Abstract:Terrestrial amphibians take up water by abducting the hind limbs and pressing a specialized portion of the ventral skin to a moist surface, using a characteristic behavior called the water absorption response. An assay of the water absorption response was used to quantify physiological factors associated with thirst and water uptake. Dramatic changes in the water absorption response resulted from subtle changes in hydration state and from altering the reserve water supply in the urinary bladder. The water absorption response could be induced by intraperitoneal and intracerebroventricular injection of angiotensin II, demonstrating that components of the renin-angiotensin system on both sides of the blood-brain barrier have a dipsogenic function in amphibians. These experiments also demonstrated that the water absorption response could be influenced by changes in barometric pressure. Toads avoided the water absorption response on hyperosmotic substrates, and behavioral experiments showed that the amphibian skin served a sensory function similar to that of the lingual epithelium of mammals. The water absorption response assay has enormous potential as a tool for the investigation of physiological processes and sensory capabilities of amphibians.
Acute hypoxia exposure rapidly triggers behavioral changes linked to cutaneous gas exchange in Lake Titicaca frogs

Abstract:Ventilation is critical to animal life-it ensures that individuals move air/water across their respiratory surface, and thus it sustains gas exchange with the environment. Many species have evolved highly specialized (if not unusual) ventilatory mechanisms, including the use of behavior to facilitate different aspects of breathing. However, these behavioral traits are often only described anecdotally, and the ecological conditions that elicit them are typically unclear. We study one such "ventilation behavior" in Lake Titicaca frogs (Telmatobius culeus). These frogs inhabit high-altitude (i.e., low oxygen) lakes in the Andean Mountains of South America, and they have become textbook examples of cutaneous gas exchange, which is essentially breathing that occurs across the skin. Accordingly, this species has evolved large, baggy skin-folds that dangle from the body to increase the surface area for ventilation. We show that individuals exposed to acute hypoxic conditions that mirror what free-living individuals likely encounter quickly (within minutes) decrease their activity levels, and thus become very still. If oxygen levels continue to decline, the frogs soon begin to perform push-up behaviors that presumably break the low-oxygen boundary layer around skin-folds to increase the conductance of the water/skin gas exchange pathway. Altogether, we suspect that individuals rapidly adjust aspects of their behavior in response to seemingly sudden changes to the oxygen environment as a mechanism to fine tune cutaneous respiration.
Comparative Morphology of the Lungs and Skin of two Anura, Pelophylax nigromaculatus and Bufo gargarizans

Abstract:Abstract The lungs and skin are important respiratory organs in Anura , but the pulmonary structure of amphibians remains unclear due to the lack of a suitable procedure. This study improved the procedure used for fixing lungs tissues and used light microscopy, transmission electron microscopy and scanning electron microscopy to reveal the differences in the lung and skin morphologies between Pelophylax nigromaculatus ( P. nigromaculatus ) and Bufo gargarizans ( B. gargarizans ). In P. nigromaculatus and B. gargarizans , the cystic lungs comprise a continuous outer pulmonary wall on which primary, secondary, and tertiary septa attach, and a number of regular lattices form from raised capillaries and the pulmonary epithelium on the surfaces of the pulmonary wall and septa. Each lattice in P. nigromaculatus consists of several elliptical sheets and flat bottom, and the septa are distributed with denser sheets and have a larger stretching range than the pulmonary wall. The lattice in B. gargarizans consists of thick folds and an uneven bottom with several thin folds, and the septa have more developed thick and thin folds than the pulmonary wall. However, the density of the pulmonary microvilli, the area of a single capillary, the thicknesses of the blood-air barrier, pulmonary wall and septum, and the lung/body weight percentage obtained for B. gargarizans were higher than those found for P. nigromaculatus . In P. nigromaculatus , the dorsal skin has dense capillaries and a ring surface structure with mucus layer on the stratum corneum, and the ventral skin is slightly keratinized. In B. gargarizans , the stratum corneum in both the dorsal and ventral skins is completely keratinized. A fine ultrastructure analysis of P. nigromaculatus and B. gargarizans revealed that the pulmonary septa are more developed than the pulmonary walls, which means that the septa have a stronger respiratory function. The more developed lungs are helpful for the adaptation of B. gargarizans to drought environments, whereas P. nigromaculatus has to rely on more vigorous skin respiration to adapt to a humid environment.

