Abstract:Cadmium pollution is becoming a serious problem due to its nondegradability and substantial negative influence on the normal growth of crops, thereby harming human health through the food chain. Rhizospheric bacteria play important roles in crop tolerance. However, there is little experimental evidence which demonstrates how various cadmium concentrations affect the bacterial community in wheat fields including rhizosphere microorganisms and nonrhizosphere (bulk) microorganisms. In this study, 16S rRNA amplicon sequencing technology was used to investigate bacterial communities in rhizosphere and bulk soils under different levels of pollution in terms of cadmium concentration. Both the richness and diversity of the rhizosphere microorganism community were higher under nonpolluted soil and very mild and mild cadmium-contaminated soils than compared with bulk soil, with a shift in community profile observed under severe cadmium pollution. Moreover, cadmium at various concentrations had greater influence on bacterial composition than for the nonpolluted site. In addition, redundancy analysis (RDA) and Spearman’s analysis elucidated the impact of exchangeable Cd and total Cd on bacterial community abundance and composition. This study suggests that cadmium imposes a distinct effect on bacterial community, both in bulk and rhizosphere soils of wheat fields. This study increases our understanding of how bacterial communities in wheat fields shaped under different concentrations of cadmium.

Abstract:Globally increasing trace metal contamination of soils requires a better mechanistic understanding of metal-stress impacts on microbially mediated nutrient cycling. Herein, a 5-month laboratory experiment was employed to assess the effects of cadmium (Cd) on soil microbial N-cycling processes and associated functional gene abundance, with and without urea amendment. In non-N-amended soils, Cd progressively stimulated microbial populations for N acquisition from initial dissolved organic N (DON) to later recalcitrant organic N. The acceleration of N catabolism was synchronously coupled with C catabolism resulting in increased CO2/N2O fluxes and adenosine triphosphate (ATP) contents. The abundance of microbes deemed inefficient in N catabolism was gradually repressed after an initial stimulation period. We posit that enhanced exergonic N processes diminished the need for endergonic activities as a survival strategy for N communities experiencing metal stress. With urea amendment, Cd exhibited an initial stimulation effect on soil nitrification and a later a promotion effect on mineralization, along with an increase in the associated microbial populations. In N-amended soils, Cd accelerated N/C transformation processes, but decreased N2O and CO2 fluxes by 19 and 14%, respectively. This implies that under eutrophic conditions, Cd synchronously altered microbial C/N metabolism from a dominance of catabolic to anabolic processes. These results infer a nutrient-based adjustment of microbial N-cycling strategies to enhance their metal resistance.

Abstract:Microbially induced carbonate precipitation (MICP) could be a potential green solution to resolve the issue of heavy metal releasing from the sulfide tailings. However, detailed mechanism of heavy metal-biomineralization in sulfide tailings and impact of procedure parameters on in-situ applications remain unexplored. We systematically investigated the biomineralization process in the column tests for a better understanding of the mechanism and effects on the inhibition of cadmium (Cd) releasing from sulfide tailings. Results revealed that uniform and efficient mineralization in the tailings column occurred under bacterial concentration of 1 x 10(8) cfu mL(-1), bacterial retention time of 3 h, concentration of mineralization solution of 0.25 mol L-1, and flow rate of 1.5 mL min(-1). The leachable Cd concentration decreased 80.7% after 7 mineralization cycles. From a suit of characterizations, bacteria can adhere on the tailings and acted as the nucleation sites to induce the mineralization of Ca and Cd (to (Ca0.67, Cd0.33)CO3 and calcite phase); eventually, tailings particles were coated with the growth of mineralized carbonates, resulting in a reduction of exposure for tailings (especially sulfur). And thus, Cd release was inhibited. Results from this study will provide a fundamental basis for future in-situ applications of MICP to mitigate heavy metal pollutions.
Colonized extremophile Deinococcus radiodurans alleviates toxicity of cadmium and lead by suppressing heavy metal accumulation and improving antioxidant system in rice

