Abstract:Calcium carbonate platforms produced by reef-building stony corals over geologic time are pervasive features around the world [1]; however, the mechanism by which these organisms produce the mineral is poorly understood (see review by [2]). It is generally assumed that stony corals precipitate calcium carbonate extracellularly as aragonite in a calcifying medium between the calicoblastic ectoderm and pre-existing skeleton, separated from the overlying seawater [2]. The calicoblastic ectoderm produces extracellular matrix (ECM) proteins, secreted to the calcifying medium [3-6], which appear to provide the nucleation, alteration, elongation, and inhibition mechanisms of the biomineral [7] and remain occluded and preserved in the skeleton [8-10]. Here we show in cell cultures of the stony coral Stylophora pistillata that calcium is concentrated in intracellular pockets that are subsequently exported from the cell where a nucleation process leads to the formation of extracellular aragonite crystals. Analysis of the growing crystals by lattice light-sheet microscopy suggests that the crystals elongate from the cells' surfaces outward.

Quantitative Interpretation of Vertical Profiles of Calcium and Ph in the Coral Coelenteron

Abstract:Scleratinian corals (hard corals) and their symbiotic algae are the ecological engineers of biodiverse and geological important coral reef habitats. The complex, linked physiological processes that enable the holobiont (coral + algae) to calcify and generate reef structures are consequently of great interest. However, the mechanism of calcification is difficult to study for several reasons including the small spatial scales of the processes and the close coupling between the symbiont and host. In this study, we explore the utility of pH and Ca2+ microelectrodes for constraining the rates and spatial distribution of photosynthesis, respiration, and calcification. The work focuses on vertical profiles of pH and Ca2+ through the coelenteron cavity, a semi-isolated compartment of modified seawater amenable to quantitative interpretation. In two studied species, Turbinaria reniformis and Acropora millepora, Ca2+ concentrations decreased in a roughly linear manner from the mouth to the base of the coelenteron, indicating the primary physiological process affecting Ca2+ concentration is removal for calcification below the coelenteron. In contrast, the H+ concentration remained relatively constant over much of the coelenteron cavity before it increased sharply toward the base of the coelenteron, indicative of proton-pumping from the calcification fluid below. The estimated H+ gradient between the coelenteron cavity and the calcification site was >10 times higher than previously predicted. Consequently, the energy required to export protons from the calcifying fluid was estimated to be ~3 times higher than previously calculated. A one-dimensional reaction-diffusion model was used to interpret the pH profile considering the effects of photosynthesis, respiration, and calcification. This model provided a good fit to the observed pH profile and helped to constrain the rates and spatial distribution of these processes. Our modeling results also suggested that adult corals with deeper polyps may be more sensitive to ocean acidification (OA) because of enhanced difficulty to transport H+ out of the coelenteron and into the surrounding seawater.
Molecular and skeletal fingerprints of scleractinian coral biomineralization: From the sea surface to mesophotic depths

Abstract:Reef-building corals, the major producers of biogenic calcium carbonate, form skeletons in a plethora of morphological forms. Here we studied skeletal modifications of Stylophora pistillata (clade 4) colonies that adapt to increasing depths with decreasing ambient light. The coral show characteristic transitions from spherical morphologies (shallow depths, 5 m deep) to flat and branching geometries (mesophotic depths, 60 m deep). Such changes are typically ascribed to the algal photosymbiont physiological feedback with the coral that host them. We find specific fine-scale skeletal variability in accretion of structure at shallow- and mesophotic depth morphotypes that suggest underlying genomic regulation of biomineralization pathways of the coral host. To explain this, we conducted comparative morphology-based analyses, including optical and electron microscopy, tomography and X-ray diffraction analysis coupled with a comprehensive transcriptomic analysis of S. pistillata. The samples originated from Gulf of Eilat in the Red Sea collected along a depth gradient from shallow to mesophotic depths (5 to 60 m). Additional samples were experimentally transplanted from 5 m to 60 m and from 60 m to 5 m. Interestingly, both morphologically and functionally, transplanted corals partly adapt by exhibiting typical depth-specific properties. In mesophotic depths, we find that the organic matrix fraction is enriched in the coralla, well matching the overrepresentation of transcripts encoding biomineralization "tool-kit" structural extracellularproteins that was observed. These results provide insights into the molecular mechanisms of calcification and skeletal adaptation that repeatedly allowed this coral group to adapt to a range of environments presumably with a rich geological past. STATEMENT OF SIGNIFICANCE: Understanding the reef coral physiological plasticity under a rapidly changing climate is of crucial importance for the protection of coral reef ecosystems. Most of the reef corals operate near their upper limit of heat tolerance. A possible rescue for some coral species is migration to deeper, cooler mesophotic depths. However, gradually changing environmental parameters (especially light) along the depth gradient pose new adaptative stress on corals with largely unknown influences on the various biological molecular pathways. This work provides a first comprehensive analysis of changes in gene expression, including biomineralization "tool kit" genes, and reports the fine-scale microstructural and crystallographic skeletal details in S. pistillata collected in the Red Sea along a depth gradient spannign 5 to 60 m.