Abstract:Amphibious fishes have evolved multiple adaptive strategies for respiring out of water, but there has been less focus on reversible plasticity. We tested the hypothesis that when amphibious fishes leave water, enhanced respiratory performance on land is the result of rapid functional phenotypic flexibility of respiratory traits. We acclimated four isogenic strains of Kryptolebias marmoratus to air for 0, 1, 3 or 7 days. We compared respiratory performance out of water with traits linked to the O2 cascade. Aerial O2 consumption rate was measured over a step-wise decrease in O2 levels. There were significant differences between strains, but time out of water had the largest impact on measured parameters. K. marmoratus had improved respiratory performance (lower aerial critical oxygen tension (Pcrit), higher regulation index (RI)) after only 1 day of air exposure and these changes were strongly associated with the change in hematocrit and dorsal cutaneous angiogenesis. Additionally, we found that 1-hour of air exposure induced the expression of four angiogenesis-associated genes, vegfa, angpt2, pecam-1 and efna1 in the skin. After 7 days in air, respiratory traits were not significantly linked to the variation in either aerial Pcrit or RI. Overall, our data indicate that there are two phases involved in the enhancement of aerial respiration; an initial rapid response (1 day) and a delayed response (7 days). We found evidence for the hypothesis that respiratory performance on land in amphibious fishes is the result of rapid flexibility in both O2 uptake and O2 carrying capacity.

Plastic mechanisms for unraveling a universal trade-off between water loss and respiration

Abstract:The expression of phenotypes is often constrained by functional conflicts between traits, and the resulting trade-offs can impose limits on phenotypic and taxonomic diversity. However, organisms can circumvent trade-offs in specific environmental contexts through phenotypic plasticity, though the mechanisms that drive trade-offs or allow organisms to resolve trade-offs are often cryptic. The trade-off between water loss and gas exchange poses a fundamental challenge for terrestrial life because the capacity to absorb oxygen is limited by the risk of desiccation from water loss across respiratory surfaces. Here, we investigated the capacity to mitigate this trade-off in a lungless salamander that breathes exclusively across its skin. We measured water loss and oxygen uptake using respirometry in field- and laboratory-based studies to identify plastic responses to environmental variation, coupled with gene expression analyses to investigate biological pathways that regulate the trade-off. Though the trade-off between oxygen uptake and water loss was generally strong, we observed that its strength changed over time in the field and laboratory. At the molecular level, we found support for antagonistic pleiotropy in multiple pathways that simultaneously affect both physiological traits (e.g., via vasoconstriction and downregulation of aerobic respiration), and for mechanisms that mitigate the trade-off by affecting only one trait (e.g., oxygen binding affinity, melanin synthesis). As organisms disentangle such conflicts, however, alternative trade-offs are likely to arise. Our study provides evidence that alternative pathways allow organisms to mitigate pleiotropic conflicts, which ultimately may allow greater phenotypic diversity and persistence in novel environments.

Water availability and temperature as modifiers of evaporative water loss in tropical frogs

Abstract:Water plays a notable role in the ecology of most terrestrial organisms due to the risks associated with water loss. Specifically, water loss in terrestrial animals happens through evaporation across respiratory tissues or epidermis. Amphibians are ideal systems for studying how abiotic factors impact water loss since their bodies often respond quickly to environmental changes. While the effect of temperature on water loss is well known across many taxa, we are still learning how temperature in combination with humidity or water availability affects water loss. Here, we tested how standing water sources (availability) and temperature (26 and 36°C) together affect water loss in anuran amphibians using a Bayesian framework. We also present a conceptual model for considering how water availability and temperature may interact, resulting in body mass changes. After accounting for phylogenetic and time autocorrelation, we determined how different variables (water loss and uptake rates, temperature, and body size) affect body mass in three species of tropical frogs (Rhinella marina, Phyllobates terribilis, and Xenopus tropicalis). We found that all variables impacted body mass changes with greater similarities between P. terribilis and X. tropicalis, but only temperature showed a notable effect in P. terribilis. Furthermore, we describe how the behavior of P. terribilis might affect its water budget. This study shows how organisms might manage water budgets across different environments and is important for developing our models of evaporative water loss and species distributions.
Convergent evolution of skin surface microarchitecture and increased skin hydrophobicity in semi-aquatic anole lizards

