCl2 ) and 25.0 mM acetic acid resulted in immediate rises in pH, the chemical reduction of the medium, and malodors. Supplementation with 3.0 mM sodium benzoate or 3.0 mM potassium sorbate enabled a decline in pH, a continuous oxidative state of the medium, and delayed rising pH spoilage. However, lactic and acetic acids eventually disappeared in fermentations supplemented with preservatives. The amount of preservatives needed to suppress growth of brined-cucumber-spoilage microbes was determined in Fermented Cucumber Juice Medium (FCJM). Supplementation of FCJM with 10.0 mM sodium benzoate was inhibitory for the spoilage yeasts, Issatchenkia occidentalis and Pichia manshurica, and the lactobacilli, Lentilactobacillus buchneri and Lentilactobacillus parafarraginis, but not of Zygosaccharomyces globiformis. Potassium sorbate inhibited the spoilage yeasts at 15.0 mM in FCJM but not the lactobacilli. Supplementation of FCJM with 20.0 mM fumaric acid had a bactericidal effect on the spoilage-associated lactobacilli. As expected, NaCl-free-commercial cucumber fermentations brined with 100 mM CaCl2 , no acetic acid, and 6 mM potassium sorbate resulted in complete fermentations, but supported rising pH, microbially induced spoilage during long-term storage. Post-fermentation supplementation with 12 mM sodium benzoate, 10 mM fumaric acid, a combination of the two, or 10 mM fumaric acid and 2 mM AITC prevented microbial activity during long-term bulk storage. PRACTICAL APPLICATION: Several preservative-based strategies for stabilizing NaCl-free cucumber fermentation in a commercial production setting were developed, enabling the implementation of a processing technology that reduces wastewater volumes and environmental impact.

Abstract:Seaweed silage has potential as an alternative feed ingredient for dairy cows. This study aims to investigate seaweed's and seaweed silageś nutrient digestibility as well as their impact on the ruminal bacterial composition. The cultivated S. latissima and A. esculenta were preserved by freezing at - 40 °C or ensiling (16 °C, 3 months) with four different treatments: no additives, 4 g formic acid/kg wet seaweed, lactic acid bacteria (LAB) inoculant, and LAB inoculant in prewilted biomass (ca. 300 g DM/kg wet biomass). The nutrient digestibility was estimated using standard feed evaluation methods. The bacterial composition in ruminal fluid after 48 h in vitro anaerobic incubation with seaweeds and common feedstuffs was analysed using 16S ribosomal RNA (rRNA) amplicon sequencing (V3-V4) and quantitative PCR (qPCR). The results suggest that S. latissima was more digestible than A. esculenta and that the preservation treatments had only a small effect on the nutrient digestibility and ruminal bacteria compositions. The rumen DM degradability of S. latissima was comparable to common perennial and corn forage; however, the total tract CP digestibility of S. latissima (460 g/kg CP) was lower than common forages (620 - 820 g/kg CP) and was not improved by ensiling. There was a lack of insoluble but rumen-degradable CP in A. esculenta, making it unsuitable as a nutrient ingredient for dairy cows. The ruminal bacterial composition changed depending on the seaweed species used as substrate: The dominant bacterial taxa when incubated with S. latissima belonged to the genus Prevotella (relative abundance: 79 - 93%), known for its ability to degrade polysaccharides in various ecosystems. Moreover, the fibrolytic bacteria including Fibrobacter succinogenes and Ruminococcus flavefaciens were > 2.5 Log2FoldChange higher when incubating with S. latissima than with A. esculenta. These bacterial taxa may play an important role in the in vitro organic matter digestibility, noted as 2 times higher in S. latissima compared to A. esculenta. The qPCR results indicated potential methane mitigation properties of the studied seaweed species, with significantly lower gene copies of Archaea 16S rRNA and methyl coenzyme-M reductase subunit A genes when the ruminal fluid was incubated with the seaweed substrates. Our study suggested that ensiled S. latissima biomass can be included in the diet of dairy cows as an alternative forage-like ingredient with the potential of methane mitigation.

