Abstract:As Olaparib has received FDA-approval for treatment of cancer patients with BRCA1 and BRCA2 mutations, the target related on Poly (ADP-ribose) polymerase (PARP) inhibitors have been emerging as a novel therapy for treatment of patients with advanced ovarian cancer. It has been observed that PARP inhibitors have optimistic antitumor activity of patients in advanced ovarian cancer bearing homologous recombination (HR) defects, supporting a substantially wider role for PARPi. Strategies to identify other potential biomarkers remain under investigation. Herein, we review recent updates on potential moleculars to predict biomarker of PARPi for cancer therapy and proposed an integrated biomarker system to predict the "appropriate users" of PARPi. It may help overcome PARPi treatment failure, which reveal novel insights into clinical use of PARPi.

Abstract:Novel PARP inhibitors with selective mode-of-action have been approved for clinical use. Herein, oxadiazole based ligands that are predicted to target PARP-1 have been synthesized and screened for the loss of cell viability in mammary carcinoma cells, wherein seven compounds were observed to possess significant IC50 values in the range of 1.4 to 25 μM. Furthermore, compound 5u, inhibited the viability of MCF-7 cells with an IC50 value of 1.4μM, when compared to Olaparib (IC50 = 3.2 μM). Compound 5s also decreased cell viability in MCF-7 and MDA-MB-231 cells with IC50 values of 15.3 and 19.2 μM, respectively. Treatment of MCF-7 cells with compounds 5u and 5s produced PARP cleavage, H2AX phosphorylation and CASPASE-3 activation comparable to that observed with Olaparib. Compounds 5u and 5s also decreased foci-formation and 3D Matrigel growth of MCF-7 cells equivalent to or greater than that observed with Olaparib. Finally, in silico analysis demonstrated binding of compound 5s towardsthe catalytic site of PARP-1, indicating that these novel oxadiazoles synthesized herein may serve as exemplars for the development of new therapeutics in cancer.

Abstract:A series of benzimidazole carboxamide derivatives have been synthesized and characterized by 1H-NMR, 13C-NMR and HRMS. PARP inhibition assays and cellular proliferation assays have also been carried out. Compounds 5cj and 5cp exhibited potential anticancer activities with IC50 values of about 4 nM against both PARP-1 and PARP-2, similar to the reference drug veliparib. The two compounds also displayed slightly better in vitro cytotoxicities against MDA-MB-436 and CAPAN-1 cell lines than veliparib and olaparib, with values of 17.4 µM and 11.4 µM, 19.8 µM and 15.5 µM, respectively. The structure-activity relationship based on molecular docking was discussed as well.

Abstract:ID 127349 Poster Board 460 Poly(ADP-ribose) polymerase 1 (PARP1), a DNA repair nuclear enzyme, is over-expressed in ovarian, breast, and prostate cancers and its targeted inhibition by small molecules (e.g., olaparib, rucaparib, niraparib, and talazoparib) has been successfully utilized in clinic. Unfortunately, frequent use of PARP inhibitors in clinics has also resulted in patients developing rapid resistance via various mechanisms. Therefore, novel class of PARP1 inhibitory scaffold that is potent and selective is urgently needed to circumvent this problem. In this study, a novel nicotinamide mimicking pharmacophore, 7-aminoquinolin-2(1H)-one, has been identified. With the aid of molecular modeling, we have designed and synthesized several derivatives as PARP1 inhibitors utilizing acid-amine coupling and reductive alkylation protocol. A recombinant in vitro PARP1 enzyme assay revealed that developed inhibitors are potent and selective and can potentially exert favorable BRCA-driven cytotoxicity profile in ovarian and breast cancer cell lines. This effort could potentially lead to development of PARP1-targeted drugs with enhanced therapeutic benefits.

