Abstract:Key messageHerein, the inoculation with strain wp-6 promoted the growth of wheat seedlings by improving the energy production and conversion of wheat seedlings and alleviating salt stress.Soil salinization decreases crop productivity due to high toxicity of sodium ions to plants. Plant growth-promoting rhizobacteria (PGPR) have been demonstrated to alleviate salinity stress. However, the mechanism of PGPR in improving plant salt tolerance remains unclear. In this study, physiological analysis, proteomics, and metabolomics were applied to investigate the changes in wheat seedlings under salt stress (150 mM NaCl), both with and without plant root inoculation with wp-6 (Bacillus sp). Under salt stress, root inoculation with strain wp-6 increased plant biomass (57%) and root length (25%). The Na+ content was reduced, while the K+ content and K+/Na+ ratio were increased. The content of malondialdehyde was decreased by 31.94% after inoculation of wp-6 under salt stress, while the content of proline, soluble sugar, and soluble protein were increased by 7.48%, 12.34%, and 4.12%, respectively. The peroxidase, catalase, and superoxide dismutase activities were increased after inoculation of wp-6 under salt stress. Galactose metabolism, phenylalanine metabolism, caffeine metabolism, ubiquinone and other terpenoid-quinone biosynthesis, and glutathione metabolism might play an important role in promoting the growth of salt-stressed wheat seedlings after the inoculation with wp-6. Interaction analysis of differentially expressed proteins and metabolites found that energy production and transformation-related proteins and six metabolites (d-arginine, palmitoleic acid, chlorophyllide b, rutin, pheophorbide a, and vanillylamine) were mainly involved in the growth of wheat seedlings after the inoculation with wp-6 under salt stress. Furthermore, correlation analysis found that inoculation with wp-6 promotes the growth of salt-stressed wheat seedlings mainly through regulating amino acid metabolism and porphyrin and chlorophyll metabolism. This study provides an eco-friendly method to increase agricultural productivity and paves a way to sustainable agriculture.

Salt Tolerant Bacillus Strains Improve Plant Growth Traits and Regulation of Phytohormones in Wheat under Salinity Stress

Abstract:Soil salinity is a major constraint adversely affecting agricultural crops including wheat worldwide. The use of plant growth promoting rhizobacteria (PGPR) to alleviate salt stress in crops has attracted the focus of many researchers due to its safe and eco-friendly nature. The current study aimed to study the genetic potential of high halophilic Bacillus strains, isolated from the rhizosphere in the extreme environment of the Qinghai–Tibetan plateau region of China, to reduce salt stress in wheat plants. The genetic analysis of high halophilic strains, NMCN1, LLCG23, and moderate halophilic stain, FZB42, revealed their key genetic features that play an important role in salt stress, osmotic regulation, signal transduction and membrane transport. Consequently, the expression of predicted salt stress-related genes were upregulated in the halophilic strains upon NaCl treatments 10, 16 and 18%, as compared with control. The halophilic strains also induced a stress response in wheat plants through the regulation of lipid peroxidation, abscisic acid and proline in a very efficient manner. Furthermore, NMCN1 and LLCG23 significantly enhanced wheat growth parameters in terms of physiological traits, i.e., fresh weight 31.2% and 29.7%, dry weight 28.6% and 27.3%, shoot length 34.2% and 31.3% and root length 32.4% and 30.2%, respectively, as compared to control plants under high NaCl concentration (200 mmol). The Bacillus strains NMCN1 and LLCG23 efficiently modulated phytohormones, leading to the substantial enhancement of plant tolerance towards salt stress. Therefore, we concluded that NMCN1 and LLCG23 contain a plethora of genetic features enabling them to combat with salt stress, which could be widely used in different bio-formulations to obtain high crop production in saline conditions.