Abstract:In spite of more than one century and half of studies, mechanisms of coral biomineralization, leading to coral growth and reef formation, still remain poorly known, although major global threats to coral reefs, such as ocean acidification, primarily affect this process. Coral skeletons are used as environmental archives but the vital processes that govern incorporation of trace elements and stable isotope are still unknown. Our knowledge on coral physiology is restricted to the organismal level due to the lack of appropriate cell model, however the advent of new approaches, such as coral genomic, is changing drastically our knowledge on these animals even if only a few data are available concerning the field of biomineralization. This chapter reviews our present knowledge and discusses the different theories on coral calcification, from the molecular to the reef level. Conclusion is presented in a list of key issues to be resolved in order to understand the intimate mechanisms of calcification of corals, essential to determine the origin of the sensitivity of corals to ocean acidification, to improve paleoceanographic reconstructions or coral reef management, or “just” to understand how genes of a soft organism control the formation of an extracellular 3D-skeleton.
A role for primary cilia in coral calcification?

Abstract:Abstract Cilia are evolutionarily conserved organelles that extend from the surface of cells and are found in diverse organisms from protozoans to multicellular organisms. Motile cilia play various biological functions by their beating motion, including mixing fluids and transporting food particles. Non-motile cilia act as sensors that signal cells about their microenvironment. In corals, cilia have been described in some of the cell layers but never in the calcifying epithelium, which is responsible for skeleton formation. In the present study, we used scanning electron microscopy and immunolabelling to investigate the cellular ciliature of the different tissue layers of the coral Stylophora pistillata , with a focus on the calcifying calicoblastic ectoderm. We show that the cilium of the calcifying cells is different from the cilium of the other cell layers. It is much shorter, and more importantly, its base is structurally distinct from the base observed in cilia of the other tissue layers. Based on these structural observations, we conclude that the cilium of the calcifying cells is a primary cilium. From what is known in other organisms, primary cilia are sensors that signal cells about their microenvironment. We discuss the implications of the presence of a primary cilium in the calcifying epithelium for our understanding of the cellular physiology driving coral calcification and its environmental sensitivity.

Abstract:Symbiotic algae of primary polyps play an important role in calcification of coral skeletons. However, the function of the symbiotic algae, including the way they influence the physical features of their host skeleton under various conditions, is not well understood. We used X‐ray microcomputed tomography to observe skeletal shape characteristics in symbiotic and aposymbiotic primary polyps of Acropora digitifera that were cultured at various temperature and pCO2 levels (temperature 27, 29, 33°C; pCO2 400, 800, 1000 µatm). Symbiotic polyps had a basal plate with a well‐developed folding margin supporting the branched skeleton, whereas aposymbiotic ones did not. The features of the folding margin suggest that it might be the initial growth stage of the epitheca. In addition, three‐dimensional (3‐D) morphological measurements made by X‐ray microcomputed tomography show that the branched skeletons of symbiotic primary polyps were taller than those of aposymbiotic ones, suggesting that zooxanthellae in coral primary polyps play a critical role in the height growth of skeletal branches. Furthermore, results of the temperature‐ and pCO2‐controlled experiments suggest that global warming might greatly affect the activity of zooxanthellae, whereas ocean acidification might reduce calcification by damaging the coral host itself. Our findings provide new knowledge about the role of zooxanthellae in coral calcification.