Abstract:Poly(ADP-ribose) polymerase 1 (PARP1) inhibition, increasing chemosensitization and conferring independent antiproliferation against defective homologous recombination cells, has provided a unique opportunity for anticancer therapeutic intervention. Therefore, PARP1, the best characterized enzyme among PARP family, is presently serving as a well-established target for the treatment of oncology attributed to its intimate role in DNA repair. Nowadays, intensified medicinal chemistry efforts have led to the discovery of 12 PARP1 inhibitors that have been advanced into clinical trial. In this article, we classify these candidates as three categories based on the chemical structure, including lactam type, pseudo ring type and untypical PARP1 inhibitors, for a sequential overview of them. In particular, the development of some candidates will serve the purpose for the future exploration of PARP1 inhibitors.

Abstract:To further explore the research of novel PARP-1 inhibitors, we designed and synthesized a series of novel amide PARP-1 inhibitors based on our previous research. Most compounds displayed certain antitumor activities against four tumor cell lines (A549, HepG2, HCT-116, and MCF-7). Specifically, the candidate compound R8e possessed strong anti-proliferative potency toward A549 cells with the IC50 value of 2.01 μM. Compound R8e had low toxicity to lung cancer cell line. And the in vitro enzyme inhibitory activity of compound R8e was better than rucaparib. Molecular docking studies provided a rational binding model of compound R8e in complex with rucaparib. The following cell cycle and apoptosis assays revealed that compound R8e could arrest cell cycle in the S phase and induce cell apoptosis. Western blot analysis further showed that compound R8e could effectively inhibit the PAR's biosynthesis and was more effective than rucaparib. Overall, based on the biological activity evaluation, compound R8e could be a potential lead compound for further developing novel amide PARP-1 inhibitors.

Abstract:Poly ADP ribose polymerase-1 (PARP-1), one of the most important members of the PARP protein family, plays a crucial role in DNA damage repair, gene transcription, and apoptosis of cancer cells. In this work, benzofuran[3,2-d]pyrimidine-4(3H)-one was used as a framework to design and synthesize a series of novel PARP-1 inhibitors by introducing thiosemicarbazone or its derivatives into the scafford. Among all the target compounds, 19b and 19c were found to exhibit more potent inhibitory activity and higher selectivity against PARP-1 than Olaparib, especially the latter had an IC50 value of 0.026 μM against PARP-1 enzyme and a PARP-2/PARP-1 selectivity of 85.19-fold over Olapanib. Apart from strong cytotoxicity against the tested cancer cell lines, 19c was most sensitive to SK-OV-3 cells, with an IC50 value of 4.98 μM superior to Olaparib. Anti-cancer mechanism studies revealed that 19c could inhibit DNA single-strand breakage repair and aggravate DNA double-strand breakage by inhibiting PARP-1 activity, and promote the apoptosis of cancer cells through the mitochondrial apoptosis pathway.

Abstract:PARP-1/2 inhibitors have become an important therapeutic strategy for the treatment of HR-deficient tumors. However, discovery of new inhibitors with an improved and distinct pharmacological file still need enormous explorations. Herein, a series of novel highly potent PARP-1/2 inhibitors bearing an N-substituted piperazinone moiety were achieved. In particular, Cpd36 was identified as a distinct PARP inhibitor, showing remarkable enzymatic activity not only toward PARP-1 (IC50 = 0.94 nM) and PARP-2 (IC50 = 0.87 nM) but also toward PARP-7 (IC50 = 0.21 nM), as well as high selectivity over other PARP isoforms. Furthermore, Cpd36 was orally bioavailable and significantly repressed the tumor growth in both breast cancer and prostate cancer xenograft model. The crystal structures of Cpd36 within PARP-1 and PARP-2 together with the predicted binding mode within PARP-7 revealed its binding features and provided insightful information for further developing highly potent and selective PARP-1 and/or PARP-7 inhibitors.