Abstract:Bacteria residing in the rhizosphere and capable of host plant growth stimulation (PGPR) through ACC-deaminase activity can ameliorate various biotic- and abiotic-stress conditions by decreasing the level of ‘stress ethylene’ in plants. The present study aimed to evaluate the effect of a PGPR strain Enterobacter cloacae ZNP-3 in augmenting wheat plant growth under salinity- and temperature-stress conditions. Among the plant growth-promoting (PGP) traits, the test organism produced ACC deaminase along with several other properties namely indole-3-acetic acid (IAA) production, mineral phosphate solubilization, hydrogen cyanide (HCN) and ammonia production. The test isolate illustrated its ability to function as a biocontrol agent as it displayed antagonistic activity against a wide range of bacterial and fungal pathogens. Its inoculation to wheat plant resulted in a considerable increase in growth parameters, biomass, and chlorophyll content under salinity stress. The inoculation also decreased the accumulation of Na+ and increased K+ uptake in shoots and roots, leading to maintenance of favorable K+/Na+ ratios in bacterial-treated plants for alleviating the toxic effect of salt stress. Moreover, application of ZNP-3 also stimulated the growth of wheat plants in terms of higher biomass and other growth parameters under temperature-stress conditions. Elevated levels of various compatible solutes following inoculation significantly alleviated the negative effect of salinity stress. The inoculation of isolate ZNP-3 also minimized salinity-induced oxidative damage by lowering H2O2 and O2− contents. Moreover, the colonization behavior of the isolate illustrates its endophytic nature. The observed results indicate that the isolate Enterobacter cloacae ZNP-3 mitigates the detrimental effects of salinity and temperature stressors, and can be used as a promising bioinoculant to decrease the deleterious effects caused by salinity and other stressors.

Rhizobacteria Isolated from Saline Soil Induce Systemic Tolerance in Wheat (Triticum aestivum L.) against Salinity Stress

Abstract:Halo-tolerant plant growth-promoting rhizobacteria (PGPR) have the inherent potential to cope up with salinity. Thus, they can be used as an effective strategy in enhancing the productivity of saline agro-systems. In this study, a total of 50 isolates were screened from the rhizospheric soil of plants growing in the salt range of Pakistan. Out of these, four isolates were selected based on their salinity tolerance and plant growth promotion characters. These isolates (SR1. SR2, SR3, and SR4) were identified as Bacillus sp. (KF719179), Azospirillum brasilense (KJ194586), Azospirillum lipoferum (KJ434039), and Pseudomonas stutzeri (KJ685889) by 16S rDNA gene sequence analysis. In vitro, these strains, in alone and in a consortium, showed better production of compatible solute and phytohormones, including indole acetic acid (IAA), gibberellic acid (GA), cytokinin (CK), and abscisic acid (ABA), in culture conditions under salt stress. When tested for inoculation, the consortium of all four strains showed the best results in terms of improved plant biomass and relative water content. Consortium-inoculated wheat plants showed tolerance by reduced electrolyte leakage and increased production of chlorophyll a, b, and total chlorophyll, and osmolytes, including soluble sugar, proline, amino acids, and antioxidant enzymes (superoxide dismutase, catalase, peroxidase), upon exposure to salinity stress (150 mM NaCl). In conclusion, plant growth-promoting bacteria, isolated from salt-affected regions, have strong potential to mitigate the deleterious effects of salt stress in wheat crop, when inoculated. Therefore, this consortium can be used as potent inoculants for wheat crop under prevailing stress conditions.
Dominance of Bacillus species in the wheat (Triticum aestivum L.) rhizosphere and their plant growth promoting potential under salt stress conditions

