Abstract:Copper (Cu) contamination dramatically affects crop growth and thus threatens crop production; while applications of melatonin (MT) serve as an effective way to tolerate Cu stress for plant development, the underlying mechanism remains largely unknown in rice. Here, we found that Cu toxicity remarkably decreased germination rates and seedling growth compared to the untreated control (CK), while seed priming with a solution of 100 μM MT significantly alleviated the adverse effects on Cu-stressed seeds. In addition, the MT treatment decreased the accumulation of Cu in seedlings at 7 days after imbibition (DAI), possibly through enhanced Cu sequestration, and improved reserve mobilization through the promoted activity of α-amylase and protease in seeds under Cu stress. Interestingly, gibberellin (GA) synthesis was restored to or even exceeded the CK levels in the MT presoaking treatment, while the abscisic acid (ABA) content decreased compared to those of the Cu-stressed seeds, suggesting crosstalk between MT and other phytohormones, e.g., GA and ABA. More importantly, MT pretreatment also significantly promoted the growth of postgermination seedlings. This was largely ascribed to the MT-ameliorated antioxidant system, which consequently reduced the accumulation of Cu stress-induced oxidative products, e.g., hydrogen peroxide (H2O2), malondialdehyde (MDA), and superoxide (O2·_). Collectively, these results demonstrate that seed priming with MT could greatly mitigate the adverse effects of Cu stress on seed germination and subsequent postgermination growth through crosstalk among various defensive response pathways. This study provides vital guidance for applications of MT in agronomic production.
Comparative Physiological and Transcriptome Analysis Reveal the Molecular Mechanism of Melatonin in Regulating Salt Tolerance in Alfalfa (Medicago sativa L.)

Abstract:As a high-quality legume forage, alfalfa is restricted by various abiotic stresses during its growth and development. Melatonin is a multifunctional signaling molecule that involves in plant defense against multiple stresses. However, little is known about its downstream signaling pathway and regulatory mechanisms in salt stress of alfalfa. In this study, we investigated the protective effects and key regulatory pathways of melatonin on alfalfa under salt tolerance. The results showed that melatonin promoted the growth of alfalfa seedlings under salt stress, as demonstrated by higher plant height, leaf area, and fresh weight. Melatonin treatment resulted in an increase in the photosynthetic capacity and starch content of alfalfa. Moreover, melatonin decreased cell membrane damage and reactive oxygen species (ROS) accumulation by enhancing antioxidant defense activity under salt stress conditions. Transcriptome sequencing (RNA-seq) analysis revealed that melatonin mainly induced the transcription of genes involved in Ca2+ signaling (cyclic nucleotide gated channel, CNGCs; cam modulin/calmodulin-like protein, CAM/CMLs and calcium-dependent protein kinase, CDPKs), starch and sucrose metabolism (α-amylase, AMYs; β-amylase, BAMs; starch synthase, SSs and sucrose synthase, SUSs), plant hormone signal transduction (auxin/indole acetic acid protein, AUX/IAAs; ABA receptor, PYL4; protein phosphatase 2C, PP2Cs; scarecrow-like protein, SCLs and ethylene-responsive transcription factor 1B, ERF1B), and key transcription factors (C3Hs, MYBs, ERFs, and WRKYs). Specifically, we focused on starch and sucrose metabolism and plant hormone signal transduction pathways. The interactions between melatonin and other phytohormones occurred via regulation of the expression of genes involved in hormone signaling pathways. In addition, melatonin increased the contents of endogenous melatonin, auxin, gibberellic acid (GA3), salicylic acid, brassinosteroids, and ethylene, while decreasing the abscisic acid content under salt stress. In summary, this study established a regulatory network for melatonin-induced key signaling pathways and functional genes under salt stress and provided a theoretical basis for salt tolerance breeding in alfalfa.

