Abstract:OBJECTIVE:During several pathological processes such as cancer progression, thermal injury, wound healing and hindlimb ischemia, the mobilization of endothelial progenitor cells (EPCs) mobilization was enhanced with an increase of sympathetic nerve activity and norepinephrine (NE) secretion, yet the cellular and molecular mechanisms involved in the effects of NE on EPCs has less been investigated.METHODS AND RESULTS:EPCs from BMs, peripheral circulation and spleens, the VEGF concentration in BM, skeletal muscle, peripheral circulation and spleen and angiogenesis in ischemic gastrocnemius were quantified in mice with hindlimbs ischemia. Systemic treatment of NE significantly increased EPCs number in BM, peripheral circulation and spleen, VEGF concentration in BM and skeletal muscle and angiogenesis in ischemic gastrocnemius in mice with hind limb ischemia, but did not affair VEGF concentration in peripheral circulation and spleen. EPCs isolated from healthy adults were cultured with NE in vitro to evaluate proliferation potential, migration capacity and phosphorylations of Akt and eNOS signal moleculars. Treatment of NE induced a significant increase in number of EPCs in the S-phase in a dose-dependent manner, as well as migrative activity of EPCs in vitro (p<0.05). The co-treatment of Phentolamine, I127, LY294002 and L-NAME with NE blocked the effects of NE on EPCs proliferation and migration. Treatment with NE significantly increased phosphorylation of Akt and eNOS of EPCs. Addition of phentolamine and I127 attenuated the activation of Akt/eNOS pathway, but metoprolol could not. Pretreatment of mice with either Phentolamine or I127 significantly attenuated the effects of NE on EPCs in vivo, VEGF concentration in BM, skeletal muscle and angiogenesis in ischemic gastrocnemius, but Metoprolol did not.CONCLUSION:These results unravel that sympathetic nervous system regulate EPCs mobilization and their pro-angiogenic capacity via α adrenoceptor, β 2 adrenoceptor and meanwhile Akt/eNOS signaling pathway.
Dexamethasone Inhibits S. Aureus-Induced Neutrophil Extracellular Pathogen-Killing Mechanism, Possibly Through Toll-Like Receptor Regulation

Abstract:Neutrophils release neutrophil extracellular traps (NETs) in a pathogen-killing process called NETosis. Excessive NETs formation, however, is implicated in disease pathogenesis. Therefore, to understand how NETosis is regulated, we examined the effect of dexamethasone (DXM), an anti-inflammatory drug, on this process and the role of toll-like receptors (TLRs). We stimulated human neutrophils with phorbol 12-myristate 13-acetate (PMA) or Staphylococcus aureus (S. aureus) and quantified NETs formation. We also examined the effect of DXM on the bactericidal effect of NETs and the role of reactive oxygen species (ROS) and nuclear factor (NF)-κB in DXM-regulated NETosis. DXM significantly inhibited S. aureus-induced NETosis and extracellular bacterial killing. ROS production and NF-κB activation were not involved in DXM-regulated NETosis. TLR2 and TLR4, but not TLR5 or TLR6, modified S. aureus-induced NETs formation. Neither DXM nor TLRs were involved in PMA-induced NETosis. Furthermore, TLR2 and TLR4 agonists rescued DXM-inhibited NETosis, and neither TLR2 nor TLR4 antagonists could further inhibit NETosis reduction induced by DXM, indicating that DXM may inhibit NETosis by regulating TLR2 and TLR4. In conclusion, the mechanisms of S. aureus- and PMA-induced NETosis are different. DXM decreases NETs formation independently of oxidant production and NF-κB phosphorylation and possibly via a TLR-dependent mechanism.
ERK1/2 pathway-mediated norepinephrine regulates function of endothelial progenitor cells