Abstract:Cadmium (Cd) and lead (Pb) are the major toxic heavy metals accumulated in rice and pose a serious threat to human health. The most important remediation strategy is to reduce the translocation of these heavy metals from polluted soil to rice. Bioremediation using microorganisms had been widely used for preventing environmental heavy metal pollution, and the interaction between microorganisms and plants is critical to reduce the heavy metal stress. In this study, we demonstrated that an extremophile Deinococcus radiodurans, especially its mutant strain-Δdr2577 which is deficient in cell surface-layer, could efficiently prevent the translocation and damages of Cd or Pb in rice. The bacterial cells efficiently removed Cd or Pb from culture medium. Following colonization of Δdr2577 cells in rice root, Cd level decreased to 71.6% in root and 60.9% in shoot, comparing to the plants treated with Cd alone; Pb level decreased to 73.3% in root and 56.9% in shoot, comparing to the plants treated with Pb alone. Meanwhile, the bacterial cells released their intracellular antioxidant-related molecules including glutamate and manganese ions into culture medium. Accumulation of glutamate and manganese ions detected in rice root and shoot ameliorate Cd/Pb-induced oxidative stress as indicated by reduced levels of ROS and enhanced activities of antioxidant enzymes in rice. Our results provide a potential application of an extremophile bacterium in alleviating heavy metal toxicity in rice.
Cadmium Pollution Impact on the Bacterial Community Structure of Arable Soil and the Isolation of the Cadmium Resistant Bacteria

Abstract:Microorganisms play an important role in the remediation of cadmium pollution in the soil and their diversity can be affected by cadmium. In this study, the bacterial community in arable soil samples collected from two near geographical sites, with different degrees of cadmium pollution at three different seasons, were characterized using Illumina MiSeq sequencing. The result showed that cadmium is an important factor to affect the bacterial diversity and the microbial communities in the high cadmium polluted area (the site H) had significant differences compared with low cadmium polluted area (the site L). Especially, higher concentrations of Cd significantly increased the abundance of Proteobacteria and Gemmatimonas whereas decreased the abundance of Nitrospirae. Moreover, 42 Cd-resistant bacteria were isolated from six soil samples and evaluated for potential application in Cd bioremediation. Based on their Cd-MIC [minimum inhibitory concentration (MIC) of Cd 2+ ], Cd 2+ removal rate and 16S rDNA gene sequence analyses, three Burkholderia sp. strains (ha-1, hj-2, and ho-3) showed very high tolerance to Cd (5, 5, and 6 mM) and exhibited high Cd 2+ removal rate (81.78, 79.37, and 63.05%), six Bacillus sp. strains (151-5,151-6,151-13, 151-20, and 151-21) showed moderate tolerance to Cd (0.8, 0.4, 0.8, 0.4, 0.6, and 0.4 mM) but high Cd 2+ removal rate (84.78, 90.14, 82.82, 82.39, 81.79, and 84.17%). Those results indicated that Burkholderia sp. belonging to the phylum Proteobacteria and Bacillus sp. belonging to the phylum Firmicutes have developed a resistance for cadmium and may play an important role in Cd-contaminated soils. Our study provided baseline data for bacterial communities in cadmium polluted soils and concluded that Cd-resistant bacteria have potential for bioremediation of Cd-contaminated soils.

Abstract:The acute respiratory distress syndrome (ARDS) is a form of severe hypoxemic respiratory failure that is characterized by inflammatory injury to the alveolar capillary barrier, with extravasation of protein-rich edema fluid into the airspace. Although many modalities to treat ARDS have been investigated over the past several decades, supportive therapies remain the mainstay of treatment. In this article, we briefly review the definition, epidemiology, and pathophysiology of ARDS and present emerging aspects of ARDS pathophysiology that encompass modulators of the innate immune response, damage signals, and aberrant proteolysis that may serve as a foundation for future therapeutic targets.
A Novel Constructed Carbonate-Mineralized Functional Bacterial Consortium for High-Efficiency Cadmium Biomineralization.

