Abstract:The present study provides a comparative assessment of non-invasive sensors as means of estimating the microbial contamination and time-on-shelf (i.e. storage time) of leafy green vegetables, using a novel unified spectra analysis workflow. Two fresh ready-to-eat green salads were used in the context of this study for the purpose of evaluating the efficiency and practical application of the presented workflow: rocket and baby spinach salads. The employed analysis workflow consisted of robust data normalization, powerful feature selection based on random forests regression, and selection of the number of partial least squares regression coefficients in the training process by estimating the knee-point on the explained variance plot. Training processes were based on microbiological and spectral data derived during storage of green salad samples at isothermal conditions (4, 8 and 12C), whereas testing was performed on data during storage under dynamic temperature conditions (simulating real-life temperature fluctuations in the food supply chain). Since an increasing interest in the use of non-invasive sensors in food quality assessment has been made evident in recent years, the unified spectra analysis workflow described herein, by being based on the creation/usage of limited sized featured sets, could be very useful in food-specific low-cost sensor development.

Improving SSC Detection Accuracy of Cherry Tomatoes by Feature Synergy and Complementary Spectral Bands Combination

Abstract:The visible/near-infrared (VIS/NIR) spectroscopy technique has been extensively employed for the nondestructive detection of soluble solids content (SSC) in fruit. However, some existing algorithms and modeling optimization methods for improving the detection accuracy of SSC may be applicable to specific conditions and not universal. To break through this bottleneck problem, we propose a potentially universal strategy based on feature synergy and spectral bands combination of multiple detection modes. Firstly, compared with the commonly used halogen lamp, the xenon lamp was utilized to cover the ultraviolet (UV) range and match the characteristic information of different sugars (glucose, fructose, and sucrose). Furthermore, the spectral fusion method was employed to combine the reflectance and transmittance spectra to improve the cherry tomatoes SSC prediction accuracy with comprehensive data in the UV/VIS/NIR region. Finally, the best results were achieved by the partial least square regression (PLSR) model with spectral bands fusion, yielding the results of R2p, RMSEP, and RPD as 0.9653, 0.1998%, and 5.31, respectively. The same conclusion can also be verified when predicting the SSC in strawberries. Overall, this strategy is potentially universal to improve the prediction accuracy of SSC in fruit by matching light source spectra with characteristic absorption, as well as utilizing comprehensive spectral information in the 200-1100 nm range, providing valuable insights for the practical application of nondestructive detection of fruit internal quality.
Predicting sensory evaluation of spinach freshness using machine learning model and digital images

Abstract:The visual perception of freshness is an important factor considered by consumers in the purchase of fruits and vegetables. However, panel testing when evaluating food products is time consuming and expensive. Herein, the ability of an image processing-based, nondestructive technique to classify spinach freshness was evaluated. Images of spinach leaves were taken using a smartphone camera after different storage periods. Twelve sensory panels ranked spinach freshness into one of four levels using these images. The rounded value of the average from all twelve panel evaluations was set as the true label. The spinach image was removed from the background, and then converted into a gray scale and CIE-Lab color space (L*a*b*) and Hue, Saturation and Value (HSV). The mean value, minimum value, and standard deviation of each component of color in spinach leaf were extracted as color features. Local features were extracted using the bag-of-words of key points from Oriented FAST (Features from Accelerated Segment Test) and Rotated BRIEF (Binary Robust Independent Elementary Features). The feature combinations selected from the spinach images were used to train machine learning models to recognize freshness levels. Correlation analysis between the extracted features and the sensory evaluation score showed a positive correlation (0.5 < r < 0.6) for four color features, and a negative correlation (‒0.6 < r < ‒0.5) for six clusters in the local features. The support vector machine classifier and artificial neural network algorithm successfully classified spinach samples with overall accuracy 70% in four-class, 77% in three-class and 84% in two-class, which was similar to that of the individual panel evaluations. Our findings indicate that a model using support vector machine classifiers and artificial neural networks has the potential to replace freshness evaluations currently performed by non-trained panels.

