Abstract:The goals of this study were to optimize the medium formulation for enhanced production of Aspergillus niger inulinase using Plackett–Burman Design (PBD) and to model the fermentation in optimal medium formulation. Results indicated that (NH4)2SO4 (negative effect), yeast extract and peptone (positive effect) were determined as significant factors affecting the inulinase production. Different media including Medium A (non-enriched), Medium B (contains both negative and positive factors) and Medium C (contains only positive factors) were formed and inulinase fermentations were performed. Findings showed that the best nutritional formulation was Medium C, which yielded to be 1011.02 U/mL, 834.28 U/mL, 1.22, 4383.44 U/mg, 4186 U/mg, 158.49 U/mL/day, 128.60 U/mL/day and 94.54% of PInulinase, SInulinase, I/S ratio, SInulinase, SSucrase, QInulinase, QSucrase and SUY, respectively. Additionally, fungal growth, enzyme or protein production and substrate consumption were modeled using the logistic model, Luedeking–Piret model, and modified Luedeking–Piret model, respectively, and found that enzyme or protein production was non-growth associated. Besides, maintenance value (Z) was lower than γ value, indicating that A. niger mainly utilizes the sugars for enzyme production and fungal growth. Consequently, optimum medium composition was successfully determined by PBD and also the kinetic models fitted the experimental data very well with high regression coefficient.

Abstract:Inulinases are enzymes of great interest in the food industry, especially due to their application in the synthesis of fructose and fructo-oligosaccharides. Moreover, some inulinases (I) also present invertase activity (S), making them useful for sucrose hydrolysis processes. In the present study, the production of inulinase by Aspergillus niger URM5741 was evaluated and optimized using two statistical approaches. First, the composition of the cultivation medium was determined through a simplex centroid mixture design, followed by the selection of optimal fermentation conditions using the Box-Behnken design. Based on these experimental designs, the maximum activities of inulinase (16.68 U mL-1) and invertase (27.80 U mL-1) were achieved using a mixture of wheat, soy, and oat brans (5 g), along with 2.5% inulin and 40% moisture. The inulinase exhibited optimum temperature and pH of 60 °C and 4.0, respectively, displayed a high affinity for both substrates, as evidenced by very-low Michaelis constant values (1.07-1.54 mM). A relative thermostability was observed at 55-60 °C as indicated by half-lives values (I: 169.06-137.27 min; S: 173.29-141.52 min) and D-values (I: 561.61-456.00 min; S: 575.65-470.11 min) which were further confirmed by the high activation energy (123.01 and 143.29 kJ mol-1). The enzyme demonstrated favorable results in terms of inulin and sucrose hydrolysis, being a maximum release of reducing sugars of 6.04 and 15.80 g L-1, respectively. These results indicate that the sequential statistical approach proved to be beneficial to produce inulinase by A. niger URM5741, with the obtained enzyme considered promising for long-term industrial applications.

Abstract:beta-glucosidase produced by Aspergillus niger AS 3.4309 was carried out under Solid State Fermentation (SSF). The effects of medium composition and incubation conditions on the enzyme activity were investigated and suitable solid medium components and optimum incubation conditions for fermentation process were therefore established. The maximum beta-glucosidase production (508 U g(-1) dry matter) occurred after 72 h of incubation at 28 degrees C in optimized solid medium containing wheat bran 100%, ammonium chloride 2%, MgSO4 center dot 7H(2)O 0.5% and KH2PO4, 1.5% at the optimal conditions including moisture content of solid substrate 70% (w/w) initial pH 6.0 and inoculum level at 1 x 10(7) spores g(-1) of solid substrate.
Kinetic Behaviors of Inulin Hydrolysis by Inulinase from Kluyveromyces

Abstract:Enzyme hydrolysis kinetic model of inulin by inulinase from Kluyveromyces lactis was established. The corresponding Michaelis-Menten equation model constants were found to be related to temperature through the Arrhenius model. The kinetic model was validated by experimental data and also be utilized to predict the hydrolysis reaction rates of the inulin for substrate concentrations ranging between 2 and 200 g/L and a fairly large reaction temperature range (35-55 degrees C). Furthermore, this kinetic model can provide a scientific and convenient way to choose the optimized hydrolysis conditions for the production of fermentable sugar for ethanol production.
Optimization by Response Surface Methodology of Solid-state Fermentation Medium for α-Galactosidase Production by Aspergillus niger ZJUQH