Abstract:A stable, urease-producing consortium (UPC) was constructed for high-efficiency cadmium (Cd) ion mineralization via a short-term and efficient acclimation process (five acclimation transfers). 16S rRNA gene high throughput sequencing and quantitative polymerase chain reaction (qPCR) analyses of the urease subunit C (ureC) gene suggested that the three functional genera, all belonging to the phylum Firmicutes, rapidly increased during the process and finally composed the UPC (70.22-75.41 % of Sporosarcina, 13.83-20.66 % of norank_f Bacillaceae, and 5.91-13.69 % of unclassified_f Bacillaceae). The UPC exhibited good adaptability to a wide range of environmental conditions (a pH range of 4.0-11.0, temperature range of 10 - 45 degrees C, and Cd concentration range of 0 - 200 mg L-1). After 8 h of incubation, 92.87 % of Cd at an initial concentration of 100 mg L-1 was mineralized by UPC, exhibiting a great improvement as compared to the first acclimated consortium (C-1). Furthermore, although the acclimated consortium had been successively transferred 21 times, the Cd biomineralization efficiency remained stable, and this was consistent with the observed stable microbial community structure. X-ray diffraction (XRD) spectra revealed that Cd was mineralized in a (Ca0.67, Cd0.33)CO3 phase. This research obtained a promising microbial resource for the biomineralization of Cd or other hazardous heavy metal contaminants.
Metagenomics-Guided Discovery of Potential Bacterial Metallothionein Genes from the Soil Microbiome That Confer Cu and/or Cd Resistance

Abstract:Metallothionein (MT) genes are valuable genetic materials for developing metal bioremediation tools. Currently, a limited number of prokaryotic MTs have been experimentally identified, which necessitates the expansion of bacterial MT diversity. In this study, we conducted a metagenomics-guided analysis for the discovery of potential bacterial MT genes from the soil microbiome. More specifically, we combined resistance gene enrichment through diversity loss, metagenomic mining with a dedicated MT database, evolutionary trace analysis, DNA chemical synthesis, and functional genomic validation to identify novel MTs. Results showed that Cu stress induced a compositional change in the soil microbiome, with an enrichment of metal-resistant bacteria in soils with higher Cu concentrations. Shotgun metagenomic sequencing was performed to obtain the gene pool of environmental DNA (eDNA), which was subjected to a local BLAST search against an MT database for detecting putative MT genes. Evolutional trace analysis led to the identification of 27 potential MTs with conserved cysteine/histidine motifs different from those of known prokaryotic MTs. Following chemical synthesis of these 27 potential MT genes and heterologous expression in Escherichia coli , six of them were found to improve the hosts' growth substantially and enhanced the hosts' sorption of Cu, Cd, and Zn, among which MT5 led to a 13.7-fold increase in Cd accumulation. Furthermore, four of them restored Cu and/or Cd resistance in two metal-sensitive E. coli strains. IMPORTANCE The metagenomics-guided procedure developed here bypasses the difficulties encountered in classic PCR-based approaches and led to the discovery of novel MT genes, which may be useful in developing bioremediation tools. The procedure used here expands our knowledge on the diversity of bacterial MTs in the environment and may also be applicable to identify other functional genes from eDNA.

Abstract:BackgroundSoil contaminated with the heavy metal Cadmium (Cd) is a widespread problem in many parts of the world. Based on metagenomic analysis, we investigated the functional potential and structural diversity of the microbial community in Cd-contaminated and non-contaminated soil samples and we explored the associated metabolic pathway network in cluster of orthologous groups (COG) and Kyoto Encyclopedia of Genes and Genomes (KEGG).ResultsThe results showed that microorganisms in these soils were quite abundant, and many of them possessed numerous physiological functions. However, Cd-contamination has the potential to reduce the microbial diversity and further alter the community structure in the soil. Notably, function analysis of the crucial microorganisms (e. g. Proteobacteria, Sulfuricella and Thiobacillus) indicated that these bacteria and their corresponding physiological functions were important for the community to cope with Cd pollution. The COG annotation demonstrated that the predominant category was the microbial metabolism cluster in both soil samples, while the relative abundance of metabolic genes was increased in the Cd-contaminated soil. The KEGG annotation results exhibited that the non-contaminated soil had more genes, pathways, modules, orthologies and enzymes involved in metabolic pathways of microbial communities than the Cd-contaminated soil. The relative abundance of some dominant KEGG pathways increased in the Cd contaminated soil, and they were mostly enriched to the metabolism, biosynthesis and degradation of amino acids, fatty acids and nucleotides, which was related to Cd tolerance of the microorganisms.ConclusionsCd-contamination can decrease the taxonomic species of microbes in soil and change the soil microbial composition. The functional pathways involved in the soil change with microbial structure variation, many of which are related to the heavy metal tolerance of soil microbes. The Cd-contaminated soil microbes is a potential resource for exploring cadmium resistant or tolerant bacteria.