Abstract:Salinity is one of the most common factors affecting alfalfa (Medicago sativa L.), and NaCl is one of the main factors of salinity stress which can cause heavy losses in agricultural production in the world. The application of exogenous melatonin (MT) plays a major role in numerous plants against various stress environments. The effects of exogenous MT on the NaCl tolerance of alfalfa treated with the control, 100 µmol L-1 MT, 150 mmol L-1 NaCl, or 150 mmol L-1 NaCl+ 100 µmol L-1 MT were investigated. The results showed that MT increased growth parameters, inhibited chlorophyll degradation and promoted photosynthetic gas exchange parameters (photosynthetic rate, conductance to H2O, and transpiration rate) and stomatal opening under NaCl stress. Osmotic regulation substances such as soluble sugar, proline and glycine betaine were the highest in the NaCl treatment and the second in the NaCl+MT treatment. Nitrogen, phosphorus, potassium, calcium and magnesium were reduced and sodium was increased by NaCl, whereas these levels were reversed by the NaCl+MT treatment. MT inhibited cell membrane imperfection, lipid peroxidation and reactive oxygen species (ROS) accumulation caused by NaCl stress. MT up-regulated the gene expression and activity of antioxidant enzymes and increased the content of antioxidant non-enzyme substances to scavenge excessive ROS in NaCl-treated plants. In addition, all indicators interacted with each other to a certain extent and could be grouped according to the relative values. All variables were divided into PC 1 (89.2 %) and PC 2 (4 %). They were clustered into two categories with opposite effects, and most of them were significant variables. Hence, these findings reveal that exogenous MT alleviates the inhibitory effects of NaCl stress on photosynthesis, stomata opening, osmotic adjustment, ion balance and redox homeostasis, enhancing tolerance and growth of alfalfa. Furthermore, it suggests that MT could be implemented to improve the NaCl tolerance of alfalfa.
Melatonin Mitigated Salinity Stress on Alfalfa by Improving Antioxidant Defense and Osmoregulation

Abstract:Melatonin (MT) is a growth regulator and antioxidant that can resist peroxidation damage on plants caused by environmental stresses. In this study, the alleviation effects of melatonin on alfalfa under salt stress were investigated in terms of photosynthesis, antioxidant enzymes, and osmoregulation. The alfalfa seedlings were cultured in 200 mM NaCl Hoagland solution. Five levels of MT (0, 0.1, 0.2, 0.3, and 0.4 mM) were applied as a foliar spray. Generally, the foliar spray of MT increased root length, root surface area, height, leaf length and width, aerial and root biomass, SPAD readings, the content of proline and soluble protein, and the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT). Malonaldehyde (MDA) content was decreased by MT foliar spray. The beneficial effects of MT on alfalfa under salt stress were dosage-dependent, and excessive MT levels inhibited alfalfa growth. The alleviating effects of MT on salt stress were more pronounced at 0.3 mM MT. This study suggested that exogenous MT foliar spray at appropriate levels can ameliorate the adverse effects of salt stress on alfalfa seedlings.

Abstract:Melatonin (MT) is a small molecule indole hormone that plays an important role in the regulation of biological processes and abiotic stress resistance. Previous studies have confirmed that MT promotes the normal development of plants under stress by mediating physiological regulation mechanisms. However, the physiological mechanism of exogenous MT regulating seed germination and seedling growth of wheat under salt stress is still unclear. In this study, NaCl stress decreased germination rate and inhibited seedling growth of wheat, but shoot length, root length, and plant weight of SM15 did not change significantly. The addition of 300 μM MT in the cultivation solution directly promoted the germination rate of SM15 and ZM18, and lateral root production, but decreased the germination rate of JM22 and inhibited the length of germ and radicle of three varieties under salt stress. For wheat seedling, application of MT could increase proline content, soluble protein, soluble sugar, Ca2+ content, and vital amino acid content in leaves to keep high water content, low level of H2O2 content, and low [K+]/[Na+] ratio. MT increased root vigor and [K+]/[Na+] ratio and decreased H2O2 content in root induced by salt stress. In conclusion, MT enhanced salt tolerance in wheat seeds and seedlings by regulating the synthesis of soluble protein and sugar, ion compartmentation in roots and leaves, enhancement of enzymatic systems, and changes in amino acid levels. Salt resistance varied with different varieties under the same environmental condition. SM15 was a higher salt-resistant variety and JM22 was a salt-sensitive one. In wheat production, the application of exogenous MT should consider the differences among varieties of wheat during the sowing and seedling stages.