Abstract:In order to enhance the production of α-galactosidase by Aspergillus niger ZJUQH,response surface methodology was applied to optimize the solid culture medium.A fractional factorial design was used to investigate the main factors affecting α-galactosidase yield.The results showed that two factors played important roles in the production of α-galactosidase.Central composite design and response surface analysis were applied to establish a second-order regression equation model for the yield of α-galactosidase.At last,the optimal fementation condition were obtained by response surface analysis as follows: peptone 0.6 g,Na2HPO4.12H2O 0.08 g,ratio of solid to water 0.41 and inoculum quantity 1.5 mL/5g.Under the optimal conditions,the activity of α-galactosidase reached(77.21±2.01) U/g,which was increase by 57.41% compared with(49.05 ± 2.11) U/g before the oprimization,that is,peptone 0.4 g,Na2HPO4.12H2O 0.08 g,ratio of solid to water 0.33 and inoculation quantity 1.5 mL/5 g.

Abstract:Gellan gum is an important exopolysaccharide produced with Sphingomonas paucimobilis,which is widely used in foods,pharmaceutical industries.The fermentation kinetic model of cell growth,gellan gum production and substrate utilization were established and the kinetic parameters of the model were determined.The kinetic model provides a good description of gellan gum fermentation,in which the deviations between model calculating results and experimental data are within 5%.Furthermore it shows a good prediction of the fermentation profile including substrate consumption,cell growth and gellan gum production under different initial glucose mass concentration.The deviation between actual values and predicted values is less than 10%,which indicates that the model constructed has good applicability and significantly instructive meaning to industrial-scale gellan gum fermentation.

Abstract:Interconversions of phosphoinositides play a pivotal role during phagocytosis and at the subsequent stages of phagosomal maturation into the phagolysosome. Several model systems have been used to study the role of phosphoinositides in phagosomal membrane remodelling. These include phagosomes formed by inanimate objects such as latex beads, or pathogenic bacteria, e.g. Mycobacterium tuberculosis. The latter category provides naturally occurring tools to dissect membrane trafficking processes governing phagolysosome biogenesis. M. tuberculosis persists in infected macrophages by blocking Rab conversion and affecting Rab effectors. One of the major Rab effectors involved in this process is the type III phosphatidylinositol 3-kinase hVPS34. The lipid kinase hVPS34 and its enzymatic product PtdIns3P are critical for the default pathway of phagosomal maturation into phagolysosomes. Mycobacteria block PtdIns3P production and thus arrest phagosomal maturation. PtdIns3P is also critical for the process of autophagy, recently recognized as an effector of innate immunity defenses. Induction of autophagy by pharmacological, physiological, or immunological means, overcomes mycobacterial phagosome maturation block in a PtdIns3P generation dependent manner and eliminates intracellular M. tuberculosis. PtdIns3P and PtdIns3P-dependent processes represent an important cellular nexus where fundamental trafficking processes, disease causing host-pathogen interactions, and innate and adaptive immunity defense mechanisms meet.

Abstract:The optimization of fermentation medium for alpha-galactosidase production by Aspergillus foetidus ZU-G1 was investigated in shaker flask fermentation. A one-factor-at-a-time experiment was used to screen the preferable nutriment (carbon sources, nitrogen sources, and essential elements) for alpha-galactosidase production. A fractional factorial design was used to screen the main 5 factors, soybean meal, wheat bran, KH2PO4, FeSO4.7H(2)O, and the medium initial pH, that affected the production of alpha-galactosidase. The central composite experimental design was further adopted to derive a statistical model for optimizing the composition of the fermentation medium. The experimental results showed that the optimum fermentation medium for alpha-galactosidase production by Aspergillus foetidus ZU-G1 was composed of 3.2% soybean meal (w/v), 2% wheat bran (w/v), 0.1% KH2PO4 (w/v), and 0.05% FeSO4.7H(2)O (w/v); initial medium pH was 6.31. The results further predicted that alpha-galactosidase activity reached 64.75 U/mL after 96-h incubation in this medium, which was approximately 7 times higher than that incubated in the nonoptimized medium. The time course of alpha-galactosidase production in the optimized medium composition was also carried out to validate the model.

