Abstract:In this study, alkaline metalloprotease gene 1067 was cloned from Planococcus halotolerans SCU63T and heterologously expressed in Bacillus subtilis SCK6. Using Luria Bertani (LB) broth medium as the initial medium, the optimal medium was obtained through a series of fermentation and culture optimization (g/L): yeast extract (10), soybean powder (15), urea (20), potassium chloride (6.7), calcium chloride (13.3), NaCl (10). On the basis of the optimal medium, the highest enzymatic activity of 1259.21 U/mL could be obtained by culturing at 30°C for 40 h. with pH 8, inoculation amount of 4% and filling amount of 50 mL. EDTA inhibits protease activity and PMSF promotes it, indicating that it was a metalloprotease rather than a serine protease. The optimum reaction temperature of the protease is 70°C, and the optimum pH is 9. The metal ions Zn 2+, Co2+ and surfactant β-ME, Tween 80 can improve the activity of the protease. The results of unhairing, SEM and staining show that this metalloprotease can completely dehair goatskin. Compared with the conventional chemical method, the goatskin after enzymatic unhairing has softer texture and smoother surface, and there is no obvious damage to the goatskin.

Abstract:The fermentation conditions for the production of neutral protease by Bacillus subtilis were investigated using single factor and orthogonal design experiments.The results showed that the optimal fermentation medium in the flasks contained 40 g/L glycerin,30 g/L soymeal,10 g/L bran,4 g/L Na2HPO4,and 0.3 g/L K2HPO4.The optimal fermentation conditions in the shaker were culturing with 6％ inoculum amount at 30 ℃ and 250 r/min.Under this condition,the enzyme activity reached 6 082.7U/mL.To further verify the optimized fermentation condition,the glycerol was fed and batch fermentation was carried out in 15 L fermenter.The enzyme activity reached 11 871 U/mL.

Abstract:The current study focused on the substitution of toxic Na2S with bio-enzyme G3726, a novel serine protease heterologously expressed by Bacillus subtilis strain SCK6. This protease was purified to electrophoretic homogeneity with 39.43 % recovery, 7.03-fold purification and an estimated molecular mass of 36 kDa. The maximal activity was achieved at pH 8, 75 °C and 40 g L−1 NaCl, and more than 70 % of the activity was retained across the broad salinity range of 0–200 g L−1. G3726 was activated by Sr2+ and Ca2+, displayed considerable stability to benzene and ethanediol, and exhibited excellent compatibility with Tweens (20 and 80) and Triton X-100. Simultaneously, the 3D structure of G3726 was constructed by homology modeling, and interacted well with the cementing substances glycoprotein and proteoglycan in the hair pores. Moreover, G3726 showed no collagenase activity, and could thoroughly dehair goatskin within 4 h without any damage, which was better than industrial enzyme AS1.398 and chemical Na2S. As compared to Na2S, G3726 enzymatic dehairing showed reduction in BOD5, COD and TSS by 53.81, 52.78 and 65.52 %, respectively, endowing the leather better physical properties. These outstanding findings indicate that protease G3726 may have application in dehairing for environment-friendly leather processing.

Neutral Cellulase Production by Recombinant Pichia pastoris

Abstract:Neutral cellulase has important applications in denim biofinishing.A Pichia pastoris transformant with high activity of neutral cellulase was screened from 6 recombination transformants by fermentation experiment.The research on the neutral cellulase production by the P.pastoris recombinant was conducted under shaking flask condition,and the results were shown as follows: medium volume 30mL in 250mL Erlenmeyer flask,the initial pH 4.8 and uncontrolled during the fermentation process,inoculum volume 10% and methanol addition 1% every 24h which demonstrated an obvious enhancing effect.The above conditions allowed the activity of the neutral cellulase to reach 1144U/mL.The results from the study on enzymatic properties indicate that the appropriate catalysis conditions for the neutral cellulase from the P.pastoris transformant were pH 6.0 and temperature 50℃,and that Zn~2+ and Mg~2+ were activators of the enzyme but Fe~2+,Fe~3+ and Cu~2+ were inhibitors.The outcomes of denim biofinishing test reveal that the neutral cellulase from the P.pastoris recombinant could significantly reduce the backstaining of indigo onto denim and has a promising potential in industrial application.
Characterization and Pilot-Scale Application of the ZMS-2 Serine Protease with Novel Properties for the Eco-friendly Leather Processing