Abstract:Spinach is prone to spoilage in the course of preservation. Spinach leaves stored at different temperatures for different durations will have varying degrees of freshness. In order to monitor the freshness of spinach leaves during storage, a rapid and non-destructive method—hyperspectral imaging technology—was applied in this study. Visible near-infrared reflectance (Vis-NIR) (380–1030 nm) and near-infrared reflectance (NIR) (874–1734 nm) hyperspectral imaging systems were used. Spinach leaves preserved at different temperatures with different durations (0, 3, 6, 9 days at 4 °C and 0, 1, 2 days at 20 °C) were studied. Principal component analysis (PCA) was adopted as a qualitative analysis method. The second-order derivative spectra were utilized to select effective wavelengths. Partial least squares discriminant analysis (PLS-DA), support vector machine (SVM), and extreme learning machine (ELM) were used to build models based on full spectra and effective wavelengths. All three models achieved good results, with accuracies above 92% for both Vis-NIR spectra and NIR spectra. ELM obtained the best results, with all accuracies reaching 100%. The overall results indicate the possibility of the freshness identification of spinach preserved at different temperatures for different durations using two kinds of hyperspectral imaging systems.

Abstract:Visible/near-infrared (Vis/NIR) spectroscopy was applied to determine the ripeness of melons ( Cucumis melo L.). The total soluble solids (TSS) and flesh color distribution around the equator fracture surface was studied, and a high correlation between TSS and flesh color in the outer mesocarp region was observed. A new ripeness index, defined as the edible ratio (ER), was proposed. One hundred and twenty melons with similar sizes were selected as samples and divided into a calibration set and a prediction set. Spectra of melons were acquired separately under transmittance and interactance modes. A machine vision system was established to acquire images of the melon fracture surfaces to further calculate the ER values of the melons. Partial least squares (PLS) calibration models were developed to predict the melon ripeness indices, including TSS, surface color (SC), and ER value. The results were promising for TSS, Hunter a , and ER value. For Hunter b , the results were insufficient. In addition, by comparing the performances of the calibration models developed under different detection modes, it was concluded that the best calibration results for TSS and ER were achieved under the transmittance mode, while for Hunter a , the best result was obtained using interactance spectra.

Abstract:The rapid assessment of the microbiological quality of highly perishable food commodities is of great importance. Spectroscopic data coupled with machine learning methods have been investigated intensively in recent years, because of their rapid, non-destructive, eco-friendly qualities and their potential to be used on-, in- or at-line. In the present study, the microbiological quality of chicken burgers was evaluated using Fourier transform infrared (FTIR) spectroscopy and multispectral imaging (MSI) in tandem with machine learning algorithms. Six independent batches were purchased from a food industry and stored at 0, 4, and 8 °C. At regular time intervals (specifically every 24 h), duplicate samples were subjected to microbiological analysis, FTIR measurements, and MSI sampling. The samples (n = 274) acquired during the data collection were classified into three microbiological quality groups: “satisfactory”: 4−7 log CFU/g, “acceptable”: 7−8 log CFU/g, and “unacceptable”: >8 logCFU/g. Subsequently, classification models were trained and tested (external validation) with several machine learning approaches, namely partial least squares discriminant analysis (PLSDA), support vector machine (SVM), random forest (RF), logistic regression (LR), and ordinal logistic regression (OLR). Accuracy scores were attained for the external validation, exhibiting FTIR data values in the range of 79.41−89.71%, and, for the MSI data, in the range of 74.63−85.07%. The performance of the models showed merit in terms of the microbiological quality assessment of chicken burgers.

