Abstract:As biomaterials, it is essential for polyurethane (PU) to have excellent antibacterial activity and biocompatibility at the same time. Herein, quaternary ammonium groups, sulfonic groups, and alkyl groups simultaneously introduced hyperbranched poly(amidoamine) (HPA-QSA) was synthesized, which was further used to blend with PU and prepared as films. HPA-QSA/PU blend films showed strong antibacterial activity to kill more than 99.99% of both Gram-positive and Gram-negative bacteria due to the quaternary ammonium groups and long alkyl groups, as well as good biocompatibility through protein-resistance, cytotoxicity, hemolysis assays under the combined action of cations and anions. Moreover, they exhibited stable antibacterial activity and low cytotoxicity in aquatic solutions. Overall, HPA-QSA/PU films prepared by facile blending process exhibit great promise as low-cost, effective antibacterial, good biocompatible materials, indicating an appealing prospect in biomedical fields.
Polydopamine/poly(sulfobetaine Methacrylate) Co-deposition Coatings Triggered by CuSO4/H2O2 on Implants for Improved Surface Hemocompatibility and Antibacterial Activity

Abstract:Implanted biomaterials such as medical catheters are prone to be adhered by proteins, platelets and bacteria due to their surface hydrophobicity characteristics, and then induce related infections and thrombosis. Hence, the development of a versatile strategy to endow surfaces with antibacterial and antifouling functions is particularly significant for blood-contacting materials. In this work, CuSO4/H2O2 was used to trigger polydopamine (PDA) and poly-(sulfobetaine methacrylate) (PSBMA) co-deposition process to endow polyurethane (PU) antibacterial and antifouling surface (PU/PDA(Cu)/PSBMA). The zwitterions contained in the PU/PDA(Cu)/PSBMA coating can significantly improve surface wettability to reduce protein adsorption, thereby improving its blood compatibility. In addition, the copper ions released from the metal-phenolic networks (MPNs) imparted them more than 90% antibacterial activity against E. coli and S. aureus. Notably, PU/PDA(Cu)/PSBMA also exhibits excellent performance in vivo mouse catheter-related infections models. Thus, the PU/PDA(Cu)/PSBMA has great application potential for developing multifunctional surface coatings for blood-contacting materials so as to improve antibacterial and anticoagulant properties.

Abstract:Recent advancements in bioengineering and medical devices have been greatly influenced and dominated by synthetic polymers, particularly polyurethanes (PUs). PUs offer customizable mechanical properties and long‐term stability, but their inherent hydrophobic nature poses challenges in practically biological application processes, such as interface high friction, strong protein adsorption, and thrombosis. To address these issues, surface modifications of PUs for generating functionally hydrophilic layers have received widespread attention, but the durability of generated surface functionality is poor due to irreversible mechanical wear or biodegradation. As a result, numerous researchers have investigated bulk modification techniques to incorporate zwitterionic polymers or groups onto the main or side chains of PUs, thereby improving their hydrophilicity and biocompatibility. This comprehensive review presents an extensive overview of notable zwitterionic PUs (ZPUs), including those based on phosphorylcholine (PC), sulfobetaine (SB), and carboxybetaine (CB). The review explores their wide range of biomedical applications, from blood‐contacting devices to antibacterial coatings, fouling‐resistant marine coatings, separation membranes, lubricated surfaces, and shape memory and self‐healing materials. Bioinspired Polycation Functionalization of the Polyurethane Surface for Enhanced Lubrication, Antibacterial Property, and Anticoagulation