Abstract:Objective To analyze the effects of norepinephrine(NE) on the proliferation and migration of endothelial progenitor cells(EPCs) and the related mechanism.Methods NE,adrenoceptor antagonist,and MAPK signal pathway blocker of various concentrations were used to treat peripheral EPCs derived from healthy adults.The proliferation potential,migration capacity and activation of ERK1/2 were assessed after different treatments.Results NE increased the proliferation potential of EPCs in a dose-dependent manner.The number of EPCs increased by(48.3±23.3)%,(70.5±35.6)%,(82.4±14.9)% and(100.3±48.1)% after treatment with NE at 0.01 μmol/L,0.1 μmol/L,1 μmol/L and 10 μmol/L,respectively.Addition of alpha adrenoceptor antagonist phentolamine,selective beta 2 adrenoceptor antagonist I127,JNK blocker SP600125 and ERK1/2 blocker A6355 could block the above effects of NE,and beta 1 adrenoceptor antagonist metoprolol and p38 blocker PD169318 failed to block the effects of NE.NE at 10 μmol/L significantly promoted the migration of EPCs(P<0.05).These effects could be blocked by addition of phentolamine(10 μmol/L) and I127(10 μmol/L),but not by addition of metoprolol.NE(0.1,1 and 10 μmol/L) activated ERK1/2 pathway in a dose-dependent manner,which could also be blocked by phentolamine and I127,but not by metoprolol.Conclusion NE can increase the proliferation potential and migration capacity of EPCs via activating ERK1/2 pathway with alpha and beta 2 adrenoceptor.
Propranolol Promotes Monocyte-to-Macrophage Differentiation and Enhances Macrophage Anti-Inflammatory and Antioxidant Activities by NRF2 Activation

Abstract:Adrenergic pathways represent the main channel of communication between the nervous system and the immune system. During inflammation, blood monocytes migrate within tissue and differentiate into macrophages, which polarize to M1 or M2 macrophages with tissue-damaging or -reparative properties, respectively. This study investigates whether the β-adrenergic receptor (β-AR)-blocking drug propranolol modulates the monocyte-to-macrophage differentiation process and further influences macrophages in their polarization toward M1- and M2-like phenotypes. Six-day-human monocytes were cultured with M-CSF in the presence or absence of propranolol and then activated toward an M1 pro-inflammatory state or an M2 anti-inflammatory state. The chronic exposure of monocytes to propranolol during their differentiation into macrophages promoted the increase in the M1 marker CD16 and in the M2 markers CD206 and CD163 and peroxisome proliferator-activated receptor ɣ expression. It also increased endocytosis and the release of IL-10, whereas it reduced physiological reactive oxygen species. Exposure to the pro-inflammatory conditions of propranolol-differentiated macrophages resulted in an anti-inflammatory promoting effect. At the molecular level, propranolol upregulated the expression of the oxidative stress regulators NRF2, heme oxygenase-1 and NQO1. By contributing to regulating macrophage activities, propranolol may represent a novel anti-inflammatory and immunomodulating compound with relevant therapeutic potential in several inflammatory diseases.

Comparisons of the electrophysiological effects of intravenous sotalol and propranolol on the immature mammalian heart

Abstract:Sotalol is a beta blocker that has also been reported to exert class III antiarrhythmic effects. To evaluate the effects of sotalol on the immature heart, and specifically to assess the relative importance of its class III action, the electrophysiologic effects of incremental doses of intravenous dl-sotalol (cumulative dose of 8 mg/kg) were studied in 11 intact canines (ages 4-15 days) utilizing intracardiac programmed stimulation and electrogram recording techniques. These results were compared to the electrophysiologic effects obtained in an additional 14 neonatal canines given 0.6 mg/kg of the beta blocker propranolol intravenously. Sotalol caused a greater increase than propranolol in the resting sinus cycle length (45 vs. 4%). Importantly, sotalol resulted in greater increases in atrial and ventricular muscle refractoriness than did propranolol (AERP--77 vs. 4%, AFRP--57 vs. 6%; VERP--53 vs. 4%, VFRP--51 vs. 7%). Thus, the electrophysiologic effects of sotalol include large changes in myocardial refractoriness that are not observed with simple beta blockade induced by propranolol. These results suggest that sotalol exerts a significant class III effect in the immature mammalian heart, and thus may be useful as an antiarrhythmic agent in the neonate.
Exploiting the Cellular Actions of Skca and Ikca Channels to Manipulate Endothelial Function and Vascular Tone

