Abstract:Magnesium (Mg) and its alloys posse’s great potential for the application of biodegradable medical implants. It is due to their unique properties like low density and elastic modulus, good biocompatibility, etc. But still there are many challenges for Mg alloy based implants. Due to rapid degradation of Mg and its alloys in biological fluid, it loses its mechanical integrity and fails to perform before the complete healing of bone fracture (in orthopedics application) or removal of plaque in arteries (in case of vascular implants). Using suitable alloying elements mechanical strength and corrosion resistance of Mg-alloys can be enhanced but cytotoxicity and long term inflammatory consequences of these elements are the major concern. Further modifying the surface characteristics of Mg-alloy through various surface coating, machining, mechanical working, etc., corrosion behaviour can be manipulated. In this field of biodegradable implants, the various opportunities are yet to be explored in detail to improve the clinical performance of Mg alloy implants for orthopedics and vascular applications. This review paper summarises the various challenges and opportunities in design and development of biodegradable Mg alloy implants.

Abstract:In biomedical applications, the conventionally used metallic materials, including stainless steel, Co-based alloys and Ti alloys, often times exhibit unsatisfactory results such as stress shielding and metal ion releases. Secondary surgical operation(s) usually become inevitable to prevent long term exposure of body with the toxic implant contents. The metallic biomaterials are being revolutionized with the development of biodegradable materials including several metals, alloys, and metallic glasses. As such, the nature of metallic biomaterials are transformed from the bioinert to bioactive and multi-biofunctional (anti-bacterial, anti-proliferation, anti-cancer, etc.). Magnesium-based biomaterials are candidates to be used as new generation biodegradable metals. Magnesium (Mg) can dissolve in body fluid that means the implanted Mg can degrade during healing process, and if the degradation is controlled it would leave no debris after the completion of healing. Hence, the need for secondary surgical operation(s) for the implant removal could be eliminated. Besides its biocompatibility, the inherent mechanical properties of Mg are very similar to those of human bone. Researchers have been working on synthesis and characterization of Mg-based biomaterials with a variety of composition in order to control the degradation rate of Mg since uncontrolled degradation could result in loss of mechanical integrity, metal contamination in the body and intolerable hydrogen evolution by tissue. It was observed that the applied methods of synthesis and the choice of components affect the characteristics and performance of the Mg-based biomaterials. Researchers have synthesized many Mg-based materials through several synthesis routes and investigated their mechanical properties, biocompatibility and degradation behavior through in vitro, in vivo and in silico studies. This paper is a comprehensive review that compiles, analyses and critically discusses the recent literature on the important aspects of Mg-based biomaterials.

Abstract:Magnesium-based alloys exhibit biodegradable, biocompatible and excellent mechanical properties which enable them to serve as ideal candidate biomedical materials. In particular, their biodegradable ability helps patients to avoid a second surgery. The corrosion rate, however, is too rapid to sustain the healing process. Alloying is an effective method to slow down the corrosion rate. However, currently magnesium alloys used as biomaterials are mostly commercial alloys without considering cytotoxicity from the perspective of biosafety. This article comprehensively reviews the status of various existing and newly developed degradable magnesium-based alloys specially designed for biomedical application. The effects of critical alloying elements, compositions, heat treatment and processing technology on the microstructure, mechanical properties and corrosion resistance of magnesium alloys are discussed in detail. This article covers Mg–Ca based, Mg–Zn based, Mg–Sr based, Mg–RE based and Mg–Cu-based alloy systems. The novel methods of fabricating Mg-based biomaterials and surface treatment on Mg based alloys for potential biomedical applications are summarized.