Abstract:Microbial alkaline proteases are dominating the global enzyme market with a share of over 65% due to their multifarious catalytic potentials. Hence, production of proteases with novel properties of commercial significance is highly desirable to meet the global enzyme demand. Here, we report the purification, characterization, and pilot-scale application of a serine protease from the desert soil bacterium Bacillus subtilis ZMS-2 with novel properties as dehairing agent in leather processing. The enzyme was purified 16.5-fold with a specific activity of 1543.5 U mg-1 and recovery percentage of 33.6% using ammonium sulfate precipitation, ion exchange, and gel filtration chromatography. The purified enzyme was characterized as a metal ion-, surfactant-, and denaturant-compatible alkaline serine protease having a molecular weight of 36.1 kDa with an optimum activity at pH 8.5 and 60 °C. The catalytic activity of the enzyme was enhanced by Zn+2 (204%), Ag+ (110%), H2O2 (123%), Triton X-100 (110%), iso-octane (109%), chloroform (110%), ethanol (105%), ethyl acetate (110%), and acetonitrile (128%). During pilot-scale applications, the optimum condition was found to be a combination of enzyme (1.5%, 460 U mL-1), sodium sulfide (2%), and calcium hydroxide (lime) (3%). Under this condition, the time required for complete dehairing was 90 min. Chemoenzymatically processed skins exhibited better physical properties than chemically processed skin, including tensile strength (16.35 ± 6.68 N/mm), ball burst (452.88 ± 6.06 N/mm), percent elongation (38.85 ± 1.06 N), tear strength (50.16 ± 4.42 N/mm), and softness (6.5 mm). Electron microscopy analysis of the treated skin showed complete removal of hairs with roots, confirming the keratin specificity of the enzyme. Moreover, the enzyme-assisted dehairing process reduced chemical oxygen demand (COD), biochemical oxygen demand (BOD), total dissolved solids (TDS), and total suspended solids (TSS) by 68, 77, 34, and 39%, respectively. Thus, the alkaline serine protease from B. subtilis ZMS-2 is a potential dehairing agent for the eco-friendly processing of animal skins on industrial scales.

Abstract:Among the microbial enzymes protease and amylase are the most valuable enzymes which have been has diversified applications and used extensively because of their capabilities in the degradation of organic wastes, application in biofuels, agricultural, pharmaceuticals, chemical and biotechnological industries. The aim of the current research work was the purification, characterization and application of alkaline proteases extracted from Bacillus cereus AUST-7. Various concentrations of ammonium sulphate were applied for enzyme precipitation. Sephadex-G 100 was used in FPLC system for separation of protease from other proteins. SDS-PAGE was used to measure the molecular weight of required alkaline protease. Relative activities were determined against different pH, temperature, and incubation period to measure the enzymes activity. Stability of pH, temperature and various metal ions and inhibiter were also studied. Purified enzymes were applied on the goat skin to explore the dehairing efficacy. A 6.5 purification fold and 1163.50 U/mg of specific activity were obtained at 70% saturation and 35. 91 purification fold and 8902 U/mg of specific activity were observed after FPLC separation. The 35 kDa molecular size of protease enzyme was exhibited on the SDS-PAGE. The purified enzyme was stable at pH 10, temperature 55 °C and 35 min of incubation period. The purified enzyme was found to be stable at pH 8–11, thermo-stability at 50 °C and phenyl methyl sulphonyl fluoride (PMSF) and di-isopropyl fluorophosphates (DFP) inhibited the enzyme activity. The enzyme has good potential as dehairing agent in leather industries.