Abstract:Research purpose and findings: Coralline algae are key biological substrates of many carbonate systems globally. Their capacity to build enduring crusts that underpin the formation of tropical reefs, rhodolith beds and other benthic substrate is dependent on the formation of a calcified thallus. However, this important process of skeletal carbonate formation is not well understood. We undertook a study of cellular carbonate features to develop a model for calcification. We describe two types of cell wall calcification; 1) calcified primary cell wall (PCW) in the thin-walled elongate cells such as central medullary cells in articulated corallines and hypothallial cells in crustose coralline algae (CCA), 2) calcified secondary cell wall (SCW) with radial Mg-calcite crystals in thicker-walled rounded cortical cells of articulated corallines and perithallial cells of CCA. The distinctive banding found in many rhodoliths is the regular transition from PCW-only cells to SCW cells. Within the cell walls there can be bands of elevated Mg with Mg content of a few mol% higher than radial Mg-calcite (M-type), ranging up to dolomite composition (D-type). Model for calcification: We propose the following three-step model for calcification. 1) A thin (< 0.5 μm) PCW forms and is filled with a mineralising fluid of organic compounds and seawater. Nanometer-scale Mg-calcite grains precipitate on the organic structures within the PCW. 2) Crystalline cellulose microfibrils (CMF) are extruded perpendicularly from the cellulose synthase complexes (CSC) in the plasmalemma to form the SCW. 3) The CMF soaks in the mineralising fluid as it extrudes and becomes calcified, retaining the perpendicular form, thus building the radial calcite. In Clathromorphum, SCW formation lags PCW creating a zone of weakness resulting in a split in the sub-surface crust. All calcification seems likely to be a bioinduced rather than controlled process. These findings are a substantial step forward in understanding how corallines calcify.

Abstract:The coral reef ecosystem faces increasing threats under global climate challenges. One of the core issues is the inability of fragments to quickly grow into a size that can resist environmental pressures in coral transplantation. The observation of accelerated growth during the early stages of coral regeneration provides new insights for addressing this challenge. To investigate related molecular mechanisms, our study focused on the fast-growing stony coral (with chromosome-scale reference genome). Employing diverse techniques, including single-cell RNA sequencing (scRNA-seq), we unveiled related intricate cellular dynamics. Single-cell analysis revealed notable shifts in calicoblasts and epidermal cells around 2-4 weeks post-injury. Gene expression analysis revealed enrichment in immune response and biomineralization pathways. Pseudotime analysis explained the differentiation of epidermal cells into calicoblasts, while time-course analysis identified key genes associated with dynamic biomineralization changes. This study enhances our understanding of coral regeneration, offering insights for protective strategies to foster coral growth.

From particle attachment to space-filling coral skeletons

Abstract:Reef-building corals and their aragonite (CaCO3) skeletons support entire reef ecosystems, yet their formation mechanism is poorly understood. Here we used synchrotron spectromicroscopy to observe the nanoscale mineralogy of fresh, forming skeletons from six species spanning all reef-forming coral morphologies: Branching, encrusting, massive, and table. In all species, hydrated and anhydrous amorphous calcium carbonate nanoparticles were precursors for skeletal growth, as previously observed in a single species. The amorphous precursors here were observed in tissue, between tissue and skeleton, and at growth fronts of the skeleton, within a low-density nano- or microporous layer varying in thickness from 7 to 20 µm. Brunauer-Emmett-Teller measurements, however, indicated that the mature skeletons at the microscale were space-filling, comparable to single crystals of geologic aragonite. Nanoparticles alone can never fill space completely, thus ion-by-ion filling must be invoked to fill interstitial pores. Such ion-by-ion diffusion and attachment may occur from the supersaturated calcifying fluid known to exist in corals, or from a dense liquid precursor, observed in synthetic systems but never in biogenic ones. Concomitant particle attachment and ion-by-ion filling was previously observed in synthetic calcite rhombohedra, but never in aragonite pseudohexagonal prisms, synthetic or biogenic, as observed here. Models for biomineral growth, isotope incorporation, and coral skeletons' resilience to ocean warming and acidification must take into account the dual formation mechanism, including particle attachment and ion-by-ion space filling.

