Abstract:Stimuli‐responsive materials have properties that depend on the environment in which they are used. In most cases, the material itself is formulated to react to the corresponding stimulus. However, many phenomena occur at the surface of the material. In this context, the design and the investigation of the reactivity of stimuli‐responsive surfaces are particularly interesting. More precisely, this review focuses on functional coatings that react via Diels–Alder (DA) chemistry, a thermoreversible reaction between a diene and a dienophile. According to the nature of the substrate, these coatings are mainly based on self‐assembled monolayers or silane assemblies, on polydopamine derivatives, or on polymer thin films deposited by vapor‐phase processes including plasma polymerization. The different works discussed here show that interfacial thermoreversible reactions occur between a DA‐functionalized surface and a DA reactant in solution but also between two solid substrates are possible. The direct cycloaddition is always described in the cited papers but the reversibility of the reaction is less discussed. The latter however remains very challenging for smart applications in material science. This review reports on the fabrication of functional surfaces with thermoreversible properties and the understanding of their reactivity via the Diels–Alder reaction. Based on various functionalization processes, the designed surfaces have various physico‐chemical properties, leading to different Diels–Alder reactivity. The direct cycloaddition is always described in the cited papers but the reversibility of the reaction is less discussed.

Abstract:In recent years, remarkable progress has been made in dehydro or dehydrogenative Diels-Alder (D-A) reactions. This Minireview gives an overview of the major two strategies for dehydro(genative) Diels-Alder reactions, which differ in dehydrogenation and D-A cyclization sequence. Reactions in which D-A cycloaddition is followed by dehydrogenation are useful methods for the synthesis of various aromatic compounds, whereas advancements in dehydro genative procedures with oxidants or catalysts prior to D-A cycloaddition offer yet further new routes to functionalized cycloadducts. Recent leading findings are highlighted and the current state of the art, scope, and limitations of these processes are discussed in this Minireview.

Abstract:The Diels-Alder (D-A) reaction is one of the most critical chemical transformations to construct C–C bonds with predictable regio- and stereo-selectivities. It has been widely used as a retrosynthetic disconnection in synthetic chemistry. Although significant advances have been made, synthetic challenges remain in how to precisely control the stereochemistry of intermolecular Diels-Alder reaction. Enzymes are well known for their remarkable catalytic efficiency and selectivity compared with chemo-catalysts. Therefore, identifying and designing intermolecular Diels-Alderases that can be used in organic synthesis have received considerable attention from the synthetic community. In this review, we review all the enzymes capable of catalyzing formal intermolecular Diels-Alder reactions in natural product biosynthetic pathways, discuss their catalytic mechanisms in detail, and highlight their synthetic potential in the precise and efficient synthesis of enantiopure D-A products. We also discuss the different strategies that can be used to create new artificial Diels-Alderases, especially RNA-based Diels-Alderases.
Tandem Diels-Alder reaction overrules entropy: the gate to a thermally stable, yet thermally recyclable furan-based polymers

Abstract:The study investigates the kinetics and thermodynamics of the reversible tandem Diels-Alder (tDA) reaction between difuranic compounds and maleimides, leading to the quantitative formation of tDA adducts at rates comparable to their counterparts from the “classical” Diels-Alder (DA) reaction. The tDA adducts exhibited unprecedented thermal stability up to 250 °C, which is 100 °C higher than that of the DA adducts, owing to the higher activation energy (Ea) required for the initial intramolecular step of the reverse process. The stability of the tDA adducts was exploited in the AA + BB type polymerization of tetrafuranic monomers with bis(maleimides), yielding thermally stable (up to 200 °C) yet depolymerasable linear polymers with molecular weights of 10-20 kDa. Only furanic groups were identified as the end-groups of the resulting polymers, suggesting the possibility for post-polymerization and end-group modifications. NMR and GPC kinetic data offered insights into the intermediate formation of classical adducts during polymerization, as well as the stereochemistry of tDA adducts in the polymer chains. Combined thermal analysis (DSC, TGA, and TMA) provided a comprehensive understanding of the reverse DA reaction in the resulting materials. This relatively clean, catalyst- and byproduct-free, well-controlled process, which uses derivatives of biorenewables as monomers, heralds the formation of a new class of thermally recyclable polymers.