Abstract:Wheat (Triticum aestivum L.) is a major source of calorific intake in its various forms and is considered one of the most important staple foods. Improved wheat productivity can contribute substantially to addressing food security in the coming decades. Soil salinity is the most serious limiting factor in crop production and fertilizer use efficiency. In this study, 11 bacteria were isolated from wheat rhizosphere and examined for salt tolerance ability. WGT1, WGT2, WGT3, WGT6, WGT8, and WGT11 were able to tolerate NaCl salinity up to 4%. Bacterial isolates were characterized in vitro for plant growth-promoting properties including indole-3-acetic acid (IAA) production, phosphate solubilization, nitrogen fixation, zinc solubilization, biofilm formation, and cellulase-pectinase production. Six isolates, WGT1, WGT3, WGT4, WGT6, WGT8, and WGT9 showed IAA production ability ranging from 0.7-6 µg m/L. WGT8 displayed the highest IAA production. Five isolates, WGT1, WGT2, WGT5, WGT10, and WGT11, demonstrated phosphate solubilization ranging from 1.4-12.3 µg m/L. WGT2 showed the highest phosphate solubilization. Nitrogen fixation was shown by only two isolates, WGT1 and WGT8. Zinc solubilization was shown by WGT1 and WGT11 on minimal media. All isolates showed biofilm formation ability, where WGT4 exhibited maximum potential. Cellulase production ability was noticed in WGT1, WGT2, WGT4, and WGT5, while pectinase production was observed in WGT2 and WGT3. Phylogenetic identification of potential bacteria isolates confirmed their close relationship with various species of the genus Bacillus. WGT1, WGT2, and WGT3 showed the highest similarity with B. cereus, WGT6 with B. tianshenii, WGT8 with B. subtilis, and WGT11 with B. thuringiensis. Biofertilizer characteristics of salt-tolerant potential rhizospheric bacteria were evaluated by inoculating wheat plants under controlled conditions and field experiments. B. cereus WGT1 and B. thuringiensis WGT11 displayed the maximum potential to increase plant growth parameters and enhance grain yield by 37% and 31%, respectively. Potential bacteria of this study can tolerate salt stress, have the ability to produce plant growth promoting substances under salt stress and contribute significantly to enhance wheat grain yield. These bacterial isolates have the potential to be used as biofertilizers for improved wheat production under salinity conditions and contribute to the sustainable agriculture.
Enhancing Wheat Yield and Zinc Biofortification through Synergistic Action of Potent Zinc-Solubilizing Bacteria and Zinc Sulfate in Calcareous Soil

Abstract:Zinc (Zn) is deficient all over the world, mainly in the developing countries, and zinc sulfate is applied as a source of Zn fertilizer. However, the major portions of applied zinc sulfate become unavailable to plant in the soil and can be reverted back to available by inoculating zinc-solubilizing bacteria. Given the significance of the problem, this investigation aimed to enhance wheat yield and zinc biofortification. The approach involved inoculating the crop with native zinc-solubilizing bacteria and applying zinc sulfate to calcareous soil. In this study, a pot and field experiments were conducted to evaluate the impact of treatment combinations (control (without Zn and bacterial inoculation), 4, 8, 12, 16 and 20 kg Zn ha −1 were applied to soil without and with inoculation of zinc-solubilizing bacteria to seed of wheat cultivar, i.e., Wadaan-17 and Zincol-16). Results showed that zinc-solubilizing bacteria in conjunction with zinc sulfate significantly ( P ≤ 0.05) increased the yield by 61%, grain zinc concentration by 139%, and apparent Zn recovery efficiency varied from 0.5 to 4.8% as compared to control under pot conditions. Similarly, under field conditions, the observations showed a 38% increase in yield, a 112% increase in grain zinc concentration and a 166% increase in zinc uptake. Additionally, the apparent zinc recovery efficiency ranged from 0.6% to 3.6% compared to the control. Among the treatment combinations, inoculation of Zn-solubilizing bacteria in conjunction with 8 kg Zn ha −1 substantially boosted yield and yield attributes of wheat crop under both the pot and field conditions. Hence, it can be concluded that Zn-solubilizing bacteria have immense potential to be used as inoculants as they synergize the effect of chemical Zn, increase yield and improve the nutritional value of wheat under alkaline calcareous soil.
Seed Priming by Sodium Nitroprusside Improves Salt Tolerance in Wheat (triticum Aestivum L.) by Enhancing Physiological and Biochemical Parameters.

Abstract:The germination, seedling vigor, crop establishment and yield of agronomically important crops is negatively affected by soil salinity. The current study aimed to investigate the ability of exogenous fertigation by sodium nitroprusside (SNP) to induce salt tolerance in four high yielding wheat cultivars (Sahar-06, Punjab-11, Millat-11 and Galaxy-13) that differ in their response to salt stress in terms of biomass production, oxidative defense mechanisms and grain yield. Three levels of SNP (0, 0.1 and 0.2 mM) were used for seed soaking. During soaking the seeds were kept in the dark. After soaking for 12 h the seeds were air-dried for 5 h before sowing. Salinity caused a significant reduction in biomass and grain yield, while it increased proline (Pro), ascorbic acid (AsA), hydrogen peroxide (H2O2) and malondialdehyde (MDA) contents. Cultivar Sahar-06 and Galaxy-13 were found more tolerant to salinity based on shoot length root fresh and dry wights, 100 grain weight, decreased MDA and H2O2 accumulation, phenolic and ascorbic acid (AsA) contents, accumulation of proline, activities of SOD, POD and CAT as compared to the other cultivars. Seed priming with SNP was effective in reducing the adverse effects of salt stress induced oxidative stress on plant biomass and grain yield in all the studied wheat cultivars, but maximum amelioration of salt stress tolerance by SNP treatment was found in cv. Sahar-06. The increased salt tolerance in wheat plants by SNP seed priming might be due to the role of NO in improving seed vigor and germination and early establishment of seedlings with better growth. 0.1 mM SNP was found the most effective in improving salt tolerance, as compared to other SNP concentations. Exogenous SNP fertigation increased the activities of antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) and the contents of AsA, Pro and total phenolics content (TPC) in the salt stressed wheat plants. Our data indicate that SNP-priming induced salt tolerance by up-regulating the antioxidative defense mechanisms resulting in better biomass production and grain yield.
Halotolerant Plant Growth-Promoting Rhizobacteria Induce Salinity Tolerance in Wheat by Enhancing the Expression of SOS Genes