Abstract:In this study various Aspergillus strains were explored for the production of protease using variety of agricultural byproducts as substrate. Aspergillus viridi was found to be the best among the selected strains with production of 83.0 U/mL of protease when grown on casein broth medium. Various agricultural byproducts were utilized for the growth of A. viridi and for the maximum protease production. Optimal production of enzyme 172.6 U/mL was recorded when PDB medium was supplemented with 3% banana peels powder as compared to 90.7, 84.7, 71.5, 68.7 and 47 U/mL when PDB medium was supplemented with pomegranate peels (3%), mustard oil cake (3%), arabic gum sawdust (2%), linseed oil cake (2%) and rosewood sawdust (3%) respectively. A. viridi could produce 172.6 U/mL when grown on 3% banana peels as sole substrate for growth. The protease production was increased with optimization of substrate water ratio (1:3), incubation time (72 h), inoculum size (2 mL), pH (8) and temperature (35 °C) from 172.6 to 178.1, 180.2, 182.9, 188.7 and 193.1 U/mL. Under optimal conditions, A. viridi produced 202.8 U/mL using agricultural byproducts as sole substrate. The strong potential of protease to remove hair from skin makes this enzyme a suitable candidate for its utilization in leather industry.

Abstract:In the current work, a novel thermophilic serine protease gene (P3862) from Ornithinibacillus caprae L9T was functionally expressed in Bacillus subtilis SCK6. The monomeric enzyme of about 29 kDa was purified to homogeneity with 43.91% of recovery and 2.81-folds of purification. Characterization of the purified protease revealed the optimum activity at pH 7 and 65 °C. The protease exhibited excellent activity and stability in the presence of Na+, Mg2+, Ca2+, ethanediol, n-hexane, Tween-20, Tween-80 and Triton X-100. P3862 displayed favorable caseinolytic activity, moderate keratinolytic activity but no collagenolytic activity. Besides, the homology model of P3862 possessed a globular configuration and characteristic of α/β hydrolase fold, and displayed stable interactions with casein, glycoprotein and keratin rather than collagen. Moreover, the crude enzyme could completely dehair goatskin within 6 h, resulting in decrease in BOD5, COD and TSS loads by 72.86, 74.07, and 73.79%, respectively, as compared with Na2S treatment. Biocatalytic applications revealed that it could effectively remove egg-stains from fabrics at 37 °C for 30 min with low supplementation (300 U/mL), and was able to degrade the feathers of duck and chicken. Overall, these outstanding properties make P3862 valuable in the development of environmentally friendly biotechnologies .

Abstract:Proteases have prospective financial and environment-friendly applications; hence attention is focused currently on the finding of new protease producing microorganism so as to meet the requirements of industry. A thermophilic bacterial strain producing extracellular protease activity was isolated from soil and identified as Bacillus cereus by analysis of 16S rRNA. Protease production by the microorganism was improved by studying the impact of the type of nitrogen and carbon source, fermentation period, growth temperature and initial pH of the culture medium in cultivation optimization experiments. The enzyme was purified to homogeneity in two step procedure involving Sephadex G-75 and Q-Sepharose chromatography. The molecular weight of purified enzyme was found to be 58 kDa by SDS-PAGE. Protease exhibited a pH and temperature optima of 7.5 and 60°, respectively. The enzyme was active in the pH range of 6.0-9.0 and stable up to 70°C. Histological analysis of protease treated goat and cow skin pelts showed complete removal of non leather forming structures such as hair shaft, hair follicles and glandular structures. The protease showed the stain removing property from blood stained cotton cloth and found to be compatible with six commercially available detergents. The protease could release peptides from natural proteins after digestion of coagulated egg albumin and blood clot.
Proteomic analysis and optimized production of Alkalihalobacillus patagoniensis PAT 05T extracellular proteases