Abstract:Salt stress caused by high concentrations of Na+ and Cl- in soil is one of the most important abiotic stresses in agricultural production, which seriously affects grain yield. The alleviation of salt stress through the application of exogenous substances is important for grain production. Melatonin (MT, N-acetyl-5-methoxytryptamine) is an indole-like small molecule that can effectively alleviate the damage caused by adversity stress on crops. Current studies have mainly focused on the effects of MT on the physiology and biochemistry of crops at the seedling stage, with fewer studies on the gene regulatory mechanisms of crops at the germination stage. The aim of this study was to explain the mechanism of MT-induced salt tolerance at physiological, biochemical, and molecular levels and to provide a theoretical basis for the resolution of MT-mediated regulatory mechanisms of plant adaptation to salt stress. In this study, we investigated the germination, physiology, and transcript levels of maize seeds, analyzed the relevant differentially expressed genes (DEGs), and examined salt tolerance-related pathways. The results showed that MT could increase the seed germination rate by 14.28-19.04%, improve seed antioxidant enzyme activities (average increase of 11.61%), and reduce reactive oxygen species accumulation and membrane oxidative damage. In addition, MT was involved in regulating the changes of endogenous hormones during the germination of maize seeds under salt stress. Transcriptome results showed that MT affected the activity of antioxidant enzymes, response to stress, and seed germination-related genes in maize seeds under salt stress and regulated the expression of genes related to starch and sucrose metabolism and phytohormone signal transduction pathways. Taken together, the results indicate that exogenous MT can affect the expression of stress response-related genes in salt-stressed maize seeds, enhance the antioxidant capacity of the seeds, reduce the damage induced by salt stress, and thus promote the germination of maize seeds under salt stress. The results provide a theoretical basis for the MT-mediated regulatory mechanism of plant adaptation to salt stress and screen potential candidate genes for molecular breeding of salt-tolerant maize.
Exogenous Melatonin Alleviates NaCl Injury by Influencing Stomatal Morphology, Photosynthetic Performance, and Antioxidant Balance in Maize

Abstract:Maize (Zea mays L.) is sensitive to salt stress, especially during seed germination and seedling morphogenesis, which limits maize growth and productivity formation. As a novel recognized plant hormone, melatonin (MT) participates in multiple growth and developmental processes and mediates biotic/abiotic stress responses, yet the effects of salt stress on maize seedlings remain unclear. Herein, we investigated the effects of 150 μM exogenous MT on multiple phenotypes and physiologic metabolisms in three-leaf seedlings across eight maize inbred lines under 180 mM NaCl salt stress, including growth parameters, stomatal morphology, photosynthetic metabolisms, antioxidant enzyme activities, and reactive oxygen species (ROS). Meanwhile, the six gene expression levels controlling antioxidant enzyme activities and photosynthetic pigment biosynthesis in two materials with contrasting salt resistance were examined for all treatments to explore the possible molecular mechanism of exogenous MT alleviating salt injury in maize. The results showed that 150 μM exogenous MT application protected membrane integrity and reduced ROS accumulation by activating the antioxidant system in leaves of maize seedlings under salt stress, their relative conductivity and H2O2 level average reduced by 20.91% and 17.22%, while the activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and ascorbate peroxidase (APX) averaged increased by 13.90%, 17.02%, 22.00%, and 14.24% relative to salt stress alone. The improvement of stomatal size and the deposition of photosynthetic pigments were more favorable to enhancing photosynthesis in leaves when these seedlings treated with MT application under salt stress, their stomatal size, chlorophyll content, and net photosynthetic rate averaged increased by 11.60%, 19.64%, and 27.62%. Additionally, Gene expression analysis showed that MT stimulation significantly increased the expression of antioxidant enzyme genes (Zm00001d009990, Zm00001d047479, Zm00001d014848, and Zm00001d007234) and photosynthetic pigment biosynthesis genes (Zm00001d011819 and Zm00001d017766) under salt stress. At the same time, 150 μM MT significantly promoted seedling growth and biomass accumulation. In conclusion, our study may unravel crucial evidence of the role of MT in maize seedlings against salt stress, which can provide a novel strategy for improving maize salt stress resistance.