Abstract:Soil salinity is one of the main yield-limiting factors in various crops. Under different environmental stresses, many rhizobacteria have demonstrated encouraging role in enhancing plant growth and tolerate stress conditions. In this study, three potential 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase- and exopolysaccharides (EPS)-secreting bacterial strains including Bacillus megaterium, B. tequilensis, and Pseudomonas putida have been assessed for their growth-promoting characteristics. These bacterial strains positively affected the physiology, biochemistry, and antioxidant enzymatic activities of wheat plant, under salinity stress. Results of this study depicted that the inoculation of PGPR positively invigorates growth attributes like relative water content and photosynthetic pigments of wheat seedling under saline conditions. Moreover, plants inoculated with PGPR also showed decreased concentration of malondialdehyde (MDA) and hydrogen peroxide (H2O2). Inoculation of PGPR reduced electrolytic leakage and enhanced enzymatic activity for the scavenging of reactive oxygen species (ROS). These PGPR also increased the production of proline and total soluble sugar. Expression analysis of selected genes by qPCR revealed higher expression of Salt Overly Sensitive (SOS1 and SOS4) genes and predicted their potential role in stress tolerance. These genes can be further overexpressed in wheat plant to tolerate salinity stress. On the basis of these findings, it can be concluded that the priming of seeds with aforementioned PGPR can decrease the adverse effects of salinity on wheat plant.

Abstract:Soil salinity is one of the most important abiotic stresses limiting plant growth and productivity. The breeding of salt-tolerant wheat cultivars has substantially relieved the adverse effects of salt stress. Complementing these cultivars with growth-promoting microbes has the potential to stimulate and further enhance their salt tolerance. In this study, two fungal isolates, Th4 and Th6, and one bacterial isolate, C7, were isolated. The phylogenetic analyses suggested that these isolates were closely related to Trichoderma yunnanense, Trichoderma afroharzianum, and Bacillus licheniformis, respectively. These isolates produced indole-3-acetic acid (IAA) under salt stress (200 mM). The abilities of these isolates to enhance salt tolerance were investigated by seed coatings on salt-sensitive and salt-tolerant wheat cultivars. Salt stress (S), cultivar (C), and microbial treatment (M) significantly affected water use efficiency. The interaction effect of M x S significantly correlated with all photosynthetic parameters investigated. Treatments with Trichoderma isolates enhanced net photosynthesis, water use efficiency and biomass production. Principal component analysis revealed that the influences of microbial isolates on the photosynthetic parameters of the different wheat cultivars differed substantially. This study illustrated that Trichoderma isolates enhance the growth of wheat under salt stress and demonstrated the potential of using these isolates as plant biostimulants.

Transcriptome-based analysis of the effects of compound microbial agents on gene expression in wheat roots and leaves under salt stress