Abstract:Extracellular proteolytic extracts from the haloalkalitolerant strain Alkalihalobacillus patagoniensis PAT 05T have proved highly efficient to reduce wool felting, as part of an ecofriendly treatment suitable for organic wool. In the present study, we identified the extracellular proteases produced by PAT 05T and we optimized its growth conditions for protease production through statistical methods. A total of 191 proteins were identified in PAT 05T culture supernatants through mass spectrometry analysis. Three of the 6 detected extracellular proteases belonged to the serine-endopeptidase family S8 (EC 3.4.21); two of them showed 86.3 and 67.9% identity with an alkaline protease from Bacillus alcalophilus and another one showed 50.4% identity with Bacillopeptidase F. The other 3 proteases exhibited 55.3, 49.4 and 61.1% identity with D-alanyl-D-alanine carboxypeptidase DacF, D-alanyl-D-alanine carboxypeptidase DacC and endopeptidase LytE, respectively. Using a Fractional Factorial Design followed by a Central Composite Design optimization, a twofold increase in protease production was reached. NaCl concentration was the most influential factor on protease production. The usefulness of PAT 05T extracellular proteolytic extracts to reduce wool felting was possible associated with the activity of the serine-endopeptidases closely related to highly alkaline keratinolytic proteases. The other identified proteases could cooperate, improving protein hydrolysis. This study provided valuable information for the exploitation of PAT 05T proteases which have potential for the valorization of organic wool as well as for other industrial applications.

Abstract:Proteases are a group of industrially valuable enzymes with significant demand owing to their diverse and extensive range of biotechnological applications. This study aimed to purify and optimize protease from oil seeds as oil seeds contain high oil and protein content along with the enzymes responsible for the digestion of these biomolecules. Three indigenous oil seeds including Brassica compestris, Zea mays and Helianthus annus were included in this study and proteases were isolated and characterized by germinating these seeds at different time periods. The partial purification of enzyme was carried out up to 4.8, 2.5 and 2.44-fold from 5-days old germinating oilseeds of MH 5 , BH 5 and SH 5 . Total protein content (TPC) of 30, 50 and 250 mgmL −1 were obtained for MH 5 , BH 5 and SH 5 , respectively with maximum precipitation at 60–70% saturation with (NH 4 ) 2 SO 4 . The enzyme was observed to be stable up to 50 o C at pH 8.0 using casein as substrate. The enzyme activity was alleviated using Ca 2+ , Fe 2+ , Cu 2+ and Co 2+ except for BH 5 where Ca 2+ was ineffective in enhancing or reducing enzyme-activity. The specific-activity values of 0.07, 0.094 and 0.02 mgmL −1 were observed for MH 5 , BH 5 and SH 5 respectively. The supplementation of enzyme preparation in detergent completely removed the blood stain from a stained cloth providing the basis of enzyme utilization in detergent industry. Moreover, dehairing effects were observed when applied to goat ear expanding the scope of purified enzyme to enhance the quality of leather in tannery industries by promoting the dehairing process of animal skins thereby avoiding environmental pollution.

Abstract:Optimal medium was used to improve the production of keratinase by Bacillus licheniformis ZJUEL31410, which has a promising application in the transformation of feather into soluble protein. The results of single factor design revealed that the concentration of feather at 20 g/l and the initial pH at value 8 was the best for the production of keratinase and the degradation of feather. Ammonia salt and nitrate salt strongly restricted the production of keratinase and the degradation of feather. Result of Box-Behnken design (BBD) experiment which was used to optimize concentrations of glucose, corn steep flour and K 2 HPO 4 for further improvement of keratinase productivity showed that the optimal medium was composed of glucose (20 g/l), corn steep flour (7.5 g/l), K 2 HPO 4 (1 g/l) and feather (20 g/l). The result of submerged batch cultivation of B. licheniformis ZJUEL31410 in the 5 L fermentor indicated that the optimal medium had the highest keratinase and the degree of feather degradation (DFD) at 54.9 U/ml and 72.4%; both were 5 times more than the basal medium. The degradation of feather was verified by the analysis of scanning electron microscopy (SEM). This study provides a foundation for the production of keratinase and the conversion of feather to soluble protein through submerged fermentation process by B. licheniformis ZJUEL31410. Abstract:The objective of the current study was to optimize the fermentation conditions of producing fibrinolytic enzyme through single factor experiment and response surface experiment.The optimal fermentation conditions were obtained as follows:the carbon source was bran,the nitrogen source was yeast extract,the inorganic salt was 0.1% MgSO4,the solvent of medium was bean juice,pH was 4.0.Using 2% bran,2% yeast extract,0.1% MgSO4 as the major factor,choosing the right step,designing response surface experiment,the second-order regression model was obtained fitting the experimental data results.The R2 of the model was 0.8709,and was significant at the 90% of the probability.The second-order model predicted that under the conditions of 2.97% bran,2.30% yeast extract paste,0.147% MgSO4,pH 4,the highest fibrinolytic activity predicted was 410 IU/mL.
A novel unhairing enzyme produced by heterologous expression of keratinase gene (kerT) in Bacillus subtilis

