Abstract:Introduction: The article aims to outline the features of the efforts for smallpox eradication within the pre-unitary context of the Kingdom of Sardinia, characterized by a long tradition in medical-health prevention. This tradition is partly inherited from the health magistracies of the Italian states during the ancient regime and partly adopted from policies initially outlined by Napoleon and later by other European states. In addition to prevention activities, authorities also engage in a vigorous information and awareness campaign aimed at eliminating common prejudices and doubts about vaccination among the population. Methods: In analyzing the authorities' achievements in combating smallpox, this study examines the two epidemic waves (1829-30 and 1852-54), along with the legislative developments before, during, and after these periods. It also compares these regulatory changes with those in other European contexts. Discussion: The epidemiological situation turned out to be more complex to manage than the authorities had anticipated, as evidenced by the increasing controls imposed. Scientific and political communities, both in the Kingdom of Sardinia and in other European nations, found themselves divided on the legitimacy of proposing restrictive measures by the state. Some advocated for restricted access to public places and imposed mandatory vaccination for vulnerable individuals. Conclusions: The comparison with smallpox resulted in a gradual improvement in of health security levels, although vaccination coverage did not reach the desired targets. Several factors contributed to this failure, including the limited expertise and reluctance of medical personnel, who were burdened with much of the operation's costs. Additionally, particularly in rural areas, there was widespread mistrust among the population towards doctors. Despite these challenges, the fight against smallpox enabled authorities to develop population control tools in the name of public health protection. However, it was not until 1888 that mandatory vaccination was introduced.

Abstract:The aim of this study was to determine whether Type II phospholipase A2 (PLA2) is released from late pregnant human placental tissue. Placental explants were incubated in vitro and the release of immunoreactive (ir) Type II PLA2 and PLA2 enzymatic activity into the medium was determined. Both irType II PLA2 and PL2 enzymatic activity accumulated in the incubation medium in a time-dependent manner (P < 0.0001). This release was not associated with a loss of cell membrane integrity, as indicated by measurement of the intracellular enzyme, lactate dehydrogenase, in the incubation medium. The concentration of irType II PLA2 and PLA2 enzyme activity present in incubation medium were significantly correlated (P < 0.01). Consistent with the hypothesis that Type II PLA2 may be store in secretory granules within human placental tissue, incubation in the presence of a membrane depolarising concentration of KCI (60 mM) caused the release of irType II PLA2 2.0-fold (P < 0.001). PLA2 enzyme activity released into the incubation medium displays biochemical characteristics consistent with those previously reported for secretory PLA2 isozymes, that is, a requirement for millimolar concentrations of calcium for optimal enzyme activity, inhibited by reducing agents, such as dithiothreitol and insensitive to heat inactivation. The data obtained in this study establish that irType PLA2 is released from term placenta, when incubated in vitro. The release of this extracellularly-active PLA2 isozyme may contribute to gestational and labour-associated increases in glycerophospholipid metabolism and prostaglandin formation.

Abstract:Over the past two centuries, vaccines have been critical for the prevention of infectious diseases and are considered milestones in the medical and public health history. The World Health Organization estimates that vaccination currently prevents approximately 3.5–5 million deaths annually, attributed to diseases such as diphtheria, tetanus, pertussis, influenza, and measles. Vaccination has been instrumental in eradicating important pathogens, including the smallpox virus and wild poliovirus types 2 and 3. This narrative review offers a detailed journey through the history and advancements in vaccinology, tailored for healthcare workers. It traces pivotal milestones, beginning with the variolation practices in the early 17th century, the development of the first smallpox vaccine, and the continuous evolution and innovation in vaccine development up to the present day. We also briefly review immunological principles underlying vaccination, as well as the main vaccine types, with a special mention of the recently introduced mRNA vaccine technology. Additionally, we discuss the broad benefits of vaccines, including their role in reducing morbidity and mortality, and in fostering socioeconomic development in communities. Finally, we address the issue of vaccine hesitancy and discuss effective strategies to promote vaccine acceptance. Research, collaboration, and the widespread acceptance and use of vaccines are imperative for the continued success of vaccination programs in controlling and ultimately eradicating infectious diseases.