Abstract:In order to prolong the shelf-life of fruits and vegetables, plastic films have been covered on them to improve water retention and keep external bacteria away. It is of great significance to estimate the quality of packaged fruits and vegetables accurately by predicting the shelf-life of them. In this research, hyperspectral technology combined with chemometric methods were employed to estimate the shelf-life of fresh spinach leaves in the same environment. Hyperspectral data covering the range of Vis-NIR (380～1 030 nm) and NIR (874～1 734 nm) were acquired from 300 spinach leaves (75 dishs) which were stored in 4 ℃ among 5 periods (0 d, 2 d, 4 d, 6 d, 8 d). Meanwhile, the chlorophyll contents of all spinach leaves were determined. The mean spectra of 300 spinach leaves (200 leaves in training set and 100 leaves in prediction set) were extracted. And then, principal component analysis (PCA) on the training set of 200 spectra from 5 periods of shelf-life displayed apparent cluster. Partial least-squares discriminant analysis (PLS-DA) models were established according to spectral datas and the virtual levels that we ascribed to the different storage periods previously. The total discriminant accuracy rates of prediction set were 83% (VIS-NIR) and 81% (NIR), respectively. The result indicated that the classification and prediction on the shelf-life of fresh spinach can be realized with hyperspectral technology combined with chemometric methods, which offered a theoretical guidance to evaluate the quality of packaged spinach for consumers, and provided technical supports for the development of instruments used for testing the shelf-life of fruits and vegetables in further study.

Abstract:With more attention being paid to healthy diets, vegetable freshness is a concerning issue for consumers and traders. Rapid and non-destructive evaluation of vegetable freshness plays an important role in vegetable consumption. In this study, hyperspectral imaging was used to identify the freshness of spinach and Chinese cabbage stored at different durations. Classification models were built using the extracted average spectra, including conventional machine learning methods (logistic regression (LR), support vector machine (SVM), and random forest (RF)) and deep learning methods (convolutional neural networks, long-short term memory (LSTM), and CNN combined with LSTM (CNN-LSTM)). Results showed that CNN-LSTM models outperformed the other models for both vegetables, with classification accuracy over 80 % in the training, validation and testing set. Grad-CAM++ methods showed that great similarity of important wavelengths contributing more to the freshness identification between the vegetables existed. Both fine-tuning and direct prediction using the model built on one vegetable to predict the other vegetable were explored. Good performances were obtained for both situations with the classification accuracy over 80 % for the three sets using all training samples. The overall results illustrated the great potential of hyperspectral imaging with deep learning approaches to identify the freshness of different vegetables. This study provides a basis for further assessment of vegetable freshness.

Abstract:Here we report that mutations within the DNA-binding domain of AR, shown previously to inhibit nuclear export to the cytoplasm, cause an androgen-dependent defect in intranuclear trafficking of AR. Mutation of two conserved phenylalanines within the DNA recognition helix (F582, 583A) results in androgen-dependent arrest of AR in multiple subnuclear foci. A point mutation in one of the conserved phenylalanines (DeltaF582, F582Y) is known to cause androgen insensitivity syndrome (AIS). Both AIS mutants (DeltaF582, F582Y) and the export mutant (F582, 583A) displayed androgen-dependent arrest in foci, and all three mutants promoted androgen-dependent accumulation of the histone acetyl transferase CREB binding protein (CBP) in the foci. The foci correspond to a subnuclear compartment that is highly enriched for the steroid receptor coactivator glucocorticoid receptor-interacting protein (GRIP)-1. Agonist-bound wild-type AR induces the redistribution of GRIP-1 from foci to the nucleoplasm. This likely reflects a direct interaction between these proteins because mutation of a conserved residue within the major coactivator binding site on AR (K720A) inhibits AR-dependent dissociation of GRIP-1 from foci. GRIP-1 also remains foci-associated in the presence of agonist-bound F582, 583A, DeltaF582, or F582Y forms of AR. Two-dimensional phospho-peptide mapping and analysis with a phospho-specific antibody revealed that mutant forms of AR that arrest in the subnuclear foci are hypophosphorylated at Ser81, a site that normally undergoes androgen-dependent phosphorylation. Our working model is that the subnuclear foci are sites where AR undergoes ligand-dependent engagement with GRIP-1 and CBP, a recruitment step that occurs before Ser81 phosphorylation and association with promoters of target genes.
Space-qualified, abuttable packaging for LBNL p-channel CCDs, part II