Abstract:To promote enzymatic unhairing for leather production, a new unhairing enzyme is developed. The Keratinase (kerT) gene, which is amplified from B. amyloliquefaciens TCCC11319 by PCR, is expressed in B. subtilis WB600. The recombinant KerT reduces the collagenolytic protease content as well as improving the keratinase content effectively. Therefore, the improved keratinase leads to the obviously unhairing effect, whereas the low collagenolytic protease ensures the integrity of collagen fibers in hide. It represents, the leather grain surface isn't destroyed thereby the value of finished leather can be maintained. In addition, by analyzing the properties of KerT, tits activity isn't inhibited with Na+, K+ and Ca2+ which are commonly used in leather production. The freeze-dried fermentation broth can be used directly as unhairing enzyme without addition of traditional sulfide chemicals. By evaluating the properties of unhaired hide, the results show that the collagen degradation ability of this new unhairing enzyme is slightly and it does not cause any adverse effects on the leather quality. Besides, this unhairing enzyme doesn't further degrade collagen in the time range of 8 h to 24 h, thus it is safely and easy-control in actual production. In conclusion, the enzymatic unhairing method with recombinant KerT has the potential to be more sustainable and efficient alternative than current sulphur-lime method, and it does not require the further purification thereby saving the cost.
Enzymolysis Technology Optimization for Production of Antioxidant Peptides from Goat Milk Casein

Abstract:Abstract Antioxidant peptides can inhibit lipid peroxidation and scavenging free radicals, maintain the balance of free radicals, and against a variety of diseases. Response surface methodology was used to optimize process conditions for producing antioxidative peptides from goat’s milk casein hydrolysate with Alcalase. The results suggested that the optimal process parameters were: temperature at 62.5°C, pH 8.9, E/S ration at 2.5%, substrate concentration at 4.4% and hydrolysis time was 173min). Metal-chelating effect, superoxide anion radical scavenging activity and 1, 1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity were shown to be 87.21±0.88%, 49.18±1.42% and 69.07±1.26% respectively under the optimal condition. The actual and predicated value were closely which indicated the optimized data fit well to model and the optimized parameters are reliable.

