Abstract:As a novel trend, solid carbon sources are applied to act as electron donors and biofilm carrier in biological denitrification process. In this study, simultaneous nitrate and ammonium removal process in an airlift sequencing batch reactor using 3-hydroxybutyrate-co-3-hydroxyvalerate (PHBV) as carbon source and biofilm carrier under intermittent aeration conditions was established to treat effluent of synthetic marine recirculating aquaculture system. The results showed that maximum nitrate and ammonia nitrogen removal rates of 0.45 and 0.09 kg m- 3 d-1 were achieved. No significant nitrite accumulation was found during 200-day operation, while effluent dissolved organic carbon accumulation and particle size reduction significantly increased. Microbial community analysis and batch tests illuminate that the generated sludge and attached biofilm played important roles in nitrogen removal. This study demonstrates the potential mechanism for the nitrogen removal process mediated by 3-hydroxybutyrateco-3-hydroxyvalerate and provide a new idea for the alternative solutions of solid carbon sources.
Nitrogen Removal from Sludge Dewatering Effluent Through Anaerobic Ammonia Oxidation Process.

Abstract:Anaerobic ammonia oxidation(Anammox) process is a novel and promising wastewater nitrogen removal process. The feasibility of transition of Anammox from denitrification and the performance of lab-scale Anammox biofilm reactor were investigated with sludge dewatering effluent. The results showed that Anammox process could be successfully started up after cultivation of denitrification biofilm and using it as inoculum. The transition of Anammox from denitrification was accomplished within 85 d. Anammox process was found suitable to remove ammonia from sludge dewatering effluent. The effluent ammonia concentration was detected to be 23.11 mgN/L at HRT of 28 h when influent ammonia concentration was fed 245 mgN/L, which was less than that for the national discharge standard II (25 mgN/L) of China. Volumetric total nitrogen loading rate was up to 584.99 mg TN/(L x d) at HRT of 17 h, while influent concentrations were kept 243.25 mg NH4(+) -N/L and 288.31 mg NO2(-) -N/L.

Abstract:A novel strain FDN-02 was isolated from a sequencing batch biofilm reactor. FDN-02 was identified as Paracoccus sp., and the Genbank Sequence_ID was MW652628. Comparing with the removal efficiency of ammonia nitrogen (NH4+N) by bacterium FDN-02 under different growth conditions, the optimal initial pH, carbon source, and C/N ratio were 7.0, sucrose, and 16, respectively. The maximum removal efficiency and rate of NH4+-N were respectively 96.2% and 10.06 mg-N/L/h within 8 h under anoxic condition when the concentration of NH4+-N was 44.87 mg/L. Specifically, 71.9% of NH4+-N was utilized by strain FDN-02 through heterotrophic assimilation to synthetize organic nitrogen, and approximately 24.1% of NH4+-N was lost in the form of gaseous nitrogen without the emission of nitrous oxide. Bacterium FDN-02 was also found to be a denitrifying organism, and nitrate nitrogen and nitrite nitrogen of lower concentrations were removed by denitrification after the enlargement of biomass. Further investigation showed that the biomass after the removal of NH4+-N by strain FDN-02 had resource utilization potential, and the contents of proteins and amino acids were 635 and 192.97 mg/g, respectively, especially for the usage as an alternative nutrient source for livestock and organic fertilizers. This study provided a promising environmentally friendly biological treatment method for the removal of NH4+-N in the wastewater.