Abstract:Diels-Alder reaction is a kind of reaction with high selectivity, high productivity, high reliability and condition tolerability. Ever since its discovery, it has been widely used in the organic synthetic chemistry and become a click chemistry. Recently, the Diels-Alder reaction is also gradually becoming an important linking method in the preparation for polymers with well-defined architectures, due to its broad practicability and orthogonality. Based on the classification of polymer architecture, this paper reviews the applications of Diels-Alder reaction. in preparing these polymers. Moreover, the paper gives the outlook for further prospect of Diels-Alder reaction in the polymer science. The relative key questions include methodology developments, application in the preparation of polymers with more specialized architectures, and broadening the functional usages.

Abstract:Abstract: The [4+2] Diels-Alder cycloaddition has been widely used for the synthesis of six-mem-ber scaffolds. In recent years, there have been significant developments in this area, including the discovery and design of novel dienes and dienophiles with improved reactivity and selectivity. These new building blocks can be used to develop diverse molecular structures with functional group compatibility. Additionally, there is the use of catalytic systems and metal-mediated reactions to enable asymmetric [4+2] cycloadditions, resulting in enantiomerically enriched products. Over-all, recent studies related to [4+2] Diels-Alder cycloaddition using numerous dienes, dienophiles, and catalysts in different reaction conditions have significantly improved the efficiency, selectivity, and versatility of the reaction, making it an increasingly important tool in the synthesis of complex organic molecules as presented in this review. These advancements offer exciting possibilities for the development of new methods and reagents for the construction of six-membered rings and the synthesis of bioactive compounds.

Abstract:The class of Double Diels-Alder (DDA) reactions leading the targets concisely where numerous synthetic pursuits have been rewarded with conceptually novel and streamlined method. After the tremendous development and discoveries in various Diels-Alder reactions including its key role applications in many natural products syntheses, DDA reaction is considered as a growing branch of Diels-Alder reaction where two consecutive [4+2] cycloaddition trigger the streamlined construction of polycyclic and macrocyclic architecture. In light of the ever-increasing importance of DDA reactions in chemical sciences, we review the comprehensive study and recent advances of DDA reactions in organic synthesis. This review summarizes key achievements of DDA in preparation of macrocyclic, bicyclic, heterocyclic, and polycyclic structures documented since its revelation in 1980 to 2020. Emphasis is stand on the synthesis of various structural class based on the selectivity of DDA reaction.

Abstract:Dynamic covalent polymer networks based on the Diels-Alder (DA) cycloaddition between furan and maleimide represent powerful and highly investigated tools in the design of self-healing coatings. In this minireview the most recent demonstrations of repairable coatings based on such platform chemistry are comprehensively discussed. In particular, the trends in the synthesis of DA networks, the enhancement of properties and advanced coating applications over the last ten years are highlighted. Furthermore, the current challenges in terms of the sustainability of DA coatings, performance assessment and the technological feasibility of healing procedures are outlined in the context of a future large-scale implementation.

Abstract:The development of self-healing materials has received much research attention in the last two decades. This review paper gathers recent publications on the self-healing polymeric materials with thermally reversible Diels-Alder (DA) chemistry. The DA reaction is a [4 + 2] cycloaddition involving a diene and a dienophile. The self-healing polymers employing the furan group as a diene and the maleimide group as a dienophile have been widely studied. Multifunctional furan and maleimide compounds construct thermally reversible crosslinked networks showing removability and remendability. Self-healing materials have also been utilized as healing agents for conventional thermosets like epoxy resins. Other diene-dienophile pairs, such as anthracene-maleimide and cyclopentadiene-dicyclopentadiene, have also been utilized for the development of thermally induced self-healing materials. Photo-induced self-healing polymers and some novel applications based on DA reactions have been discussed in this review. Moreover, self-healing polymer systems based on other thermally triggered and assisted reactions are also discussed. The discussed publication has provided promising molecular designs and synthetic strategies for the development of high performance self-healing polymers.