Abstract:Growing evidence indicates that small‐ and intermediate‐conductance, Ca2+‐activated K+ channels (SKCa and IKCa channels, respectively) play pivotal roles in the cellular mechanisms contributing to stimulated vasorelaxation (i.e. nitric oxide (NO) synthesis, actions of EDHF). Using single cell patch clamp and fluorimetry, along with arteriolar pressure myography, we have obtained novel results demonstrating that activators of SKCa and IKCa channels (i.e. NS309 and DCEBIO, 0.01 ‐ 1 μM) enhance a number of cellular processes associated with stimulated vasorelaxation (i.e. cytosolic Ca2+ elevations, membrane hyperpolarization and NO synthesis). These observed effects of NS309 and DCEBIO were largely prevented by the SKCa and IKCa channel blockers apamin and TRAM‐34 in combination, but not when applied separately. In rat cremaster arterioles pressurized to 70 mmHg, NS309 and DCEBIO reduced myogenic tone and further augmented ACh‐induced vasodilation. The eNOS inhibitor L‐NAME (0.1 mM) decreased ACh‐evoked inhibition of myogenic tone, but did not affect the dilatory responses to NS309 and DCEBIO. The observed inhibition of NS309/DCEBIO action following endothelial denudation confirmed that these agents acted predominantly via the vascular endothelium. Finally, we will discuss initial efforts to discern the contribution of STIM1 to SKCa and IKCa activity and endothelial vasodilatory processes.
Oxidative and adhesive responses of human neutrophils to nitrovasodilators in vitro: the role of protein kinases

Abstract:Background: Sodium nitroprusside (SNP) and molsidomine are used in the treatment of coronary heart disease. Since the neutrophils play a pathological role in ischaemic heart disease, it is important to understand the direct action of nitrovasodilators on their function. Aim: We examined the effects of SNP and 3-morpholinosydnonimine (SIN-1, molsidomine metabolite) on the respiratory burst of human neutrophils and their adhesion in vitro . The influence of nitric oxide (NO)donors on the activity of protein kinases, which are involved in the NADPH oxidase activation, was also investigated. Methods: The respiratory burst of neutrophils was determined by chemiluminescence and fluorescence methods, while the adhesion was assayed by adherence of neutrophils to the plastic surface. Results: NO donors decreased the oxidative burst of activated neutrophils. However, the effects of SNP and SIN-1 strongly depended on the treatment time of neutrophils and on the stimulus employed to cells activation. Protein kinase C inhibitor did not prevent the inhibitory effect of SIN-1, but diminished the inhibitory effect of SNP on the neutrophils' respiratory burst. Protein tyrosine kinase inhibitor did not affect the action of SNP, but diminished the inhibitory effect of SIN-1 on fMLP-stimulated but not on PMA stimulated oxidative burst of neutrophils. This suggests that SNP action is mainly associated with protein kinase C, while SIN-1 is associated with protein tyrosine kinase activity. We also found that SIN-1 but not SNP diminished the adhesive activity of neutrophils. Conclusions: Our data show that SIN-1 biological effect on some neutrophils activity is different from both spermine NONOate and SNP, and mainly depends on ONOO(-), while SNP action is mediated by NO.

Abstract:Sixteen portal cavernomas were examined with dynamic computed tomography (CT) and confirmed by sonography. Characteristic features included loss of the normal vascular structure and the presence of sinuous collateral pathways which were enhanced during the portal phase. Fifteen patients exhibited transient differences in hepatic attenuation and peripheral arterial concentration of contrast material. Increased arterial flow in the poorly perfused territories is suggested as a cause of these hemokinetic abnormalities.
Propranolol Alleviates Cardiac Injury After Acute Catecholamine Infusion via p38-MAPK Pathways

