Polydopamine Nanosheets Doped Injectable Hydrogel with Nitric Oxide Release and Photothermal Effects for Bacterial Ablation and Wound Healing

Abstract:The development of wound dressings combined with both efficient antibacterial activities and pro-healing functions has always been an intractable medical task for treating bacterial wound infection. Herein, we developed a novel hybrid hydrogel dressing loaded with two- dimensional polydopamine nanosheets (PDA NS) and the nitric oxide donor (BNN6) for skin wound healing. This three-dimensional hydrogel was fabricated in situ by dynamic Schiff base bonding from hydrazide-modified γ-polyglutamic acid (γ-PGA-ADH) and aldehyde-terminated Pluronic F127 (F127-CHO), as well as micellization of F127-CHO. The two dynamic cross-linking contributed to the hydrogel with self-healing ability, injection property, and temperature sensitivity. PDA NS could transform 808 nm near-infrared light (NIR) into heat owing to its outstanding photothermal properties (56.1%). Moreover, BNN6 loaded on PDA NS could realize NIR triggering NO release. The combination of photothermal and NO gas therapy endowed the hydrogel with a synergistic antibacterial effect on both Escherichia coli and Staphylococcus aureus in vitro. Furthermore, an in vivo full-thickness skin defect model was constructed and the evaluations demonstrated that the hybrid hydrogel showed the outstanding antibacterial properties and effectively accelerated wound healing process. Overall, this study will provide a facile and promising method for the fabrication of PDA nanosheets based multifunctional hydrogel dressing for the application of infectious skin wound healing.This article is protected by copyright.
