Abstract:Guanidinoacetic acid can improve pork quality. Previous studies have demonstrated that pork quality is closely linked to the muscle fiber type mediated by PPARGC1A. Therefore, this study aimed to evaluate the influence of dietary GAA supplementation on the skeletal muscle fiber type transformation. A total of 180 healthy Duroc × Landrace × Meishan cross castrated male pigs with a similar average weight (90 ± 1.5 kg) were randomly divided into three treatments with five replicates per treatment and 12 pigs per replicate, including a GAA-free basal diet and basal diet with 0.05% or 0.10% GAA for 15 days. Our results showed that 0.10% GAA supplementation increased the contents of Ca2+ in sarcoplasm (p < 0.05). Compared with the control group, both GAA supplementation groups upregulated the expression of Troponin I-ss (p < 0.05), and 0.10% GAA supplementation downregulated the expression of Troponin T3 (p < 0.05). GAA supplementation increased the expression of peroxisome proliferator activated receptor-γ coactivator-1alpha (PPARGC1A) (p < 0.05), and further upregulated the mitochondrial transcription factor A (TFAM), increased the level of membrane potential, and the activities of mitochondrial respiratory chain complex I, III (p < 0.05). The 0.10% GAA supplementation upregulated the protein expression of calcineurin catalytic subunit α (CnAα) and nuclear factor of activated T cells (NFATc1) (p < 0.05). Overall, dietary GAA supplementation promotes skeletal muscle fiber types transformation from fast-to-slow-twitch via increasing the PPARGC1A based mitochondrial function and the activation of CaN/NFAT pathway in finishing pigs.
Comparative effects of phosphorylation and acetylation on glycolysis and myofibrillar proteins degradation in postmortem muscle

Abstract:The study investigated the different effects between protein phosphorylation and acetylation on glycolytic enzyme activity and myofibrillar protein degradation. Lamb longissimus thoracis lumborum muscles were homogenized and then inhibitors were added for incubation at 4 °C. Phosphatase inhibitor was added to produce a high phosphorylation level (PI group) and lysine deacetylase inhibitor was added to produce a high acetylation level (DI group). The lactate and ATP content in the PI group was inhibited compared with that in the DI group ( P < 0.05). Phosphofructokinase (PFK) activity was negatively related with the phosphorylation level and was positively related with the acetylation level in the DI group ( P < 0.05). The degradation of troponin T and desmin of the DI group were restrained when compared to that in the PI group ( P < 0.05). Compared with initial PFK and desmin, the simulation of phosphorylation and acetylation of PFK and desmin showed different electrostatic potential at the surface and a more unstable structure. The phosphorylation level of the DI group was increased, suggesting that the changes of protein acetylation altered protein phosphorylation. In conclusion, compared with protein phosphorylation, protein acetylation had a greater effect on promoting glycolysis and inhibiting protein degradation.

Guanidine Acetic Acid Alters Tissue Bound Amino Acid Profiles and Oxidative Status in Finishing Pigs

Abstract:This study aims to investigate the effects of guanidine acetic acid (GAA) on carcass traits, plasma biochemical parameters, tissue antioxidant capacity, and tissue-bound amino acid contents in finishing pigs. Seventy-two 140-day-old (body weight 86.59 ± 1.16 kg) crossbred pigs (Duroc × Landrace × Large White) were randomly assigned into four treatments with six replicate pens and three pigs per pen, which were fed the basal diets supplemented with 0, 0.05%, 0.10%, or 0.15% GAA, respectively. The plasma glucose concentration decreased, and creatine kinase activity and levels of GAA and creatine increased with the dietary GAA concentration. GAA linearly improved creatine content in the longissimus thoracis muscle (LM) and heart. The activities of superoxide dismutase, total antioxidant capacity, and glutathione peroxidase increased linearly in tissue or/and plasma, while the contents of malondialdehyde and protein carbonyl decreased linearly. GAA improved the contents of multiple-bound amino acids (such as proline or isoleucine) in the myocardium and LM. In conclusion, GAA enhanced the plasma biochemical parameters, oxidative status, and bound amino acid profiles of the heart and LM in finishing pigs.