Abstract:Hyper-catecholaminergic conditions are known to cause heart failure and cardiac fibrosis when severe. Although previous investigations have studied the effects of beta-blockade in experimental models of catecholaminergic states, the detailed benefits of beta-blockade in more realistic models of hyper-adrenergic states were less clear. In this study, we examined acute cardiac changes in rats with hyper-acute catecholamine-induced heart failure with and without propranolol treatment. Male Sprague-Dawley rats (n = 12) underwent a 6-hour infusion of epinephrine and norepinephrine alone, with an additional propranolol bolus (1 mg/kg) at hour 1 (n=6). Cardiac tissues were examined after 6 hours. Cardiac immunohistochemistry revealed significantly decreased expression of phosphorylated p-38 (LV, p= 0.021; RV, p=0.021), with upregulation of reactive oxidative species and other pro-fibrosis proteins, after catecholamine infusion alone. After one propranolol 1 mg/kg bolus, the levels of phosphorylated-p38 returned to levels comparable to sham (LV, p= 0.021; RV, p= 0.043), with additional findings including downregulation of the apoptotic pathway and pro-fibrotic proteins. We conclude that catecholamine-induced heart failure exerts damage through the p-38 MAP kinase pathway, and demonstrates pro-fibrotic changes mediated by matrix metalloproteinase 9, alpha smooth muscle actin, and fibroblast growth factor-23. Changes in these pathways attenuated acute catecholamine-induced heart failure after propranolol bolus 1 mg/kg. We conclude that propranolol bolus at 1 mg/kg is able to mediate the effects of catecholamine excess through the p-38 MAP kinase pathway, pro-fibrosis, and extrinsic apoptosis pathway.

Stimulation of platelet nitric oxide production by nebivolol prevents thrombosis

Abstract:Objective: dl-Nebivolol, a selective β1-adrenergic receptor antagonist, besides its hypotensive activity exerts vasodilatory and platelet inhibitory effects in vitro by a mechanism involving nitric oxide (NO). Our aim was to evaluate whether nebivolol exerts in vivo antithrombotic effects, to unravel the mechanism of this action and to clarify the relative roles of its 2 enantiomers: d- and l-nebivolol. Methods and results: In wild-type mice, dl-nebivolol, l-nebivolol, and d-nebivolol, but not bisoprolol, reduced mortality consequent to platelet pulmonary thromboembolism induced by the intravenous injection of collagen plus epinephrine (-44%, -45%, -29%, respectively; P<0.05), whereas in eNOS(-/-) mice only dl-nebivolol and d-nebivolol were effective. dl-Nebivolol, l- and d-nebivolol reduced photochemical damage-induced femoral artery thrombosis in wild-type mice, whereas in eNOS(-/-) mice only dl-nebivolol and d-nebivolol were active. Moreover, dl-nebivolol and l-nebivolol increased plasma, urinary-, and platelet-derived nitrites and nitrates (NOx), NO degradation products, in wild-type but not in eNOS(-/-) mice. In vivo platelet activation, assessed by platelet P-selectin expression, was reduced by dl-nebivolol and l- and d-nebivolol in wild-type mice but only by dl-nebivolol and d-nebivolol in eNOS(-/-) mice. In bone marrow-transplanted, chimeric mice with only blood cells, and not the endothelium, producing NO dl-nebivolol and l-nebivolol maintained their antithrombotic activity, whereas they lose it in chimeras with only endothelium, and not blood cells, producing NO. In vitro, with isolated platelets, dl-nebivolol and l-nebivolol, but not d-nebivolol and bisoprolol, increased platelet cGMP and NOx formation. Treatment with dl-nebivolol and l-nebivolol increased phophorylated eNOS in platelets. Conclusions: Our data show that dl-nebivolol exerts an antithrombotic activity by stimulating the formation of NO by platelets, and that this effect is generated by its l-enantiomer, whereas the d-enantiomer exerts a weak antiplatelet effect because of β-adrenergic receptor-independent stimulation of adenyly cyclase. These results confirm that platelet-derived NO plays a role in thrombosis prevention and it may represent a target of pharmacological intervention.
The negative chronotropic effects of (±)-propranolol and (±)-4-NO2-propranolol in the rat isolated right atrium are due to blockade of the 6-nitrodopamine receptor