Abstract:Ammonium-nitrogen pollution is the main cause of waterbody eutrophication and has been a severe problem for the treatment of river and groundwater. In this paper, a novel Advanced Nitrogen REmoval (ANRE) process was successfully developed based on anaerobic ammonium oxidation (ANAMMOX), aiming at the treatment of low-strength ammonium-containing wastewaters (5-15 mg/L NH4+-N). The ANRE process was demonstrated to be high-efficient with the substrate (ammonium and nitrite) removal efficiency up to 97% and the effluent ammonium concentration less than 1 mg N/L. Kinetic analysis indicated that the maximum specific ammonium utilization rate was 46.40 mg N/(gVSS.d) and the half-saturation constants k(s) was 0.25 mg N/L. The ANAMMOX granular sludge was observed to disaggregate gradually due to the consumption of extracellular polymeric substances (EPS). The Anaerobic Ammonium Oxidation Bacteria (AnAOB) were proved in the state of starvation, causing the stoichiometric change for cellular synthesis and energy production and leading to more nitrate accumulation. Efficient detoxication of hydroxylamine and nitrite through heterotrophic nitrification and aerobic denitrification by Acinetobacter johnsonii EN-J1

Abstract:The co-existence of hydroxylamine (NH 2 OH) and nitrite (NO 2 – -N) can aggravate the difficulty of wastewater treatment. The roles of hydroxylamine (NH 2 OH) and nitrite (NO 2 – -N) in accelerating the elimination of multiple nitrogen sources by a novel isolated strain of Acinetobacter johnsonii EN-J1 were investigated in this study. The results demonstrated that strain EN-J1 could eliminate 100.00% of NH 2 OH (22.73 mg/L) and 90.09% of NO 2 – -N (55.32 mg/L), with maximum consumption rates of 1.22 and 6.75 mg/L/h, respectively. Prominently, the toxic substances NH 2 OH and NO 2 – -N could both facilitate nitrogen removal rates. Compared with the control treatment, the elimination rates of nitrate (NO 3 – -N) and NO 2 – -N were enhanced by 3.44 and 2.36 mg/L/h after supplementation with 10.00 mg/L NH 2 OH, and those of ammonium (NH 4 + -N) and NO 3 – -N were improved by 0.65 and 1.00 mg/L/h after the addition of 50.00 mg/L NO 2 – -N. Furthermore, the nitrogen balance results indicated that over 55.00% of the initial total nitrogen was transformed into gaseous nitrogen by heterotrophic nitrification and aerobic denitrification (HN-AD). Ammonia monooxygenase (AMO), hydroxylamine oxidoreductase (HAO), nitrate reductase (NR), and nitrite reductase (NIR), which are essential for HN-AD, were detected at levels of 0.54, 0.15, 0.14, and 0.01 U/mg protein, respectively. All findings confirmed that strain EN-J1 could efficiently execute HN-AD, detoxify NH 2 OH and NO 2 – -N, and ultimately promote nitrogen removal rates.

Abstract:Partial nitrification (PN) process is an essential pre-treatment to Anammox process for nitrogen removal from ammonium-rich wastewaters. High activity nitrifiers (HANs) are considered as the key to high-rate PN process. The HANs could be successfully enriched at free ammonia (FA) concentrations of 10–20mgNH3–NL−1, which differed significantly from those enriched at FA concentrations of 1–3mgNH3–NL−1. The maximum specific ammonium conversion rate, half saturation constant, half inhibition constant, cell yield and decay rate were 2.87gNg−1VSSd−1, 5.82mgNL−1, 8218.8mgNL−1, 0.15gVSSg−1 NH4+–N and 0.09d−1 for HANs at 30°C, respectively, which endowed the PN process a volumetric conversion rate (VCR) as high as 2.22±0.33kgNm−3d−1. The cultivation mode of HANs was workable and the HANs were promising as inoculum for the PN process.
Nitrogen removal characteristics of efficient heterotrophic nitrification-aerobic denitrification bacterium and application in biological deodorization