Abstract:It is well known that preslaughter (antemortem) stress such as rough handling, transportation, a negative environment, physical discomfort, lack of consistent routine, and bad feed quality has a big impact on meat quality. The antemortem-induced poor meat quality is characterized by low pH, a pale and exudative appearance, and a soft texture. Previous studies indicate that antemortem stress plays a key role in regulating protein acetylation and glycolysis in postmortem (PM) muscle. However, the underlying molecular and biochemical mechanism is not clearly understood yet. In this study, we investigated the relationship between antemortem and protein acetylation and glycolysis using murine longissimus dorsi muscle isolated from ICR mice and murine muscle cell line C 2 C 12 treated with epinephrine hydrochloride. Because adrenaline secretion increases in stressed animals, epinephrine hydrochloride was intraperitoneally injected epinephrine into mice to simulate pre-slaughter stress in this study to facilitate experimental operations and save experimental costs. Our findings demonstrated that protein acetylation in pyruvate kinase M1 (PKM1) form is significantly reduced by antemortem, and the reduced acetylation subsequently leads to an increase in PKM1 enzymatic activity which causes increased glycolysis in PM muscle. By using molecular approaches, we identified lysine 141 in PKM1 as a critical residue for acetylation. Our results in this study provide useful insight for controlling or improving meat quality in the future.

ACAT2 Is a Novel Negative Regulator of Pig Intramuscular Preadipocytes Differentiation

Abstract:Intramuscular fat (IMF) is considered as the fat deposited between muscle fibers. The extracellular matrix microenvironment of adipose tissue is of critical importance for the differentiation, remodeling and function of adipocytes. Therefore, in this study we extracted the muscle tissue centrifugal fluid (MTF) of the longissimus dorsi of Erhualian pigs to mimic the microenvironment of intramuscular pre-adipocytes. MTF of pigs with low intramuscular fat level can inhibit pig intramuscular pre-adipocytes differentiation. Then, proteomics technology (iTRAQ) was used to analyze the MTF with different IMF content, and it was found that individuals with high IMF had low ACAT2 (Acyl-CoA: cholesterol acyltransferases 2) levels, while individuals with low IMF had high ACAT2 levels. Significant changes took place in the pathways involved in coenzyme A, which are closely related to fat and cholesterol metabolism. Therefore, we speculate that ACAT2, as an important element involved in cholesterol metabolism, may become a potential molecular marker for the mechanism of pig intramuscular preadipocytes differentiation. Overexpression of ACAT2 in pig intramuscular pre-adipocytes can inhibit their differentiation, while adding ACAT2 inhibitor avasimibe can rescue the process. Knockdown of srebp2 or ldlr, which are two key genes closely related to ACAT2 and cholesterol metabolism, can inhibit pig intramuscular pre-adipocytes differentiation. Overall, our results suggest that ACAT2 is a novel negative regulator of intramuscular adipocyte differentiation through regulation of pparγ, cebpα signaling and srebp2/ldlr signaling involved in cholesterol metabolism.

Abstract:Objectives: The aim of this study was to investigate the effects of acorn diets on the composition of fatty acid (FA) and the intramuscular fat (IMF) content in Yuxi black pigs. Methods: Ninety Yuxi black pigs with similar body weight (99.60 ± 2.32 kg) were randomly divided into five groups. The control group was fed a basal diet, and the AD20, AD30, AD40, and AD50 groups were fed experimental diets which contained 20%, 30%, 40%, and 50% acorns, respectively. The feeding experiment lasted for 120 days. Results: The results showed that compared with the control group, the content of SFA in longissimus dorsi and biceps femoris tissues in the AD30 group decreased by 8.57% and 20.10%, and the content of MUFA increased by 5.40% and 15.83%, respectively, while the PUFA content of biceps femoris increased by 5.40% (p < 0.05). Meanwhile, the IMF content of the AD30 group was significantly higher than that of the control group in the longissimus dorsi and biceps femoris. In addition, the mRNA expression levels of the ATGL, PPARγ, and FABP4 genes in longissimus dorsi (p < 0.05) were up-regulated, and HSL were down-regulated (p < 0.05) in the AD30 group. In the biceps femoris of the AD30 group, it was observed that the expression levels of the ACC and FAS genes were up-regulated (p < 0.05), while HSL and ATGL genes were down-regulated (p < 0.05). Conclusions: These results demonstrated that the addition of appropriate amounts of acorn to the diet (a 30% acorn diet) could improve the nutritional value of pork.