Abstract:Abstract Vaccination has transformed public health, most notably including the eradication of smallpox. Despite its profound historical importance, little is known of the origins and diversity of the viruses used in smallpox vaccination. Prior to the twentieth century, the method, source and origin of smallpox vaccinations remained unstandardised and opaque. We reconstruct and analyse viral vaccine genomes associated with smallpox vaccination from historical artefacts. Significantly, we recover viral molecules through non-destructive sampling of historical materials lacking signs of biological residues. We use the authenticated ancient genomes to reveal the evolutionary relationships of smallpox vaccination viruses within the poxviruses as a whole.

Abstract:Objective This project seeks to create a differential diagnosis for lesions found on the skeletal remains of two children as a means to explore the presence of viral disease in 16th- century Peru. Materials Extremely well-preserved human remains of two children who died between the ages of 1–2 years old, recovered from the circum-contact (∼1540 CE) cemetery in Huanchaco, Peru. Methods Macroscopic and radiographic analysis. Results Both individuals present with cortical thickening, symmetrical destructive lesions, metaphyseal expansion, perforations, exposure of the medullary cavity, resorption of metaphyseal ends and necrosis of the long bones, and deposited reactive new bone. These features are consistent with osteomyelitis variolosa and bacterial osteomyelitis. Conclusions Three features of Individuals IG-124 and IG-493 suggest a highly consistent diagnosis of osteomyelitis variolosa: multiple skeletal lesions, the historical context of the area, and the high mortality rate of non-adults in the circum-contact cemetery. Significance Although viral infections are ubiquitous and well documented historically, their etiologies are often difficult to determine in archaeological populations. Orthopoxvirus variola (smallpox) is one of the many viruses whose archaeological impact is still under explored in skeletal remains. Limitations The absence of smallpox in other children from the Huanchaco cemetery creates difficulty in ascertaining true prevalence rates or information on potential outbreaks. Suggestions for further research Further research analyzing aDNA from calculus and/or residues using a DIP -GC-MS method might create a better understanding of how smallpox spread through the region.

Abstract:Latin America shows high plant biodiversity, this diversity is also reflected in all Colombian bioregions, supported by the varied topography and climates. Despite these rich natural resources, scientific understanding of Colombia's flora remains incomplete. The present study addresses the need to discuss the scientific publications around Colombian natural products derived from plants, specifically to the Coffee region. This is part of a larger initiative to explore the information available and highlight the research conducted so far. Consequently, Colombian authorities have recently launched programs to better study and utilize this biodiversity. Research related to natural products in Colombia is distributed across several information sources, making it challenging to recognize and compile. Additionally, studies focused on the structural elucidation of compounds are recent and have been reported only in the last twenty years. Likewise, it is not very abundant according to the results found. This study aims to review the current state of research on natural products derived from plants in Colombia's coffee region. To achieve this, we conducted a bibliometric analysis focusing on literature in English publications from the last few years retrieved from Web of Science (WoS) and Scopus. The analysis included identifying the most productive authors and key research topics with Bibliometrix. We also examined the impact and visibility of the publications by analyzing the H-index and quartile rankings of the journals where the research was published. Additionally, keyword analysis was performed using VOSviewer to visualize the main themes and trends in the field. This comprehensive review provides insights into the development and direction of research on natural products derived from plants in Colombia's Coffee region, highlighting the most influential studies and emerging areas of interest.

Abstract:The middle of the XIX century in Colombia (then New Granada) was marked by radical transformations, which went down in history as the revolutionary events of the 50s. The modernization of Colombian soci-ety affected all aspects of public life: political economic, social and administrative. The younger generation of radical liberals who came to power in search of ways of national identity used two mutually contra-dictory practices in their activities: the development of modern legal norms of national creation (constitutional acts) and the practice of civil wars, a tradition rooted in the era of the War of Independence of 1810–1826. The constitutional acts of this period (1853–1863) and the crea-tion of the foundations of the modern state were based on the recogni-tion of the federal structure of the republican society and the complete eradication of all vestiges of the old colonial regime. The proposed work analyzes the constitutional acts and reforms of this period in the history of Colombia (1853–1863), their role and importance for the subsequent development of the country.