Abstract:The development of environment-friendly new Poly-adenosine diphosphate (ADP)-ribose Polymerase (PARP) inhibitors are highly essential because of their involvement in the survival of cancer cells. Therefore, a library of indazolyl-substituted-1,3,4-oxadiazoles known to inhibit PARP in cancer cells was synthesized by a green protocol. Furthermore, the cytotoxic effects of these compounds were evaluated in human MCF-7 breast cancer (BC) cells, which revealed that the compound 2-(3-bromo-4-nitrophenyl)-5-(1-methyl-1H-indazol-3-yl)-1,3,4-oxadiazole (8) inhibited viability with an IC50 value of 1.57 μM. Since the oxadiazole structure was extensively used in medicinal chemistry applications, the reported environment-friendly protocol was superior to the conventional method. Further, computational mechanistic studies revealed that the oxadiazole ring formation occurred spontaneously when compared to the conventional method. Additionally, the in silico bioinformatic studies of oxadiazole binding towards PARP1 showed that compound 8 could bind to PARP1 with higher binding energy (BE) of −7.29 kcal/mol when compound to compound 5s (BE = −7.17 kcal/mol), a known PARP cleavage oxadiazole structure (2-(3,4-Dimethoxybenzyl)-5-(3-(2-fluoro-3-methylpyridin-4-yl)phenyl)-1,3,4-oxadiazole) indicative of the improvement in the optimization process. In conclusion, a newer indazolyl-oxadiazole compound is reported, which could serve as a lead in developing PARP inhibitors in BC cells.

Abstract:A series of 1H‐benzimidazole‐4‐carboxamide PARP‐1 inhibitors using different saturated nitrogen‐containing heterocycles as linker groups was designed synthesized and evaluated its anti‐proliferation activities of MDA‐MB‐436 cell to obtain the preferred structures of linker group and terminal group in the AD site. PARP‐1 inhibitors have been successfully applied in the clinical treatment of various cancer. Side effects and drug resistant cases were reported, and more effective PARP‐1 inhibitors were required. However, studies on the AD site of PARP‐1 inhibitors are currently incomplete. Therefore, to synthesize more potential candidate PARP‐1 inhibitors and disclose some AD site SAR of the PARP‐1 inhibitors, herein, a series of 2‐phenyl‐benzimidazole‐4‐carboxamide derivatives using different saturated nitrogen‐contained heterocycles as linker group (6a‐6t) have been designed, synthesized and evaluated PARP‐1 inhibitory activity and proliferation inhibitory against BRCA‐1 mutant MDA‐MB‐436 cell line in vitro. The results showed 6b (IC50 = 8.65 nM) exhibited the most PARP‐1 enzyme inhibitory activity comparable with Veliparib (IC50 = 15.54 nM) and Olaparib (IC50 =2.77 nM); 6m exhibited the strongest MDA‐MB‐436 cell anti‐proliferation activity (IC50=25.36±6.06 μM) comparable with Olaparib (IC50 = 23.89±3.81 μM). The compounds 6b, 6r, and 6m could be potential candidates for effective PARP‐1 inhibitors and valuable for further optimization. The analysis of activity data also showed that the holistically anti‐proliferation activity of the 1,4‐diazepane group was about~2 fold than that of the piperazine group. Meanwhile, the terminal 3‐methyl‐furanyl group exhibited the most robust PARP‐1 inhibitory and anti‐proliferation activity. It's hoped that the results could benefitable for further optimization of PARP‐1 inhibitors. Furthermore, we note that some compounds (6d,6g,6n,6p,6s) showed poor PARP‐1 inhibitory (>500 nM) but relatively good anti‐proliferation activity, which indicates the proliferation inhibitory mechanism against MDA‐MB‐436 cell line was worth investigating in‐depth.

Abstract:A series of pyridazino[3,4,5-de]quinazolin-3(2H)-one derivatives were designed and synthesized as PARP-1 inhibitors. Most of the synthesized compounds showed good inhibitory activities of PARP-1 and four of them achieved at the IC50 values ranging from 0.0914 μM to 0.244 μM. Two compounds, 1a and 1b, were further tested for their neuroprotective effect in the PC12 cell model injured by H2O2 and both of them exhibited excellent activities.