Abstract:Endowing the implanted biomaterial with a wide range of functions, such as lubrication and antibacterial and anticoagulation properties, is essential for their safe clinical use. In the present study, based on the superlubrication mechanism of articular cartilage, a branched polyelectrolyte polymer (PEI-PMPC) was successfully synthesized based on tert-butyl hydroperoxide-initiated grafting polymerization and then applied to the biocompatible polyurethane (PU) surface. Specifically, the PU sheet was pretreated by polydopamine (PDA), and the PEI-PMPC polymer was grafted onto the PDAmodified PU surface by Schiff base reaction to generate a solid PDA/ PEI-PMPC coating. The unmodified and modified surfaces were characterized by Fourier transform infrared spectroscopy, water contact angle, X-ray photoelectron spectroscopy, and surface zeta potential. To compare the lubrication, antibacterial behavior, and hemocompatibility, the relevant properties of the modified surfaces were evaluated by the tribological test, plate count method, surface morphology, hemolysis rate, activated partial thromboplastin time, and thrombin time. The experimental results demonstrated that the adhesion of bovine serum albumin, Escherichia coli, Staphylococcus aureus, platelets, and red blood cells was significantly decreased on the PEI-PMPC-modified PU surface compared with that of the unmodified PU surface. The multifunctional properties of the coating were attributed to the hydration layer formed by zwitterionic phosphorylcholine groups, and additionally, the cationic PEI served to kill the bacteria adhering to the surface. In summary, the bioinspired coating proposed in this study could potentially be used as a promising surface functionalization strategy for biomedical implants.

Abstract:Bacterial infection is a key challenge for implant and blood-contacting biomaterials due to the biofouling and thrombus formation at their interface. Therefore, design and fabrication of high-efficiency antifouling and bactericidal coatings on substrates are extremely important for biomedical devices. In this paper, we successfully developed a coating with high-efficiency antifouling and bactericidal properties on a polyurethane (PU) substrate through polydopamine (PDA), polyethyleneimine (PEI) assisted codeposition and amidation reaction using our synthesized copolymers of mono-2-(methylpropoxy) ethyl succinate (MSA), sulfobetaine methacrylate (SBMA) and/or ethyl methacrylate trimethyl ammonium chloride (MAC). All the coatings showed improved hydrophilicity. And due to the unique surface hydration of sulfobetaine (SB) zwitterionic groups and high ability to kill attached bacteria of quaternary ammonium cationic groups, the coatings constructed by tripolymers of MSA/MAC/SBMA (defined as PCAS) showed greatly reduced protein adsorption (BSA and Fg), platelet and cell adhesion, and excellent antifouling activity. This bioinspired and efficient surface modification strategy with dual-function coatings has potential application prospects in implantable biomedical devices.

Abstract:A new-type sulphonated polymer surface obtained by the solution technique is reported. The surface properties of the mortified polymer surfaces are characterized by SEM and ESCA, and their in vitro blood compatibility is evaluated in terms of TT, PRT, PTT, and platelet adhesion. The modified polymer surface is found to be the least thrombogenic when the modifying polymer molecule contains hydrophilic PEO chains. The PEO chains can serve as "spacers" inserted into the film surface and SO3- group. Zwitterionic Modification of Polyurethane Membranes for Enhancing the Anti-Fouling Property.

Abstract:Polyurethane (PU) is a biopolymer that has been commonly used for biomedical applications. However, the biofouling phenomenon on the hydrophobic PU surface is one of the crucial issues that embarrassing its applications. Here, we report a facile & efficient approach to improve the anti-biofouling ability of the PU substrates. Active residues were firstly generated on the PU surface by using the low temperature air-plasma treatment, promoting the immobilization of the atom transfer radical polymerization (ATRP) initiators on the surface. Then, three types of zwitterionic polymer brushes, as well as PEG brushes, have been fabricated on the PU substrates through surface-initiated ATRP (SI-ATRP). Robust surface characterizations that capable of revealing the surface chemistry (including X-ray photoelectron spectroscopy (XPS) and wettability tests), and antifouling evaluations of the PU substrates (protein adsorption, platelet adhesion, and cell adhesion measurements) were performed. Results showed that three types of zwitterionic brushes have been successful grafted on the PU surface, respectively. And the three types of zwitterionic brushes, in general, significantly inhibited the protein adsorption, the platelet adhesion, and the cell adhesion on the PU surface, endowing a significantly improved anti-fouling ability to the PU substrates. Furthermore, we found that this facial zwitterionic surface modification did not compromise the mechanical property of the PU substrates. This strategy could be easily exploited to PU-based biomaterials to improve their performance in many applications.