Competition of Cd(II) and Pb(II) on the bacterial cells: a new insight from bioaccumulation based on NanoSIMS imaging

Abstract:ABSTRACT Polymetallic exposure causes complex toxicity to microorganisms. In this study, we investigated the responses of Escherichia coli under co-existence of cadmium (Cd) and lead (Pb), primarily based on biochemical analysis and RNA sequencing. Cd completely inhibited bacterial growth at a concentration of 2.41 mmol/L, with its removal rate as low as 95% under equimolar sole Pb stress. In addition, the Raman analysis confirmed the loss of proteins for the bacterial cells. Under the co-existence of Cd and Pb, the Cd toxicity to E. coli was alleviated. Meanwhile, the biosorption of Pb cations was more intense during the competitive sorption with Cd. Transmission electron microscopy images showed that a few cells were elongated during incubation, i.e., the average cellular length increased from 1.535 ± 0.407 to 1.845 ± 0.620 μm. Moreover, NanoSIMS imaging showed that the intracellular distribution of Cd and Pb was coupled with sulfur. Genes regulating sulfate transporter were also upregulated to promote sulfate assimilation. Then, the subsequent production of biogenic sulfide and sulfur-containing amino acids was enhanced. Although this strategy based on S enrichment could resist the polymetallic stress, not all related genes were induced to upregulate under sole Cd stress. Therefore, the S metabolism might remodel the microbial resistance to variable occurrence of heavy metals. Furthermore, the competitive sorption (in contrast to sole Cd stress) could prevent microbial cells from strong Cd toxicity. IMPORTANCE Microbial tolerance and resistance to heavy metals have been widely studied under stress of single metals. However, the polymetallic exposure seems to prevail in the environment. Though microbial resistance can alleviate the effects of exogenous stress, the taxonomic or functional response to polymetallic exposure is still not fully understood. We determined the strong cytotoxicity of cadmium (Cd) on growth, and cell elongation would be driven by Cd stress. The addition of appropriate lead (Pb) showed a stimulating effect on microbial bioactivity. Meanwhile, the biosorption of Pb was more intense during co-existence of Pb and Cd. Our work also revealed the spatial coupling of intracellular S and Cd/Pb. In particular, the S assimilation was promoted by Pb stress. This work elucidated the microbial responses to polymetallic exposure and may provide new insights into the antagonistic function during metal stresses.

Abstract:Sodium sulfate stress is known to improve cadmium (Cd) mobilization in soil and microbial sulfur oxidation, Cd resistance, and the accumulation of stress tolerance-associated metabolites has been correlated with increased soil Cd availability and toxicity. In this study, aerobic soil microcosms with Cd-contamination were stimulated with sodium sulfate to investigate its effects on soil microbial community structure, functional genes, and associated metabolite profiles. Metagenomic analysis revealed that sulfur oxidizing and Cd-resistant bacteria carried gene clusters encoding sox, dsr, and sqr genes, and znt, czc, and cad genes, respectively. Exposure to sodium sulfate resulted in the reprogram of soil metabolites. In particular, intensification of sulfur metabolism triggered an up-regulation in the tricarboxylic acid (TCA) cycle, which promoted the secretion of carboxylic acids and their precursors by soil bacteria. The accumulation of organic acids induced in response to high sodium sulfate dosages potentially drove an observed increase in Cd mobility. Pseudomonas and Erythrobacter spp. exhibited a high capacity for adaptation to heavy metal- or sulfur-induced stress, evident by an increased abundance of genes and metabolites for sulfur cycling and Cd resistance. These results provide valuable insights towards understanding the microbial mechanisms of sulfur transformation and Cd dissolution under saline stress.