Abstract:ABSTRACT Animals that habitually cross the boundary between water and land face specific challenges with respect to locomotion, respiration, insulation, fouling and waterproofing. Many semi-aquatic invertebrates and plants have developed complex surface microstructures with water-repellent properties to overcome these problems, but equivalent adaptations of the skin have not been reported for vertebrates that encounter similar environmental challenges. Here, we document the first evidence of evolutionary convergence of hydrophobic structured skin in a group of semi-aquatic tetrapods. We show that the skin surface of semi-aquatic species of Anolis lizards is characterized by a more elaborate microstructural architecture (i.e. longer spines and spinules) and a lower wettability relative to closely related terrestrial species. In addition, phylogenetic comparative models reveal repeated independent evolution of enhanced skin hydrophobicity associated with the transition to a semi-aquatic lifestyle, providing evidence of adaptation. Our findings invite a new and exciting line of inquiry into the ecological significance, evolutionary origin and developmental basis of hydrophobic skin surfaces in semi-aquatic lizards, which is essential for understanding why and how the observed skin adaptations evolved in some and not other semi-aquatic tetrapod lineages.

What frog gill resorption brings: loss of function, cell death, and metabolic reorganization

Abstract:Background: Anuran metamorphosis, which is driven by thyroid hormone (TH)-mediated processes, orchestrates intricate morphological and functional transformations for the transition from aquatic tadpoles to terrestrial life, providing a valuable model for studying organ functionalization, remodeling, and regression. Larva-specific organ regression is one of the most striking phenomena observed during the anuran metamorphic climax. While previous studies extensively analyzed the regression mechanisms of the tail, the molecular processes governing gill resorption remain elusive. Results: We employed Microhyla fissipes as a model, and utilized a comprehensive approach involving histological analysis, transmission electron microscopy, and transcriptomics to unravel gill development and resorption. The pro-metamorphic stages revealed highly developed gill structures, emphasizing their crucial role as the primary respiratory organ for tadpoles. The transcriptomic analysis highlighted the upregulation of genes associated with enhanced respiratory efficiency, such as hemoglobin and mucins. However, as metamorphosis progressed, gill filaments underwent shrinkage, decreases in blood vessel density, and structural changes that signified a decline in respiratory function. The molecular mechanisms driving gill resorption involved the TH pathway-in particular, the upregulation of thyroid hormone receptor (TR) β, genes associated with the tumor necrosis factor pathway and matrix metalloproteinases. Two distinct pathways orchestrate gill resorption, involving apoptosis directly induced by TH and cell death through the degradation of the extracellular matrix. In addition, metabolic reorganization during metamorphosis is a complex process, with tadpoles adapting their feeding behavior and mobilizing energy storage organs. The gills, which were previously overlooked, have been unveiled as potential energy storage organs that undergo metabolic reorganization. The transcriptomic analysis revealed dynamic changes in metabolism-related genes, indicating decreased protein synthesis and energy production and enhanced substrate transport and metabolism during metamorphic climax. Conclusion: This study sheds light on the structural, molecular, and metabolic dynamics during gill development and resorption in M. fissipes. The findings deepen our understanding of the intricate mechanisms governing organ regression and underscore the pivotal role of the gills in facilitating the transition from aquatic to terrestrial habitats.
Novel rebreathing adaptation extends dive time in a semi-aquatic lizard