Abstract:A possible approach to improve the blood compatibility of poly(ether urethane)s (PU) involves the covalent attachment of key molecules on its surface. The purpose of the present study was to design and synthesise a novel non-thrombogenic biomaterial by modifying the surface of the PU with a zwitterionic monomer of sulfobetaine via a Jeffamine spacer. In this study, sulfobetaine was grafted onto a PU surface through the following reaction steps: (1) The PU surface was activated with hexamethylene diisocyanate (HDI) in toluene at 50degreesC in the presence of di-n-butyltin dilaurate (DBTDL) as a catalyst. The extent of the reaction was measured by ATR-FTIR spectra; a maximum number of free NCO groups was obtained after a reaction time of 90 min. (2) One cap of Jeffamine was reacted with isocyanate groups bound on the surface, so Jeffamine was introduced. (3) The PU surface terminated by Jeffamine was recoupled with isocyanate groups by HDI. (4) The hydroxyl groups of 4-dimethylamino-1-butanol (DMAB) or 2-dimethylaminoethanol (DMAE) were allowed to react with the isocyanate groups capped on Jeffamine. (5) Sulfobetaines were constructed on the surface through the ring-opening reaction between tertiary amine and 1,3-propanesultone (PS). It was confirmed by ATR-FTIR and XPS that the grafted surfaces were composed of sulfobetaine. The results of the contact-angle measurements and water absorption showed that they were strongly hydrophilic. The results of this platelet adhesion experiment as a preliminary test showed that PU grafted with sulfobetaine has good blood compatibility featured by the low platelet adhesion. Cationic Alternating Polypeptide Fixed on Polyurethane at Multiple Sites for Excellent Antibacterial and Antifouling Properties

Abstract:Bacterial infection and blockage are severe problems for polyurethane (PU) catheters and there is an urgent demand for surface-functionalized polyurethane. Herein, a cationic alternating copolymer comprising allyl-substituted ornithine and glycine (allyl-substituted poly(Orn-alter-Gly)) with abundant carbon–carbon double bond functional groups (C═C) is designed. Polyurethane is prepared with a large quantity of C═C groups (PU-D), and different amounts of allyl-substituted poly(Orn-alter-Gly) are grafted onto the PU-D surface (PU-D-2%AMPs and PU-D-20%AMPs) via the C═C functional groups. The chemical structures of the allyl-substituted poly(Orn-alter-Gly) and polyurethane samples (PU, PU-D, PU-D-2%AMPs, and PU-D-20%AMPs) are characterized and the results reveal that allyl-substituted poly(Orn-alter-Gly) is decorated on the polyurethane. PU-D-20%AMPs shows excellent antibacterial activity against Escherichia coli, Enterococcus faecalis, and Staphylococcus aureus because of the high surface potential caused by cationic allyl-substituted poly(Orn-alter-Gly), and it also exhibits excellent long-term antibacterial activity and antibiofilm properties. PU-D-20%AMPs also has excellent antifouling properties because the cationic copolymer is fixed at multiple reactive sites, thus avoiding the formation of movable long chain brush. A strong surface hydration barrier is also formed to prevent adsorption of proteins and ions, and in vivo experiments reveal excellent biocompatibility. This flexible strategy to prepare dual-functional polyurethane surfaces with antibacterial and antifouling properties has large potential in biomedical implants.This article has not yet been cited by other publications.

Abstract:Polyurethane (PU) catheters are commonly used in clinical treatment. However, the hydrophobic nature of the PU catheter surface leads to adhesion or adsorption to platelets, proteins, bacteria, and other molecules when used in human treatment. To achieve a surface with strong hydrophilicity, high stability and excellent biocompatibility, it is necessary to functionalize the PU catheters. In this paper, a coating with antifouling function was constructed on the surface of PU catheters using plasma technology and an amide coupling reaction. A series of characterization methods, including X-ray photoelectron spectroscopy (XPS), water contact angles (WCA), and atomic force microscopy (AFM), were used to prove the successful modification of the polymer coatings. The coatings showed good stability under conditions such as PBS (pH 7.4, 720 h), 75% ethanol (6 h) and 1 wt% SDS (10 min). Additionally, the coatings exhibit excellent hemocompatibility and antibacterial properties. The PU/PEI/PCSB coating has the best anti-fouling performance among them, which means that using the PCSB copolymer has the potential to modify different clinical catheters into highly effective antifouling coatings.