Abstract:The development of biodegradable implants is certainly intriguing, and magnesium and its alloys are considered significant among the various biodegradable materials. Nevertheless, the fast degradation, the generation of a significant amount of hydrogen gas, and the escalation in the pH value of the body solution are significant barriers to their use as an implant material. The appropriate approach is able to solve this issue, resulting in a decrease the rate of Mg degradation, which can be accomplished by alloying, surface adjustment, and mechanical treatment. Surface modification is a practical option because it not only improves corrosion resistance but also prepares a treated surface to improve bone regeneration and cell attachment. Metal coatings, ceramic coatings, and permanent polymers were shown to minimize degradation rates, but inflammation and foreign body responses were also suggested. In contrast to permanent materials, the bioabsorbable polymers normally show the desired biocompatibility. In order to improve the performance of drugs, they are generally encapsulated in biodegradable polymers. This study summarized the most recent advancements in manufacturing polymeric coatings on Mg alloys. The related corrosion resistance enhancement strategies and future potentials are discussed. Ultimately, the major challenges and difficulties are presented with aim of the development of polymer-coated Mg-based implant materials.

Abstract:Medical application materials must meet multiple requirements, and the designed implant must mimic the bone structure in shape and support the formation of bone tissue (osteogenesis). Magnesium (Mg) alloys, as a "smart" biodegradable material and as "the green engineering material in the twenty-first century", have become an outstanding bone implant material due to their natural degradability, smart biocompatibility, and desirable mechanical properties. Magnesium is recognised as the next generation of orthopaedic appliances and bioresorbable scaffolds. At the same time, improving the mechanical properties and corrosion resistance of magnesium alloys is an urgent challenge to promote the application of magnesium alloys. Nevertheless, the excessively quick deterioration rate generally results in premature mechanical integrity disintegration and local hydrogen build-up, resulting in restricted clinical bone restoration applicability. The condition of Mg bone implants is thoroughly examined in this study. The relevant approaches to boost the corrosion resistance, including purification, alloying treatment, surface coating, and Mg-based metal matrix composite, are comprehensively revealed. These characteristics are reviewed to assess the progress of contemporary Mg-based biocomposites and alloys for biomedical applications. The fabricating techniques for Mg bone implants also are thoroughly investigated. Notably, laser-based additive manufacturing fabricates customised forms and complicated porous structures based on its distinctive additive manufacturing conception. Because of its high laser energy density and strong controllability, it is capable of fast heating and cooling, allowing it to modify the microstructure and performance. This review paper aims to provide more insight on the present challenges and continued research on Mg bone implants, highlighting some of the most important characteristics, challenges, and strategies for improving Mg bone implants.

Abstract:Magnesium (Mg) has attracted considerable attention as a biodegradable material for medical implants owing to its excellent biocompatibility, mitigating long-term toxicity and stress shielding. Nevertheless, challenges arise from its rapid degradation and low corrosion resistance under physiological conditions. To overcome these challenges, titanium (biocompatibility and corrosion resistance) has been integrated into Mg. The incorporation of titanium significantly improves mechanical and corrosion resistance properties, thereby enhancing performance in biological settings. Mg–Ti alloys are produced through mechanical alloying and spark plasma sintering (SPS). The SPS technique transforms powder mixtures into bulk materials while preserving structural integrity, resulting in enhanced corrosion resistance, particularly Mg80-Ti20 alloy in simulated body fluids. Moreover, Mg–Ti alloy revealed no more toxicity when assessed on pre-osteoblastic cells. Furthermore, the ability of Mg–Ti-based alloy to create composites with polymers such as PLGA (polylactic-co-glycolic acid) widen their biomedical applications by regulating degradation and ensuring pH stability. These alloys promote temporary orthopaedic implants, offering initial load-bearing capacity during the healing process of fractures without requiring a second surgery for removal. To address scalability constraints, further research is necessary to investigate additional consolidation methods beyond SPS. It is essential to evaluate the relationship between corrosion and mechanical loading to confirm their adequacy in physiological environments. This review article highlights the importance of mechanical characterization and corrosion evaluation of Mg–Ti alloys, reinforcing their applicability in fracture fixation and various biomedical implants.

Abstract:In recent years, materials science research based on magnesium (Mg) alloys has increased significantly due to their notable advantages over traditional metals. However, magnesium alloys are susceptible to excessive degradation and subsequent disruption of mechanical integrity; this phenomenon limits the utility of these materials. Mg alloys can thus be combined with other materials to form composites for medical applications. The present article describes key advances in and considerations for the development of biodegradable Mg-based composites (BMCs). The primary characteristics of these materials include their controllable degradation rates, tunable mechanical properties, adjustable structures to promote tissue repair, improved biocompatibility, and added functionality according to the purpose of the applications. Here we provide an overview of the current research on and development status of BMCs for biomedical materials, including the present limitations and challenges of their use. Finally, this paper comprehensively discusses the most promising directions of future development for these materials.