Abstract:The positive chronotropic action induced by 6-nitrodopamine (6-ND) is selectively blocked by β 1 -adrenoceptor antagonists at concentrations that do not affect the positive chronotropic effect induced by dopamine, noradrenaline, and adrenaline. Here, the effects of ( ±)-propranolol, ( ±)-4-NO 2 -propranolol, and ( ±)-7-NO 2 -propranolol were investigated in the rat isolated right atrium. The atrium was mounted in glass chambers containing gassed (95%O 2 :5%CO 2 ) and warmed (37 °C) Krebs–Henseleit's solution, and the isometric tension registered (PowerLab system). ( ±)-Propranolol, ( ±)-4-NO 2 -propranolol, and ( ±)-7-NO 2 -propranolol caused concentration-dependent falls in the spontaneous atrial frequency (pIC 50 : 4.80 ± 0.10, 4.64 ± 0.10, and 4.95 ± 0.10, respectively). The calculated pA 2 values for ( ±)-propranolol, ( ±)-4-NO 2 -propranolol, and ( ±)-7-NO 2 -propranol on noradrenaline-induced positive chronotropism were 8.44 ± 0.08, 6.41 ± 0.07, and 9.21 ± 0.29, respectively. The positive chronotropism induced by 6-ND (10 pM) was blocked by ( ±)-propranolol (1 μM) and ( ±)-4-NO 2 -propranolol (30 nM), whereas ( ±)-7-NO 2 -propranolol (1 μM) had no effect on 6-ND-induced responses. The pIC 50 of ( ±)-propranolol, ( ±)-4-NO 2 -propranolol, and ( ±)-7-NO 2 -propranolol were significantly shifted to the right in L-NAME-treated atria. The discrepancy between pA 2 values of ( ±)-propranolol and its respective pIC 50 indicates that the falls in atrial rate induced by ( ±)-propranolol should not be attributed to b-adrenergic antagonism. The reduced chronotropism by ( ±)-propranolol was unaffected by the sodium channel inhibitors tetrodotoxin and lidocaine but that was abolished in atria pre-treated with ( ±)-4-NO 2 -propranolol. The finding that ( ±)-propranolol reduces spontaneous atrial rate only in concentrations that affect 6-ND-induced positive chronotropism confirms the role of this catecholamine as an endogenous modulator of heart chronotropism. ( ±)-4-NO 2 -Propranolol behaves as a selective antagonist of 6-ND in the rat isolated atrium.

Abstract:The primary goal of this study was to compare the effects of isoproterenol which stimulates beta-adrenergic receptors and forskolin, and NKH 477, a water soluble derivative of forskolin, which stimulate adenylyl cyclase in stunned myocardium of conscious pigs, previously instrumented for measurements of left ventricular pressure and dP/dt, arterial pressure, and wall thickening. Ten min of coronary artery occlusion induced transmural reductions in blood flow (radioactive microspheres) in subepicardium (-98 +/- 2%) and subendocardium (-99 +/- 1%). Wall thickening (piezoelectric crystals) fell from 2.50 +/- 0.26 mm to -0.26 +/- 0.26 mm and remained depressed at 1.37 +/- 0.19 mm after 20-30 min coronary artery reperfusion, reflecting myocardial stunning. At that time, isoproterenol (0.2 microgram/kg) increased wall thickening in stunned myocardium (+1.40 +/- 0.16 mm, P < 0.05) more than in control (+0.71 +/- 0.22 mm), while forskolin elicited the opposite effects. NKH 477 (30 micrograms/kg), which does not penetrate the blood-brain barrier, increased systolic wall thickening similarly before (+0.95 +/- 0.25 mm) and during (+1.01 +/- 0.24 mm) myocardial stunning. The reflex inotropic responses to inferior vena caval occlusion on wall thickening were diminished, P < 0.05, in the stunned myocardium (+0.53 +/- 0.05 mm) compared with control (+0.95 +/- 0.07 mm). beta-adrenergic receptor density, which was quantitated with 125I-cyanopindolol binding, was increased transmurally in stunned myocardium compared with non-ischemic myocardium (subepicardium: +23 +/- 5%, subendocardium: +34 +/- 13%, P < 0.05). Basal and forskolin-stimulated adenylyl cyclase activities were decreased slightly, but significantly, in the stunned subendocardium but not in the subepicardium, while isoproterenol stimulation of adenylyl cyclase activity showed no differences. In summary, paradoxical responses to beta-adrenergic receptor stimulation were observed in stunned myocardium, with pharmacological stimulation with isoproterenol evoking enhanced responses, and neural stimulation with inferior vena caval occlusion eliciting depressed responses. The diminished responses to forskolin in vivo, in stunned myocardium were out of proportion to the biochemical measurements, and may be attributed to neurally mediated cardiac effects of forskolin, since the responses to direct stimulation of adenylyl cyclase by NKH 477 were preserved.
Catecholamine-induced Regulation in Vitro and Ex Vivo of Intralymphocyte Ionized Magnesium.