Genomic and transcriptomic analyses reveal insights into cadmium resistance mechanisms of Cupriavidus nantongensis strain E324

Abstract:The cadmium-resistant Cupriavidus sp. strain E324 has been previously shown to have a high potential for use in cadmium (Cd) remediation, due to its high capacity for cadmium bioaccumulation. According to the comparative genomic analysis, the strain E324 was most closely related to C. nantongensis X1T, indicating that the strain E324 should be re-identified as C. nantongensis. To unravel the Cd tolerance mechanisms of C. nantongensis strain E324, the transcriptional response of this strain to acute Cd exposure was assessed using RNA-seq-based transcriptome analysis, followed by validation through qRT-PCR. The results showed that the upregulated Differentially Expressed Genes (DEGs) were significantly enriched in categories related to metal binding and transport, phosphate transport, and oxidative stress response. Consistently, we observed significant increases in both the cell wall and intracellular contents of certain essential metals (Cu, Fe, Mn, and Zn) upon Cd exposure. Among these, only the Zn pretreatment resulting in high Zn accumulation in the cell walls could enhance bacterial growth under Cd stress conditions through its role in inhibiting Cd accumulation. Additionally, the promotion of catalase activity and glutathione metabolism upon Cd exposure to cope with Cd-induced oxidative stress was demonstrated. Meanwhile, the upregulation of phosphate transport-related genes upon Cd treatment seems to be the bacterial response to Cd-induced phosphate depletion. Altogether, our findings suggest that these adaptive responses are critical mechanisms contributing to increased Cd tolerance in C. nantongensis strain E324 via the enhancement of metal-chelating and antioxidant capacities of the cells.
Study on the regulatory mechanism of the earthworm microbial community in vitro and in vivo under cadmium stress

Abstract:In this paper, cadmium (Cd) stress tests were performed on Eisenia fetida in sterile artificial soil, and its regulatory mechanism between microbial communities in vivo and in vitro after Cd stress was explored. In the test, 0, 50, 100, 125, 250 and 500 mg.kg-1 Cd stress concentrations were implemented. After long-term and short-term stress, the microbes in the earthworms and the soil were cultured with ECO plates. The data statistics of carbon source utilization intensity were carried out using the method developed by our team. CCA was scientifically integrated into TOPSIS to establish a new data analysis model to find the regulatory nodes after stress[<a class="workspace-trigger" href="#bib1">1</a>]. Macro gene sequencing technology revealed that the species with the highest absolute abundance in the microbial communities in vivo and in vitro were all unnamed new species. It was confirmed that the HBA gene, NEUROD1 gene and ABCA3 gene were the regulatory genes of the microbial community in the earthworms under Cd stress, while the TC.FEV.OM gene and cheBR gene were the main regulatory genes of the microbial community in the soil. These results provide a scientific and theoretical reference and model basis for the bioremediation of Cd-contaminated soil and the detoxification mechanism of earthworms.

Biomineralization of Cd2+ and inhibition on rhizobacterial Cd mobilization function by Bacillus Cereus to improve safety of maize grains