Effects of γ-aminobutyric acid on growth performance and antioxidant capacity in finishing pigs housed in thermal environment

Abstract:The experiment was conducted to study the effects of γ-aminobutyric acid on performance and metabolism of finishing pigs in thermal environment.According to sex and body weight,48 three-way crossbred Duroc ×(Landrace ×Yorkshire)finishing pigs(43 kg) were randomly divided into 2 groups and each group had 3 replicates of 8 pigs.Pigs of the control group were fed basal diets and the treatment group were fed basal diets supplemented with γ-aminobutyric acid 10 mg/kg.The results showed that the treatment group showed better growth performance and its feed efficiency than the control group(P < 0.01).The serum urea nitrogen (SUN) concentration of the treatment group was not significantly difference from that of the control group(P > 0.05),and its serum glucose (GLU) concentration was higher than that of the control group(P < 0.05),and its lactate dehydrogenase (LDH) was lower than that of the control group(P < 0.05),creatine kinase (CK) was higher than that of the control group(P < 0.01).The content of malondialdehyde decreased(P < 0.05)while glutathione peroxidase activities increased(P < 0.01),but superoxide dismutase was not significantly difference from that of the control group(P > 0.05).It was concluded that γ-aminobutyric acid had better effect on improving growth performance and metabolism response and improved the ability of resisting heat stress in finishing pigs in thermal environment.
miR-152 targets pyruvate kinase to regulate the glycolytic activity of pig skeletal muscles and affects pork quality

Abstract:As a type of non-coding RNA, microRNAs are widely involved in the biological processes of animals. In the present study, the expression of miR-152 in glycolytic muscle fibers (Longissimus thoracis, LT) was lower than that of oxidative muscle fibers (Psoas major, PM). Using dual luciferase assay, miR-152 was shown to target muscle pyruvate kinase (PKM) to perform biological functions. Moreover, overexpression of miR-152 in primary porcine cells inhibited PKM gene expression and reduced lactic acid production in cells, whereas inhibition of miR-152 expression promoted PKM gene expression and increased lactic acid production. Correlation analysis showed that the expression of miR-152 was significantly positively correlated with the ultimate pH of LT after slaughter, while the expression of the PKM gene was significantly negatively correlated with the final pH of LT. In vivo and in vitro experiments discussed herein suggest that miR-152 may affect muscle pH by targeting the expression of the PKM gene. Our findings enrich the understanding of the genetic regulatory network that influences pork quality.
The Effects of 8-week Acetic Acid Feeding on Endurance Performance and Fat Metabolism in Skeletal Muscle of Mice

Abstract:PURPOSE:The combined effect of acetic acid supplementation and endurance exercise training on fatty acid metabolism and skeletal muscle functions are not well known. Therefore, the purpose of this study was to investigate the effects of 8-week acetic acid administration with or without endurance exercise training on fatty acid metabolism and skeletal muscle functions using mice.METHODS: Fourty-eight male wild-type ICR mice (10-week old) were randomly divided into 4 groups: sedentary control (Sed+Con), sedentary sodium acetate (Sed+NaAc), exercise control (Exe+Con) and exercise sodium acetate (Exe+NaAc) groups. For acetic acid diet, sodium acetate was incorporated into the chow diet at 5% (w/w). For the exercise training, mice performed 4 days/week of 20 min treadmill running exercise training for 8 weeks. Following 8 weeks of combined treatments of acetic acid and endurance running exercise training, visceral fat mass and skeletal muscle mass, blood parameters, and the markers for fatty acid metabolism were analyzed. The results were analyzed with one-way ANOVA (p<.05) using the SPSS 21 program.RESULTS: Eight weeks of Exe+NaAc treatment significantly increased maximal running time compared with Sed+Con and Exe+Con groups (p<.05). Eight weeks of Exe+NaAc treatment significantly decreased fatty acid synthesis-related FAS (fatty acid synthase) protein levels compared with the Sed+Con group, and increased fatty acid oxidation-related CPT1B (carnitine palmitoyltransferase 1B) protein levels compared with Sed+Con and Sed+NaAc groups (p<.05). This combined treatment of acetic acid and endurance exercise training also increased AMPK activation compared with the Sed+Con and the Sed+NaAc groups (p<.05).CONCLUSIONS: It was concluded that there were synergistic effects of the treatment of 8-week acetic acid supplementation and endurance exercise training on the reductions of body weight and visceral fat mass and an increase in endurance performance.
Apoptosis and autophagy of muscle cell during pork postmortem aging