Abstract:Alfalfa (Medicago sativa L.) is an important and widely cultivated forage grass. The productivity and forage quality of alfalfa are severely affected by salt stress. Melatonin is a bioactive molecule with versatile physiological functions and plays important roles in response to various biotic and abiotic stresses. Melatonin has been proven efficient in improving alfalfa drought and waterlogging tolerance in recent studies. In our reports, we applied melatonin exogenously to explore the effects of melatonin on alfalfa growth and salt resistance. The results demonstrated that melatonin application promoted alfalfa seed germination and seedling growth, and reduced oxidative damage under salt stress. Further application research found that melatonin alleviated salt injury in alfalfa plants under salt stress. The electrolyte leakage, malondialdehyde (MDA) content and H2O2 content were significantly reduced, and the activities of catalase (CAT), peroxidase (POD), and Cu/Zn superoxide dismutase (Cu/Zn-SOD) were increased with melatonin pretreatment compared to control plants under salt stress with the upregulation of genes related to melatonin and antioxidant enzymes biosynthesis. Melatonin was also involved in reducing Na+ accumulation in alfalfa plants. Our study indicates that melatonin plays a primary role as an antioxidant in scavenging H2O2 and enhancing activities of antioxidant enzymes to improve the salt tolerance of alfalfa plants.

The Physiological Mechanism of Melatonin Enhancing the Tolerance of Oat Seedlings under Saline–Alkali Stress

Abstract:Exogenous melatonin (MT) regulates plant growth and mitigates stress in response to stress. To analyze the machinery of exogenous melatonin, which improves salt and alkaline tolerance in oats, MT’s function was identified in the oat seed germination stage in our previous study. In this study, morphogenesis, photosynthetic physiology, hormone levels, and ion homeostasis were evaluated using the same MT treatment concentration. The results revealed that compared to the S45 treatment, the 100 μmol·L−1 MT treatment efficiently increased the seedling height and main root length of oat seedlings; promoted secondary root development; enhanced the root volume and root surface area; maintained a higher photosynthetic pigment content (carotenoids; chlorophyll a; chlorophyll b); raised the leaf photosynthetic rate (Pn), intercellular CO2 concentration (Ci), conductance to H2O (Gs), and transpiration rate (Tr); enhanced the light energy absorption and conversion of leaves; increased the leaf GA3, Tryptamine (TAM), and IAA contents; and decreased ABA levels. Hierarchical cluster analysis revealed that MT treatment also increased the contents of P, K, Ca, Mn, Cu, Mg, Fe, Zn, Mo, Cd, Al, Se, Ni, Co, and Ti; decreased the Na/K ratio; and maintained cellular ionic homeostasis in oat seedlings under saline–alkali stress, as compared with the untreated group. These findings showed that MT treatment enhanced the adaptation of oat to saline–alkali stress through regulating the physiological process of seedling growth. This suggests that MT plays a different role in improving saline–alkali tolerance in the germination and seedling stages of oat.

Abstract:Saline–alkali stress is one of the major factors limiting oat seed germination. The regulatory role of melatonin (MT) as a naturally occurring active substance is well known, but the mechanism of MT-mediated intrinsic physiological regulation of oat seed germination under saline–alkali stress is unclear. Therefore, this study investigated (1) the variability of different MT seed soaking concentrations and times on the germination of oat seeds under saline–alkali stress, and (2) the possible physiological regulatory mechanisms of MT on the germination of oat seeds under saline–alkali stress. The results showed that seed vigor was significantly reduced under saline–alkali stress, and seed germination of oats was significantly inhibited; different concentrations of MT seed soaking treatments improved the germination rate, germination potential, germination index, vigor index, root length, germ length, fresh weight, and dry weight; and, overall, treatment improved seed germination and exhibited the phenomenon of “low promotion and high inhibition”. The 100 μmol·L−1 MT seed soaking treatment effectively enhanced the activities of seed antioxidant enzymes (SOD, POD, CAT, and APX) and nonenzymatic antioxidants (GSH and AsA), affected the AsA-GSH cycle, and effectively increased the contents of osmoregulatory substances (proline, protein, and soluble sugar) by reducing levels of H2O2, OH−, and MDA, thus enhancing the tolerance of oats to saline–alkali and promoting seed germination. In conclusion, MT has a positive effect on the saline–alkali tolerance of oat seeds.