Abstract:An integrative laboratory study addressing fundamentals of enzyme catalysis and their application to reactors operation and modeling is presented. Invertase, a β-fructofuranosidase that catalyses the hydrolysis of sucrose, is used as the model enzyme at optimal conditions (pH 4.5 and 45 °C). The experimental work involves 3 h of laboratory time for each student per week over four weeks. Students, organized in laboratory sessions of three groups with 3 or 4 students per group, work in a collaborative manner to obtain a set of replicates of initial reaction rate for different substrate concentrations, using both free and calcium alginate-immobilized Saccharomyces bayanus cells containing biologically active invertase. The results are shared by all of the groups for statistical data treatment, calculation of Michaelis–Menten kinetic parameters, and modeling of three types of classical reactors, a Continuous Stirred Tank Reactor (CSTR), a Plug Flow Reactor (PFR), and a Fluidized Bed Reactor (FBR), operating in continuous mode with immobilized invertase. For each reactor, the experimental conversion profile as a function of the feed flow rate is compared to predicted profiles based upon the kinetic parameters obtained.

Abstract:An unstructured kinetic model for xanthan production is described and fitted to experimental data obtained in a stirred batch reactor. The culture medium was composed of several nitrogen sources (soybean hydrolysates, ammonium and nitrate salts) consumed sequentially. The model proposed is able to describe this sequential consumption of nitrogen sources, the consumption of inorganic phosphate and carbon, the evolution of biomass, and production of xanthan. The parameter estimation has been performed by fitting the kinetic model in differential form to experimental data. Runs of the model for simulating xanthan gum production as a function of the initial concentration of inorganic phosphate have shown the positive effect of phosphate limitation on xanthan yield, though diminishing rates of production. The model was used to predict the kinetic parameters for a medium containing a 2-fold lower initial phosphate concentration. When tested experimentally, the measured fermentation parameters were in close agreement with the predicted model values, demonstrating the validity of the model.

Abstract:The kinetic models for elastase batch cultivation in a 70 L fermentor by Bacillus sp. EL31410 were discussed. Based on the Logistic and Luedeking-Piret equation, the mathematical dynamic models were proposed for cell growth, product formation and substrate consumption. The parameters of the kinetic models were established with experimental data. The models for cell growth, elastase formation and glucose consumption model were described by the following equations:dx/dt=0.180(1-x/9.77)x,dp/dt=-dx/dt=4.278x and -ds/dt=0.03(dx/dt)-7.53x, respectively. It showed that the predict values of the models and experimental data were in a good agreement.
Response Surface Method for the Optimization of Bacillus sp. strain ZEID-14 Growth on Acrylamide as a Nitrogen Source

Abstract:Acrylamide contamination in food is mainly from raw material of plant-based origin. Acrylamide biodegradation by soil bacteria is an important remediation process. Bacillus sp. strain ZEID-14, which had previously been identified and exhibited the ability to break down amides, was examined further to determine the crucial parameters that contribute to the optimum growth of acrylamide. The Box-Behnken design was used to optimize the previously identified three significant components (pH, incubation time and acrylamide concentration). The model was supported by the diagnostic plots including the half-normal, Cook's distance, leverage vs runs, residual vs runs, Box-Cox, DFFITS, and DFBETAS. Predicted optimal conditions were determined using "Numerical Optimisation" toolbox of the Design Expert software. Two optimal conditions were tested. The model predicted a maximum growth of 10.686 (95% C.I., 10.458 to 10.913) which was verified through experimental results with a growth of 11.257 (95% C.I., 11.051 to 11.462) with the actual results being near to the predicted values but was significantly higher than the predicted values. The second numerical optimization gave a solution with a predicted maximum growth of 9.305 Log CFU/mL (95% C.I. from 9.011 to 9.614) which was verified through experimental results with a growth of 9.978 Log CFU/mL (95% C.I. from 9.830 to 10.126) with the actual results were also significantly higher than the predicted values. The RSM exercise gave far better growth on acrylamide than OFAT with a higher response of about 2 log CFU/mL unit indicating the utility of RSM over OFAT in the optimization of growth of this bacterium on acrylamide.