Abstract:We describe a case of transient cortical blindness following internal mammary artery to left anterior descending coronary artery graft angiography. This dramatic, infrequent, and self-limiting complication so far has not been described in the cardiovascular literature. In the present era of internal mammary artery use for the left coronary artery grafting, the angiographer should be familiar with the diagnosis, prevention, and management of complications previously seen mostly during the posterior cerebral circulation angiography.

Abstract:Fermented nut-based products, obtained after soaking and fermentation, are gaining increasing interest as animal food substitutes because of ethical, environmental and health reasons. In these products, Lactic Acid Bacteria (LAB) perform the fermentation, leading to matrix acidification and contributing to controlling spoilage and pathogenic microbiota. In this work, LAB strains isolated from an artisanal product and combined with a commercial strain were added as starter cultures during nut soaking to produce a cheese-like fermented plant-based product. Three different LAB consortia were used in challenge tests at laboratory scale against Listeria monocytogenes, Escherichia coli or Salmonella Enteritidis, inoculated in nuts at 5 log CFU/g, and monitored for pathogen survival and matrix acidification. The combination of Lactiplantibacillus plantarum 82 and Leuc. carnosum 4010 resulted in faster acidification (pH value < 4.4 after 18 h instead of 48 h) and the reduction of target pathogens; L. monocytogenes was already absent after seven days from production, and the counts of E. coli or S. Enteritidis were lower with respect to other samples. Thus, this microbial consortium was used for a pilot-scale production in which, beyond safety, the fermented plant-based product was also characterized for aroma profile and phenolic compounds, parameters that are known to be affected by LAB fermentation. The results showed an enhancement of the aroma profile, with an accumulation of molecules able to confer cheese-like notes (i.e., acetoin and diacetyl) and higher phenolic content, as well as the presence of compounds (i.e., phenyllactic acid and hydroxyphenyllactic acid) that could exert antimicrobial activity. This study allowed us to set up a guided fermentation for a cheese-like vegan product, guaranteeing safety and improving aromatic and functional features.
Microbial Regulation of Deterioration and Preservation of Salted Kelp under Different Temperature and Salinity Conditions

Abstract:High salinity is an effective measure to preserve kelp, but salted kelp can still deteriorate after long-term preservation. In order to clarify the key conditions and microbial behavior of salted kelp preservation, 10% (S10), 20% (S20), and 30% (S30) salt concentrations were evaluated at 25 °C (T25) and 4 °C (T4). After 30 days storage, these salted kelps showed different states including rot (T25S10), softening (T25S20), and undamaged (other samples). By detecting polysaccharide lyase activity and performing high-throughput sequencing of the prokaryotic 16S rRNA sequence and metagenome, we found that deteriorated kelps (T25S10 and T25S20) had significantly higher alginate lyase activity and bacterial relative abundance than other undamaged samples. Dyella, Saccharophagus, Halomonas, Aromatoleum, Ulvibacter, Rhodopirellula, and Microbulbifer were annotated with genes encoding endonuclease-type alginate lyases, while Bacillus and Thiobacillus were annotated as the exonuclease type. Additionally, no alginate lyase activity was detected in undamaged kelps, whose dominant microorganisms were halophilic archaea without alginate lyase-encoding genes. These results indicated that room-temperature storage may promote salted kelp deterioration due to the secretion of bacterial alginate lyase, while ultra-high-salinity and low-temperature storage can inhibit bacterial alginate lyase and promote the growth of halophilic archaea without alginate lyase, thus achieving the preservation of salted kelp.