Abstract:Introduction Salt stress inhibits the beneficial effects of most plant growth-promoting rhizobacteria. The synergistic relationship between beneficial rhizosphere microorganisms and plants helps achieve more stable growth-promoting effects. This study aimed 1) to elucidate changes in gene expression profiles in the roots and leaves of wheat after inoculation with compound microbial agents and 2) to determine the mechanisms by which plant growth-promoting rhizobacteria mediate plant responses to microorganisms. Methods Following inoculation with compound bacteria, transcriptome characteristics of gene expression profiles of wheat, roots, and leaves at the flowering stage were investigated using Illumina high-throughput sequencing technology. Gene ontology (GO) function and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses were performed on the genes that were significantly differentially expressed. Results The expression of 231 genes in the roots of bacterial preparations (BIO) -inoculated wheat changed significantly (including 35 upregulated and 196 downregulated genes) compared with that of non-inoculated wheat. The expression of 16,321 genes in leaves changed significantly, including 9651 upregulated genes and 6670 downregulated genes. The differentially expressed genes were involved in the metabolism of carbohydrates, amino acids, and secondary compounds as well as signal transduction pathways. The ethylene receptor 1 gene in wheat leaves was significantly downregulated, and genes related to ethylene-responsive transcription factor were significantly upregulated. GO enrichment analysis showed that metabolic and cellular processes were the main functions affected in the roots and leaves. The main molecular functions altered were binding and catalytic activities, among which the cellular oxidant detoxification enrichment rate was highly expressed in the roots. The expression of peroxisome size regulation was the highest in the leaves. KEGG enrichment analysis showed that linoleic acid metabolism expression was highest in the roots, and the expression of photosynthesis-antenna proteins was the highest in leaves. After inoculation with a complex biosynthesis agent, the phenylalanine ammonia lyase (PAL) gene of the phenylpropanoid biosynthesis pathway was upregulated in wheat leaf cells while 4CL, CCR, and CYP73A were downregulated. Additionally, CYP98A and REF1 genes involved in the flavonoid biosynthesis pathway were upregulated, while F5H, HCT, CCR, E2.1.1.104, and TOGT1-related genes were downregulated. Discussion Differentially expressed genes may play key roles in improving salt tolerance in wheat. Compound microbial inoculants promoted the growth of wheat under salt stress and improved disease resistance by regulating the expression of metabolism-related genes in wheat roots and leaves and activating immune pathway-related genes.

Abstract:We report the growth promoting potential in wheat under saline conditions by an endophytic fungus Aspergillus terreus BTK-1. The isolated BTK-1 from the root of Chenopodium album was identified as Aspergillus terreus through 18S rDNA sequence analysis. BTK-1 secreted indole acetic acid (IAA), exhibited 1- aminocyclopropane-1- carboxylate deaminase (ACC) and siderophores activity, and solubilized phosphate. Wheat seedlings were exposed to a saline environment (0, 60, 120, and 180 mM) with or without BKT-1 inoculation. Seedlings inoculated with BTK-1 showed higher concentrations of IAA and gibberellins, whereas they showed low concentrations of abscisic acid compared to the BTK-1 non-inoculated plants. Also, BTK-1 inoculated wheat plants revealed significantly ( P = 0.05) longer shoots and roots, biomass, and chlorophyll contents. On the contrary, plants without BTK-1 inoculation indicated significantly ( P = 0.05) low amounts of carbohydrates, phenolics, prolines, potassium, magnesium, and calcium, with high amounts of Na and malonaldehyde under salt stress. Likewise, BTK-1 inoculated wheat plants showed high activity of reduced glutathione, and low activity of ascorbate, catalase, and peroxidase under salt stress. The mitigation of salinity stress by BTK-1 inoculated wheat plants suggested its use as a bio-stimulator in salt affected soils.