Abstract:A heterotrophic nitrifying aerobic denitrifying (HN-AD) strain HY-1 with excellent capacity, identified as Paracoccus denitrificans, was isolated from activated sludge. HY-1 was capable of removing NH4+, NO2-, and NO3- with the corresponding rate of 17.33 mg-N L-1 h-1, 21.83 mg-N L-1 h-1, and 32.37 mg-N L-1 h-1, as well as the mixture of multiple nitrogen sources. Meanwhile, HY-1 could execute denitrification function under anaerobic conditions with a rate of 14.56 mg-N L-1 h-1. HY-1 required less energy investment, which exhibited average denitrification rate of 5.19 mg-N L-1 h-1 at carbon-nitrogen ratio was 1. After nitrification-denitrification metabolic pathway analysis, HY-1 was applied in a biological trickling filter reactor for compost deodorization. The results showed that adding of HY-1 greatly reduced the ionic concentration of NH4+ and NO3- in the circulating liquid without impairing the deodorization effect (NH3 removal rate＞98.07%). These findings extend the field of application of HN-AD and provide new insights for biological deodorization.
Simultaneous Nitrification and Aerobic Denitrification by a Novel Isolated Ochrobactrum Anthropi HND19

Abstract:The study aimed to isolate a novel strain with heterotrophic nitrification and aerobic denitrification ability and evaluate the nitrogen removal characteristics. Results showed that Ochrobactrum anthropi HND19 could remove approximately 98.6% of NH4+-N (104.3 mg.L-1) and 97.6% of NO3--N (98.6 mg.L-1 ), and the removal rates achieved 4.28 and 4.01 mg-N/(L.h) by heterotrophic nitrification and aerobic denitrification. The optimal incubate conditions of strain HND19 were 120 rpm (shaking speed), 5 parts per thousand (salinity), 30 degrees C (temperature), 7.5 (C/N ratio) with sodium acetate as carbon resource. And the removal efficiency of the total nitrogen (TN) realized 73.4% under the optimal conditions. Functional genes (hao, napA, nirK, norB, and nosZ) involved in the nitrogen removal processes were successfully amplified from strain HND19. These findings indicate that the strain HND19 possesses great application feasibility in treating wastewater with high-intensity nitrogen.

Abstract:Introduction: Nitrogen and arsenic contaminants often coexist in groundwater, and microbes show the potential for simultaneous removal of nitrogen and arsenic. Here, we reported that Hydrogenophaga sp. H7 was heterotrophic nitrification and aerobic denitrification (HNAD) and arsenite [As(III)] oxidation bacterium. Methods: The appearance of nitrogen removal and As(III) oxidation of Hydrogenophaga sp. H7 in liquid culture medium was studied. The effect of carbon source, C/N ratio, temperature, pH values, and shaking speeds were analyzed. The impact of strains H7 treatment with FeCl3 on nitrogen and As(III) in wastewater was assessed. The key pathways that participate in simultaneous nitrogen removal and As(III) oxidation was analyzed by genome and proteomic analysis. Results and discussion: Strain H7 presented efficient capacities for simultaneous NH 4 + -N, NO 3 − -N, or NO 2 − -N removal with As(III) oxidation during aerobic cultivation. Strikingly, the bacterial ability to remove nitrogen and oxidize As(III) has remained high across a wide range of pH values, and shaking speeds, exceeding that of the most commonly reported HNAD bacteria. Additionally, the previous HNAD strains exhibited a high denitrification efficiency, but a suboptimal concentration of nitrogen remained in the wastewater. Here, strain H7 combined with FeCl3 efficiently removed 96.14% of NH 4 + -N, 99.08% of NO 3 − -N, and 94.68% of total nitrogen (TN), and it oxidized 100% of As(III), even at a low nitrogen concentration (35 mg/L). The residues in the wastewater still met the V of Surface Water Environmental Quality Standard of China after five continuous wastewater treatment cycles. Furthermore, genome and proteomic analyses led us to propose that the shortcut nitrification-denitrification pathway and As(III) oxidase AioBA are the key pathways that participate in simultaneous nitrogen removal and As(III) oxidation.