Abstract:Fully depleted, back-illuminated, p-channel CCDs developed at Lawrence Berkeley National Laboratory exhibit high quantum efficiency in the near-infrared (700-1050nm), low fringing effects, low lateral charge diffusion (and hence small, well-controlled point spread function), and high radiation tolerance. Building on previous efforts, we have developed techniques and hardware that have produced space-qualified 4-side abuttable, high-precision detector packages for 10.5μm pixel, 3.5k x 3.5k p-channel LBNL CCDs. These packages are built around a silicon carbide mounting pedestal, providing excellent rigidity, thermal stability, and heat transfer. Precision fixturing produces packages with detector surface flatness better than 10μm P-V. These packages with active areas of 36.8mm square may be packed on a detector pitch as small as 44mm. LBNL-developed Front End Electronics (FEE) packages can mount directly to the detector packages within the same footprint and detector pitch. This combination, along with identically interfaced NIR detector/FEE packages offers excellent opportunities for high density, high pixel count focal planes for space-based, ground-based, and airborne astronomy.

Abstract:In food industry, quality and safety are considered important issues worldwide that are directly related to health and social progress. The use of vision technology for quality testing of food production has the obvious advantage of being able to continuously monitor a production using non-destructive methods, thus increasing the quality and minimizing cost. The performance of an intelligent decision support system has been evaluated in monitoring the spoilage of minced beef stored either aerobically or under modified atmosphere packaging, at different storage temperatures (0, 5, 10, and 15 °C) utilising multispectral imaging information. This paper utilises a neuro-fuzzy model which incorporates a clustering pre-processing stage for the definition of fuzzy rules, while its final fuzzy rule base is determined by competitive learning. Initially, meat samples are classified according to their storage conditions, while identification models are then utilised for the prediction of the Total Viable Counts of bacteria. The innovation of the proposed approach is further extended to the identification of the temperature used for storage, utilizing only imaging spectral information. Results indicated that spectral information in combination with the proposed modelling scheme could be considered as an alternative methodology for the accurate evaluation of meat spoilage.

Prediction of various freshness indicators in fish fillets by one multispectral imaging system

Abstract:Abstract In current study, a simple multispectral imaging (430–1010 nm) system along with linear and non-linear regressions were used to assess the various fish spoilage indicators during 12 days storage at 4 ± 2 °C. The indicators included Total-Volatile Basic Nitrogen (TVB-N) and Psychrotrophic Plate Count (PPC) and sensory score in fish fillets. immediately, after hyperspectral imaging, the reference values (TVB-N, PPC and sensory score) of samples were obtained by traditional method. To simplify the calibration models, nine optimal wavelengths were selected by genetic algorithm. The prediction performance of various chemometric models including partial least-squares regression (PLSR), multiple-linear regression (MLR), least-squares support vector machine (LS-SVM) and back-propagation artificial neural network (BP-ANN) were compared. All models showed acceptable performance for simultaneous predicting of PPC, TVB-N and sensory score (R 2 P ≥ 0.853 and RPD ≥ 2.603). Non-linear models were considered better quantitative model to predict all of three freshness indicators in fish fillets. Among the three spoilage indices, the best predictive power was obtained for PPC value and the weakest one was acquired for TVB-N content prediction. The best model for prediction TVB-N (R 2 p = 0.862; RMSEP = 3.542 and RPD = 2.678) and sensory score (R 2 p = 0.912; RMSEP = 1.802 and RPD = 3.33) belonged to GA-LS-SVM and for prediction of PPC value was BP-ANN (R 2 p = 0.921; RMSEP = 0.504 and RPD = 3.64). Therefore, developing multispectral imaging system based on LS-SVM model seems to be suitable for simultaneous prediction of all three indicators (R 2 P > 0.862 and RPD > 2.678). Further studies needed to improve the accuracy and applicability of HSI system for predicting freshness of rainbow-trout fish.