Abstract:Bubble use evolved in many small invertebrates to enable underwater respiration, but, until recently, there has been no evidence that vertebrate animals use bubbles in a similar manner. Only one group of vertebrates, semi-aquatic Anolis lizards, may be an exception: these lizards dive underwater when threatened and, while underwater, rebreathe a bubble of air over their nostrils. Although it seems that rebreathing should be adaptive, possibly functioning to extend the time that lizards remain in underwater refugia, this has not been empirically tested. Here, I demonstrate that rebreathing serves to extend dive time in a semi-aquatic anole, Anolis aquaticus . I prevented the formation of normal rebreathing bubbles by applying a commercial emollient on the skin surface where bubbles form to assess the impact of bubbles on rebreathing cycles, gular pumps, and dive times. Lizards that were allowed to rebreathe normally remained underwater an average of 32% longer than those with impaired rebreathing, suggesting a functional role of rebreathing in underwater respiration. Unlike rebreathing, gular pumping was unaffected by treatment and may warrant further research regarding its role in supplementing underwater respiration. This study provides evidence that vertebrates can use bubbles to respire underwater and raises questions about adaptive mechanisms and potential bio-inspired applications.

Abstract:Dragonflies (Odonata, Anisoptera) are amphibiotic; the nymph is aquatic and breathes water using a rectal gill before transitioning to the winged adult that breathes air through spiracles. While the evolutionary and developmental transition from water- to air-breathing is known to be associated with a dramatic rise in internal CO2 levels, the changes in blood-gas composition experienced by amphibiotic insects, which represent an ancestral air-to-water transition, are unknown. This study measured total CO2 (TCO2) in hemolymph collected from aquatic nymphs and air-breathing adults of Anax junius, Aeshna multicolor (Aeshnidae), Libellula quadrimaculata, and L. forensis (Libellulidae). Hemolymph PCO2 was also measured in vivo in both Aeshnid nymphs and marbled crayfish (Procambarus fallax. f. virginalis) using a novel fiber-optic CO2 sensor. The hemolymph TCO2 of the pre- and early-final instar nymphs was found to be significantly lower than that of the air-breathing adults. However, the TCO2 of the late-final instar Aeshnid nymphs was not significantly different from the air-breathing adult, despite the late-final nymph still breathing water. TCO2 and PCO2 were also significantly higher in the hemolymph of early-final Aeshnid nymphs compared to the water-breathing crayfish. Thus, while dragonfly nymphs show an increase in internal CO2 as they transition from water to air, from an evolutionary standpoint, the nymph's ability to breathe water is associated with a comparatively minor decrease in hemolymph TCO2 relative to the air-breathing adult.

The Xenopus respiratory system reveals common tetrapod mechanisms for growth, regeneration and healing

Abstract:In recent years, we have seen a significant increase in our understanding of the mechanisms of development, regeneration, and healing of the respiratory system. However, most of these studies have been limited by their focus on mammalian systems. Here, we aimed to identify the underlying molecular mechanisms that are active during lung growth and tissue repair in amphibians, specifically Xenopus tropicalis (X. tropicalis). First, we analyzed the stem cell composition and signaling pathways that are active in epithelial and mesenchymal cells during lung growth. Then, we established a protocol for lung injury to assess the types of stem cells underlying tissue repair. In mammals, Sftpc+ (AT2) cells are alveolar stem cells that can differentiate to Krt8+ cells during lung homeostasis and post-injury repair. In this study, we identified Sftpc+ cells and Krt8+ cells, along with the activity of key developmental signaling pathways, Hippo and Wnt, during lung maturation at post-metamorphosis stages. We then established a protocol for lung injury using chemically induced injury with bleomycin, which damages the lung through oxidative stress. The results show an elevation in collagen post-injury, indicating bleomycin's effect in causing lung fibrosis. X. tropicalis froglets survived 42 days post-injury, with a continuous decrease in fibrosis. To explore this effect, we analyzed the distribution of lung stem cells; Sox9 protein levels and Sftp gene expression were downregulated at the alveoli 42 days post-injury. The decrease in stem cell marker expression 42 days post-injury suggests they are differentiating as part of the healing process. Nevertheless, we could still detect them after a few weeks of healing. These results suggest that X. tropicalis has a regenerative capacity for lung tissue repair and that the same signaling pathways and stem cells are active in both amphibians and mammalian lungs during lung growth, regeneration, and healing. These findings show for the first time the physiological similarities between the anuran and the mammalian lung during growth and tissue repair processes, suggesting X. tropicalis as a potential animal model to study lung regeneration.