Abstract:Melatonin is an important phytohormone influencing plant growth and defense responses. However, the mechanism mediating the regulatory effects of melatonin on the salt tolerance of germinating maize seeds remains unexplored. In this study, the application of exogenous melatonin enhanced the salt tolerance of germinating maize seeds. A transcriptome analysis indicated that complex regulatory pathways may be associated with the melatonin-mediated salt tolerance. Remarkably, antioxidant activities, transcriptional regulation, and phytohormone (e.g., cytokinin and auxin) pathways were induced following the exogenous application of melatonin. The microRNA (miRNA)-seq analysis result indicated that exogenous melatonin obviously altered the expression of a set of miRNAs. Notably, many differentially expressed miRNAs and their target genes, including several transcription factor genes, were revealed to contribute to salt stress responses. According to metabolite profiles, the abundance of diverse metabolites, like secondary metabolites, nucleotides, cofactors, and vitamins, increased significantly after using exogenous melatonin. The combined analysis of the transcriptome and metabolome indicated that several gene–metabolite networks related to amino acid metabolism and secondary metabolite biosynthetic pathways are essential for melatonin-mediated maize tolerance to salt stress. Consistent with these findings, exogenously applied melatonin altered the phytohormone levels and increased the antioxidant enzyme activities and energy supply. Our results reflect the significance of melatonin for enhancing the salt tolerance of germinating maize seeds, which is achieved through the regulation of antioxidant capacity, phytohormone content, and metabolic adaptation.
Exogenous Melatonin Reinforces Photosynthesis, Antioxidant Defense and Gene Expression to Ameliorate Na2CO3 Stress in Maize

Abstract:Salt stress can seriously affect the growth and development of maize (Zea mays L.), resulting in a great yield loss. Melatonin (MT), an indole hormone, is a potential enhancer of plant tolerance against salt stress. However, the complex mechanisms of MT application in enhancing maize salt tolerance are still unclear. Herein, three-leaf seedlings of salt-susceptible P138 and its salt-resistant ethyl methane sulfonate (EMS)-104 mutant were cultured with or without 150 μM MT application under 0 and 100 mM Na2CO3 treatments for seven days, to systematically explore the response mechanisms of exogenous MT in improving the salt tolerance of maize. The results showed that salt stress triggered an escalation in reactive oxygen species production, enhanced multiple antioxidant enzymes' activities, impaired cellular membrane permeability, inhibited photosynthetic pigment accumulation, and ultimately undermined the vigor and photosynthetic prowess of the seedlings. While suitable MT application counteracted the detrimental impacts of Na2CO3 on seedlings' growth and photosynthetic capacity, the seedling length and net photosynthetic rate of P138 and EMS-104 were increased by 5.5% and 18.7%, and 12.7% and 54.5%, respectively. Quantitative real-time PCR (qRT-PCR) analysis further showed that MT application activated the expression levels of antioxidant enzyme-related genes (Zm00001d025106, Zm00001d031908, Zm00001d027511, and Zm00001d040364) and pigment biosynthesis-related genes (Zm00001d011819 and Zm00001d017766) in both maize seedlings under Na2CO3 stress; they then formed a complex interaction network of gene expression, multiple physiological metabolisms, and phenotype changes to influence the salt tolerance of maize seedlings under MT or Na2CO3 stress. To sum up, these observations underscore that 150 μM MT can alleviate salt injury of maize seedlings, which may provide new insights for further investigating MT regulation mechanisms to enhance maize seedlings' salt resistance.