Abstract:Reducing cadmium (Cd) bioavailability and rhizobacterial Cd mobilization functions in the rhizosphere via the inoculation of screened microbial inoculum is an environmental-friendly strategy to improve safety of crop grains. In this study, Bacillus Cereus, a model Cd resistant strain, was selected to explore its effects on Cd bioavailability and uptake, bacterial metabolic functions related to Cd mobilization. Results indicated that inoculation of Bacillus Cereus in maize roots of sand pot with water-soluble Cd (0.06–0.15 mg/kg) and soil pot with high Cd-contaminated soil (total Cd: 2.33 mg/kg; Cd extracted by NH4NO3: 38.6 μg/kg) could decrease water-soluble Cd ion concentration by 7.7–30.1% and Cd extracted with NH4NO3 solution by 7.8–22.5%, inducing Cd concentrations in maize grains reduced by 10.6–39.9% and 17.4–38.6%, respectively. Even for a single inoculation in soil, Cd concentration in maize grains still satisfy food safety requirements (Cd content: 0.1 mg/kg dry weight) due to its successful colonization on root surface of maize. Bacillus Cereus could enrich more plant growth promotion bacteria (PGPB) and down-regulate the expression of genes related to bacterial motility, membrane transports, carbon and nitrogen metabolism in the rhizosphere soil, decreasing Cd bioavailability in soil. Approximately 80% Cd2+ in media was transferred into intracellular, meanwhile Cd salts (sulfide and/or phosphate) were produced in Bacillus Cereus through biomineralization process. Overall, this study could provide a feasible method for improving safety of maize grains via the inoculation of Bacillus Cereus under Cd pollution.

Abstract:Microbial communities as the most important and active component of soil play a crucial role in the geochemical cycling of toxic metal(loid)s in the Pb and Zn smelting site soils. However, the relationships between soil microbial communities and the fractions of toxic metal(loid)s and the succession of soil microbial community and functions after enrichment cultivation have rarely been analyzed. In this study, the diversity and composition of microbial communities in soils before and after enrichment cultivation were investigated by high-throughput sequencing. And the co-occurrence relationships between soil microbial community after enrichment cultivation and MRGs genes were also analyzed through the BacMet database. Results showed that the dominant genus in the soils was Lactobacillus and Stenotrophomonas. The soil microbial community exhibited a notable correlation with Cd, Pb, and As, among which Cd exerted the most profound impact. Alishewanella, Pseudomonas, Massilia and Roseibacillus were significantly correlated with the fraction of Cd. After enrichment cultivation, the number of genera decrease to 96. And the dominant genus changed to Acinetobacter, Bacillus, Comamonas, Lysobacter, and Pseudoxanthomonas. High abundance of metal resistance genes (MRGs) including zntA, fpvA, zipB, cadA, czcA, czcB, czcC, zntA, arsR, pstS and pstB was found in the microbial community after enrichment cultivation. The potential host genus for MRGs was Acinetobacter, Comamonas, Lysinibacillus, Azotobacter, Bacillus, Lysobacter, Cupriavidus, Pseudoxanthomonas, and Thermomonas. Additionally, these microbial community after enrichment cultivation possessing pathways of bacterial chemotaxis and two-component systems was enabled them to adapt to the polluted environment. These observations provided potential guidance for microbe isolation and the development of strategies for the bioremediation of toxic metal(loid)s polluted soils.

Abstract:Interferon inhibits the early phase of mitogenic action of Concanavalin A on lymphocytes as measured by incorporation of labelled precursors in RNA, DNA or protein. There was no evidence for a role of cyclic AMP in the process of inhibition. Dialysis of interferon against high salt and urea did not separate cell inhibitory activity from antiviral activity. Thus, these results are compatible with the possibility that the antiviral and cell inhibitory activities are in the same molecule. However, further purification of the interferon will be required to verify this assumption.