Abstract:Background: The market for fruits and vegetables is mainly controlled by the mass distribution channel (MDC). MDC buyers do not have useful instruments to rapidly evaluate the quality of the products. Decisions by the buyers are driven primarily by pricing strategies rather than product quality. Simple, rapid and easy-to-use methods for objectively evaluating the quality of postharvest products are needed. The present study aimed to use visible and near-infrared (vis/NIR) spectroscopy to estimate some qualitative parameters of two low-price products (carrots and tomatoes) of various brands, as well as evaluate the applicability of this technique for use in stores. Results: A non-destructive optical system (vis/NIR spectrophotometer with a reflection probe, spectral range 450-1650 nm) was tested. The differences in quality among carrots and tomatoes purchased from 13 stores on various dates were examined. The reference quality parameters (firmness, water content, soluble solids content, pH and colour) were correlated with the spectral readings. The models derived from the optical data gave positive results, in particular for the prediction of the soluble solids content and the colour, with better results for tomatoes than for carrots. Conclusion: The application of optical techniques may help MDC buyers to monitor the quality of postharvest products, leading to an effective optimization of the entire supply chain. © 2017 Society of Chemical Industry.

Abstract:The consumption of ready-to-eat (RTE) leafy vegetables has increased rapidly due to changes in consumer diet. RTE products are perceived as fresh, high-quality, and health-promoting. The monitoring of the RTE quality is crucial in relation to safety issues. This study aimed to evaluate the maintenance of RTE rocket salad freshness packed under modified atmospheres. A portable E-nose, the electrolyte leakage test (which measures the index of leaf damage—ILD), and NIR spectroscopy and Aquaphotomics were employed. Two trials were carried out, using the following gas mixtures: (A) atmospheric air (21% O2, 78% N2); (B) 30% O2, 70% N2; (C) 10% CO2, 5% O2, 85% N2. Samples were stored at 4 °C and analyzed at 0, 1, 4, 7, 11, and 13 days. ANOVA, PCA, PLS were applied for data processing. E-nose and ILD results identified the B atmosphere as the best for maintaining product freshness. NIR spectroscopy was able to group the samples according to the storage time. Aquaphotomics proved to be able to detect changes in the water structure during storage. These preliminary data showed a good agreement NIR/ILD suggesting the use of NIR for non-destructive monitoring of the damage to the plant membranes of RTE rocket salad.

Abstract:The market demand for baby leaf lettuce is constantly increasing, while safety has become one of the most important traits in determining consumer preference driven by human health hazards concerns. In this study, the performance of visible and near-infrared (vis/NIR) spectroscopy was tested in discriminating pesticide-free against pesticide-treated lettuce plants. Two commercial fungicides (mancozeb and fosetyl-al) and two insecticides (deltamethrin and imidacloprid) were applied as spray solutions at the recommended rates on baby leaf lettuce plants. Untreated-control plants were sprayed with water. Reflectance data in the wavelength range 400–2500 nm were captured on leaf samples until harvest on the 10th day upon pesticide application, as well as after 4 and 8 days during post-harvest storage at 5 °C. In addition, biochemical components in leaf tissue were also determined during storage, such as antioxidant enzymes' activities (peroxidase [POD], catalase [CAT], and ascorbate peroxidase [APX]), along with malondialdehyde [MDA] and hydrogen peroxide [H2O2] content. Partial least square discriminant analysis (PLSDA) combined with feature-selection techniques was implemented, in order to classify baby lettuce tissue into pesticide-free or pesticide-treated ones. The genetic algorithm (GA) and the variable importance in projection (VIP) scores identified eleven distinct regions and nine specific wavelengths that exhibited the most significant effect in the detection models, with most of them in the near-infrared region of the electromagnetic spectrum. According to the results, the classification accuracy of discriminating pesticide-treated against non-treated lettuce leaves ranged from 94% to 99% in both pre-harvest and post-harvest periods. Although there were no significant differences in enzyme activities or H2O2, the MDA content in pesticide-treated tissue was greater than in untreated ones, implying that the chemical spray application probably induced a stress response in the plant that was disclosed with the reflected energy. In conclusion, vis/NIR spectroscopy appears as a promising, reliable, rapid, and non-destructive tool in distinguishing pesticide-free from pesticide-treated lettuce products.