Abstract:OBJECTIVE To investigate the general anesthetic's effect upon cholinergic system to examine whether the regional and progressive cholinergic changes may lead to postoperative cognitive dysfunction (POCD). METHOD A model of hippocampus microdialysis was established in aging rats (18 months old). The tissue levels of choline (Ch) and acetylcholine (Ach) were determined in hippocampus. The post-anesthesia learning capability and spatial memory were tested in Morris maze. Using in vivo microdialysis, the releases of Ach and Ch, functional parameters of cholinergic system, were determined in freely moving rats. The contents of both in perfusate were quantified by HPLC-ECD as described for the function of cholinergic system. RESULTS The learning curve of the control group demonstrated differences from the experiment group. Changes in hippocampal Ach and Ch levels were observed in both cognition markedly impaired group and cognition lightly impaired group, accompanied by performance failure in water maze test. In the experiment group, the hippocampal releases of Ach and Ch were markedly different between cognition markedly impaired group and cognition lightly impaired group. CONCLUSION Cholinergic system dysfunction in hippocampus may be responsible for behavioral abnormality in learning and spatial memory tasks in senescent rats.
Responses of intraspecific metabolic scaling to temperature and activity differ between water‐ and air‐breathing ectothermic vertebrates

Abstract:Metabolic scaling (i.e. size‐dependence of metabolism) varies widely with temperature and locomotor activity. We here show that intraspecific changes in metabolic scaling with warming and increasing activity differ between water‐ and air‐breathing ectothermic vertebrates. By combining evolved responses to oxygen limitation and influences of volume‐related processes, our study quantitatively incorporates temperature, activity and respiration modes to explain animal energetics. Metabolism underpins all life‐sustaining processes and varies profoundly with body size, temperature and locomotor activity. A current theory explains some of the size‐dependence of metabolic rate (its mass exponent, b) through changes in metabolic level (L). We propose two predictive advances that: (a) combine the above theory with the evolved avoidance of oxygen limitation in water‐breathers experiencing warming, and (b) quantify the overall magnitude of combined temperatures and degrees of locomotion on metabolic scaling across air‐ and water‐breathers. We use intraspecific metabolic scaling responses to temperature (523 regressions) and activity (281 regressions) in diverse ectothermic vertebrates (fish, reptiles and amphibians) to show that b decreases with temperature‐increased L in water‐breathers, supporting surface area‐related avoidance of oxygen limitation, whereas b increases with activity‐increased L in air‐breathers, following volume‐related influences. This new theoretical integration quantitatively incorporates different influences (warming, locomotion) and respiration modes (aquatic, terrestrial) on animal energetics.

Allometric scaling of heat and water exchanges in the mammals' lung

Abstract:Mammals have a high metabolism that produces heat proportionally to the power 3/4 of their mass at rest. Any excess of heat has to be dissipated in the surrounding environment to prevent overheating. Most of that dissipation occurs through the skin, but the efficiency of that mechanism decreases with the animal's mass. The role of the other mechanisms for dissipating heat is then raised, more particularly the one linked to the lung that forms a much larger surface area than the skin. The dissipation occurring in the lung is however often neglected, even though there exists no real knowledge of its dynamics, hidden by the complexity of the organ's geometry and of the physics of the exchanges. Here we show, based on an original and analytical model of the exchanges in the lung, that all mammals, independently of their mass, dissipate through their lung the same proportion of the heat they produced, about 6-7 %. We found that the heat dissipation in mammals' lung is driven by a number, universal among mammals, that arises from the dynamics of the temperature of the bronchial mucosa. We propose a scenario to explain how evolution might have tuned the lung for heat exchanges. Furthermore, our analysis allows to define the pulmonary heat and water diffusive capacities. We show in the human case that these capacities follow closely the oxygen consumption. Our work lays the foundations for more detailed analysis of the heat exchanges occurring in the lung. Future studies should focus on refining our understanding of the universal number identified. In an ecological framework, our analysis paves the way to a better understanding of the mammals' strategies for thermoregulation and of the effect of warming environments on mammals' metabolism.
Hypoxic pulmonary vasoconstriction as a regulator of alveolar-capillary oxygen flux: A computational model of ventilation-perfusion matching