Abstract:Objective: Pork is an important source of animal protein in many countries. Subtle physiochemical changes occur during pork postmortem aging. The changes of apoptosis and autophagy in pork at 6 h to 72 h after slaughter were studied to provide evidence for pork quality. Methods: In this article, morphological changes of postmortem pork was observed through Hematoxylin-eosin staining, apoptotic nuclei were observed by TdT-mediated dUTP nick end labeling assay, protein related to apoptosis and autophagy expressions were tested by western blot and LC3 level were expressed according to immunofluorescence assay. Results: In this study, we found the occurrence of apoptosis in postmortem pork, and the process was characterized by nucleus condensation and fragmentation, formation of apoptotic bodies, increase in apoptosis-related Bax/Bcl-2 levels, and activation of caspases. Autophagy reached its peak between 24 and 48 h after slaughter, accompanied by the formation of autophagosomes on the cell membrane and expression of autophagy-related proteins beclin-1, P62, LC3-I, LC3-II, and ATG5. Conclusion: Obvious apoptosis was observed at 12 h and autophagy reached its peak at 48 h. The present work provides the evidence for the occurrence of apoptosis and autophagy during postmortem aging of pork. In conclusion, the apoptosis and autophagy of muscle cells discovered in this study have important implications for pork in the meat industry.

Abstract:Abstract In this study, we explored the role of protein transcriptional modification (PTM) in the adipogenesis and fat accumulation of intramuscular adipocytes. Intramuscular fat (IMF) is mainly stored in adipocytes interspersed in the perimysial space or within fascicles, and its content is a key factor attributing to meat quality in farm animals, especially in terms of tenderness, flavor and juiciness. The early stage from the fetus to the neonate is crucial for skeletal muscle development involving myogenesis, adipogenesis, and fibrogenesis, and is susceptible to environmental factors. Previous studies have been subjected to explore the molecular mechanism involved in IMF development. However, it was obscured by muscular background of IMF adipocytes. Therefore, we investigated PTM dynamics from three aspects: 1) first analyzed PTM levels of lysine acetylation (Kac), 2-hydroxyisobutyrylation (Khib), crotonylation (Kcr) and succinylation (Ksu) in three distinct differentiation potenctial primary stem cells, including myogenic precursor cells (MPCs), fibro-adipogenic progenitors (FAPs) and MSCs isolated from neonatal longissimus dorsi muscle of pigs within 3-d of age; 2) Comparing the Khib levels of proliferative and differentiated FAPs; and 3) Exploring the role of Khib modification in the adipose tissue of obese- and lean-types of pigs. We found that only Khib level in FAPs was significantly greater than MSCs. During adipogenic differentiation of FAPs, the modification level of Khib was significantly increased quadratically in parallel with protein levels of acylation regulatory enzymes KAT5 (writer) and HDAC2 (Eraser). In comparison of Khib levels in FAPs in proliferation and in differentiation and Khib levels in backfat tissues from obese pigs (Laiwu pigs) and lean pigs (Duroc pigs), we also demonstrated that Khib modification level was positive correlated with adipogenic differentiation and fat accumulation. Accordingly, the expression patterns of KAT5 and HDAC2 were parallel to the degree of lipid accumulation in the backfat adipose tissue of finishing pigs. Furthermore, we showed that KAT5 knockdown in FAPs inhibited adipogenic differentiation, while HDAC2 knockdown enhanced adipogenic differentiation of FAPs. By employing C3H10T1/2 cells, we identified EON1 involved in Khib modification in regulation of adipo gesis and fat accumulation with analysis of Liquid chromatography-tandem mass spectrometry (LC-MS/MS) using the Tandem Mass Tag (TMT Through conditional loss- and gain-of-function mutations, we further demonstrated that khib modification of ENO1 is necessary for preadipocyte differentiation and subsequent fat accumulation by mediating cellular glycolysis. In conclusion, we are the first to reveal the role of Khib modification in adipogenesis and fat deposition in pigs, and provided new clues for the improvement of fat accumulation and distribution via genetic selection and nutritional strategy in pigs.