Abstract:A heterotrophic nitrifier wsw-1001 was isolated from Songhua River and identified as Pseudomonas fluorescens. Ammonium removal by the strain at low temperature was investigated. The effect of initial ammonium concentration (from 5 to 1000 mg/L) and culture temperature (from 4°C to 30°C) on ammonium removal efficiency was studied. Biodegradation product, [Formula: see text], [Formula: see text], N2, N2O and intercellular N were monitored. The results indicated that the strain had potential for water and wastewater treatment. Ammonium could be removed by the strain at low temperature. Ammonium removal efficiency increased with temperature from 4°C to 20°C and decreased with ammonium concentration from 5 to 1000 mg/L. The strain exhibited a capability of heterotrophic nitrification and aerobic denitrification using [Formula: see text] as the sole nitrogen source at 8°C. [Formula: see text] and [Formula: see text] were reduced by the strain. Nitrogen balance analysis in the presence of 39.7 mg/L [Formula: see text] indicated that 71.2% [Formula: see text] was removed by converting to N2 (46.3%) and assimilating as biomass (42.5%). Substances such as [Formula: see text], [Formula: see text] and N2O were detected at very low concentrations. Ammonium mono-oxygenase, hydroxylamine oxidase, nitrite reductase and nitrate reductase activity were measured. The ammonium removal pathway of the strain was speculated to be [Formula: see text].

Abstract:A novel oligotrophic heterotrophic nitrification-aerobic denitrification bacterium designated as Pseudomonas sp. N31942, was isolated from a eutrophic lake. Strain N31942 exhibits high ammonia nitrogen removal ability in oligotrophic environment as ammonia nitrogen can be efficiently (86.97 %) removed within 10 h with no accumulation of nitrite. In the nitrification process, strain N31942 can convert ammonia into nitrate in the absence of hydroxylamine oxidase and nitrite oxidoreductase. As for the denitrification process, nitrate or nitrite were reduced to ammonia and further converted into glutamate by dissimilatory nitrate reduction pathway. Transcriptomic analysis detected 2080 differentially expressed genes. Among them, the expression of the related genes in dissimilatory nitrate reduction process was all up-regulated at low ammonia concentrations, which indicates that the strain has excellent nitrogen removal efficiency for further nitrogen removal. Integrative omics analyses revealed that strain N31942 may have two possible pathways for the NH4+-N removal as direct GDH/GS-GOGAT pathway (NH4+-N → Glutamate) and indirect GDH/GS-GOGAT pathway (NH4+-N → NH2OH → NO2−-N → NO3−-N → NO2−-N → NH4+-N → Glutamate). Moreover, strain N31942 also has excellent nitrogen removal ability for real sewage and 77.21 % total nitrogen could be removed within 48 h. The results presented here provide new insights into ammonia nitrogen removal characteristics and mechanism of heterotrophic nitrification-aerobic denitrification bacterium under oligotrophic conditions.

Abstract:Recently, the functional microorganisms capable of eliminating nitrogenous waste have been applied in mariculture systems. As a potential candidate for treating mariculture wastewater, strain DN3 eliminated 100% of ammonia and nitrate and 96.61%–100% of nitrite within 72 h, when single nitrogen sources at concentrations of 0–50 mg/L. Strain DN3 also exhibited the efficient removal performance of mixed-form nitrogen (ammonia, nitrate, and nitrite) at salinity 30 ‰, C/N ratio 20, and 180 rpm. The nitrogen assimilation pathway dominated inorganic nitrogen metabolism, with less nitrogen (14.23%-25.02% of TN) lost into the air via nitrification and denitrification, based on nitrogen balance analysis. Moreover, the bacterial nitrification pathway was explored by enzymatic assays and inhibition assays. These complex nitrogen assimilation and dissimilation processes were further revealed by bacterial genome analysis. These results provide important insight into nitrogen metabolism of Halomonas sp. and theoretical support for treating mariculture wastewater with strain DN3.