Abstract:Washing and sanitation are vital steps during the postharvest processing of fresh produce to reduce the microbial load on the produce surface. Although current process control and validation tools effectively predict sanitizer concentrations in wash water, they have significant limitations in assessing sanitizer effectiveness for reducing microbial counts on produce surfaces. These challenges highlight the urgent need to improve the validation of sanitation processes, especially considering the presence of dynamic organic contaminants and complex surface topographies. This study aims to provide the fresh produce industry with a novel, reliable, and highly accurate method for validating the sanitation efficacy on the produce surface. Our results demonstrate the feasibility of using a food-grade, catalase (CAT)-immobilized biomimetic leaf in combination with vibrational spectroscopy and machine learning to predict microbial inactivation on microgreen surfaces. This was tested using two sanitizers: sodium hypochlorite (NaClO) and hydrogen peroxide (H2O2). The developed CAT-immobilized leaf-replicated PDMS (CAT@L-PDMS) effectively mimics the microscale topographies and bacterial distribution on the leaf surface. Alterations in the FTIR spectra of CAT@L-PDMS, following simulated sanitation processes, indicate chemical changes due to CAT oxidation induced by NaClO or H2O2 treatments, facilitating the subsequent machine learning modeling. Among the five algorithms tested, the competitive adaptive reweighted sampling partial least squares discriminant analysis (CARS-PLSDA) algorithm was the most effective for classifying the inactivation efficacy of E. coli on microgreen leaf surfaces. It predicted bacterial reduction on microgreen surfaces with 100% accuracy in both training and prediction sets for NaClO, and 95% in the training set and 86% in the prediction set for H2O2. This approach can improve the validation of fresh produce sanitation processes and pave the way for future research.

Abstract:Gas sensors present an alternative to traditional off-package food quality assessment, due to their high sensitivity and fast response without the need of sample pre-treatment. The safe integration of gas sensors into packaging without compromising sensitivity, response rate, and stability, however, remains a challenge. Such packaging integration of spoilage sensors is crucial towards preventing food waste and transitioning towards more sustainable supply chains. Here, we demonstrate a wide-ranging solution to enable the use of gas sensors for the continuous monitoring of food spoilage, building upon our previous work on Paper-based Electrical Gas Sensors (PEGS). By comparing various materials commonly used in the food industry, we analyze the optimal membrane to encapsulate PEGS for packaging integration. Focusing on spinach as a high-value crop, we assess the feasibility of PEGS to monitor the gases released during its spoilage at low and room temperatures. Finally, we integrate the sensors with wireless communication and batteryless electronics, creating a user-friendly system to evaluate the spoilage of spinach, operated by a smartphone via Near Field Communication (NFC). The work reported here provides an alternative approach that surpasses traditional on-site and in-line monitoring, ensuring comprehensive monitoring of food shelf life.

In Search of Optimum Fresh-Cut Raw Material: Using Computer Vision Systems as a Sensory Screening Tool for Browning-Resistant Romaine Lettuce Accessions

Abstract:The popularity of ready-to-eat (RTE) salads has prompted novel technology to prolong the shelf life of their ingredients. Fresh-cut romaine lettuce is widely used in RTE salads; however, its tendency to quickly discolor continues to be a challenge for the industry. Selecting the ideal lettuce accessions for use in RTE salads is essential to ensure maximum shelf life, and it is critical to have a practical way to assess and compare the quality of multiple lettuce accessions that are being considered for use in fresh-cut applications. Thus, in this work we aimed to determine whether a computer vision system (CVS) composed of image acquisition, processing, and analysis could be effective to detect visual quality differences among 16 accessions of fresh-cut romaine lettuce during postharvest storage. The CVS involved a post-capturing color correction, effective image segmentation, and calculation of a browning index, which was tested as a predictor of quality and shelf life of fresh-cut romaine lettuce. The results demonstrated that machine vision software can be implemented to replace or supplement the scoring of a trained panel and instrumental quality measurements. Overall visual quality, a key sensory parameter that determines food preferences and consumer behavior, was highly correlated with the browning index, with a Pearson correlation coefficient of −0.85. Other important sensory decision parameters were also strongly or moderately correlated with the browning index, with Pearson correlation coefficients of −0.84 for freshness, 0.79 for off odor, and 0.57 for browning. The ranking of the accessions according to quality acceptability from the sensory evaluation produced a similar pattern to those obtained with the CVS. This study revealed that multiple lettuce accessions can be effectively benchmarked for their performance as fresh-cut sources via a CVS-based method. Future opportunities and challenges in using machine vision image processing to predict consumer preferences for RTE salad greens is also discussed.