Abstract:The formation of skeletal structures composed of different calcium carbonate polymorphs (e.g. aragonite and calcite) appears to be both biologically and environmentally regulated. Among environmental factors influencing aragonite and calcite precipitation, changes in seawater conditions—primarily in the molar ratio of magnesium and calcium during so‐called 'Calcite' (mMg:mCa below 2) or 'Aragonite' seas (mMg:mCa above 2)—have had profound impacts on the distribution and performance of marine calcifiers throughout Earth's history. Nonetheless, the fossil record shows that some species appear to have counteracted such changes and kept their skeleton polymorph unaltered. Here, the aragonitic octocoral Heliopora coerulea and the aragonitic scleractinian Montipora digitata were exposed to Calcite Sea‐like mMg:mCa with various levels of magnesium and calcium concentration, and changes in both the mineralogy (i.e. CaCO3 polymorph) and gene expression were monitored. Both species maintained aragonite deposition at lower mMg:mCa ratios, while concurrent calcite presence was only detected in M. digitata. Despite a strong variability between independent experimental replicates for both species, the expression for a set of putative calcification‐related genes, including known components of the M. digitata skeleton organic matrix (SkOM), was found to consistently change at lower mMg:mCa. These results support the previously proposed involvements of the SkOM in counteracting decreases in seawater mMg:mCa. Although no consistent expression changes in calcium and magnesium transporters were observed, down‐regulation calcium channels in H. coerulea in one experimental replicate and at an mMg:mCa of 2.5, pointing to a possible active calcium uptake regulation by the corals under altered mMg:mCa.

Abstract:Abstract Boron isotopic and elemental analysis of coral aragonite can give important insights into the calcification strategies employed in coral skeletal construction. Traditional methods of analysis have limited spatial (and thus temporal) resolution, hindering attempts to unravel skeletal heterogeneity. Laser ablation mass spectrometry allows a much more refined view, and here we employ these techniques to explore boron isotope and co-varying elemental ratios in the tropical coral Siderastrea siderea . We generate two-dimensional maps of the carbonate parameters within the calcification medium that deposited the skeleton, which reveal large heterogeneities in carbonate chemistry across the macro-structure of a coral polyp. These differences have the potential to bias proxy interpretations, and indicate that different processes facilitated precipitation of different parts of the coral skeleton: the low-density columella being precipitated from a fluid with a carbonate composition closer to seawater, compared to the high-density inter-polyp walls where aragonite saturation was ~ 5 times that of external seawater. Therefore, the skeleton does not precipitate from a spatially homogeneous fluid and its different parts may thus have varying sensitivity to environmental stress. This offers new insights into the mechanisms behind the response of the S. siderea skeletal phenotype to ocean acidification.

Abstract:Understanding the mechanisms of coral calcification is critical for accurately projecting coral reef futures under ocean acidification and warming. Recent suggestions that calcification is primarily controlled by organic molecules and the biological activity of the coral polyp imply that ocean acidification may not affect skeletal accretion. The basis for these suggestions relies heavily on correlating the presence of organic matter with the orientation and disorder of aragonite crystals in the skeleton, carrying the assumption that organic matter observed in the skeleton was produced by the polyp to control calcification. Here we use Raman spectroscopy to test whether there are differences in organic matter content between coral skeleton and abiogenic aragonites precipitated from seawater, both before and after thermal annealing (heating). We measured the background fluxorescence and intensity of C-H bonding signals in the Raman spectra, which are commonly attributed to coral polyp-derived skeletal organic matrix (SOM) and have been used to map its distribution. Surprisingly, we found no differences in either fluorescence or C-H bonding between abiogenic aragonite and coral skeleton. Annealing reduced the molecular disorder in coral skeleton, potentially due to removal of organic matter, but the same effect was also observed in the abiogenic aragonites. The presence of organic molecules in the abiogenic aragonites is further supported by measurements of N content and δ15N. Together, our data suggest that some of what has been interpreted in previous studies as polyp-derived SOM may actually be seawater-sourced organic matter or some other signal not unique to biogenic aragonite. Finally, we create a high-resolution Raman map of a Pocillopora skeleton to demonstrate how patterns of fluorescence and elevated calcifying fluid aragonite saturation state (ΩAr) along centers of calcification are consistent with both biological and physico-chemical controls. Our aim is to advance discussion on biological mediation of calcification and the implications for coral resilience in a high-CO2 world.