Abstract:ctive: In this study, we further explored the discovery of novel PARP-1 inhibitors based on previous studies. We designed and synthesized 6 compounds with different structures, which further enriched the structural types of novel PARP-1 inhibitors. Methods: Firstly, the structures of all target compounds were determined by 1 H NMR, 13 C NMR, and high resolution MS. Next, the target compounds were evaluated for enzyme activity in vitro . Finally, ADMET prediction and molecular docking studies were used for experimental analysis. Results and Discussion: Compound ( S10a ) (IC 50 = 23.74 nM) was found to have PARP-1 enzyme inhibitory activity similar to Rucaparib (IC 50 = 23.88 nM). We also predicted the ADMET activity of all compounds, and their corresponding index values were similar to those of the reference drug Rucaparib. The molecular docking studies showed that compounds ( X10a ) and ( S10a ) could penetrate deep into the active pocket. Conclusions: Overall, based on the above analysis, the compounds reported in this paper have better research potential, and are expected to be the lead compounds for the discovery of novel PARP-1 inhibitors.

Abstract:Poly(ADP-ribose) polymerase (PARP) belongs to family of nuclear proteins having a critical role in base excision DNA repair system by catalysing the process of poly(ADP-ribosyl)ation leading to cell proliferation. The role of PARP inhibitors as anticancer agents was accepted after the approval of olaparib for the treatment of BRCA mutation-induced ovarian cancer. A combined ligand-based and receptor-based approach has been reported here for identification of PARP-1 inhibitors. Seventy one analogues of phthalazinone and 4-carboxamide benzimidazole showing activities against PARP-1 were used to develop ligand-based pharmacophore hypothesis using PHASE. The five-point pharmacophore hypothesis ADHRR.1031 was developed, and atom-based 3D-QSAR studies were carried. The developed 3D QSAR model has good correlation coefficient value (R2 = 0.952) and cross validated with correlation coefficient value (Q2 = 0.764). To further ascertain the structural requirement for PARP-1 inhibition, molecular interaction studies of inhibitors with PARP-1 were carried out by performing docking studies using Glide 5.6. Hydrogen bond interaction with Gly202 and Ser243 is an important interaction observed by the carboxyl amide group. The pharmacophore model depicts the importance of the hydrogen bond donor and acceptor nature of the carboxyl amide group. Further multistage virtual screening was performed to screen molecules with possible PARP-1 inhibition. Initial screening was performed based on pharmacophoric features followed by docking that consisted of HTVS, SP, and XP docking and final optimisation by Prime MM-GBSA calculation. The screened molecules were further scrutinised for their ADME-toxicity properties.

Carbamoylation at C-8 position of natural 3-arylcoumarin scaffold for the discovery of novel PARP-1 inhibitors with potent anticancer activity

Abstract:Structural modification based on natural privileged scaffolds has proven to be an attractive approach to generate potential antitumor candidates with high potency and specific targeting. As a continuation of our efforts to identify potent PARP-1 inhibitors, natural 3-arylcoumarin scaffold was served as the starting point for the construction of novel structural unit for PARP-1 inhibition. Herein, a series of novel 8-carbamyl-3-arylcoumarin derivatives were designed and synthesized. The antiproliferative activities of target compounds against four BRCA-mutated cancer cells (SUM149PT, HCC1937, MDA-MB-436 and Capan-1) were evaluated. Among them, compound 9b exhibited excellent antiproliferative effects against SUM149PT, HCC1937 and Capan-1 cells with IC50 values of 0.62, 1.91 and 4.26 μM, respectively. Moreover, 9b could significantly inhibit the intracellular PARP-1/2 activity in SUM149PT cells with IC50 values of 2.53 nM and 6.45 nM, respectively. Further mechanism studies revealed that 9b could aggravate DNA double-strand breaks, increase ROS production, decrease mitochondrial membrane potential, arrest cell cycle at G2/M phase and ultimately induce apoptosis in SUM149PT cells. In addition, molecular docking study demonstrated that the binding mode of 9b with PARP-1 was similar to that of niraparib, forming multiple hydrogen bond interactions with the active site of PARP-1. Taken together, these findings suggest that 8-carbamyl-3-arylcoumarin scaffold could serve as an effective structural unit for PARP-1 inhibition and offer a valuable paradigm for the structural modification of natural products.