Abstract:Sucrose hydrolysis was carried out in a constant-volume batch reactor, using recombinant Pichia pastoris BfrA4X whole cells expressing Thermotoga maritima invertase, entrapped in calcium alginate beads. The kinetics of the enzymatic hydrolysis of sucrose by the biocatalyst was examined at substrate concentrations ranging between 0.03 M and 2.04 M. The reaction rate increases until 0.31 M after which the reaction velocity was constant until 1.16 M, above this concentration, the reaction rate decreases with increasing sucrose concentration. The experimental data obtained with two weight of the biocatalyst were incorporated into two kinetic models to predict the reaction time needed for sucrose hydrolysis. One model was applied for sucrose concentrations bellow 1.16 M while a second one could be used at inhibitory range between 1.46 and 2.04 M with a k value as function of initial sucrose concentration and biocatalyst weight.
Application of Response Surface Methodology for Optimization of Extracellular Glucoamylase Production by Candida guilliermondii

Abstract:Objective: Glucoamylase is among the most important enzymes in biotechnology. The present study aims to determine better conditions for growth and glucoamylase production by Candida guilliermondii and to reduce the overall cost of the medium using Box-Behnken design with one central point and response surface methodology. Methodology: Box-Behnken factorial design based on three levels was carried out to obtain optimal medium combination of five independent variables such as initial pH, soluble starch, CH4N2O, yeast extract and MgSO4. Forty one randomized mediums were incubated in flask on a rotary shaker at 105 rpm for 72 h at 30°C. Results: The production of biomass was found to be pH and starch dependent, maximum production when the starch concentration was 8 g L-1 and the initial pH was 6, while maximum glucoamylase production was found at 6.5 of initial pH, 4 g L-1 yeast extract and 6 g L-1 starch, whereas yeast extract and urea were highly significant, but interacted negatively. Box-Behnken factorial design used for the analysis of treatment combinations gave a second-order polynomial regression model with R2 = 0.976 for Biomass and R2 = 0.981 for glucoamylase. Conclusion: The final biomass and glucoamylase activity obtained was very close to the calculated parameters according to the p-values (p<0.001), the predicted optimal parameters were confirmed and provides a basis for further studies in baking additives and in the valuation of starch waste products.
Selection of Non-Mycotoxigenic Inulinase Producers in the Group of Black Aspergilli for Use in Food Processing

Abstract:Research background: Inulinases are used for fructooligosaccharide production and they are of interest for both scientific community and industry. Black aspergilli represent a diverse group of species that has use for enzyme production, in particular some species are known as potent inulinase producers. Finding new potential producers from the environment is as important as improving the production with known strains. Safe use of enzymes produced by aspergilli in food industry is placed ahead of their benefit for inulinase production. Experimental approach: Here we show a specific approach to finding/screening of newly isolated fungal inulinase producers that combines a newly developed screening method and an equally important assessment of the toxigenic potential of the fungus. In this study 39 black aspergilli collected from different substrates in Serbia were identified and assessed for inulinase production. Results and conclusions: The most common species were Aspergillus tubingensis (51.2%), followed by A. niger (23.1%), A. welwitschiae (23.1%) and A. uvarum (2.6%). The isolates for inulinase production were selected using a cheap and easy, fast and non-hazardous alternative inulinase screening test developed in this work. Enzymatic activity of selected inulinase-producing strains was confirmed spectrophotometrically. Since some A. niger and A. welwitschiae strains are able to produce mycotoxins ochratoxin A (OTA) and fumonisins (FB), the toxigenic potential of selected inulinase producers was assessed analytically and genetically. Fungal enzyme producer can be considered safe for use in food industry only after comparing the results of both approaches for investigating toxic potential, the direct presence of mycotoxins in the enzyme preparation (analytically) and the presence of mycotoxin gene clusters (genetically). In some strains the absence of OTA and FB production capability was molecularly confirmed by the absence of complete or critical parts of biosynthetic gene clusters, respectively. The two best inulinase producers and mycotoxin non-producers (without mycotoxin production capability as additional safety) were selected as potential candidates for further development of enzyme production. Novelty and scientific contribution: The presented innovative approach for the selection of potential fungal enzyme producer shows that only non-toxigenic fungi could be considered as useful in food industry. Although this study was done on local isolates, the approach is applicable globally.