Abstract:Octocorallia (class Anthozoa, phylum Cnidaria) is a group of calcifying corals displaying a wide diversity of mineral skeletons. This includes skeletal structures composed of different calcium carbonate polymorphs (aragonite and calcite). This represents a unique feature among anthozoans, as scleractinian corals (subclass Hexacorallia), main reef builders and focus of biomineralization research, are all characterized by an aragonite exoskeleton. From an evolutionary perspective, the presence of aragonitic skeletons in Octocorallia is puzzling as it is observed in very few species and has apparently originated during a Calcite sea (i.e., time interval characterized by calcite-inducing seawater conditions). Despite this, octocorals have been systematically overlooked in biomineralization studies. Here we review what is known about octocoral biomineralization, focusing on the evolutionary and biological processes that underlie calcite and aragonite formation. Although differences in research focus between octocorals and scleractinians are often mentioned, we highlight how strong variability also exists between different octocoral groups. Different main aspects of octocoral biomineralization have been in fact studied in a small set of species, including the (calcitic) gorgonian Leptogorgia virgulata and/or the precious coral Corallium rubrum. These include descriptions of calcifying cells (scleroblasts), calcium transport and chemistry of the calcification fluids. With the exception of few histological observations, no information on these features is available for aragonitic octocorals. Availability of sequencing data is also heterogeneous between groups, with no transcriptome or genome available, for instance, for the clade Calcaxonia. Although calcite represents by far the most common polymorph deposited by octocorals, we argue that studying aragonite-forming could provide insight on octocoral, and more generally anthozoan, biomineralization. First and foremost it would allow to compare calcification processes between octocoral groups, highlighting homologies and differences. Secondly, similarities (exoskeleton) between Heliopora and scleractinian skeletons, would provide further insight on which biomineralization features are driven by skeleton characteristics (shared by scleractinians and aragonitic octocorals) and those driven by taxonomy (shared by octocorals regardless of skeleton polymorph). Including the diversity of anthozoan mineralization strategies into biomineralization studies remains thus essential to comprehensively study how skeletons form and evolved within this ecologically important group of marine animals.

Abstract:The coralligenous build-ups located on the Mediterranean shelf in front of Marzamemi (SE Sicily, Italy) represent useful natural examples to use in studying the relationship between skeletal organisms and non-skeletal components in marine bioconstructions. Coralligenous build-ups are formed in open marine systems, and their comparison with coeval bioconstructions (biostalactites) of confined environments, like submarine caves, allows depicting the complex interactions between metazoans and microbial communities in the formations of recent bioconstructions in different Mediterranean settings. In this study, two coralligenous build-ups were characterized in terms of organisms and sediments involved in their formation. The framework mainly consists of coralline algae and subordinate bryozoans and serpulids. Sponges affect the general morphology of the bioconstructions both interacting with skeletonized organisms and through bioerosion activity. The micrite or microcrystalline calcite is present in minor amounts compared to other components that form the build-ups and consists of two types: autochthonous (in situ) and allochthonous (detrital). Fine autochthonous micrite mineralized directly inside the framework cavities and shows aphanitic or peloidal fabric, produced by organomineralization processes of soft sponge tissues and microbial metabolic activity, respectively. The detrital micrite occurring inside cavities derives from external sources or erosion processes of the bioconstructions themselves. This component has been classified as organic or inorganic based on the organic matter contents deduced by UV epifluorescence. A great quantity of sponges live in cavities of the coralligenous build-ups and compete with carbonatogenic bacteria for the same cryptic spaces, limiting the production of microbialites. The sharing of a similar relationship between sponges and microbial communities by coralligenous concretion and biotic crusts of particular submarine caves suggests that this competition is not habitat-specific. On the contrary, it may develop in a range of environmental settings, from open to cryptic systems, and could be used to clarify the role of metazoans vs. microbialites in palaeoecological reconstructions.

Abstract:Biocalcification encompasses the kinetic and structural, abiotic and biologically mediated processes involved in the formation of calcium carbonate skeletons by marine organisms and represents a key process in the global carbon cycle. Throughout the geological record, this process has evolved repetitively and has altered global biogeochemical cycles. Besides the structural variability of calcium carbonate polymorphs laid down by different organisms, biogenic carbonate skeletons are characterized by the presence of organic molecules that are incorporated into the growing skeleton. Major advances have identified the macromolecules associated to the organic matrix within marine calcifiers, however, it has yet to be established the actual role these organic molecules play in the calcification process. In this study, we isolated the effect of skeletal organic molecules (SOM) on the precipitation of calcium carbonate on coral skeleton fragments by adding extracted SOM or coral mucus (CM) to oversaturated calcium carbonate solutions. We found that the precipitation rate did not change regardless if organic molecules were present or not. However, the primary polymorph did change between the treatments, suggesting that organic molecules influence the surface processes that lead to the formation of the crystal lattice but not the kinetic processes that transport ions to the crystal surface. Since SOM and CM both altered the crystal polymorph but not the crystallization rate, we argue that SOM may not represent a specialized biomineralization toolkit, but that SOM originate from CM and the requirement of the polyp to adhere to the substratum.