Abstract:Post-mortem muscle energy metabolism plays an important role in pork quality. To analyse the differences of meat quality and energy metabolism, three commercial pig crossbreeds frequently used in China were studied, they were DT (Duroc × Taihu; n = 16), PIC (five-way crossbreed from Pig Improvement Co., UK; n = 29) and DLY (Duroc × (Landrace × Yorkshire); n = 19) pigs. The results showed that DT pigs had a higher post-mortem pH45 min and pH24 h, lower shear force and drip loss, higher muscle free-glucose and glycogen contents, and lower lactic acid content than did PIC and DLY pigs. Post-mortem muscle free-glucose content of these three pig crossbreeds changed little, from 45 min to 96 h post-mortem. The expression levels of PRKAG3 (encoding a regulatory subunit of the AMP-activated protein kinase) and GYS1 (encoding muscle glycogen synthase) genes of DT pigs were significantly lower than those of PIC and DLY pigs. DT pigs had a higher expression level of glycogenin-1-like (encoding glycogenin) gene than did PIC and DLY pigs. In conclusion, DT pigs had better meat quality than did the other two pig crossbreeds. We deduced that the post-mortem muscle energy status and metabolism of DT pigs might be an important reason for their good meat quality, and future research should focus on the molecular and physiological mechanism of post-mortem muscle energy metabolism to find ways to improve meat quality.
Heat-induced formation of advanced glycation end-products in ground pork as affected by the addition of acetic acid or citric acid and the storage duration prior to the heat treatments

Abstract:The heat-induced (121 °C, 10 or 30 min) formation of two potentially hazardous advanced glycation end-products (AGEs), protein-bound Nɛ-carboxymethyllysine (CML) and Nɛ-carboxyethyllysine (CEL), in pork as affected by citric or acetic acid (0.5, 1 g/100 pork) and the storage duration (0 °C, 0 – 8 d) prior to the heating was investigated. A longer storage time of raw pork resulted in higher levels of AGEs produced during the later heating, likely due to the accumulation of some AGE precursors during the storage. Depending on the acid level and heating time, adding acid in pork led to 30 – 54% (citric acid) or 14 – 48% (acetic acid) average reduction of heat-induced production of CML/CEL, which corresponded to the reduction of thiobarbituric acid reactive substances and Schiff bases. The marinating time of raw pork with an acid did not significantly affect (P = 0.959 – 0.998) the acid’s inhibition effect on heat-induced formation of CML/CEL.

Abstract:The Laiwu pig is an indigenous fatty pig breed distributed in North China, characterized by an extremely high level of intramuscular fat (IMF) content (9% ~ 12%), but the regulatory mechanism underlying intramuscular fat deposition in skeletal muscle is still unknown. In this study, the TMT-labeled quantitative malonylome of the longissimus dorsi muscle in Laiwu pigs at the fastest IMF deposition stage (240 d vs 120 d) was compared to analyze the molecular mechanism of IMF variation in pigs. In Laiwu pigs aged 240 days/120 days, we identified 291 malonylated lysine sites across 188 proteins in the longissimus dorsi muscle. Among these, 38 sites across 31 proteins exhibited differential malonylation. Annotation analysis and enrichment analysis were performed for differentially malonylated proteins (DMPs). These DMPs were mainly clustered into 12 GO functional categories accounting for 5 biological processes, 4 cellular components and 3 molecular functions, and 2 signaling pathways by KEGG enrichment analysis. The function of differentially malonylated protein ACOT7 in the process of fat deposition was further investigated during the differentiation of 3 T3-L1 cells. The results showed that the protein level of ACOT7 in 3 T3-L1 cells decreased but the malonylated level of ACOT7 increased significantly. The malonyl-CoA that is synthesized by ACSF3 affected the malonylation level of ACOT7 in 3 T3-L1 cells. Significance: The intramuscular fat (IMF) content, by affecting sensory quality traits of meat, such as tenderness, flavor and juiciness, plays an important role in meat quality. Using TMT-based quantitative malonylated proteome analysis, we identified malonylated proteins in LD muscle samples in two stages (120 d and 240 d) of development and further identified differentially malonylated proteins, such as SLC25A4, ANXA5, TPM3 and ACOT7, that are associated with intramuscular fat deposition and fat metabolism in pigs. These differentially malonylated proteins could serve as candidates for elucidating the molecular mechanism of IMF deposition in pigs. In addition, we found that the malonyl-CoA in 3 T3-L1 cells is mainly synthesized by ACSF3, affecting the malonylated level of ACOT7. The study provides some data concerning the role of protein malonylation in regulating the variation in porcine IMF content. Abstract:The aim of this study was to investigate effects of dietary malic acid supplementation on skeletal muscle fiber-type transition during nursery period and the subsequent meat quality of finishing pigs. Results showed that malic acid supplementation for 28 days increased oxidative fiber percentage of weaned piglets, accompanied by the increased aerobic oxidation in serum and longissimus thoracis (LT) muscle. Additionally, activities of total antioxidant capacity and glutathione peroxidase in serum were increased. Moreover, dietary malic acid supplementation during nursery period tended to increase pH 24h and significantly decreased drip loss in LT muscle of finishing pigs. The content of total saturated fatty acid (SFA) and total monounsaturated fatty acid in LT muscle was significantly decreased, whereas the ratio of polyunsaturated fatty acid to SFA tended to increase. Together, dietary malic acid supplementation during nursery period can effectively increase antioxidant capacity and oxidative fibers percentage of weaned piglets, and further improve water holding capacity and nutritional values of pork in finishing pigs.