Abstract:Developing an efficient yet uncomplicated method to achieve anticoagulant coatings on biomedical PU surfaces remains a challenge. In this research, we have proposed a very simple way to synthesize fluorinated urethane prepolymers (FPUs) that possess extended fluorocarbon side chains and terminal groups, innovatively using a new fluorinated chain extender and a fluorinated terminal capping agent. These FPUs have been utilized as additives to fabricate biomedical PU blends (BPUB). Incorporating a minimal quantity of FPUs, as little as 2.5 %, bestows BPUB with exceptional capabilities to resist protein and platelet adhesion, which can further prevent platelet activation, facilitated by the self-migrating nature of the fluorocarbon chains toward the film's upper surfaces, as substantiated by contact angle measurements and XPS. DSC and TG analysis indicate that the addition of FPUs marginally affects the thermal stability of the BPUB films, yet it reduces the glass transition temperature of the PUs. Remarkably, the BPUB films demonstrate enhanced degradation resistance to physiological solutions, exhibit excellent hemocompatibility, and maintain extremely low cytotoxicity levels. Thus, the FPUs present themselves as highly encouraging candidates for the development of anticoagulant coatings on biomedical polymers.

Abstract:To improve the antifouling properties of polymer membranes, a novel phosphate-based zwitterionic polymer (methacryloyloxyethylphosphorylcholine-co-poly(propylene glycol) methacrylate, i.e., MPC-PPGMA) is introduced into the polyvinyl chloride (PVC) membrane matrix. The solubility of this zwitterionic copolymer in common organic solvents and the compatibility between this copolymer and PVC are studied. A series of PVC blend membranes are prepared via non-solvent induced phase separation based on the good compatibility of the copolymer with PVC. Surface chemical compositions, wettability and porous structures of the membranes are characterized. Fluorescence-labeled proteins static adsorption and organic foulant filtration are conducted to evaluate the fouling resistance of the blend membranes. Antibiofouling properties of the membranes are also confirmed by the immersing method. Doping MPC-PPGMA in the casting solution can significantly improve the surface hydrophilicity and antifouling properties of the PVC blend membranes due to the strong hydration ability of the zwitterionic segments. The blend membranes show excellent and stable resistance to both organic fouling and biofouling. Overall, the novel zwitterionic copolymer MPC-PPGMA is a promising candidate to develop antifouling blend membranes due to its high effectiveness and long-term stability in the membranes.

Abstract:Interaction of surface with water plays a key role in the blood compatibility of biomedical material.In this article,the research progress in hydrophilic functionalization for the surface of medical polyurethane as the typical model of biomedical polymer was reviewed,and the functions of water to block nonspecific adsorption of blood proteins and the formation of thrombosis were also discussed.The behavior of water molecules and their effect on the blood compatibility of the modified polyurethane,functionalized by the active materials such as hydrophilic vinyl monomer,polyethylene glycol and its derivates,heparin and zwitterionic materials,was explored.The knowledge of the functionalities of water and hydration layer during the interactions between blood and biomedical materials would benefit surface design and application of biocompatible polymer.