Magnesium (Mg)-based materials are a new generation of alloys with the exclusive ability to be biodegradable within the human/animal body. In addition to biodegradability, their inherent biocompatibility and similar-to-bone density make Mg-based alloys good candidates for fabricating surgical bioimplants for use in orthopedic and traumatology treatments. To this end, nowadays additive manufacturing (AM) along with three-dimensional (3D) printing represents a promising manufacturing technique as it allows for the integration of bioimplant design and manufacturing processes specific to given applications. Meanwhile, this technique also faces many new challenges associated with the properties of Mg-based alloys, including high chemical reactivity, potential for combustion, and low vaporization temperature. In this review article, various AM processes to fabricate biomedical implants from Mg-based alloys, along with their metallic microstructure, mechanical properties, biodegradability, biocompatibility, and antibacterial properties, as well as various post-AM treatments were critically reviewed. Also, the challenges and issues involved in AM processes from the perspectives of bioimplant design, properties, and applications were identified; the possibilities and potential scope of the Mg-based scaffolds/implants are discussed and highlighted.

Abstract:Magnesium (Mg) or its alloys are widely tested as potential orthopedic implants, particularly as biodegradable alloys for fracture fixation due to their mechanical properties are close to those of bone. Currently, available Mg or its alloys are confronted with challenges in passing regulatory biosafety tests prior to clinical trials due to its fast degradation and associated degradation products. The degradation of Mg is accompanied by the release of Mg ions, the rise of pH and osmolality in surrounding environments. According to the standard of ISO 10993 Part 13, the pH value shall be appropriate to the site of intended use maintaining in an appropriate range. Approaches to overcome these challenges include the selection of adequate alloying elements, proper surface treatment techniques and control of the degradation rate of Mg or its alloys developed as orthopedic implants. To date, Mg or its alloy-based bone implants have not yet been widely used in clinical applications as medical implants. This review critically summarized published methods to improve the corrosion resistance of Mg and its alloys. The current progress on in vitro cytotoxicity and in vivo biocompatibility properties of these metals was also reviewed. This review aimed to provide a reference for further research and development (R&D) of biodegradable Mg and its alloys with regard to the evaluation of their corrosion process and biocompatibility and facilitation of their translation to clinical applications.

In this article, we review the latest research progress in biodegradable Mg alloys, including research on high-performance Mg alloys, bioactive coatings and actual or potential clinical applications of Mg alloys. Finally, we review the research and development direction of biodegradable Mg alloys. This article has a guiding significance for future development and application of high-performance biodegradable Mg alloys, promoting the future advancement of the magnesium alloy research field, especially in biomedicine.

Abstract:Magnesium-based biomaterials hold remarkable promise for various clinical applications, offering advantages such as reduced stress-shielding and enhanced bone strengthening and vascular remodeling compared to traditional materials. However, ensuring the quality of preclinical research is crucial for the development of these implants. To achieve implant success, an understanding of the cellular responses post-implantation, proper model selection, and good study design are crucial. There are several challenges to reaching a safe and effective translation of laboratory findings into clinical practice. The utilization of Mg-based biomedical devices eliminates the need for biomaterial removal surgery post-healing and mitigates adverse effects associated with permanent biomaterial implantation. However, the high corrosion rate of Mg-based implants poses challenges such as unexpected degradation, structural failure, hydrogen evolution, alkalization, and cytotoxicity. The biocompatibility and degradability of materials based on magnesium have been studied by many researchers in vitro; however, evaluations addressing the impact of the material in vivo still need to be improved. Several animal models, including rats, rabbits, dogs, and pigs, have been explored to assess the potential of magnesium-based materials. Moreover, strategies such as alloying and coating have been identified to enhance the degradation rate of magnesium-based materials in vivo to transform these challenges into opportunities. This review aims to explore the utilization of Mg implants across various biomedical applications within cellular (in vitro) and animal (in vivo) models.