Identification of Metabonomics Changes in Longissimus Dorsi Muscle of Finishing Pigs Following Heat Stress through LC-MS/MS-Based Metabonomics Method

Abstract:Heat stress (HS) negatively affects meat quality by affecting material and energy metabolism, and exploring the mechanism underlying the muscle response to chronic HS in finishing pigs is important for the global pork industry. This study investigated changes in the metabolic profiles of the longissimus dorsi (LD) muscle of finishing pigs under high temperature using ultra-performance liquid chromatography coupled with electrospray ionization quadrupole time-of-flight mass spectrometry (UPLC-ESI-QTOF-MS) and multivariate data analysis (MDA). Castrated male DLY pigs (Duroc × Landrance × Yorkshire pigs, n = 24) from 8 litters were divided into three treatment groups: constant optimal ambient temperature at 22 °C and ad libitum feeding (CR, n = 8); constant high ambient temperature at 30 °C and ad libitum feeding (HS, n = 8); and constant optimal ambient temperature 22 °C and pair-feeding to the control pigs (PF, n = 8). The metabolic profile data from LD muscle samples were analyzed by MDA and external search engines. Nine differential metabolites (L-carnosine, acetylcholine, inosinic acid, L-carnitine, L-anserine, L-α-glycerylphosphorylcholine, acetylcarnitine, thiamine triphosphate, and adenosine thiamine diphosphate) were involved in antioxidant function, lipid metabolism, and cell signal transduction, which may decrease post mortem meat quality and play important roles in anti-HS. Four metabolites (L-carnosine, acetylcholine, inosinic acid, and L-carnitine) were verified, and it was indicated that the muscle L-carnitine content was significantly lower in HS than in CR (p < 0.01). The results show that constant HS affects the metabolites in the LD muscle and leads to coordinated changes in the endogenous antioxidant defense and meat quality of finishing pigs. These metabonomics results provide a basis for researching nutritional strategies to reduce the negative effects of heat stress on livestock and present new insights for further research.

Abstract:Abstractα-Ketoglutarate (AKG) is a crucial intermediate in the tricarboxylic acid (TCA) cycle and can be used for the production of ATP and amino acids in animal tissues. However, the effect of AKG on the expression patterns of genes involved in muscle protein metabolism is largely unknown, and the underlying mechanism remains to be elucidated. Therefore, we used young pigs to investigate the effects of a low crude protein (CP) diet and a low CP diet supplemented with AKG on protein accretion in their skeletal muscle. A total of 27 growing pigs with an initial body weight of 11.96 ± 0.18 kg were assigned randomly to one of the three diets: control (normal recommended 20% CP, NP), low CP (17% CP, LP), or low CP supplemented with 1% AKG (ALP). The pigs were fed their respective diets for 35 days. Free amino acid (AA) profile and hormone levels in the serum, and the expression of genes implicated in protein metabolism in skeletal muscle were examined. Results showed that compared with the control group or LP group, low-protein diets supplemented with AKG enhanced serum and intramuscular free AA concentrations, the mRNA abundances of AA transporters, and serum concentrations of insulin-like growth factor-1 (IGF-1), activated the mammalian target of rapamycin (mTOR) pathway, and decreased serum urea concentration and the mRNA levels for genes related to muscle protein degradation (P < 0.05). In conclusion, these results indicated that addition of AKG to a low-protein diet promotes amino acid synthesis in tissues and improves protein metabolism in skeletal muscle.