The main aim of this research study was to isolate, screen, and identify alkaline protease-producing bacteria that were sampled from leather industry effluent present in the outer skirts of Addis Ababa, Ethiopia. Purpose To isolate and characterize the alkaline protease-producing bacteria from leather industrial effluents. Methods Samples are collected from Modji leather industrial effluents and stored in the microbiology lab. After isolated bacteria from effluent using serial dilution and followed by isolated protease-producing bacteria using skim milk agar media. After studying primary and secondary screening using zonal inhibition methods to select potential protease-producing bacteria using skim milk agar media. Finally, to identify the potential bacteria using biochemical methods, bacterial biomass, protease activity, and gene sequencing (16S rRNA) method to finalize the best alkaline protease producing bacteria identified. Results First twenty-eight different bacterial colonies were isolated initially from the leather industry effluent sample situated at the Modjo town of Ethiopia. The isolated bacteria were screened using the primary and secondary screening method with skim milk agar medium. At the primary level, we selected three isolates namely ML5(14 mm), ML12(18 mm), and MS12 (15 mm), showing the highest zone of proteolysis as a result of casein degradation on the agar plates were selected and subjected to primary screening. Further secondary screening confirmed that the zone of inhibition methods ML5 (14.00±0.75 mm), ML12 (19.50±0.66 mm), and MS12 (15.00±1.32 mm) has efficient proteolytic activity and can be considered as effective protease producer. The three isolates were then subjected to morphological and biochemical tests to identify probably bacterial species, and all the three bacterial isolates were found out to be of Bacillus species. The shake flask method was carried out to identify the most potent one having greater biomass production capabilities and protease activity. ML12 isolated from leather effluent waste showed the highest protease activity (19 U/ml), high biomass production, and the same was subjected to molecular identification using 16s sequencing and a phylogenetic tree was constructed to identify the closest neighbor. The isolate ML12 (Bacillus cereus strain -MN629232.1) is 97.87% homologous to Bacillus cereus strain (KY995152.1) and 97.86% homologous to Bacillus cereus strain (MK968813.1). Conclusions This study has exposed that from twenty-eight different bacterial samples isolated from leather industry effluent; further primary and secondary screening methods were selected three potential alkaline protease strains. Finally, based on its biochemical identification, biomass, and protease activity, ML12 (Bacillus cereus strains) is the best strain identified. The alkaline protease has the significant feature of housing potent bacterial species for producing protease of commercial value.

Abstract:In this study, neutral proteinase, alkaline proteinase, and papain were used to hydrolyze fresh pig skin respectively. The effects of pH value ,temperature, enzyme content and substrate content on hydrolysis were studied by the single factor test, and orthogonal design, by assessing the parameter ‐ degree of hydrolysis (DH), was adopted to determine the optimum condition of hydrolysis. The optimal hydrolysis condition of neutral proteinase was: pH 6.5, 45°C, enzymatic content 0.5%, and substrate content 15%; while The optimal hydrolysis condition of alkaline proteinase was: pH 11, 55°C, enzymatic content 0.5%, and substrate content 35%; and the most suitable hydrolysis parameters of papain were: pH 5, temperature of 60°C, 0.9% of enzyme content and 30% of substrate content. The ability of hydrolysis was sequenced as: alkaline proteinase > papain > neutral proteinase. On the basis of the optimum condition of each single proteinase, any two of the proteinases were chosen to hydrolyze fresh pig skin by two‐step‐hydrolysis. The results showed that any combination of the enzymes has a higher DH value than its single component and the adding sequence inflects DH value. Overall, the two‐step‐hydrolysis of papain plus alkaline shows the highest DH value (22.31%). Response Surface Methodology (RSM) was used for the optimization of all the culture conditions. Casein (1% w/v) was used as a substrate in nutrient agar medium for the screening of enzyme production potential and showed maximum zone of clearance (4.6 cm). It was identified as Bacillus subtilis K-5 by genetic identification based on 16S rRNA and blast technology of NCBI. Among culture conditions, incubation temperature, incubation time, pH of the medium and moisture level of the substrate were optimized. Maximum protease production was observed at 37oC, pH 9.0 with incubation time of 36 h and moisture to substrate ratio of 1: 0.75. Maximum protease production of 70.21 U/mL was recorded when wheat bran was used as an agro-industrial substrate. The activity of crude protease was observed 99.63 % at 60oC and pH 10.0 with protein concentration 0.63 mg/mL and specific activity of 111.56 U/mg. Protein contents of 0.57 mg/mL (specific activity of 124.72 U/mg) and protein contents of 0.44 mg/mL (specific activity of 143.65 U/mg) were observed by 70% saturation with ammonium sulphate and gel chromatography, respectively. Line Weaver Burk plot was used to find its Vmax and Km, which were 344. 83 mg/mL/min and 100.04 mg/mL, respectively. The study concluded that Bacillus subtilis K-5 is thermophilic and alkaliphilic strain which produces active protease and can be used as potential microorganism for industries.