Abstract:Introduction: The nematode species Meloidogyne incognita has been responsible for significant financial losses within the agricultural sector. Nematophagous bacteria, characterised by their extensive distribution and broad spectrum of hosts, exhibit remarkable efficacy as natural antagonists against nematodes. Sneb518 ( Clostridium beijerinckii ) fermentation broth displayed substantial biocontrol activity against M. incognita in previous research. Optimizing fermentation conditions is a fundamental technique for dramatically enhancing end product performance. There has been no such study conducted yet on enhancing the nematicidal activities of Sneb518 ( Clostridium beijerinckii ) fermentation using response surface methodology (RSM). Methods: The influence of strain Sneb518 fermentation media and conditions on nematicidal activity was examined using the three-factor technique and a Plackett-Burman design, and the interaction between various fermentation factors was examined using a Box-Behnken design. The present study employed response surface methodology (RSM) to examine and enhance the nematicidal activity of Sneb518 culture filtrates by identifying and optimising the influential components. Results: Glucose, peanut cake flour, and potassium chloride as carbon, nitrogen, and inorganic salts displayed considerably increased nematicidal potential in the present study. Furthermore, the corrected mortality of J2 ranged from 52.24% to 91.15% when utilizing the Box-Behnken design. These findings clearly support the application of RSM for medium optimization. Moreover, the outcomes of the validation experiment corresponded to the model predictions. Discussion: This research has enhanced the biocontrol ability of C. beijerinckii to control M. incognita and this research has led to the advancement of new biocontrol agents.

Adaptive neuro-fuzzy inference system (ANFIS), response surface method (RSM), and artificial neural network (ANN) were comparatively used in the simulation and modeling of the treatment process for improved biogas yield. Waste plantain peels were pretreated and used as substrate. FTIR and SEM results revealed that the pretreatment improved the substrate's desirable qualities. The amount of biogas yield was controlled by time, NaOH concentration, and temperature of the substrate pretreatment. Optimum pretreatment conditions obtained were a temperature of 102.7 °C, time of 31.7 min and NaOH concentration of 0.125 N. RSM, ANN, and ANFIS modeling techniques were proficient in simulating the biogas production, as evidenced by high R2values of 0.9281, 0.9850, and 0.9852, respectively. Furthermore, the values of the calculated error terms such as RMSE (RSM = 0.04799, ANN = 0.00969, and ANFIS = 0.00587) and HYBRID (RSM = 18.556, ANN = 0.803, and ANFIS = 0.0447) were low, indicating a satisfactory correlation between experimental and predicted values. Scrubbing of the biogas with caustic soda and activated charcoal increased the methane content to 94 %. The kinetics of the cumulative biogas yield were best fit with the Logistics and Modified Logistics models. The low C/N ratio in addition to the presence of potassium, nitrogen, and phosphorus suggested that the spent plantain peel slurry can be utilized as an agricultural fertilizer in crop production. The observations of this study therefore recommends the pre-treatment of biodigestion substrates as a key means to enhance beneficiation of methane production.

Abstract:Bioethanol production from renewable biomass sources has garnered significant interest due to its potential as a sustainable alternative to fossil fuels. In this study, we investigated the optimization of bioethanol production from molasses, a by-product of the sugar production process using Saccharomyces cerevisiae through Response Surface Methodology (RSM). Initially, the fermentation process was optimized using RSM, considering four independent variables: substrate concentration, pH, temperature, and fermentation time. Subsequently, the effects of these variables on bioethanol yield were evaluated, and a quadratic model was developed to predict the optimum conditions. Analysis of variance (ANOVA) indicated a high coefficient of determination (R 2 ) for the model, suggesting its adequacy for prediction. The optimized conditions for bioethanol production were determined as follows: substrate concentration of 200 g L −1 , pH of 5.0, temperature of 30 °C and fermentation time of 72 h. Under these conditions, the predicted bioethanol yield was 84%. Overall, this study demonstrates the successful application of RSM for optimizing bioethanol production from molasses using S. cerevisiae , highlighting its potential as a promising feedstock for biofuel production.