Abstract:Toxic metal-contaminated farmland from Cadmium (Cd) can enhance the accumulation of Cd and impair the absorption of mineral elements in brown rice. Although several studies have been conducted on Cd exposure on rice, little has been reported on the relationship between Cd and mineral elements in brown rice and the regulatory mechanism of rhizosphere microorganisms during element uptake. Thus, a field study was undertaken to screen japonica rice cultivars with low Cd and high mineral elements levels, analyze the quantitative relationship between Cd and seven mineral elements, and investigate the cultivar-specific response of rice rhizosphere bacterial communities to differences in Cd and mineral uptake in japonica rice. Results showed that Huaidao-9 and Xudao-7 had low Cd absorption and high amounts of mineral nutrient elements (Fe, Zn, Mg, and Ca, LCHM group), whereas Zhongdao-1 and Xinkedao-31 showed opposite accumulation characteristics (HCLM group). Stepwise regression analysis showed that zinc, iron, and potassium are the key minerals that affect Cd accumulation in japonica rice and zinc was the most important factor, accounting for 68.99 %. The accumulation of Cd and mineral elements is potentially associated with rhizosphere soil bacteria. Taxa enriched in the LCHM rhizosphere (phyla Acidobacteriota and MBNT15) indicated the high nutrient characteristics of the soil and reduced activity of Cd in soil. The HCLM rhizosphere was highly colonized by metal-activating bacteria (Actinobacteria), lignin-degrading bacteria (Actinobacteria and Chlorofexi), and bacteria scavenging nutrients and trace elements (Anaerolinea and Ketobacter). Moreover, the differences in the uptake of Cd and mineral elements affected predicted functions of microbial communities, including sulfur oxidation and sulfur derivative formation, human or plant pathogen, and functions related to the iron oxidation and nitrate reduction. The results indicate a potential association of Cd and mineral elements uptake and accumulation with rhizosphere bacteria in rice, thus providing theoretical basis and a new perspective on the maintenance of rice security and high quality simultaneously.

Abstract:Enterobacter sp. are widely used in bioremediation, but the mechanism of Cadmium (Cd) toxicity in Enterobacter sp. has been poorly studied. In the present study, we determined the tolerance of Enterobacter sp. FM-1 to Cd by analyzing the physiological and biochemical responses of FM-1 induced under Cd stress. Differentially expressed proteins (DEPs) under exposure to different Cd environments were analyzed by 4D-label-free proteomics to provide a comprehensive understanding of Cd toxicity in FM-1. The greatest total number of DEPs, 1148, was found in the High concentration vs. Control comparison group at 10 h. When protein expression was compared after different incubation times, FM-1 showed the highest Cd tolerance at 48 h. Additionally, with an increasing incubation time, different comparison groups gradually began to show similar growth patterns, which was reflected in the GO enrichment analysis. Notably, only 815 proteins were identified in the High concentration vs. Control group, and KEGG enrichment analysis revealed that these proteins were significantly enriched in the pyruvate metabolism, oxidative phosphorylation, peroxisome, glyoxylate and dicarboxylate metabolism, and citrate cycle pathways. These results suggested that an increased incubation time allows FM-1 adapt and survive in an environment with Cd toxicity, and protein expression significantly increased in response to oxidative stress in a Cd-contaminated environment during the pre-growth period. This study provides new perspectives on bacterial participation in bioremediation and expands our understanding of the mechanism of bacterial resistance under Cd exposure.

Abstract:The utility of endophytic bacteria in Cadmium (Cd) remediation has gained significant attention due to their ability to alleviate metal-induced stress and enhance plant growth. Here, we investigate C. metallidurans CML2, an endophytic bacterial strain prevalent in rice, showing resilience against 2400 mg/L of Cd(II). We conducted an in-depth integrated morphological and transcriptomic analysis illustrating the multifarious mechanisms CML2 employs to combat Cd, including the formation of biofilm and CdO nanoparticles, upregulation of genes involved in periplasmic immobilization, and the utilization of RND efflux pumps to extract excess Cd ions. Beyond Cd, CML2 exhibited robust tolerance to an array of heavy metals, including Mn2+, Se4+, Ni2+, Cu2+, and Hg2+, demonstrating effective Cd(II) removal capacity. Furthermore, CML2 has exhibited plant growth-promoting properties through the production of indole-3-acetic acid (IAA) at 0.93 mg/L, soluble phosphorus compounds at 1.11 mg/L, and siderophores at 22.67%. Supportively, pot experiments indicated an increase in root lengths and a decrease in Cd bioaccumulation in rice seedlings inoculated with CML2, consequently reducing Cd translocation rates from 43% to 31%. These findings not only contribute to the understanding of Cd resistance mechanisms in C. metallidurans, but also underscore CML2's promising application in Cd remediation within rice farming ecosystems.