Abstract:Based on the reported synthetic lethality of the combination of PARP inhibitor olaparib with the natural product alantolactone, we designed several series of new PARP1 inhibitors by structurally merging both compounds into a single hybrid compound. Among them, compounds 20e and 25a displayed not only high biochemical activity (IC50 = 2.99 nM and 5.91 nM vs 11.36 nM), but also higher inhibitory effects against proliferation of BRCA1-deficient UWB1.289 cells than olaparib (IC50 = 0.27 μM and 0.41 μM vs 0.66 μM). Much weak activity was observed in BRCA1 wild-type human fetal lung IMR-90 and WI-38 cells (IC50s > 10 μM). Treatment with compounds 20e and 25a was found to induce increased levels of γH2AX in a concentration-dependent manner in both MDA-MB-436 and Capan-1 cells to a degree comparable with that of olaparib. Further mechanism study indicated that these compounds activated the cell cycle checkpoints, and subsequently induced G2/M arrest and apoptosis. The results validated that merging PARP inhibitors with other DNA-damage related compounds would produce more potent PARP inhibitors for anticancer studies. However, the poor aqueous solubility and low cell penetration of the current hybrid compounds call for further structural optimization.

Abstract:Poly(ADP-ribose) polymerase-1 (PARP-1) is a new potential target for anticancer drug discovery. A series of bromophenol-thiosemicarbazone hybrids as PARP-1 inhibitors were designed, synthesized, and evaluated for their antitumor activities. Among them, the most promising compound, 11, showed excellent selective PARP-1 inhibitory activity (IC50 = 29.5 nM) over PARP-2 (IC50 > 1000 nM) and potent anticancer activities toward the SK-OV-3, Bel-7402 and HepG2 cancer cell lines (IC50 = 2.39, 5.45, and 4.60 μM), along with inhibition of tumor growth in an in vivo SK-OV-3 cell xenograft model. Further study demonstrated that compound 11 played an antitumor role through multiple anticancer mechanisms, including the induction of apoptosis and cell cycle arrest, cellular accumulation of DNA double-strand breaks, DNA repair alterations, inhibition of H2O2-triggered PARylation, antiproliferative effects via the production of cytotoxic reactive oxygen species, and autophagy. In addition, compound 11 displayed good pharmacokinetic characteristics and favorable safety. These observations demonstrate that compound 11 may serve as a lead compound for the discovery of new anticancer drugs.

Abstract:Poly(ADP-ribose) polymerase-1 (PARP1) is an enzyme that catalyzes the polymerization of ADP-ribose units to target proteins, and it is a potential target for anti-cancer drug discovery, especially for BRAC1/2 mutated tumors. In this study, a series of 2-aminoimidazole Lissodendrins B derivatives were designed, synthesized, and evaluated as PARP1 inhibitors. We found that compound D3 is better due to its PARP enzyme inhibitory activity and in vitro anti-cancer activity compared with other tested compounds. It could inhibit PARP1 enzymatic activity (IC50 = 17.46 μM) in the non-cell system and BRCA1-deficient HCC1937 and MDA-MB-436 cells growth (IC50 = 17.81 and 12.63 μM, respectively). Further study demonstrated that compound D3 inhibits tumor growth through multiple mechanisms, such as reduction of PARylation, accumulation of cellular DNA double-strand breaks, induction of G2/M cell cycle arrest, and subsequent apoptosis of BRCA1-deficient cells. Besides, the molecular docking study also confirmed that compound D3 could effectively occupy the active pocket of PARP1. Our findings provide a new skeleton structure for PARP1 inhibitor, and the results suggested that the compound D3 may serve as a potential lead compound to develop novel PARP1 inhibitors for cancer therapy.