Abstract:Contamination is a critical issue that affects food consumption adversely. Therefore, efficient detection and classification of food contaminants are essential to ensure food safety. This study applied a visible and near-infrared (VNIR) hyperspectral imaging technique to detect and classify organic residues on the metallic surfaces of food processing machinery. The experimental analysis was performed by diluting both potato and spinach juices to six different concentration levels using distilled water. The 3D hypercube data were acquired in the range of 400–1000 nm using a line-scan VNIR hyperspectral imaging system. Each diluted residue in the spectral domain was detected and classified using six classification methods, including a 1D convolutional neural network (CNN-1D) and five pre-processing methods. Among them, CNN-1D exhibited the highest classification accuracy, with a 0.99 and 0.98 calibration result and a 0.94 validation result for both spinach and potato residues. Therefore, in comparison with the validation accuracy of the support vector machine classifier (0.9 and 0.92 for spinach and potato, respectively), the CNN-1D technique demonstrated improved performance. Hence, the VNIR hyperspectral imaging technique with deep learning can potentially afford rapid and non-destructive detection and classification of organic residues in food facilities.

Abstract:Fresh-cut leafy vegetables are one of the most perishable products because they readily deteriorate in quality even during cold storage and have a relatively short shelf life. Since these products are in high demand, methods for rigorous quality control and estimation of freshness that are rapid and non-destructive would be highly desirable. The objective of the present research was to develop a rapid, non-destructive near-infrared spectroscopy (NIRS)-based method for the evaluation of changes during cold storage of lettuce using an aquaphotomics approach to monitor the water molecular structure in lettuce leaves. The reference measurements showed that after 6 days of dark, cold storage, the weight and water activity of lettuce leaves decreased and β-carotene decreased, while chlorophylls slightly increased. Aquaphotomics characterization showed large differences in the lettuce leaves' spectra depending on their growth zone. Difference spectra, principal component analysis (PCA) and linear discriminant analysis (LDA) confirmed the differences in the inner and outer leaves and revealed that spectra change as a function of storage time. Partial least squares regression (PLSR) allowed the prediction of the time spent in storage with a coefficient of determination of R2 = 0.80 and standard error of RMSE = 0.77 days for inner, and R2 = 0.86 and RMSE = 0.66 days for outer leaves, respectively. The following water absorbance bands were found to provide the most information in the spectra: 1348, 1360, 1373, 1385, 1391, 1410, 1416, 1422, 1441, 1447, 1453, 1466, 1472, 1490, 1503, 1515, 1521, 1534 and 1571 nm. They were further used as water matrix coordinates (WAMACs) to define the water spectral patterns (WASPs) of lettuce leaves. The WASPs of leaves served to succinctly describe the state of lettuces during storage. The changes in WASPs during storage reveled moisture loss, damage to cell walls and expulsion of intracellular water, as well as loss of free and weakly hydrogen-bonded water, all leading to a loss of juiciness. The WASPs also showed that damage stimulated the defense mechanisms and production of vitamin C. The leaves at the end of the storage period were characterized by water strongly bound to collapsed structural elements of leaf tissues, mainly cellulose, leading to a loss of firmness that was more pronounced in the outer leaves. All of this information was reflected in the changes of absorbance in the identified WAMACs, showing that the water molecular structure of lettuce leaves accurately reflects the state of the lettuce during storage and that WASPs can be used as a multidimensional biomarker to monitor changes during storage.