Abstract:Mg-based alloys as revolutionary implantable biomaterials have increasingly attracted considerable attention, owing to their biodegradability in vivo and beneficial effects on biological systems. The degradation process and products of Mg-based alloys have been reported to exhibit significant biological effects on host-tissue responses. However, these effects have not yet been fully understood. This review systemically summarizes and analyses the current understandings and recent research progress in this area. The primary focal points are the biological effects and related mechanisms associated with the degradation behaviour of Mg-based alloys. The biological impacts of the degradation products are elucidated and the arguable or controversial issues are also discussed, providing a pathway toward a greater understanding of the biological implications of Mg-based alloys. Furthermore, based on these biological implications, the restorative potential of Mg-based alloys for applications in tissue repair and regeneration is summarized. Finally, outlooks on biosafety evaluation and design strategies for Mg-based alloy implants are briefly discussed.

Magnesium (Mg) based materials show great promise as temporary implant applications owing to their biocompatibility and biodegradability. These characteristics remove the risk of subsequent surgery to extract the implant once the process of bone tissue healing is finished. Additionally, its density and elastic modulus are near to those of natural bone, thereby reducing the stress-shielding effect. Mg is mostly recognized for its osteoconductive abilities, which implies that it encourages the generation of fresh bone tissue. It also has antimicrobial properties, which lower the possibility of infections leading to implant failure. Moreover, the rapid bio-corrosion of pure Mg in the presence of physiological fluids is a serious concern. The implant's mechanical integrity deteriorates as a result of this corrosion before the surrounding tissue has completely recovered. To address these issues, this review focused on approaches, including alloying, the creation of composites, and surface coating, which can increase their biomechanical and bio-corrosion properties. In vitro analysis of biomechanical and bio-corrosion characteristics of newly manufactured Mg-based implant material is presented in this article. In addition to this application, a list of approved devices made from Mg-based material is highlighted. Furthermore, the present challenges and prospects for future research are also discussed.

Abstract:The combination of high strength, light weight, and natural biodegradability renders magnesium (Mg)-based alloys promising in orthopedic implants and cardiovascular stents. Being metallic materials, Mg and Mg alloys made for scaffolds provide the necessary mechanical support for tissue healing and cell growth in the early stage, while natural degradation and reabsorption by surrounding tissues in the later stage make an unnecessarily follow-up removal surgery. However, uncontrolled degradation may collapse the scaffolds resulting in premature implant failure, and there has been much research in controlling the degradation rates of Mg alloys. This paper reviews recent progress in the design of novel Mg alloys, surface modification and corrosion mechanisms under different conditions, and describes the effects of the structure, composition, and surface conditions on the degradation behavior in vitro and in vivo. Statement of significance: Owing to their unique mechanical properties, biodegradability, biocompatibility, Mg based biomaterials are becoming the most promising substitutes for tissue regeneration for impaired bone, vascular and other tissues because these scaffolds can provide not only ideal space for the growth and differentiation of seeded cells but also enough strength before the formation of normal tissues. The most important is that these scaffolds can be fully degraded after tissue regeneration, which can satisfy the increasing demand for better biomedical devices and functional tissue engineering biomaterials in the world. However, the rapid degradation rate of these scaffolds restricts the wide application in clinic. This paper reviews recent progress on how to control the degrdation rate based on the relevant corrosion mechanisms through the design of porous structure, phase structure, grains, and amorphous structure as well as surface modification, which will be beneficial to the better understanding and functional design of Mg-based scaffolds for wide clinical applications in tissue reconstruction in near futures.

Abstract:In recent years, much progress has been made on the development of biodegradable magnesium alloys as "smart" implants in cardiovascular and orthopedic applications. Mg-based alloys as biodegradable implants have outstanding advantages over Fe-based and Zn-based ones. However, the extensive applications of Mg-based alloys are still inhibited mainly by their high degradation rates and consequent loss in mechanical integrity. Consequently, extensive studies have been conducted to develop Mg-based alloys with superior mechanical and corrosion performance. Herein, the critical criteria and the state-of-the-art strategies for controlling the degradation process of Mg alloys are reported. Furthermore, future developments of biodegradable Mg and its alloys systems with satisfactory specifications for clinical trials and deployment, are discussed. This review aims to provide information to materials scientists and clinical practitioners in the context of developing practical biodegradable Mg alloys.