Abstract:Salinity stress (SS) is a challenging abiotic stress that limits crop growth and productivity. Sustainable and cost effective methods are needed to improve crop production and decrease the deleterious impacts of SS. Zinc (Zn) nano-particles (NPs) have emerged as an important approach to regulating plant tolerance against SS. However, the mechanisms of SS tolerance mediated by Zn-NPs are not fully explained. Thus, this study was performed to explore the role of Zn-NPs (seed priming and foliar spray) in reducing the deleterious impacts of SS on wheat plants. The study comprised different SS levels: control, 6 and 12 dS m −1 , and different Zn-NPs treatments: control, seed priming (40 ppm), foliar spray (20 ppm), and their combination. Salinity stress markedly reduced plant growth, biomass, and grain yield. This was associated with enhanced electrolyte leakage (EL), malondialdehyde (MDA), hydrogen peroxide (H 2 O 2 ), sodium (Na), chloride (Cl) accumulation, reduced photosynthetic pigments, relative water contents (RWC), photosynthetic rate (Pn), transpiration rate (Tr), stomata conductance (Gs), water use efficiency (WUE), free amino acids (FAA), total soluble protein (TSP), indole acetic acid (IAA), gibberellic acid (GA), and nutrients (Ca, Mg, K, N, and P). However, the application of Zn-NPs significantly improved the yield of the wheat crop, which was associated with reduced abscisic acid (ABA), MDA, H 2 O 2 concentration, and EL, owing to improved antioxidant activities, and an increase in RWC, Pn, Tr, WUE, and the accumulation of osmoregulating compounds (proline, soluble sugars, TSP, and FAA) and hormones (GA and IAA). Furthermore, Zn-NPs contrasted the salinity-induced uptake of toxic ions (Na and Cl) and increased the uptake of Ca, K, Mg, N, and P. Additionally, Zn-NPs application substantially increased the wheat grain Zn bio-fortification. Our results support previous findings on the role of Zn-NPs in wheat growth, yield, and grain Zn bio-fortification, demonstrating that beneficial effects are obtained under normal as well as adverse conditions, thanks to improved physiological activity and the accumulation of useful compounds. This sets the premise for general use of Zn-NPs in wheat, to which aim more experimental evidence is intensively being sought. Further studies are needed at the genomic, transcriptomic, proteomic, and metabolomic level to better acknowledge the mechanisms of general physiological enhancement observed with Zn-NPs application.

Abstract:Using salt-tolerant bacteria to protect plants from salt stress is a promising microbiological treatment strategy for saline-alkali soil improvement. Here, we conducted research on the growthpromoting effect of Brevibacterium frigoritolerans on wheat under salt stress, which has rarely been addressed before. The synergistic effect of B. frigoritolerans combined with representative salttolerant bacteria Bacillus velezensis and Bacillus thuringiensis to promote the development of wheat under salt stress was also further studied. Our approach involved two steps: investigation of the plant growth-promoting traits of each strain at six salt stress levels (0, 2, 4, 6, 8, and 10%); examination of the effects of the strains (single or in combination) inoculated on wheat in different salt stress conditions (0, 50, 100, 200, 300, and 400 mM). The experiment of plant growth-promoting traits indicated that among three strains, B. frigoritolerans had the most potential for promoting wheat parameters. In single-strain inoculation, B. frigoritolerans showed the best performance of plant growth promotion. Moreover, a pot experiment proved that the plant growth-promoting potential of co-inoculation with three strains on wheat is better than single-strain inoculation under salt stress condition. Up to now, this is the first report suggesting that B. frigoritolerans has the potential to promote wheat growth under salt stress, especially combined with B. velezensis and B. thuringiensis.
Application of Plant-Growth-Promoting Fungi Trichoderma longibrachiatum T6 Enhances Tolerance of Wheat to Salt Stress through Improvement of Antioxidative Defense System and Gene Expression

Abstract:Soil salinity is a serious problem worldwide that reduces agricultural productivity. Trichoderma longibrachiatum T6 (T6) has been shown to promote wheat growth and induce plant resistance to parasitic nematodes, but whether the plant-growth-promoting fungi T6 can enhance plant tolerance to salt stress is unknown. Here, we determined the effect of plant-growth-promoting fungi T6 on wheat seedlings' growth and development under salt stress, and investigated the role of T6 in inducing the resistance to NaCl stress at physiological, biochemical, and molecular levels. Wheat seedlings were inoculated with the strain of T6 and then compared with non-inoculated controls. Shoot height, root length, and shoot and root weights were measured on 15 days old wheat seedlings grown either under 150 mM NaCl or in a controlled setting without any NaCl. A number of colonies were re-isolated from the roots of wheat seedlings under salt stress. The relative water content in the leaves and roots, chlorophyll content, and root activity were significantly increased, and the accumulation of proline content in leaves was markedly accelerated with the plant growth parameters, but the content of leaf malondialdehyde under saline condition was significantly decreased. The antioxidant enzymes-superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) in wheat seedlings were increased by 29, 39, and 19%, respectively, with the application of the strain of T6 under salt stress; the relative expression of SOD, POD, and CAT genes in these wheat seedlings were significantly up-regulated. Our results indicated that the strain of T6 ameliorated the adverse effects significantly, protecting the seedlings from salt stress during their growth period. The possible mechanisms by which T6 suppresses the negative effect of NaCl stress on wheat seedling growth may be due to the improvement of the antioxidative defense system and gene expression in the stressed wheat plants.