Abstract:Salt stress can affect various physiological processes in plants, ultimately hindering their growth and development. Melatonin (MT) can effectively resist multiple abiotic stresses, improving plant stress resistance. To analyze the mechanism of exogenous MT to enhance salt tolerance in red clover, we conducted a comprehensive study to examine the influence of exogenous MT on various parameters, including seed germination indices, seedling morphological traits, and physiological and photosynthetic indicators, using four distinct red clover varieties (H1, H2, H3, and H4). This investigation was performed under various salt stress conditions with differing pH values, specifically utilizing NaCl, Na2SO4, NaHCO3, and Na2CO3 as the salt stressors. The results showed that MT solution immersion significantly improved the germination indicators of red clover seeds under salt stress. The foliar spraying of 50 μM and 25 μM MT solution significantly increased SOD activity (21-127%), POD activity, soluble sugar content, proline content (22-117%), chlorophyll content (2-66%), and the net photosynthetic rate. It reduced the MDA content (14-55%) and intercellular CO2 concentration of red clover seedlings under salt stress. Gray correlation analysis and the Mantel test further verified that MT is a key factor in enhancing seed germination and seedling growth of red clover under salt stress; the most significant improvement was observed for NaHCO3 stress. MT is demonstrated to improve the salt tolerance of red clover through a variety of mechanisms, including an increase in antioxidant enzyme activity, osmoregulation ability, and cell membrane stability. Additionally, it improves photosynthetic efficiency and plant architecture, promoting energy production, growth, and optimal resource allocation. These mechanisms function synergistically, enabling red clover to sustain normal growth and development under salt stress.

Abstract:Melatonin is a ubiquitous signaling plant hormone that plays a crucial role in regulating the growth and development of plants under stress conditions. Since a few species have been investigated to unveil the effect of exogenous melatonin on salt stress, in the current research study, we investigated the effects of melatonin by measuring the photosynthetic characteristics and antioxidant defense system of maize seedling under salt stress (200 mM NaCl), along with different concentrations of melatonin (MT1—30, MT2–60, and MT3–90 µM) treatments. Salt stress reduced the plant growth characteristics and photosynthetic efficiency by increasing the ROS accumulation and reducing the antioxidant enzyme activities of maize seedling. However, pretreatment with melatonin on roots alleviated NaCl-induced decrease in photosynthetic rate and oxidative stress in a dose-dependent manner. Our results revealed that exogenous application of melatonin at an optimum concentration (60 µM) under salt stress conditions significantly increased the growth of plants, chlorophyll content, photosynthetic efficiency, antioxidant enzymes activities, i.e., superoxide dismutase (SOD), peroxide (POD), catalase (CAT) and ascorbate peroxidase (APX), and reduced the reactive oxygen species (ROS). This study unraveled the crucial role of melatonin in salt stress mitigation and thus can be implicated in the management of salinity in maize seedling.

Abstract:Salt stress severely restricts the growth of plants and threatens the development of agriculture throughout the world. Worldwide studies have shown that exogenous melatonin (MT) can effectively improve the growth of plants under salt stress. Through a meta-analysis of 549 observations, this study first explored the effects of salt stress characteristics and MT application characteristics on MT regulated plant growth under salt stress. The results show that MT has a wide range of regulatory effects on plant growth indicators under salt stress, of which the regulatory effect on root indexes is the strongest, and this regulatory effect is not species-specific. The intensity of salt stress did not affect the positive effect of MT on plant growth, but the application effect of MT in soil was stronger than that in rooting medium. This meta-analysis also revealed that the foliar application of a concentration between 100–200 μM is the best condition for MT to enhance plant growth under salt stress. The results can inspire scientific research and practical production, while seeking the maximum improvement in plant salt tolerance under salt stress.
Exogenous Melatonin Improves the Growth of Rice Seedlings by Regulating Redox Balance and Ion Homeostasis Under Salt Stress