Abstract:The sources and mechanisms of inorganic carbon transport for scleractinian coral calcification and photosynthesis were studied using a double labelling technique with H(14)CO(3) and (45)Ca. Clones of Stylophora pistillata that had developed into microcolonies were examined. Compartmental and pharmacological analyses of the distribution of(45)Ca and H(14)CO(3) in the coelenteron, tissues and skeleton were performed in dark or light conditions or in the presence of various seawater HCO(3)(−) concentrations. For calcification, irrespective of the lighting conditions, the major source of dissolved inorganic carbon (DIC) is metabolic CO(2) (70–75% of total CaCO(3) deposition), while only 25–30% originates from the external medium (seawater carbon pool). These results are in agreement with the observation that metabolic CO(2) production in the light is at least six times greater than is required for calcification. This source is dependent on carbonic anhydrase activity because it is sensitive to ethoxyzolamide. Seawater DIC is transferred from the external medium to the coral skeleton by two different pathways: from sea water to the coelenteron, the passive paracellular pathway is largely sufficient, while a DIDS-sensitive transcellular pathway appears to mediate the flux across calicoblastic cells. Irrespective of the source, an anion exchanger performs the secretion of DIC at the site of calcification. Furthermore, a fourfold light-enhanced calcification of Stylophora pistillata microcolonies was measured. This stimulation was only effective after a lag of 10 min. These results are discussed in the context of light-enhanced calcification. Characterisation of the DIC supply for symbiotic dinoflagellate photosynthesis demonstrated the presence of a DIC pool within the tissues. The size of this pool was dependent on the lighting conditions, since it increased 39-fold after 3 h of illumination. Passive DIC equilibration through oral tissues between sea water and the coelenteric cavity is insufficient to supply this DIC pool, suggesting that there is an active transepithelial absorption of inorganic carbon sensitive to DIDS, ethoxyzolamide and iodide. These results confirm the presence of CO(2)-concentrating mechanisms in coral cells. The tissue pool is not, however, used as a source for calcification since no significant lag phase in the incorporation of external seawater DIC was measured.

Abstract:Different techniques have been used to characterize the physical and chemical structure of the red coral calcitic skeleton. A section normal to the axis of the skeleton shows a medullar zone surrounded by a circular domain composed of concentric rings. Growth rings are revealed by the cyclic variation of organic matter (OM) and Mg/Ca ratio. These growth rings are annual; thus, both OM and Mg/Ca ratio can be used to date red coral colonies. Growth rings display wavelets. The internal structure of each wavelet results from the stacking of layers with tortuous interfaces. Tortuosity is due to the presence of microprotuberances. Interfaces between layers may display sharp discontinuities indicative of interruption of the mineralizing process. SEM and TEM studies show that each layer is made of (1) fibers, organized or not in fan-shaped structures; and (2) submicrometer (apparently mono-) crystalline units. Fibers are superstructures made of submicrometer units possibly assembled by an oriented aggregation mechanism. HRTEM studies show that in spite of displaying single-crystal scattering behavior, the submicrometer crystalline units are made of 2-5 nm nanograins again possibly aggregated by a mechanism of oriented attachment. Thus, submicrometer crystalline units and polycrystalline fibers can be both defined as mesocrystals. The red coral skeleton is a hierarchically organized organic-inorganic composite that exhibits porosity and structural and compositional order on length scales from the nanoscale to the macroscale.

Abstract:Coral reefs, the largest bioconstruction on Earth, are formed by calcium carbonate skeletons of corals. Coral skeleton formation commonly referred to as calcification occurs in a specific compartment, the extracellular calcifying medium (ECM), located between the aboral ectoderm and the skeleton. Calcification models often assume a direct link between the surrounding seawater and the ECM. However, the ECM is separated from the seawater by several tissue layers and the cœlenteron, which contains the cœlenteric fluid found in both polyps and cœnosarc (tissue connecting the polyps). Symbiotic dinoflagellate-containing cells line the cœlenteron and their photosynthetic activity contributes to changes in the chemistry of the cœlenteric fluid, particularly with respect to pH. The aim of our study is to compare cœlenteron pH between the cœnosarc and polyps and to compare areas of high or low dinoflagellate density based on tissue coloration. To achieve this, we use liquid ion exchange (LIX) pH microsensors to profile pH in the cœlenteron of polyps and the cœnosarc in different regions of the coral colony in light and darkness. We interpret our results in terms of what light and dark exposure means for proton gradients between the ECM and the coelenteron, and how this could affect calcification.