Abstract:Oligo-2'-O-methylribonucleotides conjugated with 4-(2-aminooxyethoxy)-2-(ethylureido)quinoline (AOQ) and 4-ethoxy-2-(ethylureido)quinoline (EOQ) were prepared by reaction of the AOQ or EOQ phosphoramidite with the protected oligonucleotide on a controlled pore glass support. Deprotection with ethylenediamine enabled successful isolation and purification of the highly reactive AOQ-conjugated oligomer. Polyacrylamide gel electrophoresis mobility shift experiments showed that the dissociation constants of complexes formed between an AOQ- or EOQ-conjugated 8-mer and complementary RNA or 2'-O-methyl-RNA targets (9- and 10-mers) were in the low nM concentration range at 37 degrees C, whereas no binding was observed for the corresponding nonconjugated oligomer, even at a concentration of 500 nM. Fluorescence studies suggested that this enhanced affinity is most likely due to the ability of the quinoline ring of the AOQ or EOQ group to stack on the last base pair formed between the oligomer and target, thus stabilizing the duplex. The binding affinity of a 2'-O-methyl RNA 15-mer, which contained an alternating methylphosphonate/phosphodiester backbone, for a 59-nucleotide stem-loop HIV TAR RNA target, increased 2.3 times as a consequence of conjugation with EOQ. The aminooxy group of AOQ-conjugated oligomers is a highly reactive nucleophile, which reacts readily with aldehydes and ketones to form stable oxime derivatives. This feature was used to couple an AOQ-oligomer with leupeptin, a tripeptide that contains a C-terminus aldehyde group. A simple method was developed to introduce a ketone functionality into peptides that contain a cysteine residue by reacting the peptide with bromoacetone. The resulting keto-peptide was then coupled to the AOQ-oligomer. This procedure was used to prepare oligonucleotide conjugates of a tetrapeptide, RGDC, and a derivative of HIV tat peptide having a C-terminus cysteine. The combination of the unique reactivity of the aminooxy group and enhanced binding affinity conferred by its quinoline ring suggests that AOQ may serve as a useful platform for the preparation of novel oligonucleotide conjugates.
Optimization of sulfonated polyethyleneimine zwitterionic coating mediated by polydopamine for poly(vinyl chloride) antifouling

Abstract:Although poly(vinyl chloride) (PVC) has been widely used as blood‐contacting materials in extracorporeal circuits and indwelling medical devices, its hydrophobicity also is a limitation due to the adsorption of plasma proteins and the immediately followed thrombosis cascade and bacterial infections. Herein, we developed a mild and simple strategy for improving the hydrophilicity and antifouling properties of the surface via mussel‐inspired chemistry. Different amounts of 1,3‐Propanesultone (1,3‐PS) were attached onto the polyethyleneimine (PEI), so as to modify it into the zwitterion, called PESX. Two layer‐by‐layer approaches about PESX grafting on PVC substrates were designed. One strategy was by coating PESX after coating polydopamine (PDA) on PVC surface, obtaining PDA‐PESX coatings. The other was by grafting PESX after co‐deposition treatment of PDA and PESX with appropriate feeding ratio on PVC, obtaining PDA/PESX‐PESX coatings. The study results indicated that the PDA/PES0.5‐PES0.5 coatings with optimized PEI molecular weight and sulfonation degree, presented about 2.5–3 times reduction in BSA adsorption (0.77 μg/cm2) compared with PDA/PES0.5 (1.93 μg/cm2) and PDA‐PES0.5 (2.26 μg/cm2), and kept long time antifouling stability. Moreover, the more excellent antifouling performance of PDA/PES0.5‐PES0.5 was also seen on fibrinogen (Fg), γ‐globulin (γ‐GL), and the mixture of these proteins. Meanwhile, PDA/PES0.5‐PES0.5 could prevent the adhesion of blood components and bacteria on the surface as well as maintain excellent hemocompatibility and cytocompatibility. We had hopes that our simple and mild strategy will find practical application in surface engineering of clinical use in blood‐contacting medical devices.

Blood Compatibility of Two Novel Polyurethane Coating Materials

Abstract:Amphiphilic coupling-polymer of stearyl poly (ethylene oxide)-co-4, 4'-methylendiphenyl diisocyanate-co-stearyl poly(ethylene oxide), MSPEO, was specially designed as surface-modifying additives. The blends of MSPEO in both polyether urethane (PEU) and chitosan(Chi), as the coating materials for intravascular device were investigated. Two kinds of static clotting time tests, plasma recalcification time (PRT) and prothrombin time(PT), as well as the static platelet adhesion experiment were carried out. And the dynamic anti-coagulation experiment was performed with a closed-loop tubular system under a blood shear rate of 1,500 s-1. The results demonstrate that both blend coatings can improve the anti-coagulation of polyurethane greatly and will not lead to hemolysis, and that more platelets adhere to the surface modified by Chi-MSPEO blend coating as compared with those adhere to the surface modified by PEU-MSPEO blend coating. The surface modified by Chi-MSPEO has longer PRT, whereas the surface modified by PEU-MSPEO has longer PT.
Antibiofilm and antithrombotic hydrogel coating based on superhydrophilic zwitterionic carboxymethyl chitosan for blood-contacting devices