Abstract:A novel heterotrophic nitrifying bacterium with high salt and high ammonia nitrogen tolerance, Alcaligenes faecalis TF-1, was isolated from the leachate of a landfill. The verification of nitrogen removal efficiency of different nitrogen sources and PCR amplification electrophoresis results showed that the HN-AD pathway of the strain TF-1 was NH 4 + → NH 2 OH → NO → N 2 O → N 2 . The results of parameter optimization showed that the optimal nitrogen removal conditions were as follows: sodium citrate as carbon source, C/N = 16, pH = 7, and NH 4 + -N loading of 808.21 mg/L. The strain TF-1 could remove about 94.60% of ammonia nitrogen (1963.94 mg/L). The salinity tolerance range of the strain TF-1 was 0–70 g/L, and the removal efficiency was 52.87% at salinity 70 g/L and NH 4 + -N concentration 919.20 mg/L and 55.67% at pH = 10 and NH 4 + -N concentration 994.82 mg/L. The extreme environmental adaptability and remarkable nitrogen removal performance make this strain a promising candidate in leachate treatment.

Abstract:The NH2OH dosing strategy for nitrogen removal was investigated in a single return continuous-flow anaerobic/aerobic/anoxic/aerobic (AnOAO) reactor fed with real municipal wastewater. A high nitrite accumulation ratio of 98% was achieved in only two days by continuously adding 10 mg/L NH2OH. When gradually reducing dosing frequency to one day every four days, effluent total nitrogen was as low as 4.8 ± 2.2 mg N/L with removal efficiency of 88.7±5.3%, under aerobic HRT of 4.6 h, DO below 1.0 mg/L, and C/N of 2.8 without external carbon sources. Batch test showed that nitrite oxidizing bacteria (NOB) activity decreased by 81% after adding NH2OH, while ammonia oxidizing bacteria (AOB) activity remained stable. qPCR confirmed that NOB abundance decreased, and 16S rRNA sequencing further showed that g_Nitrospira belonging to NOB decreased significantly (P < 0.001). Overall, this study provides a novel strategy for advanced nitrogen removal from municipal wastewater in continuous flow systems.

Genomics and nitrogen metabolic characteristics of a novel heterotrophic nitrifying-aerobic denitrifying bacterium Acinetobacter oleivorans AHP123

Abstract:A novel aerobic strain of Acinetobacter oleivorans AHP123 was isolated from activated sludge, which could conduct heterotrophic nitrification and denitrification simultaneously. This strain has excellent NH4+-N removal ability, with 97.93% removal rate at 24-hour. To identify the metabolic pathways of this novel strain, genes of gam, glnA, gdhA, gltB, nirB, nasA, nar, nor, glnK and amt were detected by genome analysis. Through RT-qPCR, it was found that the expression of key genes confirmed two possible ways of nitrogen removal in strain AHP123: nitrogen assimilation and heterotrophic nitrification aerobic denitrification (HNAD). However, the absence of some common HNAD genes (amo, nap and nos) suggested that strain AHP123 might have a different HNAD pathway from other HNAD bacteria. Nitrogen balance analysis revealed that strain AHP123 assimilated most of the external nitrogen sources into intracellular nitrogen.

Abstract:Low grade mucinous tubular and spindle cell carcinoma of kidney was newly established as a distinct renal cell carcinoma in the World Health Organization (WHO) classification of 2004. Until now, less than 60 cases have been reported and the largest series represented approximately 15 patients with this type of tumor. Herein, we report a case of mucinous tubular and spindle cell carcinoma in a 63-year-old male presented with right flank pain which was diagnosed after nephrectomy. Pathologists should consider this diagnosis and its spectrum of histopathologic features in mind to ensure an accurate diagnosis.