Abstract:This study evaluated the effects of foliar spraying melatonin (MT) on the growth of salt-stressed rice. Seedlings were treated with 50 and 100 mM of NaCl and different concentrations of MT (25, 50, 100, 200, 300, and 400 μM) for 14 days. Different concentrations of MT could promote plant growth significantly under salt stress, particularly at concentrations of 200, 300, and 400 μM. A concentration of 200 μM MT was considered as optimal and used in a subsequent experiment on biomass, water content, antioxidation, mineral nutrition, salt absorption, and distribution of salt-stressed rice seedlings. Results showed that MT's promoting effect on plant growth under salt stress was evident with time, particularly under high salt stress. MT improved the activities of antioxidant enzymes, reduced membrane lipid peroxidation, alleviated cell injury in plant leaves, and increased N content and Si accumulation in the leaves and roots under salt stress, particularly under high salinity. This compound also inhibited Na uptake and upward transport, but it promoted or maintained the uptake and upward transport of K and Ca in salt-stressed rice. Thus, MT improved the ion homeostasis of K/Na and Ca/Na in plants, particularly in the leaves. Foliar spraying of MT alleviated salt stress on rice by promoting nutrient accumulation or translocation, improving ion homeostasis, which is evident in the leaves, and consequently enhancing its salt resistance. The antioxidative improvement caused by MT might also be related to the improved ion homeostasis.

Abstract:Salt hazard is one of the major environmental stresses that seriously threaten world food security. Although melatonin has been proven to improve stress tolerance in plants, the metabolic changes and gene transcription affected by melatonin (MT) under salt stress remain uncertain, especially in maize. In the present study, we investigated the differences in the diurnal metabolism and transcription of maize seedlings to reveal the MT-mediated changes in diurnal metabolism in response to salt stress. Transcriptome analysis identified melatonin crosstalk with other hormones affecting their signal transduction pathways. The transcriptome and metabolome co-analysis revealed that MT maintained osmotic potential and regulated energy supply by regulating sucrose and starch metabolism. In addition, the regulatory network showed that MT and its metabolites were significantly correlated with a series of stress and hormone-related transcription factor (TF) genes, such as JAZ3-1 , JAZ9 , WRKY28 , and ERF020 . Taken together, our results indicated that MT alleviated salt stress-induced damage, influenced diurnal metabolic changes in maize under salt stress, and crosstalk with other endogenous hormones to improve salt tolerance in maize.

Abstract:Melatonin is an important phytohormone in plant development and stress responses. However, the functions of melatonin in rice salt tolerance during seed germination is largely undetermined. In this study, we investigated its potential molecular mechanism during rice seed germination under salinity through comprehensive transcriptome sequencing, metabolome profiling, and physiological assays. Transcriptome analysis revealed 4794 differentially expressed genes (DEGs) after melatonin pretreatment under salinity, including 2843 upregulated and 1951 downregulated genes. Functional annotation showed that many DEGs were associated with antioxidative activity and phytohormone biosynthesis and signal transduction. Consistent with the transcriptome results, physiological analysis revealed enhanced activity of antioxidant capacity, increased indole-3-acetic acid (IAA) level, and reduced abscisic acid (ABA) level under melatonin pretreatment. The enhanced antioxidative activity was also supported by metabolome assay that melatonin pretreatment increased the levels of non-enzymatic antioxidant organic acids and amino acids. The results demonstrated that exogenous melatonin can enhance rice seed tolerance to salinity during germination via activating antioxidants and modulating phytohormones.