Abstract:Despite the importance of the adrenergic activity and of the metabolism of magnesium in some important cardiovascular pathologies, very little is known about how intracellular ionized magnesium (Mg i 2+ ) is regulated by catecholamines. We made an in-vitro study of the variations in the concentration of ionized magnesium in human lymphocytes using the fluorescent probe furaptra in response to different catecholamines. We also made an ex-vivo study of the changes in intracellular ionized magnesium in lymphocytes in 20 subjects with essential arterial hypertension, 10 treated with 120 mg/d of propranolol and 10 with placebo. Norepinephrine and isoproterenol significantly decrease Mg i 2+ and this effect is blocked by β-blockers but not by α-blockers. The EC 50 of the effect of norepinephrine is within the range of concentrations physiologically present in plasma. The substitution of extracellular sodium with choline blocks the decrease in intracellular ionized magnesium induced by norepinephrine, which leads us to suppose that the magnesium-reducing effect of catecholamines is a result of the activation of a Na+-Mg2+ exchanger. We were not able to demonstrate any change in intracellular ionized magnesium after 1 and 17 days of active treatment in essential hypertensives. The impossibility of demonstrating ex vivo the mechanism of catecholamine-mediated regulation that is evident in vitro is perhaps due to our experimental conditions or to substances which in vivo inhibit the action of the catecholamines on magnesium, such as insulin and/or glucose.
The DRIFT Directional Dark Matter Detector and First Studies of the Head-Tail Effect

Abstract:Measurement of the direction of the elastic nuclear recoil track and ionization charge distribution along it, gives unique possibility for unambiguous detection of the dark matter WIMP particle. Within current radiation detection technologies only Time Projection Chambers filled with low pressure gas are capable of such measurement. Due to the character of the electronic and nuclear stopping powers of low energy nuclear recoils in the gas, an asymmetric ionization charge distribution along their tracks may be expected. Preliminary study of this effect, called Head-Tail, has been carried out here using the SRIM simulation program for Carbon and Sulfur in 40 Torr carbon disulfide, as relevant to the DRIFT detector. Investigations were focused on ion tracks projected onto the axis of the initial direction of motion in the energy range between 10 and 400 keV. Results indicate the likely existence of an asymmetry influenced by two competing effects: the nature of the stopping power and range straggling. The former tends to result in the Tail being greater than the Head and the latter the reverse. It has been found that for projected tracks the mean position of the ionization charge flows from 'head' to 'tail' with the magnitude depending on the ion type and its energy.
Blood Flow Changes in the Rabbit Cranial and Caudal Venae Cavae during Postural Loads after Propranolol, Bisoprolol and Methyldopa Pretreatment

Abstract:The β 1,2 -adrenergic receptor blocker propranolol, β 1 -blocker bisoprolol, and central presynaptic α 2 -adrenergic receptor agonist methyldopa are used in clinical practice to treat the hyperadrenergic type of postural orthostatic tachycardia syndrome. There are no literature data on the effects of these drugs on vena cava flows during postural tests. In acute experiments on anesthetized rabbits, we studied blood flow changes in the cranial vena cava (CrVC) and caudal vena cava (CdVC) during orthostatic (head-up tilt by 25°) and antiorthostatic (head-down tilt by –25°) tests for 20 s after propranolol, bisoprolol and methyldopa intravenous preadministration. Before drug pretreatment, both CrVC and CdVC flows decreased in response to orthostasis at the 4th and 20th s. During antiorthostasis, CdVC flow increased at the 4th s and decreased to the baseline value by the 20th s, while CrVC flow decreased by the 4th s and exceeded the baseline level by the 20th s. After propranolol pretreatment, CdVC flow decreased to a greater extent than in response to bisoprolol and methyldopa pretreatment. After methyldopa pretreatment during orthostasis, by the 20th s, CrVC flow decreased more notably than CdVC flow, while after propranolol and bisoprolol pretreatment under conditions of orthostasis, both CrVC and CdVC flows decreased about equally. During antiorthostasis, 20 s after propranolol pretreatment, CdVC flow increased more considerably compared to CrVC flow. After bisoprolol and methyldopa pretreatment, CrVC flow increased in response to antiorthostasis not only to a greater extent than CdVC flow but even more pronouncedly compared to its initial increment. Thus, postural tests after the pretreatment with the above drugs reveal differences in the mechanisms of blood flow redistribution in CrVC and CdVC basins.