Abstract:The Diels-Alder reaction stands as one of the most pivotal transformations in organic chemistry. Its efficiency, marked by the formation of two carbon-carbon bonds and up to four new stereocenters in a single step, underscores its versatility and indispensability in synthesizing natural products and pharmaceuticals. The most significant stereoselectivity feature is the "endo rule". While this rule underpins the predictability of the stereochemical outcomes, it also underscores the challenges in achieving the opposite diastereoselectivity, making the exo-Diels-Alder reactions often considered outliers. This review delves into recent examples of exo-Diels-Alder reactions, shedding light on the factors inverting the intrinsic tendency. We explore the roles of steric, electrostatic, and orbital interactions, as well as thermodynamic equilibriums in influencing exo/endo selectivity. Furthermore, we illustrate strategies to manipulate these factors, employing approaches such as bulky substituents, s-cis conformations, transient structural constraints, and innovative control physics. Through these analyses, our aim is to provide a comprehensive understanding of how to predict and design exo-Diels-Alder reactions, paving the way for new diastereoselective catalyst systems and expanding the chemical scope of Diels-Alder reactions.

Abstract:The Diels-Alder (DA) reaction refers to a [4 + 2] cycloaddition reaction that falls under the category of pericyclic reactions. It is a reaction that allows regio- and stereo-selective construction of two carbon-carbon bonds simultaneously in a concerted manner to generate a six-membered ring structure through a six-electron cyclic transition state. The DA reaction is one of the most widely applied reactions in organic synthesis, yet its role in biological systems has been debated intensely over the last four decades. A survey of secondary metabolites produced by microorganisms suggests strongly that many of the compounds possess features that are likely formed through DA reactions, and most of them are considered to be catalyzed by enzymes that are commonly referred to as Diels-Alderases (DAases). In recent years, especially over the past 10 years or so, we have seen an accumulation of a substantial body of work that substantiates the argument that DAases indeed exist and play a critical role in the biosynthesis of complex metabolites. This review will cover the DAases involved in the biosynthesis of decalin moieties, which are found in many of the medicinally important natural products, especially those produced by fungi. In particular, we will focus on a subset of secondary metabolites referred to as pyrrolidine-2-one-bearing decalin compounds and discuss the decalin ring stereochemistry and the biological activities of those compounds. We will also look into the genes and enzymes that drive the biosynthetic construction of those complex natural products, and highlight the recent progress made on the structural and mechanistic understanding of DAases, especially regarding how those enzymes exert stereochemical control over the [4 + 2] cycloaddition reactions they catalyze.

Abstract:In this review, potential applications of the Diels–Alder reaction are explored within the nexus of biomaterials and drug delivery. This includes an emphasis on key platforms such as polymers, nanoparticles, and hydrogels which utilize Diels–Alder for drug delivery, functionalized surfaces, bioconjugation, and other diverse applications in tissue engineering and cancer therapies. Recent studies, leveraging click chemistry reactions, have significantly advanced the fields of biomaterials and drug delivery. Of these click reactions, the Diels–Alder cycloaddition is exceptionally valuable for synthetic organic chemistry and biomaterial design, as it occurs under mild reaction conditions and can undergo a retrograde reaction, under physiologically relevant conditions, to yield the initial reactants. Experimental And Theoretical Studies On The Hydrogen-Bond-Promoted Enantioselective Hetero-Diels-Alder Reaction Of Danishefsky'S Diene With Benzaldehyde

Abstract:The enantioselective hetero-Diels-Alder (HDA) reaction of Danishefsky's diene with benzaldehyde has been achieved catalytically by a series of alpha,alpha,alpha',alpha'-tetraaryl-1,3-dioxolane-4,5-dimethanol (TADDOL) derivatives through hydrogen-bonding activation, affording 2-phenyl-2,3-dihydro-4H-pyran-4-one after trifluoroacetic acid workup in moderate yield and good enantioselectivity. The alpha,alpha'-aryl substituents in the TADDOL molecules are found to exert a significant impact on both the activity and the enantioselectivity of the catalysis. In combination with the experimental investigations, the mechanism of the present reaction has also been studied theoretically using the ONIOM (133LYP/6-31G*:PM3) method with trans-1,3-dimethoxy-1,3-butadiene as the model for Danishefsky's diene. In agreement with the experimental findings, the calculation results indicate that this TADDOL-catalyzed HDA reaction proceeds via a concerted mechanism through an asynchronous and zwitterionic transition structure (TS). The carbonyl group of benzaldehyde is activated by forming an intermolecular hydrogen bond with one of the hydroxy groups of TADDOL. Meanwhile, the intramolecular hydrogen bond between the two hydroxy groups in TADDOL is found to facilitate the intermolecular hydrogen bonding with benzaldehyde. The sense of asymmetric induction is well-rationalized by the analysis of the relative energies of TADDOL-complexed TSs, while the different stereocontrol capabilities exhibited by TADDOLs in the reaction can be qualitatively established on the basis of the structural features of their corresponding TSs.
Accurate Prediction of Rate Constants of Diels-Alder Reactions and Application to Design of Diels-Alder Ligation.