Design, synthesis and antitumor activities of phthalazinone derivatives as PARP-1 inhibitors and PARP-1/HDAC-1 inhibitors

Abstract:In recent years, poly(ADP-ribose)polymerase-1 (PARP-1) and histone deacetylase (HDAC) have emerged as significant targets in tumor therapy, garnering widespread attention. In this study, we designed and synthesized two novel phthalazinone PARP-1 inhibitors and dual PARP-1/HDAC-1 inhibitors, named DLC-1-46 containing dithiocarboxylate fragments and DLC-47-63 containing hydroxamic acid fragments, and evaluated their inhibitory activities on enzymes and cells. Among the PARP-1 inhibitors, most compounds exhibited high inhibitory activity against the PARP-1 enzyme, with DLC-1-6 being particularly notable, showing IC50 values <0.2 nM. Notably, DLC-1 demonstrated significant anti-proliferative activity, with IC50 values for inhibiting the proliferation of MDA-MB-436, MDA-MB-231, and MCF-7 cells reaching 0.08, 26.39, and 1.01 μM, respectively. Further investigation revealed that DLC-1 arrested MDA-MB-231 cells in the G1 phase and induced apoptosis in a dose-dependent manner. Among the designed dual PARP-1/HDAC-1 inhibitors, several compounds exhibited potent dual-target inhibitory activity, with DLC-49 displaying IC50 values of 0.53 nM and 17 nM for PARP-1 and HDAC-1, respectively. DLC-50 demonstrated the most potent anti-proliferative activity, with IC50 values for inhibiting the proliferation of MDA-MB-436, MDA-MB-231, and MCF-7 cells at 0.30, 2.70, and 2.41 μM, respectively. Cell cycle arrest and apoptosis assays indicated that DLC-50 arrested the cell cycle in the G2 phase and induced apoptosis in HCT-116 cells. Our findings present a novel avenue for further exploration of PARP-1 inhibitors and dual PARP-1/HDAC-1 inhibitors.

Abstract:Polypharmacology has emerged as a promising approach to drug discovery, especially antitumor drug. This study reports the design, synthesis, and biological evaluation of novel phosphatidylinositol 3-kinases (PI3Ks) and histone deacetylases (HDACs) dual inhibitors on the basis of GSK2126458 under clinical evaluation and vorinostat approved. Among these hybrid molecules, GYB-4 and GYB-5 possessed potent inhibition against both PI3K alpha (1.0 and 1.3 nmol/L, respectively) and HDAC1 (4.2 and 4.8 nmol/L, respectively). Antiproliferative assays with HCT116, PC3 and A2780 cell lines subsequently were performed. The structure-activity relationship study will guide to optimization of dual PI3K and HDAC inhibitors.

Abstract:Both poly(ADP-ribose)polymerase-1 (PARP-1) and histone deacetylase (HDAC) are important antitumor targets and have attracted extensive attention. In this work, a total of fourteen PARP-1/HDAC dual targeting inhibitors were designed and synthesized using either benzopyrazole or benzimidazole as core structures. Two leading compounds 1-8-6 and 1-8-7 were proven to be dual targeting inhibitors of PARP-1 and HDAC6, and showed high antiproliferative activities against six human cancer cell lines with IC50 values in micromole range. Moreover, compounds 1-8-6 and 1-8-7 could impair tumor cell proliferation in 48 h and 72 h with much higher potency than co-treatment of Olaparib and Tubastatin A. 1-8-6 displayed remarkable anti-migration and anti-angiogenesis activities. Meanwhile, western blot experiment result showed that 1-8-6 was able to heighten expression level of acetylated alpha-tubulin with marginal effects to acetylated histones H3 and H4. Finally, docking simulation work showed that 1-8-6 could fit into the active sites of PARP-1 and HDAC6. All results indicated that 1-8-6 is a promising candidate for further preclinical studies.