Abstract:Abstract The idea that smallpox could be eradicated was not necessarily the ultimate aim when inoculation was introduced in Europe in the 1720s. This potentiality was not clearly articulated as an aim until the end of the eighteenth century. This article argues that during most of the eighteenth century, the main aim of inoculation was to lead people as safely as possible through what was regarded as an unavoidable disease. Inoculation became safer, simpler and less expensive from the 1760s, but the changing ideas about its potentiality had more complex roots. A new understanding was produced through an interaction between inoculation practice, more general medical theory and developments within probabilistic thinking and political arithmetic. The first part of the article explores how smallpox inoculation was incorporated into existing medical thinking based on traditional humoral pathology. Inoculation was a new technology, but as it was perceived in the early eighteenth century, the innovation did not first and foremost concern the medical principles of the treatment. The second part of the article investigates arguments about why and when to inoculate: what kind of remedy was inoculation for eighteenth-century agents? The article concludes with a discussion on changes emerging towards the end of the century, and relates them to developments during the preceding decades rather than seeing them as inspired precursors of events and ideas to come.

Abstract:A mediados del siglo XVIII llegó la primera imprenta a Nueva Granada, debido a la congregación jesuita. Pero fue a finales de esa centuria cuando, con ocasión del sismo que asoló a la capital, apareció el Aviso del Terremoto, primer periódico del país que alcanzó una gran popularidad. En ese sentido, en el siguiente artículo de reflexión, se analizan los efectos de la imprenta en la opinión pública neogranadina en el periodo de la llamada Reconquista española. Mediante el estudio de fuentes primarias y secundarias, concluimos que la labor de la imprenta resultó fundamental para desdibujar el dominio simbólico de la monarquía, al integrar parcialmente a las clases populares dentro de la deliberación general, la participación y la transparencia relativa sobre las tareas de gobierno.

Abstract:En este artículo, estudiamos la elaboración de dos métodos para combatir la gripe española o influenza en la Argentina: el suero y la vacuna. Las investigaciones sobre el agente etiológico y la elaboración de estos instrumentos se llevaron a cabo en el Instituto Bacteriológico, institución estatal que al momento en que ingresó y se expandió la enfermedad estaba dirigida por el médico bohemio Rudolf Kraus y junto a él trabajaba una parte de la élite médica porteña y extranjera. Realizamos un análisis de las metodologías que se utilizaron tanto para la elaboración como en las pruebas que se desarrollaron a lo largo de los años 1918 y 1919. Consideramos que el mismo Estado que permitió y alentó la elaboración de ambos métodos de prevención y de cura, en especial de la vacuna, también fue un factor que, por su misma dinámica, impidió que se llevaran adelante las pruebas para validarla y aplicarla. El estudio se llevó a cabo a partir de diversas fuentes, como documentación estatal, artículos científicos y notas periodísticas, desde un análisis hermenéutico.

Abstract:In the last 20 years, the Dental Anthropology and Forensic Dentistry Research Group at the Universidad del Valle (Cali, Colombia) has integrated knowledge from anthropology, dentistry, biology, paleontology and paleopathology to characterize the dental morphology of living populations in southwestern Colombia. This has been done by studying the frequency and variability of dental morphological features in populations with different ancestries, including Euro-descendants, Afro-descendants and Native Americans. The group has employed strategies such as formative research and the creation of cooperative research networks to publish and disseminate their findings on dental morphology mainly within the Colombian dental clinical context. However, these studies have been limited in their impact on the international anthropological academic community due to a lack of publication in English and refusals from some specialized journals to publish research on contemporary Colombian populations. To address this issue, this article aims to provide a literature review of the research on dental anthropology carried out at the School of Dentistry of the Universidad del Valle (Cali, Colombia) between 2002 and 2021. Despite the high amount of available information, the results of this scientific research have been difficult to make visible, search, access, and recover.