Abstract:Soil salinity is an important abiotic stress factor that seriously affects the crop growth and yield. Use of plant-derived microorganisms is a promising strategy to alleviate salt stress. In a previous study, the endophytic strain Bacillus altitudinis WR10 isolated from wheat roots showed high salt resistance. In this study, we investigated the efficacy of WR10 in improving the salt tolerance of wheat and its potential mechanisms using a hydroponic test. Under salt stress, WR10 inoculation significantly increased the lengths and dry weights of the roots and shoots, indicating that WR10 improves wheat salt tolerance at the seedling stage. WR10 inoculation significantly reduced Na+ accumulation and enhanced K+, P, and Ca2+ uptake in salt-stressed plants, which can be attributed to the upregulated gene expression of H+-ATPase as well as the P-solubilizing and biofilm-producing characteristics of WR10. At the transcriptional level, L-ascorbate peroxidase (APX), glutathione (GSH) synthetase related to GSH biosynthesis, and phenylpropanoid biosynthesis genes (CYP73A, 4CL, and CAD) were significantly upregulated, whereas those of GSH metabolism genes (glutathione S-transferase and gamma-glutamyltranspeptidase) were significantly downregulated in WR10-applied wheat roots under salt stress. These changes increased the APX activity and GSH levels and resulted in a decrease in hydrogen peroxide levels. Additionally, a decrease in proline content was observed in WR10-inoculated plants under salt stress because of WR10-induced upregulation of proline dehydrogenase gene expression. These results provide supporting evidence that WR10 improves wheat salt tolerance via more than one mechanism and open a window of opportunity for WR10 application in salinized soil.
Effects of ischemia on the hind limb of the rat.

Abstract:The effects of ischemia, induced by a tourniquet, were investigated on 36 adult Holtzman female rates in terms of damage to the ventral horn cell of the spinal cord and tibial nerve and motor end-plate degeneration and regeneration. Clinically, the rats were tested for sensory and motor loss and recovery and these results were compared with the histological findings. Ischemic periods of 2, 4, 6, and 8 hours were used followed by survival times of 3 weeks to 9 months. Histologically, there was degeneration and regeneration found to varying degrees in the nerve and motor end-plate. There were no changes found in the ventral horn cell. There was a loss of pain sensation in all animals, except the 2 hour group, and a loss of motor function. Motor function, preceded by sensation, returned in all animals. With the absence of ventral horn cell damage it was not surprising to find adequate regeneration histologically and, therefore, a return of both sensory and motor function.
Effects of Plant Growth Promoting Rhizobacteria on the Content of Abscisic Acid and Salt Resistance of Wheat Plants

Abstract:Although salinity inhibits plant growth, application of appropriate rhizosphere bacteria can diminish this negative effect. We studied one possible mechanism that may underlie this beneficial response. Wheat plants were inoculated with Bacillus subtilis IB-22 and Pseudomonas mandelii IB-Ki14 and their consequences for growth, water relations, and concentrations of the hormone abscisic acid (ABA) were followed in the presence of soil salinity. Salinity alone increased ABA concentration in wheat leaves and roots and this was associated with decreased stomatal conductance, but also with chlorophyll loss. Bacterial treatment raised ABA concentrations in roots, suppressed accumulation of leaf ABA, decreased chlorophyll loss, and promoted leaf area and transpiration. However, water balance was maintained due to increased water uptake by inoculated plants, brought about in part by a larger root system. The effect may be the outcome of ABA action since the hormone is known to maintain root extension in stressed plants. Root ABA concentration was highest in salt-stressed plants inoculated with B. subtilis and this contributed to greater root hydraulic conductivity. We conclude that bacteria can raise salt resistance in wheat by increasing root ABA, resulting in larger root systems that can also possess enhanced hydraulic conductivity thereby supporting better-hydrated leaves.