Abstract:The relationship between regional variabilities in airflow (ventilation) and blood flow (perfusion) is a critical determinant of gas exchange efficiency in the lungs. Hypoxic pulmonary vasoconstriction is understood to be the primary active regulator of ventilation-perfusion matching, where upstream arterioles constrict to direct blood flow away from areas that have low oxygen supply. However, it is not understood how the integrated action of hypoxic pulmonary vasoconstriction affects oxygen transport at the system level. In this study we develop, and make functional predictions with a multi-scale multi-physics model of ventilation-perfusion matching governed by the mechanism of hypoxic pulmonary vasoconstriction. Our model consists of (a) morphometrically realistic 2D pulmonary vascular networks to the level of large arterioles and venules; (b) a tileable lumped-parameter model of vascular fluid and wall mechanics that accounts for the influence of alveolar pressure; (c) oxygen transport accounting for oxygen bound to hemoglobin and dissolved in plasma; and (d) a novel empirical model of hypoxic pulmonary vasoconstriction. Our model simulations predict that under the artificial test condition of a uniform ventilation distribution (1) hypoxic pulmonary vasoconstriction matches perfusion to ventilation; (2) hypoxic pulmonary vasoconstriction homogenizes regional alveolar-capillary oxygen flux; and (3) hypoxic pulmonary vasoconstriction increases whole-lobe oxygen uptake by improving ventilation-perfusion matching.

Adaptation of skin structures to environmental variations in anurans from southern and southwestern China

Abstract:We studied the environmental adaptation of the skin structures of 102 Chinese anuran species across varying geographical distributions. We found that the volume of granular glands and capillary density in the dorsal skin significantly increased with increasing latitude. Our findings suggest that phenotypic variation in anuran skin is associated with latitude. Variations in skin structures can possibly reflect local adaptation to distinct environmental factors. As the primary interface with the surrounding environment, amphibian skin undergoes phenotypic innovations that play a key role in protection, water absorption, and respiration. However, the effects of environmental factors on skin structures have been examined in only a limited number of species. Here, we conducted a comparative analysis of the skin structures of 102 Chinese anuran species across varying geographical distributions and habitat types. Our results revealed that the total volume of granular glands and capillary density in the dorsal skin significantly increased with increasing latitude. We also found that the thickness of calcified layers in both dorsal and ventral skin was positively correlated with annual temperature and negatively correlated with humidity. Additionally, terrestrial species exhibited the largest dorsal granular gland, whereas arboreal species had the smallest one. Likewise, the largest dorsal mucous gland was observed in aquatic species, while the smallest was found in terrestrial species. These results highlighted the importance of understanding the relationship between skin phenotypes and environmental variables and thus providing conservation strategies based on the evolutionary adaptations in anurans. Our study can contribute to the broader knowledge of evolutionary biology in anurans by demonstrating how specific skin traits are linked to survival and fitness across various ecological contexts.

Spiracular fluttering decouples oxygen uptake and water loss: a stochastic PDE model of respiratory water loss in insects

Abstract:In insect respiration, oxygen from the air diffuses through a branching system of air-filled tubes to the cells of the body and carbon dioxide produced in cellular respiration diffuses out. The tracheal system has a very large surface area, so water loss is a potential threat and the question of how insects regulate oxygen uptake and water loss has been an important issue in insect physiology for the past century. The tracheal system starts at spiracles on the surface of the body that insects can open and close, and three phases are observed experimentally, open or closed for relatively long periods of time and opening and closing rapidly, which is called fluttering. In previous work we have shown that during this flutter phase, no matter how small the percentage of time that the spiracles are open, the insect can absorb almost as much oxygen as if the spiracle were always open, if the insect flutters fast enough. This left open the question of water loss during the flutter phase, which is the question addressed in this paper. We formulate a stochastic diffusion-convection model for the concentration of water vapor in the tracheae. Mathematical analysis of the model yields an explicit formula for water loss as a function of six non-dimensional parameters and we use experimental data from various insects to show that, for parameters in the physiological ranges, water loss during the flutter phase is approximately proportional to the percentage of time open. This means that the insect can solve the oxygen uptake versus water loss problem by choosing to have their spiracles open a small percentage of time during the flutter phase and fluttering rapidly.