Abstract:Although it is well established that Hispanics generally have a mixed Native American, African, and European ancestry, the dynamics of admixture at the foundation of Hispanic populations is heterogeneous and poorly documented. Genetic analyses are potentially very informative for probing the early demographic history of these populations. Here we evaluate the genetic structure and admixture dynamics of a province in northwest Colombia (Antioquia), which prior analyses indicate was founded mostly by Spanish men and native women. We examined surname, Y chromosome, and mtDNA diversity in a geographically structured sample of the region and obtained admixture estimates with highly informative autosomal and X chromosome markers. We found evidence of reduced surname diversity and support for the introduction of several common surnames by single founders, consistent with the isolation of Antioquia after the colonial period. Y chromosome and mtDNA data indicate little population substructure among founder Antioquian municipalities. Interestingly, despite a nearly complete Native American mtDNA background, Antioquia has a markedly predominant European ancestry at the autosomal and X chromosome level, which suggests that, after foundation, continuing admixture with Spanish men (but not with native women) increased the European nuclear ancestry of Antioquia. This scenario is consistent with historical information and with results from population genetics theory.
Consumers' and Professionals' Responses to Situations Raising Ethical Questions

Abstract:In this study we have attempted, in collaboration with an agrifood company, to compare and contrast the responses of consumers and professionals to the same set of situations raising ethical questions, in order to highlight the differences and the similarities of viewpoint between these two groups of stakeholders. In order to do this, we constructed multi-stakeholder scenarios comprising a description of the situation and the various possible approaches to managing the situation. We constructed two scenarios, dealing with the origin of the gelatin used in the companyâ€™s products and the companyâ€™s product information on livestock feed (GMO, genetically modified organisms). This allowed us to: - study the extent of the professionalsâ€™ ethical perceptions and the possible compromises between ethical values and business interests in the decision-making process. - compare and contrast the ethical perceptions of consumers and professionals. In the first part we discuss the exploratory phase which enabled us to construct the scenarios. In the second part we compare the ethical value of the decisions suggested for each scenario according to consumers and professionals and the probability of adopting the various decisions according to professionals. The third part compares consumersâ€™ and professionalsâ€™ responses on the matching of each decision to consumer expectations and on the decision which would appear to them to be ideal. If there is a convergence between consumers and professionals in relation to the ethical classification of the various decisions, we can note, however, that consumersâ€™ expectations with regard to ethics are little understood by professionals. This study leads to a better understanding of the significance of considerations of an ethical nature in consumer perceptions and allows them to be compared and contrasted with the ethical judgments of professionals. It gives professionals a better insight into consumer expectations on sensitive issues and highlights certain differences between consumer expectations and managersâ€™ suggestions.

WVU NLP Class Participation in ShARe/CLEF Challenge

Abstract:The spring 2013 graduate class in NLP decided to participate in the ShARe/CLEF challenge Tasks 1 & 2. The timing for the challenge coincided nicely with the spring semester session. There were six students in the graduate class, and the challenge tasks appeared to be good material to expose students to a practical task faced by healthcare industry. The general approach used by the class is to use CRF learning algorithm using Factorie – a scala-language based toolkit. The F-scores for best results for Task 1a relaxed – 0.801, 1a strict – 0.554, 1b relaxed – 0.625 & 1b strict 0.349, respectively. Task 2 was attempted as a group task. F-scores were 0.426 and 0.428 for strict and relaxed respectively. It was a real challenge to focus on the challenge as a class project. The students did learn how to apply NER CRF engine to a practical problem.

Abstract:The historical significance of the poxviruses is profound, largely due to the enduring impact left by smallpox virus across many centuries. The elimination of smallpox is a remarkable accomplishment in the history of science and medicine, with centuries of devoted efforts resulting in the development and widespread administration of smallpox vaccines. This review provides insight into the pivotal historical events involving medically significant poxviruses. Understanding the remarkable saga of combatting smallpox is crucial, serving as a guidepost for potential future encounters with poxvirus infections. There is a continual need for vigilant observation of poxvirus evolution and spillover from animals to humans, considering the expansive range of susceptible hosts. The recent occurrence of monkeypox cases in non-endemic countries stands as a stark reminder of the ease with which infections can be disseminated through international travel and trade. This backdrop encourages introspection about our journey and the current status of poxvirus research.