Abstract:Melatonin (MT) is an important plant growth regulator that significantly regulates the growth and development of plants. Previous studies confirmed the effectiveness of MT in improving plant stress tolerance. In this study, annual M9-T337 seedlings were selected as subjects, and five treatments were applied: control (CK), in which only half the concentration of Hoagland was applied; Saline-alkaline stress treatment (SA, 100 mmol·L −1 saline-alkaline solution); melatonin treatment (MT, CK + 200 μmol L −1 exogenous MT); Saline-alkaline + melatonin treatment (MS, SA + 200 μmol L −1 exogenous MT); and saline-alkaline stress + melatonin + inhibitor treatment (HS, additional 100 μmol L −1 p-CPA treatment to MS). The results showed that saline-alkaline stress negatively affected the growth of M9-T337 seedlings by reducing photosynthetic capacity, increasing Na + , promoting reactive oxygen species such as H 2 O 2 , and changing the osmotic content and antioxidant system. However, the application of exogenous MT effectively alleviated saline-alkaline damage and significantly promoted the growth of M9-T337 seedlings. It significantly increased plant height, diameter, root length, root surface area, volume and activity. Furthermore, MT alleviated osmotic stress by accumulating proline, soluble sugars, soluble proteins and starch. MT improved photosynthetic capacity by delaying chlorophyll degradation and regulating gas exchange parameters as well as fluorescence parameters in leaves. Additionally, MT reduced the Na + /K + ratio to reduce ion toxicity by upregulating the expression of Na + transporter genes ( MhCAX5 , MhCHX15 , MhSOS1 , and MhALT1 ) and downregulating the expression of K + transporter genes ( MhSKOR and MhNHX4 ). In addition, MT can increase antioxidant enzyme activity (superoxide dismutase (SOD), peroxidase(POD), catalase (CAT), ascorbic acid oxidase (AAO), ascorbate peroxidase (APX) and monodehydroascorbate reductase (MDHAR)) in the ASA-GSH cycle and increase ascorbic acid (AsA), reduced glutathione (GSH) and oxidized glutathione (GSSG) levels to counteract the accumulation of reactive oxygen species (ROS) such as hydrogen peroxide (H 2 O 2 ) and Superoxide anion free radicals (O 2 − ), reducing oxidative damage. Exogenous MT promotes M9-T337 seedlings growth under saline-alkaline stress by responding synergistically with auxin (IAA), gibberellin (GA 3 ) and zeatin (ZT) to saline-alkaline stress. Our results confirm that MT has the potential to alleviate Saline-alkaline stress by promoting root growth, increasing biomass accumulation and photosynthetic capacity, strengthening the antioxidant defense system, maintaining ionic balance, the ascorbate–glutathione cycle and the Osmoregulation facilitates and regulates endogenous hormone levels in M9-T337 seedlings.

Abstract:Seed priming is a useful seed pretreatment technology, which is widely applied in crop cultivation. Salt stress is one of the important environmental stresses that always inhibited cotton (Gossypium hirsutum L.) growth. However, melatonin could alleviate abiotic and biotic stress in plants. In this study melatonin solution was used for seed priming and the role of melatonin priming in cotton seedlings tolerance to salt stress was investigated. As a widely extended upland cotton cultivar in China, CCRI12 was used as a material to study the effects of seed priming with melatonin on the seedling tolerance to the salt stress. Through morphological characterization, 25 mu M melatonin primed seedlings exhibited the best phenotype after 14 days salt stress treatment. To reveal the mechanism of melatonin priming alleviating the salt stress, the photosynthesis indexes and antioxidant enzymes activities were detected. It was displayed that seedlings from 25 mu M melatonin primed seeds showed better photosynthetic efficiency and stronger ability to scavenge reactive oxygen species. Besides, transcription sequencing analysis, it was found that almost all the photosynthesis-related genes in the melatonin primed seedlings were significantly down-regulated after 48 h salt stress treatment. It might be a way to decrease the photosynthetic rate to adapt salt stress and mitigate the damage. Moreover, amounts of genes encoding antioxidant enzymes were up-regulated by melatonin priming under salt stress, which was in accordance with the result of enzymes activities. Furthermore, a large number of genes participating in eight phytohormones signal pathways were significantly and differentially expressed by melatonin priming under salt stress, which suggested that melatonin could cooperated with phytohormone signal pathways to respond against salt stress. Therefore, it was indicated that melatonin priming might improve the tolerance of cotton seedling by regulating photosynthetic efficiency, enhancing the active oxygen scavenging system and coordinating to phytohormone signal pathways. Therefore, the application of melatonin, enhancing seedling tolerance to salt stress displayed its great potential for cotton production in the salinized land.