Abstract:Bioorthogonal reactions are useful tools to gain insights into the structure, dynamics, and function of biomolecules in the field of chemical biology. Recently, the Diels-Alder reaction has become a promising and attractive procedure for ligation in bioorthogonal chemistry because of its higher rate and selectivity in water. However, a drawback of the previous Diels-Alder ligation is that the widely used maleimide moiety as a typical Michael acceptor can readily undergo Michael addition with nucleophiles in living systems. Thus, it is important to develop a nucleophile-tolerant Diels-Alder system in order to extend the scope of the application of Diels-Alder ligation. To solve this problem, we found that the theoretical protocol M06-2X/6-31+G(d)//B3LYP/6-31G(d) can accurately predict the activation free energies of Diels-Alder reactions with a precision of 1.4 kcal mol(-1) by benchmarking the calculations against the 72 available experimental data. Subsequently, the electronic effect and ring-strain effect on the Diels-Alder reaction were studied to guide the design of the new dienophiles. The criteria of the design is that the designed Diels-Alder reaction should have a lower barrier than the Michael addition, while at the same time it should show a similar (or even higher) reactivity as compared to the maleimide-involving Diels-Alder ligation. Among the designed dienophiles, three substituted cyclopropenes (i.e. 1,2-bis(trifluoromethyl)-, 1,2-bis(hydroxylmethyl)- and 1,2-bis(hydroxylmethyl)-3-carboxylcyclopropenes) meet our requirements. These substituted cyclopropene analogs could be synthesized and they are thermodynamically stable. As a result, we propose that 1,2-bis(trifluoromethyl)-, 1,2-bis(hydroxylmethyl)- and 1,2-bis(hydroxylmethyl)-3-carboxylcyclopropenes may be potential candidates for efficient and selective Diels-Alder ligation in living systems.

Abstract:The advent of the click chemistry paradigm has profoundly transformed the design of functional polymeric materials. Among the various reactions in the click chemistry toolkit, the inverse electron demand Diels–Alder (IEDDA) reaction is particularly notable for its rapid kinetics, high specificity, and bioorthogonality. Of the different electron-deficient dienes, the tetrazine (Tz) building block has become the most extensively employed component in the IEDDA reaction-based synthesis and functionalization of polymeric materials. This review begins with an overview of the IEDDA reaction involving Tz, followed by a discussion of examples where the Tz moiety has been utilized as a reactive handle for polymer synthesis and functionalization. We then focus on the application of Tz-based IEDDA reactions in the development of functional hydrogels and surface modifications. The selected examples highlight the diversity and versatility of this highly efficient chemistry. This review aims not only to survey recent progress but also to demonstrate the significant potential of IEDDA chemistry in advancing the design of functional polymeric materials.

Organocatalytic Multicomponent Alpha-Methylenation/Diels-Alder Reactions: A Versatile Route to Substituted Cyclohexenecarbaldehyde Derivatives