Abstract:A key component of amphibian antipredator strategies is the chemical defenses that make them toxic and/or distasteful, like the cardiotoxic bufadienolides synthesized by the true toads and stored in granular skin glands. The morphology of adult toad glands is well-described, but the ontogenetic timing and distribution of glands during larval stages are poorly understood. A comparative understanding of granular gland development is needed to elucidate the mechanisms underlying the diversification of toad chemical defenses and their ecological roles across lineages and ontogeny. Herein, we analyzed gland morphology before and after metamorphosis of Rhinella horribilis, Incilius melanochlorus, and I. luetkenii . We hypothesized that granular gland development would begin earlier and progress faster in relatively more toxic Rhinella species. Our results showed that the timeline of skin development, the relative dimensions and quantity of structures, and the appearance of protein and mucin content in granular and mucous glands did not vary among Rhinella and Incilius . Furthermore, epidermal mucus and/or giant cells in larval toads may act as sources of chemical defenses prior to gland development. Our findings suggest that gland development is well-conserved among these genera; it is possible that reported differences in toxin profiles are not due to divergent morphogenesis during larval or metamorphic stages, but rather to differences in molecular function or structural differentiation in juveniles or adults. Therefore, complementary studies using integrative techniques such as immunohistochemistry and comparative transcriptomics are needed to uncover the mechanisms responsible for the diversity of chemical defenses found in bufonid toads across development.

Seasonal variation of behavioural thermoregulation in the Spotted Salamander ( )

Abstract:Mounting evidence suggests that temperature seasonality plays a pivotal role in shaping the thermal biology of ectotherms. However, we still have a limited understanding of how amphibians maintain thermal balance in the face of varying temperatures, especially in fossorial species. Due to thermal buffering underground, theory predicts relaxed selection pressure over thermoregulation in fossorial ectotherms. As a result, fossorial ectotherms typically show low thermoregulatory precision and low evidence of thermotactic behaviours when tested in laboratory thermal gradients. In this study, we evaluated how temperature selection ( ) and behavioural thermoregulation differed between seasons in the fossorial Spotted Salamander ( ). By comparing thermoregulatory parameters between the activity and overwintering seasons, we provide evidence that . engages in active behavioural thermoregulation despite its fossorial habit. In both seasons, we found to be consistently offset higher than prevailing thermal conditions. Thermoregulation differed between seasons, with salamanders having higher and showing greater evidence of thermophilic behaviours in the active season compared to the overwintering season. Our study highlights that the combination of behavioural and thermal biology measurements is a necessary step to better understand the mechanisms that underlie body temperature control in amphibians. Ultimately, our study provides a broader understanding of thermoregulation in amphibians, particularly in the context of behavioural responses to seasonality in fossorial species.

Abstract:Terrestrial crabs (brachyurans and anomurans) have invaded land following a variety of pathways from marine and/or via freshwater environments. This transition from water to land requires physiological, ecological, and behavioral adaptations to allow the exploitation of these new environmental conditions. Arguably, the management of salt and water balance (e.g., osmoregulation) is integral for their survival and success in an environment where predominantly low-salinity aquatic (e.g., freshwater) water sources are found, sometimes in only minimal amounts. This requires a suite of morphological and biochemical modifications, especially at the branchial chamber of semi-terrestrial and terrestrial crabs to allow reprocessing of urine to maximize ion uptake. Using knowledge gained from electrophysiology, biochemistry, and more recent molecular biology techniques, we present summarized updated models for ion transport for all major taxonomic groups of terrestrial crabs. This is an exciting and fast-moving field of research, and we hope that this review will stimulate further study. Terrestrial crabs retain their crown as the ideal model group for studying the evolutionary pathways that facilitated terrestrial invasion.