Abstract:Background Technosoils in Inowrocław, central Poland, are impacted by human activities and exhibit high salinity (ECe up to 70 dS/m) due to a soda lime repository. These saline environments pose challenges to plant growth and soil health. However, they also offer an opportunity for the evolution of microorganisms adapted to such conditions, including plant growth-promoting rhizospheric (PGPR) bacteria. The hypothesis tested here was that introducing PGPR bacteria from these environments could boost degraded soil performance, leading to better plant biomass and improved pathogen defense. Methods Azotobacter chroococcum W4ii was isolated from the rhizosphere of wheat ( Triticum aestivum L.) for its plant growth properties on wheat plants under salt stress. Results Wheat seeds co-inoculated with A. chroococcum W4ii under 200 mM salt stress showed significant improvement in various growth parameters such as seeds germination (by 130%), shoot biomass (15%), chlorophyll b content (40%) compared to un-inoculated ones. Bacterial inoculation decreased the level of malondialdehyde (MDA) by 55.5% (P<0.001), whereas it elevated the antioxidative enzymatic activities of peroxidase (POD) by 33.69% (P<0.001). The test isolate also significantly (P<0.05) enhanced the level of defense enzymes like β-1,3-glucanase, which can protect plants from infection by pathogens. The bacterium could also successfully colonize the wheat plants. Conclusions These results indicate that A. chroococcum isolated from the technosoil has the potential to promote wheat growth under salt stress and can be further used as a bioinoculant in the salt affected agricultural fields.
Plant Growth Stimulators Improve Two Wheat Cultivars Salt-Tolerance: Insights into Their Physiological and Nutritional Responses

Abstract:Spermine (SPM) and salicylic acid (SA), plant growth stimulators, are involved in various biological processes and responses to environmental cues in plants. However, the function of their combined treatment on wheat salt tolerance is unclear. In this study, wheat (Triticum aestivum L. cvs. Shandawel 1 and Sids 14) plants were grown under non-saline and saline (6.0 and 12.0 dS m–1) conditions and were foliar sprayed with 100 mgL−1 SA and/or 30 mgL−1 SPM. Exogenously applied SA and/or SPM relieved the adverse effects caused by salt stress and significantly improved wheat growth and production by inducing higher photosynthetic pigment (chlorophyll a, chlorophyll b, carotenoids) content, nutrient (N, P, K+, Ca2+, Mg2+, Fe, Zn, Cu) acquisition, ionic (K+/Na+, Ca2+/Na+, Mg2+/Na+) homeostatics, osmolyte (soluble sugars, free amino acids, proline, glycinebetaine) accumulation, grains, carbohydrate, and protein content, along with significantly lower Na+ accumulation and chlorophyll a/b ratio. The best response was registered with SA and SPM combined treatment, especially in Shandawel 1. This study highlighted the recovery impact of SA and SPM combined treatment on salinity-damaged wheat plants. The newly discovered data demonstrate that this treatment significantly improved the photosynthetic pigment content, mineral homeostasis, and osmoprotector solutes buildup in salinity-damaged wheat plants. Therefore, it can be a better strategy for ameliorating salt toxicity in sustainable agricultural systems.

Abstract:Combined application of (69,71)Ga NMR spectroscopy and quantum mechanical calculations reveals the chemical bonding in the digallides of Ca, Sr, and Ba. An analysis of the electron localization function (ELF) shows honeycomb-like 6(3) nets of the Ga atoms as the most prominent structural features in SrGa(2) and BaGa(2). For CaGa(2) a description of a 3+1-coordinated Ga atom is revealed by the ELF and by an analysis of interatomic distances. The NMR spectroscopic signal shift is mainly due to the Knight shift and is almost equal for the investigated digallides, whereas the anisotropy of the signal shift decreases with the radius of the alkaline-earth metals. Calculated and observed values of the electric field gradient (EFG) are in good agreement for CaGa(2) and BaGa(2) but differ by about 21 % for SrGa(2) indicating structural instability. Better agreement is achieved by considering a puckering of the Ga layers. For BaGa(2) an instability of the structure is indicated by a peak in the density of states at the Fermi level, which is shifted to lower energies when taking puckering of the Ga layers into account. Both structural modifications are confirmed by crystallographic information. The Fermi velocity of the electrons is strongly anisotropic and is largest in the (001) plane of the crystal structure. This results in an alignment of the crystallites with the [001] axis perpendicular to the magnetic field as observed in (69,71)Ga NMR spectroscopy and magnetic susceptibility experiments. The electron transport is predominantly mediated by the Ga-Ga p(x)- and p(y)-like electrons in the (001) plane. The specific heat capacity of BaGa(2) was determined and indicated the absence of phase transitions between 1.8 and 320 K.