Propranolol therapy in experimental heart failure in rabbits improves cardiac response to catecholamines without beta-adrenoceptor up-regulation

Abstract:Beta-blockade has been shown to improve cardiac response to catecholamines in heart failure but cellular mechanisms of the improvement are unknown. The effect on left ventricular function of a 14 day propranolol treatment was studied in seven treated and eight non-treated rabbits with experimental heart failure. All animals were subjected to a volume (aortic insufficiency) plus pressure (aortic constriction) overload and were instrumented with a left ventricular catheter and ultrasonic crystals measuring anteroposterior left ventricular diameter. Beta-adrenoceptors were measured using 125I-Cyanopindolol in crude membranes. With isoproterenol, the heart rate was slower in treated rabbits than in non-treated rabbits (p < 0.005) and isoproterenol increased more systolic diameter shortening in treated than in non-treated rabbits (p < 0.05). With norepinephrine, for matched pressures, % delta D increased in the treated group but it did not change in the non-treated group. This improvement of ventricular function was due, in a large part, to an increased diastolic response to norepinephrine: end-diastolic diameter increased in the treated group but not in the non-treated group. In contrast with the improved ventricular response to catecholamines, beta-adrenergic receptor density in the treated group was identical to that of the non-treated group (27.8 fmoles/mg/proteins) and was significantly lower than that of normal rabbits (58.2 fmoles/mg, p < 0.01). The improvement of ventricular response to catecholamines appears to be due to a myocardial protection by propranolol against the toxic effect of catecholamines in heart failure and not, at least in this model, to an up-regulation of beta-adrenoceptors.
Noradrenaline inhibits neurogenic propulsive motor patterns but not neurogenic segmenting haustral progression in the rabbit colon

Abstract:Background: Excessive sympathetic inhibition may be a cause of colon motor dysfunction. Our aim was to better understand the mechanisms of sympathetic inhibition on colonic motor patterns using the rabbit colon, hypothesizing that noradrenaline selectively inhibits propulsive motor patterns. Methods: Changes in motor patterns of the rabbit colon were studied ex vivo using noradrenaline and adrenoceptor antagonists and analyzed using spatiotemporal diameter maps. Key results: Noradrenaline abolished propulsive contractions: it abolished the long-distance contractions (LDCs) from a baseline frequency of 0.8 ± 0.3 and the clusters of fast propagating contractions (FPCs) at a frequency of 14.4 ± 2.8 cpm. Both motor patterns recovered after addition of the α2 -adrenoceptor antagonist yohimbine to a frequency of 0.5 ± 0.2 and 9.9 ± 3.3 cpm, respectively. The β-adrenoceptor antagonist propranolol did not prevent the loss of propulsive motor patterns with noradrenaline. Noradrenaline did not inhibit haustral boundary contractions and increased the frequency of the myogenic ripples from 8.3 ± 1.4 to 10.5 ± 1.3 cpm which was not affected by yohimbine, propranolol nor the α1 -adrenoceptor blocker prazosin. Conclusions and inferences: Noradrenergic inhibition of propulsive motor patterns is mediated by the α2 -adrenoceptor to inhibit the neurogenic LDCs and the neurogenic clustering of FPCs. The neurogenic haustral boundary contractions are not affected, suggesting that α2- receptors are on selective neural circuits. The excitatory effect of noradrenaline on ripples may be due to the activation of adrenoceptors on interstitial cells of Cajal, but action on α1- receptors was excluded. No role for the β-adrenoceptor was found.