Abstract:In this study for bioaugmentation in the treatment of high nitrogen content wastewater,a well functioning heterotrophic nitrification-aerobic denitrification bacteria strain was isolated from activated sludge and named HN-02.By morphological,physiological,biochemical identification and 16S rDNA sequence analysis,the short rod-shaped Gram-negative bacteria strain was identified as Aeromonas bacteria.It's partial napA gene was cloned.The best carbon source for HN-02 bacteria was succinic acid,followed by sucrose and sodium citrate;the optimum temperature was 30 ℃;and the optimum pH range was 6 to 9.Under aerobic conditions,various forms of nitrogen could be removed by HN-02 independently.The 24 h TN removal efficiencies were 77.71%,53.34%,56.80%,respectively,with ammonia nitrogen,nitrite,nitrate as sole nitrogen source.By qualitative verification,the nitrogen removal product gas contained nitrogen oxides.

Abstract:A thermotolerant strain isolated from sewage sludge (SS) composting was identified as Brevibacillus Agri N2, which showed the efficient capability for heterotrophic nitrification under high-temperature conditions. Incubation at 60 °C, strain N2 could utilize 45.47% of ammonium nitrogen (99.64 mg/L), 68.89% of hydroxylamine nitrogen (51.14 mg/L) and 76.77% of nitrite nitrogen (55.20 mg/L), with a minor part of nitrogen loss for 1.64%, 2.82% and 5.01%, respectively. The successful detection of ammonia monooxygenase, hydroxylamine oxidase, and nitrate oxidoreductase and PCR amplification of amoA, hao and nxrA genes provided evidence of nitrification ability by strain N2. Furthermore, single-factor experiments indicated that the optimal conditions for efficient nitrification performance by strain N2 were succinate as carbon source, 50 °C, C/N 12, pH 8 and 200 r/min. Strain N2 could perform the complete nitrification process, with minimal nitrogen loss at high temperature conditions, which indicated it had the potential for practical application for reducing nitrogen loss of SS composting.

Abstract:Partial nitritation–Anammox process is regarded as a promising nitrogen removal method for nitrogen removal from low C/N wastewaters. In the present work, stable partial nitrification was successfully achieved by 10mg/l hydroxylamine dosing in aerobic granules under experimental conditions of pH 7.8–8.2, dissolved oxygen (DO) above 5mg/l and temperature around 25°C. When the NH4–N and COD were fixed at 100mg/l and 400mg/l, the TN removal efficiency was 57% with a NO2–N/NOx–N (NO2+NO3)–N ratio of 99.8% in the effluent. Meanwhile, 79% COD was removed by aerobic granules, which was regarded as beneficial to the following Anammox. Sequentially, a mixture of the SBR effluent and raw influent with an average NO2–N/NH4–N ratio of 1.67 was fed to an Anammox reactor. The COD and TN removal efficiencies reached 77% and 81%, respectively, through coupling of Anammox and denitrification. SEM detection showed that the main composition of aerobic granules was coccus and bacilli bacteria. FISH results demonstrated that ammonia-oxidizing bacteria (AOB) became the dominant bacteria of aerobic granules, which further proved the important effects of hydroxylamine on the achievement of partial nitrification by aerobic granules under conditions of high DO and ambient temperature.

Abstract:Partial nitrification-anaerobic ammonium oxidation (PN-anammox) was started up within 40 days by bioaugmentation and aeration control, and its performance in the treatment of anaerobic digestate of pig manure (ADPM) was evaluated. Inhibitors in ADPM decreased the nitrogen removal rate (NRR) by 0.24 g N/L/d. The effect and mechanism of hydroxylamine (NH 2 OH) alleviation of PN-anammox inhibition during ADPM treatment were investigated. As an intermediate product of anammox and ammonia-oxidizing bacteria, NH 2 OH strengthened energy metabolism, improved the activity and abundance of functional bacteria, and eliminated miscellaneous bacteria, increasing the average NRR by 31%. However, the average nitrous oxide emission was increased by 10.1% via hydroxylamine oxidation. The results showed that synergy and competition among nitrogen-transforming microorganisms were crucial for NRR and that NH 2 OH played an essential role in maintaining efficient operation. This study lays a foundation for restoring PN-anammox for treating livestock wastewater.