Abstract:Blood-contacting devices must be designed to minimize the risk of bloodstream-associated infections, thrombosis, and intimal lesions caused by surface friction. However, achieving effective prevention of both bloodstream-associated infections and thrombosis poses a challenge due to the conflicting nature of antibacterial and antithrombotic activities, specifically regarding electrostatic interactions. This study introduced a novel biocompatible hydrogel of sodium alginate and zwitterionic carboxymethyl chitosan (ZW@CMC) with antibacterial and antithrombotic activities for use in catheters. The ZW@CMC hydrogel demonstrates a superhydrophilic surface and good hygroscopic properties, which facilitate the formation of a stable hydration layer with low friction. The zwitterionic-functionalized CMC incorporates an additional negative sulfone group and increased negative charge density in the carboxyl group. This augmentation enhances electrostatic repulsion and facilitates the formation of hydration layer. This leads to exceptional prevention of blood clotting factor adhesion and inhibition of biofilm formation. Subsequently, the ZW@CMC hydrogel exhibited biocompatibility with tests of in vitro cytotoxicity, hemolysis, and catheter friction. Furthermore, in vivo tests of antithrombotic and systemic inflammation models with catheterization indicated that ZW@CMC has significant advantages for practical applications in cardiovascular-related and sepsis treatment. This study opens a new avenue for the development of chitosan-based multifunctional hydrogel for applications in blood-contacting devices.
Novel Amphiphilic Poly(Dimethylsiloxane) Based Polyurethane Networks Tethered with Carboxybetaine and Their Combined Antibacterial and Anti-Adhesive Property

Abstract:The traditional nonfouling materials are powerless against bacterial cells attachment, while the hydrophobic bactericidal surfaces always suffer from nonspecific protein adsorption and dead bacterial cells accumulation. Here, amphiphilic polyurethane (PU) networks modified with poly(dimethylsiloxane) (PDMS) and cationic carboxybetaine diol through simple crosslinking reaction were developed, which had an antibacterial efficiency of 97.7%. Thereafter, the hydrolysis of carboxybetaine ester into zwitterionic groups brought about anti-adhesive properties against bacteria and proteins. The surface chemical composition and wettability performance of the PU network surfaces were investigated by attenuated total reflectance Fourier transform infrared spectroscopy (ATR-FTIR), X-ray photoelectron spectroscopy (XPS) and contact angle analysis. The surface distribution of PDMS and zwitterionic segments produced an obvious amphiphilic heterogeneous surface, which was demonstrated by atomic force microscopy (AFM). Enzyme-linked immunosorbent assays (ELISA) were used to test the nonspecific protein adsorption behaviors. With the advantages of the transition from excellent bactericidal performance to anti-adhesion and the combination of fouling resistance and fouling release property, the designed PDMS-based amphiphilic PU network shows great application potential in biomedical devices and marine facilities. Antibacterial and antifouling properties of a polyurethane surface modified with perfluoroalkyl and silver nanoparticles.

Abstract:Inspired by mussel-adhesion phenomena in nature, a simple, mild surface modification process was elaborated to endow the polyurethane (PU) substrate with antibacterial/antifouling properties. In the present study, polydopamine was coated directly onto polyurethane surfaces. AgNO3 was then added and absorbed onto the surface by the active catechol and amine groups of the polydopamine coating. Meanwhile, the adsorbed Ag+ ions were reduced in situ into metallic silver nanoparticles by the bridge of the polydopamine coating which yielded a coating with good antimicrobial properties. Finally, 1H, 1H, 2H, 2H-perfluorodecanethiol (CF3(CF2)(7)CH2CH2SH, F-SH) was attached on the PDA coating via the Michael addition reaction. The hydrophobic F-SH layer above the antibacterial layer yielded a surface with excellent antifouling properties. Preliminary antibacterial assays indicate that the coated surfaces show enhanced antibacterial activity against Escherichia coli (Gram-negative bacteria) and Staphylococcus aureus (Gram-positive bacteria). Platelet adhesion was significantly reduced for the F-SH-coated PU film. These results suggest that the modified PU could be used as an antibacterial material for future biomedical applications.