Abstract:L-malic acid is a vital intermediate in the citric acid cycle and has been reported to improve the antioxidant capacity and aerobic oxidation of weaned piglets; however, its application in finishing pigs is limited at present. This study explored the effects of dietary L-malic acid supplementation on the carcass traits and meat quality of finishing pigs. In a 45-day experiment, 192 Duroc × Landrace × Yorkshire pigs (75.01 ± 0.51 kg) were divided into four treatments, i.e., a basal diet supplemented with 0, 0.5%, 1%, and 2% L-malic acid complex. The results showed that L-malic acid supplementation had no effects on the growth performance of finishing pigs. Importantly, L-malic acid significantly increased redness (a*) value at 24-h postmortem (quadratic, p < 0.05) and tended to increase the proportion of oxymyoglobin (OMb) (quadratic, p = 0.10), as well as the total antioxidant capacity (T-AOC) activity (quadratic, p = 0.08) in the longissimus dorsi (LD) muscle. Further, dietary supplementation of 1% L-malic acid markedly increased the protein expression level of slow skeletal myosin heavy chain (MyHC) in the LD muscle (p < 0.05). Moreover, 0.5% and 2% L-malic acid supplementation significantly increased carcass length and loin eye area (p < 0.05). In conclusion, dietary L-malic acid could effectively improve the meat color and carcass traits of finishing pigs.

Effect of ultrasound-assisted L-lysine treatment on pork meat quality and myofibrillar protein properties during postmortem aging

Abstract:This paper aimed to investigate the effects of ultrasound-assisted L-lysine treatment on meat quality and myofibrillar proteins (MPs) properties of pork longissimus dorsi during postmortem aging. The results revealed that the L-lysine (Lys) and/or ultrasound treatment significantly increased (p < 0.05) the water-holding capacity and tenderness of the pork during postmortem aging, while the ultrasound-assisted Lys treatment had the lowest cooking loss, pressurization loss, Warner-Bratzler shear force, and hardness. In addition, L-lysine and/or ultrasound treatment increased (p < 0.05) pH value, T21, and myofibrillar fragmentation index, while the ultrasound-assisted Lys treatment had the highest value. Meanwhile, the protein solubility was increased with Lys and/or ultrasound treatment during postmortem aging, and ultrasound-assisted Lys treatment had the highest solubility, reaching 88.19%, 92.98%, and 91.73% at 0, 1, and 3 days, respectively. The result of protein conformational characteristics showed that Lys and/or ultrasound treatment caused the unfolding of the α-helix structure, resulting in the exposure of more hydrophobic amino acids and buried sulfhydryl groups, ultimately enhancing MPs solubility. In summary, ultrasound-assisted Lys treatment altered the structure of MPs, resulting in the enhancement of the water-holding capacity and tenderness of the pork. PRACTICAL APPLICATION: This study showed that ultrasound-assisted L-lysine (Lys) treatment could enhance the water-holding capacity and tenderness of pork during postmortem aging. The results might provide a reference for the application of ultrasound-assisted Lys treatment on the improvement of pork meat quality. To facilitate practical applications in production, the development of medium and large-sized ultrasound equipment for conducting small-scale and pilot experiments is crucial for future research.

Abstract:This study aimed to investigate effects of apple polyphenols (APPs) on myofiber-type transformation in longissimus dorsi muscle of finishing pigs and its mechanism. In this study, 36 healthy castrated Duroc × Landrace × Yorkshire pigs with an average body weight of 71.25 ± 2.40 kg were randomly divided into three treatment groups (control, 0.04% APPs, 0.08% APPs). The experiment lasted for 49 days. Results showed that dietary APP supplementation increased the protein expression of MyHC I and the activities of succinic dehydrogenase and malate dehydrogenase, as well as decreased the protein expression of MyHC IIb and the activity of lactate dehydrogenase, suggesting that APPs promoted muscle fiber-type transformation from fast-twitch to slow-twitch in finishing pigs. We also showed that dietary 0.08% APP supplementation increased the expressions of mitochondrial biogenesis and function-related proteins PGC-1α, Sirt1 and Cytc. In addition, dietary supplementation with 0.08% APPs increased the activities of T-SOD, GSH-PX and CAT and decreased the MDA content. Together, we provided the first evidence that APP promotes muscle fiber-type transformation from fast-twitch to slow-twitch in finishing pigs, which may be achieved by improving the mitochondrial biogenesis and function and increasing the antioxidant capacity of skeletal muscle.