Abstract:Medical implants made of biodegradable materials could be of advantage for temporary applications such mechanical support during bone-healing or as vascular stents to keep blood vessels open. After completion of the healing process the implant would disappear, avoiding long-term side effects or the need for surgical removal. Various corrodible metal alloys based on magnesium, iron or zinc have been proposed as sturdier and potentially less inflammatory alternative to degradable organic polymers, in particular for load-bearing applications. Despite the recent introduction of magnesium-based screws the remaining hurdles to routine clinical applications are still challenging, such as limiting mechanical material characteristics or unsuitable corrosion characteristics. Here, salient features and clinical prospects of currently investigated biodegradable implant materials are summarized with a main focus on magnesium alloys. A mechanism of action for the stimulation of bone growth due to the exertion of mechanical force by magnesium corrosion products is discussed. To explain divergent in vitro and in vivo effects of magnesium a novel model for bacterial biofilm infections is proposed which predicts crucial consequences antibacterial implant strategies.

Abstract:Magnesium (Mg) based biodegradable materials are a new generation orthopedic implant materials that are intended to possess same mechanical properties as that of bone. Mg alloys are considered as promising substitutes to permanent implants due to their biodegradability in the physiological environment. However, rapid corrosion rate is one of the major constraints of using Mg alloys in clinical applications in spite of their excellent biocompatibility. Approaches to overcome the limitations include the selection of adequate alloying elements, proper surface treatment, surface modification with coating to control the degradation rate. This review focuses on current advances on surface engineering of Mg based biomaterials for biomedical applications. The review begins with a description of corrosion mechanism of Mg alloy, the requirement for appropriate surface functionalization/coatings, their structure-property-performance relationship, and suitability for biomedical applications. The control of physico-chemical properties such as wettability, surface morphology, surface chemistry, and surface functional groups of the coating tailored by various approaches forms the pivotal part of the review. Chemical surface treatment offers initial protection from corrosion and inorganic coating like hydroxyapatite (HA) improves the biocompatibility of the substrate. Considering the demand of ideal implant materials, multilayer hybrid coatings on Mg alloy in combination with chemical pretreatment or inorganic HA coating, and protein-based polymer coating could be a promising technique to improve corrosion resistance and promote biocompatibility of Mg-based alloys.

Abstract:In recent years, a new generation of biodegradable metallic materials, magnesium alloys, has been called a revolutionary material for biomedical applications (i.e. in orthopedics applications as a bone-implant material), thanks to the reasonable strength (similar to bone tissue, compared to available metallic alloys) and high biocompatibility of magnesium and its alloys. However, pure magnesium can corrode too quickly in the physiological pH (7.4–7.6) and high chloride environment of the physiological system and therefore lose their mechanical integrity before tissues have sufficiently. Engineering approach to this challenge (high corrosion rate of Mg) can be (i) alloying of element additions, (ii) surface treatment and (iii) development of metal (magnesium) matrix composites (MMCs). Magnesium-based composites, as bio-materials, can provide a combination of unique characteristics including adjustable mechanical properties (i.e. tensile strength, elastic modulus, ductility) and corrosion resistance. This is the main advantage of magnesium-based composites as compared with alloying and surface treatment approaches. Here, the matrix materials are biomedical magnesium alloys based on Mg–Zn, Mg–Ca and Mg–REE alloy systems (REE stands for rare earth elements including yttrium, Y, cerium, Ce, lanthanum, La). The reinforcement phases are mainly based on hydroxyapatite (HAP), calcium polyphosphate (CPP), and β-tricalcium phosphate (β-TCP) particles, and hybrid HAP + β-TCP particles. In this paper a comprehensive review is provided on different grades of biodegradable magnesium matrix composites, with focus on their mechanical properties.