Abstract:This article describes the design and optimization of an effective organocatalytic three-component domino alpha-methylenation/Diels-Alder reaction to produce vinyl-substituted cyclohexenecarboxaldehydes in a highly regioselective fashion. In these one-pot transformations, 2-formyl-1,3-butadienes (4) were prepared in situ from alpha,beta-unsaturated aldehydes and formalin and were subsequently trapped with a variety of buta-1,3-dienes. The outcomes of the reactions were dependent on the electronic properties of the dienes. 1-Vinylcyclohexenecarbaldehydes 6 were formed by use of acyclic electron-rich dienes, while the initially formed cycloadducts of 4 with cyclopentadiene underwent Cope rearrangements, leading to the formation of tetrahydro-3H-indene-5-carbaldehyde compounds 7. The mechanisms involved in these reactions were deduced from experimental findings. Furthermore, the method was also extended to one-pot domino methylenation/Diels-Alder reactions of dihydrofurans and dihydropyrans to yield spirocyclic lactols 22. In these reactions, the unstable intermediate hydroxyethyl and hydroxypropyl acroleins behaved as dienophiles, undergoing cycloaddition reactions with dienes with good yields and selectivities. The wide variety of functionalized 1-vinylcyclohex-3-enecarbaldehydes 6, 4-vinylcyclohex-1-enecarbaldehydes 7, and spiro lactols 22 generated through the use of these organocatalytic domino processes as a diversity-oriented synthesis provided useful intermediates for the construction of novel odorants.
Highly Regio-, Enantio-, and Exo-selective Diels–Alder Reactions Enabled by a Bispyrrolidine Diboronate

Abstract:Catalytic asymmetric Diels−Alder reaction is one of the most powerful reactions in organic chemistry. It is still a challenge to achieve high and general exo-selectivity. A novel bispyrrolidine diboronate compound has been derived from the reaction of two molecules of oxazaborolidine with one molecule of water. Upon activation, it effectively catalyzes (0.4-5 mol % loading) the Diels–Alder reaction between a variety of mono- and di-carbonyl activated dienophiles and dienes with better than 20:1 regioselectivity, up to >99:1 enantioselectivity, and better than 20:1 exo/endo selectivity. Mechanistic studies show a remarkable nonlinearity in enantioselectivity, and a second-order kinetics with respect to the catalyst concentration. A model is proposed to rationalize the above observations. The reaction is expected to find wide applications in organic synthesis.
Efficient synthesis of fully renewable, furfural-derived building blocks via formal Diels-Alder cycloaddition of atypical addends

Abstract:Diels-Alder (DA) cycloaddition of furanics is emerging as a key transformation in circular chemistry, providing access to highly versatile, biobased platform molecules. Further development of this technology into viable industrial applications faces major challenges, a notorious one being the lack of reactivity of the most readily available furans, i.e. the furfural derivatives. Herein we describe the remarkably-facile intramolecular DA reaction of allyl acetals of different furfurals to efficiently afford formal DA adducts with the atypical, unreactive dienophile allyl alcohol. Our methodology gives access to unprecedented oxanorbornene derivatives in high chemo-, regio- and stereoselectivity, which can be readily diversified into valuable products. This offers the potential of scalable production of renewable chemical building blocks from cheap, bioderived platform molecules.

Dft Study Of The Mechanism And Stereochemistry Of The Rh(I)Catalyzed Diels-Alder Reactions Between Electronically Neutral Dienes And Dienophiles

Abstract:DielsAlder reaction between electronically neutral dienes and dienophiles is usually sluggish under thermal conditions and has to be catalyzed by transition metal catalysts. We report here our DFT study of the mechanism and stereochemistry of the Rh-catalyzed DielsAlder reaction between electronically neutral dienes and dienophiles (alkenes and alkynes), finding that this reaction includes a reaction sequence of oxidative cyclization between diene and alkene/alkyne and a reductive elimination step. The alkynes oxidative cyclization is much faster than alkenes due to the additional coordination of alkyne to the Rh center in the oxidative cyclization transition state. For both intermolecular and intramolecular reactions, the reductive elimination step in the catalytic cycle is rate-determining. The different reactivity of ene-diene and yne-diene substrates can be rationalized by the model that reductive elimination to form a C(sp(2))C(sp(3)) bond is easier than that for the formation of a C(sp(3))C(sp(3)) bond, due to the additional coordination of the double bond to the Rh center in the transition state in the former. We also uncovered the reasons for the high para-selectivity of the intermolecular DielsAlder reaction of dienes and alkynes. In addition, DFT calculations aiming to understand the high diastereoselectivity of an intramolecular [4 + 2] reaction of ene-dienes with substituents adjacent to the diene and ene moieties of the substrates found that the substituents in the substrates favor staying away from the Rh center in the oxidative cyclization transition states. This preference leads to the generation of the final [4 + 2] products with the substituents and the bridgehead hydrogen atoms in a cis-configuration.