Abstract:Recent consumer demand for non-dairy alternatives has forced many manufacturers to turn their attention to cereal-based non-alcoholic fermented products. In contrast to fermented dairy products, there is no defined and standardized starter culture for manufacturing cereal-based products. Since spontaneous fermentation is rarely suitable for large-scale commercial production, it is not surprising that manufacturers have started to adopt centuries-known dairy starters based on lactic acid bacteria (LABs) for the fermentation of cereals. However, little is known about the fermentation processes of cereals with these starters. In this study, we combined various analytical tools in order to understand how the most common starter cultures of LABs affect the most common types of cereals during fermentation. Specifically, 3% suspensions of rice, oat, and wheat flour were fermented by the pure cultures of 16 LAB strains belonging to five LAB species-Lacticaseibacillus paracasei, Lactobacillus delbrueckii, Lactobacillus helveticus, Streptococcus thermophilus, and Lactococcus lactis. The fermentation process was described in terms of culture growth and changes in the pH, reducing sugars, starch, free proteins, and free phenolic compounds. The organoleptic and rheological features of the obtained fermented products were characterized, and their functional properties, such as their antioxidant capacity and angiotensin-converting enzyme inhibitory activity, were determined.
Extending viability of Lactobacillus plantarum and Lactobacillus johnsonii by microencapsulation in alginate microgels

Abstract:Aim: To investigate whether microencapsulation of Lactobacillus in alginate microbeads will lead to increased longevity during refrigerated storage or simulated digestion. Materials and methods: Microscopy was used to confirm that Lactobacillus plantarum ATCC BAA-793 and Lactobacillus johnsonii ATCC 33200 were immobilised within the microbeads and laser scattering analysis was used to determine the mean diameter of the microbeads. The number of viable cells were enumerated throughout refrigerated storage and simulated digestion experiments. Results: Microencapsulation was shown to have differing effects on viability depending on the species, but led to extended viability during refrigerated storage and simulated digestion in L. johnsonii and L. plantarum respectively. Conclusion: Fermented functional foods contain microbes beneficial to human health. However, extended shelf storage and the harsh environment of the GI tract significantly reduces the number of viable microbes reaching the consumer. Microencapsulation allows beneficial microbes to reach the gut of the consumer in higher numbers, and thus confer greater health benefits.
Salmon Gravlax Biopreservation With Lactic Acid Bacteria: A Polyphasic Approach to Assessing the Impact on Organoleptic Properties, Microbial Ecosystem and Volatilome Composition

Abstract:Seafood and fishery products are very perishable commodities with short shelf-lives owing to rapid deterioration of their organoleptic and microbiological quality. Microbial growth and activity are responsible for up to 25% of food losses in the fishery industry. In this context and to meet consumer demand for minimally processed food, developing mild preservation technologies such as biopreservation represents a major challenge. In this work, we studied the use of six lactic acid bacteria (LAB), previously selected for their properties as bioprotective agents, for salmon dill gravlax biopreservation. Naturally contaminated salmon dill gravlax slices, with a commercial shelf-life of 21 days, were purchased from a French industrial company and inoculated by spraying with the protective cultures (PCs) to reach an initial concentration of 106 log CFU/g. PC impact on gravlax microbial ecosystem (cultural and acultural methods), sensory properties (sensory profiling test), biochemical parameters (pH, TMA, TVBN, biogenic amines) and volatilome was followed for 25 days of storage at 8°C in vacuum packaging. PC antimicrobial activity was also assessed in situ against Listeria monocytogenes. This polyphasic approach underlined two scenarios depending on the protective strain. Carnobacterium maltaromaticum SF1944, Lactococcus piscium EU2229 and Leuconostoc gelidum EU2249, were very competitive in the product, dominated the microbial ecosystem, and displayed antimicrobial activity against the spoilage microbiota and L. monocytogenes. The strains also expressed their own sensory and volatilome signatures. However, of these three strains, C. maltaromaticum SF1944 did not induce strong spoilage and was the most efficient for L. monocytogenes growth control. By contrast, Vagococcus fluvialis CD264, Carnobacterium inhibens MIP2551 and Aerococcus viridans SF1044 were not competitive, did not express strong antimicrobial activity and produced only few organic volatile compounds (VOCs). However, V. fluvialis CD264 was the only strain to extend the sensory quality, even beyond 25 days. This study shows that C. maltaromaticum SF1944 and V. fluvialis CD264 both have a promising potential as bioprotective cultures to ensure salmon gravlax microbial safety and sensorial quality, respectively.
